A new human adenovirus has been isolated from patients with keratoconjunctivitis and/or genital infection since 1976. This adenovirus, designed candidate adenovirus 37 (Ad 37) is serologically distinct but related to Ad 10, 13, 19, and 30 (see the accompanying paper by de Jong et al). SDS-Polyacrylamide gel electrophoresis of Ad 37 virion polypeptides showed that this adenovirus is a member of subgroup D. DNA restriction endonuclease analysis of DNA from Ad 37 and related serotypes belonging to subgroup D showed that Ad 37 is a new genome type belonging to subgroup D but clearly distinct from the 20 serotypes classified into this subgroup.

Nonionic detergent solubilised polypeptides from highly purified human cytomegalovirus virions were used as immunogens to produce murine monoclonal antibody secreting hybridomas. One monoclonal antibody was shown, by immunoprecipitation followed by SDS-polyacrylamide gel electrophoresis (SDS-PAGE), to precipitate three glycoproteins with molecular weights 52, 95, and 130 (all X 10(3)) and one minor component with a molecular weight of 50 X 10(3). When virion envelope components were first separated by SDS-PAGE and electrophoretically transferred to nitrocellulose membranes, this monoclonal antibody recognised two related components with molecular weights 50 and 52 (both X 10(3)). Immunofluorescence studies suggested that these viral antigens were associated with membrane systems of virus-infected cells and were particularly abundant late in infection.

The polypeptide composition of the lymphoproliferative disease virus (LPDV) of turkeys was shown to comprise several polypeptides with apparent molecular weights of 76, 31, 28, 20 and 15 kDa. This polypeptide pattern is distinctly different from the protein profiles of avian leukosis viruses, reticuloendotheliosis virus, or murine leukemia viruses. Moreover, LPD virions contain 2 major structural proteins (p31 and p28), in contrast to only one major internal protein present in most other retroviruses. The 76 kDa protein was established as the major viral envelope glycoprotein. The uniqueness of the LPDV polypeptide pattern is consistent with the lack of genetic relatedness of LPDV genome to other retroviruses, establishing LPDV as a representative of a distinct group of retroviridae.

Midway virus, a new tick-borne virus isolated from two species of Ornithodoros (Alectorobius) ticks of the capensis group (O capensis, O denmarki), is described from Midway, Kure, and Manana islands in the Central Pacific (Hawaiian Archipelago) and from northern Honshu (Japan). Midway virion is enveloped, unusually large, acid and temperature sensitive, and its type of nucleic acid is RNA. Complement-fixation (CF) tests show a close relation of Midway to Nyamanini virus, which has been isolated from ardeid birds and Argas ticks in Africa, the Indian subcontinent, and Southeastern Asia. However, cross-box tests (CF, mouse and tissue culture neutralization, immunofluorescence) show that these two viruses are quite distinct. Midway virus is lethal for newborn Swiss mice inoculated by intracerebral, but not intraperitoneal route. It fails to kill four-week-old mice by either route. Midway virus causes cytopathic effects in BHK-21 cells and titerable plaques in Vero cells. Antibodies to it were prevalent among nestlings of Larus crassirostris (Black-tailed Gull) on Aomatsushima I., but were scarce among those of Nycticorax nycticorax (Black-crowned Night Heron) of the same island.

In order to establish the role of human papillomavirus (HPV) in carcinogenesis of epidermodysplasia verruciformis (EV), the presence, the molecular characteristics and the physical state of HPV DNA in a benign lesion, a primary carcinoma and a metastatic carcinoma developing in the same EV patient were studied and compared. Of the 2 HPV DNAs isolated from benign macular lesions, only one (a subtype of HPV 5) was detected in both primary and metastatic tumors. Only one normal species of viral DNA molecule was detected in the benign lesion, whereas most, if not all, viral DNA molecules present in the carcinoma (both primary and metastatic) were aberrant ones. The major viral DNA molecule in the primary carcinoma was a large HPV DNA with duplicated 40% subgenomic segments, and was present as free episomes. The major viral DNA molecule in the metastatic carcinoma was the 40% subgenomic segment itself, lacking the remaining 60% segment of the viral genome, and was integrated within cellular DNA. Thus, HPV DNA was present in tumors at any stage of malignancy, and its molecular characteristics and physical state changed not only with the development but also with the enhancement of malignancy, consistently conserving its defined 40% subgenomic segment as the predominant viral sequences. Our results suggest that HPV 5 may be actually involved in carcinogenesis in EV patients.

The structural proteins in purified preparations of variola, monkeypox, and vaccinia viruses were separated and compared by using a high resolution SDS-polyacrylamide gel electrophoresis system. About 30 proteins were resolved for each virus by autoradiography of longitudinally-sliced gel rods. Although the autoradioelectropherograms of each virus were similar, it was possible to differentiate them by their unique protein pattern in the 30,000 to 40,000 molecular weight region of the gels. A single virion glycoprotein (mol. wt. = 38 X 10(3)) and a virion phosphoprotein (mol. wt. = 12 X 10(3)) were associated with each of the virus preparations. Cross-absorbed monospecific immune sera against variola, monkeypox, and vaccinia virus-infected cells were used in immunodiffusion tests to precipitate radiolabeled, homologous, soluble antigen proteins. The predominant antigen protein associated with each immunospecific precipitate had a molecular weight of approximately 73,000.
