Background: The use of mesh is controversial in the treatment of female pelvic organ prolapse.Objectives: To systematically review the outcomes of sacrocolpopexy compared with transvaginal mesh surgery and to provide evidence-based suggestions.Search Strategy: The MEDLINE, EMBASE, Cochrane Library and clinicaltrials.gov databases were searched on 21 November 2018.Selection Criteria: Randomised controlled trials and prospective and retrospective cohort studies were included.Data Collection and Analysis: Data were extracted by one reviewer and examined by a second reviewer for accuracy. Odds ratios and 95% CI were calculated using random-effects models.Main Results: Twenty comparative studies were included. The meta-analysis was performed with subgroups. The summary odds ratios of the randomised controlled group were 1.84 (95% CI 0.79-4.29, I2  = 75%) for anatomical success, 1.41 (95% CI 0.47-4.24, I2  = 38%) for subjective success, 0.42 (95% CI 0.18-0.98, I2  = 0%) for mesh complications, 0.61 (95% CI 0.20-1.91, I2  = 0%) for prolapse reoperation and 0.44 (95% CI 0.23-0.88, I2  = 0%) for de novo dyspareunia. The mean differences were 0.77 (95% CI 0.31-1.23, I2  = 66%) for total vaginal length and -1.28 (95% CI -2.00 to -0.55, I2  = 66%) for point C after surgery.Conclusions: Very-low-quality evidence indicated that the anatomical and subjective success rates of sacrocolpopexy were similar to those of transvaginal mesh surgery; sacrocolpopexy might be more beneficial than transvaginal mesh surgery in terms of mesh-related complication rates, prolapse recurrence and de novo dyspareunia. We report the direct observation on bursty bulk flow (BBF) deceleration down‐tail of X < −15 RE by MMS satellite. Two typical events are presented in the paper. In the first event on 05 June 2018, MMS1 is located at X ~ −16.1 RE and records four individual bursty flows (BFs). Each burst flow has distinctly lower velocity than the preceding one. Accompanying with the decelerated BFs, the Bz/Bx continuously increases/decreases. Simultaneous Vx‐decrease and Bz‐increase are in coincidence with the scenario of the local BBF deceleration and formation of the magnetic pileup region. In the second event on 03 July 2017, MMS stays in the neutral sheet of X ~ −24.5 RE, and encounters similar BBF deceleration process. For both events, the decelerated BF series exhibit prominent medium‐energy ion component (2–10 KeV). Analyses show enhanced parallel current (J//) and Kinetic Alfvénic wave (KAW) emitting during the BF intervals. The strength of the emitted KAW has a clear tendency to decay with the BBF decreasing. Power spectra density analysis confirms the substantial Joule dissipation during the BBF deceleration, both J// and J⊥. Combined analyses support BBF dissipation via Joule heating as well as KAW emitting. Finally, we propose a possible mechanism on the BBF deceleration, i.e., "collision" with the tailward flow. Abstract: The ability to recover from drought stress after re‐watering is an important feature that will enable plants to cope with the predicted increase in episodic drought. The effects of pre‐drought and re‐watering conditions on leaf spectral properties and their relationships with the biochemical processes that underlie the recovery from pre‐drought conditions should be better understood. The reflectance spectra, 10 spectral reflectance indices (SRIs) and biochemical characteristics of maize (Zea mays) leaves were monitored 7, 14, 21 and 28 days after the initiation of soil drought stress during two successive cycles of drought and re‐watering periods. The leaf reflectance of the two inbred maize lines increased under the drought stress, especially in the visible spectral range. In addition, an obvious recovery of the leaf reflectance was only observed in the first re‐watering period, and its value remained higher than that of the control plants during the second recovery period. A recovery lag in the pigment contents was also observed during the second cycle. The recovery variations in the pattern and magnitude of the SRIs and the total contents of C, N and P that were measured in response to the re‐watering during both cycles were diverse and complex; both full and partial recoveries were observed. The SRIs representing different physiological attributes of plant growth, including the water index, red edge position, photochemical reflectance index and near‐infrared reflectance at 800 nm, showed strong linear relationships (P < 0.01 or 0.05) with the growth and biochemical traits across the successive drought and re‐watering cycles. The results suggest that maize plants can adjust their leaf reflectance properties and employ growth and biochemical strategies to adapt to cyclic drought stress and recover from drought stress after re‐watering. Micro RNAs (mi RNAs) are one of a growing class of noncoding RNAs that are involved in the regulation of a wide range of metabolic processes including cellular differentiation, cell proliferation and apoptosis. The generation of mi RNA is regulated in complex ways, for example by small interfering RNAs (small nucleolar and nuclear RNAs) and various other metabolites. This complexity of control is likely to explain how a relatively small part of the DNA that codes for proteins has enabled the evolution of such complex organisms as mammals. Non-protein-coding DNA is therefore thought to carry the memory of early evolutionary steps that led to progressively complex metabolic controls. Clinically, mi RNAs are becoming increasingly important following the recognition that some congenital abnormalities can be traced to defects in mi RNA processing. The potential for manipulating metabolism and affecting disease processes by the pharmaceutical or biological targeting of specific mi RNA pathways is now being tested. mi RNAs are also released into the extracellular milieu after packaging by cells into nano-sized extracellular vesicles. Such vesicles can be taken up by adjacent and possibly more distant cells, thereby allowing coordinated intercellular communication in specific tissues. Extracellular mi RNAs found in the blood stream may also serve as novel biomarkers for both diagnosing specific forms of cancer and assessing the likelihood of metastasis, and as powerful prognostic indices for various cancers. Here, we discuss the role of intracellular and extracellular mi RNAs in nutritional control of various (patho)physiological processes. This study aimed to evaluate the effects of single nucleotide polymorphisms ( SNPs) in candidate genes for meat quality using a custom 96- SNP panel ( Illumina Vera Code Golden Gate Assay) on 15 traits collected from 400 commercial pigs. Meat quality measurements included muscle p H, color ( L*, a* and b*), drip loss, cooking loss, peak shear force and six sensory traits including appearance (outside and inside), tenderness, juiciness, flavor and overall liking as well as carcass weight and probe yield. Thirty-five SNPs with minor allele frequencies > 0.10 remained for the multimarker association using the GLM procedure of sas 9.2. Results showed that 20 SNPs were significantly associated with at least one of the traits with either additive or dominance or both effects ( P < 0.05). Among these significant SNPs, five of them in ADIPOQ, FTO, TNF, LEPR and AMPD1 had an effect on more than three traits simultaneously; those in MC4 R, CAST, DGAT1 and MYF6 had an effect on two traits, while the others were associated with one trait. The results suggest that these markers could be incorporated into commercial pigs for marker-assisted selection and breeding programs for carcass and meat quality trait improvement. This study was carried out in 121 pigs to develop a population pharmacokinetic ( PPK) model by oral (p.o.) administration of valnemulin at a single dose of 10 mg/kg. Serum biochemistry parameters of each pig were determined prior to drug administration. Three to five blood samples were collected at random time points, but uniformly distributed in the absorption, distribution, and elimination phases of drug disposition. Plasma concentrations of valnemulin were determined by high-performance liquid chromatography-tandem mass spectrometry ( HPLC- MS/ MS). The concentration-time data were fitted to PPK models using nonlinear mixed effect modeling ( NONMEM) with G77 FORTRAN compiler. NONMEM runs were executed using Wings for NONMEM. Fixed effects of weight, age, sex as well as biochemistry parameters, which may influence the PK of valnemulin, were investigated. The drug concentration-time data were adequately described by a one-compartmental model with first-order absorption. A random effect model of valnemulin revealed a pattern of log-normal distribution, and it satisfactorily characterized the observed interindividual variability. The distribution of random residual errors, however, suggested an additive model for the initial phase (<12 h) followed by a combined model that consists of both proportional and additive features (≥12 h), so that the intra-individual variability could be sufficiently characterized. Covariate analysis indicated that body weight had a conspicuous effect on valnemulin clearance ( CL/ F). This study was performed in 145 pigs to develop a population pharmacokinetics ( PPK) model by i.m. administration of cefquinome (CEQ) at the dose of 2 mg/kg in the neck muscle. Serum physiological and biochemical parameters for each pig were determined before administration. After administration, 2-4 samples were collected at random, with the sampling point evenly distributed in the three periods (<1 h, 1-4 h and >4 h). The plasma concentration of CEQ was determined by high performance liquid chromatography with UV detector. The pharmacostatistical analyses of concentration-time data, weight, age, gender, serum physiological and biochemical parameters were performed with nonlinear mixed effect modeling ( NONMEM). A one-compartmental model with first-order absorption and elimination adequately described the data from the study group. The optimal random effect model of pharmacokinetics parameters was of log-normal distribution and the residual errors assumed a mixed-type model (proportional and additive) to best explain intra-individual variability. Covariate analysis showed that body weight is positively correlated with apparent volume of distribution ( V/ F) and body clearance (CL/ F). The typical PPK parameters of Ka, CL, and V were 0.564/h, 5.15 L/h, and 1.36 L, respectively. [ABSTRACT FROM AUTHOR]

The nanohydroxyapatite/chitosan/poly( L-lactic acid) (HA/CS/PLLA) ternary biocomposites were prepared by blending the hydroxyapatite/chitosan (HA/CS) nanocomposites with poly( L-lactic acid) (PLLA) solution. Surface modification by grafting D-, L-lactic acid onto the HA/CS nanocomposites was designed to improve the bonding with PLLA. The FTIR and 13C-NMR spectrum confirmed that the oligo(lactic acid) was successfully grafted onto the HA/CS nanocomposites, and the time-dependent phase monitoring showed that the grafted copolymers were stable. The TEM morphology of the HA/CS/PLLA ternary nanocomposites showed that nano-HA fibers were distributed homogeneously, compacted closely and wrapped tightly by the CS and PLLA matrix. The ternary biocomposites with the HA content of 60 and 67 wt % exhibited high compressive strength of about 160 MPa and suitable hydrophilicity. The in vitro tests exhibited that the ternary biocomposites have good biodegradability and bioactivity when immersed in SBF solutions. Nano-fibrous hydroxyapatite/chitosan (HA/CS) composites with HA contents of 0-70 wt % were prepared by in situ hybridization with a preprepared chitosan film as semipermeable membrane to slow down the hybridization process. The phase composition, microstructure, and morphology of the composites were characterized by means of Fourier transform infrared spectroscopy, X-ray diffraction, and high-resolution transmission electron microscopy. It was found that the in situ prepared inorganic phase was hydroxyapatite nano-fibers that were uniformly dispersed in the chitosan matrix. The average diameter of the fibers were about 3 nm, while the length of the fibers increases from 20 to 60 nm when the hydroxyapatite content increased from 10 to 70 wt %. The compressive strength and Young's modulus of these nano-fibrous HA/CS composites increased with the increasing HA content and reached the highest values of 170 and 1.7 GPa, respectively, at the HA content of 50-70 wt %, which were much higher than the values of samples prepared by coprecipitation. © 2011 Wiley Periodicals, Inc. The objective of this study was to investigate the pharmacokinetics and bioavailability of valnemulin in broiler chickens after intravenous (i.v.), intramuscular (i.m.) and oral administrations of 10 mg/kg body weight (bw). Plasma samples were analyzed by high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). Pharmacokinetic characterization was performed by non-compartmental analysis using WinNonlin program. After intravenous administration, distribution was wide with the volume of distribution based on terminal phase(V) of 4.27 ± 0.99 L /kg. Mean valnemulin t(h), Cl(L /h /kg), V (L /kg) and AUC(μg·h /mL) values were 2.85, 0.99, 2.72 and 10.34, respectively. After intramuscular administration, valnemulin was rapidly absorbed with a C of 2.2 μg/mL achieved at 0.43 h (t), and the absolute bioavailability (F) was 88.81%; and for the oral route the same parameters were 0.66 ± 0.15 μg/mL, 1.54 ± 0.27 h and 74.42%. A multiple-peak phenomenon was present after oral administration. The plasma profile of valnemulin exhibited a secondary peak during 2-6 h and a tertiary peak at 32 h. The favorable PK behavior, such as the wide distribution, slow elimination and acceptable bioavailability indicated that it is likely to be effective in chickens. [ABSTRACT FROM AUTHOR]

Chemokine receptor-2 (CCR2) is a co-receptor for the entry of human immunodeficiency virus-1 (HIV-1) into the target cells. A mutation in CCR2 (CCR2-64I) exhibited a protective effect to delay the progression of acquired immunodeficiency syndrome (AIDS). To study the mutant frequency and polymorphism of CCR2 in Chinese populations, 1082 individuals from 15 Chinese populations distributing widely from north to south were collected. The genotypes of CCR2-64I were determined by polymerase chain reaction–restriction fragment length polymorphism (PCR-RFLP) with the digestion of restriction endonuclease FokI. Of the 1082 individuals, 352 (32.53%) were carriers of CCR2-64I allele, 257 of whom (23.75%) were heterozygotes (CCR2-64V/I), whereas 95 (8.78%) were homozygotes (CCR2-64V/V). The frequency of the CCR2-64I allele in those tested individuals was 20.66%. This prevalence of CCR2-64I was higher than what was known for American and European populations. Moreover, the frequencies of CCR2-64I were generally higher in northern China than they were in southern China, and the frequencies had significant variance in 15 populations of China (χ2 = 27.135, P = 0.018). The pharmacological effects of 14-deoxyandrographolide on rat isolated thoracic aorta were examined. 2. 14-Deoxyandrographolide (2.5-120 mumol/L) inhibited contractions induced by phenylephrine (PE; 0.1 mumol/L) and high K+ (80 mmol/L) in a concentration-dependent manner in endothelium-intact aorta. The effect was attenuated in endothelium-denuded aorta without modifying the maximal response. Like verapamil, 14-deoxyandrographolide produced a much greater vasorelaxant effect in aorta precontracted by KCl than by PE. 14-Deoxyandrographolide (20-60 mumol/L) also inhibited responses of the rat aorta to PE. 3. In Ca(2+)-free medium (KCl 55 mmol/L), 14-deoxyandrographolide (20-80 mumol/L) antagonized Ca(2+)-induced vasocontraction in a concentration-dependent manner and transient contractions induced by both caffeine (10 mmol/L) and nor-adrenaline (1 mumol/L) were suppressed or almost abolished by 14-deoxyandrographolide. 4. The vasorelaxant effect of 14-deoxyandrographolide was partially antagonized by NG-nitro-L-arginine methyl ester (25 mumol/L), a specific and competitive nitric oxide synthase (NOS) inhibitor, and methylene blue (10 mumol/L), a soluble guanylate cyclase inhibitor, but was not affected by indomethacin (20 mumol/L), a cyclo-oxygenase inhibitor, or glibenclamide (10 mumol/L), an ATP-sensitive K(+)-channel blocker. 5. These results suggest that the vasorelaxant activity of 14-deoxyandrographolide may be mediated via the activation of NOS and guanylate cyclase, as well as the blockade of Ca2+ influx through both voltage- and receptor-operated Ca2+ channels.

1. The hypotensive activity of an aqueous extract of Andrographis paniculata was studied using chronic intraperitoneal (i.p.) infusions by osmotic pumps. The extract exhibited a dose-dependent hypotensive effect on the systolic blood pressure (SBP) of spontaneously hypertensive rats (SHR). 2. The optimum hypotensive dose determined was repeated in a study in SHR and their normotensive controls, Wistar-Kyoto (WKY) rats, to demonstrate its comparative effects on the SBP, plasma and lung angiotensin-converting enzyme (ACE) activities, as well as on lipid peroxidation in the kidneys, as measured by the thiobarbituric acid (TBA) assay. 3. The extract significantly lowered the SBP of both SHR and WKY rats. 4. Plasma, but not lung, ACE activity and kidney TBA level were significantly lower in extract-treated SHR when compared with vehicle-treated SHR controls. 5. Plasma and lung ACE activities as well as kidney TBA levels were not significantly different between extract- and vehicle-treated WKY rats. 6. This study indicates that the aqueous extract of A. paniculata lowers SBP in the SHR possibly by reducing circulating ACE in the plasma as well as by reducing free radical levels in the kidneys. The mechanism(s) of hypotensive action seems to be different in WKY rats.
