The Kirkpatrick–Baez (KB) microscope is an effective instrument for x-ray imaging of hot spot. However, the non-uniform distribution of response efficiency in the field of view is a drawback of the KB microscope. A more accurate hot spot image requires the correction of the measured image by combining the hot spot position and the response efficiency distribution. Here, we describe a method to locate the position of the hot spot in the field of view during hot spot imaging with a KB microscope. The position of the hot spot in the field of view can be obtained by measuring the grazing incidence angle change during hot spot imaging. In the experiment of hot spot self-emission imaging with a four-channel KB microscope, the location of the hot spot with an accuracy of 15 μm was realized, and the intensity corrected hot spot image was obtained. This will solve the problem of the non-uniform distribution of the response efficiency of the KB microscope and enable quantitative measurement of hot spot radiation intensity.

Abstract In laser-driven inertial confinement fusion, driving pressure boosting and smoothing are major challenges. A proposed hybrid-drive (HD) scheme can offer such ideal HD pressure performing stable implosion and nonstagnation ignition. Here we report that in the hemispherical and planar ablator targets installed in the semicylindrical hohlraum scaled down from the spherical hohlraum of the designed ignition target, under indirect-drive (ID) laser energies of ~43–50 kJ, the peak radiation temperature of 200 ± 6 eV is achieved. And using only direct-drive (DD) laser energies of 3.6–4.0 kJ at an intensity of 1.8 × 1015 W/cm2, in the hemispherical and planar targets the boosted HD pressures reach 3.8–4.0 and 3.5–3.6 times the radiation ablation pressure respectively. In all the above experiments, significant HD pressure smoothing and the important phenomenon of how a symmetric strong HD shock suppresses the asymmetric ID shock pre-compressed fuel are demonstrated. The backscattering and hot-electron energy fractions both of which are about one-third of that in the DD scheme are also measured.

Diagnosing the layered cryogenic DT implosions with traditional absorption based x-ray backlight radiography in inertial confinement fusion is a challenge because of the low opacity of the cold fuel. Refraction enhanced x-ray phase-contrast imaging was proposed for diagnosing optically opaque material. In this paper, A Monte Carlo tool based on Geant4 is employed to model the x-ray phase-contrast imaging for diagnosing cold fuel layer in cryogenic implosions. This model can successfully explain the x-ray phase-contrast imaging experimental results on a micro focus x-ray tube with triple-layer ablator capsules. Furthermore, the radiographs of ignition-scale capsule target is calculated. The fuel layer of DT ice can be observed with the phase contrast imaging and the image is faded using absorption imaging only. Our simulations show that the shape of cold fuel and implosion velocity can be inferred directly with the phase contrast imaging in inertial confinement fusion(ICF).

Two neutron time-of-flight (nToF) detectors have been employed to measure the neutron time-of-flight spectrum in different lines-of-sight, i.e., at the equator plane and the south pole, on Shenguang-III (SG-III) laser facility. The contribution of scattered neutrons has been calculated with the Monte Carlo code JMCT for each nToF detector. The results show that the scattered neutron spectrum is dominated by neutrons scattered on materials in the experiment hall, including the vacuum chamber. The shape of the scattered neutron spectrum depends on the view line, which has been observed with nToF detectors located in the experiment hall of the SG-III laser facility. A method based on the convolution of the calculated neutron time-of-flight spectrum and the instrument response function has been developed for the ion temperature determination. The calculated neutron spectra with the contribution of scattered neutrons can fit the measured results. No obvious ion temperature anisotropy has been observed on the SG-III laser facility at present.

The interactions between the bacterial Hsp70, DnaK and the sequence of a model all-alpha-helical globin (apoMb) are explored by cellulose-bound peptide scanning. It is found that the simple fractional mean buried area correlates well with Hsp70's binding site locations along the apoMb sequence.

The high-resolution secondary structure of relatively large N-terminal protein fragments bound to the substrate-binding domain of the cotranslationally active chaperone DnaK is reported, taking the [alpha]-helical protein apomyoglobin (apoMb) as a model substrate. Data show evidence for weak, residual helical content of chaperone-bound N-terminal protein fragment chains away from the chaperone binding site and local helical unwinding at the binding site.

A coaxial, high performance diamond detector has been developed for neutron diagnostics of inertial confinement fusion at ShenGuangIII laser facility. A Φ10 mm × 1 mm "optical grade" chemical-vapor deposition diamond wafer is assembled in coaxial-designing housing, and the signal is linked to a SubMiniature A connector by the cathode cone. The coaxial diamond detector performs excellently for neutron measurement with the full width at half maximum of response time to be 444 ps for a 50 Ω measurement system. The average sensitivity is 0.677 μV ns/n for 14 MeV (DT fusion) neutrons at an electric field of 1000 V/mm, and the linear dynamic range is beyond three orders of magnitude. The ion temperature results fluctuate widely from the neutron time-of-flight scintillator detector results because of the short flight length. These characteristics of small size, large linear dynamic range, and insensitive to x-ray make the diamond detector suitable to measure the neutron yield, ion temperature, and neutron emission time. The response of coastal wetlands to future sea-level rise remains uncertain. Palaeoecological data are essential to constrain the still conflicting ecological models. However, obtaining detailed palaeo-coastal stratigraphic records before Holocene is often difficult due to repeated Quaternary marine transgression-regressions. Here we utilize pollen data from a deep-sea sedimentary archive in northern South China Sea to explore the historical behavior of coastal wetlands at a large river estuary over the last 140-kyr. A recurrent wetland (Cyperaceae)-pioneer species (Selaginella)-zonal forest (Pinus) succession throughout the last glacial-interglacial cycle implies a coastal salt marsh origin of the Cyperaceae pollen. Comparing with global sea-level reconstructions, the increases in Cyperaceae pollen abundance, and hence the expansions of coastal salt marsh, were found to be closely linked with rapid large-scale sea-level rises. This finding indicates a resilience of coastal wetlands to future sea-level rise, and highlights the probable importance of conventionally ignored horizontal adaptability in long-term survival of coastal wetlands. Overall, marine pollen records provide an opportunity to supplement existing palaeoecological observations of coastal wetlands during the Quaternary.

Neutron emission measurement at the HL-2A tokamak device with a liquid scintillation detector is described. The detector was placed at a location with little structure material in the field of view, and equipped with a gain monitoring system which could provide the possibility to evaluate the gain variation as well as to correct for the detector response. Time trace of the neutron emissivity was obtained and it was consistent with the result of a standard 235U fission chamber. During the plasma discharge the neutron yield could vary by about four orders of magnitude and the fluctuation of the detector gain was up to about 6%. Pulse height spectrum of the liquid scintillation detector was constructed and corrected with the aid of the gain monitoring system, and the correction was found to be essential for the assessment of the neutron energy spectrum. This successful measurement offered experience and confidence for the application of liquid scintillation detectors in the upcoming neutron camera system. [ABSTRACT FROM AUTHOR]

Background Although increased volume of pericardial fat has been associated with decreased cardiac function, it is unclear whether this association is mediated by systemic overall obesity or direct regional fat interactions. We hypothesized that if local effects dominate, left ventricular (LV) function would be most strongly associated with pericardial fat that surrounds the left rather than the right ventricle (RV). Methods Female obese subjects (n = 60) had cardiovascular magnetic resonance (CMR) scans to obtain measures of LV function and pericardial fat volumes. LV function was obtained using the cine steady state free precession imaging in short axis orientation. The amount of pericardial fat was determined volumetrically by the cardiac gated T1 black blood imaging and normalized to body surface area. Results In this study cohort, LV fat correlated with several LV hemodynamic measurements including cardiac output (r = -0.41, p = 0.001) and stroke volume (r = -0.26, p = 0.05), as well as diastolic functional parameters including peak-early-filling rate (r = -0.38, p = 0.01), early late filling ratio (r = -0.34, p = 0.03), and time to peak-early-filling (r = 0.34, p = 0.03). These correlations remained significant even after adjusting for the body mass index and the blood pressure. However, similar correlations became weakened or even disappeared between RV fat and LV function. LV function was not correlated with systemic plasma factors, such as C-reactive protein (CRP), B-type natriuretic peptide (BNP), Interleukin-6 (IL-6), resistin and adiponectin (all p > 0.05). Conclusions LV hemodynamic and diastolic function was associated more with LV fat as compared to RV or total pericardial fat, but not with systemic inflammatory markers or adipokines. The correlations between LV function and pericardial fat remained significant even after adjusting for systemic factors. These findings suggest a site-specific influence of pericardial fat on LV function, which could imply local secretion of molecules into the underlying tissue or an anatomic effect, both mechanisms meriting future evaluation. The ability of human serum albumin (HSA) to bind fatty acids (FA) in multiple sites has been revealed by many studies. Here we detect and characterize nine individual binding sites by two-dimensional (2D) nuclear magnetic resonance (NMR) spectroscopy of 18-[13C]-oleic acid (OA) complexed with HSA We characterize site-specific FA binding by addition of (i) different FA molar ratios (from 1:1 to 4:1 OA:HSA) to observe the order of filling and occupancy of binding sites; (ii) methyl-β-cyclodextrin, as a FA acceptor, to observe the dissociation of FA; and (iii) drugs (with known binding sites in the crystal structure) to reveal the correspondence of three NMR peaks with sites in the crystal structure. At 1:1 and 2:1 OA:HSA ratios, three sites were shown to fill sequentially. These high-affinity sites were well resolved from additional sites (one medium-affinity and five low-affinity) observed at 3:1 and 4:1 0A:HSA ratios. Methyl-#946;-cycIodextrin extracted OA from individual sites in the reverse order of filling. FA bound in three low-affinity sites were displaced by drugs shown to bind in crystalline HSA to FA sites 7 and 3 (Sudlow's drug sites I and II, respectively) and FA site 6. With this strategy, 2D NMR spectral analysis permits site-specific characterization of the binding of drugs and FA and provides a sensitive probe of the mutual effects of FA and ligand binding. Storage of lipid in ectopic depots outside of abdominal visceral and subcutaneous stores, including within the pericardium and liver, has been associated with obesity, insulin resistance, and cardiovascular risk. We sought to determine whether anatomically distinct ectopic depots were physiologically correlated and site-specific effects upon cardiovascular function could be identified. Obese subjects (n = 28) with metabolic syndrome but without known atherosclerotic disease and healthy controls (n = 18) underwent magnetic resonance imaging (MRI) and proton MR spectroscopy (MRS) to quantify pericardial and periaortic lipid volumes, cardiac function, aortic compliance, and intrahepatic lipid content. Fasting plasma lipoproteins, glucose, insulin, and free-fatty acids were measured. Pericardial and intrahepatic (P < 0.01) and periaortic (P < 0.05) lipid volumes were increased in obese subjects vs. controls and were strongly and positively correlated (P ≤ 0.01) but independent of BMI (P = NS) among obese subjects. Intrahepatic lipid was associated with insulin resistance (P < 0.01) and triglycerides (P < 0.05), whereas pericardial and periaortic lipid were not (P = NS). Periaortic and pericardial lipid positively correlated to free-fatty acids (P ≤ 0.01) and negatively correlated to high-density lipoprotein (HDL) cholesterol (P < 0.05). Pericardial lipid negatively correlated to cardiac output (P = 0.03) and stroke volume (P = 0.01) but not to left ventricular ejection fraction (P = 0.46). None of the ectopic depots correlated to aortic compliance. In conclusion, ectopic storage of lipid in anatomically distinct depots appeared tightly correlated but independent of body size. Site-specific functional abnormalities were observed for pericardial but not periaortic lipid. Little is known about polypeptide conformation and folding in the presence of molecular chaperones participating in protein biosynthesis. In vitro studies on chaperone-substrate complexes have been mostly carried out with small peptide ligands. However, the technical challenges associated with either competing aggregation or spectroscopically unfavorable size and exchange rates have typically prevented analysis of larger substrates. Here, we report the high-resolution secondary structure of relatively large N-terminal protein fragments bound to the substrate-binding domain of the cotranslationally active chaperone DnaK. The all-α-helical protein apomyoglobin (apoMb), bearing the ubiquitous globin fold, has been chosen as a model substrate. On the basis of NMR secondary chemical shift analysis, we identify, for the first time, weak helical content (similar to that found in the chemically unfolded full-length protein) for the assigned residues of the chaperone-bound chain away from the chaperone binding sites. In contrast, we found that the residues corresponding to the strongest specific binding site for DnaK, examined via a short 13-mer apoMb peptide fragment matching the binding site sequence, display highly reduced helical content in their chaperone-bound form. Given that the free state of the peptide is weakly helical in isolation, we conclude that the substrate residues corresponding to the chaperone binding site undergo helix unwinding upon chaperone binding. Low density and low convergence implosion occurs in the exploding-pusher target experiment, and generates neutrons isotropically to develop a high yield platform. In order to validate the performance of ShenGuang (SG) laser facility and test nuclear diagnostics, all 48-beam lasers with an on-target energy of 48 kJ were firstly used to drive room-temperature, DT gas-filled glass targets. The optimization has been carried out and optimal drive uniformity was obtained by the combination of beam repointing and target. The final irradiation uniformity of less than 5% on polar direct-drive capsules of in diameter was achieved, and the highest thermonuclear yield of the polar direct-drive DT fuel implosion at the SG was 1.04×1013. The experiment results show neutron yields severely depend on the irradiation uniformity and laser timing, and decrease with the increase of the diameter and fuel pressure of the target. The thin CH ablator does not impact the implosion performance, but the laser drive uniformity is important. The simulated results validate that the distribution laser design is reasonable and can achieve a symmetric pressure distribution. Further optimization will focus on measuring the symmetry of the hot spot by self-emission imaging, increasing the diameter, and decreasing the fuel pressure. [ABSTRACT FROM AUTHOR]

Double-shell implosion is proposed as a non-cryogenic target design for ignition, offering a robust implosion platform for inertial confinement fusion research. Hohlraum-driven double-shell implosions have been performed at the ShenGuang-III laser facility; a vacuum hohlraum with a high CCR value was employed. With 96kJ laser energy in a squared temporal profile, the implosion neutron yield was close to 1010, and the YOC1D was about 27%. A fuel areal density of 14 mg cm−2 was inferred from the measured yield ratio of secondary and primary neutrons. The implosion trajectory was diagnosed via the 4.75 keV x-ray backlight. The measured implosion velocity and hot-spot x-ray emission history indicate a predictable hydrodynamic process in the double-shell implosion. Two crucial factors which might detrimentally affect implosion performance are the implosion symmetry and pusher–gas mix. The loss of four driven laser beams in the backlight shot caused the YOC1D to decrease to 5% and fuel areal density to decrease to 4 mg cm−2. In future, optimization of the implosion symmetry and the capsule fabrication will be implemented to improve the double-shell implosion performance. [ABSTRACT FROM AUTHOR]

The new neutron spectrometer time-of-flight enhanced diagnostics (TOFED) for the EAST tokamak is presented and its characteristics are described in terms of simulation results, as well as the interface in the torus hall along with new neutral beam (NB) injectors. The use of TOFED for studies of the slowing down of NB-injected deuterons is illustrated. The implications of measuring the neutron emission on a long pulse machine are discussed together with the experimental challenges and diagnostic possibilities approaching those to be encountered in continuous operation. A single crystal chemical vapor deposition (scCVD) diamond detector has been characterized and employed for the neutron measurement at the HL-2A tokamak device. The scCVD diamond detector has been deposited with 5μm of lithium fluoride (LiF) layer to enhance the sensitivity to thermal neutrons. Time stability of the detector has been studied with α-source and good performance has been found for more than 12h. Neutron irradiations have been performed in four quasi-monoenergetic neutron fields in the energy range from 2.50MeV to 16.03MeV. The measured response function of the scCVD diamond detector to 14.13MeV neutrons shows a narrow 12C (n, α)9Be reaction peak which is well isolated from other structures by about 1MeV in energy, indicating the great potential as a fast neutron spectrometer. Neutron measurement of deuterium plasma discharge was established at the HL-2A tokamak device, and good consistence has been revealed among this detector signal and other related signals.
