IntroductionThere is no established scoring model focused on viral hepatitis patients to predict the prognosis after transjugular intrahepatic portosystemic shunt (TIPS). We aimed to develop and validate a novel model based on the largest cohort for better prediction of both short-term (1 year) and long-term (3 years) postoperative prognoses after TIPS in viral hepatitis cirrhosis-related portal hypertension patients.MethodsA total of 925 viral hepatitis cirrhosis-related portal hypertension patients who underwent TIPS from nine hospitals were divided into the training and external validation cohorts. A novel Viral-associated Index of Post-TIPS score (VIPs) model was developed after performing Cox regression analysis. The VIPs model was compared to five previous models, namely, Child–Pugh, MELD, ALBI, CCG, and FIPS. Furthermore, X-tile software was used to stratify patients into low-, medium-, and high-risk groups.ResultsThe VIPs model included age, ascites, albumin, prothrombin time, total bilirubin, and sodium for post-TIPS prognosis prediction. The model demonstrated satisfying predictive efficiency in both discrimination and calibration, with an area under the curve of 0.781/0.774 (1 year/3 years) in the training cohort and 0.771/0.775 (1 year/3 years) in the external validation cohort, respectively.DiscussionWe first developed and externally validated a novel VIPs model for better prediction of both short-term and long-term postoperative prognoses after TIPS in Chinese patients with viral hepatitis cirrhosis-related portal hypertension.

Abstract To gain a deeper understanding of the genetic factors influencing the growth and development of Eriocheir sinensis, a well-known species of hairy crab found in Yangcheng Lake, this study focused on the de novo genome and full-length transcriptome information of the selected subjects. Specifically, Yangcheng Lake hairy crabs were chosen as the experimental samples. Initially, a genome analysis was performed, resulting in the identification of gene fragments with a combined length of 1266,092,319 bp. Subsequently, a transcriptome analysis was conducted on a mixture of tissues from four different sites, namely muscle, brain, eye, and heart, to further investigate the genetic characteristics at the transcriptome level. The Pacific Biosciences (Pacio) single-molecule real-time sequencing system generated a total of 36.93 G sub-fragments and 175,90041 effective inserts. This research contributes to the indirect comprehension of genetic variations underlying individual traits. Furthermore, a comparison of the obtained data with relevant literature emphasizes the advantages of this study and establishes a basis for further investigations on the Chinese mitten crab.

Tandem solar cells have the potential to surpass conventional single-junction photovoltaics by harnessing a wider range of the solar spectrum and reducing losses caused by thermalization and transmission. This study examines the performance of a kesterite-kesterite tandem solar cell with two different absorber layers in each subcell using SCAPS-1D. The upper subcell comprises CZTS and ACZTS with wider bandgaps, while the lower subcell comprises ACZTSe and CZTSe with narrower bandgaps. The CZTS- and CZTSe-based solar cells were first validated using experimental data to determine the feasibility of the tandem solar cell design. The CZTS-based solar cell was modified by replacing the back contact, adding a Cd-free buffer layer of ZnMnO, and using a SnMnO2 layer as a window layer. The band gap and electron affinity of ZnMnO and SnMnO2 layers were adjusted by changing their manganese contents for optimal conduction band offset. A thin ACZTS layer was also added as a second absorber layer in the CZTS-based solar cell to enhance open circuit voltage. The CZTSe-based solar cell was thereafter modified by including an ACZTSe layer as a second absorber layer and by selecting ZnSe as a buffer layer and optimizing the thicknesses of all layers. Both upper and lower subcells were analyzed for tandem configuration using filtered spectra and current-matching techniques. The thickness of the CZTS layer in the upper subcell was adjusted to achieve current matching; consequently, the proposed tandem cell had an efficiency of ∼24%. These findings provide valuable insights for future advancements in kesterite-kesterite-based tandem solar cells.

Monolayer molybdenum disulfide (MoS2) has weak light absorption due to its atomically-thin thickness, thus hindering the development of MoS2-based optoelectronic devices. CdSxSe[Formula: see text] has excellent photoelectric performance in the visible light range, and its nanostructure shows great potential in new nanoscale electronic and optoelectronic devices. In this work, a composite photodetector device with the combination of monolayer MoS2 nanosheets and CdS[Formula: see text]Se[Formula: see text] nanobelts has been successfully prepared, which can not only maintain the inherent excellent properties of the two blocks, but also play a synergistic role between them, thus improving the photoelectric performance of the device. The monolayer MoS2 nanosheet /CdS[Formula: see text]Se[Formula: see text] nanobelt photodetector has a wide spectral response range (400–800[Formula: see text]nm), high responsivity (527.22[Formula: see text]A/W) and large external quantum efficiency (EQE) ([Formula: see text]%). Compared with the isolated monolayer MoS2 nanosheet, both the responsivity and EQE of the hybrid photodetector are increased by 117.4 times under 620[Formula: see text]nm illumination. This study provides a way to prepare hybrid photodetectors with wide spectral response and high responsivity.

Abstract Aim The aim of this study is to compare the prognostic values of the Child–Pugh, integrated model for end‐stage liver disease (iMELD), albumin–bilirubin (ALBI), and Freiburg index of postsurvival (FIPS) scores in patients undergoing transjugular intrahepatic portosystemic shunt (TIPS). Methods We conducted a multicenter retrospective study including patients who underwent TIPS by collecting data from several hospitals in southwest China between January 2014 and February 2021. We compared the performance of different scoring models for survival prediction in these patients. The performance of each scoring model was assessed via area under the receiver‐operating characteristic (AUROC) curve analysis. Results The study included 378 TIPS patients (268 men, 110 women; median age 52 [interquartile range, 45–60] years). Age; cirrhosis etiology; ascites severity; albumin levels; international normalized ratio; total bilirubin levels; sodium levels; and Child–Pugh, iMELD, ALBI, and FIPS scores were significant prognostic factors in cirrhotic patients who underwent TIPS. The Child–Pugh, iMELD, ALBI, and FIPS scores were all independent predictors of survival in TIPS patients. Survival analysis showed that all scoring models effectively stratified the prognostic risk of these patients. The Child–Pugh score was the best predictor of postoperative survival, followed by the ALBI and FIPS scores. The iMELD score was the worst predictor. The Child–Pugh, iMELD, ALBI, and FIPS scores predicted the 1‐year postoperative survival, with AUROC values of 0.832, 0.677, 0.761, and 0.745, respectively, and the 3‐year postoperative survival, with AUROC values of 0.710, 0.668, 0.721, and 0.658, respectively. The calibration curve showed that the Child–Pugh, ALBI, and FIPS models performed well in predicting 1‐ and 3‐year survival, whereas the iMELD model was a poor predictor. Conclusions The four scoring models can predict survival in cirrhotic patients after TIPS and can effectively stratify prognostic risk. The Child–Pugh score may be more suitable for predicting survival after TIPS in patients with liver cirrhosis.

The opening and closing of a vacuum circuit breaker in the selected phase of the system voltage and the associated electrical equipment plug in or disconnect the electrical system with minimal impact on itself and the system. To realize this and ensure the control precision and stability of its action process, this study designs the drive motor and controller of a vacuum circuit breaker actuator. Additionally, a new function-switching pseudo-differential control (FPD) strategy is based on the feedback element principle. The control strategy avoids the direct differentiation of the controlled variables and has the ability of a fast dynamic response and anti-interference. At the same time, according to the moving process of the circuit breaker contacts, the control parameters are directly assigned to the system in stages, which reduces the amount of calculation and enhances the response speed of the control system. According to the research on the motor actuator movement process, the motor actuator and its control system of a 40.5 kV vacuum circuit breaker are developed and verified by closing operation in the selected phase, and the experimental results show the effectiveness of the proposed control strategy.

A combined-type axial-flux permanent magnet machine is proposed in this paper. The structure of the proposed machine is introduced. The torque density characteristics of the proposed combined-type axial-flux permanent magnet machine are studied, and the torque density advantages of this kind of machine are analyzed by comparing with radial-flux permanent magnet machines. The magnetic field analysis method of the machine is studied. In the process of magnetic field analysis, an air gap permeance element is proposed to improve the traditional magnetic equivalent circuit method, which can facilitate the establishment of magnetic equivalent circuit model at different times. The calculation results of the proposed method are verified by finite element method. The cogging torque optimization of this machine is investigated, which includes magnet skewing with different reference modes. The optimal magnet skewing angle is calculated and verified, and the characteristics of different cogging torque optimization methods are summarized.

The effect of seed power on the inter-modal four-wave mixing (IM-FWM) effect in a distributed side-pumped fiber amplifier is investigated for the first time, to the best of our knowledge. By using a seed source smaller than 70 W, the effect of seed power on the IM-FWM is studied experimentally. Surprisingly, it is found that the IM-FWM effect becomes stronger with the weaker seed. To analyze the physics behind this result, a numerical model is introduced, and the numerical results are in good agreement with the experimental observations. It is revealed that the IM-FWM enhancement is induced by the gain competition between the signal light and the anti-Stokes light of the IM-FWM. It is also found that there is an optimized seed power for suppressing the IM-FWM effect. The experimental and numerical results which are presented in this work can provide significant guidance for understanding the IM-FWM in fiber lasers and amplifiers.

Switched reluctance motor has been widely used in various fields because of its simple, solid structure and high reliability. However, the disadvantage of large torque ripple limits its application in high-speed occasions. In this paper, a new type of rotor dislocation permanent magnet assisted high-speed reluctance motor (RDPMA-HSRM) is proposed, which realizes the axial magnetic flux through the structure of two groups of stator and rotor, and a permanent magnet is connected in series for auxiliary excitation. On this basis, the rotor dislocation is developed and a unique structural design is developed. The working principle of the motor is explained, the magnetic circuit model of the motor is analyzed, the mathematical model of the motor is given, and the contribution of permanent magnet assistance to torque is discussed. The magnetic characteristics of the motor in terms of flux density, flux linkage, inductance and torque are obtained by finite element simulation, and compared with the characteristics of the improved motor. The effects of rotor misalignment and permanent magnet auxiliary excitation on the performance of the motor are analyzed. Finally, the prototype is tested and the experimental results are obtained. Simulation and experimental results show the RDPMA-HSRM has smaller torque ripple and higher torque density.

Abstract Gearbox oil temperature is one of the important indicators for gearbox condition monitoring and faults early warning. Accurately predicting the gearbox oil temperature change trend can maintain the gearbox in advance and ensure the safety and reliability of the wind turbine gearbox. The purpose of this article is to analyze the supervisory control and data acquisition (SCADA) data in wind turbines. A method based on multi‐input improved ant lion optimization and support vector regression (M‐IALO‐SVR) proposed, which can accurately predict the gearbox oil temperature. The prediction method is compared with back propagation neural network (BPNN) and ALO‐SVR methods to verify the effectiveness of the M‐IALO‐SVR method. To further analyze the prediction results, the 95% confidence interval processing is performed on the residuals of the prediction model, and then the trends of the mean and standard deviation of the moving window residuals are calculated. Testing SCADA data from a wind farm in northeast China, the test results show that when the gearbox is operating normally, the predicted value of the gearbox oil temperature follows the measured value very well. When the gearbox operates abnormally, its temperature deviates from the normal range, and the statistical characteristics of the residuals also change. According to the trend of the residuals statistical characteristics, the abnormal state of the gearbox can be found in time.

Highly toxic pollutants such as dioxins and metals emitted from municipal solid waste (MSW) incineration have caused a serious not-in-my-backyard social phenomenon. However, relatively few studies have comprehensively analyzed the contaminant characteristics, coexistence relationships and potential health risks of dioxins and metals in soils near MSW incinerator. Therefore, this study monitors 17 dioxin homologues and 13 metals in soils around MSW incinerator to evaluate their contaminant and coexistence characteristics as well as associated potential health risks. The empirical study focused on Hainan Province, a relatively pure area with no heavy industry and low population density in southern China. The contamination characteristics analysis shows that the total International Toxic Equivalence Quantity (I-TEQ) concentrations of dioxins are comparatively lower than the risk control standards and that in previous studies, whereas the concentrations of specific metals (e.g., Ni, Cu, Cd and Cr) are higher than the risk control standards. To explore the coexistence relationships, the statistical analysis is conducted and indicates that Sb, Cd, Zn and Pb metals (extracted as principal components, with a variance contribution rate of 30.89%) and most dioxin homologues (accounting for 82.83% of the total I-TEQ values) are clustered into one group, suggesting that they may have similar distribution characteristics and origins. Furthermore, the health risk analysis of two toxic pollutants reveals that the carcinogenic and non-carcinogenic risks of dioxins and most metals (except for Ni and Cr) are all below the acceptable levels, implying that the studied incinerator posed no obvious health effect to the surrounding residents.

Abstract Background To examine association between gestational weight gain (GWG) in women with gestational diabetes mellitus (GDM) and adverse pregnancy outcomes (APOs). Methods This retrospective cohort study enrolled women with GDM who delivered at 2010–2020 in Changzhou, Jiangsu. Total GWG, rates of GWG in second trimester and third trimesters were stratified into three categories according to IOM guidelines: within, below, and above IOM guidelines. Univariable and multivariable logistic regression analyses were used. Results Overall, 1606 women with GDM fulfilled inclusion criteria. Compared with within IOM guidelines and after adjusting for confounders, total GWG above IOM guidelines in pregnancy was associated with an increased odds of caesarean delivery [adjusted odds ratio (aOR) = 1.34, 95% confidence interval (CI): 1.04–1.72], hypertensive disorders of pregnancy (HDP) (aOR = 2.00, 1.28–3.12), preeclampsia (aOR = 2.06, 1.01–3.12), macrosomia (aOR = 1.55, 1.13–2.13) and large for gestational age (LGA) (aOR = 2.82, 1.94–3.23), and a decreased odds of premature rupture of membrane (PROM) (aOR = 0.46, 0.36–0.60) and preterm birth (aOR = 0.35, 0.26–0.44); total GWG below IOM guidelines in pregnancy was associated with an increased risk of preterm birth (aOR = 1.96, 1.44–2.66), small for gestational age (SGA) (aOR = 1.32, 1.11–1.50) and a decreased odds of macrosomia (aOR = 0.35, 0.23–0.53) and LGA (aOR = 0.54, 0.42–0.72). Further, in both second and third trimesters of pregnancy, rates of GWG above IOM guidelines was found to be associated with a high odds of HDP (aOR = 2.55, 1.86–3.38; aOR = 1.93, 1.08–2.98), preeclampsia (aOR = 2.28, 1.21–3.81; aOR = 2.17, 1.35–4.37), macrosomia (aOR = 1.20, 1.02–1.82; aOR = 2.02, 1.51–2.64) and LGA (aOR = 1.42, 1.24–1.97; aOR = 1.79, 1.51–2.54). Rates of GWG above IOM guidelines in third trimester of pregnancy also increased odds of caesarean delivery (aOR = 1.48, 1.16–2.34) when compared with within IOM guidelines. While rates of GWG below IOM guidelines in both second and third trimesters of pregnancy was associated with a decreased odds of macrosomia (aOR = 0.66, 95% CI: 0.52–0.78; aOR = 0.52, 0.39–0.63) and LGA(aOR = 0.71, 0.51–0.82; aOR = 0.67, 0.55–0.79). In addition, rate of GWG below IOM guidelines in third trimester of pregnancy was associated with an increased odds of preterm birth (aOR = 1.52, 1.12–2.05) and SGA (aOR = 1.21, 1.10–1.69). Conclusion GWG, outside IOM guidelines has increased risks of APOs among women with GDM, implying that careful surveillance for GWG during different stages of pregnancy is warranted.

The kynurenine pathway (KP) is the primary route for the catabolism of the essential amino acid tryptophan. The central KP metabolites are neurologically active molecules or biosynthetic precursors to critical molecules, such as NAD+. Within this pathway are three enzymes of interest, HAO, ACMSD, and AMSDH, whose substrates and/or products can spontaneously cyclize to form side products such as quinolinic acid (QA or QUIN) and picolinic acid. Due to their unstable nature for spontaneous autocyclization, it might be expected that the levels of these side products would be dependent on tryptophan intake; however, this is not the case in healthy individuals. On top of that, the regulatory mechanisms of the KP remain unknown, even after a deeper understanding of the structure and mechanism of the enzymes that handle these unstable KP metabolic intermediates. Thus, the question arises, how do these enzymes compete with the autocyclization of their substrates, especially amidst increased tryptophan levels? Here, we propose the formation of a transient enzyme complex as a regulatory mechanism for metabolite distribution between enzymatic and non-enzymatic routes during periods of increased metabolic intake. Amid high levels of tryptophan, HAO, ACMSD, and AMSDH may bind together, forming a tunnel to shuttle the metabolites through each enzyme, consequently regulating the autocyclization of their products. Though further research is required to establish the formation of transient complexation as a solution to the regulatory mysteries of the KP, our docking model studies support this new hypothesis.

Background and AimsEmergency endoscopy is recommended for patients with acute esophageal variceal bleeding (EVB) and their prognosis has improved markedly over past decades due to the increased specialization of endoscopic practice. The study aimed to compare outcomes following emergency endoscopic injection sclerotherapy (EIS) and endoscopic variceal ligation (EVL) in cirrhotic patients with acute EVB.MethodsCirrhotic patients with acute EVB who underwent emergency endoscopy were retrospectively enrolled from 2013 to 2020 across 34 university hospitals from 30 cities. The primary outcome was the incidence of 5-day rebleeding after emergency endoscopy. Subgroup analysis was stratified by Child-Pugh class and bleeding history. A 1:1 propensity score matching (PSM) analysis was performed.ResultsA total of 1,017 and 382 patients were included in EIS group and EVL group, respectively. The 5-day rebleeding incidence was similar between EIS group and EVL group (4% vs. 5%, P = 0.45). The result remained the same after PSM (P = 1.00). Among Child-Pugh class A, B and C patients, there were no differences in the 5-day rebleeding incidence between the two groups after PSM (P = 0.25, 0.82, and 0.21, respectively). As for the patients with or without bleeding history, the differences between EIS group and EVL group were not significant after PSM (P = 1.00 and 0.26, respectively).ConclusionThe nationwide cohort study indicates that EIS and EVL are both efficient emergency endoscopic treatment strategies for acute EVB. EIS should not be dismissed as an economical and effective emergency endoscopic treatment strategy of acute EVB. ClincialTrials.gov number NCT04307264.

The effect of tens-of-Watt seed light on the stimulated Raman scattering (SRS) in a distributed side-pumped Yb-doped fiber amplifier is studied. By using two sorts of seed sources with the power smaller than 70 W, it is observed in experiment that the SRS can be enhanced by lowering the seed power. To the best of our knowledge, this is the first observation of the weak-seed enhancement of SRS in the distributed side-pumped Yb-doped fiber amplifier. In order to reveal the physical mechanism of the phenomenon, the numerical study is carried out. It is found that the Yb-gain at Raman wavelength, although much smaller than that at signal wavelength (i.e., 1080 nm), plays an important role in the phenomenon. It is also revealed that the weak-seed enhancement of SRS can only be presented when the dopant concentration is large enough (e.g., larger than 3 × 1025 in our numerical calculation). Besides, with more effective suppression of Raman seed, the weak-seed induced enhancement of SRS could be observed in a larger seed power range. The pertinent results and conclusion can provide significant guidance for designing high-power fiber lasers and amplifiers.

Portal hypertensive gastropathy (PHG) is a complication of cirrhotic or noncirrhotic portal hypertension. PHG is very important in the clinic because it can cause acute or even massive blood loss, and its treatment efficacy and prognosis are poor. Currently, the incidence of PHG in patients with cirrhosis is 20–80%, but its pathogenesis is complicated and poorly understood. Studies have shown that portal hypertension can cause changes in gastric mucosal microcirculation hemodynamics, leading to changes in gastric mucosal histology and function and thereby weakening the mucosal defense barrier. However, no specific drug treatment plans are currently available. This article reviews the current literature to further our understanding of the mechanism underlying PHG and the relationship between PHG and the posterior mucosal defense barrier and to explore new therapeutic targets.

An aluminum-containing solvate ionic liquid, DMI–AlCl3 (1 M)–MgCl2 (0.5 M) was used as the electrolyte for magnesium electrodeposition. The results of Raman spectroscopy and 27Al nuclear magnetic resonance spectroscopy indicated the existence of aluminum-containing [AlCl4]- and [AlCl2(DMI)4]+ in the DMI–AlCl3 (1 M) system. When 0.5 M MgCl2 was added to DMI–AlCl3, the Mg(II) species was [MgCl(DMI)n]+. X-ray diffraction analysis indicated that a coating of Al3Mg2 can be prepared by potentiostatic electrodeposition at −1.45 V (vs. Al) at 323 K on a copper sheet cathode.

In this paper, a 500-W-level all-fiber amplifier operating near 980 nm is demonstrated firstly, to the best of our knowledge. The amplifier is fabricated with a 105/250-μm core/cladding-diameter double-cladding Yb-doped fiber (DCYF). By optimizing the length of DCYF, the record 556-W output power and 50.5% slope efficiency are obtained with a 1.85-W seed light, and about 30.9 dB peak-to-peak suppression of 1030-nm amplified spontaneous emission (ASE) is realized. Furthermore, the strong in-band ASE is also observed in experiment, which limits the power up-scaling of the fiber amplifier. By analyzing the origin of the in-band ASE, it is revealed that optimizing the seed light should be beneficial to the suppression of in-band ASE. Besides, the beam quality is measured at the maximum output power and the M2 factor is about 23.6. The experimental study should be able to provide significant guidance for understanding and designing the high-power three-level fiber lasers and amplifiers.

In this paper, the scalability of slope efficiency of a Yb-doped fiber amplifier operating near 980 nm is studied with the core-pumping scheme. By means of numerical prediction, it is found that the theoretical limit of slope efficiency should be about 92.2%. Then, the experiment study is carried out. An 85.3% slope efficiency of emission around 980 nm is achieved with the seed light around 976.5 nm, and the strong in-band amplified spontaneous emission (ASE) is supposed to be a factor limiting the upscaling of slope efficiency. In order to suppress the in-band ASE, the double-wavelength fiber oscillator near 980 nm is fabricated and used as the seed source, with which the slope efficiency is elevated to 90.7%. Such slope efficiency is very close to the theoretical limit and sets a new record of slope efficiency for the Yb-doped fiber amplifier operating near 980 nm, to the best of our knowledge. It is also revealed that the suppression of in-band ASE should be of great importance to elevate the slope efficiency of a Yb-doped fiber amplifier operating near 980 nm.

Mutations in WDR62 are the second most common genetic cause of autosomal recessive primary microcephaly, yet the molecular mechanisms underlying this pathogenesis remain unclear. Here, authors demonstrate that WDR62 depletion leads to neural precursor cell depletion and microcephaly via WDR62-CEP170-KIF2A pathway that promotes cilium disassembly.

In this paper, the in-band amplified spontaneous emission (ASE) in the Yb-doped continuous-wave (CW) fiber amplifier operating near 980 nm is experimentally studied for the first time, to the best of our knowledge. A core-pumped single-mode Yb-doped fiber amplifier is fabricated and the effects of pump power, seed power, and active fiber length on the in-band ASE are investigated in the experiment. It is found that a strong in-band ASE around 980 nm can be observed even with no obvious ASE around 1030 nm present. It is also found that with the increment of pump power, the in-band ASE can be enhanced faster than the signal light. By studying the effects of seed power and active fiber length, it is found that, although increasing the seed power and shortening the active fiber can both suppress the in-band ASE, the latter method is less effective than the former one. The theoretical study is also carried out in order to understand the difference between the in-band ASE and 1030-nm ASE. The experimental observations are also discussed qualitatively with the theoretical results. We believe that the pertinent results and discussions can provide significant guidance for understanding the in-band ASE in the Yb-doped fiber amplifier operating near 980 nm.

Genome association studies in human and genetic studies in mouse implicated members of the transmembrane protein 132 (TMEM132) family in multiple conditions including panic disorder, hearing loss, limb and kidney malformation. However, the presence of five TMEM132 paralogs in mammalian genomes makes it extremely challenging to reveal the full requirement for these proteins in vivo. In contrast, there is only one TMEM132 homolog, detonator (dtn), in the genome of fruit fly Drosophila melanogaster, enabling straightforward research into its in vivo function. In the current study, we generate multiple loss-of-function dtn mutant fly strains through a polycistronic tRNA-gRNA approach, and show that most embryos lacking both maternal and paternal dtn fail to hatch into larvae, indicating an essential role of dtn in Drosophila reproduction.

In this work, the Zn–Ta coating was electrodeposited from DMI–ZnCl2–TaCl5 solvate ionic liquid. The electrochemical reaction was studied by cyclic voltammetry using titanium as working electrode. The onset potential of zinc and tantalum co-deposition was −0.7 V (vs. Ag). Moreover, potentiostatic electrodeposition experiments were conducted to investigate the influence of potential and temperature on the morphology of the coating. X-ray photoelectron spectroscopy analysis confirmed that the coating obtained by electrodeposition at −1.6 V (vs. Ag) and 353 K was composed of Zn, Ta, ZnO, and Ta2O5. The mass fraction of element tantalum was 6.21 %, and metallic tantalum accounted for 11.54 % of the mass of element tantalum. The corrosion current density of the Zn–Ta coating was 1.83 × 10−4 A cm−2.

Intu is a planar cell polarity protein known to regulate ciliogenesis during embryonic development. Here, Wang et al. identify a role for Intu in adult kidneys, where they find it promotes degradation of STAT1 and thus prevents cilia loss and cell death upon ischemia-reperfusion injury.

A membrane bioreactor (MBR) was used for treating biological aerated filter effluent in a municipal wastewater plant, and chemical phosphorus removal was accomplished in the MBR. The results showed that ferric chloride of 20 mg/L and aluminum sulfate of 30 mg/L were the optimal dosages for total phosphorus (TP) removal, and the TP removal efficiency was over 80%. In long-term continuous operations, both ferric chloride and aluminum sulfate effectively mitigated membrane fouling, with the corresponding growth rate of transmembrane pressure decreased to 0.08 and 0.067 kPa/d, respectively. Sludge particle sizes analysis demonstrated that the decrease of particle sizes lower than 50 μm was the main reason for membrane fouling control. Simultaneously, the proteins and polysaccharide (PS) concentrations in the MBR supernatant were analyzed, and the PS concentration significantly decreased to 2.02 mg/L at aluminum sulfate of 30 mg/L, indicating the flocculation of aluminum sulfate on PS was the main reason for mitigation of membrane fouling.

An eco-driving strategy is established in this article which includes four parts, namely, initial judging model, speed prediction model based on back-propagation neural network, MATLAB curve fitting, and integral. First, based on vehicle infrastructure cooperative systems, the initial judging model is instructed and vehicle road test is conducted. Then, a speed forecast model based on back-propagation neural network had been set up using test data obtained in the previous step. Next eco-driving strategy had been specified using curve fitting based on the forecast speed data and integral in MATLAB. Finally, a verification test had been done using VISSIM simulation tool. The conclusion of the test showed that using eco-driving strategy was conducive to decrease fuel consumption efficiently when driving at intersection. This article provided a specific case in application of vehicle infrastructure cooperative system to study on fuel consumption and emission in city traffic.

HupZ is an expected heme degrading enzyme in the heme acquisition and utilization pathway in Group A Streptococcus. The isolated HupZ protein containing a C-terminal V5-His6 tag exhibits a weak heme degradation activity. Here, we revisited and characterized the HupZ-V5-His6 protein via biochemical, mutagenesis, protein quaternary structure, UV–vis, EPR, and resonance Raman spectroscopies. The results show that the ferric heme-protein complex did not display an expected ferric EPR signal and that heme binding to HupZ triggered the formation of higher oligomeric states. We found that heme binding to HupZ was an O2-dependent process. The single histidine residue in the HupZ sequence, His111, did not bind to the ferric heme, nor was it involved with the weak heme-degradation activity. Our results do not favor the heme oxygenase assignment because of the slow binding of heme and the newly discovered association of the weak heme degradation activity with the His6-tag. Altogether, the data suggest that the protein binds heme by its His6-tag, resulting in a heme-induced higher-order oligomeric structure and heme stacking. This work emphasizes the importance of considering exogenous tags when interpreting experimental observations during the study of heme utilization proteins.

Interdigitated back contact (IBC) n-type silicon solar cells with a different front surface layer doping concentration were fabricated and studied and the influence of the front surface doping level was analyzed via simulation (PC1D). The IBC cells were processed by industrially feasible technologies including laser ablation and screen printing; photolithography was not used. A maximum efficiency of up to 20.88% was achieved at an optimal front surface field (FSF) peak doping concentration of 4.8 × 1019 cm−3 with a sheet resistance of approximately 95 Ω/square, corresponding to Jsc = 40.05 mA/cm2, Voc = 671 mV and a fill factor of 77.70%. The effects of the front surface doping level were studied in detail by analyzing parameters related to carrier transmission mechanisms such as minority carrier concentration, minority carrier lifetime and the saturation current density of the FSF (J0e). The influence of the front surface recombination velocity (FSRV) on the performance of IBC solar cells with different FSF layer doping concentrations was also investigated and was verified by examining the variation in the minority carrier density as a function of the distance from the front surface. In particular, the impact of the FSF doping concentration on the Jsc of the IBC cells was clarified by considering carrier transmission mechanisms and the charge-collection probability. The trends revealed in the simulations agreed with the corresponding experimental data obtained from the fabricated IBC solar cells. This study not only verifies that the presented simulation is a reasonable and reliable guide for choosing the optimal front surface doping concentration in industrial IBC solar cells but also provides a deeper physical understanding of the impact that front surface layer doping has on the IBC solar cell performance considering carrier transmission mechanisms and the charge-collection probability.

In this work, we propose an analysis approach to determine the individual surface recombination velocities (S1 and S2) on each surface of an unequally passivated wafer, which precludes the crude assumption of S1=S2 in conventional methods. Taking advantage of the surface distributed excess charge carriers relatively sensitive to the surface recombination, we probe the sample using quasi-steady-state illumination of the xenon flash lamp equipped with a short pass filter (FSP1). A set of samples passivated by SiO2 and SiNx, as well as bare silicon wafers, are prepared in the experiment. On the basis of fitting the measured time-dependent-excess charge carriers, S1 and S2 are determined based on our analysis approach. The spatial and the temporal distributions of excess charge carrier density are presented. The dependence of τeff on the wavelength, S and τbulk is also discussed in detail. The reliability of this method is finally verified with a long pass filter (FLP2).

With the exploitation of offshore oil and gas gradually moving to deep water, higher temperature differences and pressure differences are applied to the pipeline system, making the global buckling of the pipeline more serious. For unburied deep-water pipelines, the lateral buckling is the major buckling form. The initial imperfections widely exist in the pipeline system due to manufacture defects or the influence of uneven seabed, and the distribution and geometry features of initial imperfections are random. They can be divided into two kinds based on shape: single-arch imperfections and double-arch imperfections. This paper analyzed the global buckling process of a pipeline with 2 initial imperfections by using a numerical simulation method and revealed how the ratio of the initial imperfection's space length to the imperfection's wavelength and the combination of imperfections affects the buckling process. The results show that a pipeline with 2 initial imperfections may suffer the superposition of global buckling. The growth ratios of buckling displacement, axial force and bending moment in the superposition zone are several times larger than no buckling superposition pipeline. The ratio of the initial imperfection's space length to the imperfection's wavelength decides whether a pipeline suffers buckling superposition. The potential failure point of pipeline exhibiting buckling superposition is as same as the no buckling superposition pipeline, but the failure risk of pipeline exhibiting buckling superposition is much higher. The shape and direction of two nearby imperfections also affects the failure risk of pipeline exhibiting global buckling superposition. The failure risk of pipeline with two double-arch imperfections is higher than pipeline with two single-arch imperfections.

Dynamic characteristic analysis of a two-stand reversible cold rolling mill in the startup process was carried out. The delay algorithm of the interstand thickness was proposed. A new method combined with the accelerated secant and the tangent methods was established to solve the simultaneous equations. The thickness and interstand tension transition processes with different static tension establishing processes were analyzed. Both mills were operated under constant rolling force control mode in the above process. The results show that the strip thickness in the rolling gap reduces in the static mill screwdown process. The entry stand runs inversely to establish the static interstand tension. This area becomes an abnormal thickness reduction area of the incoming strip. It results in several abnormal interstand tension increases in the subsequent startup process. The tension increase leads to an impact force on the strip that is the main reason of the strip breakage in the startup process. So the static tension establishing process was optimized, and the interstand tension fluctuation and the strip breakage accidents both reduced significantly. The results are beneficial to the startup process of the two-stand reversible cold rolling mill.

Fe-Ni-based alloys are promising materials of inert anodes for use in aluminum electrolysis and adding Al can further improve the corrosion resistance. Fe-Ni-Al alloys with 1.4–8.6 wt.% Al were prepared by vacuum melting, and their corrosion as anodes during the production of pure Al (98.14–99.68%) by electrolysis was studied in a melt of NaF-AlF3-NaCl-CaF2-Al2O3 at 850 °C. The corrosion layer on the anode contains fluorine salt that corrodes the oxide film, and the inner layer is Ni-enriched while the outer layer is enriched with Fe and O due to the preferential oxidation of Fe. The electrolytically deposited oxide films on Fe-Ni-Al alloys with different compositions contains Fe2O3, Fe3O4, NiO, Al2O3, FeAl2O4, NiFe2O4, and other protective oxides, making the alloys very corrosion-resistant. The linear voltammetric curves can be divided into three parts: active dissolution, passivation transition, and over-passivation zones. The alloy with 3.9 wt.% Al (57.9Fe-38.2Ni-3.9Al) has a relatively negative passivation potential, and therefore, is easier to become passivated. According to the Tafel curve, this alloy shows a relatively positive corrosion potential as anode (1.20 V vs. Al/AlF3), and thus can form a protective film.

The primary cilia play essential roles in Hh-dependent Gli2 activation and Gli3 proteolytic processing in mammals. However, the roles of the cilia in Gli1 activation remain unresolved due to the loss of Gli1 transcription in cilia mutant embryos, and the inability to address this question by overexpression in cultured cells. Here, we address the roles of the cilia in Gli1 activation by expressing Gli1 from the Gli2 locus in mouse embryos. We find that the maximal activation of Gli1 depends on the cilia, but partial activation of Gli1 by Smo-mediated Hh signaling exists in the absence of the cilia. Combined with reduced Gli3 repressors, this partial activation of Gli1 leads to dorsal expansion of V3 interneuron and motor neuron domains in the absence of the cilia. Moreover, expressing Gli1 from the Gli2 locus in the presence of reduced Sufu has no recognizable impact on neural tube patterning, suggesting an imbalance between the dosages of Gli and Sufu does not explain the extra Gli1 activity. Finally, a non-ciliary Gli2 variant present at a higher level than Gli1 when expressed from the Gli2 locus fails to activate Hh pathway ectopically in the absence of the cilia, suggesting that increased protein level is unlikely the major factor underlying the ectopic activation of Hh signaling by Gli1 in the absence of the cilia.

OBJECTIVES: Familial steroid-sensitive idiopathic nephrotic syndrome is rare, and only approximately 3% of patients have affected siblings. METHODS: Herein, we report seven cases of patients with steroid-sensitive idiopathic nephrotic syndrome from three Chinese families. Mutational screening of the Nphs2 gene was performed in all the patients. RESULTS: All seven of the familial steroid-sensitive idiopathic nephrotic syndrome cases in our sample exhibited minimal change disease, and one case also presented with mesangial proliferative glomerulonephritis, according to the renal pathology. No significant was associations were found between Nphs2 gene mutations and the onset of proteinuria and nephrotic syndrome in these familial cases. CONCLUSIONS: The presence of minimal change disease is important, but it is not an unusual finding in patients with familial steroid-sensitive idiopathic nephrotic syndrome, which appears to be clinically benign and genetically distinct from other types of nephrosis.

The purpose of this study was to investigate the potential effects of Huaiqihuang (HQH) granule, a Chinese herbal medicine, in treating proteinuria and to reveal its possible mechanism. MPC5 podocytes were cultured in vitro at 37°C and induced with tunicamycin (TM). The TM-induced cells were treated with HQH at different concentrations. The cell proliferation was detected using the MTT assay. The optimal effective dose of HQH for MPC5 cells was determined by the MTT assay and LDH assay respectively. The influences of HQH on the proteinuria-related protein expression and the signaling pathway associated protein expression were also detected using quantitative reverse transcription PCR and Western blotting analysis. The results showed that the MPC5 cell model was successfully constructed in vitro. The HQH application could improve the harmful effects induced by TM on the MPC5 cells, including promoted cell proliferation and suppressed cell apoptosis. Furthermore, the protein expression, including podocin, nephrin, and synaptopodin was down-regulated by the TM treatment in the MPC5 cells. On contrary, the expression of these proteins was up-regulated after the HQH application. Also, the effect of TM on integrin α3 and integrin β1 expressions was also reversed by the HQH treatment. Moreover, the HQH application decreased the expression of p-ERK and DNA-damage-inducible transcript 3 (DDIT3 or CHOP) in the MPC5 cells, which was opposite to the effect observed in the cells treated with TM. Taken together, our study suggest that HQH application may protect podocytes from TM damage by suppressing the p-ERK/CHOP signaling pathway.

This work is concerned with the quadratic-mean asymptotically almost periodic mild solutions for a class of stochastic functional differential equations dxt=Atxt+Ft,xt,xtdt+H(t,xt,xt)∘dW(t). A new criterion ensuring the existence and uniqueness of the quadratic-mean asymptotically almost periodic mild solutions for the system is presented. The condition of being uniformly exponentially stable of the strongly continuous semigroup {Tt}t≥0 is essentially removed, which is generated by the linear densely defined operator A∶D(A)⊂L2(ℙ,ℍ)→L2(ℙ,ℍ), only using the exponential trichotomy of the system, which reflects a deeper analysis of the behavior of solutions of the system. In this case the asymptotic behavior is described through the splitting of the main space into stable, unstable, and central subspaces at each point from the flow’s domain. An example is also given to illustrate our results.

The performance of black silicon solar cells with various passivation films was characterized. Large area (156×156 mm2) black silicon was prepared by silver-nanoparticle-assisted etching on pyramidal silicon wafer. The conversion efficiency of black silicon solar cell without passivation is 13.8%. For the SiO2 and SiNx:H passivation, the conversion efficiency of black silicon solar cells increases to 16.1% and 16.5%, respectively. Compared to the single film of surface passivation of black silicon solar cells, the SiO2/SiNx:H stacks exhibit the highest efficiency of 17.1%. The investigation of internal quantum efficiency (IQE) suggests that the SiO2/SiNx:H stacks films decrease the Auger recombination through reducing the surface doping concentration and surface state density of the Si/SiO2 interface, and SiNx:H layer suppresses the Shockley-Read-Hall (SRH) recombination in the black silicon solar cell, which yields the best electrical performance of b-Si solar cells.

BackgroundPodocytes serve as an important constituent of the glomerular filtration barrier. Recently, we and others identified Myo1e as a key molecular component of the podocyte cytoskeleton.ResultsMyo1e mRNA and protein was expressed in human and mouse kidney sections as determined by Northern blot and reverse transcriptase PCR, and its expression was more evident in podocytes by immunofluorescence. By specific knock-down of MYO1E in zebrafish, the injected larvae exhibited pericardial edema and pronephric cysts, consistent with the appearance of protein in condensed incubation supernate. Furthermore, specific inhibition of Myo1e expression in a conditionally immortalized podocyte cell line induced morphological changes, actin cytoskeleton rearrangement, and dysfunction in cell proliferation, migration, endocytosis, and adhesion with the glomerular basement membrane.ConclusionsOur results revealed that Myo1e is a key component contributing to the functional integrity of podocytes. Its impairment may cause actin cytoskeleton re-organization, alteration of cell shape, and membrane transport, and podocyte drop-out from the glomerular basement membrane, which might eventually lead to an impaired glomerular filtration barrier and proteinuria.

Intracellular transduction of Hedgehog (Hh) signals in mammals requires functional primary cilia. The Hh signaling effectors, the Gli family of transcription factors, and their negative regulator, Suppressor of Fused (Sufu), accumulate at the tips of cilia; however, the molecular mechanism regulating this localization remains elusive. In the current study, we show that the ciliary localization of mammalian Gli proteins depends on both their N-terminal domains and a central region lying C-terminal to the zinc-finger DNA-binding domains. Invertebrate Gli homologs Ci and Tra1, when over-expressed in ciliated mouse fibroblasts, fail to localize to the cilia, suggesting the lack of a vertebrate-specific structural feature required for ciliary localization. We further show that activation of protein kinase A (PKA) efficiently inhibits ciliary localization of Gli2 and Gli3, but only moderately affects the ciliary localization of Gli1. Interestingly, variants of Gli2 mimicking the phosphorylated or non-phosphorylated states of Gli2 are both localized to the cilia, and their ciliary localizations are subjected to the inhibitory effect of PKA activation, suggesting a likely indirect mechanism underlying the roles of PKA in Gli ciliary localization. Finally, we show that ciliary localization of Sufu is dependent on ciliary-localized Gli proteins, and is inhibited by PKA activation, suggesting a coordinated mechanism for the ciliary translocation of Sufu and Gli proteins.

OBJECTIVE: To investigate the outcomes of childhood diffuse endocapillary proliferation Henoch-Schönlein purpura nephritis (DEP-HSPN) in response to early diagnosis and prompt treatment. METHODS: Eleven cases of DEP-HSPN in children were investigated in comparison to HSPN without diffuse endocapillary proliferation (non-DEP-HSPN). RESULTS: DEP-HSPN had a higher prevalence of nephrotic syndrome but a lower prevalence of hematuria compared to non-DEP-HSPN. IgA, IgG and IgM antibody deposition was found in DEP-HSPN by histopathological examination. Proteinuria cleared in all 11 cases through treatment with steroids and/or immunosuppressive drugs. However, half of the DEP-HSPN patients continuously had hematuria after treatment. CONCLUSION: The early diagnosis and prompt initiation of immunosuppressive treatment based on renal biopsy are important for achieving favorable outcomes.
