[Purpose/Significance] Many scholars have studied the influencing factors of mobile government users' usage intention. Reviewing a large amount of literature, we found that the existing empirical research results in this field are inconsistent. Therefore, we need to clarify further the primary factors and moderating variables that affect mobile government users' willingness and reveal the specific reasons for the differences in results. Finally, we will try to obtain the universal conclusion and provide a reference for improving the relevant theories and researches on user information behavior. [Method/Process] We selected 8 antecedent variables and 4 moderating variables affecting users' usage intention, 45 Chinese and foreign literatures, 27 795 samples, and 139 independent effect values in the meta-analysis. [Result/Conclusion] Performance expectation (r=0.586), effort expectation (r=0.538), trust (r=0.590), social influence (r=0.549), safety (r=0.489), compatibility (r=0.645), quality of service (r=0.588), and information quality (r=0.549) have significant positive effects on the usage intention of mobile government users. Group category, education level, social culture, and mobile government category moderate the relationship between the above 8 variables and mobile government users' usage intention.

Currently, complex scene classification strategies are limited to high-definition image scene sets, and low-quality scene sets are overlooked. Although a few studies have focused on artificially noisy images or specific image sets, none have involved actual low-resolution scene images. Therefore, designing classification models around practicality is of paramount importance. To solve the above problems, this paper proposes a two-stage classification optimization algorithm model based on MPSO, thus achieving high-precision classification of low-quality scene images. Firstly, to verify the rationality of the proposed model, three groups of internationally recognized scene datasets were used to conduct comparative experiments with the proposed model and 21 existing methods. It was found that the proposed model performs better, especially in the 15-scene dataset, with 1.54% higher accuracy than the best existing method ResNet-ELM. Secondly, to prove the necessity of the pre-reconstruction stage of the proposed model, the same classification architecture was used to conduct comparative experiments between the proposed reconstruction method and six existing preprocessing methods on the seven self-built low-quality news scene frames. The results show that the proposed model has a higher improvement rate for outdoor scenes. Finally, to test the application potential of the proposed model in outdoor environments, an adaptive test experiment was conducted on the two self-built scene sets affected by lighting and weather. A deep learning‐powered VS approach combined with two free docking programs are proposed and evaluated for screening an ultra‐large compound library to obtain diverse potential active compounds rapidly and efficiently. We found that it is a practical and transferable strategy to significantly reduce computational cost. Background: Molecular docking‐based virtual screening (VS) aims to choose ligands with potential pharmacological activities from millions or even billions of molecules. This process could significantly cut down the number of compounds that need to be experimentally tested. However, during the docking calculation, many molecules have low affinity for a particular protein target, which waste a lot of computational resources. Methods: We implemented a fast and practical molecular screening approach called DL‐DockVS (deep learning dock virtual screening) by using deep learning models (regression and classification models) to learn the outcomes of pipelined docking programs step‐by‐step. Results: In this study, we showed that this approach could successfully weed out compounds with poor docking scores while keeping compounds with potentially high docking scores against 10 DUD‐E protein targets. A self‐built dataset of about 1.9 million molecules was used to further verify DL‐DockVS, yielding good results in terms of recall rate, active compounds enrichment factor and runtime speed. Conclusions: We comprehensively evaluate the practicality and effectiveness of DL‐DockVS against 10 protein targets. Due to the improvements of runtime and maintained success rate, it would be a useful and promising approach to screen ultra‐large compound libraries in the age of big data. It is also very convenient for researchers to make a well‐trained model of one specific target for predicting other chemical libraries and high docking‐score molecules without docking computation again. The delivery of targeted family education guidance services is essential for establishing a three-in-one education model that encompasses families, schools, and communities. It is also the key to creating a favorable educational environment that promotes the comprehensive and healthy development of individual students. To gain a comprehensive understanding of family education characteristics and trends in Chinese counties, accurately tap into the educational needs of parents, and effectively identify strengths and weaknesses in family education guidance services, this paper employed a quantitative research method based on a questionnaire survey and interview data from J district of C city after years of continuous monitoring of the quality of family education in the area, aiming to explore these issues and put forward several suggestions for improvement. The results indicate that the family learning environment and atmosphere in the district are generally favorable, while parental educational competence requires further improvement. Parents express a strong desire for interactive, systematic, and fragmented family education guidance from schools. Most parents acknowledge the significance of receiving guidance on family education, yet they are disinclined to actively engage in learning-related activities. While the operational mechanisms of county-level family education guidance services have been continuously improved, there is a need for greater overall coordination. Furthermore, the potential of school-led family education guidance services has yet to be fully tapped, and the scientific and professional standards of such guidance need to be further enhanced. Based on the aforementioned findings, we propose recommendations for improvement. Using J district's effective improvement practices in management, curriculum, and teams as an example, we intend to explore practical ways to promote county-level family education guidance services and provide a valuable reference for the establishment of a comprehensive and effective system to improve such services. Location and properties of ligand binding sites provide important information to uncover protein functions and to direct structure-based drug design approaches. However, as binding site detection depends on the three-dimensional (3D) structural data of proteins, functional analysis based on protein ligand binding sites is formidable for proteins without structural information. Recent developments in protein structure prediction and the 3D structures built by AlphaFold provide an unprecedented opportunity for analyzing ligand binding sites in human proteins. Here, we constructed the CavitySpace database, the first pocket library for all the proteins in the human proteome, using a widely-applied ligand binding site detection program CAVITY. Our analysis showed that known ligand binding sites could be well recovered. We grouped the predicted binding sites according to their similarity which can be used in protein function prediction and drug repurposing studies. Novel binding sites in highly reliable predicted structure regions provide new opportunities for drug discovery. Intermediate neural progenitors (INPs) boost the number and diversity of neurons generated from neural stem cells (NSCs) by undergoing transient proliferation. In the developing Drosophila brains, INPs are generated from type II neuroblasts (NBs). In order to maintain type II NB identity and their capability to produce INPs, the proneural protein Asense (Ase) needs to be silenced by the Ets transcription factor pointed P1 (PntP1), a master regulator of type II NB development. However, the molecular mechanisms underlying the PntP1-mediated suppression of Ase is still unclear. In this study, we utilized genetic and molecular approaches to determine the transcriptional property of PntP1 and identify the direct downstream effector of PntP1 and the cis-DNA elements that mediate the suppression of ase. Our results demonstrate that PntP1 directly activates the expression of the transcriptional repressor, Tailless (Tll), by binding to seven Ets-binding sites, and Tll in turn suppresses the expression of Ase in type II NBs by binding to two hexameric core half-site motifs. We further show that Tll provides positive feedback to maintain the expression of PntP1 and the identity of type II NBs. Thus, our study identifies a novel direct target of PntP1 and reveals mechanistic details of the specification and maintenance of the type II NB identity by PntP1. The key to the generation of INPs from type II NBs is the suppression of proneural protein Asense (Ase) in type II NBs by the Ets family transcription factor Pointed P1 (PntP1), but how PntP1 suppresses Ase expression remains unclear. In this study, we provided evidence to demonstrate that PntP1 directly activates the orphan nuclear receptor Tailless (Tll), which in turn suppresses Ase expression to maintain the capability of type II NBs to produce INPs. Meanwhile, Tll provides positive feedback to maintain the expression of PntP1 and type II NB identity. We further identified seven PntP1 binding sites in the tll enhancer regions and two Tll binding sites in the ase regulatory regions that mediate the activation of tll and the suppression of ase, respectively. Large scale fire tests of building external wall insulation system were conducted. In the experiment, thermal-couples were mounted to measure the insulation system surface temperature and the gas temperature inside rooms at the second and third floors. Photos were also taken during the fire tests. The measurement provides information of the ignition and fire spread of the external insulation system which consists of surface protection layer, glass fibre net, bonding thin layer, anchor and the load bearing wall. Comprehensive simulations of the fire tests were carried out using an advanced CFD fire simulation software Simtec (Simulation of Thermal Engineering Complex) [1, 2], which is now released by Simtec Soft Sweden, with the turbulent flow, turbulent combustion, thermal radiation, soot formation, convective heat transfer, the fully coupled three dimensional heat transfer inside solid materials, the ‘burn-out' of the surface protection layer and the pyrolysis of the insulation layer, etc, all computed. The simulation is compared with experimental measurement for validation. The simulation well captured the burning and fire spread of the external insulation wall.

The anti-oxidative enzyme, glutathione peroxidase 4 (GPX4), helps to promote inflammation resolution by eliminating oxidative species produced by the arachidonic acid (AA) metabolic network. Up-regulating its activity has been proposed as a promising strategy for inflammation intervention. In the present study, we aimed to study the effect of GPX4 activator on the AA metabolic network and inflammation related pathways. Using combined computational and experimental screen, we identified a novel compound that can activate the enzyme activity of GPX4 by more than two folds. We further assessed its potential in a series of cellular assays where GPX4 was demonstrated to play a regulatory role. We are able to show that GPX4 activation suppressed inflammatory conditions such as oxidation of AA and NF-κB pathway activation. We further demonstrated that this GPX4 activator can decrease the intracellular ROS level and suppress ferroptosis. Our study suggests that GPX4 activators can be developed as anti-inflammatory or cyto-protective agent in lipid-peroxidation-mediated diseases. [ABSTRACT FROM AUTHOR]

Various substituted thiazolidin-2-ones were synthesized from the corresponding thiazolidine-2-thiones with bromoethanol in ethanol with sodium ethoxide as a base. The optimal reaction conditions and mechanism were reinvestigated in detail. The bioassay indicated that (S)-4-isobutyl and (S)-4-benzylthiazolidin-2-ones show certain inhibitive activities against Candida albicans and Escherichia coli. Summary Dendrite pruning of Drosophila sensory neurons during metamorphosis is induced by the steroid hormone ecdysone through a transcriptional program. In addition, ecdysone activates the eukaryotic initiation factor 4E-binding protein (4E-BP) to inhibit cap-dependent translation initiation. To uncover how efficient translation of ecdysone targets is achieved under these conditions, we assessed the requirements for translation initiation factors during dendrite pruning. We found that the canonical cap-binding complex eIF4F is dispensable for dendrite pruning, but the eIF3 complex and the helicase eIF4A are required, indicating that differential translation initiation mechanisms are operating during dendrite pruning. eIF4A and eIF3 are stringently required for translation of the ecdysone target Mical, and this depends on the 5′ UTR of Mical mRNA. Functional analyses indicate that eIF4A regulates eIF3-mRNA interactions in a helicase-dependent manner. We propose that an eIF3-eIF4A-dependent alternative initiation pathway bypasses 4E-BP to ensure adequate translation of ecdysone-induced genes. In this work, microwave-assisted hydrothermal process was applied to modify the municipal solid waste incineration (MSWI) fly ash and get zeolitic production. The condition of hydrothermal process was fixed at 200 °C and 30 min. The 1 mol/L Na 2 HPO 4 was selected as the additive, and the liquid to solid ratio was 3 ml/g. X-ray diffraction results revealed that analogous zeolite crystals formed by the MSWI fly ash during the hydrothermal process. The cation exchange capacity (CEC) was 0.498 meq/g of the modified fly ash, which increased about 22 times compared to the raw MSWI fly ash. To obtain the adsorption mechanism of the modified fly ash, the experiments of adsorption isotherm and kinetics were researched. The adsorption isotherms of heavy metal cations in mixed solution could be described by Langmuir isotherm equations, with a high correlation coefficient value. The experimental products were better fitted by the pseudo second-order kinetics rather than the pseudo first-order kinetics. Although the adsorption capacity of the modified fly ash needs to be promoted, it is a potential utilization for MSWI fly ash to be the original materials as adsorbents. [ABSTRACT FROM AUTHOR]

Abstract: A series of 6-nitro-3-(m-tolylamino) benzo[d]isothiazole 1,1-dioxide analogues were synthesized and evaluated for their inhibition activity against 5-lipoxygenase (5-LOX) and microsomal prostaglandin E2 synthase (mPGES-1). These compounds can inhibit both enzymes with IC50 values ranging from 0.15 to 23.6μM. One of the most potential compounds, 3g, inhibits 5-LOX and mPGES-1 with IC50 values of 0.6μM, 2.1μM, respectively. [Copyright &y& Elsevier]
