UDP-glucuronic acid (UDP-GlcA) is the precursor of many plant cell wall polysaccharides and is required for production of seed mucilage. Following synthesis in the cytosol, it is transported into the lumen of the Golgi apparatus, where it is converted to UDP-galacturonic acid (UDP-GalA), UDP-arabinose, and UDP-xylose. To identify the Golgi-localized UDP-GlcA transporter, we screened Arabidopsis thaliana mutants in genes coding for putative nucleotide sugar transporters for altered seed mucilage, a structure rich in the GalA-containing polysaccharide rhamnogalacturonan I. As a result, we identified UUAT1, which encodes a Golgi-localized protein that transports UDP-GlcA and UDP-GalA in vitro. The seed coat of uuat1 mutants had less GalA, rhamnose, and xylose in the soluble mucilage, and the distal cell walls had decreased arabinan content. Cell walls of other organs and cells had lower arabinose levels in roots and pollen tubes, but no differences were observed in GalA or xylose contents. Furthermore, the GlcA content of glucuronoxylan in the stem was not affected in the mutant. Interestingly, the degree of homogalacturonan methylation increased in uuat1 These results suggest that this UDP-GlcA transporter plays a key role defining the seed mucilage sugar composition and that its absence produces pleiotropic effects in this component of the plant extracellular matrix.

International audience; Les plantes reviviscentes une solution au réchauffement climatique? La sécheresse se traduit par des pertes considérables dans le domaine de l'agronomie. Améliorer la production des cultures est un enjeu extrêmement important dans les décennies à venir. Les plantes reviviscentes constituent un excellent modèle pour l'étude des mécanismes moléculaires, cellulaires et physiologiques impliqués dans la tolérance à la sécheresse. Les plantes reviviscentes encore appelées « plantes de la résurrection » présentent la remarquable capacité de survivre à une déshydratation quasi-totale, la dessiccation. 9 milliards d'êtres humains en 2050… moins de terres agricoles disponibles… des ressources en eau limitées… les effets du réchauffement climatique… …..et si les plantes «de la résurrection » pouvaient contribuer à lutter contre la faim dans le monde ??? Objectifs: sélectionner et / ou produire des plantes d'intérêt agronomiques capables: d'atteindre une production élevée dans des conditions de manque d'eau modéré. d'améliorer la tolérance des plantes de grandes cultures dans les régions semi-arides d'Afrique. Les plantes des zones arides ou semi-désertiques ont développé d'étonnantes stratégies pour pallier au manque d'eau. (A) Les éphémèrophyes du Namaqualand Afrique du Sud. (B) Welvitchia mirabilis Désert du Namib. (C)Crassula columnaris Afrique du Sud. Adaptée à des conditions de sécheresse extrême, C.wilmsii peut survivre plusieurs mois dans un état totalement déshydraté: son métabolisme, (photosynthèse et respiration) s'arrête, la plante paraît « comme morte » (A). Aux premières pluies, cette plante reviviscente, comme son nom l'indique, va « revenir à la vie » (B). Craterostigma wilmsii: une petite plante sud africaine très surprenante !!
