Objective: The objective of this study was to retrospectively assess the effect of Radiofrequency Volumetric Tissue Reduction (RFVTR) on hypertrophic turbinates and clinical outcome in brachycephalic dogs when included in multi-level surgery (MLS). Study design: Clinical retrospective multicenter study. Animals: 132 client-owned brachycephalic dogs. Methods: 132 brachycephalic dogs with high-grade Brachycephalic Obstructive Airway Ayndrome (BOAS) and hypertrophic turbinates were treated with RFVTR as part of MLS of the upper airways. Intranasal obstruction was evaluated by computer tomography (CT) and antero-/retrograde rhinoscopy before and 6 months after RFVTR. The clinical records, the CT images and the rhinoscopy videos were reviewed and clinical evolution was evaluated using a standardized questionnaire. The data was scored semi-quantitatively. Results: In this study, 132 patients were included for a follow-up period of 120 weeks. RFVTR resulted in minor complications, including serous nasal discharge within the first postoperative week in all dogs, and intermittent nasal congestion between 3–8 weeks after treatment in 24.3% of the patients. Rhinoscopy and CT follow-ups were available for 33 patients. Six months after treatment intranasal airspace was increased (p = 0.002) and the presence and overall amount of mucosal contact points was reduced (p = 0.039). Conclusion: MLS with RFVTR led to a significant reduction in turbinate volume at the 6-month follow-up examination and significant clinical improvement over a long-term period of 120 weeks. This suggests the viability of RFVTR as a turbinate-preserving treatment for intranasal obstruction in dogs with BOAS. Diffusion-weighted imaging is increasingly available for brain investigation. Image interpretation of intracranial space-occupying lesions often includes the derived apparent diffusion coefficient (ADC) analysis. In human medicine, ADC can help discriminate between benign and malignant lesions in intracranial tumors. This study investigates the difference in ADC values depending on the sample strategies of image analysis. MRI examination, including diffusionweighted images of canine and feline patients presented between 2015 and 2020, were reviewed retrospectively. Patients with single, large intracranial space-occupying lesions were included. Lesions homogeneity was subjectively scored. ADC values were calculated using six different methods of sampling (M1-M6) on the ADC map. M1 included as much as possible of the lesion on a maximum of five consecutive slices; M2 included five central and five peripheral ROIs; M3 included a single ROI on the solid part of the lesion; M4 included three central ROIs on one slice; M5 included three central ROIs on different slices; and M6 included one large ROI on the entire lesion. A total of 201 animals of various breeds, genders, and ages were analyzed. ADC values differed significantly between M5 against M2 (peripheral) (p < 0.001), M5 against M6 (p = 0.009), and M4 against M2 (peripheral) (p = 0.005). When lesions scored as homogeneous in all sequences were excluded, an additional significant difference in three further sampling methods was present (p < 0.005). ADC of single, large, intracranial space-occupying lesions differed significantly in half of the tested methods of sampling. Excluding homogeneous lesions, additional significant differences among the sampling methods were present. Simple Summary: Diseases involving the subchondral bone are common in horses and are usually evaluated with conventional imaging techniques, such as radiography, conventional computer tomography, and magnetic resonance imaging. Characterization of the normal subchondral bone as well as exercise-related changes is of special interest in ongoing research. Single-source dual energy and detector-based spectral computed tomography are emerging technologies allowing the characterization of tissues that have different attenuation properties at different energies. Both technologies enable the calculation of effective atomic number, an index to determine tissue composition, and electron density, which is assumed to be associated with cellularity in tissues. In the present study, both values were determined in normal subchondral and trabecular bones in the distal limb of horses with single-source dual energy and detector-based spectral computed tomography. Several effective atomic number and electron density values differed between both technologies and within and between bones of the distal equine limb with both technologies. Effective atomic number values increased with age in the palmar/plantar fetlock joint zone. The reported effective atomic number and electron density values in the subchondral and trabecular bone of the equine distal limb may serve as preliminary reference values and aid future classification of diseases. Single-source dual energy (SSDECT) and detector-based spectral computed tomography (DBSCT) are emerging technologies allowing the interrogation of materials that have different attenuation properties at different energies. Both technologies enable the calculation of effective atomic number (EAN), an index to determine tissue composition, and electron density (ED), which is assumed to be associated with cellularity in tissues. In the present prospective observational study, EAN and ED values were determined for 16 zones in normal subchondral and trabecular bone of 37 equine cadaver limbs. Using both technologies, the following findings were obtained: 1. palmar/plantar EAN zone values in the fetlock increased significantly with increasing age of the horse; 2. all EAN and ED values were significantly lower in the trabecular bone than in the subchondral bone of all phalanges; 3. in the distal phalanx and navicular bone, most EAN and ED values were significantly lower compared to the proximal and middle phalanx; and 4. some EAN and ED values were significantly different between front and hind limbs. Several EAN and ED values significantly differed between SSDECT and DBSCT. The reported EAN and ED values in the subchondral and trabecular bone of the equine distal limb may serve as preliminary reference values and aid future evaluation and classification of diseases. Delayed ischemic neurological deficit (DIND) is a major driver of adverse outcomes in patients with aneurysmal subarachnoid hemorrhage (aSAH), defining an unmet need for therapeutic development. Cell-free hemoglobin that is released from erythrocytes into the cerebrospinal fluid (CSF) is suggested to cause vasoconstriction and neuronal toxicity, and correlates with the occurrence of DIND. Cell-free hemoglobin in the CSF of patients with aSAH disrupted dilatory NO signaling ex vivo in cerebral arteries, which shifted vascular tone balance from dilation to constriction. We found that selective removal of hemoglobin from patient CSF with a haptoglobin-affinity column or its sequestration in a soluble hemoglobin-haptoglobin complex was sufficient to restore physiological vascular responses. In a sheep model, administration of haptoglobin into the CSF inhibited hemoglobin-induced cerebral vasospasm and preserved vascular NO signaling. We identified 2 pathways of hemoglobin delocalization from CSF into the brain parenchyma and into the NO-sensitive compartment of small cerebral arteries. Both pathways were critical for hemoglobin toxicity and were interrupted by the large hemoglobin-haptoglobin complex that inhibited spatial requirements for hemoglobin reactions with NO in tissues. Thermally evaporated C60 is a near-ubiquitous electron transport layer in state-of-the-art p–i–n perovskite-based solar cells. As perovskite photovoltaic technologies are moving toward industrialization, batch-to-batch reproducibility of device performances becomes crucial. Here, we show that commercial as-received (99.75% pure) C60 source materials may coalesce during repeated thermal evaporation processes, jeopardizing such reproducibility. We find that the coalescence is due to oxygen present in the initial source powder and leads to the formation of deep states within the perovskite bandgap, resulting in a systematic decrease in solar cell performance. However, further purification (through sublimation) of the C60 to 99.95% before evaporation is found to hinder coalescence, with the associated solar cell performances being fully reproducible after repeated processing. We verify the universality of this behavior on perovskite/silicon tandem solar cells by demonstrating their open-circuit voltages and fill factors to remain at 1950 mV and 81% respectively, over eight repeated processes using the same sublimed C60 source material. Notably, one of these cells achieved a certified power conversion efficiency of 30.9%. These findings provide insights crucial for the advancement of perovskite photovoltaic technologies towards scaled production with high process yield. Introduction: The understanding of epileptic seizure pathogenesis has evolved over time, and it is now generally accepted that not only are cortical and subcortical areas involved but also the connection of these regions in the white matter (WM). Recent human neuroimaging studies confirmed the involvement of the WM in several epilepsy syndromes. Neuroimaging studies investigating WM integrity with diffusion tensor imaging (DTI) in canine idiopathic epilepsy are lacking. This study aimed to test the hypothesis that WM diffusion changes can be found in dogs affected by idiopathic epilepsy. Method: Twenty-six dogs with idiopathic epilepsy (15 Border Collies and 11 Greater Swiss Mountain dogs) and 24 healthy controls (11 Beagle dogs, 5 Border Collies, and 8 Greater Swiss Mountain dogs) were prospectively enrolled. Most dogs with idiopathic epilepsy (17/26) were enrolled within 3 months after seizure onset. Diffusion tensor imaging of the brain with 32 diffusion directions (low b value = 0 s/mm2; maximal b value = 800 s/mm2) was performed in a 3 Tesla scanner. Tract-based spatial statistics (TBSS), a voxel-based approach, was used to investigate changes in fractional anisotropy (FA) and mean diffusivity (MD) in the idiopathic epilepsy group compared to the healthy control group. Additionally, FA and MD were investigated in the region of corpus callosum and cingulate white matter in both groups. Results: We observed subtle changes in WM DTI between the idiopathic epilepsy group and the healthy control group limited to cingulate WM, with a significantly lower FA in the idiopathic epilepsy group compared to the healthy control group in the region of interest (ROI) approach (p = 0.027). No significant changes were found between the idiopathic epilepsy group and the healthy control group in the TBSS analysis and in the corpus callosum in the ROI approach. Conclusion: This study supports the cingulate area as a target structure in canine epilepsy. The subtle changes only might be explained by the short duration of epilepsy, small sample sizes, and the higher variability in canine brain anatomy. Furthermore, all included dogs showed generalized tonic-clonic seizures, possibly affected by generalized epilepsy syndrome, which are also associated with less pronounced DTI changes in humans than focal epilepsy syndromes. G-ratio is crucial for understanding the nervous system's health and function as it measures the relative myelin thickness around an axon. However, manual measurement is biased and variable, emphasizing the need for an automated and standardized technique. Although deep learning holds promise, current implementations lack clinical relevance and generalizability. This study aimed to develop an automated pipeline for selecting nerve fibers and calculating relevant g-ratio using quality parameters in optical microscopy. Histological sections from the sciatic nerves of 16 female mice were prepared and stained with either p-phenylenediamine (PPD) or toluidine blue (TB). A custom UNet model was trained on a mix of both types of staining to segment the sections based on 7,694 manually delineated nerve fibers. Post-processing excluded nonrelevant nerves. Axon diameter, myelin thickness, and g-ratio were computed from the segmentation results and its reliability was assessed using the intraclass correlation coefficient (ICC). Validation was performed on adjacent cuts of the same nerve. Then, morphometrical analyses of both staining techniques were performed. High agreement with the ground truth was shown by the model, with dice scores of 0.86 (axon) and 0.80 (myelin) and pixel-wise accuracy of 0.98 (axon) and 0.94 (myelin). Good inter-device reliability was observed with ICC at 0.87 (g-ratio) and 0.83 (myelin thickness), and an excellent ICC of 0.99 for axon diameter. Although axon diameter significantly differed from the ground truth (p = 0.006), g-ratio (p = 0.098) and myelin thickness (p = 0.877) showed no significant differences. No statistical differences in morphological parameters (g-ratio, myelin thickness, and axon diameter) were found in adjacent cuts of the same nerve (ANOVA p-values: 0.34, 0.34, and 0.39, respectively). Comparing all animals, staining techniques yielded significant differences in mean g-ratio (PPD: 0.48 ± 0.04, TB: 0.50 ± 0.04), myelin thickness (PPD: 0.83 ± 0.28 μm, TB: 0.60 ± 0.20 μm), and axon diameter (PPD: 1.80 ± 0.63 μm, TB: 1.78 ± 0.63 μm). The proposed pipeline automatically selects relevant nerve fibers for g-ratio calculation in optical microscopy. This provides a reliable measurement method and serves as a potential pre-selection approach for large datasets in the context of healthy tissue. It remains to be demonstrated whether this method is applicable to measure g-ratio related with neurological disorders by comparing healthy and pathological tissue. Trapped ion mobility spectrometry (TIMS) adds an additional separation dimension to mass spectrometry (MS) imaging, however, the lack of fragmentation spectra (MS2) impedes confident compound annotation in spatial metabolomics. Here, we describe spatial ion mobility-scheduled exhaustive fragmentation (SIMSEF), a dataset-dependent acquisition strategy that augments TIMS-MS imaging datasets with MS2 spectra. The fragmentation experiments are systematically distributed across the sample and scheduled for multiple collision energies per precursor ion. Extendable data processing and evaluation workflows are implemented into the open source software MZmine. The workflow and annotation capabilities are demonstrated on rat brain tissue thin sections, measured by matrix-assisted laser desorption/ionisation (MALDI)-TIMS-MS, where SIMSEF enables on-tissue compound annotation through spectral library matching and rule-based lipid annotation within MZmine and maps the (un)known chemical space by molecular networking. The SIMSEF algorithm and data analysis pipelines are open source and modular to provide a community resource. There is a need for dataset-dependent MS2 acquisition in trapped ion mobility spectrometry imaging. Computed tomography (CT) has become a routine method to examine the equine skull. Its clinical use for the diagnosis of diseases of the incisors and canine teeth has not been reported so far. The goal of this study was to study the prevalence and relationship of single CT features and equine odontoclastic tooth resorption and hypercementosis (EOTRH) in equine incisors and canine teeth. In this descriptive retrospective study, helical CT studies of 115 Warmblood horses of the age of 5 years or older examined between 2007 and 2020 for reasons unrelated to the incisors and canine teeth were included. Resorption in the crown or root, hypercementosis, widening of the periodontal space, clubbing of the root, lysis of the lamina dura, an abnormal pulp cavity and fracture of the root were recorded in all incisors and canine teeth. The length of the pulp cavity and the labial and lingual/palatal length of the enamel and root was measured in each incisor and canine tooth. Additionally, the angulation was assessed in each incisor. The variable EOTRH was defined based on the presence of resorptive lesions, hypercementosis and clubbing. In result median age of the included horses was 12 years (range, 5–29 years). With regard to the investigated population of horses, 44.3% of all horses had normal incisors whereas 55.7% of the horses had mild EOTRH changes in one incisor at least. Regarding the canine teeth, 54.7% of the horses had normal canine teeth whereas 43.3% horses had mild EOTRH in one canine tooth at least. With regard to all investigated teeth, 868 teeth (53.0%) were classified as normal, whereas 769 incisors and canine teeth (47.0%) showed at least one abnormal CT criterion. Clubbing of the root and hypercementosis were most common (37.1% and 22.7%, respectively) and EOTRH was present in 27.1% of all teeth. Mild changes were more common than moderate or severe abnormalities. In the lower jaw, hypercementosis and widening of the periodontal space at the anatomical root and apex of the root was significantly more frequent (p=0.004 and 0.02), whereas clubbing of the root was more common in the upper jaw (p=0.009). In canine teeth, resorption in the anatomical crown and anatomical root, widening of the periodontal space, clubbing and lysis of the lamina dura were significantly more common whereas hy)percementosis was more common in incisors (p<0.001). Frequency of certain CT changes significantly increased from central to middle and corner incisors. Severity of all single CT criteria as well as prevalence and severity of EOTRH significantly increased with age (r=0.08–0.56). Linear forward and backward multivariate regression analysis confirmed a significant association between prevalence and severity of EOTRH and age, changes of the pulp cavity, widening of the periodontal space at the anatomical rootand incisor group (central, middle, corner) (p=0.01 – < 0.001). In conclusion computed tomographic changes of the incisors and canine teeth are common and age-related in Warm-blood horses. External quartz abrasives are one of the driving forces of macrowear in herbivorous animals. We tested to what extent different sizes and concentrations influence their effect on tooth wear. We fed seven pelleted diets varying only in quartz concentration (0%, 4%, and 8%) and size (fine silt: ∼4 μm, coarse silt: ∼50 μm, fine sand: ∼130 μm) to rabbits (Oryctolagus cuniculus, n = 16) for 2 weeks each in a randomized serial experiment. Measurements to quantify wear and growth of incisors and the mandibular first cheek tooth, as well as heights of all other cheek teeth, were performed using calipers, endoscopic examination, and computed tomography scans before and after each feeding period. Tooth growth showed a compensatory correlation with wear. Absolute tooth height (ATH) and relative tooth height (RTH); relative to the 0% quartz "control" diet) was generally lower on the higher concentration and the larger size of abrasives. The effect was more pronounced on the maxillary teeth, on specific tooth positions and the right jaw side. When offered the choice between different sizes of abrasives, the rabbits favored the silt diets over the control and the fine sand diet; in a second choice experiment with different diets, they selected a pelleted diet with coarse‐grained sand, however. This study confirms the dose‐ and size‐dependent wear effects of external abrasives, and that hypselodont teeth show compensatory growth. The avoidance of wear did not seem a priority for animals with hypselodont teeth, since the rabbits did not avoid diets inducing a certain degree of wear. Research Highlights: Exposure of rabbit teeth to external quartz abrasives results in distinct macrowear patterns depending on the size and concentration they are fed in as well as tooth position. The rabbits prefer the quartz enriched to control diets especially at when silt or coarse sand is added, but the higher wear rates induced by these abrasives are eventually compensated by regrowth of the hypselodont dentition. Diagnostic imaging is a mainstay in the investigation of equine neck and back pain, but interpretation of radiographic and ultrasonographic findings in the spinal column of horses is not straightforward for a variety of reasons including individual anatomical variations, progressive degenerative nature of most pathologies and superimposition as well as technical limits. These issues are even more relevant in the context of examinations of apparently sound horses with absent, mild or unclear complaints, like it is often the case in pre-purchase examinations or cases of failure to meet expected performance. The first aim was to report on the spectrum, degree and location of first-line imaging findings in the spine of a large population of normally performing horses. Limited data is available about agreement of interpretation of equine vertebral column imaging by radiologists. The second aim of this prospective study was to determine interobserver agreement on radiographic and ultrasonographic diagnostic imaging findings in the vertebral column of the same population between multiple observers with longstanding experience in equine diagnostic imaging at two different institutions. Seventy-one horses randomly selected from a larger population of 250 normally performing horses participating in a swiss project on equine back health were examined at one referral center. Radiographic and ultrasonographic examinations were performed in a standardized fashion and images graded separately by two experienced radiologists at two different institutions. Focus was placed on osteoarthrosis of the synovial intervertebral articulations (SIVAs), impinging and overriding of the spinous processes (SPs) and spondylosis. Cohen’s weighted kappa was calculated for each pathology, location and segment in each modality. Interobserver agreement was calculated for findings at specific locations, single pathologies and single grades. Most horses showed no changes and were allocated grades 0 at most locations. Few abnormalities were found throughout the vertebral column, with clusters of abnormalities of the SIVAs in the caudal cervical segment and cranial lumbar segment as well as a cumulation of changes at the dorsal spinous processes in the caudal thoracic segment. These coincide with previously reported predilection sites of imaging findings in symptomatic populations. Overall mean value of agreement for imaging findings was moderate (k= 0.7). Agreement was moderate for imaging findings regarding the synovial intervertebral articulations of the thoracolumbar spine in radiography (k=0.66) but weak in ultrasonography (k=0.58). There was moderate agreement in the imaging findings of the cervical spine in ultrasonography (k=0.61) as well as radiography (k=0.62). Strong agreement was found in the radiographic assessment of changes of the thoracolumbar spinous processes (k=0.80). Almost perfect agreement was found in the radiographic assessment of thoracolumbar spondylosis (k=0.95). Agreement between radiologists in detection and grading of pathologies of the equine vertebral column is weak to almost perfect depending on pathology and location. Our results confirm findings about distribution of specific pathologies found in other studies and support the importance of interpreting imaging findings along with clinical findings for definitive case management and decision making. In recent years, insect husbandry has seen an increased interest in order to supply in the production of raw materials, food, or as biological/environmental control. Unfortunately, large insect rearings are susceptible to pathogens, pests and parasitoids which can spread rapidly due to the confined nature of a rearing system. Thus, it is of interest to monitor the spread of such manifestations and the overall population size quickly and efficiently. Medical imaging techniques could be used for this purpose, as large volumes can be scanned non-invasively. Due to its 3D acquisition nature, computed tomography seems to be the most suitable for this task. This study presents an automated, computed tomography-based, counting method for bee rearings that performs comparable to identifying all Osmia cornuta cocoons manually. The proposed methodology achieves this in an average of 10 seconds per sample, compared to 90 minutes per sample for the manual count over a total of 12 samples collected around lake Zurich in 2020. Such an automated bee population evaluation tool is efficient and valuable in combating environmental influences on bee, and potentially other insect, rearings. [ABSTRACT FROM AUTHOR]

Objectives: Diffusion-weighted imaging (DWI) MRI is increasingly available in veterinary medicine for investigation of the brain. However, apparent diffusion coefficient (ADC) values have only been reported in a small number of cats or in research settings. The aim of this study was to investigate the ADC values of different anatomical regions of the morphologically normal brain in a feline patient population. Additionally, we aimed to assess the possible influence on the ADC values of different patient-related factors, such as sex, body weight, age, imaging of the left and right side of the cerebral hemispheres and white vs grey matter regions. Methods: This retrospective study included cats undergoing an MRI (3T) examination with DWI sequences of the head at the Vetsuisse Faculty of the University Zurich between 2015 and 2021. Only cats with morphologically normal brains were included. On the ADC maps, 10 regions of interest (ROIs) were manually drawn on the following anatomical regions: caudate nucleus; internal capsule (two locations); piriform lobe; thalamus; hippocampus; cortex cerebri (two locations); cerebellar hemisphere; and one ROI in the centre of the cerebellar vermis. Except for the ROI at the cerebellar vermis, each ROI was drawn in the left and right hemisphere. The ADC values were calculated by the software and recorded. Results: A total of 129 cats were included in this study. The ADC varied in the different ROIs, with the highest mean ADC value in the hippocampus and the lowest in the cerebellar hemisphere. ADC was significantly lower in the white cerebral matter compared with the grey matter. ADC values were not influenced by age, with the exception of the hippocampus and the cingulate gyrus. A common feature of domestic animals is tameness--i.e., they tolerate and are unafraid of human presence and handling. To gain insight into the genetic basis of tameness and aggression, we studied an intercross between two lines of rats (Rattus norvegicus) selected over >60 generations for increased tameness and increased aggression against humans, respectively. We measured 45 traits, including tameness and aggression, anxiety-related traits, organ weights, and levels of serum components in >700 rats from an intercross population. Using 201 genetic markers, we identified two significant quantitative trait loci (QTL) for tameness. These loci overlap with QTL for adrenal gland weight and for anxiety-related traits and are part of a five-locus epistatic network influencing tanxeness. Au additional QTL influences the occurrence of white coat spots, but shows no significant effect on tameness. The loci described here are important starting points for finding the genes that cause tameness in these rats and potentially in domestic animals in general. DOI: 10.1534/genefics.109.102186
