Abstract

We examine several conducting spheres moving through a magnetic field gradient. An analytical approximation is derived and an experiment is conducted to verify the analytical solution. The experiment is simulated as well to produce a numerical result. Both the low and high magnetic Reynolds number regimes are studied. Deformation of the sphere is noted in the high Reynolds number case. Abstract

MUSTANG is a 90 GHz bolometer camera built for use as a facility instrument on the 100 m Robert C. Byrd Green Bank radio telescope (GBT). MUSTANG has an 8 by 8 focal plane array of transition edge sensor bolometers read out using time-domain multiplexed SQUID electronics. As a continuum instrument on a large single dish MUSTANG has a combination of high resolution (8") and good sensitivity to extended emission which make it very competitive for a wide range of galactic and extragalactic science. Abstract

We present performance results based on the first astronomical use of multiplexed superconducting bolometers. The Fabry-Perot Interferometer Bolometer Research Experiment (FIBRE) is a broadband submillimeter spectrometer that achieved first light in June 2001 at the Caltech Submillimeter Observatory (CSO). FIBRE's detectors are superconducting transition edge sensor (TES) bolometers read out by a SQUID multiplexer. The Fabry-Perot uses a low resolution grating to order sort the incoming light. A linear bolometer array consisting of 16 elements detects this dispersed light, capturing 5 orders simultaneously from one position on the sky. With tuning of the Fabry-Perot over one free spectral range, a spectrum covering Delta lambda/lambda = 1/7 at a resolution of delta lambda/lambda approx. 1/1200 can be acquired. This spectral resolution is sufficient to resolve Doppler-broadened line emission from external galaxies. FIBRE operates in the 350 m and 450 m bands. These bands cover line emission from the important star formation tracers neutral carbon (CI) and carbon monoxide (CO). We have verified that the multiplexed bolometers are photon noise limited even with the low power present in moderate resolution spectrometry.

Abstract

Carex roanensis is a globally rare species endemic in moderate to high elevation forests in the southern Appalachian Mountains. All known collections were reviewed, including previously unpublished records from Pennsylvania, West Virginia, Virginia, North Carolina, and Tennessee, and an annotated distribution map is presented. A preliminary assessment of its ecological affinities is provided, especially with respect to the closely related C. aestivalis and C. virescens.

Abstract

1. Many physiologically important activities of oligodendrocyte progenitor cells (O-2A cells), including proliferation, migration and differentiation, are regulated by cytosolic Ca2+ signals. However, little is known concerning the mechanisms of Ca2+ signalling in this cell type. We have studied the interactions between Ca2+ entry, Ca2+ release from endoplasmic reticulum and Ca2+ regulation by mitochondria in influencing cytosolic Ca2+ responses in O-2A cells. 2. Methacholine (MCh; 100 microM) activated Ca2+ waves that propagated from several initiation sites along O-2A processes. 3. During a Ca2+ wave evoked by MCh, mitochondrial membrane potential was often either depolarized (21 % of mitochondria) or hyperpolarized (20 % of mitochondria), as measured by changes in the fluorescence of 5,5',6,6'-tetrachloro-1,1',3, 3'-tetraethylbenzimidazole carbocyanine iodide (JC-1). 4. Stimulation with kainate (100 microM) evoked a slowly rising, sustained cytosolic Ca2+ elevation in O-2A cells. This also, in some cases, resulted in either a depolarization (15 % of mitochondria) or hyperpolarization (12 % of mitochondria) of mitochondrial membrane potential. 5. Simultaneous measurement of cytosolic (fluo-3 AM) and mitochondrial (rhod-2 AM) Ca2+ responses revealed that Ca2+ elevations in the cytosol evoked by either MCh or kainate were translated into long-lasting Ca2+ elevations in subpopulations of mitochondria. In some mitochondria, Ca2+ signals appeared to activate Ca2+ release into the cytosol. 6. Inhibition of the mitochondrial Na+-Ca2+ exchanger by CGP-37157 (25 microM) decreased kainate Ca2+ response amplitude and increased the rate of return of the response to basal Ca2+ levels. 7. Thus, both ionotropic and metabotropic stimulation evoke changes in mitochondrial membrane potential and Ca2+ levels in O-2A cells. Ca2+ uptake into some mitochondria is activated by Ca2+ entry into cells or release from stores. Mitochondrial Ca2+ release appears to play a key role in shaping kainate-evoked Ca2+ responses.

Abstract

The effect of bovine respiratory syncytial virus (BRSV) upon alveolar macrophage (AM) function was investigated using an in vivo calf inoculation model. Alveolar macrophages were collected sequentially from live calves at multiple time points during the 14 day period following viral inoculation. Alveolar macrophages from bronchoalveolar lavage fluids were purified by density gradient centrifugation (> 95% AM) prior to in vitro evaluation of cell functions. There were significant but variable and inconsistent differences in the functions of AM from the BRSV inoculated calves compared to the control calves. Fc-receptor mediated phagocytosis was either increased or unchanged by BRSV inoculation. Nonopsonized phagocytosis was decreased during the early postinoculation period and later increased. There was a variable effect on AM phagosome lysosome fusion with increased fusion activity on postinoculation days 2 through 5, 7 and 12 but reduced activity on days 6 and 10. The AM respiratory burst, as measured by nitroblue tetrazolium dye reduction, was essentially unaffected with a reduction in activity on day 10 only. In this model, BRSV inoculation of calves primarily resulted in an alteration of the membrane associated phagocytic functions of the alveolar macrophages (p < 0.05).

Tyler D Ames,1 Corinne E Wee,1 Khoi M Le,1 Tiffany L Wang,1 Julie Y Bishop,2 Laura S Phieffer,2 Carmen E Quatman2 1The Ohio State University College of Medicine, 2Department of Orthopaedics, The Ohio State University Wexner Medical Center, Columbus, OH, USA Purpose: To identify inexpensive, noninvasive, portable, clinical assessment tools that can be used to assess functional performance measures that may put older patients at risk for falls such as balance, handgrip strength, and lumbopelvic control.Patients and methods: Twenty fragility fracture patients and 21 healthy control subjects were evaluated using clinical assessment tools (Nintendo Wii Balance Board [WBB], a handheld dynamometer, and an application for the Apple iPod Touch, the Level Belt) that measure functional performance during activity of daily living tasks. The main outcome measurements were balance (WBB), handgrip strength (handheld dynamometer), and lumbopelvic control (iPod Touch Level Belt), which were compared between fragility fracture patients and healthy controls.Results: Fragility fracture patients had lower scores on the vertical component of the WBB Torso Twist task (P=0.042) and greater medial–lateral lumbopelvic sway during a 40 m walk (P=0.026) when compared to healthy controls. Unexpectedly, the fracture patients had significantly higher scores on the left leg (P=0.020) and total components (P=0.010) of the WBB Single Leg Stand task as well as less faults during the left Single Leg Stand task (P=0.003).Conclusion: The clinical assessment tools utilized in this study are relatively inexpensive and portable tools of performance measures capable of detecting differences in postural sway between fragility fracture patients and controls. 27. Detection of immunoglobulin G to Pasteurella haemolytica capsular polysaccharide by enzyme-linked immunosorbent assay

Abstract

An enzyme-linked immunosorbent assay (ELISA) was developed for the detection of immunoglobulin G (IgG) to the capsular polysaccharide (CP) of Pasteurella haemolytica serotype 1. Purified CP was first covalently coupled to poly-L-lysine and then optimally adsorbed at a concentration of 5 micrograms/ml to microtiter plates in the presence of carbonate-bicarbonate buffer (pH 9.8). The ELISA was used to evaluate and compare the CP-specific IgG response of calves vaccinated with different P. haemolytica-derived experimental vaccines. Elevated levels of ELISA IgG titers were detected in postvaccination sera and lung lavage from calves vaccinated intradermally with live logarithmic-phase organisms or the culture supernatants. The ELISA was found to be a rapid, reproducible, and sensitive technique for the detection of CP-specific antibodies and may be useful to delineate the protective role of these antibodies in bovine pneumonic pasteurellosis.

Abstract

Calves were inoculated intratracheally with 5 X 10(7), 5 X 10(8), or 5 X 10(9) colony forming units of either 18-hour stationary phase cultures or 4-hour log phase cultures of Pasteurella haemolytica. The log phase culture at all concentrations produced more severe clinical signs, hematological changes and pulmonary lesions at postmortem examination than did the corresponding stationary phase culture. More severe effects were seen with the larger doses especially with the log phase culture. Fibrinous bronchopneumonia with focal or multifocal necrosis was consistently produced by both the stationary and log phase cultures. To determine if this lesion was peculiar to P. haemolytica or whether it could be produced generally by rapidly growing Gram negative organisms, a 4-hour log phase culture of Pasteurella multocida was prepared in an identical manner to that used for the culture of P. haemolytica and given to calves intratracheally at the high bacterial dose (5 X 10(9]. The P. haemolytica produced more severe clinical, hematological and morphological changes than did the P. multocida. The lesions observed with P. multocida differed morphologically from those of P. haemolytica; there was a suppurative exudative component and minimal to no necrosis with P. multocida. It appears that an important pathogenic principle is produced by the rapidly growing P. haemolytica that causes it to produce a more severe clinical disease and more necrotizing pulmonary lesions than P. multocida.

Abstract

An extensive materials and equipment investigation was conducted in response to an U. S. Air Force requirement which specified the development of improved materials and methods to provide a rapid and permanent runway repair capability. Materials studies, tests, and evaluation covered both organic and inorganic materials. Both were found to meet the contract requirements, but an inorganic compound containing calcium sulfate hemihydrate, cement, TF-4 and water was selected as the most satisfactory end product. This material was selected on the basis of an impartial grading system. Both the organic and inorganic field tests included set time checks and load applications within 10 minutes to several hours after application of the mixture over various subsoil and fill materials, under specified environmental conditions. Paralleling materials testing, small-scale mixing and pumping machines were developed and built. Material set-up rates were adjusted to approximately 12 minutes. Slurry viscosity was reduced to the required pumping level by varying the TF-4 additive. The Fast-Fix formula and equipment will undergo further tests, including actual bomb crater repairs at Eglin Air Force Base, allowing Air Force technicians to operate the equipment. Details of these tests and further Fast-Fix improvement will be provided in Volume II of this report. (Author)

In September 2006 MUSTANG had first light and became the first instrument to use the GBT at 90 GHz. During two months of subsequent testing we showed that MUSTANG behaved as well on the telescope as it did in the lab. Observations were made of many astronomical sources including Saturn and the star forming region W3. We were able to confirm our beam shape and different observing strategies were tested. Procedures were put in place for pointing and focusing and a data analysis pipeline was set up. Several problems were identified during commissioning and are currently being addressed. One of these, excessive 1/f noise, has already been solved and we plan to take our first science observations in late 2007.

Abstract

MUSTANG is a 90 GHz bolometer camera built for use as a facility instrument on the 100 m Robert C. ByrdGreen Bank radio telescope (GBT). MUSTANG has an 8 by 8 focal plane array of transition edge sensorbolometers read out using time-domain multiplexed SQUID electronics. As a continuum instrument on a largesingle dish MUSTANG has a combination of high resolution (8) and good sensitivity to extended emissionwhich make it very competitive for a wide range of galactic and extragalactic science. Commissioning finishedin January 2008 and some of the first science data have been collected.

Abstract

MUSTANG, the MUltiplexed SQUID TES Array at Ninety GHz, is an 8 by 8 array of TES bolometers designed as a user instrument for the 100 m diameter Green Bank radio telescope (GBT). MUSTANG is the GBT’s first 90 GHz instrument and its first focal plane array. As a continuum receiver on a large single dish, MUSTANG offers a unique combination of 8 arcsecond angular resolution and high sensitivity to extended structure. In this poster we present an overview of MUSTANG. Observations of a range of objects including M87 and the Crab nebula will be used to illustrate MUSTANG's capabilities. Our first scientific observations in the spring of 2008 were of the Orion star forming region. These results will be presented in greater detail in a separate talk by Philip Korngut. Currently, work is under way to improve the high frequency performance of the GBT and there are plans to increase MUSTANG's sensitivity further. A paper containing more technical details will appear in the proceedings of the SPIE conference on astronomical instrumentation (Dicker et. al. 2008). MUSTANG will be available for proposals by the general astronomical community in the winter high frequency season of 2008/2009. Calls for proposals will be announced at NRAO's website, www.gb.nrao.edu.

This data, obtained with the 100m Green Bank Telescope yields a high resolution (8") map while maintaining good sensitivity to extended structure. This observation of Orion at a wavelength of 3mm is uniquely sensitive to both the free-free emission from the nebular gas ionized by the O stars in the Trapezium and thermal dust emission from the dense molecular cloud OMC 1. Combined with other data sets of comparable spatial resolution (e.g. VLA and/or GBT continuum for the free-free and SCUBA 450 and 850microns, BOLOCAM 1mm for the thermal dust emission), spectral decomposition of the MUSTANG map will be used to study the dust emissivity and its possible variation through this region which is among the most active sites of star formation near the Sun.

Abstract

Abstract Background Metagenomic sequence data are proving to be a vast resource for the discovery of biological components. Yet analysis of this data to identify functional RNAs lags behind efforts to characterize protein diversity. The genome of 'Candidatus Pelagibacter ubique' HTCC 1062 is the closest match for approximately 20% of marine metagenomic sequence reads. It is also small, contains little non-coding DNA, and has strikingly low GC content. Results To aid the discovery of RNA motifs within the marine metagenome we exploited the genomic properties of 'Cand. P. ubique' by targeting our search to long intergenic regions (IGRs) with relatively high GC content. Analysis of known RNAs (rRNA, tRNA, riboswitches etc.) shows that structured RNAs are significantly enriched in such IGRs. To identify additional candidate structured RNAs, we examined other IGRs with similar characteristics from 'Cand. P. ubique' using comparative genomics approaches in conjunction with marine metagenomic data. Employing this strategy, we discovered four candidate structured RNAs including a new riboswitch class as well as three additional likely cis-regulatory elements that precede genes encoding ribosomal proteins S2 and S12, and the cytoplasmic protein component of the signal recognition particle. We also describe four additional potential RNA motifs with few or no examples occurring outside the metagenomic data. Conclusion This work begins the process of identifying functional RNA motifs present in the metagenomic data and illustrates how existing completed genomes may be used to aid in this task.

Abstract

Proliferative enteropathy (PE) is an intestinal disease that affects equine and several other species. The goal of this study is to determine whether proliferative enteropathy could be experimentally reproduced in foals. Seven foals were divided into two groups: A (n=5), and B (n=2). The foals were inoculated intragastrically with porcine intestinal mucosal homogenate (group A), or a placebo challenge (group B). Ante mortem and post mortem tests were performed. All foals were euthanised on day 22 post challenge. Physical examination detected variable signs of depression, colic, decreased appetite, diarrhoea, dehydration and emaciation in three foals in group A. At necropsy, one foal (foal 5, group A) was severely emaciated, the wall of the ileum and distal jejunum was thickened and the mucosa was corrugated and hyperaemic. Histologically, hyperplasia of immature enterocytes and reduced number of Paneth and goblet cells were observed. PCR confirmed the presence of Lawsonia intracellularis. The foals in groups A were positive for L. intracellularis by immunohistochemistry. The control foals (group B) remained physically normal, had no pathologic lesions and were negative by immunohistochemistry. PE was reproduced in foals using intestinal mucosal homogenate. Physical, gross and histopathologic alterations typical of PE were detected in some foals. The presence of L. intracellularis was confirmed by immunohistochemistry and/or PCR. The role of L. intracellularis from different species in the etiopathogenesis of PE in the horse was established. [ABSTRACT FROM AUTHOR]
