Purpose: Few studies have reported the integrated characteristics of hepatitis B virus (HBV)-related hepatocellular carcinoma (HCC) after long-term antiviral therapy. This study aimed to investigate the HBV integration features in HBV-HCC patients who had undergone long-term antiviral therapy, evaluate their impact on clinical indicators, and analyze the potential mechanisms involved. Patients and Methods: We utilized genome-wide association study (GWAS) to analyze liver cancer tissues and detect the presence of HBV integration. Seventeen patients with HBV integration were included in the integration (Int) group, while the remaining five patients were included in the non-integration (N-int) group. Clinical indicators were regularly monitored and compared between the two groups. The characteristics of HBV integration patterns were analyzed, and differences between the groups were explored at the chromosome and genomic levels. Results: After long-term antiviral therapy, although the frequency of HBV integration in HBV-HCC was reduced, residual HBV integration still accelerated the development of HCC. It affected the diagnosis, treatment, and prognosis of patients. HBV integration events led to changes in chromosome structure, which were closely related to HCC. Novel fusion genes were detected at a high frequency and had the potential to be specific detection sites for HBV-HCC. Conclusion: HBV integration events are synergistically involved in the human genome and HBV, which can lead to chromosome structural instability, gene rearrangement events closely related to HCC production, and the formation of new specific fusion genes. An abrupt decrease in egg production and quality in laying hens can be brought on by the avian adenovirus known as the egg drop syndrome virus (EDSV). So far, there are no efficient commercial diagnostic approaches for EDSV. In the present study, we first purified two monoclonal antibodies (mAbs 5G4 and 6G6) specifically against the EDSV fibre protein. The mAb 5G4 was then employed as capture antibody. Additionally, the detection antibody 6G6 (HRP-6G6) was conjugated with horseradish peroxidase. Consequently, based on these two mAbs, we developed a sandwich enzyme-linked immunosorbent assay (ELISA) for EDSV detection. Specificity evaluation demonstrated that only EDSV gave a positive result while other avian viruses showed negative results. Sensitivity investigation showed that the limit of detection of EDSV was 102.9 TCID50/ml and the limit of detection of the purified His-Fiber was 5 ng/ml. Furthermore, the screening of samples from infected chickens showed that this method was very effective and could be applied to the detection of EDSV in infected samples. In summary, the sandwich enzyme-linked immunosorbent assay developed here provides an efficient, sensitive, and large-scale method for the specific diagnosis of EDSV. A sandwich ELISA was developed to detect EDSV using the mAbs 5G4 and HRP-6G6. The sandwich ELISA maintained high specificity and sensitivity. Marbofloxacin (MBF) was once widely used as a veterinary drug to control diseases in animals. MBF residues in animal food endanger human health. In the present study, an immunochromatographic strip assay (ICSA) utilizing a competitive principle was developed to rapidly detect MBF in beef samples. The 50% inhibitory concentration (IC50) and the limit of detection (LOD) of the ICSAs were 2.5 ng/mL and 0.5 ng/mL, respectively. The cross-reactivity (CR) of the MBF ICSAs to Ofloxacin (OFL), enrofloxacin (ENR), norfloxacin (NOR), and Ciprofloxacin (CIP) were 60.98%, 32.05%, 22.94%, and 23.58%, respectively. The CR for difloxacin (DIF) and sarafloxacin (SAR) was less than 0.1%. The recovery rates of MBF in spiked beef samples ranged from 82.0% to 90.4%. The intra-assay and interassay coefficients of variation (CVs) were below 10%. In addition, when the same authentic beef samples were detected in a side-by-side comparison between the ICSAs and HPLC‒MS, no statistically significant difference was observed. Therefore, the proposed ICSAs can be a useful tool for monitoring MBF residues in beef samples in a qualitative and quantitative manner. [ABSTRACT FROM AUTHOR]

Simple Summary: Bovine IgG1 Fc receptor (boFcγRIII) is a homologue to human FcγRIII (CD16) that has two extracellular Ig-like domains and can bind bovine IgG1 with medium–low affinity. We identified the Fc-binding site as well as its key residues for IgG1 binding using synthetic peptides, which is located in the second extracellular domain of boFcγRIII. It provides new insights for the IgG–Fcγ interaction and FcγR-targeting drugs development. The bovine IgG1 Fc receptor (boFcγRIII) is a homologue to human FcγRIII (CD16) that binds bovine IgGI with medium–low affinity. In order to identify the Fc-binding site on the bovine IgG1 Fc receptor (boFcγRIII), peptides derived from the second extracellular domain (EC2) of boFcγRIII were synthesized and conjugated with the carrier protein. With a Dot-blot assay, the ability of the peptides to bind bovine IgG1 was determined, and the IgG1-binding peptide was also identified via truncation and mutation. The minimal peptide AQRVVN corresponding to the sequence 98–103 of boFcγRIII bound bovine IgG1 in Dot-blot, suggesting that it represents a linear ligand-binding site located in the putative A–B loop of the boFcγRIII EC2 domain. Mutation analysis of the peptide showed that the residues of Ala98, Gln99, Val101, Val102 and Asn103 within the Fc-binding site are critical for IgG1 binding on boFcγRIII. Summary: Pestiviruses, including classical swine fever virus, remain a concern for global animal health and are responsible for major economic losses of livestock worldwide. Despite high levels of vaccination, currently available commercial vaccines are limited by safety concerns, moderate efficacy, and required high doses. The development of new vaccines is therefore essential. Vaccine efforts should focus on optimizing antigen presentation to enhance immune responses. Here, we describe a simple herringbone‐dimer strategy for efficient vaccine design, using the classical swine fever virus E2 expressed in a rice endosperm as an example. The expression of rE2 protein was identified, with the rE2 antigen accumulating to 480 mg/kg. Immunological assays in mice, rabbits, and pigs showed high antigenicity of rE2. Two immunizations with 284 ng of the rE2 vaccine or one shot with 5.12 μg provided effective protection in pigs without interference from pre‐existing antibodies. Crystal structure and small‐angle X‐ray scattering results confirmed the stable herringbone dimeric conformation, which had two fully exposed duplex receptor binding domains. Our results demonstrated that rice endosperm is a promising platform for precise vaccine design, and this strategy can be universally applied to other Flaviviridae virus vaccines. A total of 587 individuals from 12 indigenous chicken breeds from South China and two commercial breeds were genotyped for 26 microsatellites to investigate the genetic diversity and population structure. All microsatellites were found to be polymorphic. The number of alleles per locus ranged from 5 to 36, with an average of 12.10 ± 7.00 (SE). All breeds, except White Recessive Rock, had high allelic polymorphism (>0.5). Higher genetic diversity was revealed in the indigenous chicken breeds rather than in the commercial breeds. Potential introgression from the commercial breeds into the indigenous chickens was also detected. The population structure of these indigenous chicken breeds could be explained by their geographical distribution, which suggested the presence of independent history of breed formation. Data generated in this study will provide valuable information to the conservation for indigenous chicken breeds in future.

Soil, which exhibits difference in nutrient contents and aggregate sizes, provides spatially distinct habitats for biota. Cover crops influence the compositions of soil organism communities, playing an indispensable role in regulation of underground food webs and ecosystem functions. However, the effect of cover crops on soil microbes and nematodes distribution within different aggregate sizes remains unknown. Thus, a field experiment in a kiwifruit orchard with cover crops was conducted to estimate the distribution of soil nematodes and bacteria with different soil aggregate sizes (mega-aggregate (>2 mm, LMA), macro-aggregate (0.25–2 mm, SMA), and micro-aggregate (<0.25 mm, MA)) and cover crop treatments (four cover crop species (CC) and no cover crop as control (CK)). The results showed that bacterial compositions varied with both aggregate sizes and cover crop treatments. The composition of bacterial community was significantly different between mega-aggregate and micro-aggregate, and bacterial community diversity was significantly higher in micro-aggregate compared with mega-aggregate. Moreover, cover crop treatment dramatically changed the compositions of bacterial communities. However, the nematode communities were mainly impacted by soil aggregate sizes. Larger aggregates (mega- and macro-aggregates) contained higher abundance of omnivores/predators and lower abundance of fungivores. In contrast to bacterial community, the richness of nematode community was lower in micro-aggregates compared with larger aggregates (mega- and macro-aggregates). Redundancy analysis (RDA) and structural equation model (SEM) showed soil organic carbon (SOC) was the main soil factor that directly and indirectly affected both bacterial and nematode communities. Iron is one of the essential mineral elements for the human body and this nutrient deficiency is a worldwide public health problem. Iron is essential in oxygen transport, participates in many enzyme systems in the body, and is an important trace element in maintaining basic cellular life activities. Iron also plays an important role in collagen synthesis and vitamin D metabolism. Therefore, decrease in intracellular iron can lead to disturbance in the activity and function of osteoblasts and osteoclasts, resulting in imbalance in bone homeostasis and ultimately bone loss. Indeed, iron deficiency, with or without anemia, leads to osteopenia or osteoporosis, which has been revealed by numerous clinical observations and animal studies. This review presents current knowledge on iron metabolism under iron deficiency states and the diagnosis and prevention of iron deficiency and iron deficiency anemia (IDA). With emphasis, studies related to iron deficiency and bone loss are discussed, and the potential mechanisms of iron deficiency leading to bone loss are analyzed. Simple Summary: Newcastle disease (ND) is an Office International Des Epizooties (OIE) notifiable terrestrial and aquatic animal disease that can infect chickens of all ages with a high mortality rate, causing ongoing damage to the poultry industry in China currently. In this study, we have developed a blocking lateral flow assay (bLFA) strip for the rapid detection of Newcastle disease virus (NDV) antibodies, using neutralizing monoclonal antibodies against hemagglutinin-neuraminidase. The results showed that the chicken NDV hyperimmunized serum had a complete blocking titer of 11 log 2, and half-blocking titer of 13 log 2, which are 4 times less than and the same as that of the Hemagglutination inhibition (HI) test (13 log 2), and 8 and 2 times less than that of the virus neutralization test (14 log 2), respectively. Additionally, the bLFA strip has no cross-reactivity with positive serum of other avian pathogens. A total of 510 clinical samples were tested for NDV antibodies. The coincidence rate between the results of the bLFA strip and HI test was 97.65%. Therefore, it is an ideal alternative method for assessing the clinical immunity of ND vaccines in the field in real-time. Considering that vaccination is currently the main modality for the prevention of ND, it is essential to assess the effectiveness of clinical immunization. In this study, we have developed a blocking lateral flow assay (bLFA) strip for the rapid detection of NDV antibodies using the monoclonal antibody 9C1 against haemagglutinin-neuraminidase (HN), which allows for the determination of an NDV-specific antibody titer within 10 min at room temperature. In addition, the bLFA strip has no cross-reactivity with the positive serum of other avian pathogens including avian influenza subtypes H5, H7, and H9, MD, IBD, IB, EDS, and avian adenovirus. The ability of the bLFA strip for detecting a neutralizing antibody was also estimated. The results showed that the chicken NDV hyperimmunized serum had a complete blocking (100%) titer of 11 log 2, and half-blocking titer of 13 log 2, which are 4 times less than and the same as that of the HI test (13 log 2), and 8 and 2 times less than that of the VN test (14 log 2), respectively. To investigate the clinical effect of lymphatic preserving varicocelectomy (LPV) versus lymphatic non-preserving varicocelectomy (nLPV). Literature retrieval was performed for literatures published until January 2021 using the terms "varicoceles" and "lymphatic" through databases including Embase, PubMed, Cochrane Review database, Wanfang, and China National Knowledge Infrastructure (CNKI). Randomized controlled trials (RCTs) comparing LPV and nLPV were included. A meta-analysis was conducted using risk ratios (RR) for dichotomous variables. A total of 8 RCTs involving 419 cases with LPV and 434 cases with nLPV met inclusion criteria. LPV was with lower incidence of hydrocele compared with nLPV (RR = 0.099, 95% CI 0.046, 0.212; p = 0.000). In addition, no significant difference was found in recurrence rate (RR = 0.883, 95% CI 0.468, 1.665; p = 0.700) or testicular atrophy rate (RR = 0.750, 95% CI 0.170, 3.311; p = 0.704). This meta-analysis provided evidence that LPV was with lower incidence of hydrocele compared with nLPV, LPV may be preferable to nLPV in the treatment of varicoceles. [ABSTRACT FROM AUTHOR]
