Providing an anodic potential in a bio‐electrochemical system to the obligate aerobe Pseudomonas putida enables anaerobic survival and allows the cells to overcome redox imbalances. In this setup, the bacteria could be exploited to produce chemicals via oxidative pathways at high yield. However, the absence of anaerobic growth and low carbon turnover rates remain as obstacles for the application of such an electro‐fermentation technology. Growth and carbon turnover start with carbon uptake into the periplasm and cytosol. P. putida KT2440 has three native transporting systems for glucose, each differing in energy and redox demand. This architecture previously led to the hypothesis that internal redox and energy constraints ultimately limit cytoplasmic carbon utilization in a bio‐electrochemical system. However, it remains largely unclear which uptake route is predominantly used by P. putida under electro‐fermentative conditions. To elucidate this, we created three gene deletion mutants of P. putida KT2440, forcing the cells to exclusively utilize one of the routes. When grown in a bio‐electrochemical system, the pathway mutants were heavily affected in terms of sugar consumption, current output and product formation. Surprisingly, however, we found that about half of the acetate formed in the cytoplasm originated from carbon that was put into the system via the inoculation biomass, while the other half came from the consumption of substrate. The deletion of individual sugar uptake routes did not alter significantly the secreted acetate concentrations among different strains even with different carbon sources. This means that the stoichiometry of the sugar uptake routes is not a limiting factor during electro‐fermentation and that the low rates might be caused by other reasons, for example energy limitations or a yet‐to‐be‐identified oxygen‐dependent regulatory mechanism. Redox mediators are commonly used in microbial electrochemical systems to enable or enhance the electron transfer between microorganisms and electrodes. In recent studies, new insights into the mechanism of mediated extracellular electron transfer were gained, but some questions remain unanswered. In this review, some of the most outstanding research questions regarding the use of redox mediators in microbial electrochemical systems were discussed. These included the recycling of artificial and natural redox mediators, limitations in electron transfer rates by mediator turnover, metabolic burden, membrane permeability, and the putative interaction sites between commonly used redox mediators and the proteins of the electron transport chain of diverse electroactive microorganisms. To simplify the planning of mediator‐based bioelectrochemical systems, these molecular interaction sites were defined by their redox potential and are assigned to redox mediators, known or hypothesized to be able to transfer electrons from or to the specific interaction site. Phosphatases are a diverse family of enzymes, comprising at least 10 distinct protein folds. Like most other enzyme families, many have sequence variations that predict an impairment or loss of catalytic activity classifying them as pseudophosphatases. Research on pseudoenzymes is an emerging area of interest, with new biological functions repurposed from catalytically active relatives. Here, we provide an overview of the pseudophosphatases identified to date in all major phosphatase families. We will highlight the degeneration of the various catalytic sequence motifs and discuss the challenges associated with the experimental determination of catalytic inactivity. Objective: Childhood cancer survivors are at high risk for late effects. Regular attendance to long-term follow-up care is recommended and helps monitoring survivors' health. Using the theory of planned behavior, we aimed to (1) investigate the predictors of the intention to attend follow-up care, and (2) examine the associations between perceived control and behavioral intention with actual follow-up care attendance in Swiss childhood cancer survivors.Methods: We conducted a questionnaire survey in Swiss childhood cancer survivors (diagnosed with cancer aged <16 years between 1990 and 2005; ≥5 years since diagnosis). We assessed theory of planned behavior-related predictors (attitude, subjective norm, perceived control), intention to attend follow-up care, and actual attendance. We applied structural equation modeling to investigate predictors of intention, and logistic regression models to study the association between intention and actual attendance.Results: Of 299 responders (166 [55.5%] females), 145 (48.5%) reported attending follow-up care. We found that subjective norm, ie, survivors' perceived social pressure and support (coef = 0.90, P < 0.001), predicted the intention to attend follow-up; attitude and perceived control did not. Perceived control (OR = 1.58, 95%CI:1.04-2.41) and intention to attend follow-up (OR = 6.43, 95%CI:4.21-9.81) were positively associated with attendance.Conclusions: To increase attendance, an effort should be made to sensitize partners, friends, parents, and health care professionals on their important role in supporting survivors regarding follow-up care. Abstract: Activation of AMPK has been associated with pro‐atrophic signaling in muscle. However, AMPK also has anti‐inflammatory effects, suggesting that in cachexia, a syndrome of inflammatory‐driven muscle wasting, AMPK activation could be beneficial. Here we show that the AMPK agonist AICAR suppresses IFNγ/TNFα‐induced atrophy, while the mitochondrial inhibitor metformin does not. IFNγ/TNFα impair mitochondrial oxidative respiration in myotubes and promote a metabolic shift to aerobic glycolysis, similarly to metformin. In contrast, AICAR partially restored metabolic function. The effects of AICAR were prevented by the AMPK inhibitor Compound C and were reproduced with A‐769662, a specific AMPK activator. AICAR and A‐769662 co‐treatment was found to be synergistic, suggesting that the anti‐cachectic effects of these drugs are mediated through AMPK activation. AICAR spared muscle mass in mouse models of cancer and LPS induced atrophy. Together, our findings suggest a dual function for AMPK during inflammation‐driven atrophy, wherein it can play a protective role when activated exogenously early in disease progression, but may contribute to anabolic suppression and atrophy when activated later through mitochondrial dysfunction and subsequent metabolic stress. Abstract: Cellular senescence is a physiological response by which an organism halts the proliferation of potentially harmful and damaged cells. However, the accumulation of senescent cells over time can become deleterious leading to diseases and physiological decline. Our data reveal a novel interplay between senescence and the stress response that affects both the progression of senescence and the behavior of senescent cells. We show that constitutive exposure to stress induces the formation of stress granules (SGs) in proliferative and presenescent cells, but not in fully senescent cells. Stress granule assembly alone is sufficient to decrease the number of senescent cells without affecting the expression of bona fide senescence markers. SG‐mediated inhibition of senescence is associated with the recruitment of the plasminogen activator inhibitor‐1 (PAI‐1), a known promoter of senescence, to these entities. PAI‐1 localization to SGs increases the translocation of cyclin D1 to the nucleus, promotes RB phosphorylation, and maintains a proliferative, non‐senescent state. Together, our data indicate that SGs may be targets of intervention to modulate senescence in order to impair or prevent its deleterious effects. [ABSTRACT FROM AUTHOR]

In accordance with simulation theories of empathy, the somatosensory cortex is involved in the perception of pain of others. Cognitive processes, like perspective taking, can alter empathy-related activity within the somatosensory cortex. The current study investigates whether this modulation is caused by the imagined sensation of pain or by the cognitive load of a perspective-taking task. Applying a within-subject design, participants (N = 30) watched pictures of painful and nonpainful actions, while imagining reduced, normal, or increased pain perception of the observed individual. Mu activity (8-13 Hz), which is inversely correlated with sensorimotor-cortex activity, was measured via EEG. To calculate mu activity (central electrodes) and alpha activity (occipital electrodes), which served as a control for effects of cognitive load, a fast Fourier transform was applied. Mu suppression linearly increased from reduced to normal to increased imagined pain (p<.05), while alpha activity was unaffected by the imagined pain (p>.80). Suppression of the 8-13 Hz band at central and occipital electrodes was stronger in response to painful actions compared to nonpainful actions (p<.01). These results indicate that modulation of mu activity through perspective taking reflects the imagined pain intensity and not the cognitive load induced by the task. [ABSTRACT FROM AUTHOR]

Abstract: TA-TMA is a pathology that occurs after allogenic HSC transplantation with an incidence of 4-13%, and represents one of the most severe vascular damage related with this therapy. We report here the case of a nine-yr-old girl suffering from a severe refractory aplastic anemia who received an unrelated, 9/10 HLA-matched HSC. Soon after transplantation, the patient developed a graft-versus-host disease (GvHD), a TA-TMA, and renal insufficiency. These pathologies remained refractory to the various treatments undertaken and required several hospitalizations in the intensive care unit. On day 106 post-HSC transfusion, after several episodes of intensive care, the patient was infused with mismatched, third-party MSCs. Schizocyte levels rapidly decreased after MSC infusion, and two wk later, most biological parameters returned to normal. Erythrocyte and thrombocyte transfusions were discontinued, and the patient remained stable for 10 wk. Thereafter, TA-TMA symptoms, viral reactivation, pleural and cardiac effusions reappeared and lead to the death of the patient. Our observations suggest that allogenic MSC infusion may decrease the symptoms of TA-TMA, but further investigation is required to determine how and when MSC should be infused to develop a long-lasting protective effect. Summary Remarkable morphological diversity of virus-like particles was observed by transmission electron microscopy in a hypersaline water sample from Lake Retba, Senegal. The majority of particles morphologically resembled hyperthermophilic archaeal DNA viruses isolated from extreme geothermal environments. Some hypersaline viral morphotypes have not been previously observed in nature, and less than 1% of observed particles had a head-and-tail morphology, which is typical for bacterial DNA viruses. Culture-independent analysis of the microbial diversity in the sample suggested the dominance of extremely halophilic archaea. Few of the 16S sequences corresponded to known archeal genera ( Haloquadratum, Halorubrum and Natronomonas) , whereas the majority represented novel archaeal clades. Three sequences corresponded to a new basal lineage of the haloarchaea. Bacteria belonged to four major phyla, consistent with the known diversity in saline environments. Metagenomic sequencing of DNA from the purified virus-like particles revealed very few similarities to the NCBI non-redundant database at either the nucleotide or amino acid level. Some of the identifiable virus sequences were most similar to previously described haloarchaeal viruses, but no sequence similarities were found to archaeal viruses from extreme geothermal environments. The Australian dugong ( Dugong dugon) and Florida manatee ( Trichechus manatus latirostris) are threatened species of aquatic mammals in the order Sirenia. Sirenian conservation and management actions would benefit from a more complete understanding of genetic diversity and population structure. Generally, species-specific microsatellite markers are employed in conservation genetic studies; however, robust markers can be difficult and costly to isolate. To increase the number of available markers, dugong and manatee microsatellite primers were evaluated for cross-species amplification. Furthermore, one manatee and four dugong novel primers are reported. After polymerase chain reaction optimization, 23 (92%) manatee primers successfully amplified dugong DNA, of which 11 (48%) were polymorphic. Of the 32 dugong primers tested, 27 (84%) yielded product in the manatee, of which 17 (63%) were polymorphic. Dugong and manatee primers were compared and the most informative markers were selected to create robust and informative marker-panels for each species. These cross-species microsatellite marker-panels can be employed to assess other sirenian populations and can provide beneficial information for the protection and management of these unique mammals. AIHA can complicate solid organ and bone marrow transplantation early after transplant. We describe the first case report of a 16-month-old boy with mixed type warm-acting IgM and warm IgG autoantibodies AIHA, occurring eight months after liver transplantation. This case describes the complexity of this very rare form of AIHA. It also illustrates the efficacy of rituximab in this indication, as well as the transfusion support with extremely rare blood, along with the importance of international collaboration to provide it. Florida manatees ( Trichechus manatus latirostris) are marine mammals that inhabit the coastal waters and rivers of the southeastern USA, primarily Florida. Previous studies have shown that Florida manatees have low mitochondrial DNA variability, suggesting that nuclear DNA loci are necessary for discriminatory analyses. Here we report 10 polymorphic microsatellite loci with an average of 4.2 alleles per locus, and average heterozygosity of 50.1%. It is controversial whether or not humans convey specific compounds within their body odours which can potentially affect the physiology and behaviour of others. Such compounds are called pheromones and have been discovered in many other species, including mammals. It has been suggested that humans might have a special organ within their nose that can transmit such chemosensory information. However, the evidence for this organ is highly questionable. In any case, the main olfactory system is a highly diverse system, capable of transmitting pheromonal information. So far, no single substance has been found that acts as a chemical messenger for erotic attraction. On the other hand, studies investigating the pheromonal properties of natural complex body odour have proven that it does deliver information about the sender and that it has an effect on the physiology and likely behaviour of other humans. The aim of the present study was to investigate the similarities and differences in the olfactory and visual processing of emotional stimuli in healthy subjects and in patients with major depressive disorder (MDD). Twenty-five inpatients were investigated after admission to the psychiatric clinic. Fifteen of them participated a second time, shortly before their discharge from the hospital. A group of healthy subjects, matched according to age and sex, served as a control. Chemsosensory event-related potentials (CSERPs) were recorded using the constant flow method. In addition, event-related potentials (ERPs), in response to colors and emotional slides, were obtained to control modality and emotion-specific effects. The subjects' task was to discriminate the colors (red/yellow) and odors (phenyl-ethyl alcohol = rose/isobutyraldehyde = rotten butter) according to their quality and to judge the valence of the emotional slides (IAPS slides). The EEG was recorded from 32 scalp locations. At the beginning of the therapy, visual stimulus processing was attenuated in depressive subjects at a relatively late processing level (reduced amplitudes of the P3 and pSW in response to colors and emotional slides), whereas olfactory stimulus processing had already been affected at an early level (reduced amplitudes of the P2 and P3-1 peaks in MDD patients). However, after successful medical treatment, ERPs did not differentiate between depressive patients and healthy controls. We discuss whether functional deviations within the primary olfactory cortex are responsible for the lower olfactory sensitivity, as well as for the altered emotional stimulus processing in MDD patients. The eukaryotic translation initiation factor 4E (eIF4E) plays an important role in the control of cell growth. eIF4E binds to the mRNA 5′ cap structure m7GpppN (where N is any nucleotide), and promotes ribosome binding to the mRNA in the cytoplasm. However, a fraction of eIF4E localizes to the nucleus. Here we describe the cloning and functional characterization of a new eIF4E-binding protein, referred to as 4E-T (eIF4E-Transporter). We demonstrate that 4E-T is a nucleocytoplasmic shuttling protein that contains an eIF4E-binding site, one bipartite nuclear localization signal and two leucine-rich nuclear export signals, eIF4E forms a complex with the importin αβ heterodimer only in the presence of 4E-T. Over-expression of wild-type 4E-T, but not of a mutant defective for eIF4E binding, causes the nuclear accumulation of HA-eIF4E in cells treated with leptomycin B. Taken together, these results demonstrate that the novel nucleocytoplasmic shuttling protein 4E-T mediates the nuclear import of eIF4E via the importin αβ pathway by a piggy-back mechanism. The olfactory event-related potential (OERP) has been described as being dependent on exogenous stimulus features, bur no effort has been made to examine possible endogenous determinants. We wanted to separate exogenous and endogenous components of the OERP by using an olfactory oddball paradigm. A high concentration of citral was used as the target stimulus, and a low concentration was used as the standard stimulus. Odors were presented within a constantly flowing air stream. We found that the early components of the OERP (N1, P2) are modulated by the stimulus concentration, whereas the late positive components (P3-1, P3-2) vary depending on the subjective stimulus significance and stimulus probability. It is concluded that the positive component of the OERP, which has been formerly explained by chemical and physical stimulus features, is actually determined by endogenous processes. Spatial vegetation patterns can be used as a proxies of the underlying soil and soil water conditions. The study examines the use of hyperspectral remote sensing techniques for quantifying geophysical parameters from the hyperspectral reflectance of the vegetation canopy. The best prediction of EMI and gamma-ray measurements was found by a combination of elevation and hyperspectral remote sensing information. The identification of spatial and temporal patterns of soil properties and moisture structures is an important challenge in environmental and soil monitoring as well as for soil landscape model approaches. This work examines the use of hyperspectral remote sensing techniques for quantifying geophysical parameters from the hyperspectral reflectance of the vegetation canopy. These can be used as proxies of the underlying soil and soil water conditions. Different spectral index derivatives, single band reflectance, and spectral indices from the airborne hyperspectral sensor AISA were quantified and tested in univariate and multivariate regression models for their correlation with geophysical measurements with electromagnetic induction (EMI) and gamma-ray spectrometry. The best univariate models for predicting electrical conductivity based on spectral information were based on the vertical dipole of an EM38DD with an R² = 0.54 with the spectral index Normalized Pigments Reflectance Index (NPCI) as well as for the horizontal dipole of an EM38DD with an R² = 0.65 with the spectral index NPCI. For predicting soil characteristics measured with gamma-ray spectrometry we received the best model results for γTh with an R² = 0.55 with the spectral index NPCI and γK with an R² = 0.44 with the spectral index Triangular Vegetation Index (TVI) and NPCI. The combination of variables including the geographical elevation was tested as the input for a multivariate regression analysis. For EMI and gamma-ray measurements, the "elevation" was found to be the most predictive variable and an integration of spectral indices into the elevation-based model led to only a slight improvement in the predictive power for EMI.
