2. Transhiatal bilateral cervical approach for mediastinoscopy‐assisted esophagectomy: A retrospective cohort study.

Background: The McKeown minimally invasive esophagectomy (McMIE) procedure has various limitations, including surgical contraindications and a high rate of postoperative pulmonary complications. A novel mediastinoscopic esophagectomy procedure was described in this study by using esophageal invagination and a transhiatal and bilateral cervical approach (EITHBC). Methods: According to the mode of operation, a total of 259 patients were divided into two groups, among which 106 underwent EITHBC and 153 underwent McMIE. The number of lymph nodes dissected, intraoperative outcomes, and postoperative outcomes were compared between the two groups of patients. Results: The results revealed that the average number of resected lymph node in the EITHBC group was significantly higher in the recL106 and TbL106 stations (recL106: 1.75 vs. 1.51, p = 0.016, TbL106: 1.53 vs. 1.19, p = 0.016) and significantly lower in the 107 stations (1. 74 vs. 2. 07, p < 0.001) than in the McMIE group. The intraoperative blood loss in the EITHBC group was significantly lower than that in the McMIE group (63.30 vs. 80.45 mL, p < 0.001). The incidence of postoperative pulmonary complications in the EITHBC group was lower than that in the McMIE group (14.15% vs. 27.45%, p = 0.008). The incidence of recurrent laryngeal nerve paralysis in the EITHBC group was significantly higher than that in the McMIE group (26.41% vs. 10.46%, p = 0.003). Conclusion: Compared with the McMIE procedure, the EITHBC procedure has advantages in terms of removing the upper mediastinal lymph nodes and reducing postoperative pulmonary complications. This article discusses the generation of an induced pluripotent stem cell (iPSC) line carrying a specific genetic mutation associated with cardiac syndrome. The mutation, known as PRKAG2 R302Q, is responsible for a rare autosomal dominant genetic heart disease characterized by glycogen accumulation and ventricular septal hypertrophy. The study successfully differentiated the iPSCs into cardiomyocytes that exhibited beating behavior and expressed specific cardiac genes. This research provides a valuable model for understanding the underlying mechanisms of the disease and exploring potential therapeutic strategies. 4. Learning‐based encoded target detection on iteratively orthorectified images for accurate fisheye calibration.

Fisheye camera calibration is an essential task in photogrammetry. However, previous calibration patterns and the robustness of the adjoint processing methods are limited due to the fisheye distortion and various lighting. This problem leads to additional manual intervention in the data collection. Moreover, it is arduous to accurately detect the board target under fisheye's distortion. To increase the robustness in this task, we present a novel encoded board "Meta‐Board" and a learning‐based target detection method. Additionally, an automatic image orthorectification is integrated to alleviate the distortion effect on the target iteratively until convergence. A low‐cost control field with the proposed boards is built for the experiment. Results on both virtual and real camera lenses and multi‐camera rigs show that our method can be robustly used in calibrating the fisheye camera and reaches state‐of‐the‐art accuracy. 5. View‐graph key‐subset extraction for efficient and robust structure from motion.

Structure from motion (SfM) is used to recover camera poses and the sparse structure of real scenes from multiview images. SfM methods construct a view‐graph from the matching relationships of images. Redundancy and incorrect edges are usually observed in it. Redundancy inhibits the efficiency and incorrect edges result in the misalignment of structures. In addition, the uneven distribution of vertices usually affects the global accuracy. To address these problems, we propose a coarse‐to‐fine approach in which the poses of an extracted key‐subset of images are first computed and then all remaining images are oriented. The core of this approach is view‐graph key‐subset extraction, which not only prunes redundant data and incorrect edges but also obtains properly distributed key‐subset vertices. The extraction approach is based on a replaceability score and an iteration‐update strategy. In this way, only vertices with high SfM importance are preserved in the key‐subset. Different public datasets are used to evaluate our approach. Due to the absence of ground‐truth camera poses in large‐scale datasets, we present new datasets with accurate camera poses and point clouds. 6. CCL3‐CCR5 axis promotes cell migration and invasion of colon adenocarcinoma via Akt signaling pathway.

Background: Infiltration of tumor‐associated macrophages (TAMs) can promote tumorigenesis and development. C‐C motif chemokine ligand 3 (CCL3) was reported to be derived from TAMs and tumor cells and facilitate the progression of several cancers. Nevertheless, whether CCL3 can be derived from TAMs and tumor cells of colon adenocarcinoma (COAD) is unclarified. Methods: Peripheral blood monocytes‐derived macrophages were polarized by the conditioned medium from COAD cells to establish TAM‐like macrophages (TAM1/2). RT‐qPCR and western blotting were used for detection of expression levels of CCL3 and its receptors C‐C motif chemokine receptor 1 (CCR1) and CCR5 in TAM1/2 and COAD cells. Immunofluorescence staining was utilized for evaluating CCL3, CD163 and CCR5 expression. The Akt signaling pathway‐associated protein levels were measured by western blotting. Transwell assays were used for assessing cell migration and invasiveness. Results: CCL3 displayed a high level in TAMs and cancer cells of COAD. CCL3 activated the Akt signaling pathway by binding to CCR5. CCL3‐CCR5 axis facilitated COAD cell migration and invasiveness by activating the Akt signaling. CCL3 derived from both TAMs and cancer cells contributed to the malignant behaviors of COAD cells. High expression of CCL3/CCR5 was closely associated with poor prognoses of COAD patients. Conclusion: CCL3‐CCR5 interaction promotes cell migration and invasiveness, and functions as a prognostic biomarker for COAD. [ABSTRACT FROM AUTHOR]

Haitaite‐(La), (La, Ce)(U4+, U6+, Fe2+)(Fe3+, Al)2(Ti, Fe2+, Fe3+)18O38, is a new member of the crichtonite group. It is named after the Haita Village in the Miyi County of Sichuan Province, China, where the mineral was discovered. The mineral occurs as black opaque centimeter‐sized aggregates in the external contact zone between the Neoproterozoic (∼800 Ma) alkali feldspar granite and the Mesoproterozoic (∼1700 Ma) micaschist. In the studied sample, haitaite‐(La) is associated with other minerals, including ilmenite, magnetite, rutile, zircon, brannerite and uraninite. The new mineral is a black, metallic phase and has a Mohs hardness of 6, with a density of 4.99 g/cm3 (calculated) and 5.03 g/cm3 (measured). Haitaite‐(La) is opaque in transmitted light and grayish‐white under reflected light, with a reflectivity between 22.5% and 16.42% in the 400–700 nm band (SiC, in the air). The compositions of the mineral were measured by EPMA, the U4+/U6+ ratio was determined by X‐ray photoelectron spectroscopy and the Fe2+/Fe3+ ratio was determined by Mössbauer spectroscopy. Haitaite‐(La) is trigonal, belongs to and has unit‐cell parameters a = 10.3678(5) Å, c = 20.8390(11) Å, V = 1939.9(2) Å3, Z = 3. The crystalline structure is composed of octahedra with 9 layers of close‐packed octahedra (M1, M3, M4, M5), tetrahedra (M2) and contains large 12‐coordinated M0 sites. [ABSTRACT FROM AUTHOR]

9. Effect of Preheating on the Microstructure, Impact Properties, and Fracture Mechanisms of a Laser Surface‐Hardened High‐Carbon–Chromium Steel.

To optimize the toughness and hardness of laser surface‐hardened high‐carbon–chromium (52100) steel, the effect of preheating on microstructure evolution and impact property is studied, and the impact fracture mechanism is analyzed. The results indicate that preheating deepens the hardened depth, facilitating grain coarsening and cementite dissolution near the surface. Regardless of preheating, the size distribution of cementite follows a typical lognormal distribution. Laser surface hardening leads to a decrease in impact absorbed energy. Preheating at 160 °C increases the impact absorbed energy by ≈25%, at which point the maximum hardness is still as high as 860 HV0.3. The impact absorbed energy of spheroidized microstructure tends to be larger before laser hardening and reduces after laser hardening. The impact toughness is the combined result of the brittleness of the hardened layer, residual stress, and substrate microstructure. Under the impact load, microcracks nucleate near the surface of the hardened layer and propagate in the form of intergranular fracture and then propagate in the form of transgranular fracture toward the inside of the hardening layer and substrate. Herein, a novelty research work on optimizing the impact properties during laser hardening through preheating and adjusting initial microstructure is provided. [ABSTRACT FROM AUTHOR]

Monochiral single‐walled carbon nanotubes (SWCNTs) are promising materials with potential applications in 3D integrated circuits and optoelectronic hybrid circuits. However, the purity and device performance of monochiral SWCNTs are still far lower than expected. Here, the authors demonstrate that specific monochiral SWCNTs can be wrapped by conjugated polymers containing pyridine units, and the supramolecular assemblies show surprising suspension stability even after high‐intensity ultracentrifugation. Additionally, two novel methods are developed, namely, enhanced ultracentrifugation (E‐UCG) and stepwise extraction processing (STEP), which successfully achieve isolation of (10,8) and (12,5) SWCNTs with respective diameters of 1.24 and 1.2 nm at high monochiral purity (92.3% and 95.6%). Their S11 absorption and fluorescence emission peaks are both at ≈1.5 µm (optical telecommunications C‐band). Both micro‐ and nanoscale field‐effect transistor (FET) devices can be fabricated from the as‐isolated (10,8) SWCNTs, and these FETs exhibit excellent electrical performance and a high semiconducting purity of up to 99.94%. Long noncoding RNAs (lncRNAs) exert crucial effects on the development of many malignancies, including gastric cancer. Herein, we investigated the role of lncRNA noncatalytic region of tyrosine kinase adaptor protein 1 (NCK1) divergent transcript (NCK1‐DT, also known as NCK1‐AS1) in gastric cancer. Reverse transcription quantitative polymerase chain reaction demonstrated that NCK1‐AS1 exhibited high expression in gastric cancer tissues and cells. In vitro assays including MTT, colony formation, Transwell, wound healing and sphere formation assays indicated that NCK1‐AS1 depletion inhibited cell proliferation, migration, invasion and stemness maintenance. Luciferase reporter and RIP assays suggested that NCK1‐AS1 functioned as a competitive endogenous RNA (ceRNA) for miR‐22‐3p to positively modulate BCL9 expression. BCL9 was a target gene of miR‐22‐3p. According to western blot analysis and TOP/FOP flash assay, NCK1‐AS1 activated the Wnt/β‐catenin signaling via the miR‐22‐3p/BCL9 axis. Furthermore, rescue experiments verified that NCK1‐AS1 affected cellular processes by activating the Wnt/β‐catenin signaling pathway via the miR‐22‐3p/BCL9 axis. Tumor xenograft model validated that NCK1‐AS1 promoted tumor growth in vivo via the Wnt/β‐catenin signaling by upregulating BCL9 expression. Overall, NCK1‐AS1 functions as an oncogene and promotes gastric cancer progression via the miR‐22‐3p/BCL9‐Wnt/β‐catenin signaling pathway. [ABSTRACT FROM AUTHOR]

This protocol enables to get quinoline, quinoxaline, quinazolin‐4(3H)‐one and benzo[d]thiazole attached 2,4,5‐trisubstituted oxazoles from methyl azaarenes, benzoins, and NH4OAc in the presence of iodine and molecular sieves without any metal catalyst. A general method for the catalytic asymmetric synthesis of 4‐alkyl‐4H‐chromenes was developed. With readily available β‐alkyl‐substituted enones and 2‐hydroxylated arylboronic acids, a rhodium‐catalysed asymmetric conjugate addition/intramolecular hemi‐acetalization/acid‐promoted dehydration sequence leads to the formation of 4‐alkyl‐4H‐chromenes in up to 99% yield and with up to >99% ee. 15. Effect of acaricidal compound extracted from Arachis hypogaea Linn against Tetranychus cinnabarinus.

Six plant crude extracts were chosen to evaluate their acaricidal activity against Tetranychus cinnabarinus adults. The crude extract from the stems and leaves of Arachis hypogaea L. presented the highest activity in leaf‐dip bioassays, with an LC50 value of 3,545.98 mg/L. Further extraction using four different solvents (petroleum ether, ethyl acetate, n‐butyl alcohol and distilled water) demonstrated that the active components mainly existed in the petroleum ether phase. Then, the active compound, palmitic acid, was isolated from the petroleum ether phase via two‐step column chromatography using columns filled with silica gel and C‐18 and identified via mass spectrometry and nuclear magnetic resonance analyses. The LC50 value of active palmitic acid against T. cinnabarinus was 534.58 mg/L. The present study demonstrated that the active compound extracted from A. hypogaea is a potential novel botanical acaricide for controlling T. cinnabarinus. [ABSTRACT FROM AUTHOR]

This paper describes the recent National Institute of Standards and Technology (NIST) work on incorporating an optical clock into a time scale. We simulate a time scale composed of continuously operating commercial hydrogen masers and an optical frequency standard that does not operate continuously as a clock. The simulations indicate that to achieve the same performance of a continuously operating Cs‐fountain time scale, it is necessary to run an optical frequency standard 12 minutes per half a day, or 1 hour per day, or 4 hours per 2.33 day, or 12 hours per week. Following the simulations, a Yb optical clock at NIST was frequently operated during the periods of 2017 March – April and 2017 late October – late December. During this operation, comb‐mediated measurements between the Yb clock and a hydrogen maser had durations ranging from a few minutes to a few hours, depending on the experimental arrangements. 17. Biological role and clinical value of miR‐99a‐5p in head and neck squamous cell carcinoma (HNSCC): A bioinformatics‐based study.

MicroRNAs (miRNAs) are confirmed to be tumor promoters or suppressors in multiple squamous cell carcinomas (SCCs). miR‐99a‐5p has been demonstrated to be downregulated in cancerous tissues, but its functional role in head and neck SCC (HNSCC) and its mechanism of action have not been fully elucidated. Here, we studied the expression of miR‐99a‐5p in HNSCC and performed a clinical value assessment and then extracted mature expression data from The Cancer Genome Atlas (TCGA) and microarrays from Gene Expression Omnibus (GEO). Furthermore, biological analysis was constructed via online prediction tools. The results revealed that miR‐99a‐5p expression was markedly lower in HNSCC tissues than in normal tissues, which also showed significance in the prognosis of HNSCC. However, its diagnostic value could not be verified due to the lack of body fluid samples. Additionally, miR‐99a‐5p was expressed at higher levels in patients with low histological grade neoplasms than those with high histological grade neoplasms. The age of the patient might also be a possible clinical parameter affecting miR‐99a‐5p expression. Furthermore, miR‐99a‐5p significantly influenced HNSCC progression by regulating the PI3K‐Akt signaling pathway, in which the key target genes were upregulated in 519 HNSCC tissues compared to 44 normal tissues, as determined by the Gene Expression Profiling Interactive Analysis (GEPIA). In conclusion, our study may provide insights into the expression and mechanism of miR‐99a‐5p in HNSCC. Further studies are required to elucidate the role of miR‐99a‐5p and its potential clinical applications for HNSCC. [ABSTRACT FROM AUTHOR]

Abstract: Philadelphia chromosome (Ph)/BCR‐ABL‐positive (ph+) ALL is the most common genetic abnormality associated with ALL and has been shown to confer the worst prognosis to both children and adults. Increasing evidence has revealed that the dysregulation of prolyl isomerase Pin 1 contributes to multicancer development and progression, including ALL, although the underlying molecular mechanisms remain unclear. Here, we report that the expression of Pin 1 was enhanced in ph+ ALL patient samples and was associated positively with the expression of BCR‐ABL. Genetically or pharmacologically inhibiting Pin 1 expression or activity produces potent therapeutic efficacy against ph+ ALL. We further demonstrated that BCR‐ABL enhances the prolyl isomerase activity of Pin 1 by decreasing the phosphorylated level of Pin 1 at Ser 71 and interacting with DAPK1. The inhibition of BCR‐ABL activity by imatinib in human ph+ ALL cells reduces the prolyl isomerase activity of Pin 1, further suggesting a key role of the newly identified BCR‐ABL‐Pin 1 axis in ph+ ALL progression. Thus, the combined suppression of Pin 1 and BCR‐ABL proteins may be exploited as an additional target therapy for ph+ ALL. [ABSTRACT FROM AUTHOR]

Sargassum polycystum is used as a model to identify putative marine glacial refugia and explore biogeographical patterns driven by climate change in the late Pleistocene ice ages. Location: South‐East Asia. Methods: Mitochondrial cox1 and cox3 and nuclear internal transcribed spacer‐2 (ITS2) were obtained from 310, 325 and 313 individuals ofS. polycystum (23 localities), respectively. Phylogenetic trees (maximum likelihood and Bayesian inference) and haplotype/ribotype networks were constructed to elucidate phylogeographical patterns. Analysis of molecular variance (AMOVA), neutrality tests (Tajima'sD and Fu & Li'sD* ), current (θπ ) and historical (θw ) genetic diversities and extended Bayesian skyline plots (EBSP) were used to estimate historical demography. Results: The populations from the south‐west of Hainan Island harboured much higher genetic diversity and unique endemism in comparison with other populations in the distribution range.Sargassum polycystum experienced relatively long‐term stable population size followed by a continued period of demographic expansion in the late Pleistocene. Main conclusions: Our phylogeographical evidence revealed the existence of a previously unidentified marine refugium specific toS. polycystum in the south‐west of Hainan Island, China (the Central Depression of the Yinggehai Basin), along with a possible secondary refugium around the Bali Island, Indonesia. These biogeographical findings provide important insights regarding speciation, adaptation and evolution of marine organisms in South‐East Asia and the conservation of unique biodiversity under climate change. Abstract: Providing scientific explanations for natural phenomena is a fundamental aim of science; therefore, scientific explanation has been selected as one of the key practices in science education policy documents around the world. To further elaborate on existing educational frameworks of scientific explanation in K–12, we propose a learning progression of scientific explanation based on the Phenomenon‐Theory‐Data‐Reasoning (PTDR) framework. To examine the validation of the PTDR framework for scientific explanation and the learning progression based on it, we developed the
Scientific Explanation Progression Assessment to investigate students’ performance (Grades 8–12 students,N = 4,554) on scientific explanations. A Rasch analysis and other statistical tests together indicated the validity of the PTDR framework for scientific explanation and the learning progression derived from this framework. Carbenoxolone ( CBX) is a clinically prescribed drug for the treatment of digestive ulcer and inflammation. It is also a widely used pharmacological inhibitor of several channels in basic research. Given that the overactivity of several channels, including those inhibitable by CBX, underlies bladder dysfunction, we tested the potential therapeutic application and mechanism of CBX in the treatment of voiding dysfunction. In a mouse model of cystitis induced by cyclophosphamide ( CYP), CBX administration prevented the CYP-elicited increase in bladder weight, oedema, haemorrhage, and urothelial injury. CBX also greatly improved micturition pattern, as manifested by the apparently decreased micturition frequency and increased micturition volume. Western blot results showed that CBX suppressed CYP-induced increase in protein carbonyls, COX-2, and iNOS. Further analysis using cultured urothelial cells revealed that acrolein, the major metabolite of CYP, caused protein oxidation, p38 activation, and urothelial injury. These effects of acrolein were reproduced by TRPV4 agonists and significantly prevented by antioxidant NAC, p38 inhibitor SB203580, TRPV4 antagonist RN-1734, and CBX. Further studies showed that CBX potently suppressed TRPV4 agonist-initiated calcium influx and subsequent cell injury. CBX attenuated CYP-induced cystitis in vivo and reduced acrolein-induced cell injury in vitro, through mechanisms involving inhibition of TRPV4 channels and attenuation of the channel-mediated oxidative stress. Understanding the evolutionary processes that have created diversity and the genetic potential of species to adapt to environmental change is an important premise for biodiversity conservation. Herein, we used mitochondrial trnW-L and cox3 and plastid rbcL-S data sets to analyze population genetic variation and phylogeographic history of the brown alga Sargassum fusiforme, whose natural resource has been largely exterminated in the Asia-Northwest Pacific in the past decades. Phylogenetic trees and network analysis consistently revealed three major haplotype groups (A, B, and C) in S. fusiforme, with A and B distributed in the Japan-Pacific coast. Group C consisted of three subgroups (C1, C2, and C3) which were distributed in the Sea of Japan, the Yellow-Bohai Sea, and East China Sea, respectively. Isolation-with-migration ( IM a) analysis revealed that the three groups diverged approximately during the mid-Pleistocene ( c. 756-1,224 ka). Extended Bayesian skyline plots ( EBSP) showed that groups A and B underwent relatively long-term stable population size despite a subsequent rapid demographic expansion, while subgroups C2 and C3 underwent a sudden expansion at c. 260 ka. F
ST and AMOVA detected low population-level genetic variation and high degrees of divergence between groups. The cryptic diversity and phylogeographic patterns found in S. fusiforme not only are essential to understand how environmental shifts and evolutionary processes shaped diversity and distribution of coastal seaweeds but also provide additional insights for conserving and managing seaweed resources and facilitate predictions of their responses to future climate change and habitat loss. Objective This randomized, double-blind, placebo-controlled study assessed the necessity of early intervention, safety and efficacy of intravenous zoledronic acid 5 mg/year in East China women with newly diagnosed osteoporosis at high risk of fracture during a 24-month treatment period. Methods Subjects (57 [52-62] years old) were randomized 3:2 to zoledronic acid versus placebo (randomized at baseline, zoledronic acid [175 cases], placebo-zoledronic acid [110 cases]). The bone mineral density of the lumbar spine and total hip was measured every 6 months with the use of dual-energy X-ray absorptiometry. Serum procollagen I N-terminal pro-peptide ( PINP) and serum C-telopeptide of type I collagen ( CTX) levels were measured every 6 months. The primary end point was the rate of change in the bone mineral density at the posteroanterior spine. Results For subjects with measurements at 24 months, zoledronic acid significantly increased bone mineral density ( BMD) at the lumbar spine (mean percent change ± SD, zoledronic acid 5.390% ± 0.854% versus placebo-zoledronic acid −1.038% ± 0.599%), the total hip (zoledronic acid 1.900% ± 0.262% versus placebo-zoledronic acid −1.631% ± 0.649%). Serum procollagen I N-terminal pro-peptide ( PINP) and CTX decreased rapidly with zoledronic acid 5 mg treatment ( P < 0.001 versus placebo at 6 month and 24 months) and changed from baseline in the zoledronic acid 5 mg and placebo-zoledronic acid 5 mg at 6 months by a mean of −66.348% and −75.375%, respectively ( P < 0.001), and at 24 months by −49.950% and −52.325%, respectively ( P < 0.001). No cases of serious adverse events were observed in two groups. Headache, pyrexia and myalgia occurred more commonly within the first 3 days after infusion with zoledronic acid 5 mg than with placebo (13.7% versus 2.1%, P = 0.0018; 28.0% versus 3.2%, P < 0.001; 21.7% versus 4.2%, P < 0.001, respectively). Conclusions These data show that early application of zoledronic acid 5 mg/year was well stimulated and tolerated for bone mass in newly diagnosed east china subjects with osteoporosis in a 24-month treatment. It remains controversial whether contemporary cerebral perfusion techniques, utilized during deep hypothermic circulatory arrest ( DHCA), establish adequate perfusion to deep structures in the brain. This study aimed to investigate whether selective antegrade cerebral perfusion ( SACP) or retrograde cerebral perfusion ( RCP) can provide perfusion equally to various anatomical positions in the brain using metabolic evidence obtained from microdialysis. Eighteen piglets were randomly assigned to 40 min of circulatory arrest ( CA) at 18° C without cerebral perfusion ( DHCA group, n = 6) or with SACP ( SACP group, n = 6) or RCP ( RCP group, n = 6). Microdialysis parameters (glucose, lactate, pyruvate, and glutamate) were measured every 30 min in cortex and striatum. After 3 h of reperfusion, brain tissue was harvested for Western blot measurement of α-spectrin. After 40 min of CA, the DHCA group showed marked elevations of lactate and glycerol and a reduction in glucose in the microdialysis perfusate (all P < 0.05). The changes in glucose, lactate, and glycerol in the perfusate and α-spectrin expression in brain tissue were similar between cortex and striatum in the SACP group (all P > 0.05). In the RCP group, the cortex exhibited lower glucose, higher lactate, and higher glycerol in the perfusate and higher α-spectrin expression in brain tissue compared with the striatum (all P < 0.05). Glutamate showed no difference between cortex and striatum in all groups (all P > 0.05). In summary, SACP provided uniform and continuous cerebral perfusion to most anatomical sites in the brain, whereas RCP resulted in less sufficient perfusion to the cortex but better perfusion to the striatum. Electrospun poly ( p-phenylene terephthalamide) (PPTA) fibers with diameters between 275 nm and 15 µm were obtained from anisotropic solutions, albeit that the spinning process was difficult to control. Key solution parameters like conductivity, surface tension, and viscosity were used to compare the spinnability of these anisotropic solutions with common polymer solutions in order to better understand the challenges of spinning such solutions. Mechanical properties of the electrospun p-aramid single fibers were evaluated and it was found that Young's modulus and tensile strength increased dramatically with decreasing fiber diameter. A maximum Young's modulus and tensile strength of 59 and 1.1 GPa, respectively were found and a geometrical model for the size-dependent Young's modulus was developed to predict fiber properties based on rigid-rod chains down to the nanoscale. 31. A novel TXNIP-based mechanism for Cx43-mediated regulation of oxidative drug injury.

Gap junctions ( GJs) play an important role in the regulation of cell response to many drugs. However, little is known about their mechanisms. Using an in vitro model of cytotoxicity induced by geneticin (G418), we explored the potential signalling mechanisms involved. Incubation of cells with G418 resulted in cell death, as indicated by the change in cell morphology, loss of cell viability and activation of caspase-3. Before the onset of cell injury, G418 induced reactive oxygen species ( ROS) generation, activated oxidative sensitive kinase P38 and caused a shift of connexin 43 (Cx43) from non-phosphorylated form to hyperphosphorylated form. These changes were largely prevented by antioxidants, suggesting an implication of oxidative stress. Downregulation of Cx43 with inhibitors or si RNA suppressed the expression of thioredoxin-interacting protein ( TXNIP), activated Akt and protected cells against the toxicity of G418. Further analysis revealed that inhibition of TXNIP with si RNA activated Akt and reproduced the protective effect of Cx43-inhibiting agents, whereas suppression of Akt sensitized cells to the toxicity of G418. Furthermore, interference of TXNIP/Akt also affected puromycin- and adriamycin-induced cell injury. Our study thus characterized TXNIP as a presently unrecognized molecule implicated in the regulatory actions of Cx43 on oxidative drug injury. 34. Estimating the effects of climate change on the intensification of monsoonal-driven stream discharge in a Himalayan watershed.

Understanding potential hydrologic influences to continued climate change in Himalayan watersheds is important for management of transnational water resources. This study estimates the climate change impacts on hydrologic processes of the Kali Gandaki watershed from central Himalayan region using the Soil and Water Assessment Tool. Daily predicted stream discharge of the basin for 1981-95 following calibration was accurate with Nash Sutcliffe Efficiency value >0.75. Sensitivity analysis of the hydrologic parameters showed the precipitation and temperature lapse rates as the most sensitive parameters to the stream discharge. To assess the influence of continued climate change on hydrologic processes, we modified the weather inputs for the model using average, minimum and maximum temperature, and precipitation changes for the Special Report on Emission Scenarios B1, A1B and A2 derived from 16 General Circulation Models for 2080s. Mean annual stream discharge was approximately 39% higher than current values for the maximum temperature and precipitation changes of the A2 scenario and 22% less for minimum changes of the same scenario. Stream discharge was projected to be changed by +9% during monsoon season and by −6% during pre-monsoon season. Snowfall and snow melt were projected to be 30% and 29%, respectively, less than the current average for the maximum temperature and precipitation changes of the A2 scenario. Future simulations showed potential increase in monsoonal stream discharge associated with projected higher precipitation which when coupled with enhanced summer glacier melt might influence the downstream water availability of the basin. A bis-branched [3]rotaxane, with two [2]rotaxane arms separated by an oligo( para-phenylenevinylene) (OPV) fluorophore, was designed and investigated. Each [2]rotaxane arm employed a difluoroboradiaza- s-indacene (BODIPY) dye-functionalized dibenzo[24]crown-8 macrocycle interlocked onto a dibenzylammonium in the rod part. The chemical structure of the [3]rotaxane was confirmed and characterized by 1H and 13C NMR spectroscopy and high-resolution ESI mass spectrometry. The photophysical properties of [3]rotaxane and its reference systems were investigated through UV/Vis absorption, fluorescence, and time-resolved fluorescence spectroscopy. An efficient energy-transfer process in [3]rotaxane occurred from the OPV donor to the BODIPY acceptor because of the large overlap between the absorption spectrum of the BODIPY moiety and the emission spectrum of the OPV fluorophore; this shows the important potential of this system for designing functional molecular systems. Background and Purpose Suramin is a clinically prescribed drug for treatment of human African trypanosomiasis, cancer and infection. It is also a well-known pharmacological antagonist of P2 purinoceptors. Despite its clinical use and use in research, the biological actions of this molecule are still incompletely understood. Here, we investigated the effects of suramin on membrane channels, as exemplified by its actions on non-junctional connexin43 ( Cx43) hemichannels, pore-forming α-haemolysin and channels involved in ATP release under hypotonic conditions. Experimental Approach Hemichannels were activated by removing extracellular Ca
2+ . The influences of suramin on hemichannel activities were evaluated by its effects on influx of fluorescent dyes and efflux of ATP. The membrane permeability and integrity were assessed through cellular retention of preloaded calcein and LDH release. Key Results Suramin blocked Cx43 hemichannel permeability induced by removal of extracellular Ca2+ without much effect on Cx43 expression and gap junctional intercellular communication. This action of suramin was mimicked by its analogue NF023 and NF449 but not by another P2 purinoceptor antagonist PPADS. Besides hemichannels, suramin also significantly blocked intracellular and extracellular exchanges of small molecules caused by α-haemolysin from S taphylococcus aureus and by exposure of cells to hypotonic solution. Furthermore, it prevented α-haemolysin- and hypotonic stress-elicited cell injury. Conclusion and Implications Suramin blocked membrane channels and protected cells against toxin- and hypotonic stress-elicited injury. Our finding provides novel mechanistic insights into the pharmacological actions of suramin. A new approach was developed for the magnetic separation of copper(II) ions with easy operation and high efficiency. p-Mercaptobenzoic acid served as the modified tag of Fe
2 O3 @Au nanoparticles both for the chelation ligand and Raman reporter. Through the chelation between the copper(II) ions and carboxyl groups on the gold shell, the Fe2 O3 @Au nanoparticles aggregated to form networks that were enriched and separated from the solution by a magnet. A significant decrease in the concentration of copper(II) ions in the supernatant solution was observed. An extremely sensitive method based on surface-enhanced Raman spectroscopy was employed to detect free copper(II) ions that remained after the magnetic separation, and thus to evaluate the separation efficiency. The results indicated the intensities of the surface-enhanced Raman spectroscopy bands from p-mercaptobenzoic acid were dependent on the concentration of copper(II) ions, and the concentration was decreased by several orders of magnitude after the magnetic separation. The present protocol effectively decreased the total amount of heavy metal ions in the solution. Palynological analysis of 24 samples from four types of natural pollen traps (Lugu Lake bottom sediments, surface soil, bark samples, and moss cushions) in four sites at different altitudes from the Lugu Lake area, southwest China, has been undertaken to investigate pollen dispersal and deposition in a mountainous area and assist with the interpretation of fossil pollen analysis. Detailed comparisons between the palynological assemblage and the modern vegetation in the Lugu Lake region have been carried out. Preliminary interpretations of the correlation between pollen assemblage and vegetation at the different vertical vegetational zones can be recognized by the percentages of the main taxa, and most of the pollen taxa except Pinus are expected to be underrepresented. Exotic pollen grains can be transported over mountains more than 70 km away by wind. Upslope or downslope transport of pollen grains is crucial when reconstructing palaeoclimate in mountainous areas. We summarize the altitudinal distributions of modern woody plants whose pollen grains are present at three sites, and reveal that pollen grains are more readily carried uphill than downhill. These findings have important implications regarding the reconstruction of vegetation in mountainous regions and the interpretion of palaeoelevations. [ABSTRACT FROM AUTHOR]

A new conifer, Austrohamia acanthobractea, sp. nov., is described from the Jurassic Daohugou flora, Inner Mongolia Autonomous Region, China. The material consists of impressions represented by well-preserved leafy twigs and branches as well as ovulate cones. Leafy shoots with at least two orders of branching; ultimate branchlets alternate or sub-opposite with helically arranged leaves, decurrent at base with distal rounded tip; dorsiventrally flattened and univeined. Ovuliferous cones elliptical, less than 1 cm long, terminally borne on ultimate and penultimate branches, composed of helically arranged bracts with ovules disposed on their adaxial surfaces. The presence of similar, if not identical taxa, on both sides of the Pacific indicates the cosmopolitan distribution of primitive Cupressaceae between East Asia (Eurasia) and South America in the Pangaea. 41. Development of chondroitin sulfate encapsulated PLGA microsphere delivery systems with controllable multiple burst releases for treating osteoarthritis.

The purpose of the study was to design and develop unique drug delivery systems with controllable multiple burst releases of drugs for treating osteoarthritis. Chondroitin sulfate (CS) was encapsulated into four types of PLGA materials, that is, PLGA 50:50, PLGA 65:35, PLGA 75:25, and PLGA 85:15. The effects of microsphere size and various combinations of blend PLGA microspheres on CS release were investigated. The cytotoxicity of the CS-encapsulated microspheres was investigated according to the ISO 10993 guideline. Our study showed that the encapsulation efficiency of CS into PLGA 50:50 microspheres varied with the size of microspheres; however, the encapsulation efficiencies of CS into PLGA microspheres were independent of the types of PLGA materials. The size of PLGA microspheres was shown to affect the rate of CS release. With the increase of microsphere size from 75-150 μm to 300-355 μm, the initial CS release decreased. Further increase in microsphere size led to an increase in the initial CS release. In addition, combination of different types of PLGA microspheres was shown to be capable of achieving multiple burst CS releases. Moreover, the CS encapsulated PLGA microspheres were shown to be non-cytotoxic. This study proved the concept of multiple burst drug releases that were achieved by encapsulating CS into different types of PLGA microspheres and delivering CS from systems consisting of mixed types of PLGA microspheres, which may be applied to treat osteoarthritis by mimicking multiple intra-joint injection of therapeutic agents. © 2011 Wiley Periodicals, Inc. The red turpentine beetle (RTB), Dendroctonus valens LeConte, as a destructive invasive pest, has become one of the most economically important forest pest in China. Effective control measures are desperately needed. Entomopathogenic fungi, such as Beauveria bassiana, have shown great potential for the management of some bark beetle species. In this study, 12 isolates of B. bassiana from bark beetle were examined for biological characteristics and virulence, to assess their potential as biocontrol agents for RTB. There were significant differences (at P= 0.05) in colony growth rate, conidial yield, conidial germination, tolerance to UV light and extracellular proteases activity among the tested B. bassiana isolates. Isolates, including Bb1801, Bb1906, Bb789 and Bb773, exhibited the best characteristics, because they have faster hyphal growth rate, higher spore production and faster spore germination, higher UV tolerance and protease (Pr1) production. The results of a pathogenicity test of B. bassiana on RTB larvae showed that most isolates of B. bassiana have demonstrated high efficacy and the highest virulent isolate was Bb1801, which killed 100% of the treated insects and had a median lethal time (LT) of 4.60 days at a concentration of 1×10 conidia/mL. Therefore, isolate Bb1801 has a great potential for sustainable control of RTB in the forest. The correlation between biological characteristics and virulence of the fungal isolates is discussed and the possibility of combination of entomopathogenic fungi with semiochemicals, as one of the promising strategy for RTB control, is considered. The wear, delamination, and fatigue resistance of artificially aged gamma irradiation-sterilized conventional polyethylene (CPE) and gas-plasma-sterilized melt-annealed highly crosslinked polyethylene tibial inserts (HXPE) were compared. Six CPE and 12 HXPE (six irradiated at 58 kGy and six at 72 kGy) left knee inserts were wear tested for 5.5 million cycles (Mc) under loads and motions that mimic activities of daily living, such as walking, chair rise, stair ascent, and deep squatting. Another six HXPE (72 kGy) and six CPE inserts were also tested under conditions that could produce severe delamination for 8 Mc. Ten other knees (five 72 kGy HXPE and five CPE) were subjected to posterior edge loading fatigue testing for 5 Mc. The HXPE inserts had an average wear rate reduction of about 80% relative to their CPE counterparts during all activities. All of the CPE inserts delaminated and fractured during high cycle deep squat (152° flexion) motions, while all the HXPE remained intact. None of the HXPE inserts delaminated after 8 Mc, while all of the CPE inserts developed delamination damage within 1.5-5.8 Mc of delamination testing. All CPE inserts developed subsurface cracks and delamination within 2.8 Mc during posterior edge loading fatigue studies, while none of the HXPE inserts showed cracking or delamination after 5 Mc. These results show that aged HXPE has higher wear and fatigue resistance than aged CPE, and offers potential long-term advantages for young active patients with sustained activities of daily living. © 2010 Orthopaedic Research Society. Background and Purpose: Gap junctions play important roles in the regulation of cell phenotype and in determining cell survival after various insults. Here, we investigated the role of gap junctions in aminoglycoside-induced injury to renal tubular cells.Experimental Approach: Two tubular epithelial cell lines NRK-E52 and LLC-PK1 were compared for gap junction protein expression and function by immunofluorescent staining, Western blot and dye transfer assay. Cell viability after exposure to aminoglycosides was evaluated by WST assay. Gap junctions were modulated by transfection of the gap junction protein, connexin 43 (Cx43), use of Cx43 siRNA and gap junction inhibitors.Key Results: NRK-E52 cells expressed abundant Cx43 and were functionally coupled by gap junctional intercellular communication (GJIC). Exposure of NRK-E52 cells to aminoglycosides, G418 and hygromycin, increased Cx43 phosphorylation and GJIC. The aminoglycosides also decreased cell viability that was prevented by gap junction inhibitors and Cx43 siRNA. LLC-PK1 cells were gap junction-deficient and resistant to aminoglycoside-induced cytotoxicity. Over-expression of a wild-type Cx43 converted LLC-PK1 cells to a drug-sensitive phenotype. The gap junction inhibitor alpha-glycyrrhetinic acid (alpha-GA) activated Akt in NRK-E52 cells. Inhibition of the Akt pathway enhanced cell toxicity to G418 and abolished the protective effects of alpha-GA. In addition, gentamycin-elicited cytotoxicity in NRK-E52 cells was also significantly attenuated by alpha-GA.Conclusion and Implications: Gap junctions contributed to the cytotoxic effects of aminoglycosides. Tissue‐engineered osteochondral grafts have been synthesized from a variety of materials, with some success at repairing chondral defects in animal models. We hypothesized that in tissue‐engineered osteochondral grafts synthesized by bonding mesenchymal stem cell‐loaded hydrogels to a porous material, the choice of the porous scaffold would affect graft healing to host bone, and the quality of cell restoration at the hyaline cartilage surface. Bone marrow‐derived allogeneic mesenchymal stem cells were suspended in hydrogels that were attached to cylinders of porous tantalum metal, allograft bone, or a bioactive glass. The tissue‐engineered osteochondral grafts, thus created were implanted into experimental defects in rabbit knees. Subchondral bone restoration, defect fill, bone ingrowth‐implant integration, and articular tissue quality were compared between the three subchondral materials at 6 and 12 weeks. Bioactive glass and porous tantalum were superior to bone allograft in integrating to adjacent host bone, regenerating hyaline‐like tissue at the graft surface, and expressing type II collagen in the articular cartilage. © 2010 Wiley Periodicals, Inc. 49. Gap junctions sensitize cancer cells to proteasome inhibitor MG132-induced apoptosis.

( Cancer Sci 2010; 101: 713–721) Proteasome inhibition is a promising approach for cancer therapy. However, the mechanisms involved have not been fully elucidated. Gap junctions play important roles in the regulation of tumor cell phenotypes and mediation of the bystander effect in cancer therapy. Because the degradation of gap junction proteins involves the proteasome, we speculated that altered gap junctions might contribute to the antitumor activities of proteasome inhibition. Incubation of Hepa-1c1c7 cells with the proteasome inhibitor MG132 elevated the levels of gap junction protein connexin 43 (Cx43) and promoted gap junctional intercellular communication. This was associated with a marked accumulation of ubiquitylated Cx43 and a significantly decreased rate of Cx43 degradation. The elevated Cx43 contributed to MG132-induced cell apoptosis. This is shown by the observations that: (i) overexpression of Cx43 in the gap junction-deficient LLC-PK1 cells rendered them vulnerable to MG132-elicited cell injury; (ii) fibroblasts derived from Cx43-null mice were more resistant to MG-132 compared with Cx43 wild-type control; and (iii) the gap junction inhibitor flufenamic acid significantly attenuated cell damage caused by MG132 in Hepa-1c1c7 cells. Further studies demonstrated that MG132 activates endoplasmic reticulum stress. Exposure of cells to the endoplasmic reticulum stress inducers thapsigargin and tunicamycin also led to cell apoptosis, which was modulated by Cx43 levels in a way similar to MG132. These results suggested that elevated Cx43 sensitizes cells to MG132-induced cell apoptosis. Regulation of gap junctions could be an important mechanism behind the antitumor activities of proteasome inhibitors. [ABSTRACT FROM AUTHOR]
