Objective: To enhance the accuracy in predicting lymph node metastasis (LNM) preoperatively in patients with papillary thyroid microcarcinoma (PTMC), refining the "low‐risk" classification for tailored treatment strategies. Methods: This study involves the development and validation of a predictive model using a cohort of 1004 patients with PTMC undergoing thyroidectomy along with central neck dissection. The data was divided into a training cohort (n = 702) and a validation cohort (n = 302). Multivariate logistic regression identified independent LNM predictors in PTMC, leading to the construction of a predictive nomogram model. The model's performance was assessed through ROC analysis, calibration curve analysis, and decision curve analysis. Results: Identified LNM predictors in PTMC included age, tumor maximum diameter, nodule‐capsule distance, capsular contact length, bilateral suspicious lesions, absence of the lymphatic hilum, microcalcification, and sex. Especially, tumors larger than 7 mm, nodules closer to the capsule (less than 3 mm), and longer capsular contact lengths (more than 1 mm) showed higher LNM rates. The model exhibited AUCs of 0.733 and 0.771 in the training and validation cohorts respectively, alongside superior calibration and clinical utility. Conclusion: This study proposes and substantiates a preoperative predictive model for LNM in patients with PTMC, honing the precision of "low‐risk" categorization. This model furnishes clinicians with an invaluable tool for individualized treatment approach, ensuring better management of patients who might be proposed observation or ablative options in the absence of such predictive information. Sedimentary successions of the Frasnian–Tournaisian (Late Devonian to Earliest Carboniferous) and their spatial distributions in the Hainan Island, South China are critically reviewed based on updated information of the previously published data and newly obtained fossils. The Upper Devonian strata are assigned to the Frasnian Lanyang Formation and Famennian Changjiang Formation, which are distributed in north‐western Hainan Island. The Lanyang Formation is represented by limestone, siltstone, slate, and intercalated with tuff, and yields tentaculites: Polycylindrites sp., stromatoporoids, and tabulate corals. The Changjiang Formation yields abundant conodonts Palmatolepis gracilis sigmoidalis and Polynodosus germanus, typical of Famennian assemblages, and the associated faunas include corals, radiolarians, foraminifers, and calcareous algae of Famennian age. Of these, corals are assignable to the upper Famennian Cystophrentis Zone of South China. The strata of the Tournaisian Jishi Formation are categorized into two types, typically exposed in the Shilu and Jinbo areas, respectively, both of which are located north to the Gancheng‐Wanning Fault. The lower Jishi Formation yields brachiopods Unispirifer (Unispirifer) extensus, Unispirifer sp., Schuchertella magna, and gastropods. The middle Jishi Formation yields conodonts: Siphonodella isosticha and S. levis, brachiopods: Unispirifer (Unispirifer) forbesi, Atylephorus tornacensis, coral: Pseudouralinia irregularis, trilobite: Weberiphillipsia linguiformis, ostracods, bryozoans, and foraminifers. The conodonts are assignable to the zonation equivalent to the Siphonodella sulcata Zone, implying the Devonian–Carboniferous boundary is placed at the base of the middle part of the formation. The upper part of the Jishi Formation yields abundant brachiopods characterized by Mesochorispira subgrandis, Atylephorus tornacensis, and Marginatia huaqiaoensis. The presence of corals Cystophrentis, Pseudouralinia, Zaphrentites, and Kueichowpora, and trilobite Hunanoproetus, endemic to South China from the Changjiang and Jishi formations implies that the Hainan Island was palaeobiogeographically close to the South China Block of the Tethyan Realm, rather than of Gondwana affinity during the Late Devonian and Tournaisian. Identification of Xinanosprifer flabellum and Homotoma sp., typical of early Silurian from the 'Nanhao Formation' previously assigned to the Tournaisian suggests that no Carboniferous strata are distributed in southern Hainan Island and the area south of the Gancheng‐Wanning Fault. Introduction: To assess the clinical performance and correlations of automated chemiluminescence assay (CIA) and enzyme‐linked immunosorbent assay (ELISA) for detecting antiphospholipid (aPL) antibodies in the diagnosis of antiphospholipid syndrome (APS). Methods: The study recruited 505 subjects, including 192 with APS, 193 with connective tissue diseases other than APS, and 120 healthy donors. We measured anticardiolipin (aCL) and anti‐β2‐glycoprotein I (anti‐β2GPI) antibodies IgG, IgM, and IgA in all the samples using both CIA and ELISA. Results: Total agreement between the two methods ranged from 83.50% for anti‐β2GPI IgG antibodies to 92.76% for anti‐β2GPI IgM antibodies in all the groups. Anti‐β2GPI and aCL IgG assays showed the highest Spearman's rho coefficients (anti‐β2GPI IgG = 0.742, aCL IgG = 0.715). Anti‐β2GPI IgG CIA showed the highest sensitivity for diagnosis of APS at 80.21%, which was significantly higher than the sensitivity of anti‐β2GPI IgG ELISA (52.08%). For diagnosis of APS, anti‐β2GPI IgG CIA had the best discrimination power with the area under the curves (AUC) of 0.922, followed by aCL IgG CIA (AUC of 0.905). While the CIA AUC was slightly higher in all cases, the difference was not statistically significant. Conclusion: CIA measurements had a good agreement and correlation with comparative ELISA assays. The CIA anti‐β2GPI IgG however was significantly more sensitive for APS diagnosis. Summary: Oestrogens play an important role in the development and progression of papillary thyroid carcinoma (PTC) through oestrogen receptor (ER)‐α and ‐β, which may exert different or even opposing actions in PTC. The roles of ERβ in ERα‐negative PTC are still not clear. This study investigated the expression dynamics of ERβ1 (wild‐type ERβ) and its clinical significance in female ERα‐negative PTC patients. ERβ1 expression was detected in thyroid tissues of 136 female patients diagnosed with PTC. The relationships between ERβ1 expression and clinicopathological/biological factors were also analysed in female ERα‐negative PTC patients. The total score for ERβ1 was significantly lower in female ERα‐negative PTC patients with LNM or ETE when compared to those without LNM or ETE (Z = −2.923, P = 0.003 and Z = −3.441, P = 0.001). Accordingly, the total score for ERβ1 was significantly higher in ERα‐negative PTC patients expressing E‐cadherin compared to patients negative for E‐cadherin expression (Z = −2.636, P = 0.008). The total score was lower in ERα‐negative PTC patients positive for VEGF expression compared to those negative for VEGF expression (Z = −1.914, P = 0.056). This preliminary study indicates that reduced expression of ERβ1 in female ERα‐negative PTC patients is associated with greater progression of the disease. Objective This study aimed to investigate the effects of FTY720 on inducing cell growth inhibition and enhancing the cytotoxicity of anti-cancer drugs in the human colon cancer cell line HCT-8 and its multidrug-resistant cell line HCT-8/5-fluorouracil ( HCT-8/5- Fu). Methods Cell viability and apoptosis after being treated with FTY720 alone or in combination with doxorubicin ( DOX) and etoposide ( VP16) were tested in HCT-8 and HCT-8/5- Fu cells. The changes in P-glycoprotein ( P-gp) and multidrug resistance protein 1 ( MRP1) were determined at the mRNA and functional levels. Results FTY720 showed anti-proliferative activity against cancer cells in a dose-dependent and time-dependent manner and could enhance the cytotoxicity of DOX and VP16 in both HCT-8 and HCT-8/5- Fu cell lines. In addition, treatment with FTY720 resulted in the promotion of VP16-induced cell apoptosis and an increased accumulation of intracellular DOX and two specific fluorescent substrates of P-gp and MRP1 through the inhibition of efflux and the suppression of gene expression. Conclusion FTY720 exerts its chemosensitization effect in HCT-8 and HCT-8/5- Fu cell lines by promoting cell apoptosis and inhibiting P-gp and MRP1, which could be applied as a potential co-adjuvant therapeutic modality. OBJECTIVE: In previous studies cellular prion protein (PrPc) is confirmed to be involved in multidrug resistance (MDR) of gastric cancer. Although octarepeat peptides are important functional domains of PrPc and are closely related to the transport of Cu2+/Zn2+ and antioxidative function, the significance in MDR remains unknown. We aimed to investigate the role of octarepeat peptides in gastric cancer MDR. METHODS: Small interfering RNA (siRNA) against PrPc were transfected into adriamycin-resistant gastric cancer cell lines to inhibit the expression of wild type PrPc, and then constructs encoding PrPc without octarepeat peptides and PrPc without the fifth repeat peptide were transfected, respectively, to establish the cell models. In vitro drug sensitivity, cell apoptosis, measurement of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and glutathione (GSH), as well as changes in glutathione S-transferase (GST) were detected. RESULTS: In vitro drug sensitivity test showed that octarepeat peptides could modulate the drug resistance of gastric cancer cells, but the deletion of the fifth repeat peptide had no effect. Specifically, the anti-apoptotic capacity of gastric cancer cells decreased significantly when the octarepeat peptides of PrPc was absent. Moreover, the activities of total SOD, Cu2+/Zn2+-SOD, GSH-Px, GSH, and GST detected in different stressing periods revealed that cells lacking octarepeat peptides of PrPc exhibited weakened responses to stress. However, absence of the fifth repeat peptide did not exert any effect on stress response. Abstract: We have elucidated the cytoprotective effect of annphenone (2,4-dihyroxy-6-methoxy-acetophenone 4-O-β-d-glucopyranoside) against oxidative stress-induced apoptosis. Annphenone scavenged intracellular reactive oxygen species (ROS) and increased antioxidant enzyme activities. It thereby prevented lipid peroxidation and DNA damage, which was demonstrated by the inhibition of the formation of thiobarbituric acid reactive substance (TBARS), inhibition of the comet tail and decreased phospho-H2A.X expression. Annphenone protected Chinese hamster lung fibroblast (V79-4) cells from cell death via the inhibition of apoptosis induced by hydrogen peroxide (H2O2), as shown by decreased apoptotic nuclear fragmentation, decreased sub-G1 cell population and inhibited mitochondrial membrane potential (Δψ) loss. Taken together, these findings suggest that annphenone exhibits antioxidant properties by inhibiting ROS generation and thus protecting cells from H2O2-induced cell damage. [Copyright &y& Elsevier]

The crystal structure of the title complex, [Cu(C7H8N4)2(H2O)2](ClO4)2, consists of a discrete centrosymmetric [Cu(C7H8N4)2(H2O)2]2+ cation and two perchlorate anions. The CuII centre is six-coordinated by four N donors from the two pyrazole rings [Cu-N 1.998 (2) and 2.032 (3) Å] and two O atoms from the water mol­ecules occupying the apical sites [Cu-O 2.459 (3) Å]. The coordination geometry of the complex can be described as octahedral. There is a unique three-dimensional network in which the perchlorate units are linked by a combination of strong O-H⃛O and weak C-H⃛O hydrogen bonds. [ABSTRACT FROM AUTHOR]

To develop an HCT-8/5-FUmultidrug-resistant colorectal cancer cell line and to elucidate the effect of Andrographolid (AG), an extract from Andrographis paniculate, a medicinal herb on the HCT-8/5-FUmultidrug-resistant colorectal cancer cell line.An HCT-8 colorectal cancer cell line was used and a high concentration of 5-Fluorouracid (5-FU) was introduced at the beginning to induce drug resistance, then the concentration of 5-FU was increased in gradients. Approximately 7 months later, the cells grew stably in 2.0 µg/mL of 5-FU, and the cell line was named HCT-8/5-FUmultidrug-resistant colorectal cancer cell line. The resistant index of HCT-8/5-FUcells to 5-FU, adriamycin (ADM), cisplatin (DDP) was checked by MTT test, and a growth curve was drawn. The morphological changes were observed by both light and electron microscope. The function ofP-170 was detected by rhodamine staining. After the application of AG and co-administration of 5-FU, ADM and DDP, the growth curves and inhibition rate as well as apoptosis rate of HCT-8/5-FUat different concentrations of AG were evaluated by MTT and flow cytometry. Rhodamine staining was used to investigate the possible mechanism involved by AG.The resistance index of HCT-8/5-FUto 5-FU was 16.6, and a cross-resistance to ADM and DDP was noticed. Compared with parental cells, HCT-8/5-FUcell's growth rate did not change significantly but the cell's morphology was remarkably changed as compared with parental cells. Overexpression ofP-170 by HCT-8/5-FUcell was indicated through rhodamine staining. AG at a low concentration showed weak inhibitory effect on HCT-8/5-FU. However, a remarkable inhibitory and apoptosis rate was shown when AG was co-administered with 5-FU, ADM and DDP, respectively. Interestingly AG alone could not induce apoptosis and change the cell cycles. AG might affect the expression of P-170, which was indicated by rhodamine staining.The HCT-8/5-FUmultidrug-resistant colorectal cancer cell line has been successfully developed and because it has cross-resistance to 5-FU, ADM and DDP, it might serve as an ideal multidrug resistance (MDR) model for colorectal cancer research. The mechanism of HCT-8/5-FUresistance to chemotherapeutic agents might be related to the overexpression ofP-170. Low concentrations of AG alone have no significant inhibition on HCT-8/5-FUand fail to induce apoptosis and to change cell cycles. AG might act as a chemosensitizer when co-administered with 5-FU, ADM and DDP, and the mechanism of reversal modulation of multidrug resistance by AG in the HCT-8/5-FUresistant cell line might be related to its downregulation of overexpression ofP-170. The title compound, [Cu(2)(C(11)H(8)NO(3))(2)Cl(2)], is a bicopper(II) complex. Each Cu(II) ion is five-coordinated by two O atoms and one N atom from the (8-quinol-yloxy)acetate ligand, and by two μ(2)-chloride ligands, thus exhibiting a distorted square-pyramidal CuCl(2)NO(2) coordination environment. Each (8-quinol-yloxy)acetate anion acts as a tridentate chelating ligand. In the crystal structure, adjacent quinolyl rings are involved in strong π-π stacking inter-actions, with inter-planar distances of 3.549 (5) and 3.763 (5) Å, thereby forming a two-dimensional planar network perpendicular to the ab plane. Furthermore, a weak inter-action [2.750 (4) Å] is observed within these planes between one Cu(II) ion and a carboxyl-ate O atom from a ligand in an adjacent mol-ecule, which also contributes to the stability of the structure.
