4. Investigating the Protective Role of L-carnitine and Thymoquinone against Methotrexate-Induced Testicular Damage in Rats.

Abstract

The research aimed to investigate whether the combination of L-carnitine (LC) and thymoquinone (TQ) could lower the adverse effects of methotrexate (MTX), a widely used chemotherapy agent in cancer therapy, on testicular function. A total of seven groups were established (n=7); control group, LC (200 mg/kg PO), TQ (50 mg/kg PO), MTX (single IP dose of 20 mg/kg), LC+MTX, TQ+MTX, and LC+TQ+MTX. The use of MTX resulted in lower levels of testosterone, folliclestimulating hormone (FSH), and luteinizing hormone (LH) in the blood, accompanied by a significant rise in testicular MDA levels. Additionally, there was a notable decline in GSH, CAT, and SOD, indicating reduced antioxidant activity in the testicles. Furthermore, the histopathological examination of the testicles revealed disruptions caused by MTX, and the expression of caspase-3 in the testicles was documented. The combination of LC and TQ with MTX significantly increased testosterone, FSH, LD levels and restored oxidative damage markers (p<0.05) and improved histopathological scores and caspase-3 expression. In conclusion, our results demonstrate that LC and TQ exhibit substantial protective effects against testicular toxicity induced by MTX, primarily attributed to their antioxidant properties. Abstract

Backgorund and Objective:Acute upper GIT bleeding is a life threatening emergency. Many scoring systems have been developed to evaluate patients with acute UGIB. The purpose of this study was to confirm value of Modified Blatchford scoring system, Full rockall score , AIM65 score &lactate level as a predictor of outcome as regard patient’s mortality, their need for ICU, duration of hospitalization and need for endoscopic intervention in the emergency department. Patients an Methods:This study was performed on 100 patients who presented to the emergency department with acute UGIB in Alexandria main university hospital. All the study scores (GBS, AIMS65, PRS, and FRS) were able to predict the clinical outcomes in the UGIB patients. Results:GBS was the best performing risk score among the four scores for predicting all the clinical outcomes except the prediction of endoscopic intervention. Lactate was a sensitive predictor for ICU admission, hospital stay, rebleeding and mortality during first 48 hrs of admission in patients presenting with UGIB.

Abstract

This thesis is placed in the context of Employer Attractiveness and Employee Retention within the Pharmaceutical Industry of Egypt and Pakistan. Most of the studies conducted in the past are either specifically done on Employer Attractiveness, Employer Branding, or a combination of both, indicating a lack of research between employer attractiveness and employee retention; thus, this thesis will bridge the gap between them and will be a great contribution. The purpose of this thesis is to identify what factors attract potential employees from Egypt and Pakistan to work in the Pharmaceutical Industry and, after gaining employment, what factors retain them to keep working in the industry. The study also indicates the disappointment in the attractiveness factors by the employees after employment. The empirical data was collected through a survey with structured questionnaires. A response of 103 was collected, 50 from Egypt and 53 from Pakistan, and the responses were analyzed using a quantitative method. The findings from the empirical data indicated a positive correlation between the Attractiveness/Retention factors with Employer Attractiveness and Employee Retention in both the countries. Moreover, the regression model explained what Attractiveness/Retention factors were significant in explaining Employer Attractiveness and Employee Retention. We can conclude from our analysis that the survey results are in line with what we expected to see based on our review of previous research and our analytical model. The top attractive factors are not necessarily the same as the top retention factors, according to the study. The results also indicated that compensation and benefits in both countries are not a significant retention factor in retaining employees. HR professionals should also be mindful of the variances in branding considerations. As a result, it's vital to comprehend employees' personal lives and backgrounds in order to employ the appropriate retention strategies.

Abstract

The Pseudomonas putida strain was primarily identified and tested in vitro against antibiotic sensitivity for several antibiotics using the disc diffusion method. This isolate was also tested against sensitivity to carvacrol oil (c) and formic acid (f). The genotyping of Pseudomonas spp. and virulotyping for P. putida isolate was carried out and verified by 16S rDNA-PCR amplification. Furthermore, we assessed the efficacy of carvacrol oil and formic acid in vivo for treatment of P. Putida infection. For the in vivo challenge, 180 fish (Nile tilapia, Oreochromis niloticus) were divided into six groups: (G1: control (unchallenged), G2: carvacrol prophylaxis (3 g/kg), G3: formic acid prophylaxis (5 mL/kg), G4: control positive (challenged), G5: carvacrol treatment (3 g/kg), and G6: formic acid treatment (5 mL/kg); 30 fish per group) with three replicates. Following the challenge, nitric oxide and lysozyme activity were measured as essential indicators for fish immunity. The antioxidant parameters including SOD and catalase were computed to reflect the antioxidant status. Furthermore, relative percent survival (RPS) and mortality percent were evaluated to indicate functional immunity. The findings of the antibiotic sensitivity test showed that ciprofloxacin exhibited the largest inhibition zone. Additionally, formic acid (f) displayed the greatest inhibition zone compared to carvacrol oil (c) and was more effective in stimulating the immune-antioxidant response compared to carvacrol oil. The tested exotoxin A (tox A), exoenzyme S (exo S), and the nan1 associated-virulence genes were identified in the P. putida isolate. Overall, the current study verified the virulence of P. putida and highlighted the promising role of dietary addition of formic acid for enhancing the immune-antioxidant indicators and for mitigating P. putida infection. Abstract

The concept of probiotics is widely applied in the field of aquaculture for their beneficial and friendly influences. In this sense, the role of Saccharomyces cerevisiaeon the growth, immune and antioxidative responses of European seabass was tested in this study. Fish were distributed in 3 groups (triplicates) with ten fish in each replicate and fed 0, 1, and 2 g/kg of S. cerevisiae(15×109CFU/g) for 90 days. Fish fed S. cerevisiaeshowed higher final body, weight gain, and specific growth rate as well as lower FCR than fish fed the basal diet (P<0.05). The RBCs, WBCs, Hb, and PCV values were increased in fish fed dietary S. cerevisiaewhen compared to the control (P<0.05). The blood total protein, albumin, and globulin were higher in fish fed S. cerevisiaethan the control (P<0.05). Fish fed dietary S. cerevisiaehad enhanced phagocytic index, phagocytic, and lysozyme activity comparing the control. In a similar sense, the antioxidative enzymes (SOD, GPx, and CAT) were higher in fish fed S. cerevisiaethan the control (P<0.05). However, the level of MDA was lowered (P<0.05) by S. cerevisiaein European seabass. Gene expression of IL-8, IL-1β, GH, and IGF-1was upregulated and HSP70was downregulated by S. cerevisiae(P<0.05). It can be concluded that European seabass fed S. cerevisiaeat 1–2 g/kg (15×109CFU/g) diet had markedly enhanced growth, haemato-biochemical, and immune performances.

Abstract

Background: As different biogeographic strains and isolates of entomopathogenic fungi vary in their genetic, enzymatic and pathogenic characteristics, this study assessed the virulence of 2 indigenous strains of Beauveria bassiana(Balsam) Vuillemin and Metarhizium anisopliae(Metschn.) Sorokin (Ascomycota, Hypocreales: Clavicipitaceae), isolated from naturally infected insect cadavers, against the 3rd instar nymphs of Myzus persicae(Sulzer) (Hemiptera: Aphididae) and 3rdinstar larvae of Spodoptera frugiperda(J.E. Smith) (Lepidoptera: Noctuidae) using leaf-dip and larval-dip methods, respectively. Results: Both fungal isolates exhibited considerable pathogenicity against M. persicaeand S. frugiperda. Mortality in all bioassays was conidial concentration and exposure time dependent and increased significantly along with both factors (R2= 0.86–0.99 for B. bassianaand 0.82–0.94 for M. anisopliae). Moreover, M. anisopliaeisolate appeared more virulent to S. frugiperdalarvae than B. bassianaisolate, while the later fungal isolate was more lethal to M. persicaenymphs than the former one. At the highest conidial concentration (1.0 × 109conidia/ml), M. anisopliaecaused maximum mean mortality of S. frugiperda(88%) and M. persicae(65%) and B. bassianaexhibited maximum mean mortality of S. frugiperda(76%) and M. persicae(94%). Moreover, probit regression analyses showed LT50values for M. persicaeof 4.57 and 6.86 days at 1.0 × 109conidia/ml for the isolates of B. bassianaand M. anisopliae, respectively, while LC50values were 7.75 × 106and 8.70 × 107conidia/ml after 10th day of application, for the isolates of B. bassianaand M. anisopliae, respectively, against M. persicae. Similarly, LT50values for S. frugiperdawere 7.75 and 7.03 days for 1.0 × 109conidia/ml concentration and LC50values were 2.84 × 107and 8.84 × 105conidia/ml at 10th day data for the isolates of B. bassianaand M. anisopliae, respectively. Conclusion: Overall study results demonstrated the effectiveness of B. bassianaand M. anisopliaeagainst M. persicaeand S. frugiperda, respectively. However, field evaluations of these indigenously isolated promising fungal strains against these insect pests.

Abstract

Cisplatin (CP) is one of antineoplastic agents with a broad range of anticancer activities. This research investigated the possible protective effects of lycopene (LP) and N acetylcysteine (NAC) in rats against CP-induced cardiac toxicity. Seven groups of rats (n=7); control vehicle group was administered saline, LP (10 mg/kg, PO), NAC (150 mg/kg, PO), CP group (7.5 mg/kg, IP) on the 27th day of the experiment, LP-CP group, NAC-CP group, and LP-NAC-CP group. Following injection of CP, concentrations of cardiac biomarkers, lactate dehydrogenase (LDH), creatine kinase (CK) and creatine kinase MB (CK-MB) were increased in serum of the treated groups. In addition, CP resulted in a significant increase in malondialdehyde (MDA), as well as significant decreases in glutathione (GSH), superoxide dismutase (SOD) and catalase (CAT) in rats' heart tissues. CP resulted in changes in cardiac histopathology and increased caspase-3 expression in cardiac tissues. Abstract

Methotrexate (MTX) is a chemotherapeutic medicine frequently used to treat various forms of cancer. The objective of our research was to examine whether allicin (ALC) and lycopene (LP) could alleviate the harmful effects of MTXinduced testicular toxicity in rats. A total of 49 male white albino rats were divided into 7 groups (7 rats in each group). The rats of group 1 was kept as a control group and received normal saline orally; while rats of group 2 and group 3 administered ALC (20mg/kg/BW) LP (10mg/kg/BW) orally. On day 15, the rats of group 4 received a single dose of MTX (20mg/kg, IP). In group 5, rats received ALC and MTX; in group 6, rats received LP and MTX, and in group 7, rats received ALC, LP, and MTX. Results showed that MTX induced significant increase in values of testicular malondialdehyde (MDA) levels with a substantial reduction in glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), and lowered the serum testosterone, follicle-stimulating hormone (FSH), and lutenizing hormone (LH) levels. MTX induced a marked decline in seminiferous tubule diameter and epithelium height. MTX showed a widening of the interstitial space of seminiferous tubules, atrophy, vacuolar degeneration of the germinal epithelium and Leydig's interstitial cells had scanty cytoplasm and vesicular nuclei. Also, testicular proliferating cell nuclear antigen (PCNA) expression was recorded. In MTX-intoxicated rats, ALC and LP increased antioxidant biomarkers, decreased MDA, and attenuated changes in serum hormonal parameters.
