BACKGROUND: Ergothioneine (EGT) is a high‐value food functional factor that cannot be synthesized by humans and other vertebrates, and the low yield limits its application. RESULTS: In this study, the optimal fermentation temperature, fermentation time, initial pH, inoculum age, and inoculation ratio on EGT biosynthesis of Rhodotorula mucilaginosa DL‐X01 were optimized. In addition, the effects of three key precursor substances – histidine, methionine, and cysteine – on fungal EGT synthesis were verified. The optimal conditions were further obtained by response surface optimization. The EGT yield of R. mucilaginosa DL‐X01 under optimal fermentation conditions reached 64.48 ± 2.30 mg L−1 at shake flask fermentation level. Finally, the yield was increased to 339.08 ± 3.31 mg L−1 (intracellular) by fed‐batch fermentation in a 5 L bioreactor. CONCLUSION: To the best of our knowledge, this is the highest EGT yield ever reported in non‐recombinant strains. The Masculinizer gene, Masc, encodes a lepidopteran‐specific novel CCCH‐type zinc finger protein, which controls sex determination and dosage compensation in Bombyx mori. Considering the potential application of it in pest control, it is necessary to investigate the function of Masc gene in Hyphantria cunea, a globally invasive forest pest. In the present study, we identified and functionally characterized the Masc gene, HcMasc, in H. cunea. Sequence analysis revealed that HcMASC contained the conserved CCCH‐type zinc finger domain, nuclear localization signal, and male determining domain, in which the last was confirmed to be required for its masculinization in BmN cell line. However, expression data showed that unlike male‐biased expression in B. mori, HcMasc gene expresses in main all developmental stages or tissues in both sexes. Clustered regularly interspaced palindromic repeats (CRISPR) / CRISPR‐associated protein 9‐based disruption of the common exons 1 and 3 of the HcMasc gene resulted in imbalanced sex ratio and abnormal external genitalia of both sexes. Our results suggest that the HcMasc gene is required for both male and female sexual differentiation and dosage compensation in H. cunea and provide a foundation for developing better strategies to control this pest. Background: To compare the clinical outcomes and prognosis of total pancreatectomy (TP) and pancreaticoduodenectomy (PD) for the treatment of pancreatic ductal adenocarcinoma (PDAC), and to explore the safety and indications of TP. Methods: A systematic search was conducted on PubMed, Web of Science, and Embase databases from January 1943 to March 2023 for literatures comparing TP and PD in the treatment of PDAC. The primary outcome was postoperative overall survival (OS), and secondary outcomes included surgery time, blood loss, readmission, hospital stay, perioperative mortality, and overall morbidity. Fixed‐effect or random‐effect models were selected based on heterogeneity, and odds ratio (OR), mean difference (MD), or hazard ratio (HR) with 95% confidence intervals (CI) were calculated. Results: A total of six studies involving 8396 patients were included in the meta‐analysis. There was no statistically significant difference in OS after surgery between the two groups (HR = 1.08, 95% CI: 0.91–1.27; P = 0.38). The TP group had a longer surgery time (MD = 13.66, 95% CI: 4.57–22.75; P = 0.003) and more blood loss (MD = 133.17, 95% CI: 8.00–258.33; P = 0.04) than the PD group. There were no significant differences between the two groups in terms of hospital stay (MD = 0.09, 95% CI: −2.04 to 2.22; P = 0.93), readmission rate (OR = 1.39; 95% CI: 1.00–1.92; P = 0.05), perioperative mortality (OR = 1.29, 95% CI: 0.98–1.69; P = 0.07), and overall morbidity (OR = 0.80, 95% CI: 0.50–1.26; P = 0.33). Conclusion: The surgical process of TP is relatively complex, but there is no difference in short‐term clinical outcomes and OS compared to PD, making it a safe and reliable procedure. Background: The fall webworm, Hyphantria cunea, an invasive forest pest found worldwide, causes serious ecological and economic damage. Currently, the application of chemical pesticides is the most widely used strategy for H. cunea management. However, long‐term pesticide use leads to pest resistance, phytotoxicity, human poisoning, and environmental deterioration. RNA interference (RNAi) technology may provide an environmentally friendly and cost‐effective option for H. cunea control. However, effective RNAi targets and application methods for H. cunea are lacking. Results: We screened and obtained two highly effective RNAi targets, vATPase A (V‐type proton ATPase catalytic subunit A) and Rop (Ras opposite), from 23 candidate genes, using initial and repeat screening tests with the double‐stranded RNA (dsRNA) injection method. RNAi against these two genes was effective in suppressing each target messenger RNA level and interfering with larval growth, leading to significant larval mortality and pupal abnormality. For massive production of dsRNA and practical application of RNAi technology in H. cunea, transformed bacteria expressing dsRNAs of these two genes were prepared using the L4440 expression vector and HT115 strain of Escherichia coli. Oral administration of bacterially expressed dsRNA targeting vATPase A and Rop genes showed high mortality and the same malformed phenotype as the injection treatment. To further investigate the lethal effects of targeting these two genes on larval development, transcriptome sequencing (RNA‐seq) was performed on RNAi samples. The results demonstrated disorders in multiple metabolic pathways, and the expression levels of most genes related to insect cuticle metabolism were significantly different, which may directly threaten insect survival. In addition, some new findings were obtained via RNA‐seq analysis; for example, the progesterone‐mediated oocyte maturation and oocyte meiosis processes were significantly different after silencing vATPase A, and the insect olfactory protein‐related genes were significantly downregulated after dsHcRop treatment. Conclusion: vATPase A and Rop are two highly effective RNAi‐mediated lethal genes in H. cunea that regulate insect growth via multiple metabolic pathways. Oral delivery of bacterially expressed dsRNA specific to vATPase A and Rop can potentially be used for RNAi‐based H. cunea management. BACKGROUND: Studies have shown that either the addition of starter culture or enzyme can improve fermentation in fish or other products. However, little research has been carried out on the effects of coupling starter cultures with lipase on the microbial community and product quality. Suanzhayu is a Chinese fermented fish product that mainly relies on spontaneous fermentation, resulting in an unstable flavor and quality. The present study investigated the impact of lipase and Lactiplantibacillus plantarum 1‐24‐LJ on the quality of Suanzhayu. RESULTS: Inoculation decreased pH and 2‐thiobarbituric acid reactive substances (TBARS) values, and also helped the dominance of the strain in the ecosystem, whereas lipase addition raised TBARS values and had little effect on pH, water activity (aw) and microbiota. The addition of lipase and/or Lpb. plantarum increased the content of alcohols, aldehydes, ketones, esters and umami amino acids. The co‐additions with the most significant effect and the total contents of volatile compounds (VCs) and free amino acids (FAAs) were 1801.92 g per 100 g and 21 357.05 mg per 100 g, respectively. Former‐Lactobacillus was negatively correlated with pH, aw and Prevotella, but positively with VCs (ethyl ester of heptanoic acid, ethyl ester of octanoic acid) and FAAs (Tyr, Phe). Furthermore, adding Lpb. plantarum 1‐24‐LJ alone or in combination with lipase shortened the fermentation process. To better understand the mechanism of palmitoleic acid oxidation into volatile compounds during heating, volatile profiling was investigated by combining thermal‐desorption cryo‐trapping with gas chromatography–mass spectrometry. A total of 49 volatile compounds were detected and identified during this process, including aldehydes (18), ketones (14), alcohols (9), furans (3) and acids and ester (5). The forming temperature of each volatile was determined. Most of the volatiles with short carbon chains were generated at low temperatures, while those with long carbon chains were generated at high temperatures. Results of principal component analysis show that nearly all of the identified volatiles were considered as the characteristic ones of the high temperature points. Meanwhile, the oxidative products of the C7–C11 saturated and unsaturated aldehydes were also detected and identified during heating to intensively investigate the oxidative mechanism of palmitoleic acid during heating. Results demonstrated that only the C10 saturated and unsaturated aldehydes could continue to be oxidized during heating. Therefore, the oxidation reactions above the secondary level of lipids mainly occurred in the C10 saturated and unsaturated aldehydes. [ABSTRACT FROM AUTHOR]

Summary: Suanzhayu is a traditional Chinese fermented fish that often faces a long fermentation time (about 1 month) and unstable quality. This study evaluated the combined effects of papain and Lactiplantibacillus plantarum 1‐24‐LJ, a selected starter culture, on the quality of Suanzhayu. The addition of L. plantarum and/or papain increased the content of alcohols, esters, ketones and umami amino acids (UAAs), but decreased the content of bitter amino acids (BAAs). Besides, Lactobacillus might play a role in reducing harmful bacteria and BAAs content and increasing UAAs content, while Lactococcus and Weissella contributed to the characteristic flavour of Suanzhayu. Overall, no significant improvement in sensory quality was observed with the addition of papain alone, while better quality and faster fermentation processes were obtained with the addition of L. plantarum 1‐24‐LJ alone or in combination with papain. BACKGROUND: Velvet ash (Fraxinus velutina Torr.) is an important wood and ornamental tree species. Emerald ash borer (EAB), Agrilus planipennis Fairmaire, is a major wood borer of velvet ash. The aim of this study was to identify the secondary metabolites of velvet ash involved in regulating the dominant bacterium group of EAB. RESULTS: The amount of lignans in the phloem of infested trees had increased by 290.96% because of A. planipennis infection. The addition of lignans to the artificial diet significantly reduced the weight of the larvae and decreased the dominant bacterial group in the larval midgut, such as Pseudomonadaceae, Xanthomonadaceae, and Enterobacteriaceae. The FvPLR1, a key gene for lignan synthesis, was obtained based on the phloem transcriptome of velvet ash. The expression of FvPLR1 in the phloem of the infested tree was significantly higher than that in the noninfested tree. Meanwhile, FvPLR1 silenced by virus‐induced gene silencing showed that its expression level and the lignan content were decreased by 69.91% and 31.65%, respectively. Interestingly, silencing FvPLR1 induced alterations in the dominant bacteria group in the larvae, with the reverse trend in the lignan‐fed treatment. CONCLUSION: The evidence showed that FvPLR1 was a positive regulator. BACKGROUND: Paocai is a traditional Chinese fermented vegetable food. As the most important ingredient, salt has crucial effects on the bacterial community and volatile compounds of paocai. To demonstrate the effects of salt on the fermentation of paocai, the bacterial composition and volatile compounds were investigated using high‐throughput sequencing and gas chromatography–mass spectrometry (GC–MS). RESULTS: The salt had no significant effects on the bacterial community at the phylum level. Proteobacteria and Bacteroidetes gradually decreased during the fermentation, and Firmicutes gradually increased as the dominant bacteria in the late stage of fermentation. At the genus level, Lactobacillus and Lactococcus gradually increased in relative abundance during the fermentation and became the dominant bacteria in paocai. High salt levels can contribute to the growth of Lactobacillus, which became the dominant genus in paocai. The salt concentration affected the profiles of volatile compounds in paocai after fermentation. A total of 42 volatile components were detected by GC–MS, among which phenols, aldehydes, and nitriles were the main ones. A high salt concentration will increase the volatile compound content, mainly aldehydes and alcohols, and improve the flavor of paocai. At the same time, the electronic tongue analysis also showed that a high salt concentration made a major contribution to the flavor of paocai. Summary: The present study evaluated the relationship between the bacterial diversity and metabolites of various sour meats, a traditional fermented meat product. The results showed that sour meats had a low pH (average of 4.34), low water activity (aw, an average of 0.917), and high lactic acid (~19 177.48 mg kg−1) and free amino acid (FAA) (~4047.65 mg kg−1) concentrations. The lactic acid bacteria (LAB), especially Lactobacillus and Weissella, were predominated in the samples. The alcohols, aldehydes and esters accounted for the major components of volatile organic compounds (VOCs). Of which, (E)‐2‐octenal, nerol, cis‐4‐decenal, tetradecanal, linalyl acetate, linalool, 1‐octen‐3‐ol, hexanal, 1‐heptanol, benzaldehyde and (E)‐2‐decenal were considered as the key VOCs of sour meats. The redundancy analysis (RDA) and Pearson's correlation analysis revealed that LAB (Lactobacillus and Weissella) and salt were positively correlated with most FAAs and VOCs. Rhodosporidium toruloides has been reported as a potential biotechnological microorganism to produce carotenoids. The most commonly used molecular and genetic manipulation methods based on Agrobacterium‐mediated transformation (ATMT). However, this method was of relatively lower transformation efficiency. In this study, we optimized the ATMT method for R. toruloides on account of the promoter on T‐DNA, the ratio of A. tumefaciens to R. toruloides NP11, acetosyringone concentration, cocultivation temperature and time, and a transformation efficiency of 2,369 cells per 105 recipient cells was obtained and was 24 times as that of the previous report. With this optimized method, four redder mutants and four yellower mutants were selected out with torularhodin and β‐carotene production preference, respectively. The highest torularhodin production was 1,638.15 µg/g dry cell weight in A1‐13. The yellower mutants were found to divert the metabolic flux from torularhodin and torulene to γ‐carotene and β‐carotene, and the proportion of γ‐carotene and β‐carotene were all over 92%. TAIL‐PCR was carried out to found T‐DNA insertion in these mutants, and insertion hotspot was found. RT‐qPCR results showed that CTA1 genes in these mutants were closely related to the synthesis of total carotenoids, especially torularhodin, and was a potenial metabolic engineering site in the future. BACKGROUND Micromelalopha siversi (Staudinger) (Lepidoptera: Notodontidae) is a defoliator of poplar trees, Populus spp. (Salicaceae). In our previous study, 13,15‐octadecadienal has been conformed as a female‐produced candidate sex pheromone component of M. siversi, but the Z/E stereochemistry of the 1,3‐diene system has not been identified so far. RESULTS: Four unsaturated aliphatic aldehydes, Z13,E15‐18:Ald, Z13,Z15‐18:Ald, E13,E15‐18:Ald, and E13,Z15‐18:Ald, were synthesized from the commercially available 12‐bromo‐1‐decanol mainly by alkylation of lithium alkyne, normal Wittig or Wittig–Schlosser olefination, and hydroboration‐protonolysis. According to gas chromatography (GC) analysis of pheromone gland extracts, Z13,E15‐18:Ald was the main component, and a small amount of Z13,Z15‐18:Ald was also detected, with a ratio of approximately 7:3. However, the results of GC‐electroantennographic detection (GC‐EAD) showed that Z13,E15‐18:Ald was the only compound with electrophysiological activity, whereas Z13,Z15‐18:Ald elicited no activity. In the field, traps baited with only Z13,E15‐18:Ald resulted in much superior results to those with Z13,Z15‐18:Ald as well as the Z13,E15‐18:Ald and Z13,Z15‐18:Ald binary mixture. CONCLUSIONS: Based on geometrically selective synthesis and bioactivity tests, the active sex pheromone component of M. siversi has been identified as Z13,E15‐18:Ald, the pheromone component that has not been identified in Lepidoptera before. Dendrolimus spp. are important destructive pests of conifer forests, and Dendrolimus punctatus Walker (Lepidoptera; Lasiocampidae) is the most widely distributed Dendrolimus species. During periodic outbreaks, this species is said to make "fire without smoke" because large areas of pine forest can be quickly and heavily damaged. Yet, little is known about the molecular mechanisms that underlie the unique ecological characteristics of this forest insect. Here, we combined Pacific Biosciences (PacBio) RSII single‐molecule long reads and high‐throughput chromosome conformation capture (Hi‐C) genomics‐linked reads to produce a high‐quality, chromosome‐level reference genome for D. punctatus. The final assembly was 614 Mb with contig and scaffold N50 values of 1.39 and 22.15 Mb, respectively, and 96.96% of the contigs anchored onto 30 chromosomes. Based on the prediction, this genome contained 17,593 protein‐coding genes and 56.16% repetitive sequences. Phylogenetic analyses indicated that D. punctatus diverged from the common ancestor of Hyphantria cunea, Spodoptera litura and Thaumetopoea pityocampa ~ 108.91 million years ago. Many gene families that were expanded in the D. punctatus genome were significantly enriched for the xenobiotic biodegradation system, especially the cytochrome P450 gene family. This high‐quality, chromosome‐level reference genome will be a valuable resource for understanding mechanisms of D. punctatus outbreak and host resistance adaption. Two novel tri‐substituted γ*‐Dawson‐type polyoxotungstates H12[P2W15O59{Co(H2O)}3]·15H2O (1) and H12[P2W15O59{Ni(H2O)}3]·15H2O (2) have been successfully synthesized under conventional solution conditions, and characterized in the solid state by powder and single‐crystal X‐ray diffraction, IR spectroscopy, thermogravimetric analysis, and elemental analysis. Structural analysis indicates that they are the tri‐substituted γ*‐Dawson isomers, which are very rare in Dawson‐type polyoxometalates. The magnetic properties of both compounds were studied. Erannis ankeraria Staudinger, 1861 (Lepidoptera: Geometridae) is one of the major pests causing serious damages on Larix spp., Quercus L., and Picea Mill. To investigate the conceivable functions of antennal sensilla associated with pheromone detection, we observed the ultrastructure of antennae in female and male of E. ankeraria moths by scanning electron microscopy. Six types (including two subtypes and a new type) of sensilla were recorded and characterized, including Sensilla trichodea (ST I and ST II), Böhm bristles, Sensilla squamiformia, Sensilla chaetica, Sensilla auricillic (SAU) and newly observed serrate‐like sensilla. ST I and SAU were abundant on male antennae, displaying a sexual dismorphism. Serrate‐like sensilla were also peculiar to male antennae. In summary of reported functions of corresponding sensilla, we presumed putative functions of the recorded sensilla in E. ankeraria, providing the morphological basis for sensory mechanisms related to pest management. Aphidius gifuensis is the main enemy of Myzus persieae. While its parasitic rate can be influenced by the hyperparasitoid, Pachyneuron aphidis. As important parts of insects to sense odors from various environments, study of sensilla can lay the foundation of the further study about the parasitic mechanisms, reduce the hyperparasitic rate, and make the most effect usage of A. gifuensis. Here, we give a fundamental study about the morphology of the sensilla on the whole body of male and female P. aphidis. We observed seven main types of sensilla on them totally by using scanning electron microscopy. Including Böhm bristle (BB), chaetica sensilla (ChS), basiconic sensilla (BS), trichoid sensilla (TS), and placodea sensilla (PS), coeleoconica sensilla (CoS), basiconic capitate peg sensilla (BCPS). In addition, TS on antennae can be divided into four subtypes, on wings can be divided into two subtypes. Sensilla were most abundant on the antennae. We observed all types of sensilla on antennae. TS4 was uniporous and PS was multiporous. The other sensilla were nonporous. We did not find sexual dimorphism with regards to sensilla on the antennae except for the location of CoS. In male, CoS situated on the fourth subsegment of flagellum, but on the eighth subsegment in female. In other organs, TS has the largest number. We also found BS on compound eyes and ovipositor, BB on thoracic legs. The possible roles of these sensilla played in life activities are discussed. Aphidius gifuensis Ashmaed parasitizes several species of aphids that cause economic damage to globally important crops; however, the mechanism of host location in this species is unclear. Here, we studied the sensory system in A. gifuensis to explore the likely mechanisms of host location that could be useful in the improved application of this species in aphid biological control programs. We used scanning electron microscopy to observe the external morphologies of the sensilla recorded from the entire body of female and male A. gifuensis. We recorded 16–18 segments and five types of sensilla on female antennae, including Böhm bristle, Chaetica sensillum, basiconic sensillum, trichoid sensillum, and placodea sensillum. The male antennae consisted of 18–20 segments, and distribution of sensilla was similar to female antennae with minor differences. No hole on the surface or tip of the sensilla was observed. Interestingly, scattered pores around the TS were found on the entire body of females, but only from the antennae, abdomen, and thoracic legs in males. TS was the most abundant and widely distributed sensillum. Tympana structures were found on antennae and femur of thoracic legs. Antenna cleaner was found on the tibia of propodeum and wing cleaner was found on the tibia of metapedes. Special crevice‐shaped structures were found on the abdomen in both sexes, whereas strip‐shaped structures were found only on the abdomen of females. Possible functions of the sensilla in relation to their morphologies are discussed. We found no pores on side or tips of the Aphidius gifuensis sensilla, but, interestingly, scattered pores were found around some trichoid sensilla. The red yeast Rhodosporidium toruloides is a known lipid producer capable of accumulating large amounts of triacylglycerols and carotenoids. However, it remains challenging to study its carotenoid production profiles owing to limited biochemical information and inefficient genetic tools. Here we used an Agrobacterium tumefaciens-mediated transformation (ATMT) to change its carotenoid production and profiles. We constructed R. toruloides NP11 mutant libraries with ATMT, selected three mutants with different colours, characterized their carotenoid products by high-pressure liquid chromatography-mass spectrometry (HPLC-MS) analysis and assured differences among those strains in terms of carotenoid production and its composition profiles. We then located T-DNA insertion sites using the genome walking technology and provided discussions in terms of the new phenotypes. The oleaginous yeast Rhodosporidium toruloides is an unconventional yeast species that can accumulate a high content of lipids. Because it belongs to the basidiomycetous group of fungus, limited tools and functional elements are available for genetic engineering of R. toruloides and related red yeasts. Here we report the functional evaluation of five constitutive promoters from this yeast. We assembled a reporter gene expression cassette, consisting of a promoter, the hygromycin gene (HYG) and the nos terminator, and inserted it into the binary vector pZPK. Hygromycin-resistant transformants were obtained when R. toruloides cells were co-cultured with Agrobacterium tumefaciens AGL1 cells harbouring the engineered vector. Genomic integration of the reporter cassette was verified by successful amplification of target DNA fragments. Quantitative PCR analysis suggested that the transformant had only one copy of the reporter cassette. The strength of these promoters was demonstrated at the phenotypic level on the hygromycin-gradient plate and at the transcriptional level by real-time quantitative PCR. It was found that the strengths of these promoters varied no more than five-fold and followed a decreasing sequence of PPGI, PPGK, PFBA, PTPI, and PGPD. ABSTRACT Telenomus dendrolimusi Chu is a dominant egg parasitoid of Dendrolimus, which is the most serious conifer defoliator in China. However, the host locating mechanism of T. dendrolimusi is unclear. To take better advantage of this kind of parasitoid, it is necessary to clarify the basic knowledge of the organization of the wasp's sensory system. In this study, the external morphologies of the sensilla on the entire female and male T. dendrolimusi body were described using scanning electron microscopy. Antennae shape and sensilla types were sexually dimorphic. Ten segments and nine types of sensilla were found on the female antenna, including basiconic sensillum (BS), chaeticum sensillum (ChS), trichoid sensillum type 1 (TS1), trichoid sensillum type 2 (TS2), trichoid curvata sensillum (TCS), sickle-shaped sensillum (SSS), multiporous gustatory sensillum (MGS), campaniform sensillum (CS), and styloconica sensillum (STS). The distribution of the sensilla was directional on female antenna. The male antenna consisted of 12 segments, and 1-3 segments were similar to those of the female. Five types of sensilla were found on male antenna, including BS, ChS, TS1, TCS, and STS. The mouthparts, thoracic legs, eyes, mesonotum, and wings were all covered with longitudinally fluted TSs without hole. A tympana structure appeared on the thoracic legs. Special crevice-shaped structures were found on the abdomens of both T. dendrolimusi sexes. Possible sensilla functions are discussed according to their morphology. An imaged-based profiling and analysis system was developed to predict clinically effective synergistic drug combinations that could accelerate the identification of effective multi-drug therapies for the treatment of triple-negative breast cancer and other challenging malignancies. The identification of effective drug combinations for the treatment of triple-negative breast cancer (TNBC) was achieved by integrating high-content screening, computational analysis, and experimental biology. The approach was based on altered cellular phenotypes induced by 55 FDA-approved drugs and biologically active compounds, acquired using fluorescence microscopy and retained in multivariate compound profiles. Dissimilarities between compound profiles guided the identification of 5 combinations, which were assessed for qualitative interaction on TNBC cell growth. The combination of the microtubule-targeting drug vinblastine with KSP/Eg5 motor protein inhibitors monastrol or ispinesib showed potent synergism in 3 independent TNBC cell lines, which was not substantiated in normal fibroblasts. The synergistic interaction was mediated by an increase in mitotic arrest with cells demonstrating typical ispinesib-induced monopolar mitotic spindles, which translated into enhanced apoptosis induction. The antitumour activity of the combination vinblastine/ispinesib was confirmed in an orthotopic mouse model of TNBC. Compared to single drug treatment, combination treatment significantly reduced tumour growth without causing increased toxicity. Image-based profiling and analysis led to the rapid discovery of a drug combination effective against TNBC in vitro and in vivo , and has the potential to lead to the development of new therapeutic options in other hard-to-treat cancers. Insects rely heavily on olfaction to locate habitat, mates, and oviposition sites, while odorant molecules and the antennal olfactory sensory cells of insects are two indispensable components of olfactory response. Our previous work identified the sex pheromones and volatile compounds derived from host plant of Dendrolimus tabulaeformis Tsai et Liu, a serious economic pest of pines in northern China. However, little is known about the olfactory system of D. tabulaeformis, especially in females. To make a better understanding of the D. tabulaeformis olfactory response, we investigated the structure, innervation, numbers, and distribution of sensilla on the antennae of male and female moth, based on scanning and transmission electron microscopy. The pinniform antennae of this moth bear five types of sensilla: trichoid (TS), basiconica (BS), styloconic (StS), and chaetica (ChS) sensilla, and belt-type structures (BTSs). The BTSs are hollow and are not chemical sensilla. The ChSs occur on the scape. StS contain dendrite sheaths and four dendrites containing microtubules in their cores. The BS contents are continuous with that in the subsegments. The TSs were sexually dimorphic and could be divided into 12 subtypes based on dendrite number and form. TS1-4 contained two, three, or four dendrites and were similar in both sexes. TS 5-8 and TS 9-12 occurred only on male and female antenna, respectively. The p12 subunit of polymerase delta (Pol δ) is degraded in response to DNA damage induced by UV, alkylating agents, oxidative, and replication stresses. This leads to the conversion of the Pol δ4 holoenzyme to the heterotrimer, Pol δ3. We review studies that establish that Pol δ3 formation is an event that could have a major impact on cellular processes in genomic surveillance, DNA replication, and DNA repair. p12 degradation is dependent on the apical ataxia telangiectasia and Rad3 related (ATR) kinase and is mediated by the ubiquitin-proteasome system. Pol δ3 exhibits properties of an 'antimutator' polymerase, suggesting that it could contribute to an increased surveillance against mutagenesis, for example, when Pol δ carries out bypass synthesis past small base lesions that engage in spurious base pairing. Chromatin immunoprecipitation analysis and examination of the spatiotemporal recruitment of Pol δ to sites of DNA damage show that Pol δ3 is the primary form of Pol δ associated with cyclobutane pyrimidine dimer lesions and therefore should be considered as the operative form of Pol δ engaged in DNA repair. We propose a model for the switching of Pol δ with translesion polymerases, incorporating the salient features of the recently determined structure of monoubiquitinated proliferating cell nuclear antigen and emphasizing the role of Pol δ3. Because of the critical role of Pol δ activity in DNA replication and repair, the formation of Pol δ3 in response to DNA damage opens the prospect that pleiotropic effects may ensue. In this article, we described the ultrastructure of the sensilla on the antenna, eyes, mouthparts, wings, legs, and external genitalia of female and male Trichogramma dendrolimi using scanning electron microscopy (SEM). The antenna possessed the most sensilla types. We found 13 types of sensilla on female antenna, which were trichoid sensilla (TS) type 1-4, chaetica sensilla (ChS) type 1-2, campaniform sensilla (CaS), falcate sensilla, placoid sensilla (PS) type 1-2, basiconic capitate peg sensilla (BCPS) type 1, coeloconic sensilla (CoS), and styloconic sensilla. Ten types of sensilla were found on the male antenna, some were the same as that on female T. dendrolimi antenna, such as TS types 1 and 3, CaS, PS type 1, BCPS type 1, and CoS, but TS types 5 and 6, ChS type 3, and BCPS 2 were specific to male T. dendrolimi antenna. The leg possesses eight types of sensilla and a kind of tympana structure. Four types of TSs were found on the wings. On the mouthparts, sensilla on the maxillary and labial palps were unique, including two TSs and one ChS. The ovipositor possesses three types of sensilla, and the copulatory organ possesses two types. The eyes had only one kind of TS. Furthermore, external morphology of antenna and external genitalia revealed distinct sexual dimorphisms. According to their morphology, the possible functions of these sensilla were discussed. Red yeasts hold great promise in the production of microbial lipids and carotenoids. Genetic study of red yeasts has attracted much attention; however, rapid amplification of genes from red yeast samples remains technically challenging. Here a highly efficient method for the preparation of genomic DNA (gDNA) template, which could be directly used for PCR, was developed. Cells from colonies or liquid cultures were collected and sequentially treated by microwave, plMAN5C, proteinase K and boiling (MMPB) in a single tube to give cell lysates that were qualified as PCR templates. Single-copied gDNA fragments o up to 2.8 kb were successfully amplified. We also demonstrated successful application of this method for species in the Ascomycetes and Basidiomycetes and identification of two leucine auxotroph mutants of Rhodotorula glutinis. Oleaginous yeast Rhodosporidium toruloides is an excellent microbial lipid producer of great industrial potential, yet there is no effective genetic tool for rationally engineering this microorganism. To develop a marker recycling system, the orotidine-5′-monophosphate (OMP) decarboxylase gene of R. toruloides ( RtURA3) was isolated using methods of degenerate polymerase chain reaction (PCR) together with rapid amplification of cDNA ends. The results showed that RtURA3 contains four extrons and three introns, and that the encoded polypeptide holds a sequence of 279 amino acid residues with significant homology to those of OMP decarboxylases from other yeasts. A shuttle vector pYES2/CT- RtURA3 was constructed via site-specific insertion of RtURA3 into the commercial vector pYES2/CT. Transformation of the shuttle vector into Saccharomyces cerevisiae BY4741, a URA3-deficient yeast strain, ensured the viability of the strain on synthetic dextrose agar plate without uracil, suggesting that the isolated RtURA3 was functionally equivalent to the URA3 gene from S. cerevisiae. Worldwide, pine forests have been threatened by a devastating pine wood disease caused by Bursaphelenchus xylophilus, with Monochamus saltuarius being a newly recorded vector of the disease in Northeast China. The olfactory system plays important roles in both feeding and oviposition during the adult stage of M. saltuarius, and olfactory gene function research is essential for gaining an understanding of the olfactory mechanisms of this pest. However, there is limited information available regarding olfactory gene functions in this pest. In the present study, we selected 7 olfactory genes, including 2 chemosensory proteins, 2 odorant‐binding proteins, the odorant co‐receptor and 2 odorant receptors, which were relatively highly expressed during the adult stage. We silenced these genes by RNA interference (RNAi), and real‐time quantitative PCR was used to detect their expression levels after double‐stranded RNA (dsRNA) injection. The results indicate that these genes were significantly downregulated at 2 d post‐dsRNA injection, and this was sustained until 5 d post‐injection. Electroantennography tests indicated that the knockdown of MsalOBP14 and MsalOrco impaired the olfactory response of M. saltuarius to 11 host volatiles and 1 sex pheromone compound. Y‐tube experiments further confirmed that downregulated MsalOBP14 and MsalOrco expression led to olfactory dysfunction in M. saltuarius, which significantly lost selectivity. The results indicate that MsalOBP14 and MsalOrco play critical roles in sex communication and host volatile detection in M. saltuarius, and possibly represent 2 effective targets for controlling this forest pest through olfactory disruption.
