24. PLCL-TPU bi-layered artificial blood vessel with compliance matching to host vessel.

Abstract

Compliance mismatch between the artificial blood vessel and the host vessel leads to abnormal hemodynamics and is a major mechanical trigger of intimal hyperplasia. Efforts have been made to achieve higher compliance of artificial blood vessels. However, the preparation of artificial blood vessels with compliance matching to host vessels has not been realized. A bi-layered artificial blood vessel was successfully prepared by dip-coating and electrospinning composite method using poly(L-Lactide-co-caprolactone) (PLCL) and thermoplastic poly(ether urethane) (TPU). In the case of a certain wall thickness (200 μm), thickness ratios of the PLCL inner layer (dip-coating method) and TPU outer layer (electrospinning method) were controlled at 0:1, 1:9, 3:7, 5:5, 7:3, and 1:0 respectively and the compliance, radial tensile properties, burst pressure, and suture retention strength were investigated. Results showed compliance value of the artificial blood vessel decreased with the increase of the thickness ratio, which suggested the compliance of the bi-layered artificial blood vessel can be regulated by adjusting the ratio of the inner and outer layer thicknesses. In the six different artificial blood vessels, the one with thickness ratio of 1:9 not only had high compliance (8.768 ± 0.393%/100 mmHg) but also can guarantee the other mechanical properties, such as the radial breaking strength (6.333 ± 0.689 N/mm), burst pressure (534.473 ± 20.899 mmHg), and suture retention strength (300.773 ± 9.351 cN). The proposed artificial blood vessel preparation method is expected to achieve compliance matching with the host vessel. 40. Application of high-throughput drug sensitivity screening system in the treatment of non-small cell lung cancer with malignant pleural effusion.

Abstract

Objective · To explore the feasibility of individualized treatment of malignant pleural effusion (MPE) patients with nonsmall cell lung cancer (NSCLC) guided by in vitro high-throughput drug sensitivity screening system. Methods · Thirty NSCLC patients with MPE treated in Nanjing Chest Hospital from January 2019 to May 2020 were included. The tumor cells in MPE were collected and isolated, and primary cells were cultured. The ex vivo drug sensitivity of primary cells was assessed for 30 chemotherapeutical and molecular targeted drugs by using the high-throughput drug sensitivity screening system of automated digital fluorescence imager. The consistency between the drug sensitivity assay results and the clinical efficacy of patients was evaluated. Results · The ex vivo drug sensitivity results of primary tumor cells cultured from MPE revealed considerable heterogeneity in drug response. Most of the primary cells were sensitive to Epirubicin, Lobaplatin, Gemcitabine and Irinotecan. Clinical efficacy of 23 patients can be evaluated. For patients with previously untreated, epidermal growth factor receptor (EGFR) mutation-positive advanced NSCLC, the consistency between the efficacy of targeted therapy and the high-throughput drug sensitivity assay results was 44.4% (4/9). For the EGFR negative untreated patients and EGFR mutation-positive advanced NSCLC patients with acquired resistance to targeted treatment, the consistency between the therapeutic effect of chemotherapy and the heighthroughput drug sensitivity assay results was 85.7% (12/14). Conclusion · The high-throughput drug sensitivity assay based on MPE primary cell culture in NSCLC patients has a high consistency with the clinical efficacy of chemotherapy, which is helpful for decision-making of personalized chemotherapy. Abstract

With the rapid development of nuclear power industry, people are paying more and more attention to the acoustic environment of nuclear power plants. Apart from the main control room [1], the air conditioning noise of other plants in the nuclear power plant is basically calculated by designers manually or through empirical evaluation. However, different designers consider the system design noise from different starting points and perspectives, and the actual effect can only be tested in the application process. But at this time to adjust and modify the system, will bring serious losses. The three-dimensional layout design of nuclear power is the product of the comprehensive development of forward design of nuclear power and the high-speed advancement of the design work of three generations of nuclear power, such as HuaLong no.1 and AP1000, which has been widely promoted and applied. Based on the PDMS software of HVAC 3d layout design, a software for noise analysis and calculation of air conditioning system is developed. Designers can conduct noise checking while designing, so as to automate, intellectualize and visualize noise calculation, reduce manpower input and improve design efficiency. This paper will demonstrate it.
