Moral judgment is politically situational—people are more forgiving of transgressive copartisans and more likely to behave punitively and unethically toward political opponents. Such differences are widely observed, but not fully explained. If moral values are nonnegotiable first‐principle beliefs about right and wrong, why do similar transgressions elicit different moral judgment in the personal and political realm? We argue this pattern arises from the same forces intuitionist frameworks of moral psychology use to explain the origins of morality: the adaptive need to suppress individual behavior to ensure ingroup success. We hypothesize ingroups serve as moral boundaries, that the relative tight constraints morality exerts over ingroup relations loosen in competitive group environments because doing so also serves ingroup interests. We find support for this hypothesis in four independent samples and also find that group antipathy—internalized dislike of the outgroup—pushes personal and political moral boundaries farther apart. Intracellular plant defense against pathogens is mediated by a class of disease resistance genes known as NB‐LRRs or NLRs (R genes). Many of the diseases these genes protect against are more prevalent in regions of higher rainfall, which provide better growth conditions for the pathogens. As such, we expect a higher selective pressure for the maintenance and proliferation of R genes in plants adapted to wetter conditions. In this study, we enriched libraries for R genes using RenSeq from baits primarily developed from the common sunflower (Helianthus annuus) reference genome. We sequenced the R gene libraries of Silphium integrifolium Michx, a perennial relative of sunflower, from 12 prairie remnants across a rainfall gradient in the Central Plains of the United States, with both Illumina short‐read (n = 99) and PacBio long‐read (n = 10) approaches. We found a positive relationship between the mean effective annual precipitation of a plant's source prairie remnant and the number of R genes in its genome, consistent with intensity of plant pathogen coevolution increasing with precipitation. Advancing the seismic resilience of building systems is an active area of research in earthquake engineering. Ensuring safe egress in and out of buildings during extreme events, such as an earthquake, is essential to supporting this effort. To this end, understanding the seismic response of stairs facilitates the robust design of egress systems to ensure they can remain operable after an earthquake. From prior earthquake events and physical experiments, it is understood that stairs with a flight to landing fixed connection at multiple levels within a building are prone to damage. In addition, the stair system with flight to landing fixed attachments may affect the dynamic behavior of the building. To accommodate seismic inter‐story drifts, a kinematically free connection between the stairs and landing has been proposed. Herein this connection is referred to as a drift‐compatible stair connection. To investigate and aid in the design of such a connection, a unique set of shake table experiments were conducted at the University of Nevada, Reno. In this paper, an overview of these tests is presented, and a high‐fidelity finite element model of the tested stair system is used to predict the responses measured during these experiments. Developed in Abaqus, the robustness of the modeled stair unit is investigated considering a variety of contrasting connections, namely, drift‐compatible connections, fixed ends and one end fixed and the other free. Results from these numerical simulations offer guidance towards development of simplified models of multi‐level stair subsystems. Such models are needed when investigating seismic resilience of building systems across a wider range of hazard levels. Furthermore, best practices observed utilizing the models developed and evaluated herein against experimental data will be useful for subsequent analysis of larger stair tower models, such as the 10‐story stair system implemented in the NHERI Tall Wood mass timber building with post‐tensioned rocking walls, conducted in 2023 at the UC San Diego Large High‐Performance Outdoor Shake Table. Habitat loss is rarely truly random and often occurs selectively with respect to the plant species comprising the habitat. Such selective habitat removal that decreases plant species diversity, that is, habitat simplification or homogenization, may have two negative effects on other species. First, the reduction in plant community size (number of individuals) represents habitat loss for species at higher trophic levels who use plants as habitat. Second, when plants are removed selectively, the resulting habitat simplification decreases the diversity of resources available to species at higher trophic levels. It follows that habitat loss combined with simplification will reduce biodiversity more than habitat loss without simplification. To test this, we experimentally implemented two types of habitat loss at the plant community level and compared biodiversity of resident arthropods between habitat loss types. In the first type of habitat loss, we reduced habitats by 50% nonselectively, maintaining original relative abundance and diversity of plant species and therefore habitat and resource diversity for arthropods. In the second type of habitat loss, we reduced habitats by 50% selectively, removing all but one common plant species, dramatically simplifying habitat and resources for arthropods. We replicated this experiment across three common plant species: Asclepias tuberosa, Solidago altissima, and Baptisia alba. While habitat loss with simplification reduced arthropod species richness compared with habitat loss without simplification, neither type of habitat loss affected diversity, measured as effective number of species (ENS), or species evenness as compared with controls. Instead, differences in ENS and evenness were explained by the identity of the common plant species. Our results indicate that the quality of remaining habitat, in our case plant species identity, may be more important for multi‐trophic diversity than habitat diversity per se. Background: Immunoglobulin (IG) therapy is widely used to treat primary and secondary immune deficiencies and as immunomodulatory agent for various disorders. There is great concern that shortages of IG may rise, potentially affecting medical treatment options. Study Design and Methods: An international survey was developed to study how intravenous immunoglobulins (IVIGs) are used and managed within hospitals in case of shortages. Study data were collected and managed using REDCap electronic data capture tools hosted by the Biomedical Excellence for Safer Transfusion (BEST) Collaborative. The survey was directed to hospital pharmacists and blood bank transfusion professionals and disseminated through members of the BEST Collaborative network. Results: Survey respondents from institutions in the USA, Canada, Europe, Japan, and Australia (n = 13) confirmed that the primary specialties utilizing IG are neurology, hematology, and immunology. More than 60% of respondents reported IG supply shortages, but mitigation strategies were not well developed. Discussion: As IG is the leading driver in plasma demand, more studies are needed to understand current and future demand for IG from the clinical perspective. Oat (Avena sativa L.) production worldwide is constrained by crown rust (caused by Puccinia coronata f. sp. avenae), which can cause significant yield losses. The disease is often controlled by spraying fungicides or planting resistant cultivars. Developing host resistance, however, is a challenge due to the high genetic variability of the pathogen. Race‐specific resistance usually succumbs to new races in just a few years. As such, the USDA‐ARS Cereal Disease Laboratory developed mapping populations to identify adult plant resistance (APR) loci from Avena sativa donors. Resistant lines from the mapping populations were selected and crossed with buckthorn (Rhamnus cathartica L.) nursery selection lines BT1020‐1‐1 and BT1021‐1‐1, which possess a different gene for crown rust resistance derived from Avena strigosa. From the crosses, CDL‐111 (Reg. no. GP‐122, PI 702639) and CDL‐167 (Reg. no. GP‐123, PI 702640), both containing three APR quantitative trait loci, were selected as germplasm for resistance breeding. High‐throughput markers for selection were developed and implemented in pyramiding the APR loci. The modeling of genotype × environment interactions (GEI) is important to understand how new crops perform in different environments and management systems. Naked barley (Hordeum vulgare L.) is a type of barley where the hull threshes freely from the grain and can be used for multiple end uses, including food, malt, and animal feed. We examined the performance of a winter naked barley trial grown in organic conditions across three regions and 3 years in northern latitude regions of the United States. We recorded yield, test weight, heading date, plant height, threshability, winter survival, barley stripe rust (Puccinia striiformis f. sp. hordei Westend), barley scald (Rhynchosporium commune Zaffarano, McDonald and Linde sp. nov), and spot blotch (Cochliobolus sativus, anamorph Bipolaris sorokiniana (S. Ito & Kurib.). Barley stripe rust, plant height, heading date, preharvest sprouting, and threshability had high heritability and low GEI. Grain yield, test weight, scald, and winter survival had high GEI and were analyzed using the additive main effect and multiplicative interaction (AMMI) model, genotype plus GEI biplot (GGE), and Finlay–Wilkinson (FW) stability analysis. The environmental patterns explained by the AMMI and GGE models indicated that selection for high GEI traits should occur within target environments and across multiple years within the target environments. Environmental groups were separated by Oregon as the first group and Wisconsin and New York as the second group. We prove local Calabi and higher order estimates for solutions to the continuity equation introduced by La Nave–Tian and extended to Hermitian metrics by Sherman–Weinkove. We apply the estimates to show that on a compact complex manifold, the Chern scalar curvature of a solution must blow up at a finite‐time singularity. Additionally, starting from certain classes of initial data on Oeljeklaus–Toma manifolds, we prove Gromov–Hausdorff and smooth convergence of the metric to a particular nonnegative (1,1)‐form as t→∞$t\rightarrow \infty$. Fusarium head blight (FHB) or scab is a devastating disease of barley that severely reduces the yield and quality of the grain. Additionally, mycotoxins produced by the causal Fusarium species can contaminate harvested grain, resulting in food safety concerns and further economic losses. In the Upper Midwest region of the United States, Fusarium graminearum is the primary causal agent, and deoxynivalenol (DON) is the main mycotoxin associated with Fusarium infection. Deployment of resistant cultivars is an important component of an integrated strategy to manage this disease. Unfortunately, few good sources of FHB resistance have been identified from the evaluation of large collections of Hordeum germplasm. Over the past 25 years, many barley mapping populations have been developed with selected resistance sources to identify the number, chromosomal position and allelic effect of quantitative trait loci (QTL) contributing to FHB resistance and DON accumulation. To consolidate the genetic data generated from 14 mapping studies that included 22 bi‐ or tri‐parental mapping populations and three genome‐wide association (GWAS) mapping panels, a consensus map was constructed that includes 4145 SNP, SSR, RFLP and AFLP markers. A meta‐analysis based on this consensus map revealed 96 QTL for FHB resistance and 57 for DON accumulation scattered across the barley genome. Many of the QTL explained a low percentage (<10%) of variation for the traits and were often found significant in only one or a few environments in multi‐year/multi‐location field trials. Moreover, many of the FHB/DON QTL mapped to chromosomal positions coincided with various agro‐morphological traits that could influence the level of disease (e.g. heading date, height, spike density, and spike angle), raising the important question of whether the former are true resistance factors or are simply the result of pleiotropy with the latter. Considering the magnitude of effect, consistency of detection across environments and independence from agro‐morphological traits, only three of 96 QTL for FHB and five of 57 QTL for DON were considered priority targets for marker‐assisted selection (MAS). In spite of the challenge for having a limited number of useful QTL for breeding, genomic selection holds promise for increasing the efficiency of developing FHB‐resistant barley cultivars, an essential component of the overall management strategy for the disease. Alkylation of one of the inner‐core nitrogen atoms is one possible approach to obtain dianionic corrole ligands, suitable for the coordination of divalent metal ions, such as PdII. Inner‐core N‐methylation can be obtained by treating the corrole with CH3I, but the reaction conditions should be optimized to limit the formation of the dimethylated derivative. Two regioisomers, the N‐21 and the N‐22 methyl derivatives are obtained from the reaction, with the first product achieved in a higher amount. Structural characterization of the reaction products evidenced the distortion induced by the introduction of the methyl groups; the N‐methylcorroles are chiral compounds, and the enantiomers were separated by chromatography, with their absolute configuration assigned by ECD computation. Palladium insertion was achieved in the case of monosubstituted corroles, but not with the dimethylated macrocycle; X‐ray characterization of the complexes showed the distortion of the macrocycles. The Pd complexes do not show luminescence emission, but are able to produce singlet oxygen upon irradiation. The PdII complexes were also inserted in human serum albumin (HSA) and dispersed in water; in this case, the protein protects the corroles from photobleaching, and a switch from the type II to the type I mechanism in reactive oxygen species (ROS) production is observed. Background: The purpose of this 6‐week, single‐blinded, randomized clinical trial was to determine if the use of an interproximal brush, with or without a tracking device, is more effective than an oral irrigator in improving interproximal probing depth (PD), clinical attachment level (CAL), plaque index (PI), gingival index (GI), bleeding on probing (BOP), and inflammatory markers. Methods: Seventy‐six patients with Stages III–IV, Grade B periodontitis and a 5–7 mm posterior interproximal PD with BOP were randomized: (1) interproximal brush alone (IB; n = 26), (2) interproximal brush with tracking device (TD; n = 23), (3) oral irrigator (OI; n = 27). Participants used devices once daily for 6 weeks. Clinical measurements (PD, CAL, PI, BOP, GI) and gingival crevicular fluid (GCF) samples were collected at baseline and 6 weeks. Results: All groups showed a significant reduction in PD and CAL (≥1.1 mm, p < 0.0001) and improvement in BOP (≥56%, p < 0.0001) and GI (≥82%, p < 0.001) at the experimental site with no differences among groups. The IB and IB+TD groups showed a significant reduction in PI (≥0.9, p ≤ 0.01). Interleukin (IL)‐1β was reduced in all groups (p = 0.006), but IB+TB more than OI (p ≤ 0.05). IL‐10 was reduced among all groups (p = 0.01), while interferon‐gamma significantly increased (p = 0.01) in all groups. Conclusions: IB and OI improved clinical parameters of PD and CAL and reduced inflammatory markers (BOP, GI, GCF IL‐1β). IB had better interproximal plaque reduction. Objective: This pilot study compared non‐medically trained surf lifesavers' (SLS) ability, after infographic training, to occlude the femoral artery using a pressure point (PP) versus an arterial tourniquet (AT). Methods: Using a crossover design, eight SLS applied PP and AT to a participant's leg to occlude the femoral artery. Arterial flow, application time and perceived difficulty were recorded. Results: PP achieved 89.7% and 50.8% blood flow reduction for PP and AT, respectively. Average application time was 50.63 and 113.5 s for PP and AT, respectively. Perceived difficulty using a Likert scale from 0 to 10 (0 being no difficulty and 10 being maximal difficulty) was 2.75 and 3.50 for PP and AT, respectively. Conclusion: Infographic‐trained SLS showed superior blood flow occlusion using PP. This pilot study will inform a larger trial for untrained beachgoers. [ABSTRACT FROM AUTHOR]

Threshability, defined here as the propensity of grains to lose their hull after harvest, is a key trait in naked barley (Hordeum vulgare L.). While threshability is a defining characteristic of naked grains and has been found to be associated with grain size and shape, its genetic architecture is poorly described. The goals of this study were to identify quantitative trait loci associated with threshability and evaluate their utility as covariates in genomic prediction models. A genome‐wide association study identified two loci on chromosomes 2H and 3H associated with threshability. The locus on chromosome 2H accounted for 9.9% of the phenotypic variance explained (PVE). The locus on chromosomes 3H accounted for 7.8% of the PVE. With effects on threshability of 0.18 and 0.29 for each marker, respectively, these markers could have a limited impact when implemented in marker‐assisted selection. Predictive ability for threshability was 0.842 using a structured genomic best linear unbiased prediction model. Incorporation of the markers with significant associations as covariates in the model did not improve predictive ability. Predictive ability was improved by the use of a multi‐trait model including grain test weight. The high predictive ability for threshability overall indicate that genomic selection would be useful in selection for threshability in naked barley. Throughout the Confessions, Augustine repeatedly complains about heresy with a special focus on the heresy he once belonged to, Manicheanism. To those of us who live in a culture in which respectable people rarely, if ever, care about religious orthodoxy to such a degree, these complaints seem rather bizarre. Despite this initial appearance, Augustine presents in the Confessions several plausible reasons for thinking heresy is sinful and, therefore, detrimental to a person's sanctity and ultimate salvation. In this paper, I argue that Augustine considers heresy sinful because it involves as many as three kinds of idolatry: loving a lie/false conception of God instead of the true God, loving one's own beliefs more than the Truth, which is God, and loving the worldly praise one receives from developing novel opinions more than God. There have been numerous calls for improved pedagogical practices in biological education, and there is a clear need for such improvements in ecology and related curricula. Most ecology‐related texts lack pedagogy and are designed to be content‐rich. National initiatives, such as Vision & Change, provide guidance on undergraduate biology education, including increasing use of evidence‐based active learning, and taking a more conceptual and science practice skills approach. Biology education research is rich with evidence‐based teaching practices, which reveal that active learning approaches implemented in thoughtful ways lead to strong learning gains relative to lecture‐based course delivery. CREATE (Consider, Read, Elucidate the hypothesis, Analyze and interpret data, Think of the next Experiment) integrates evidence‐based active pedagogical practices into one approach to STEM education that focuses heavily on the process of science and science practice skills rather than content delivery by replacing the textbook with selected journal articles. The approach focuses on deep reading and analysis of primary literature; immersing students in the literature is an advantage of the pedagogy. CREATE was developed and tested in other biological disciplines (genetics and molecular biology) that have long been at the forefront of pedagogical best practices in biology. We transformed two upper‐level undergraduate ecological courses (Conservation Biology, and Biodiversity and Ecology) into CREATE courses. We provide examples of assignments, student work, and assessments of the approach, illustrating the various ways CREATE can be successfully implemented. The approach can be adopted in part, to ease into it and test it out, or in whole. We recommend that ecology teachers consider making their courses more active, if they have not already done so; adopting pedagogical practices embedded within CREATE can be a way to achieve active learning. The CREATE approach and other evidence‐based pedagogical best practices lead to strong learning gains and more inclusive learning environments. Background: Metabolic disease risk in youth is influenced by sedentary behaviors. Acute in‐lab studies show that, during a single day, interrupting a sedentary period with short bouts of physical activity improves glucometabolic outcomes. Objective: To determine if acutely improved glucose metabolism persists after multi‐day interruptions of sitting with walking brief bouts. We hypothesized that children who underwent interrupting sitting on multiple days would demonstrate lower insulin area under the curve during an oral glucose tolerance test compared to uninterrupted sitting. Methods: Healthy, normoglycemic children (N = 109) ages 7–11 years were randomized to one of two conditions: Control (3 h of daily Uninterrupted Sitting) or Interrupted Sitting (3‐min of moderate‐intensity walking every 30 min for 3 h daily); with dietary intake controlled through provision of foodstuffs for the entire experiment. Participants attended six consecutive daily visits at a research ambulatory unit. The primary outcome was insulin area under the curve during the oral glucose tolerance test on day 6 during interrupted or uninterrupted sitting; secondary outcomes included glucose and c‐peptide area under the curve, energy intake at a buffet meal on day 6, and free‐living activity. Results: Among 93 children (42 uninterrupted sitting, 51 interrupted sitting), daily interrupted sitting resulted in 21% lower insulin (β = 0.102 CI:0.032–0.172, p = 0.005) and a 10% lower C‐peptide (β = 0.043, CI:0.001–0.084, p = 0.045) area under the curve. Matsuda and Glucose Effectiveness Indices were also improved (p's < 0.05). There were no group differences in energy intake or expenditure. Trophic transfer of energy through marine food webs is strongly influenced by prey aggregation and its exploitation by predators. Rapid aggregation of some marine fish and crustacean forage species during wind‐driven coastal upwelling has recently been discovered, motivating the hypothesis that predators of these forage species track the upwelling circulation in which prey aggregation occurs. We examine this hypothesis in the central California Current Ecosystem using integrative observations of upwelling dynamics, forage species' aggregation, and blue whale movement. Directional origins of blue whale calls repeatedly tracked upwelling plume circulation when wind‐driven upwelling intensified and aggregation of forage species was heightened. Our findings illustrate a resource tracking strategy by which blue whales may maximize energy gain amid ephemeral foraging opportunities. These findings have implications for the ecology and conservation of diverse predators that are sustained by forage populations whose behaviour is responsive to episodic environmental dynamics. SUMMARY: The distribution of recombination events along large cereal chromosomes is uneven and is generally restricted to gene‐rich telomeric ends. To understand how the lack of recombination affects diversity in the large pericentromeric regions, we analysed deep exome capture data from a final panel of 815 Hordeum vulgare (barley) cultivars, landraces and wild barleys, sampled from across their eco‐geographical ranges. We defined and compared variant data across the pericentromeric and non‐pericentromeric regions, observing a clear partitioning of diversity both within and between chromosomes and germplasm groups. Dramatically reduced diversity was found in the pericentromeres of both cultivars and landraces when compared with wild barley. We observed a mixture of completely and partially differentiated single‐nucleotide polymorphisms (SNPs) between domesticated and wild gene pools, suggesting that domesticated gene pools were derived from multiple wild ancestors. Patterns of genome‐wide linkage disequilibrium, haplotype block size and number, and variant frequency within blocks showed clear contrasts among individual chromosomes and between cultivars and wild barleys. Although most cultivar chromosomes shared a single major pericentromeric haplotype, chromosome 7H clearly differentiated the two‐row and six‐row types associated with different geographical origins. Within the pericentromeric regions we identified 22 387 non‐synonymous SNPs, 92 of which were fixed for alternative alleles in cultivar versus wild accessions. Surprisingly, only 29 SNPs found exclusively in the cultivars were predicted to be 'highly deleterious'. Overall, our data reveal an unconventional pericentromeric genetic landscape among distinct barley gene pools, with different evolutionary processes driving domestication and diversification. Significance Statement: We have explored genetic variation in regions of the genome that have been genetically inaccessible, comparing wild progenitors with domesticated barleys. This paper reflects on ideas of tacit knowledge in order to examine the nature of planners' expertise. It investigates the shifting knowledge and power dynamics between and within the public and private sectors, as a means of determining how tacit expertise is transferred and re‐appropriated in new domains and geographies of practice. Tacit understanding of what facilitates successful permissions and what impedes the approval process helps planners navigate the plan‐led system and avoid inertia. Findings are two‐fold. First, public sector planners transfer their own expert tacit knowledge to influence and direct local development planning. Second, the findings illustrate that public sector planners feel their tacit expertise is increasingly undervalued and traditional networks of knowledge transference have been dismantled due to the erosion of networks of peer support. This results in a disruptive counter narrative based in private planning practice where public sector experts are re‐emerging in commercial practice due to a range of factors, including: budgets cuts; demoralisation; and seeking greater job security in the private sector. This leads to new geographies of tacit knowledge, as local government planners transfer their knowledge into new professional environments. This can result in reconfigurations of locally embedded knowledge as expertise is repurposed and used to bolster the likelihood of development applications succeeding, often for profit motives rather than the broader public good. Furthermore, it can also lead to local knowledge being uprooted and spatially diffused across wider geographies as private sector planners frequently work across broader domains of practice than public sector counterparts. Enabling these predictions may rely on extending the genomewide prediction framework to include environmental covariates (EC), but such models have generally been tested under limited, less breeding‐realistic contexts. Using a barley (Hordeum vulgare L.) multi‐environment dataset, our objectives were to compare multi‐environment prediction models and scenarios that target genotypes from different breeding generations, use different levels and timescales of ECs, and are applied to different agronomic and quality traits. When predicting the phenotypes of previously tested genotypes in untested environments, models that included the interaction of genomewide markers and pre‐selected in‐season ECs resulted in more accurate predictions (rMG or rMP) within (rMG = 0.56–0.94) and across (rMP = 0.63–0.87) environments; similar accuracy was achieved within (rMP = 0.46–0.89) and across (rMP = 0.87–0.95) locations when using only ECs from realistically available historical climate data. Shifting the prediction target to a distinct, untested offspring population slightly reduced model performance within environments or locations, but rMP across environments (rMP = 0.60–0.86) or locations (rMP = 0.87–0.94) remained very high. Though we achieved moderately high rMP for most traits in the challenging scenario of predicting the offspring population in holdout environments, the similarity between training and target environments, like that between populations, will be a limiting factor for enabling accurate predictions of new genotypes under new growing conditions. Silflower (Silphium integrifolium Michx.) has been identified as a candidate for domestication as a perennial oilseed crop. In order to successfully domesticate silflower it will be necessary to identify traits that should be targeted for selection. We have conducted the first multilocation trial of replicated silflower genotypes at six sites across four states. By observing architectural, phenological, and yield traits in a diversity panel of silflower breeding material, we were able to estimate the heritability of these traits, as well as patterns of correlation across years and environments. We found that this population expressed heritable variation for many, but not all, traits of interest, and that targeted introgression of diversity may be beneficial for some traits. In addition, we used regression analysis to identify several traits, particularly height, number of stalks per plant, and stalk diameter as important contributors to determining levels of yield and levels of lodging. These results will help to guide future silflower domestication efforts and have already shaped the direction of our breeding program. The glutamine synthetase (GS) expression system is commonly used to ensure stable transgene integration and amplification in Chinese hamster ovary (CHO) host lines. Transfected cell populations are typically grown in the presence of the GS inhibitor, methionine sulfoximine (MSX), to further select for increased transgene copy number. However, high levels of GS activity produce excess glutamine. We hypothesized that attenuating the GS promoter while keeping the strong IgG promoter on the GS‐IgG expression vector would result in a more efficient cellular metabolic phenotype. Herein, we characterized CHO cell lines expressing GS from either an attenuated promoter or an SV40 promoter and selected with/without MSX. CHO cells with the attenuated GS promoter had higher IgG specific productivity and lower glutamine production compared to cells with SV40‐driven GS expression. Selection with MSX increased both specific productivity and glutamine production, regardless of GS promoter strength. 13C metabolic flux analysis (MFA) was performed to further assess metabolic differences between these cell lines. Interestingly, central carbon metabolism was unaltered by the attenuated GS promoter while the fate of glutamate and glutamine varied depending on promoter strength and selection conditions. This study highlights the ability to optimize the GS expression system to improve IgG production and reduce wasteful glutamine overflow, without significantly altering central metabolism. Additionally, a detailed supplementary analysis of two "lactate runaway" reactors provides insight into the poorly understood phenomenon of excess lactate production by some CHO cell cultures. The synthesis and reactivity of 3,8‐dibromo‐dodecafluoro‐benzo‐fused BOPHY 2 are reported, via SNAr with O‐, N‐ S‐ and C‐nucleophiles, and in Pd(0)‐catalyzed cross‐coupling reactions (Suzuki and Stille). The resulting perfluoro‐BOPHY derivatives were investigated for their reactivity in the presence of various nucleophiles. BOPHY 3 displays reversible color change and fluorescence quenching in the presence of bases (Et3N, DBU), whereas BOPHY 7 reacts preferentially at the α‐pyrrolic positions, and BOPHY 8 undergoes regioselective fluorine substitution in the presence of thiols. The structural and electronic features of the fluorinated BOPHYs were studied by TD‐DFT computations. In addition, their spectroscopic and cellular properties were investigated; BOPHY 10 shows the most red‐shifted absorption/emission (λmax 659/699 nm) and 7 the highest fluorescence (Φf=0.95), while all compounds studied showed low cytotoxicity toward human HEp2 cells and were efficiently internalized. Cocoa butter equivalents (CBEs) are formulated by mixing palm mid fraction (PMF) with silica‐treated hard stearin fractions produced via enzymatic acidolysis of high oleic sunflower oil with various mixtures of stearic and palmitic acids. The CBE blends always contain ≈30% 1,3‐distearoyl‐2‐oleoyl‐sn‐glycerol, but vary in 1,3‐dipalmitoyl‐2‐oleoyl‐sn‐glycerol (POP; 40–45%) and rac‐1‐palmitoyl‐2‐oleoyl‐3‐stearoyl glycerol (POSt; 10‐16%) content. Overall symmetric disaturated monounsaturated (Sat‐O‐Sat) levels range from 81–86%. The effects of varying POP/POSt content on the non‐isothermal and isothermal crystallization (at 20 °C) of the CBEs are then studied using differential scanning calorimetry (DSC), stop and return DSC, and X‐ray diffraction (XRD), and compared with CB. Four polymorphs (I/sub‐α, II/α, IV/β′, and V/β) are identified which appear at different stages. The addition of PMF (raising POP levels) slows crystallization rates compared to the original stearins, but higher levels of POP also produce a small amount of V/β in under 4 h at 20 °C via apparent transformation from II/α. This does not occur with (high POSt) CB samples. However, after storage at 20 °C for 7 days, CB demonstrates greater transformation to V/β (from IV/β′) as confirmed via XRD. Solid fat contents of CBE blends are higher than CB due to higher Sat‐O‐Sat levels. Practical applications: The paper demonstrates a new route for formulating CBEs. The relative proportions of StOSt, POSt and POP in the formulation can be controlled by changing the proportions of palmitic and stearic acid used in the enzymatic acidolysis and the blend ratio with the PMF. Winter survival is a major yield-limiting factor in winter barley grown in the Upper Midwest, where winter temperatures regularly reach-20°C or lower. Here, we tested the hypothesis that improved freezing survival is associated with smaller xylem vessel diameters as a mechanism that minimizes physical damage arising from intracellular ice formation, using leaf vasculature as a proxy trait. A second goal was to test whether such anatomical differences could be captured non-destructively via gas exchange measurements. We first identified a group of 11 winter barley genotypes that exhibited differential field winter survival. We then conducted xylem diameter measurements on the first three leaves on all genotypes in two independent experiments based on 1,188 images, in addition to leaf gas exchange measurements. Freezing-tolerant genotypes consistently exhibited significantly smaller metaxylem vessel diameters irrespective of leaf rank, and this difference was not influenced by hardening, indicating that this trait is heritable. Additionally, genotypes with smaller vasculature tended to exhibit lower stomatal conductance and transpiration rates. Our data indicate that genotypes with leaf xylem diameters smaller than 30 µm are prime donor parents and could be identified using gas exchange measurements, pointing to new phenotyping approaches to accelerate breeding for freezing survival. Background: Individuals who participate in outdoor sports are subject to an increased risk of developing skin cancer. To date, there has been no review examining skin cancer and sun protective behaviours specific to outdoor, water‐based sports. Therefore, this scoping reviews objectives were to (a) summarize volume and type of the scientific literature available relating to skin cancer, (b) examine current sun protective behaviours and (c) identify knowledge gaps to inform future research. Method: A scoping review was conducted to address the objectives following the PRISMA guidelines. Seven databases were searched; identified studies were screened based on title, abstract and full text for outdoor water‐based sports, which examined skin cancer and/or sun protective behaviours. Data were extracted, synthesized and critically appraised using a modified AXIS tool. Percentage frequencies were calculated, and key results were placed in tabular or graphical presentation. Results: Nine cross‐sectional studies were identified, all defined as low‐level evidence. A combined sample of 4377 participants across six countries and five water‐based sports showed BCC (71%) was the most common skin cancer, followed by melanoma (18%) and SCC (10%). The head (41%) and upper limbs (27%) were the most common location. Conclusion: This review highlights the minimal research available and demonstrates those in water‐based sports are at high risk of developing skin cancer. Due to difficulty adhering to sun protective behaviours, the most prevalent location is the upper body. Aims: Preclinical studies of MR309, a selective sigma‐1 receptor (σ1R) antagonist, support a potential role in treating neuropathic pain. We report 2 studies that provide insight into the pharmacokinetics (PK) and brain σ1R binding of MR309. Methods: Steady‐state PK of MR309 (400 mg once daily and 200 mg twice‐daily [BID] for 10 days; EudraCT 2015–001818‐99 [PK study]) and the relationship between MR309 plasma exposure and brain σ1R occupancy (EudraCT 2017–000670‐11 [positron emission tomography study]) were investigated in healthy volunteers. Positron emission tomography using the σ1R ligand [11C]SA4503 was conducted at baseline, and 2 and 8 hours after a single dose of MR309 (200–800 mg). The relationship between brain σ1R occupancy and MR309 exposure was explored using data‐driven model fitting. Results: MR309 was well tolerated, brain σ1R occupancy ranged between 30.5 and 74.9% following single‐dose MR309 (n = 7). MR309 BID provided a plasma PK profile with less fluctuation than once daily dosing (n = 16). MR309 200 mg BID yielded average steady state plasma concentrations between 2000 and 4000 ng/mL in the PK study, which corresponded to an estimated brain σ1R occupancy of 59–74%. Conclusion: MR309 200 mg BID dose was below the 75% σ1R occupancy threshold expected to elicit maximal antinociceptive effect as observed in neuropathic pain models. Further investigations of MR309 for neuropathic pain will require higher brain σ1R occupancy, and establish the optimal dose by elucidating the clinical impact of a broad range of brain σ1R occupancy across different neuropathic pain indications. Background: Women living with HIV who misuse alcohol and live in economically disadvantaged settings in South Africa experience a multitude of contextual barriers as they navigate the HIV care continuum. The Women's Health CoOp (WHC), a brief, woman‐focused, behavioural, evidence‐based intervention, has been shown to be effective in reducing heavy drinking and improving HIV‐related outcomes among this key population. However, these women face other broader socioecological barriers to antiretroviral therapy (ART) adherence. Methods: The WHC was implemented in a modified, stepped‐wedge implementation science trial in public health clinics and substance use treatment programmes in Cape Town, South Africa. A qualitative substudy was conducted to explore barriers to HIV treatment adherence among women enrolled in this trial. Eight focus group discussions were conducted with 69 participants 6 months after completion of the WHC workshops. Focus groups were audio‐recorded (with consent), transcribed verbatim and analysed using a thematic approach. Results: The mean age of the participants was 33 years and the mean self‐reported number of drinks per day was 13. The main contextual factors influencing participants' ART adherence were intrapersonal‐level factors (substance use, financial constraints, food insecurity; community‐level factors (anticipated and enacted stigma, community violence) and institutional‐level factors (patient–provider relationships, health facility barriers, environmental stigma). Conclusion: Comprehensive interventions addressing the contextual barriers and unique challenges faced by women who misuse alcohol in low‐resource settings that intersect with HIV treatment nonadherence should be implemented in tandem with successful biobehavioural HIV interventions for long‐term effectiveness and sustainability. Naked barley (Hordeum vulgare L.) grain threshes freely from its hull during harvesting and cleaning. Much of the available naked barley germplasm is unadapted to U.S. barley growing regions, and few genotypes have been selected to thrive under organic systems. The goal of this research was to characterize a set of spring naked barley genotypes for agronomic traits in the northern United States under organic conditions and estimate the degree of genotype × environment interaction (GEI). To achieve this goal, a multienvironment trial was conducted over 3 yr. Experiments were grown under organic conditions where traits including grain yield, test weight, plant height, heading date, and threshability were evaluated. Contributions of environment, genotype, and GEI to the phenotypic variance were calculated. In the tested germplasm, test weight, plant height, heading date, and threshability were found to have higher variance attributed to genotypic effects compared to GEI. Grain yield variance attributed to GEI was five times greater than that of genotype alone. Genotype + GEI (GGE) and location‐grouping (LG) biplots showed two sets of environments where naked barley genotypes performed similarly. Sensitivity analysis by Finlay–Wilkinson regression found that grain yield was highly sensitive to changes in environment mean yield. These results show that selection for grain yield should be conducted within mega‐environments to leverage GEI patterns. Heading date, plant height, test weight, and threshability can be selected across mega‐environments. Plant breeding programs expend significant resources on multilocation testing to evaluate genotypes for advancement or potential cultivar release. The selection of genotype entries for these trials is typically based on previous phenotypic data or predictions; yet locations, important contributors to nongenetic variation, are often chosen in a less data‐driven manner. Using agronomic and quality trait data from two long‐term regional barley (Hordeum vulgare L.) nurseries, our objectives were (a) to measure the precision, repeatability, and representativeness of test locations based on multitrait data and (b) to optimize the selection of test locations for use in future trials. When considering traits individually, ideal locations could be identified simply, but a combined analysis of 11 traits indicated that very few locations were broadly favorable, and considerable tradeoffs are necessary. We developed a flexible optimization procedure to select the locations based on their precision, repeatability, and representativeness for multiple traits while simultaneously constraining the total number of locations. Optimization led to a 58–75% reduction in the number of locations, and therefore phenotyping costs, with little loss in data utility. Importantly, our approach allowed locations to be selected for phenotyping different sets of traits (e.g., either agronomic or both agronomic and malting quality), mimicking the often nested structure of trait data collection. This approach may be useful for individual plant breeding programs or collaborations wishing to increase the resource efficiency of these important regional evaluation trials. Kernel shape and ß-glucan content are important attributes for oat (Avena sativa L.) millers. The identification of genomic regions controlling these traits, as well as their effect on phenotype, will enable rapid gain fromselection. The objectives of this study were (a) to identify the genomic regions associated with kernel shape in an oat panel adapted to subtropical environments and (b) to clarify the relationship between kernel shape and ß-glucan content in oat. An elite oat panel comprising 406 inbred lines was assessed in Londrina and Eldorado do Sul, Southern Brazil. Genome-wide association analyses were performed for each environment separately and also in a multi-environment analysis. Phenotypic and genetic correlation coefficients were estimated between kernel shape traits and ß-glucan content. Genomic regions influencing kernel length were identified onMrg06, Mrg21, and Mrg24. Kernel width and kernel thickness were genetically correlated and shared a quantitative trait locus, located on Mrg13 of the oat consensus map. Phenotypic and genetic correlations were found between kernel shape traits and ß-glucan content, indicating a negative relationship between kernel width or kernel thickness and ß-glucan content. Oat breeding programs that aim to maintain minimum levels of ß-glucan for nutrition and minimum kernel widths for milling will need to screen for both traits; otherwise, indirect selection will lead to unfavorable results. Perennial ryegrass (Lolium perenne L.) is considered the most important pasture species in temperate agriculture, with over six million hectares of sown area in Australia alone. However, perennial ryegrass has poor persistence in some environments because of low tolerance to a range of both abiotic and biotic stresses. To breed perennial ryegrass, cultivars with greater persistence and productivity may require evaluation of genotypes over a number of years. Persistence assessment in pasture breeding depends on manual ground cover estimation or counting the number of surviving plants or tillers in a known area. These methods are subjective and labour intensive, which may limit data collection in large‐scale breeding programs. With the rapid development of sensors and image processing algorithms, image‐based high‐throughput phenotyping (HTP) is becoming commonplace in the breeding of major food crops. Image‐based HTP approaches consist of the deployment of a wide range of sensors on ground‐based or airborne platforms and data analysed through image processing pipelines. Image‐based HTP shows high potential for use in pasture phenotyping in breeding programs and may be able to reduce timeframes for releasing new cultivars. Moreover, existing image‐based HTP approaches could be further developed to include precise tools for phenotyping pasture persistence traits such as pasture senescence, botanical composition, pathogen and pest resistance. In this study, we reviewed existing image‐based HTP approaches in precision agriculture and discussed their feasibility for perennial ryegrass persistence estimation in pasture breeding. The frequency and intensity of high‐temperature stress events are expected to increase as climate change intensifies. Concomitantly, an increase in evaporative demand, driven in part by global warming, is also taking place worldwide. Despite this, studies examining high‐temperature stress impacts on plant productivity seldom consider this interaction to identify traits enhancing yield resilience towards climate change. Further, new evidence documents substantial increases in plant transpiration rate in response to high‐temperature stress even under arid environments, which raise a trade‐off between the need for latent cooling dictated by excessive temperatures and the need for water conservation dictated by increasing evaporative demand. However, the mechanisms behind those responses, and the potential to design the next generation of crops successfully navigating this trade‐off, remain poorly investigated. Here, we review potential mechanisms underlying reported increases in transpiration rate under high‐temperature stress, within the broader context of their impact on water conservation needed for crop drought tolerance. We outline three main contributors to this phenomenon, namely stomatal, cuticular and water viscosity‐based mechanisms, and we outline research directions aiming at designing new varieties optimized for specific temperature and evaporative demand regimes to enhance crop productivity under a warmer and dryer climate. High temperature stress could lead to increased plant transpiration to enable evaporative cooling via processes that are poorly understood. Summary: Circadian clock rhythms are shown to be intertwined with crop adaptation. To realize the adaptive value of changes in these rhythms under crop domestication and improvement, there is a need to compare the genetics of clock and yield traits.We compared circadian clock rhythmicity based on Chl leaf fluorescence and transcriptomics among wild ancestors, landraces, and breeding lines of barley under optimal and high temperatures. We conducted a genome scan to identify pleiotropic loci regulating the clock and field phenotypes. We also compared the allelic diversity in wild and cultivated barley to test for selective sweeps.We found significant loss of thermal plasticity in circadian rhythms under domestication. However, transcriptome analysis indicated that this loss was only for output genes and that temperature compensation in the core clock machinery was maintained. Drivers of the circadian clock (DOC) loci were identified via genome‐wide association study. Notably, these loci also modified growth and reproductive outputs in the field. Diversity analysis indicated selective sweep in these pleiotropic DOC loci.These results indicate a selection against thermal clock plasticity under barley domestication and improvement and highlight the importance of identifying genes underlying for understanding the biochemical basis of crop adaptation to changing environments. PREMISE: Inbreeding depression, or the reduction in fitness of progeny with related parents, has the potential to adversely affect the long‐term viability of both wild and captive plant populations. Silphium integrifolium, a prairie plant native to the central United States, has been identified as a potential candidate for domestication as a perennial oilseed crop. Little is known about the potential for inbreeding depression in this species, but it is expected to be nonnegligible because S. integrifolium is both perennial and self‐incompatible. Here, we measure lethal inbreeding depression expressed through embryo deaths, and nonlethal inbreeding depression expressed through changes in vigor and fitness phenotypes of progeny. METHODS: First, we made controlled crosses among related and unrelated individuals to determine the effect of two different levels of inbreeding on seed production. Then, we grew inbred and outbred progeny from this population to reproductive maturity and measured 11 key traits. RESULTS: We found that within an improved S. integrifolium population, individuals carried an average of slightly less than one lethal allele per gamete. In progeny, significant inbreeding depression was observed in at least one family for eight of the 11 measured traits. CONCLUSIONS: Inbreeding depression is likely to be an important challenge to S. integrifolium domestication, reducing overall population fecundity and values for important phenotypes. These effects may grow worse as selection reduces effective population size. We recommend several strategies for S. integrifolium breeding to help mitigate these problems. [ABSTRACT FROM AUTHOR]

Land‐use change is a significant cause of anthropogenic extinctions, which are likely to continue and accelerate as habitat conversion proceeds in most biomes. One way to understand the effects of habitat loss on biodiversity is through improved tools for predicting the number and identity of species losses in response to habitat loss. There are relatively few methods for predicting extinctions and even fewer opportunities for rigorously assessing the quality of these predictions. In this paper, we address these issues by applying a new method based on rarefaction to predict species losses after random, but aggregated, habitat loss. We compare predictions from three rarefaction models, individual‐based, sample‐based, and spatially clustered, to those derived from a commonly used extinction estimation method, the species–area relationship (SAR). We apply each method to a mesocosm experiment, in which we aim to predict species richness and extinctions of arthropods immediately following 50% habitat loss. While each model produced strikingly accurate predictions of species richness immediately after the habitat loss disturbance, each model significantly underestimated the number of extinctions occurring at both the local (within‐mesocosm) and regional (treatment‐wide) scales. Despite the stochastic nature of our small‐scale, short‐term, and randomly applied habitat loss experiment, we found surprisingly clear evidence for extinction selectivity, for example, when abundant species with low extinction probabilities were extirpated following habitat loss. The important role played by selective extinction even in this contrived experimental system suggests that ecologically driven, trait‐based extinctions play an equally important role to stochastic extinction, even when the disturbance itself has no clear selectivity. As a result, neutrally stochastic null models such as the SAR and rarefaction are likely to underestimate extinctions caused by habitat loss. Nevertheless, given the difficulty of predicting extinctions, null models provide useful benchmarks for conservation planning by providing minimum estimates and probabilities of species extinctions. In plant breeding, the goal of genomewide selection is to predict the merit of unobserved individuals, particularly those in the next breeding generation. Predictions of these individuals in unobserved or future environments would be of additional use to a breeder. For many of the complex traits targeted in breeding, this may require management of genotype × environment interactions by, for example, using data from homogeneous groups of environments. Our objectives were to assess the accuracy of genomewide predictions in unobserved environments both within and between breeding generations; we aimed to compare training sets that included data from all possible environments with those that included data from (a) decreasingly similar environments or (b) discrete clusters of similar environments. A 183‐line spring barley (Hordeum vulgare L.) training population and 50‐line offspring test population were phenotyped in 29 location–year environments for grain yield, heading date, and plant height. Environmental similarities were measured using phenotypic data, geographic distance, or environmental covariables. When using training data from more, but decreasingly similar environments, prediction accuracy increased, but marginal gains declined; in some cases, accuracy declined with additional data. Clusters of environments informed by phenotypes (i.e., phenotypic correlations or multiplicative models) typically improved prediction accuracy within a generation, but not between generations (offspring population). Our study suggests that, as an alternative to using data from all available environments, informative subsets may be advantageous for genomewide predictions within a single breeding generation, but not between generations. The year 2020 marks the 75th edition of Grass and Forage Science. From its beginnings as the Journal of the British Grassland Society, the journal has grown to be the major international journal with a primary emphasis on grasslands society. In this paper, to mark this anniversary we note the contributions of the editorial team, past and present, and the legacy of the journal by noting some of the highly cited papers published over the years and discuss the changing trends in science published in recent years. This includes an emphasis on the changing role of grasslands across the globe and the challenges that changing climate and altered production systems place on grazing agriculture. It is through the adoption of science that is being published now that the industries that we serve will be able to respond to these challenges. [ABSTRACT FROM AUTHOR]

We consider a cheap‐talk setting with two senders and a continuum of receivers with heterogenous preferences. Each receiver is constrained to listen to one of the senders but can choose which sender to listen to. The introduction of a second sender facilitates more informative communication and even enables full communication for a large set of sender preference pairs. We use the model to assess the size and characteristics of sender audiences, the amount of information communicated, and the impact of the senders' biases on the receivers' actions. Antimicrobial resistance (AMR) in pathogenic strains of bacteria, such as Escherichia coli (E. coli), adversely impacts personal and public health. In this study, we examine competing hypotheses for the evolution of AMR including (i) 'genetic capitalism' in which genotypes that confer antibiotic resistance are gained and not often lost in lineages, and (ii) 'stabilizing selection' in which genotypes that confer antibiotic resistance are gained and lost often. To test these hypotheses, we assembled a dataset that includes annotations for 409 AMR genotypes and a phylogenetic tree based on genome‐wide single nucleotide polymorphisms from 29 255 isolates of E. coli collected over the past 134 years. We used phylogenetic methods to count the times each AMR genotype was gained and lost across the tree and used model‐based clustering of the genotypes with respect to their gain and loss rates. We demonstrate that many genotypes cluster to support the hypothesis for genetic capitalism while a few genotypes cluster to support the hypothesis for stabilizing selection. Comparing the sets of genotypes that fall under each of the hypotheses, we found a statistically significant difference in the breakdown of resistance mechanisms through which the AMR genotypes function. The result that many AMR genotypes cluster under genetic capitalism reflects that strong positive selective forces, primarily induced by human industrialization of antibiotics, outweigh the potential fitness costs to the bacterial lineages for carrying the AMR genotypes. We expect genetic capitalism to further drive bacterial lineages to resist antibiotics. We find that antibiotics that function via replacement and efflux tend to behave under stabilizing selection and thus may be valuable in an antibiotic cycling strategy. [ABSTRACT FROM AUTHOR]

In the current study, 248 two‐rowed and another 253 six‐rowed spring barley (Hordeum vulgare L.) breeding lines from several US barley breeding programs were evaluated in multiple locations in Egypt to select for potentially adapted lines. The introduced plant materials were planted in two locations during three growing seasons (2015, 2016, and 2017). The focus of the first growing season was to increase available seeds and collect preliminary observations. During the second and third growing seasons, the advanced plant materials and local check cultivars were planted in an incomplete block design in two replicates within three locations. Grain yield, leaf rust, number of days to flowering, and plant height were measured. Based on grain yield production, the top 94 lines from the two‐rowed and 100 from the six‐rowed barley lines were selected and advanced for further evaluation in 2017. The introduced materials contained several lines that consistently outperformed the check cultivars. Therefore, these lines could be used as parents to improve barley production and enhance genetic diversity or be directly released as cultivars after appropriate testing in Egypt.
