Range expansion is a widespread biological process, with well‐described theoretical expectations associated with the colonization of novel ranges. However, comparatively few empirical studies address the genomic outcomes accompanying the genome‐wide consequences associated with the range expansion process, particularly in recent or ongoing expansions. Here, we assess two recent and distinct eastward expansion fronts of a highly mobile carnivore, the coyote (Canis latrans), to investigate patterns of genomic diversity and identify variants that may have been under selection during range expansion. Using a restriction‐associated DNA sequencing (RADseq), we genotyped 394 coyotes at 22,935 SNPs and found that overall population structure corresponded to their 19th century historical range and two distinct populations that expanded during the 20th century. Counter to theoretical expectations for populations to bottleneck during range expansions, we observed minimal evidence for decreased genomic diversity across coyotes sampled along either expansion front, which is likely due to hybridization with other Canis species. Furthermore, we identified 12 SNPs, located either within genes or putative regulatory regions, that were consistently associated with range expansion. Of these 12 genes, three (CACNA1C, ALK, and EPHA6) have putative functions related to dispersal, including habituation to novel environments and spatial learning, consistent with the expectations for traits under selection during range expansion. Although coyote colonization of eastern North America is well‐publicized, this study provides novel insights by identifying genes associated with dispersal capabilities in coyotes on the two eastern expansion fronts. Aim Understanding carnivore distribution is important for management decisions that aim to restore naturally regulated ecosystems and preserve biodiversity. Eastern Wolves, a species at risk in Canada, are centralized in Algonquin Provincial Park and their ability to disperse and establish themselves elsewhere is limited by human-caused mortality associated with hunting, trapping and vehicle collisions. Here, we refine our understanding of Eastern Wolf distribution and provide the first estimates of their effective population size. Location Southern Ontario and Gatineau Quebec. Methods We used non-invasive samples, as well as blood samples archived from other research projects, collected between 2010 and 2014 to generate autosomal microsatellite genotypes at 12 loci for 98 Canis individuals. We utilized Bayesian and multivariate clustering analyses to identify Eastern Wolves in regions that were previously unsampled. Both linkage disequilibrium and temporal approaches were used to estimate effective population size of Eastern Wolves. Results Assignment tests identified 34 individuals as Eastern Wolves, primarily in or near two provincial parks: Killarney and Queen Elizabeth II Wildlands. Eastern Coyotes were identified in Bon Echo Provincial Park, Frontenac Provincial Park and Gatineau Park, whereas many of the samples were admixed among the different Canis types. Effective population size ( Ne) estimates ranged from 24.3 to 122.1 with a harmonic mean of 45.6. Main conclusions The identification of Eastern Wolves in the regions of Killarney and Queen Elizabeth II Wildlands Provincial Parks extends the range of Eastern Wolves north of the French River and southward into previously unidentified regions. The effective population size is low and raises concerns for long-term persistence of this threatened carnivore; estimates are dangerously close to critical values recommended for short-term persistence. Social behaviour can be a vital aspect of population viability when it influences survivorship and reproductive fitness, which is often the case for especially social species, including cetaceans. Understanding this relationship is important in at-risk populations, where knowledge of network structure can provide insight into how they may respond to human impacts., Association patterns in the critically endangered eastern Taiwan Strait (ETS) population of Indo-Pacific humpback dolphins, Sousa chinensis, were investigated using the seasonally collected (2007-2010) photo-identification data of 74 individuals., Social network structure was characterized by high strength and reach, features that are thought to be atypical for the species., Standardized lagged association rates were best fitted by a two-level model: short-term associations that dissipated in less than a day, and long-term associations projected to persist nearly 10 years. These persisting relationships were primarily among calves and their inferred mothers. The short-term associations, which were probably limiting the network's modularity, seemed to occur among more weakly associated individuals temporarily participating in large mother-calf groups., When associations were defined by proximity (within one body-length), relationships involving calves were characterized by a significantly high clustering coefficient, and some calves had high associations with other calves and more than one adult, making maternal relationships ambiguous. This could be evidence for cooperative calf rearing, which may be important for calf survivorship and initiating dyadic relationships among calves., Given their apparent uniqueness among populations of this species, the association patterns observed among the ETS S. chinensis, calves and their inferred mothers in particular, could be a response to conditions (small population size, isolation, confined distribution, and anthropogenic stressors) that make long-term cooperative behaviour advantageous. Local adaptation is necessary for population survival and depends on the interplay between responses to selective forces and demographic processes that introduce or retain adaptive and maladaptive attributes. Host-parasite systems are dynamic, varying in space and time, where both host and parasites must adapt to their ever-changing environment in order to survive. We investigated patterns of local adaptation in raccoon populations with varying temporal exposure to the raccoon rabies virus ( RRV). RRV infects approximately 85% of the population when epizootic and has been presumed to be completely lethal once contracted; however, disease challenge experiments and varying spatial patterns of RRV spread suggest some level of immunity may exist. We first assessed patterns of local adaptation in raccoon populations along the eastern seaboard of North America by contrasting spatial patterns of neutral (microsatellite loci) and functional, major histocompatibility complex ( MHC) genetic diversity and structure. We explored variation of MHC allele frequencies in the light of temporal population exposure to RRV (0-60 years) and specific RRV strains in infected raccoons. Our results revealed high levels of MHC variation (66 DRB exon 2 alleles) and pronounced genetic structure relative to neutral microsatellite loci, indicative of local adaptation. We found a positive association linking MHC genetic diversity and temporal RRV exposure, but no association with susceptibility and resistance to RRV strains. These results have implications for landscape epidemiology studies seeking to predict the spread of RRV and present an example of how population demographics influence the degree to which populations adapt to local selective pressures. Mitochondrial DNA (mtDNA) sequences were analyzed from 106 bowhead whale ( Balaena mysticetus) specimens dating 471 ± 44 14C b.p.-10,290 ± 150 14C b.p. to evaluate whether historical changes in distribution and connectivity were detectable in levels of diversity and population structuring in the Central Canadian Arctic. The species has maintained levels of mtDNA diversity over 10,000 yr comparable to other nonbottlenecked large whale species. When compared to data from the Holocene East Greenland/Spitsbergen and contemporary Bering-Chuckchi-Beaufort populations, differentiation was low ( FST≤ 0.005, ΦST≤ 0.003) and no temporal or geographical genetic structuring was evident. A combination of analyses suggests that the population has expanded over the past 30,000 14C yr. This genetic signature of expansion could result from population growth, admixture of multiple gene pools, or a combination of both scenarios. Despite known climatic change that altered bowhead distribution and led to isolation of populations, there is no detectable population structuring or change in genetic diversity during the Holocene. This may be due to long generation time, occasional population connectivity and a historically large global population. These characteristics warrant caution when interpreting contemporary bowhead whale DNA data, as it is unlikely that any population will be in mutation-drift equilibrium. [ABSTRACT FROM AUTHOR]

Interpretation of the genetic composition and taxonomic history of wolves in the western Great Lakes region (WGLR) of the United States has long been debated and has become more important to their conservation given the recent changes in their status under the Endangered Species Act. Currently, the two competing hypotheses on WGLR wolves are that they resulted from hybridization between (i) grey wolves ( Canis lupus) and western coyotes ( C. latrans) or (ii) between grey wolves and eastern wolves ( C. lycaon). We performed a genetic analysis of sympatric wolves and coyotes from the region to assess the degree of reproductive isolation between them and to clarify the taxonomic status of WGLR wolves. Based on data from maternal, paternal and bi-parental genetic markers, we demonstrate a clear genetic distinction between sympatric wolves and coyotes and conclude that they are reproductively isolated and that wolf-coyote hybridization in the WGLR is uncommon. The data reject the hypothesis that wolves in the WGLR derive from hybridization between grey wolves and western coyotes, and we conclude that the extant WGLR wolf population is derived from hybridization between grey wolves and eastern wolves. Grey-eastern wolf hybrids ( C. lupus × lycaon) comprise a substantial population that extends across Michigan, Wisconsin, Minnesota and western Ontario. These findings have important implications for the conservation and management of wolves in North America, specifically concerning the overestimation of grey wolf numbers in the United States and the need to address policies for hybrids. During the 1990s, North Atlantic right whales had significantly decreased reproduction and showed signs of compromised health, prompting the initiation of noninvasive fecal-based studies to investigate potential causal factors. The interpretation of these studies is enhanced when the defecator is identified, as data can then be linked to individual life history information. Fecal samples ( n= 118) were either collected from single photoidentified whales, associated with several individuals by photoidentification of whales in the vicinity upon sample collection, or were collected when no whales were in the vicinity. Genetic profiles from fecal DNA comprising sex, mitochondrial haplotype, and five microsatellite loci helped assign specific samples to individual right whales based on existing genetic profiles. Profiles were informative in assigning 61 fecal samples to known individuals, 24 of which were collected when no whales were in the vicinity. Whales identified genetically were typically photographed in the same habitat area and on the same day of sample collection ( n= 35/48). Twelve profiles new to the genetic database were identified, suggesting fecal sampling provides a means to obtain genetic profiles from previously unsampled individuals, which may help refine estimates of population size and habitat use patterns if annual fecal sampling continues. The article presents a study which aims at conserving habitat for the endangered Indo-Pacific humpback dolphins (Sousa chinensis) in Eastern Taiwan Strait. The study describes the habitat requirements of the dolphins and discusses the causes of the degradation of the species. It found that the dolphins are exposed to sewage and chemicals, as well as noise and disturbances. It recommends that priority habitat must be available to areas of high dolphin density or intensive dolphin use.

Sampling of feces for genetic studies of wild populations can be problematic because of the low quality and quantity of template DNA obtained. We used cotton swabs in the field to isolate the mucous layer on the surface of fresh wolf (Canis lupus, C. lycaon, and their hybrids) scats followed by immediate preservation, and compared microsatellite genotyping of DNA from these fresh field swabs (FS) to that of previously frozen laboratory swabs (LS). In single polymerase chain reactions (PCRs) of 2 multiplexes, amplification at 8 loci was higher in the FS samples (FS = 50%, LS = 15%; P 5 0.02) because proportion, quantity, and quality of large fragment wolf nuclear DNA from these samples was greater (2.5-25%, 6.25-62.5 ng/swab, 35% amplified at 1,000 base pairs [bp]) than from the LS samples (1.9%-10%, 4.7-25 ng/ swab, 10% amplified at 1,000 bp). Paired blood and fresh field-swabbed samples had identical genotypes. In 84 multiplex PCRs we found no evidence of allelic dropout associated with low template quality or quantity. We conclude that field swabbing of fresh wolf scat facilitates field storage and reduces the need for multiple amplifications at single microsatellite loci, thereby reducing the genotyping costs for wildlife projects that use noninvasive samples. The correlation of landscape features with genetic discontinuities reveals barriers to dispersal that can contribute to understanding present and future spread of wildlife diseases. This knowledge can then be used for targeting control efforts. The impact of natural barriers on raccoon dispersal was assessed through genetic analysis of samples from two regions, Niagara ( N = 666) and St. Lawrence ( N = 802). These areas are transected by major rivers and are at the northern front of a raccoon rabies epizootic. Genetic clusters were identified in each region using Bayesian clustering algorithms. In the Niagara region, two clusters were identified corresponding to either side of the Niagara River. For the St. Lawrence region, spatially congruent clusters were not identified, despite the presence of the intervening St. Lawrence River. These genetic data are consistent with raccoon rabies incidence data where rabies has been detected across the St. Lawrence River in Ontario while no cases have been detected in Ontario across the Niagara River. This is despite expectations of rabies incidence in Niagara before the St. Lawrence based on the progression of rabies from New York. The results from the two regions suggest different permeabilities to raccoons between New York and Ontario that may be attributed to the rivers. However, other factors have also been explored that could contribute to this difference between these study sites including the shape of the landscape and resource distribution. We have isolated and characterized 17 tetranucleotide microsatellite loci for Blanchard's cricket frog ( Acris crepitans blanchardi), an anuran common in the central USA. Sixteen loci were organized into four multiplex amplification reactions. These loci were highly polymorphic when screened in 55 individuals from two distant populations, with 11–48 alleles per locus (average = 24.8). Observed and expected heterozygosities ranged from 0.18 to 0.97 and from 0.17 to 0.96, respectively. Nine loci were also polymorphic in Acris crepitans crepitans, with seven polymorphic in Acris gryllus. Five loci amplified in all three taxa. AbstractIn order to assess levels of major histocompatibility complex (Mhc) variation within the St Lawrence beluga (Delphinapterus leucas) the variation at the beluga Mhc DRB1 class II locus was assessed by single-strand conformation polymorphism (SSCP) analysis of the peptide-binding region for 313 whales collected from 13 sampling locations across North America. In addition, samples from west Greenland and the St Lawrence were also typed at the DQB locus, allowing comparison to a previous study and assessment of linkage disequilibrium of alleles at the two loci. Comparisons of DRB1 and DQB allele frequencies among all sampling locations indicated genetic structure (α < 0.005). Most of this structure resulted from differences between the different wintering groups. Significant genetic structure (α = 0.05) exists among each pair of the following groups at both the DRB1 and DQB loci; St Lawrence, Hudson Strait, Bering Sea, Cunningham Inlet, and Davis Strait (minus Cunningham Inlet), except the St Lawrence and Hudson Strait for the DQB locus. In the St Lawrence population, six of the eight DRB1 alleles are present representing all five known allelic lineages. Evidence of linkage disequilibrium between the DRB1 and DQB is present in two sampling locations, the St Lawrence and Nuussuaq (α = 0.05). Analysis of probable DRB1–DQB haplotypes among groups of beluga suggests a haplotype reduction in the St Lawrence. [ABSTRACT FROM AUTHOR]

In this study we report the first documentation of mating between harp (Phoca groenlandica) and hooded seals (Cystophora cristata), The production of this hybrid was quite unusual, being the result of a cross between parents of different genera which are morphologically dissimilar and have quite different mating behavior and dramatically different body sizes. The molecular techniques (mtDNA and macrosatellite nuclear DNA banding patterns) used in this study will undoubtedly be applied widely to many different taxa in the near future, allowingus to re-examine many suspected cases of hybridization among marinemammals and, in a larger context, the meaning of the species concept.
