Search

Assessments of climate‐change effects on ecosystem processes and services in high‐latitude regions are hindered by a lack of decision‐support tools capable of forecasting possible future landscapes. We describe a collaborative effort to develop and apply the Integrated Ecosystem Model (IEM) for Alaska and northwestern Canada to explore how climate change influences interactions among disturbance regimes, permafrost integrity, hydrology, and vegetation, and how these dynamics in turn influence resource management decisions. This process emphasizes co‐production of knowledge among decision makers, scientists, major funders, partners, and stakeholders. We highlight research findings based on IEM applications in Arctic Alaska, as well as successes and challenges of the co‐production process. The overall framework and lessons from our work with the IEM are relevant to other collaborative efforts outside the Arctic that aim to develop a decision‐support tool or an undertaking of equivalent scope. Abstract: We summarize the results of a recent interagency assessment of land carbon dynamics in Alaska, in which carbon dynamics were estimated for all major terrestrial and aquatic ecosystems for the historical period (1950–2009) and a projection period (2010–2099). Between 1950 and 2009, upland and wetland (i.e., terrestrial) ecosystems of the state gained 0.4 Tg C/yr (0.1% of net primary production, NPP), resulting in a cumulative greenhouse gas radiative forcing of 1.68 × 10−3 W/m2. The change in carbon storage is spatially variable with the region of the Northwest Boreal Landscape Conservation Cooperative (LCC) losing carbon because of fire disturbance. The combined carbon transport via various pathways through inland aquatic ecosystems of Alaska was estimated to be 41.3 Tg C/yr (17% of terrestrial NPP). During the projection period (2010–2099), carbon storage of terrestrial ecosystems of Alaska was projected to increase (22.5–70.0 Tg C/yr), primarily because of NPP increases of 10–30% associated with responses to rising atmospheric CO2, increased nitrogen cycling, and longer growing seasons. Although carbon emissions to the atmosphere from wildfire and wetland CH4 were projected to increase for all of the climate projections, the increases in NPP more than compensated for those losses at the statewide level. Carbon dynamics of terrestrial ecosystems continue to warm the climate for four of the six future projections and cool the climate for only one of the projections. The attribution analyses we conducted indicated that the response of NPP in terrestrial ecosystems to rising atmospheric CO2 (~5% per 100 ppmv CO2) saturates as CO2 increases (between approximately +150 and +450 ppmv among projections). This response, along with the expectation that permafrost thaw would be much greater and release large quantities of permafrost carbon after 2100, suggests that projected carbon gains in terrestrial ecosystems of Alaska may not be sustained. From a national perspective, inclusion of all of Alaska in greenhouse gas inventory reports would ensure better accounting of the overall greenhouse gas balance of the nation and provide a foundation for considering mitigation activities in areas that are accessible enough to support substantive deployment. Abstract: Wetlands are critical terrestrial ecosystems in Alaska, covering ~177,000 km2, an area greater than all the wetlands in the remainder of the United States. To assess the relative influence of changing climate, atmospheric carbon dioxide (CO2) concentration, and fire regime on carbon balance in wetland ecosystems of Alaska, a modeling framework that incorporates a fire disturbance model and two biogeochemical models was used. Spatially explicit simulations were conducted at 1‐km resolution for the historical period (1950–2009) and future projection period (2010–2099). Simulations estimated that wetland ecosystems of Alaska lost 175 Tg carbon (C) in the historical period. Ecosystem C storage in 2009 was 5,556 Tg, with 89% of the C stored in soils. The estimated loss of C as CO2 and biogenic methane (CH4) emissions resulted in wetlands of Alaska increasing the greenhouse gas forcing of climate warming. Simulations for the projection period were conducted for six climate change scenarios constructed from two climate models forced under three CO2 emission scenarios. Ecosystem C storage averaged among climate scenarios increased 3.94 Tg C/yr by 2099, with variability among the simulations ranging from 2.02 to 4.42 Tg C/yr. These increases were driven primarily by increases in net primary production (NPP) that were greater than losses from increased decomposition and fire. The NPP increase was driven by CO2 fertilization (~5% per 100 parts per million by volume increase) and by increases in air temperature (~1% per °C increase). Increases in air temperature were estimated to be the primary cause for a projected 47.7% mean increase in biogenic CH4 emissions among the simulations (~15% per °C increase). Ecosystem CO2 sequestration offset the increase in CH4 emissions during the 21st century to decrease the greenhouse gas forcing of climate warming. However, beyond 2100, we expect that this forcing will ultimately increase as wetland ecosystems transition from being a sink to a source of atmospheric CO2 because of (1) decreasing sensitivity of NPP to increasing atmospheric CO2, (2) increasing availability of soil C for decomposition as permafrost thaws, and (3) continued positive sensitivity of biogenic CH4 emissions to increases in soil temperature. Increasing wildfire activity in Alaska's boreal forests has led to greater fuel-reduction management. Management has been implemented to reduce wildfire spread, but the ecological impacts of these practices are poorly known. We quantified the effects of hand-thinning and shearblading on above- and belowground stand characteristics, plant species composition, carbon (C) and nitrogen (N) pools, and soil thaw across 19 sites dominated by black spruce ( Picea mariana) in interior Alaska treated 2-12 years prior to sampling. The density of deciduous tree seedlings was significantly higher in shearbladed areas compared to unmanaged forest (6.4 vs. 0.1 stems/m2), and unmanaged stands exhibited the highest mean density of conifer seedlings and layers (1.4 stems/m2). Understory plant community composition was most similar between unmanaged and thinned stands. Shearblading resulted in a near complete loss of aboveground tree biomass C pools while thinning approximately halved the C pool size (1.2 kg C/m2 compared to 3.1 kg C/m2 in unmanaged forest). Significantly smaller soil organic layer ( SOL) C and N pools were observed in shearbladed stands (3.2 kg C/m2 and 116.8 g N/m2) relative to thinned (6.0 kg C/m2 and 192.2 g N/m2) and unmanaged (5.9 kg C/m2 and 178.7 g N/m2) stands. No difference in C and N pool sizes in the uppermost 10 cm of mineral soil was observed among stand types. Total C stocks for measured pools was 2.6 kg C/m2 smaller in thinned stands and 5.8 kg C/m2 smaller in shearbladed stands when compared to unmanaged forest. Soil thaw depth averaged 13 cm deeper in thinned areas and 46 cm deeper in shearbladed areas relative to adjacent unmanaged stands, although variability was high across sites. Deeper soil thaw was linked to shallower SOL depth for unmanaged stands and both management types, however for any given SOL depth, thaw tended to be deeper in shearbladed areas compared to unmanaged forest. These findings indicate that fuel-reduction management alters plant community composition, C and N pools, and soil thaw depth, with consequences for ecosystem structure and function beyond those intended for fire management. It is important to understand how upland ecosystems of Alaska, which are estimated to occupy 84% of the state (i.e., 1,237,774 km2), are influencing and will influence state-wide carbon (C) dynamics in the face of ongoing climate change. We coupled fire disturbance and biogeochemical models to assess the relative effects of changing atmospheric carbon dioxide (CO2), climate, logging and fire regimes on the historical and future C balance of upland ecosystems for the four main Landscape Conservation Cooperatives (LCCs) of Alaska. At the end of the historical period (1950-2009) of our analysis, we estimate that upland ecosystems of Alaska store ~50 Pg C (with ~90% of the C in soils), and gained 3.26 Tg C/yr. Three of the LCCs had gains in total ecosystem C storage, while the Northwest Boreal LCC lost C (−6.01 Tg C/yr) because of increases in fire activity. Carbon exports from logging affected only the North Pacific LCC and represented less than 1% of the state's net primary production (NPP). The analysis for the future time period (2010-2099) consisted of six simulations driven by climate outputs from two climate models for three emission scenarios. Across the climate scenarios, total ecosystem C storage increased between 19.5 and 66.3 Tg C/yr, which represents 3.4% to 11.7% increase in Alaska upland's storage. We conducted additional simulations to attribute these responses to environmental changes. This analysis showed that atmospheric CO2 fertilization was the main driver of ecosystem C balance. By comparing future simulations with constant and with increasing atmospheric CO2, we estimated that the sensitivity of NPP was 4.8% per 100 ppmv, but NPP becomes less sensitive to CO2 increase throughout the 21st century. Overall, our analyses suggest that the decreasing CO2 sensitivity of NPP and the increasing sensitivity of heterotrophic respiration to air temperature, in addition to the increase in C loss from wildfires weakens the C sink from upland ecosystems of Alaska and will ultimately lead to a source of CO2 to the atmosphere beyond 2100. Modern climate change in Alaska has resulted in widespread thawing of permafrost, increased fire activity, and extensive changes in vegetation characteristics that have significant consequences for socioecological systems. Despite observations of the heightened sensitivity of these systems to change, there has not been a comprehensive assessment of factors that drive ecosystem changes throughout Alaska. Here we present research that improves our understanding of the main drivers of the spatiotemporal patterns of carbon dynamics using in situ observations, remote sensing data, and an array of modeling techniques. In the last 60 yr, Alaska has seen a large increase in mean annual air temperature (1.7°C), with the greatest warming occurring over winter and spring. Warming trends are projected to continue throughout the 21st century and will likely result in landscape-level changes to ecosystem structure and function. Wetlands, mainly bogs and fens, which are currently estimated to cover 12.5% of the landscape, strongly influence exchange of methane between Alaska's ecosystems and the atmosphere and are expected to be affected by thawing permafrost and shifts in hydrology. Simulations suggest the current proportion of near-surface (within 1 m) and deep (within 5 m) permafrost extent will be reduced by 9-74% and 33-55% by the end of the 21st century, respectively. Since 2000, an average of 678 595 ha/yr was burned, more than twice the annual average during 1950-1999. The largest increase in fire activity is projected for the boreal forest, which could result in a reduction in late-successional spruce forest (8-44%) and an increase in early-successional deciduous forest (25-113%) that would mediate future fire activity and weaken permafrost stability in the region. Climate warming will also affect vegetation communities across arctic regions, where the coverage of deciduous forest could increase (223-620%), shrub tundra may increase (4-21%), and graminoid tundra might decrease (10-24%). This study sheds light on the sensitivity of Alaska's ecosystems to change that has the potential to significantly affect local and regional carbon balance, but more research is needed to improve estimates of land-surface and subsurface properties, and to better account for ecosystem dynamics affected by a myriad of biophysical factors and interactions. Factors underlying the amplitude of exercise performance reduction at altitude and the development of high-altitude illnesses are not completely understood. To better describe these mechanisms, we assessed cardiorespiratory and tissue oxygenation responses to hypoxia in elite high-altitude climbers. Eleven high-altitude climbers were matched with 11 non-climber trained controls according to gender, age, and fitness level (maximal oxygen consumption, VO2max). Subjects performed two maximal incremental cycling tests, in normoxia and in hypoxia (inspiratory oxygen fraction: 0.12). Cardiorespiratory measurements and tissue (cerebral and muscle) oxygenation were assessed continuously. Hypoxic ventilatory and cardiac responses were determined at rest and during exercise; hypercapnic ventilatory response was determined at rest. In hypoxia, climbers exhibited similar reductions to controls in VO2max (climbers −39 ± 7% vs controls −39 ± 9%), maximal power output (−27 ± 5% vs −26 ± 4%), and arterial oxygen saturation ( Sp O2). However, climbers had lower hypoxic ventilatory response during exercise (1.7 ± 0.5 vs 2.6 ± 0.7 L/min/%; P < 0.05) and lower hypercapnic ventilatory response (1.8 ± 1.4 vs 3.8 ± 2.5 mL/min/mmHg; P < 0.05). Finally, climbers exhibited slower breathing frequency, larger tidal volume and larger muscle oxygenation index. These results suggest that elite climbers show some specific ventilatory and muscular responses to hypoxia possibly because of genetic factors or adaptation to frequent high-altitude climbing. Adaptation planning in Alaska, as in other snowy parts of the world, will require snow projections, yet snow is a challenging variable to measure, simulate and downscale. Here we describe the construction and evaluation of 771-m-resolution gridded historical and statistically downscaled projections of snow/rain partitioning for the state of Alaska at decadal temporal resolution. The method developed here uses observational data to describe the relationship between average monthly temperature and the fraction of wet days in that month receiving snow, the snow-day fraction. Regionally and seasonally specific equations were developed to accommodate variability in synoptic scale climatology of rain and snow events. These equations were then applied to gridded decadal temperature data and projections. The gridded products provide a reasonable characterization of snow-day fraction throughout the state. However, there are local deviations from the regional relationships, particularly in the topographically complex areas ringing the Gulf of Alaska and Cook Inlet. When applied to questions about changing precipitation regimes in northern, western and south-eastern Alaska, these data demonstrate the potential for marked changes from snow-dominated to mixed precipitation regimes and also exhibit a wide range of potential future conditions. Recent research has linked climate warming to global declines in caribou and reindeer (both Rangifer tarandus) populations. We hypothesize large-scale climate patterns are a contributing factor explaining why these declines are not universal. To test our hypothesis for such relationships among Alaska caribou herds, we calculated the population growth rate and percent change of four arctic herds using existing population estimates, and explored associations with indices of the Arctic Oscillation (AO) and the Pacific Decadal Oscillation (PDO). The AO, which more strongly affects eastern Alaska, was negatively associated with the population trends of the Porcupine Caribou Herd and Central Arctic Herd, the easternmost of the herds. We hypothesize that either increased snowfall or suboptimal growing conditions for summer forage plants could explain this negative relationship. Intensity of the PDO, which has greatest effects in western Alaska, was negatively associated with the growth rate of the Teshekpuk Caribou Herd in northwestern Alaska, but the Western Arctic Herd in western Alaska displayed the opposite trend. We suggest that the contrasting patterns of association relate to the spatial variability of the effects of the PDO on western and northwestern Alaska. Although predation and winter range quality have often been considered the primary causes of population variation, our results show that large-scale climate patterns may play an important role in caribou population dynamics in arctic Alaska. Our findings reveal that climate warming has not acted uniformly to reduce caribou populations globally. Interactions between vegetation and fire have the potential to overshadow direct effects of climate change on fire regimes in boreal forests of North America. We develop methods to compare sediment-charcoal records with fire regimes simulated by an ecological model, ALFRESCO (Alaskan Frame-based Ecosystem Code) and apply these methods to evaluate potential causes of a mid-Holocene fire-regime shift in boreal forests of the south-central Brooks Range, Alaska, USA. Fire-return intervals (FRIs, number of years between fires) are estimated over the past 7000 calibrated 14C years (7-0 kyr BP [before present]) from short-term variations in charcoal accumulation rates (CHARs) at three lakes, and an index of area burned is inferred from long-term CHARs at these sites. ALFRESCO simulations of FRIs and annual area burned are based on prescribed vegetation and climate for 7-5 kyr BP and 5-0 kyr BP, inferred from pollen and stomata records and qualitative paleoclimate proxies. Two sets of experiments examine potential causes of increased burning between 7-5 and 5-0 kyr BP. (I) Static-vegetation scenarios: white spruce dominates with static mean temperature and total precipitation of the growing season for 7-0 kyr BP or with decreased temperature and/or increased precipitation for 5-0 kyr BP. (2) Changed-vegetation scenarios: black spruce dominates 5-0 kyr BP, with static temperature and precipitation or decreased temperature and/or increased precipitation. Median FRIs decreased between 7-5 and 5-0 kyr BP in empirical data and changed-vegetation scenarios but remained relatively constant in static-vegetation scenarios. Median empirical and simulated FRIs are not statistically different for 7-5 kyr BP and for two changed-vegetation scenarios (temperature decrease, precipitation increase) for 5-0 kyr BP. In these scenarios, cooler temperatures or increased precipitation dampened the effect of increased landscape flammability resulting from the increase in black spruce. CHAR records and all changed-vegetation scenarios indicate long-term increases in area burned between 7-5 and 5-0 kyr BP. The similarity of CHAR and ALFRESCO results demonstrates the compatibility of these independent data sets for investigating ecological mechanisms causing past fire-regime changes. The finding that vegetation flammability was a major driver of Holocene fire regimes is consistent with other investigations that suggest that landscape fuel characteristics will mediate the direct effects of future climate change on boreal fire regimes. Aim We examined the interactive effects of mammalian herbivory and fluvial dynamics on vegetation dynamics and composition along the Tanana River in interior Alaska. Location Model parameters were obtained from field studies along the Tanana River, Alaska between Fairbanks (64°50.50′ N, 147°43.30′ W) and Manley Hot Springs (65°0.0′ N, 150°36.0′ W). Methods We used a spatially explicit model of landscape dynamics (ALFRESCO) to simulate vegetation changes on a 1-year time-step. The model was run for 250 years and was replicated 100 times. Results Increases in herbivory decreased the proportion of early successional vegetation and increased the proportion of late successional vegetation on the simulated landscape. Erosion and accretion worked as antagonists to herbivory, increasing the amount of early successional vegetation and decreasing the amount of late successional vegetation. However, the interactive effects of herbivory and erosion/accretion were especially important in determining system response, particularly in early seral vegetation types. High erosion rates, when coupled with low herbivory, greatly increased the proportion of willow on the landscape. When coupled with high herbivory, however, they greatly increased the proportion of alder on the landscape. At low levels of herbivory, alder abundance peaked at intermediate levels of erosion/accretion. Main conclusions Neither erosion/accretion nor herbivory produced consistent landscape patterns that could be predicted independently of the other. These findings underscore the importance of the interactive effects of biotic and abiotic disturbances in shaping large-scale landscape vegetation patterns in boreal floodplain ecosystems – systems traditionally thought to be driven primarily by abiotic disturbance alone. The article tracks the frequency, extent and spatial distribution of the simulated fires through the use of four different fire scenario models. Changes were associated to vegetation composition and distribution. Results suggest that shorter fire frequencies on the winter range of the Nelchina caribou herd in eastern interior Alaska will result in large decreases of available winter habitat, relative to that currently available, in both the short and long term. A 30 percent shortening of the fire frequency resulted in a 3.5-fold increase in the area burned annually and an associated 41 percent decrease in the amount of spruce-lichen forest found on the landscape.

The article explores the relationships between weather variables and the annual area burned in Alaska. The boreal forest contains roughly 40% of the world's reactive soil carbon, an amount similar to that held in the atmosphere. The biophysical phenomena affecting carbon storage and high-latitude albedo make the boreal forest an integral component of the global climate system. Fire-initiated succession underlies the biophysical factors, and there is a pressing need to characterize sensitivities and potential responses of the boreal forest disturbance regime to climatic change. The article addresses variation in the seasonality of the atmospheric circulation-fire linkage through an evaluation of both the East Pacific teleconnection field and a Pacific Decadal Oscillation index keyed to an annual fire index. It uses multiple linear regression (MLR) method. In the MLR, seven explanatory variables and an interaction term collectively explain 79% of the variability in the natural logarithm of the number of hectares burned annually by lightning-caused fires in Alaska from 1950 to 2003. Average June temperature alone explains one-third of the variability in the logarithm of annual area burned.

To understand drivers of vegetation type distribution and sensitivity to climate change.Interior Alaska.A logistic regression model was developed that predicts the potential equilibrium distribution of four major vegetation types: tundra, deciduous forest, black spruce forest and white spruce forest based on elevation, aspect, slope, drainage type, fire interval, average growing season temperature and total growing season precipitation. The model was run in three consecutive steps. The hierarchical logistic regression model was used to evaluate how scenarios of changes in temperature, precipitation and fire interval may influence the distribution of the four major vegetation types found in this region.At the first step, tundra was distinguished from forest, which was mostly driven by elevation, precipitation and south to north aspect. At the second step, forest was separated into deciduous and spruce forest, a distinction that was primarily driven by fire interval and elevation. At the third step, the identification of black vs. white spruce was driven mainly by fire interval and elevation. The model was verified for Interior Alaska, the region used to develop the model, where it predicted vegetation distribution among the steps with an accuracy of 60–83%. When the model was independently validated for north-west Canada, it predicted vegetation distribution among the steps with an accuracy of 53–85%. Black spruce remains the dominant vegetation type under all scenarios, potentially expanding most under warming coupled with increasing fire interval. White spruce is clearly limited by moisture once average growing season temperatures exceeded a critical limit (+2 °C). Deciduous forests expand their range the most when any two of the following scenarios are combined: decreasing fire interval, warming and increasing precipitation. Tundra can be replaced by forest under warming but expands under precipitation increase.The model analyses agree with current knowledge of the responses of vegetation types to climate change and provide further insight into drivers of vegetation change. The development of policies that promote ecological, economic, and cultural sustainability requires collaboration between natural and social scientists. We present a modeling approach to facilitate this communication and illustrate its application to studies of wildfire in the interior of Alaska. We distill the essence of complex fire-vegetation interactions that occur in the real world into a simplified landscape model, and describe how equally complex fire-human interactions could be incorporated into a similar modeling framework. Simulations suggest that fire suppression is likely to increase the proportion of flammable vegetation on the landscape and reduce the long-term effectiveness of wildfire suppression. Simple models that test the consequences of assumptions help natural and social scientists to communicate objectively when exploring the long-term consequences of alternative policy scenarios. SummaryUnderstanding the response of terrestrial ecosystems to climatic warming is a challenge because of the complex interactions of climate, disturbance, and recruitment across the landscape. We use a spatially explicit model (ALFRESCO) to simulate the transient response of subarctic vegetation to climatic warming on the Seward Peninsula (80 000 km2) in north-west Alaska. Model calibration efforts showed that fire ignition was less sensitive than fire spread to regional climate (temperature and precipitation). In the model simulations a warming climate led to slightly more fires and much larger fires and expansion of forest into previously treeless tundra. Vegetation and fire regime continued to change for centuries after cessation of the simulated climate warming. Flammability increased rapidly in direct response to climate warming and more gradually in response to climate-induced vegetation change. In the simulations warming caused as much as a 228% increase in the total area burned per decade, leading to an increasingly early successional and more homogenous deciduous forest-dominated landscape. A single transient 40-y drought led to the development of a novel grassland–steppe ecosystem that persisted indefinitely and caused permanent increases in fires in both the grassland and adjacent vegetation. These simulated changes in vegetation and disturbance dynamics under a warming climate have important implications for regional carbon budgets and biotic feedbacks to regional climate. The construction of artificial muscles is one of the most challenging developments in today’s biomedical science. The application of artificial muscles is focused both on the construction of orthotics and prosthetics for rehabilitation and prevention purposes and on building humanoid walking machines for robotics research. Research in biomechanics tries to explain the functioning and design of real biologicalmuscles and therefore lays the fundament for the development of functional artificialmuscles. Recently, the hyperbolic Hill-type force-velocity relation was derived from simple mechanical components. In this contribution, this theoretical yet biomechanical model is transferred to a numerical model and applied for presenting a proof-of-concept of a functional artificial muscle. Additionally, this validated theoretical model is used to determine force-velocity relations of different animal species that are based on the literature data from biological experiments.Moreover, it is shown that an antagonistic muscle actuator can help in stabilising a single inverted pendulum model in favour of a control approach using a linear torque generator. In this study, we investigated the effect of hypoxic exposure on cerebral haemodynamics at different cortical locations during exercise at the same relative intensity after 1 (D1) and 5 days (D5) at 4350 m. Eleven male subjects performed a submaximal bout of cycling exercise (6 min at 35% + 6 min at 55% + time-to-exhaustion at 75% of peak work rate achieved in the same conditions, i.e. normoxia or hypoxia at sea level) on D1 and D5. Transcranial Doppler and near-infrared spectroscopy were used to assess middle cerebral artery blood velocity and prefrontal and motor cortex oxygenation, respectively. Despite using the same relative intensity, the duration of exercise was reduced on D1 (22.7 ± 5.1 min) compared with sea level (32.2 ± 9.0 min; P < 0.001), with no improvement on D5 (20.9 ± 6.3 min; P > 0.05). Middle cerebral artery blood velocity during exercise was elevated on D1 (+18.2%) and D5 (+15.0%) compared with sea level (P < 0.001). However, prefrontal and motor cortex oxygenation was reduced on D1 and D5 compared with sea level (P < 0.001). This pattern was of similar magnitude between cortical locations, whereas the total haemoglobin concentration increased to a greater extent in the prefrontal versus motor cortex at exhaustion on D1 and D5. In contrast to our primary hypothesis, prefrontal and motor cortex oxygenation and exercise performance did not improve over 5 days at 4350 m. New Findings: What is the central question of this study? This study is the first to investigate the effects of high-altitude trekking on biventricular mechanics, including measurements of left ventricular subendocardial and subepicardial function. What is the main finding and its importance? We provide new evidence that an increased contractility and untwisting efficiency, a key element of diastolic function, probably plays a key role in preservation of cardiac function during high-altitude trekking. Persistent increased loading conditions during several weeks at high altitude might have a key role in the appearance of left or right ventricular dysfunction., Abstract: Cardiac responses to acute hypoxic exposure have been thoroughly investigated. We analysed the effects of high-altitude trekking (i.e. prolonged hypoxic exposure) on biventricular function, including the evaluation of subendocardial and subepicardial function in the left ventricle (LV). Resting evaluations of LV and right ventricular (RV) function and mechanics were assessed by speckle tracking echocardiography on 20 subjects at sea level and at high altitude (5085 m, after a 10 day ascent). Pulmonary artery systolic pressure was increased at high altitude (sea level, 13.1 ± 5.9 mmHg; high altitude, 26.6 ± 10.8 mmHg; P < 0.001). Left ventricular volumes were decreased, whereas RV volumes were increased at high altitude. Alterations in pulmonary artery systolic pressure and cardiac volumes were correlated with hypoxaemia. We observed neither RV nor LV systolic dysfunction, including analysis of LV subendocardial and subepicardial function. Left ventricular systolic strain rates were enhanced at high altitude. Transmitral and transtricuspid diastolic filling ratios were decreased at high altitude. Diastolic apical rotational rate, untwisting rate and untwisting rate/peak twist ratio (i.e. untwisting efficiency) were enhanced at high altitude. We observed no echocardiographic signs of LV and RV pathological dysfunction at rest at high altitude. In contrast, our data highlighted major changes in the LV mechanics, with an increased LV contractility and a higher untwisting efficiency at high altitude. Biventricular interaction, alterations in loading conditions and an increase in plasma catecholamine concentration might partly explain these modifications. Background Information: Tumor stroma remodeling is a key feature of malignant tumors and can promote cancer progression. Laminins are major constituents of basement membranes that physically separate the epithelium from the underlying stroma., Results: By employing mouse models expressing high and low levels of the laminin α1 chain (LMα1), we highlighted its implication in a tumor-stroma crosstalk, thus leading to increased colon tumor incidence, angiogenesis and tumor growth. The underlying mechanism involves attraction of carcinoma-associated fibroblasts by LMα1, VEGFA expression triggered by the complex integrin α2β1-CXCR4 and binding of VEGFA to LM-111, which in turn promotes angiogenesis, tumor cell survival and proliferation. A gene signature comprising LAMA1, ITGB1, ITGA2, CXCR4 and VEGFA has negative predictive value in colon cancer., Conclusions: Together, we have identified VEGFA, CXCR4 and α2β1 integrin downstream of LMα1 in colon cancer as of bad prognostic value for patient survival., Significance: This information opens novel opportunities for diagnosis and treatment of colon cancer., (© 2018 Société Française des Microscopies and Société de Biologie Cellulaire de France. This study tested the hypothesis that during fatiguing quadriceps exercise, supraspinal fatigue develops late, is associated with both increased corticospinal excitability and inhibition and recovers quickly. Eight subjects performed 20 s contractions [15 s at 50% maximal voluntary contraction (MVC) followed by 5 s MVC] separated by a 10 s rest period until task failure. Transcranial magnetic stimulation (TMS) and electrical femoral nerve stimulation (PNS) were delivered ∼ 2 s apart during 50% MVC, during MVC and after MVC in relaxed muscle. Voluntary activation was assessed by TMS (VATMS) immediately before and after exercise and then three times over a 6 min recovery period. During exercise, MVC and twitch force evoked by PNS in relaxed muscle decreased progressively to 48 ± 8 and 36 ± 16% of control values, respectively (both P < 0.01). Significant changes in voluntary activation assessed by PNS and twitch evoked by TMS during MVC were observed during the last quarter of exercise only (from 96.4 ± 1.7 to 86 ± 13%, P = 0.03 and from 0.76 ± 0.8 to 4.9 ± 4.7% MVC, P = 0.02, from baseline to task failure, respectively). The TMS-induced silent period increased linearly during both MVC (by ∼ 79 ms) and 50% MVC (by ∼ 63 ms; both P < 0.01). Motor-evoked potential amplitude did not change during the protocol at any force levels. Both silent period and VATMS recovered within 2 min postexercise, whereas MVC and twitch force evoked by PNS in relaxed muscle recovered to only 84 ± 9 and 73 ± 17% of control values 6 min after exercise, respectively. In conclusion, high-intensity single-joint quadriceps exercise induces supraspinal fatigue near task failure, with increased intracortical inhibition and, in contrast to previous upper-limb results, unchanged corticospinal excitability. These changes recover rapidly after task failure, emphasizing the need to measure corticospinal adaptations immediately at task failure to avoid underestimation of exercise-induced corticospinal changes., (© 2014 The Authors.
