Cactus pear plant (Opuntia ficus-indica (L.) Mill.) is a type of cactus with a high tolerance to heat and drought due to the geographical and climatic conditions in which it grows. The fruits and decaying cladodes of the cactus pear plant provide a suitable environment for yeasts and other microorganisms to grow. Yeast cells accumulate trehalose and glycogen as a stress metabolite under stress conditions. Our study, it was aimed to isolate stress-tolerant yeast species from cactus pear fruit and identify them with molecular methods, and determine their storage carbohydrate accumulations. Sequence analysis of the 26S rDNA-D1/D2 gene region was used for molecular identification of thermotolerant and osmotolerant yeast strains. Trehalose and glycogen accumulations of stress-tolerant yeast strains were determined enzymatically under stress conditions. All isolated strains belonging to K. marxianus, K. lactis and P. kudriavzevii yeast species were determined as thermotolerant and osmotolerant. It was determined that K. marxianus and K. lactis yeast species preferred glycogen as a stress metabolite while P. kudriavzevii yeast species accumulated trehalose. Stress tolerance and the type and amount of reserve carbohydrate were found to be various among the other isolated yeast strains of A. pullulans, H. opuntiae and P. kluyveri depending on the degree and type of stress. It has been evaluated that the isolated K. marxianus and K. lactis yeast strains have the potential to be used in the dairy industry, while the P. kudriavzevii yeast strains are suitable for use in the bioethanol and fermentation industry., Kaktüs armut bitkisi (Opuntia ficus-indica (L.) Mill.) yetiştiği coğrafya ve iklim koşulları nedeniyle sıcaklığa ve kuraklığa toleransı oldukça yüksek olan bir kaktüs türüdür. Bu kaktüs türünün bozulmaya başlayan kladotları ve meyveleri mayaların ve diğer mikroorganizmaların çoğalabilmesi için uygun alan oluşturmaktadır. Çalışmamızda kaktüs armut meyvesinden stres dirençli maya türlerinin izole edilerek moleküler yöntemlerle tanımlanması ve depo karbonhidrat birikimlerinin belirlenmesi amaçlandı. Termotolerant ve osmotolerant maya suşlarının moleküler tanımlanmasında 26S rDNA-D1/D2 gen bölgesinin dizi analizi kullanıldı. Stres dirençli maya suşlarının trehaloz ve glikojen birikimleri stres koşullarında enzimatik olarak belirlendi. İzole edilen K. marxianus, K. lactis ve P. kudriavzevii maya türlerine ait tüm maya suşlarının termotolerant ve osmotolerant maya suşları olduğu gözlendi. K. marxianus ve K. lactis maya türünde stres metaboliti olarak glikojenin P. kudriavzevii maya türünde ise trehalozun tercih edildiği belirlendi. Diğer izole edilen A. pullulans, H. opuntiae ve P. kluyveri türlerine ait maya suşları arasında stres toleransının ve depo karbonhidratın stresin derecesine ve çeşidine bağlı olarak değiştiği tespit edildi. İzole edilen K. marxianus ve K. lactis maya suşlarının süt ürünleri endüstrisinde, P. kudriavzevii maya suşlarının ise biyoetanol ve fermentasyon endüstrisinde kullanımı için uygun potansiyele sahip oldukları değerlendirildi.

Transit authorities have been searching for the indicators to measure transit service quality and the key factors to attract citizens who do not prefer public transport. The recent advent of data collection technologies such as AVL, APC, GPS and Smart Card (SC) promise opportunities for conducting comprehensive transit system performance measures, improving the quality of service while meeting passenger needs and reducing operation costs. The aim of this study is to propose metrics to improve transit service. The focus is on bus transportation since it is more flexible compared to rail transportation and widely preferred by the masses in cities. The primary data source of this study comes from the Department of Transportation for the City of Antalya. We load the complete boarding data of December 18,2019 which is a standard weekday. Most of the analysis is done using Knime software. As an outcome of this research, the analysis may propose to modify or eliminate inefficient routes, suggest new lines, identify inefficient bus stops, and potentially modify path of a route. Maya türlerinin tanımlanmasında kullanılan geleneksel tanımlama yöntemleri ve kit sistemlerinin yanında moleküler tanımlama yöntemleri de kullanılmaktadır. Araştırma kapsamında nar meyvesinin yüzey maya florası ile ahududu meyvesinin barındırdığı maya florası maya suşlarının morfolojik özellikleri, API ID32C kit sistemi ve moleküler identifikasyon yöntemleri kullanılarak belirlenmesi ve endüstriyel öneme sahip enzim üretebilen maya türlerinin tespit edilmesi planlandı. Maya suşlarının genomik DNA izolasyonlarının ardından ITS1-5.8S rDNA-ITS2 bölgesi ITS1 ve ITS4 evrensel primerleri kullanılarak PCR yöntemiyle çoğaltıldı. PCR ürünlerine göre oluşturulan gruplara ait maya suşlarının ITS1-5.8S rDNA-ITS2 bölgeleri beş farklı restriksiyon enzimi (HinfI, HaeIII, MspI, AluI ve HhaI) kullanılarak kesimleri yapıldı. PCR grupları içinde farklılık gösteren maya suşları seçilerek DNA dizi analizi yapıldı ve BLAST programı kullanılarak maya türleri tanımlandı. Tanımlanan türlerin hücre dışı enzim aktiviteleri API-ZYM test sistemi kullanarak belirlendi. Çalışmamızın sonucunda, maya suşlarının DNA dizi analizlerine göre nar meyvesi yüzeyinden izole edilen maya suşları Metschnikowia pulcherrima, Cryptococcus bestiolae, Aureobasidium pullulans, Candida zeylanoides, Hanseniaspora uvarum, Kluyveromyces lactis ve Metschnikowia sp. türleriyle, ahududu meyvesinden izole edilen maya suşları ise M. pulcherrima, A. pullulans, Candida sp., H. uvarum ve Metschnikowia sp. türleriyle yüksek homoloji gösterdi. API-ZYM test sistemi sonucuna göre nar meyvesinden izole edilen maya türlerinin alkalin fosfataz, lösin arilamidaz, esteraz (C 4), esteraz lipaz (C 8), asit fosfataz, naftol-AS-BI-fosfohidrolaz enzimlerinin; ahududu meyvesinden izole edilen maya türlerinde ise Lösin arilamidaz ve β- Glukosidaz enzimlerinin yüksek aktivite gösterdiği belirlendi. Moleculer identification methods beside the conventional identification methods and kit systems using for identification of yeast species have also been using recently. As a part of research, determination of yeast flora on pomegranate surface and raspberry fruits were planned using morphologic features of yeast strains, API ID32C kit system and molecular identification methods and determination of yeast species that can produce industrial importance enzymes were aimed. After genomic DNA isolation from yeast strains, ITS1-5.8S rDNA-ITS2 gene regions were amplified by PCR method using ITS1 and ITS4 universal primer. The groups obtained from the size of PCR products that belongs to yeast strains were cut by using restriction enzymes (HinfI, HaeIII, MspI, AluI ve HhaI). The yeast strains which differ in PCR groups were selected as restriction profile and DNA sequence analysis were carried out and yeast strains were identified using DNA sequences by BLAST program. Extracellular enzyme activities were determined of identified yeast species by API-ZYM test system. Consequence of my study, according to DNA sequence analysis of the pomegranate yeast strains showed high homology with Metschnikowia pulcherrima, Cryptococcus bestiolae, Aureobasidium pullulans, Candida zeylanoides, Hanseniaspora uvarum, Kluyveromyces lactis and Metschnikowia sp. species. Raspberry yeast strains showed high homology M. pulcherrima, A. pullulans, Candida sp. H. uvarum ve Metschnikowia sp. species. According to API-ZYM results, high enzyme activity were determined for pomegranate in alkaline phosphatase, leucine arylamidase, esterase (C 4), esterase lipase (C 8), acid phosphatase, naftol-AS-BI-fosfohidrolase enzymes, were determinated for raspberry yeast strains in leucine arylamydase and β- glukosidase enzymes. 140

Farklı kaynaklardan elde edilen hedef bilgilerindeki bilinmeyen korelasyonun ele alınması tutarlı bir iz füzyonu yapılması açısından önemli bir problemdir. Chernoff birleştirme tekniği bu tutarlılığı sağlamak adına önerilen popüler yöntemlerden biridir. Bu yöntem girdi fonksiyonlarının üstel olarak ağırlıklandırılmasına dayanmakta ve Gauss dağılımlı fonksiyonlar için analitik çözüm önermektedir. Bu tezde Gauss Karışımlı fonksiyonlar için Chernoff birleştirme yönteminin geliştirilmesi ve tutarlı ve gürbüz sonuçlar elde edilmesine yönelik çözüm önerilmektedir. Önerilen teknik Gauss karışım fonksiyonunun yaklaşık üstel değerini bulmayı gerektirir. Üstel değer bulma işlemi girdi fonksiyonlara sigma nokta yaklaştırımı uygulanması ile sağlanmaktadır. Sonuçta, Gauss karışımlı fonksiyonlar için kapalı formda bir maliyet fonksiyonu üretilmekte ve füzyon sonucu yeni bir Gauss karışımı olarak elde edilmektedir. Önerilen yöntemin etkinliği basit ve aydınlatıcı örneklerde gösterilmiştir. Bu örneklerde olasılık yoğunluk fonksiyonlarının bileştirilmesi problemi ele alınmış ve önerilen yöntem optimal çözüm ve nümerik Chernoff birleştirme çözümleri ile kıyaslanmıştır. Bu teknik, hedef izlemede güncel bir problem olan IMM süzgeci içeren füzyon mimarilerinin çıktılarının birleştirilmesi amacı ile kullanılmıştır. Diğer bir güncel problem olan PHD filtresi içeren füzyon mimarilerinin çıktılarının birleştirilmesi problemi için de aynı yöntem kullanılmıştır. Sonuçlar gerek değişik füzyon mimarileri için gerekse, olabildiği durumlarda, değişik füzyon yöntemleri için karşılaştırılmıştır. Handling of unknown correlation in the target information obtained from different sources is an important problem for consistent track fusion. Chernoff fusion technique is one of the popular approaches which produce conservative fusion results to bring this consistency. This method is based on exponential scaling of the input functions and it provides an analytical solution when input functions are Gaussian densities. The thesis mainly discusses the extension of the Chernoff fusion method to Gaussian Mixtures in a consistent and robust way and proposes an approximate approach for the computation of the fused output. The exponential scaling, required for Chernoff fusion, is based on a sigma-point approximation of the underlying functions. The resulting general fusion rule yields a closed form problem formulation that gives the fused function as a Gaussian mixture. Effectiveness of the fusion method is presented for simple but illustrative density fusion problems and compared to the optimal solutions and exact numerical Chernoff fusion. The technique is applied to the IMM filter used in target tracking problems. The results show the effectiveness of the method. The second application of the method is to fuse the PHD filter outputs that are Gaussian Mixture intensities. PHD filters are again used in target tracking. Different fusion architectures are investigated and their results are compared with each other. The comparison is also made with other available methods whenever they are applicable. 175

Covariance Matrix in target tracking algorithms has a critical role at multi-sensor track fusion systems. This matrix reveals the uncertainty of state es-timates that are obtained from different sensors. So, many subproblems of track fusion usually utilize this matrix to get more accurate results. That is why this matrix should be interchanged between the nodes of the multi-sensor tracking system. This thesis mainly deals with analysis of approximations of the covariance matrix that can best represent this matrix in order to effectively transmit this matrix to the demanding site. Kullback-Leibler (KL) Distance is exploited to derive some of the representations for Gaussian case. Bu tez çalışması, Linux işletim sistemindeki veri mimarilerinin doğru kurulum, yapılandırma ve optimizasyon işlemlerini içerir. Çekirdek olarak alacağımız ve özelleştireceğimiz işletim sistemi, şu anda BAP (Bilimsel Araştırma Projeleri) tarafından desteklenen Milis Linux projesini taban alacaktır. Bu projede kullanılacak büyük veri platformu yaygın olarak kullanılan ve iyi bilinen Apache Hadoop dosya sistemini uyarlar. Apache Hadoop platformu, temel programlama çerçevesine sahip bilgisayar ve disk kümelerine büyük miktarda veri dağıtılmasını sağlar. Apache Hadoop'un üstüne topluluk tarafından işletilen veri işleme, güvenlik, erişim yönetimi, performans izleme ve diğer yardımcı yazılımlar kurulması bu tez kapsamında gerçeklenecektir. Kararlı bir Büyük Veri işleme yazılımı üretmek ve dağıtım yapmak için, yazılım paketlerinin en yeni ve uyumlu sürümlerinin bir araya getirilmesini sağlayan bir sistem kurulmalıdır. Bu tür yazılım seçim sürecinin geliştirilmesi ve otomasyonu ve birim testleriyle doğrulanması bu araştırmanın kapsamıdır. The thesis covers the proper installation, configuration and optimization processes of the big data architectures for Linux operating system. The operating system that we will take as a core and customize is based on MILIS Operating System which is currently supported by Akdeniz University Scientific Research Projects Coordination Unit (BAP). Data platform and file system that is planned to be used in this project is well known and widely adapted Apache Hadoop. Apache Hadoop platform enables large volumes of data to be distributed across cluster of computer and disks with a basic programming framework. On top of Apache Hadoop, community-run data processing, security, access management, performance monitoring and other utility software is installed. In order to produce a stable Big Data processing software and distribution, a system should be put in place that includes the latest and compatible versions of the software packages to be assembled together. Development and automation of such software selection process and verification through unit tests is part of this research. In this research, a system image that contains the widely used big data modules is prepared for installation on virtual/physical disks. 107

Bu tez çalışmasında endüstriyel soğuk hava depolarında oda veya depo bazlı enerji tüketiminin tahminlenebilmesi için gereken teorik ve pratik çalışmalar yer almaktadır. Tez çalışmaları aynı zamanda Türkiye Cumhuriyeti Sanayi ve Teknoloji Bakanlığı tarafından desteklenen bir ArGe Merkezi proje çalışmalarının bir kısmını kapsamaktadır. Merkezi sistem soğutma ile soğutulan endüstriyel soğuk hava depolarında genellikle oda veya depo bazında enerji ölçümü yapılmamaktadır. Oda veya depo bazlı enerji tüketim verileri olmaması operasyonel pek çok çalışmanın verimsiz şekilde yapılmasına sebep olmaktadır. Oda veya depo tüketimleri elde edilerek operasyonel işlerin veriye dayalı olacak şekilde yapılması mümkün hale gelebilir. Bu durumda tez çalışmasında detaylarıyla bahsedildiği üzere büyük oranda enerji tasarrufları yapılabilir. Bu kapsamda başlatılan ArGe Merkezi projesinin analiz çalışmaları gerçekleştirilmiştir. Analiz çalışmasının sonucunda merkezi sistem ile soğutulan bir tesisin oda veya depo tüketimlerinin elde edilmesi için en büyük bilinmezliğin evaporatör kapasiteleri olduğu anlaşılmıştır. Tez çalışmaları kapsamında makine öğrenmesi teknolojisi derinlemesine araştırılmış ve Feature Importance Scoring yöntemleri ile tesislerde bulunan oda veya depoların soğutucu komponenti olan evaporatör ünitelerinin kapasiteleri farklı algoritmalar kullanılarak tahminlenmiştir. Tezin yazılım geliştirme çalışmaları sonucunda kapasite tahminlemesi yapan yazılım modülü elde edilmiştir. Bu modül REST Api teknolojisi ile bir web servis hizmeti verecek şekilde tasarlanmıştır. Bu modülün ArGe Merkezi projesinde nasıl bir konumda yer alacağı da tez çalışmasında tartışma ve bulgular kısmında bir diagram olarak yer almaktadır.

Bu tez çalışması, masa tenisi sporunda oyun içerisinde atılan bir topun yörüngesinin en ekonomik yolla tahmin edilmesi için kullanılacak metod ve dataset bilgilerini içerir. Profesyonel/yarı profesyonel masa tenisi oyuncuları için en önemli sorunlardan biri, gelişmelerine katkıda bulunacak bir antrenör veya partner bulmaktır. Farklı oyun tarzlarına karşı beceriler geliştirmek için, bu tarzlardaki oyuncularla antrenman yapmak gerekmektedir. Diğer sporlara göre daha az popüler olması nedeniyle masa tenisi spor dalında bu fırsatı bulmak çok zordur. Bu sorunu çözmek için masa tenisi robotunun geliştirilmesi 1980'lerin sonundan beri yıllarca araştırma konusu olmuş ve bu alanda onlarca çalışma yapılmıştır. Tüm yöntemler için ortak sorun; topa iyi vuruş yapan masa tenisi robotu için uygun vuruş noktasını, raket yörüngesini, vuruş hızını ve açısını bulmaktır. Bu yöntemlerde ana hedef, doğru zamanda yörünge tahminini elde etmektir. Atak bir oyuncu için top uçuş süresi 200 ms'nin altında olabildiği için yörünge tahmini 100ms'nin altında bitmelidir. Farklı yöntemlerle yapılan çalışmalarda bu soruna kesin bir çözüm bulunamamış olması tahmin algoritmasının daha ekonomik ve zamana duyarlı olması gerekliliğini ortaya koymaktadır. Bu araştırma ile var olan çalışmalardaki metotlar incelenerek en ekonomik metot ve bu metodun üzerinde çalışacağı dataset yapısı kurgulanacaktır.

Therefore, it is challenge to evaluate and compare the research performance of individual researchers and departments within the university.In this study, a software has been developed to evaluate performance, support research and collaboration at Akdeniz University. The data for this study is obtained from the database of HR, Web of Science (Wos) and InCites. The data has been analyzed by data mining techniques and evaluated according to research areas. In this study, an original software for the evaluation of academic publication performance was implemented for the Higher Education Institutes. Research performance for an academic unit or staff may be accessible in real time. As publication performance is a good indicator of quality of research and impact, it can be used to determine the effectiveness of academicians and universities. 55

Bu tez kapsamında, Java Web teknolojileri kullanılarak web tabanlı esnek bilgisayar tabanlı değerlendirme sistemi geliştirilmiştir. Sistem tasarlanırken esnek olması ve mevcuttaki sistemlerin problemlerini giderecek şekilde olması sağlanmıştır. Geliştirilen sistem bir web sunucusuna konuşlandırıldıktan sonra otuz adet elektronik sınav Akdeniz Üniversitesi Mühendislik Fakültesi öğrencilerine uygulanmıştır. Sisteme eğitmenler tarafından eklenen sorular ve oluşturulan testler, madde cevap kuramı ilkelerine göre analiz edilmektedir. Bu analizlerin sonuçları eğitmene yansıtılarak, sorulmuş sorular hakkında geri dönüş sağlanmaktadır. Bu tez kapsamında referans seçilen programlamaya giriş dersinin soru bankası irdelenmiştir. Yapılan analizler sonucunda sınavlarda sorulan bazı soruların uygun olmadığı, bazılarının ise revize edilmesi gerektiği saptanmıştır. Araştırmacılar bir çok çalışmada bilgisayar tabanlı değerlendirme sistemlerinin kullanımının öğrencilerin sınav başarılarına dair olumsuz etkisinin olup olmadığını araştırmışlardır. Çoktan seçmeli soruların sorulduğu sınavların elektronik ortamda yapılmasının öğrencilerin sınav başarısına bir etkisinin olmadığı ortaya konsa da açık uçlu sorular için yapılmış bir çalışma bulunmamaktadır. Bu tez kapsamında Doğa Bilimleri (Natural Science) dersi bu karşılaştırmayı yapmak için referans ders olarak seçilmiştir. 2017-2018 güz döneminde dersin sınavları kağıt ortamında açık uçlu sorular üzerinden sınavlar yapılmıştır. 2018-2019 gün döneminde ise yine açık uçlu sorular ile elektronik ortamda sınavlar yapıldı. Ortalama sınav puanları üzerinden yapılan bağımsız student t-test analizine göre iki grup arasında anlamlı bir fark bulunmadığı anlaşılmıştır ($p$ = 0,441). Elde edilen bulgular ile elektronik sınav sistemi kullanımının açık uçlu sorulardan oluşan sınavlarda da öğrencilerin başarısına dair olumsuz bir etkisinin olmadığı kanıtlanmıştır.Elektronik sınavların yapıldığı ortamlar genellikle okulların bilgisayar laboratuvarlarıdır. Bu ortamlar ders verilmek için tasarlandığından dolayı, sınav esnasında öğrencilerin birbirlerinin ekranlarına bakmaları çok zor olmamaktadır. Bilgisayarların yerleşim planında değişiklik yapmak her zaman mümkün olmasa da sınavlardan sonra öğrenci cevaplarının birbirine olan benzerlikleri araştırılarak kopya çeken öğrencileri tespit etmek mümkündür. Geliştirilen sistem kullanılarak yapılan sınavlarda, Jaccard ve Kosinüs benzerlik analizi yöntemleri kopya çeken öğrencilerin tespitinde kullanılmıştır. Bu iki yöntem elde ettiği sonuçlar bakımından kıyaslanmıştır. Kosinüs benzerlik analizi yönteminin kullanımının, kopya tespiti konusunda Jaccard benzerlik yöntemine göre daha uygun olduğu yapılan çalışmalar sonucunda görülmüştür.Tüm bu yapılan çalışmaların sonunda, açık kaynak kodlu teknolojiler kullanılarak eğitim kurumlarının kullanımına uygun olarak tasarlanan ve tüm ölçme ve değerlendirme süreçlerini kapsayan, web ortamında çalışan bilgisayar tabanlı değerlendirme sistemi geliştirilmiştir. In the scope of this thesis, a web based flexible computer based assessment system was developed using Java web technologies. The system is designed to be flexible in order to eliminate the problems of existing systems. After the deployment on a web server of the developed system, thirty electronic exams were applied to the Akdeniz University Engineering Faculty students.The questions and tests added by the instructors to the system are analyzed according to the principles of item-response theory. The results of these analyses are presented the instructor and feedback are given about the questions. Within the scope of this thesis, introduction to programming course was selected as a reference course and its question bank was examined. According to the analysis results, it was determined that some questions were inappropriate for asking and some questions need to be revised.Researchers have investigated in many studies, whether the use of computer based assessment system has a negative impact on the student's exam success. Although it is proved that the exams in which multiple-choice questions are asked in the electronic environment have no effect on the student's exam success, there are no studies for open-ended questions. In the scope of this thesis, Natural Science course was selected as a reference course. In 2017-2018 fall semester, exams of this course were held in the paper environment with the open-ended questions. In 2018-2019 fall semester, exams were held in the electronic environment with the open-ended questions. According to the independent student t-test analysis performed on the average exam scores, there was no significant difference between the two groups ($p$=0,441). The findings of the study revealed that the use of the electronic exam system did not have a negative effect on student's success in the exams which consisted of open-ended questions.Electronic exams are usually held in computer laboratories of schools. This laboratories are designed to teach courses. Because of this, it is not difficult for students to look at each other's screens. Although it is not always possible to make changes in the settlement of the computers, it is possible to detect the students who are copying by examining the similarities of the student's answers after the exams. In the exams conducted using the developed system, the methods of Jaccard and Cosine similarity analysis were used to detect the students who were copying. These two methods were compared in terms of the results. It has been seen that the use of Cosine similarity analysis method is more suitable than the Jaccard similarity analysis method for copy detection.As a results of all these studies, by using open source technologies, a computer based assessment system designed in accordance with the use of educational institutions and covering all measurement and evaluation processes is developed. 61
