Objective: This study aimed to evaluate the role of pepsin inhibitors in the inflammatory response and their effects on laryngeal mucosal integrity during gastroesophageal reflux (GERD) under in vivo conditions. Methods: A surgical model of GERD was used, in which mice were treated with pepstatin (0.3 mg/kg) or darunavir (8.6 mg/kg) for 3 days. On the third day after the experimental protocol, the laryngeal samples were collected to assess the severity of inflammation (wet weight and myeloperoxidase activity) and mucosal integrity (transepithelial electrical resistance and paracellular epithelial permeability to fluorescein). Results: The surgical GERD model was reproduced. It showed features of inflammation and loss of barrier function in the laryngeal mucosa. Pepstatin and darunavir administration suppressed laryngeal inflammation and preserved laryngeal mucosal integrity. Conclusion: Pepsin inhibition by the administration of pepstatin and darunavir improved inflammation and protected the laryngeal mucosa in a mouse experimental model of GERD. Taking advantage of a large ensemble of Convection Permitting‐Regional Climate Models on a pan‐Alpine domain and of an object‐oriented dedicated analysis, this study aims to investigate future changes in high‐impact fall Mediterranean Heavy Precipitation Events at high warming levels. We identify a robust multi‐model agreement for an increased frequency from central Italy to the northern Balkans combined with a substantial extension of the affected areas, for a dominant influence of the driving Global Climate Models for projecting changes in the frequency, and for an increase in intensity, area, volume and severity over the French Mediterranean. However, large quantitative uncertainties persist despite the use of convection‐permitting models, with no clear agreement in frequency changes over southeastern France and a large range of plausible changes in events' properties, including for the most intense events. Model diversity and international coordination are still needed to provide policy‐relevant climate information regarding precipitation extremes. Plain Language Summary: Despite growing computational resources and multiple model developments, projecting future changes in the high‐impact Mediterranean Heavy Precipitation Events remains both a numerical and scientific challenge. The present study takes advantage of the recent availability of a relatively large ensemble of high resolution Regional Climate Models (2–3 km), which represent a step change in the simulation of precipitation extremes, and of an object‐oriented approach, allowing us to track the convective precipitating systems on an hourly basis. Looking at future changes in fall Mediterranean Heavy Precipitation Events at high warming levels, we identify a robust multi‐model agreement for an increased frequency from central Italy to the northern Balkans combined with a substantial expansion of the affected areas, and an increase in intensity, area, volume and severity over the French Mediterranean. However, considerable uncertainties remain in terms of frequency over parts of the domain arising from uncertainty in changes in large scale weather patterns, and in terms of degree of intensification for the most intense events. Wave phenomena impact high commercial value coastal and offshore activities, infrastructures and transportation. The knowledge of future wave conditions allows for consistent long‐term planning and decision‐making. The present study aims to provide robust, reliable projections of the potential future wave conditions of the Black Sea under the influence of climate change. For this purpose, an eight‐member dynamic wave climate ensemble that accurately represents the Black Sea's present wave climate has been produced, and future projections are assessed and analysed here. The wave climate ensemble was obtained by forcing the Simulating Waves Nearshore spectral wave model with eight regional wind fields from the EURO‐CORDEX. The future simulations of the regional wind fields are based on the RCP8.5 high‐emission scenario. The historical wave climate of the ensemble was evaluated against ERA5 reanalysis data. Projected changes in the mean wind and wave characteristics are examined by comparing historical and future simulations. In addition, the projected trends in the annual means during the future period of the wave simulations for significant wave height (Hs) and wave energy flux are also analysed. The projections for Hs and wind speeds are pronounced with significant decreases down to −10% across the basin, especially in the eastern region of the Black Sea. Changes in the Hs 99% percentiles of up to 16% are projected to occur. The projected changes in the annual mean of the wave energy flux are close to the projected changes in Hs means, while the seasonal changes (between −15% and 12%) are expected to be higher. New geomagnetically induced current (GIC) computations for mainland Portugal include the entire power network, with network parameters and topology provided by the transmission grid operator for all the high voltage lines (150, 220, and 400 kV). The first 3D conductivity model for the west region of the Iberian Peninsula, based on 31 broadband magnetotelluric soundings, is used in calculations, revealing the effect of different crustal domains in GIC distribution. Geomagnetic field variations are taken from Coimbra or San Fernando magnetic observatories, according to the Nearest Neighbor method, and used together with surface impedance values predicted from the new conductivity model to calculate the induced electric field on a regular grid. The global distribution of GICs over the power network is characterized based on results derived for the eight most significant storms registered in the Iberia during solar cycle 24. Substations susceptible to the highest GICs are found near the transition between the granitic geotectonic unit of Central Iberian Zone and the Lusitanian Basin. A prototype of a Hall effect sensor has been installed at a substation and is active since the end of August 2021. In order to validate our GIC model, recent measurements are compared with simulations. GIC computation is prone to uncertainties from various sources, possibly contributing with different weights to the final error in computed values. Here, we evaluate the contribution of substation earthing resistance and nonuniqueness of the conductivity model to the final GIC uncertainties. Plain Language Summary: Magnetic storms on the Sun can trigger geomagnetic disturbances (GMD) on Earth, affecting technological infrastructures and gadgets we depend upon. As an example, they can interfere with the normal operation of power networks transporting electricity by inducing spurious currents along the conducting lines, which are called geomagnetically induced currents (GICs). We computed GICs at all substations of the transmission power network in Portugal mainland, gathering information on the electric circuit parameters, the most intense GMD events registered at the magnetic observatories of Coimbra (Portugal) and San Fernando (Spain), and measuring the Earth response to electromagnetic waves of different frequencies to obtain a conductivity model. Our results show the influence of local geotectonic structures on the geographic distribution of GICs during a given GMD. GICs tend to be stronger close to sharp discontinuities of the ground resistivity, which occur at the transition between different geotectonic units. Among different possible sources of errors in GICs, we compute error estimates due to uncertainties in the ground conductivity model and in the grounding resistances at certain substations. Finally, we give account of a first GIC sensor installed in the power network, and use its data to test different conductivity models. Microbe domestication has a major applied relevance but is still poorly understood from an evolutionary perspective. The yeast Torulaspora delbrueckii is gaining importance for biotechnology but little is known about its population structure, variation in gene content or possible domestication routes. Here, we show that T. delbrueckii is composed of five major clades. Among the three European clades, a lineage associated with the wild arboreal niche is sister to the two other lineages that are linked to anthropic environments, one to wine fermentations and the other to diverse sources including dairy products and bread dough (Mix‐Anthropic clade). Using 64 genomes we assembled the pangenome and the variable genome of T. delbrueckii. A comparison with Saccharomyces cerevisiae indicated that the weight of the variable genome in the pangenome of T. delbrueckii is considerably smaller. An association of gene content and ecology supported the hypothesis that the Mix‐Anthropic clade has the most specialized genome and indicated that some of the exclusive genes were implicated in galactose and maltose utilization. More detailed analyses traced the acquisition of a cluster of GAL genes in strains associated with dairy products and the expansion and functional diversification of MAL genes in strains isolated from bread dough. In contrast to S. cerevisiae, domestication in T. delbrueckii is not primarily driven by alcoholic fermentation but rather by adaptation to dairy and bread‐production niches. The Iberian Peninsula is a known climate change hotspot. In the last decades, the Coupled Model Intercomparison Project (CMIP) has allowed for thousands of Global Climate Model (GCM) simulations to be conducted, an important tool to assess and understand future changes in Earth's climate. The comparison of past future projections with observations provides a robust evaluation of the models' skill according to different emission scenarios. Here, a comprehensive performance assessment of GCM‐simulated mean, maximum, and minimum temperatures, and accumulated precipitation is conducted for Iberia, by retrospectively comparing historical simulations and past future projections from CMIPs 1 to 6 with reference datasets. From means to extremes, and multi‐year intra‐annual cycles to inter‐annual trends, GCM simulations are compared with Iberia0.1 and E‐OBS observational gridded datasets, and ERA5 reanalysis. The matching between the variables' distributions is assessed through the distribution added values (DAVs), a measure of gain or loss in performance between CMIPs. Results show relevant improvements in the description of the Iberian climate throughout the CMIP effort, for historical and past future periods. While the representation of intra‐annual cycles (inter‐annual trends) is enhanced after CMIP3 (CMIP2), GCMs from all CMIPs are generally able to depict the observed warming trend. Nevertheless, until 2021, a slight detrimental effect in the performance of CMIP6 models is found, in comparison with CMIP5 ones, with positive DAVs obtained only for past future temperature projections (less than 2%). In the context of the CORDEX project, an ensemble of regional climate simulations (RCMs) of high resolution on a 0.11° grid has been generated for Europe with the objective of improving the representation of regional to local‐scale atmospheric phenomena. However, such simulations are computationally expensive and do not always reveal added value. Here, a recently proposed metric (the distribution added value [DAV]) is used to determine the added value of all available EURO‐CORDEX high‐resolution simulations at 0.11° for daily mean wind speed compared to their respective coarser‐gridded 0.44° counterparts and their driving fields, hindcast and historical experiments. The analysis consists in comparing the degree of similarity between normalized wind probability density function (PDF) of simulations and observations. In addition, the use of a normalized PDF allows for a direct spatial comparison among the different regions and time periods. Results show that RCMs add value to their reanalysis or forcing global model, but the nature and magnitude of the improvement on the representation of wind speed may vary depending on the model, region and season. We found most RCMS at 0.11° to outperform models at the 0.44° resolution in terms of their quality in capturing the measured wind speed PDF. When looking at the upper tail of the wind speed PDF, the benefits of downscaling are generally larger. At the regional scale, added value is obtained for 0.11° with respect to 0.44° resolution for all subdomains studied, particularly over the Mediterranean, the Iberian Peninsula and the Alps. When analysing the added value dependence on altitude, runs at 0.11° models represent better the locations below 50 m and above 350 m of altitude, and the 0.44° increasingly under‐perform for higher altitudes. Fighter pilots (FP) are exposed to flight accelerations and stressful situations that defy cardiovascular control during and after flight. FP presents a smaller adjustment in sympatho‐vagal balance during tilt test after flight compared to baseline, suggesting a huge impact of flight on autonomic modulation to the heart. We undertake to test the hypothesis that FP will have a smaller vagal reentrance and lower sympathetic withdrawal during the recovery at the supine position after a prolonged active standing test that mimics flight hemodynamic demands. Twenty‐one military personnel (20–34 years old), composed of 9 FP and 12 non‐pilots (NP) matched by age, V̉O2max and body mass index were enroled in the experimental protocol. R‐R intervals were continuously recorded in the supine position for 15 min (SUPbaseline), during the prolonged active standing test (45 min) windowed in six 5 min time frames (from ORT1 to ORT6), and a recovery period in the supine position for 15 min (SUPrecovery). Heart rate variability was performed by spectral analysis to obtain the normalized low (LFn) and high (HFn) frequency components. The variation (Δ) from baseline (Δ = ORT6 − SUPbaseline) and from recovery (Δ = SUPrecovery–ORT6) periods were calculated. FP had a smaller ΔLFn (sympathetic mediated) and ΔHFn (vagal meditated) during recovery after active standing as compared to NP. Both groups showed similar changes in ΔLFn and ΔHFn during orthostatic stress compared to baseline, with no differences over time. Objective: This study aimed to evaluate the in vivo protective effect of the angico gum biopolymer in reducing the inflammatory response and preserving the integrity of the laryngeal and esophageal mucosa. Study Design: Animal study. Methods: A murine surgical model of gastroesophageal reflux disease was accomplished and subsequently treated with angico gum or omeprazole. On days 3 and 7 post surgery, samples of the larynx and esophagus, respectively, were collected to measure the level of inflammation (wet weight and myeloperoxidase activity) and mucosal integrity (transepithelial electrical resistance and mucosal permeability to fluorescein). Results: Angico gum and omeprazole decreased laryngeal inflammation (wet weight and myeloperoxidase activity) and dramatically improved the integrity of the laryngeal mucosa. It also reduced inflammation (decreased wet weight and myeloperoxidase activity) of the esophagus and preserved the barrier function (inferred by assessing the integrity of the mucosa). Conclusion: This study demonstrates the protective effect of angico gum in an experimental gastroesophageal reflux disease model. Angico gum attenuates inflammation and impairment of the mucosal barrier function not only in the larynx but also in the esophagus. This study evaluates the performance of a dynamic ensemble of wave climate simulations for the Black Sea. The ensemble members (and ensemble means) are evaluated for the historical period (1979–2005). The spectral wave model (simulating waves nearshore [SWAN]) was forced with eight Coupled Model Intercomparison Project Phase 5 (CMIP5) based Regional Climate Models (RCMs) wind fields to obtain each of the eight ensemble members. The RCM‐forcings used belongs to the European branch of the Coordinated Regional Downscaling Experiment (EURO‐CORDEX). The ensemble members' historical wave climate is thoroughly compared against the ERA5 reanalysis, in situ observations, and altimeter measurements. In addition to the individual evaluation of ensemble members, the results for the 8‐member ensemble mean are also examined. The performance of the ensemble members in reproducing the mean and extreme significant wave heights, mean wave periods, and mean wave directions is found to be good, with differences generally between +25 and −15% in comparison to ERA5. Differences between +10% and −30% were found for the comparison with altimeter measurements. We conclude that the ensemble better represents the wave climate of the Black Sea, than its individual members. The 8‐member ensemble provides a more balanced dataset for the Black Sea wave climate and a better representation of the associated uncertainty. Ultra‐high molecular weight polyethylene (UHMWPE) exhibits high wear resistance, low coefficient of friction (COF), and high impact strength. The operating conditions can influence the performance of the UHMWPE in specific applications. Graphene is applied as reinforcement to the UHMWPE matrix in order to improve the tribological and mechanical properties. In the present work, the effects of the test parameters on the hardness and tribological behavior of nanocomposites are evaluated. The Vickers microhardness of the UHMWPE/rGO nanocomposites increases by up to 25% at 0.50 wt% of rGO, regardless of the indentation applied load. Pin‐on‐disc tests are performed for the polymer at several sliding speeds and applied normal loads to get information about friction responses. Once the most suitable parameters have been defined, the pin‐on‐disc tests of the UHMWPE/rGO nanocomposites are performed, resulting in a decrease of up to 48% of the COF at 0.25 wt% of rGO. In this work, the effect of the addition of reduced graphene oxide (rGO) nanofillers to UHMWPE was investigated. The UHMWPE/rGO nanocomposites were prepared at rGO contents of 0.010, 0.10, 0.25, and 0.50 wt%. The morphological, mechanical, and surface properties of the nanocomposites and the neat UHMWPE were evaluated and related with their tribological behavior. For that, DSC, SEM, contact angle, MO, and profilometry techniques, besides shore D hardness tests, instrumented indentation tests and pin‐on‐disc sliding wear tests were performed. The results showed that nanocomposites with rGO contents of 0.10 and 0.25 wt% exhibited higher crystallinity degree than the neat UHMWPE, which improved significantly their mechanical and tribological behavior through increases of around 40% in the instrumented hardness and the elastic modulus, and a significant reduction of up to 48% in the coefficient of friction when compared to the neat UHMWPE. Additionally, the presence of rGO decreased the work of adhesion of the nanocomposites/counterface pairs, resulting in friction and wear mechanisms changes in comparison with the neat polymer. The recent ECMWF (European Centre of Medium‐Range Weather Forecasts) reanalysis product (ERA5) is a 3D‐atmospheric dataset of unparalleled value to advance the characterization and understanding of the main global coastal low‐level jet (CLLJ) systems, due to its finer resolution (horizontal and vertical) and hourly output. Regional climate studies have revealed the added value of higher spatial and temporal resolutions of models and reanalyses for the representation of CLLJs, which play an important role in the definition of the regional climate along the Eastern Boundary Current Systems. ERA5 reveals an improvement in the representation of the wind speed and temperature vertical profiles within the boundary layer in CLLJ regions. This improvement enables a better representation of CLLJs and therefore a new view on their features. ERA5 show larger CLLJ frequencies of occurrence and larger wind speed intensities, when compared with previous reanalysis. The maximum frequency of occurrence ranges from 20% in the West Australian and 73% in the Oman CLLJs, in the summer season, which constitutes an increase of 10% compared to ERA‐Interim. Moreover, a shift for higher wind speeds is also found, where the maximum wind speed approach 30 m·s−1. Typically, CLLJs are found below 500 m above sea level, where the finer vertical discretization results clearly in a better description of the CLLJ peak height. Regarding the diurnal cycles of CLLJs frequency of occurrence, ERA5 exposes higher peaks and larger amplitudes, where the maximum hourly frequency is about 38% in the Oman CLLJ and the diurnal amplitude can vary between 7 and 14%. The diurnal maximum and minimum frequency of CLLJ occurrence can be found between 1900 and 2200 UTC, and between 1000 and 1400 LST, respectively. The momentum and temperature budget profiles reveal significant differences between CLLJ regions and emphasize the relevance of local processes in its occurrences. [ABSTRACT FROM AUTHOR]

Regional climate simulations with high horizontal resolutions are becoming increasingly common and although model development has continually enhanced the representation of atmospheric phenomena, the model improvements are variable, region and time scale‐dependant. The high computational costs of increasingly smaller grid‐spacing underline the need for a robust assessment of the benefits or losses associated to the dynamical downscaling of coarser resolution models (reanalysis, global climate models or ~tenths km runs), that is, quantitative added value evaluation. In the current study, a probability density function (PDF) matching score is used to determine the distribution added value (DAV) of the EURO‐CORDEX maximum and minimum temperatures from the hindcast simulations at 0.44° and 0.11° resolutions. The gridded maximum and minimum temperatures from European Climate Assessment & Dataset (E‐Obs) were used as benchmarks for the matching scores and DAVs were determined for the 0.44° and 0.11° simulations individually against ERA‐Interim as well as against each other. Temperature added value against ERA‐Interim is difficult to find due to the assimilation of surface temperatures in the reanalysis. Nonetheless, there is positive added value in Tmax for half of the models at the yearly time scale, and the increase in resolution also implies positive DAV in half of the models, with DAVs between 0.1 and 3.8%. More importantly, the benefits of downscaling are significantly visible in the extreme end tail of Tmax, where all models have high added value at all resolutions and seasons except in summer (from ~1 to ~21%). In minimum temperature there is added value in winter and autumn at both resolutions. The Mediterranean and the British Isles are the regions where larger positive DAV values for both variables and resolutions are identified. In Tmax, added value is also associated to the increase in resolution (from 50 to 10 km) for half of the models in all regions. Scandinavia, the Alps, and eastern Europe are regions where negative added value is clearly linked with the highest mountain peaks for both variables. [ABSTRACT FROM AUTHOR]

Common bean is one of the main legumes used for food worldwide. Among the main limitations for its production is common bacterial blight (CBB) caused by Xanthomonascitri pv. fuscans (Xcf). The most desirable method to control the disease is the use of plant genetic resistance, but resistance against Xcf has not been sufficiently investigated. Genetic studies have shown that resistance to CBB is of quantitative type, but no gene associated with resistance has been identified. Here, we aimed to determine the resistance to CBB of 103 Mesoamerican common bean cultivars by evaluating disease severity and the area under the disease progress curve (AUDPC) after inoculation with a Xcf strain. In addition, 80 of the evaluated cultivars were used to investigate the association of single nucleotide polymorphisms (SNPs) with the resistance phenotype in a genome‐wide association study (GWAS). Twenty‐nine cultivars were classified as highly resistant, 15 of which are shown to be resistant to CBB for the first time in this work. A positive correlation between AUDPC and disease severity was observed. The 10 SNPs with the highest −log10(p) values in the GWAS are located on bean chromosomes Pv01, Pv03, Pv06, Pv07, Pv08, Pv09, Pv10, and Pv11 within or near genes predicted to code for biochemical functions such as serine/threonine‐protein kinase, glutamine synthase, lectin domain protein, MADS‐domain transcription factor, and pentatricopeptide repeat protein, previously associated with immunity. Biodiversity studies greatly benefit from molecular tools, such as DNA metabarcoding, which provides an effective identification tool in biomonitoring and conservation programmes. The accuracy of species‐level assignment, and consequent taxonomic coverage, relies on comprehensive DNA barcode reference libraries. The role of these libraries is to support species identification, but accidental errors in the generation of the barcodes may compromise their accuracy. Here, we present an R‐based application, Barcode, Audit & Grade System (BAGS) (https://github.com/tadeu95/BAGS), that performs automated auditing and annotation of cytochrome c oxidase subunit I (COI) sequences libraries, for a given taxonomic group of animals, available in the Barcode of Life Data System (BOLD). This is followed by implementing a qualitative ranking system that assigns one of five grades (A to E) to each species in the reference library, according to the attributes of the data and congruency of species names with sequences clustered in barcode index numbers (BINs). Our goal is to allow researchers to obtain the most useful and reliable data, highlighting and segregating records according to their congruency. Different tests were performed to perceive its usefulness and limitations. BAGS fulfils a significant gap in the current landscape of DNA barcoding research tools by quickly screening reference libraries to gauge the congruence status of data and facilitate the triage of ambiguous data for posterior review. Thereby, BAGS has the potential to become a valuable addition in forthcoming DNA metabarcoding studies, in the long term contributing to globally improve the quality and reliability of the public reference libraries. In a recent study, Coppola et al. assessed the ability of an ensemble of convection‐permitting models (CPM) to simulate deep convection using three case studies. The ensemble exhibited strong discrepancies between models, which were attributed to various factors. In order to shed some light on the issue, we quantify in this article the uncertainty associated to different physical parameterizations from that of using different initial conditions, often referred to as the internal variability. For this purpose, we establish a framework to quantify both signals and we compare them for upper atmospheric circulation and near‐surface variables. The analysis is carried out in the context of the CORDEX Flagship Pilot Study on Convective phenomena at high resolution over Europe and the Mediterranean, in which the intermediate RCM WRF simulations that serve to drive the CPM are run several times with different parameterizations. For atmospheric circulation (geopotential height), the sensitivity induced by multi‐physics and the internal variability show comparable magnitudes and a similar spatial distribution pattern. For 2‐m temperature and 10‐m wind, the simulations with different parameterizations show larger differences than those launched with different initial conditions. The systematic effect over 1 year shows distinct patterns for the multi‐physics and the internal variability. Therefore, the general lesson of this study is that internal variability should be analysed in order to properly distinguish the impact of other sources of uncertainty, especially for short‐term sensitivity simulations. Land‐atmosphere coupling plays a crucial role in determining the evolution of weather and climate. In the current study, the full ensemble of CORDEX‐Africa climate change simulations is used to understand how strong and weak coupling regions in Africa will evolve in the future. The ability of the regional climate models to capture the coupling signal relies on a reasonable representation of near surface air temperature, precipitation, surface fluxes, and soil moisture. A thorough model evaluation reveals typical shortcomings in the representation of the African climate, in particular seasonal precipitation. The multimodel ensemble mean outperforms the individual models and is therefore used for the investigation of the land‐atmosphere coupling. This ensemble mean shows a widespread warming in Africa and changes in precipitation, such as a decrease in the Sahel during summer and an increase in western Africa during summer and autumn. The coupling analysis relies on surface fluxes, the related evaporative fraction and their correlations as well as the correlation between evaporative fraction and soil moisture. Overall, water‐limited regions that exhibit a strong land‐atmosphere coupling are projected to expand further southward in West Africa and further northward in southern Africa. This is particularly true over the Sahel during spring and summer, when the strong coupling region shifts southward, indicating a potential expansion of the semiarid and arid regions. A transition of energy limited regimes, with weak coupling, to water‐limited regimes where soil moisture plays a more important role, is projected for the end of the 21st century as drying continues. The spatial dependence of meteorological variables is crucial for many impacts, for example, droughts, floods, river flows, energy demand, and crop yield. There is thus a need to understand how well it is represented in downscaling (DS) products. Within the COST Action VALUE, we have conducted a comprehensive analysis of spatial variability in the output of over 40 different DS methods in a perfect predictor setup. The DS output is evaluated against daily precipitation and temperature observations for the period 1979–2008 at 86 sites across Europe and 53 sites across Germany. We have analysed the dependency of correlations of daily temperature and precipitation series at station pairs on the distance between the stations. For the European data set, we have also investigated the complexity of the downscaled data by calculating the number of independent spatial degrees of freedom. For daily precipitation at the German network, we have additionally evaluated the dependency of the joint exceedance of the wet day threshold and of the local 90th percentile on the distance between the stations. Finally, we have investigated regional patterns of European monthly precipitation obtained from rotated principal component analysis. We analysed Perfect Prog (PP) methods, which are based on statistical relationships derived from observations, as well as Model Output Statistics (MOS) approaches, which attempt to correct simulated variables. In summary, we found that most PP DS methods, with the exception of multisite analog methods and a method that explicitly models spatial dependence yield unrealistic spatial characteristics. Regional climate model‐based MOS methods showed good performance with respect to correlation lengths and the joint occurrence of wet days, but a substantial overestimation of the joint occurrence of heavy precipitation events. These findings apply to the spatial scales that are resolved by our observation network, and similar studies with higher resolutions, which are relevant for small hydrological catchment, are desirable. This work analyses three uncertainty sources affecting the observation‐based gridded data sets: station density, interpolation methodology and spatial resolution. For this purpose, we consider precipitation in two countries, Poland and Spain, three resolutions (0.11, 0.22 and 0.44°), three interpolation methods, both areal‐ and point‐representative implementations, and three different densities of the underlying station network (high/medium/low density). As a result, for each resolution and interpolation approach, nine different grids have been obtained for each country and inter‐compared using a variance decomposition methodology. Results indicate larger differences among the data sets for Spain than for Poland, mainly due to the larger spatial variability and complex orography of the former region. The variance decomposition points out to station density as the most influential factor, independent of the season, the areal‐ or point‐representative implementation and the country considered, and slightly increasing with the spatial resolution. In contrast, the decomposition is stable when extreme precipitation indices are considered, in particular for the 50‐year return value. Finally, the uncertainty due to station sub‐sampling inside a particular grid box decreases with the number of stations used in the averaging/interpolation. In the case of spatially homogeneous grid boxes, the interpolation approach obtains similar results for all the parameters, excepting the wet day frequency, independently of the number of stations. When there is a more significant internal variability in the grid box, the interpolation is more sensitive to the number of stations, pointing out to a minimum stations' density for the target resolution (six to seven stations). Credible information about the properties and changes of extreme events on the regional and local scales is of prime importance in the context of future climate change. Within the EU‐COST Action VALUE a comprehensive validation framework for downscaling methods has been developed. Here we present validation results for extremes of temperature and precipitation from the perfect predictor experiment that uses reanalysis‐based predictors to isolate downscaling skill. The raw reanalysis output reveals that there is mostly a large bias with respect to the extreme index values at the considered stations across Europe, clearly pointing to the necessity of downscaling. The performance of the downscaling methods is closely linked to their specific structure and setup. All methods using parametric distributions require non‐standard distributions to correctly represent marginal aspects of extremes. Also, the performance is much improved by explicitly including a seasonal component, particularly in case of precipitation. With respect to the marginal aspects of extremes the best performance is found for model output statistics (MOS), weather generators (WGs) as well as perfect prognosis (PP) methods using analogues. Spell‐length‐related extremes of temperature are best assessed by MOS and WGs, spell‐length‐related extremes of precipitation by MOS and PP methods using analogues. The influence of uncertainties in gridded observational reference data on regional climate model (RCM) evaluation is quantified on a pan‐European scale. Three different reference data sets are considered: the coarse‐resolved E‐OBS data set, a compilation of regional high‐resolution gridded products (HR) and the European‐scale MESAN reanalysis. Five high‐resolution ERA‐Interim‐driven RCM experiments of the EURO‐CORDEX initiative are evaluated against each of these references over eight European sub‐regions and considering a range of performance metrics for mean daily temperature and daily precipitation. The spatial scale of the evaluation is 0.22°, that is, the grid spacing of the coarsest data set in the exercise (E‐OBS). While the three reference grids agree on the overall mean climatology, differences can be pronounced over individual regions. These differences partly translate into RCM evaluation uncertainty. For most cases observational uncertainty is smaller than RCM uncertainty. Nevertheless, for individual sub‐regions and performance metrics observational uncertainty can dominate. This is especially true for precipitation and for metrics targeting the wet‐day frequency, the pattern correlation and the distributional similarity. In some cases the spatially averaged mean bias can also be considerably affected. An illustrative ranking exercise highlights the overall effect of observational uncertainty on RCM ranking. Over individual sub‐domains, the choice of a specific reference can modify RCM ranks by up to four levels (out of five RCMs). For most cases, however, RCM ranks are stable irrespective of the reference. These results provide a twofold picture: model uncertainty dominates for most regions and for most performance metrics considered, and observational uncertainty plays a minor role. For individual cases, however, observational uncertainty can be pronounced and needs to be definitely taken into account. Temporal variability is an important feature of climate, comprising systematic variations such as the annual cycle, as well as residual temporal variations such as short‐term variations, spells and variability from interannual to long‐term trends. The EU‐COST Action VALUE developed a comprehensive framework to evaluate downscaling methods. Here we present the evaluation of the perfect predictor experiment for temporal variability. Overall, the behaviour of the different approaches turned out to be as expected from their structure and implementation. The chosen regional climate model adds value to reanalysis data for most considered aspects, for all seasons and for both temperature and precipitation. Bias correction methods do not directly modify temporal variability apart from the annual cycle. However, wet day corrections substantially improve transition probabilities and spell length distributions, whereas interannual variability is in some cases deteriorated by quantile mapping. The performance of perfect prognosis (PP) statistical downscaling methods varies strongly from aspect to aspect and method to method, and depends strongly on the predictor choice. Unconditional weather generators tend to perform well for the aspects they have been calibrated for, but underrepresent long spells and interannual variability. Long‐term temperature trends of the driving model are essentially unchanged by bias correction methods. If precipitation trends are not well simulated by the driving model, bias correction further deteriorates these trends. The strong coastal upwelling associated to the Benguela eastern boundary upwelling system makes the ocean along coast of this current one of the most productive ecosystems in the world. The Benguela Coastal Low‐Level Jet (BCLLJ) is one of the most important mesoscale feature that shape the climate of this region. The main synoptic forcing of the BCLLJ is the Angola thermal low over land and the St. Helen anticyclone over the ocean, resulting in southwesterly winds along the coast. This study investigates how the BCLLJ might change due to climate warming, with the help of uncoupled and coupled simulations from a 25‐km horizontal resolution regional climate model (ROM). In general, the coupled simulation displays the best performance in representing the present time near‐surface wind speed, with a decrease on the known warm bias of sea surface temperature in the Benguela eastern boundary upwelling system region. The analysis of the projected changes of the BCLLJ climate toward the end of the 21st century (2070–2099), following the RCP8.5 emissions scenario, shows an increase in the frequency of the BCLLJ occurrence along the southern area with higher changes in the coupled simulation (between 6% and 8%). These changes are related to a southerly shift of the St. Helen High, which intensifies the flow offshore the west coast of South Africa and causes a sharpening of the land‐sea thermal contrasts. However, during spring, associated with the decrease in near‐surface wind speed due to higher sea surface temperatures, the future frequency and intensity of the BCLLJ are lower. The Benguela coastal low‐level jet (CLLJ) is characterized by intense winds that occur around 400 m above sea level, within or at the top of the marine atmospheric boundary layer. The semipermanent St. Helen high‐pressure system, over the South Atlantic Ocean, and the inland thermal low‐pressure system, over the Namib Desert, are the synoptic forcing behind the Benguela CLLJ. This coastal jet is an important mesoscale feature in the Namibia and Angola coastal areas, since it is present virtually all year round, with a marked seasonal cycle. This study investigates the climatology of the frequency and the intensity of the Benguela CLLJ and its relationship with synoptic and local forcing's, using high‐resolution modeling. An uncoupled ROM (REMO‐OASIS‐MPIOM) hindcast simulation, with 25‐km horizontal resolution, for the period 1980–2014, is used to analyze the features of the Benguela CLLJ. It is shown that Benguela CLLJ is characterized by two local maxima of frequency of occurrence at around 26°S and 17.5°S. During austral summer, the jet has a frequency of occurrence of about 60% and it is in its southern core. During autumn and winter, the frequency of occurrence decreases to nearly half and migrates equatorward. During spring, the jet has a frequency of occurrence of 45% and is found at the northern core. The jet wind speed is higher in the south core but is found at higher altitudes in the north core. The seasonal cycle of horizontal momentum budget defines the seasonality of Benguela CLLJ frequency of occurrence. Diversification and speciation of terrestrial organisms are anticipated in oceanic islands such as Macaronesia, a group of Atlantic islands that have remained unconnected to continental landmasses. Hitherto, the diversification of marine organisms in oceanic islands, especially those with low vagility, has received little direct empirical analysis using molecular markers. Here, we focus on such a case study, through applying a multilocus molecular approach to investigate the diversity and evolution of a group that lacks a planktonic larval stage, the isopod genus Dynamene, in Macaronesia and Northeast Atlantic. Sequences of two mitochondrial (cytochrome c oxidase subunit I and 16S rRNA) and two nuclear (18S rRNA and 28S rRNA) loci were obtained from specimens of Dynamene edwardsi (Lucas, 1849), Dynamene magnitorata Holdich, 1968 and Dynamene bidentata (Adams, 1800) collected along the Northeast Atlantic and Macaronesia. Although no major phylogeographic structure was detected in D. bidentata and D. magnitorata, from five to nine deeply divergent lineages were evident within D. edwardsi. The divergent lineages displayed genetic distances comparable to those found among established species of peracarids. D. edwardsi exhibits a long, rich and complex phylogeographic history in Macaronesia, where the geodynamics of the islands possibly associated with founder effects and subsequent lack of gene flow among populations confounds patterns based on geographic proximity of targeted populations. Our findings collectively suggest a much larger role of oceanic islands in the diversification of marine invertebrates than previously anticipated. Aim: To investigate the influence of breathing patterns and blood pressure behavior postural control in older adults. Methods: A total of 20 older adults carried out spontaneous, controlled (15 cycles/min) and deep (6 cycles/min) breathing trials, in random order. In this session, the heart rate was recorded continuously and blood pressure measured every 3 min. In addition, the challenge from sitting to an active standing position was carried out with eyes closed during 6 min on a baropodometer plate with electromyography recording. The root mean square (RMS) of electromyography was calculated. Results: There were significant differences in the postural control with spontaneous breathing from the first minute (center of pressure [COP]‐area 216.40 ± 116.70 mm2) in comparison with the third minute (COP‐area 79.35 ± 68.11 mm2; d = 0.89; P < 0.01) and the sixth minute (COP‐area 70.24 ± 41.26 mm2; d = 0.60; P < 0.05) of active standing. Furthermore, deep breathing significantly increases the COP‐area (158.50 ± 126.2 mm2) at the sixth minute of active standing in comparison with spontaneous breathing (70.24 ± 41.26 mm2; d = 0.80; P < 0.01). There were also significant differences in systolic blood pressure during spontaneous breathing between the baseline measures (sitting position 123 ± 11 mmHg) and the first minute of active standing (117 ± 13 mmHg; d = 0.24; P < 0.05). Furthermore, electromyography activity of the tibialis anterior increased during the deep (17.52 ± 9.21 RMS) and controlled breathing (16.75 ± 5.26 RMS) compared with the spontaneous condition (14.93 ± 5.56 RMS; d = 0.17; P < 0.05). Conclusions: The present data provide important insight into the respiratory and hemodynamics effects of postural control in older adults. Objectives/hypothesis: Evaluate the effect of in vitro exposure of mice laryngeal mucosa to solutions that simulated human gastric juice and to assess the topical protective effect of cashew gum on mice laryngeal mucosal integrity in vitro.Study Design: Animal study.Methods: Murine (Swiss) laryngeal samples were mounted in Ussing chambers. The luminal side of biopsies was exposed to solutions of different acidity with or without pepsin and/or taurodeoxycholic acid (TDC). Transepithelial electrical resistance (TER) was continuously recorded. The topical protective effect of cashew gum solution was evaluated by precoating the biopsies before the exposure with a solution at pH 5 containing 5 mM TDC. Changes in TER and mucosal permeability to fluorescein were measured.Results: Exposure of laryngeal mucosa to acidic solutions containing pepsin and TDC provoked a pH-dependent drop in TER with the maximal effect at pH 1, but still present at pH 5 (weakly acidic). The exposure of the laryngeal mucosa to a solution of pH 5 with TDC, but not with pepsin, produced a dose-dependent decrease in TER. Precoating the mucosa with cashew gum prevented the reduction of TER and increased transepithelial permeability by exposure to a solution at pH5 containing TDC.Conclusions: Weakly acidic solutions containing bile acids can produce impairment of laryngeal epithelial barrier, which may be protected by topical treatment with cashew gum.Level Of Evidence: NA. The spatial variability and the climatic characteristics of the coastal low-level jet along the west coast of the Iberian Peninsula are presented in this study, based on a 9-km resolution downscaling data set, produced using the weather research and forecasting model, forced by the ERA-Interim reanalysis. The west coast of Iberia is divided into two (north and south) subareas, where the Iberian Peninsula coastal jet (IPCJ) characteristics are studied separately, and compared to the whole western Iberia. It is shown that while IPCJ occurrences are more frequent in the southern area, during summer (June–August), the jet mean wind speed maxima is highest in the northernmost area. The along- and cross-coast sea surface temperature (SST) structure is studied and linked to the coastal wind speed. It is also shown that the local cross-coast marine atmospheric boundary layer gradient at the coast is considerably higher than the synoptic one, and that this difference is responsible for the local intensification of the wind speed. The flow interaction with the three major capes along the west coast of Iberia (Finisterre, Roca, and Saint Vincent) is responsible for local higher occurrence of the IPCJ, as well as the downwind intensification of the wind speed. The along-coast west Iberia wind speed structure due to IPCJ occurrences is shown. Regional climate models (RCMs) are used with increasing resolutions seeking to represent in an improved way regional to local-scale atmospheric phenomena. The EURO-CORDEX simulations at 0.11° and simulations exploiting finer grid spacing approaching the convective-permitting regimes are representative examples. These climate runs are computationally very demanding and do not always show improvements, which depend on the region, variable and object of study. The gains or losses associated with the use of higher resolution in relation to the forcing model (global climate model or reanalysis), or to different resolution RCM simulations, are widely known as added value. Its characterization is a long-standing issue, and many different added-value measures have been proposed. In the current study, a new method is proposed to assess the added value of finer-resolution simulations, in comparison to its forcing data or coarser-resolution counterparts. This approach builds on a probability density function (PDF) matching score, giving a normalized measure of the difference between diverse resolution PDFs, mediated by the observational ones. The distribution added value (DAV) is an objective added-value measure that can be applied to any variable, region or temporal scale, from hindcast or historical (non-synchronous) simulations. The DAVs metric and its application to the EURO-CORDEX hindcast daily precipitation data are presented here. Generally, the EURO-CORDEX simulations at both resolutions (0.44° and 0.11°) display a clear added value in relation to ERA-Interim, with maximum values around 30% in summer and 20% in the intermediate seasons. When both RCM resolutions are directly compared, only three of five models (0.11°) show added value, with a maximum of ∼10%. The regions with the larger DAVs are areas where convection is relevant, e.g. Alps and Iberia. The economic damage to Brazilian soybean [Glycine max (L.) Merr.] production attributed to Pratylenchus brachyurus has increased in recent years. The objective of this study was to determine the influence of soil properties on nematode variability in a soybean crop. Soil and root samples (0–0.20 m) were collected from 142 points in an area that was 180 by 180 m. Root samples were analyzed for nematodes, and soil samples were analyzed for chemical attributes. The data were analyzed using principal component analysis and geostatistics. The spatial variability maps of the attributes S, Ca, Mg, K, sum of bases, base saturation index, and cation exchange capacity were aligned wiThthe water runoffdirection. Soil organic matter and K were associated wiThvery low number of nematodes in the root (NNR) (NNR £ 3614), low (3614 < NNR £ 10,496), and moderate (10,496 < NNR £ 21,248). On the other hand, the regions wiThhigh Mg, S, and soybean productivity were associated wiThregions wiThhigh (21,248 < NNR £ 43,648) and very high (NNR > 43,648) NNR. The number of people affected by neurodegenerative disorders such as Alzheimer's disease and Parkinson's disease is rapidly increasing owing to the global increase in life expectancy. Small molecules with neurotrophic effects have great potential for management of these neurological disorders. In this study, different (C1-C12) alkyl ester derivatives of hydroxycinnamic acids ( HCAs) were synthesized (a total of 30 compounds). The neurotrophic capacity of the test compounds was examined by measuring promotion of survival in serum-deprived conditions and enhancement of nerve growth factor ( NGF)-induced neurite outgrowth in PC12 neuronal cells. p-Coumaric, ferulic, and sinapic acids and their esters did not alter cell survival, while caffeic acid and all its alkyl esters, especially decyl and dodecyl caffeate, significantly promoted neuronal survival at 25 μ m. Methyl, ethyl, propyl, and butyl caffeate esters also significantly enhanced NGF-induced neurite outgrowth, among which the most effective ones were propyl and butyl esters, which at 5 μ m led to 25- and 22-fold increases in the number of neurites, respectively. The findings of the docking study suggested phosphatidylinositol 3-kinase ( PI3K) as the potential molecular target. In conclusion, our findings demonstrate that alkyl esters of caffeic acid can be useful as scaffolds for the discovery of therapeutic agents for neurodegenerative diseases. [ABSTRACT FROM AUTHOR]

ABSTRACT The summer time cloud diurnal cycle over western Iberia is analysed here using a satellite climate data record of fractional cloud cover based on 9 years of Meteosat Second Generation observations which is distributed by the EUMETSAT's Climate Monitoring Satellite Applications Facility. These observations were complemented with a corresponding mean cloud diurnal cycle using SYNOP reports on six locations over the studied domain. It is shown that the main coastal mountain range separates regions that are characterized by two very different cloud regimes: stratocumulus-topped boundary layer convection dominates the region towards the coast and continental cumulus convection dominates the region to the east of these mountains. To explain the observed variability, a long-term regional climate model [Weather Research and Forecasting model ( WRF)] simulation over Iberia was used. A comparison of the observations against model output for the common period between observations and simulation shows that although the model generally underestimates cloudiness, it is able to represent the diurnal cycle in a realistic manner. It is shown that the observed cloud diurnal evolution is linked to the thermal circulations generated by the land-sea contrast and orography. The extent to which the cloud deck penetrates inland is closely related to the coastal orography: although smaller hills tend to enhance cloudiness, larger mountains block the progression of the marine boundary layer further inland, as it behaves as a density current. Larger mountains also produce katabatic flow and a rather strong subsidence aloft during the night. The warming due to this subsidence helps the blocking of the cloud deck as it is partially responsible for evaporating clouds, as shown by a potential temperature budget analysis. In this study, the amino acid sequence and anti-inflammatory effect of Bauhinia bauhinioides (BBL) lectin were evaluated. Tandem mass spectrometry revealed that BBL possesses 86 amino acid residues. BBL (1 mg/kg) intravenously injected in rats 30 min prior to inflammatory stimuli inhibited the cellular edema induced by carrageenan in only the second phase (21% - 3 h, 19% - 4 h) and did not alter the osmotic edema induced by dextran. BBL also inhibited carrageenan peritoneal neutrophil migration (51%), leukocyte rolling (58%) and adhesion (68%) and the neutrophil migration induced by TNF-α (64%). These effects were reversed by the association of BBL with galactose, demonstrating that the carbohydrate-binding domain is essential for lectin activity. In addition, BBL reduced myeloperoxidase activity (84%) and TNF-α (68%) and IL1-β (47%) levels. In conclusion, the present investigation demonstrated that BBL contains highly homologous isolectins, resulting in a total of 86 amino acid residues, and exhibits anti-inflammatory activity by inhibiting neutrophil migration by reducing TNF-α and IL1-β levels via the lectin domain. The presence of sub-Saharan L-type mtDNA sequences in North Africa has traditionally been explained by the recent slave trade. However, gene flow between sub-Saharan and northern African populations would also have been made possible earlier through the greening of the Sahara resulting from Early Holocene climatic improvement. In this article, we examine human dispersals across the Sahara through the analysis of the sub-Saharan mtDNA haplogroup L3e5, which is not only commonly found in the Lake Chad Basin (~17%), but which also attains nonnegligible frequencies (~10%) in some Northwestern African populations. Age estimates point to its origin ~10 ka, probably directly in the Lake Chad Basin, where the clade occurs across linguistic boundaries. The virtual absence of this specific haplogroup in Daza from Northern Chad and all West African populations suggests that its migration took place elsewhere, perhaps through Northern Niger. Interestingly, independent confirmation of Early Holocene contacts between North Africa and the Lake Chad Basin have been provided by craniofacial data from Central Niger, supporting our suggestion that the Early Holocene offered a suitable climatic window for genetic exchanges between North and sub-Saharan Africa. In view of its younger founder age in North Africa, the discontinuous distribution of L3e5 was probably caused by the Middle Holocene re-expansion of the Sahara desert, disrupting the clade's original continuous spread. In the present study we evaluated the effects of short-term pyridostigmine bromide (0.14 mg/mL) treatment started early after myocardial infarction ( MI) on left ventricular ( LV) and autonomic functions in rats., Male Wistar rats were divided into control, pyridostigmine, infarcted and infarcted + pyridostigmine-treated groups. Pyridostigmine was administered in the drinking water, starting immediately after MI or sham operation, for 11 days. Left ventricular function was evaluated indirectly by echocardiography and directly by LV catheterization. Cardiovascular autonomic control was evaluated by baroreflex sensitivity ( BRS), heart rate variability ( HRV) and pharmacological blockade. All evaluations started after 7 days pyridostigmine treatment and were finalized after 11 days treatment., Pyridostigmine prevented the impairment of +d P/d T and reduced the MI area in infarcted + pyridostigmine compared with infarcted rats (7 ± 3% vs 17 ± 4%, respectively). Mean blood pressure was restored in infarcted + pyridostigmine compared with infarcted rats (103 ± 3 vs 94 ± 3 mmHg, respectively). In addition, compared with the infarcted group, pyridostigmine improved BRS, as evaluated by tachycardic (1.6 ± 0.2 vs 2.5 ± 0.2 b.p.m./mm Hg, respectively) and bradycardic (−0.42 ± 0.01 vs −1.9 ± 0.1 b.p.m./mm Hg) responses, and reduced the low frequency/high frequency ratio of HRV (0.81 ± 0.11 vs 0.24 ± 0.14, respectively). These improvements are probably associated with increased vagal tone and reduced sympathetic tone in infarcted + pyridostigmine compared with infarcted rats., In conclusion, the data suggest that short-term pyridostigmine treatment started early after MI can improve BRS, HRV and parasympathetic and sympathetic tone in experimental rats. It is now known that several population movements have taken place at different times throughout southern Arabian prehistory. One of the principal questions under debate is if the Early Holocene peopling of southern Arabia was mainly due to input from the Levant during the Pre-Pottery Neolithic B, to the expansion of an autochthonous population, or some combination of these demographic processes. Since previous genetic studies have not been able to include all parts of southern Arabia, we have helped fill this lacuna by collecting new population datasets from Oman (Dhofar) and Yemen (Al-Mahra and Bab el-Mandab). We identified several new haplotypes belonging to haplogroup R2 and generated its whole genome mtDNA tree with age estimates undertaken by different methods. R2, together with other considerably frequent southern Arabian mtDNA haplogroups (R0a, HV1, summing up more than 20% of the South Arabian gene pool) were used to infer the past effective population size through Bayesian skyline plots. These data indicate that the southern Arabian population underwent a large expansion already some 12 ka. A founder analysis of these haplogroups shows that this expansion is largely attributed to demographic input from the Near East. These results support thus the spread of a population coming from the north, but at a significantly earlier date than presently considered by archaeologists. Our data suggest that some of the mtDNA lineages found in southern Arabia have persisted in the region since the end of the Last Ice Age. Eucalyptol, also known as 1,8-cineole, is a monoterpene traditionally used to treat respiratory disorders due to its secretolytic properties. In addition to its myorelaxant effects, it also has anti-inflammatory actions in vitro. In this study, we aimed to evaluate the efficacy of acute treatment with 1,8-cineole on reducing airway inflammatory parameters. Ovalbumin (OVA)-sensitized guinea pigs were submitted to antigenic challenge (OVA) with or without pre-treatment with a single dose of 1,8-cineole administered by inhalation. Airway inflammatory parameters were reduced or absent in 1,8-cineole-treated animals as compared with untreated guinea pigs. Acute treatment with 1,8-cineole impaired the development of airway hyperresponsiveness to carbachol in isolated tracheal rings. Levels of the pro-inflammatory cytokines TNFα and IL-1β was lower in bronchoalveolar lavage fluid (BALF) of 1,8-cineol-treated guinea pigs than in untreated animals. 1,8-Cineole impaired the OVA-induced increase of the myeloperoxidase activity in BALF. 1,8-Cineole also prevented the reduction of the mucociliary clearance induced by the antigen presentation. 1. The aim of the present study was to investigate the cardiovascular effects of anabolic androgenic steroid (AAS) abuse by comparing the electrocardiographic parameters before and after submaximal exercise between AAS users and non-AAS users. 2. A total of 22 men who regularly engaged in both resistance and aerobic exercise at fitness academies volunteered for the study (control group: n = 11, age 25 ± 4 years; AAS group: n = 11, age 27 ± 5 years). All subjects were submitted to submaximal exercise testing using an Astrand-Rhyming protocol. Heart rate and electrocardiography parameters were measured at rest and at the third minute of the post-exercise recovery time. 3. AAS users presented higher QTc and QTd at rest (10% and 55%, respectively) and at the post-exercise period (17% and 43%, respectively), compared with control subjects. The maximal and minimum QTc interval of the AAS group was significantly prolonged at the post-exercise period (12% and 15%, respectively). The haemodynamic parameters were similar in both groups ( P > 0.05). The AAS group showed a lower heart rate recovery at the first minute after the test ( P = 0.0001), and a higher exertion score ( P < 0.0001) at a lower workload, compared with the control group. 4. Our results show that the QTc interval and dispersion are increased in individuals who abuse AAS, suggesting the presence of ventricular repolarization abnormalities that could potentially increase the risk of cardiac arrhythmias and sudden cardiac death. BACKGROUND AND PURPOSE Within the spectrum of reversible neuroimaging abnormalities induced by status epilepticus (SE) tumor-like lesions (TLL) have been rarely described. Their etiology, pathophysiology, and long-term outcome remain uncertain. These issues could be clarified by long-term magnetic resonance imaging (MRI) studies in TLL induced by presenting SE. METHODS Prospective multi-sequence MRI and clinical and electroencephalographic long-term (18 to 60 months) follow-up studies were performed in 3 patients with reversible TLL induced by presenting SE. RESULTS In the peri-ictal MRI, TLL are hypointense in T1-weighted, hyperintense in T2-weighted, and fluid-attenuated inversion recovery images with a marked subcortical component and sulci effacement. The diffusion and MR-spectroscopy studies disclosed intermixed areas of increased/decreased diffusivity associated with a lactate peak and a decreased N-acetylaspartate. At long-term follow-up, none of the patients had seizure recurrence or electroencephalographic epileptiform abnormalities; MRI showed residual focal atrophy and gliosis associated with neuronal loss/dysfunction. CONCLUSIONS SE per se may induce TLL. MRI multi-sequence studies disclosed that they are mainly formed by focal vasogenic and cytotoxic edema resulting from the hypermetabolism associated with seizure activity. 1: We have investigated the inhibitory effects of blockers of volume-activated (Clvol) and calcium-activated (ClCa) chloride channels on hypotonic solution (HS)-induced contractions of rat trachea, comparing their effects with those of the voltage-dependent calcium channel (VDCC) blocker nifedpine. 2: HS elicited large, stable contractions that were partially dependent on the cellular chloride gradient; a reduction to 41.45±7.71% of the control response was obtained when extracellular chloride was removed. In addition, HS-induced responses were reduced to 26.8±5.6% of the control by 1?µM nifedipine, and abolished under calcium-free conditions, indicating a substantial requirement for extracellular calcium entry, principally via VDCCs. 3: The established Clvol blockers tamoxifen (?10?µM) and 4,4'-diisothiocyanatostilbene-2,2'-disulphonic acid (1-100?µM), at concentrations previously reported to inhibit Clvol in smooth muscle, did not significantly inhibit HS-induced contractions. 4: In contrast, the recognized ClCa blocker niflumic acid (NFA; 1-100?µM) produced a reversible, concentration-dependent inhibition of HS responses, with a reduction to 36.6±6.4% of control contractions at the highest concentration. The mixed Clvol and ClCa blocker, 5-nitro 2-(3-phenylpropylamine) benzoic acid (NPPB; 10-100?µM) also elicited concentration-related inhibition of HS-induced contractions, producing a decrease to 35.9±11.3% of the control at 100?µM. 5: Our results show that HS induces reversible, chloride-dependent contractions of rat isolated trachea that were inhibited by NFA and NPPB, while exhibiting little sensitivity to recognized blockers of Clvol.
