Though sterile diet, post‐transplantation surgery is a clinical strategy for patient care to prevent the infiltration of gut pathogens, less is known about its effects on the gut microbiome. Here, the gut microbiome dynamics of leukemia patients following a 120‐day "sterile‐normal" diet strategy posthematopoietic cell transplantation are examined. In contrast to the traditional idea, a sterile diet leads to the lowest gut microbiota diversity (p < 0.05) and short‐chain fatty acids, promoted the proliferation of potential pathogens such as Streptococcus (up by 16.93%) and Lactobacillus (up by 40.30%), and 43.32% reduction in nodes and an 85.33% reduction in edges within the microbial interaction's network. Interestingly, a normal diet allows the gut microbiome recovery and significantly promotes the abundance of beneficial bacteria. These results indicate that a sterile diet leads to a collapse of the patient's gut microbiome and promoted the proliferation of potential pathogens. This assay is a starting point for a more sophisticated assessment of the effects of a sterile diet. The work also suggests a basic principle for the re‐establishment of microbial equilibrium that supplementation of microbial taxa may be the key to the restoration of the degraded ecosystem. The insect resistance of Bacillus thuringiensis (Bt) transgenic rice (Oryza sativa L.) is mainly determined by the transcription of the CRY1C gene and the translation process of the Cry1C protein. With this in mind, we analyzed CRY1C expression and Cry1C protein content in a transgenic Bt insect‐resistant restorer line and its F1 hybrids, evaluated the resistance to Chilo suppressalis of various lines of rice, and determined the 50% lethal concentration (LC50) of Cry1C for C. suppressalis. In four transgenic rice lines, the relative expression of CRY1C was highest at the heading stage in most tissues. Among different tissues from the same developmental stage, CRY1C expression was highest in leaves, followed by stems and panicles. The relative expression of CRY1C was higher in the parent restorer line than in the F1 hybrids. The LC50 of the Cry1C protein for C. suppressalis was 4.016 μg g−1, and the Cry1C protein expression level of transgenic insect‐resistant rice exceeded this threshold at the heading stages and in stems. We next analyzed the lethality of each strain toward C. suppressalis. After 48 h, the mortality rate of second‐instar C. suppressalis larvae feeding on transgenic stem tissue was higher at the heading stage than at the tiller stage and varied from 66.7 to 91.7%. By documenting the temporal and spatial expression of CRY1C and evaluating C. suppressalis resistance to Bt transgenic rice, the risk of pests and diseases during the rice production process can be greatly reduced. Abstract: YL143 was identified as a novel wild‐type sparing EGFRT790M inhibitor with good pharmacokinetic properties. It potently suppresses EGFRL858R/T790M with an 50% inhibitory concentration (IC50) value of 2.0 ± 0.3 nmol/L, but is approximately 92‐folds less potent against EGFRWT kinase. YL143 suppresses cellular proliferation and induces G0/G1 phase arrest and apoptosis in H1975 cells with EGFRL858R/T790M mutation at 30 nmol/L. It also exhibits acceptable pharmacokinetics (PK) parameters with an oral bioavailability value of 25.0% after oral administration in rats and exhibits promising antitumor efficacy in a gefitinib‐resistant human H1975 xenografted model after oral administration of 30 mg/kg/day. These data supported that YL143 could be a promising lead compound for overcoming clinical EGFRT790M resistance of patients with non‐small‐cell lung cancer (NSCLC). [ABSTRACT FROM AUTHOR]

Manure fertilizers are widely used in agriculture and highly impacted the soil microbial communities such as ammonia oxidizers. However, the knowledge on the communities of archaeal versus bacterial ammonia oxidizers in paddy soil affected by manure fertilization remains largely unknown, especially for a long-term influence. In present work, the impact of manure fertilization on the population of ammonia oxidizers, related potential nitrification rates (PNRs) and the key factors manipulating the impact were investigated through studying two composite soil cores (long-term fed with manure fertilization versus undisturbed). Moreover, soil incubated with NH4+ for 5 weeks was designed to verify the field research. The results showed that the copy numbers of bacterial amoA gene in the manure fed soil were significant higher than those in the unfed soil ( p < 0.05), suggesting a clear stimulating effect of long-term manure fertilization on the population of ammonia-oxidizing bacteria (AOB). The detected PNRs in the manure fed soil core (14-218 nmol L−1 N g−1 h−1) were significant higher than those in the unfed soil core (5-72 nmol L−1 N g−1 h−1; p < 0.05). Highly correlations between the PNRs and the bacterial amoA gene copies rather than archaeal amoA gene were observed, indicating strong nitrification capacity related to bacterial ammonia oxidizers. The NH4+-N significantly correlated to the abundance of AOB ( p < 0.01) and explained 96.1% of the environmental variation, showing the NH4+-N was the main factor impacting the population of AOB. The incubation experiment demonstrated a clear increase of the bacterial amoA gene abundance (2.0 × 106 to 8.4 × 106 g−1 d.w.s. and 1.6 × 104 to 4.8 × 105 g−1 d.w.s.) in both soil but not for the archaeal amoA gene, in agreement with the field observation. Overall, our results suggested that manure fertilization promoted the population size of bacterial ammonia oxidizers rather than their archaeal counterparts whether in long-term or short-term usage and the NH4+-N was the key impact factor. The antiinflammatory effect of the total flavonoids of Laggera pterodonta (TFLP) was evaluated with various in vivo models of both acute and chronic inflammation. In the acute inflammation tests, TFLP significantly inhibited xylene-induced mouse ear oedema, carrageenan-induced rat paw oedema and acetic acid-induced mouse vascular permeability. In the carrageenan-induced rat pleurisy model, TFLP efficiently suppressed inflammatory exudate and leukocyte migration, reduced the serum levels of lysozyme (LZM) and malondialdehyde (MDA), increased the activity of serum superoxide dismutase (SOD), and also decreased the contents of total protein, nitric oxide (NO) and prostaglandin E2 (PGE2) in the pleural exudates. No marked effect of TFLP on the activity of serum glutathione peroxidase (GSH-PX) was observed. In the chronic inflammation experiment, TFLP inhibited cotton pellet-induced rat granuloma. The antiinflammatory mechanisms of TFLP are probably associated with the inhibition of prostaglandin formation, influence on the antioxidant systems and the suppression of LZM release.
