We present a newly developed instrument for 22Na-based positron-annihilation lifetime spectroscopy, designed to facilitate the simultaneous control of temperature, gas atmosphere, and humidity in a single experimental system. The spectrometer operates within a temperature range of 50–480 K and pressures from 10−6 mbar to 1.5 bars. It features a novel gas dosing chamber that allows in situ adsorption studies with gases such as but not limited to CO2, N2, Ar, O2, and their mixtures, with precise control over mixing ratios. Additionally, the device supports in situ humidity exposure, allowing for comprehensive studies of sample interactions with both humidity and humid gases. Fully automated, the system provides seamless data acquisition and environmental control, including pressure and temperature regulation. We demonstrate the instrument’s capability to elucidate alterations in the free volume of maltodextrin under humidity exposure. Additionally, we illustrate the instrument’s efficacy through case studies on CPO-27 metal-organic frameworks (MOFs), highlighting its versatility in analyzing adsorption phenomena across diverse gas adsorbates and temperatures. This state-of-the-art spectrometer stands as an indispensable tool for probing the physicochemical attributes of materials under varying conditions, providing pivotal insights into gas adsorption mechanisms and material dynamics.

In this, the tenth annual update for the F1000Research Tics collection, we summarize research reports from 2023 on Gilles de la Tourette syndrome and other tic disorders. The authors welcome article suggestions and thoughtful feedback from readers.

Abstract Single-molecule FRET (smFRET) has become a versatile tool for probing the structure and functional dynamics of biomolecular systems, and is extensively used to address questions ranging from biomolecular folding to drug discovery. Confocal smFRET measurements are amongst the widely used smFRET assays and are typically performed in a single-well format. Thus, sampling of many experimental parameters is laborious and time consuming. To address this challenge, we extend here the capabilities of confocal smFRET beyond single-well measurements by integrating a multiwell plate functionality to allow for continuous and automated smFRET measurements. We demonstrate the broad applicability of the multiwell plate assay towards DNA hairpin dynamics, protein folding, competitive and cooperative protein–DNA interactions, and drug-discovery, revealing insights that would be very difficult to achieve with conventional single-well format measurements. For the adaptation into existing instrumentations, we provide a detailed guide and open-source acquisition and analysis software.

Abstract Many charged organic molecules behave as persistent and hazardous pollutants with harmful effects on human health and ecosystems. They are widely distributed related to their charged molecular structure that provides water solubility. In order to track the fate and behavior of such pollutants, charged dyes with specific absorption in the visible spectra serve as convenient model compounds. We provide a platform of smart adsorbers that efficiently remediate positively and negatively charged dyes (crystal violet and Amaranth) from water. Metal oxide nanoparticles serve as a core with an intrinsically large surface area. The surface potential was tuned towards positive or negative by decorating the cores with self‐assembled monolayers of dedicated long‐chained phosphonic acid derivatives. Selective remediation of the dyes was obtained with corresponding oppositely charged core‐shell nanoparticles. Mixed dye solution can be cleaned by a cascade approach or by applying both particle systems simultaneously. The removal efficiency was determined as a function of particle concentration via UV‐spectroscopy. The results of remediation experiments at different pH values and using superparamagnetic iron oxide nanoparticle cores lead to a simple process with recycling capability.

Abstract The actin cortex is an active polymer network underneath the plasma membrane at the periphery of mammalian cells. It is a major regulator of cell shape through the generation of active cortical tension. In addition, the cortex constitutes a mechanical shield that protects the cell during mechanical agitation. Cortical mechanics is tightly controlled by the presence of actin cross‐linking proteins that dynamically bind and unbind actin filaments. Cross‐linker actin bonds are weak non‐covalent bonds whose bond lifetime is likely affected by mechanical tension in the actin cortex making cortical composition inherently mechanosensitive. Here, a quantitative study of changes in cortex composition and turnover dynamics upon short‐lived peaks in active and passive mechanical tension in mitotic HeLa cells is presented. These findings dsclose a twofold mechanical reinforcement strategy of the cortex upon tension peaks entailing i) a direct catch‐bond mechanosensitivity of cross‐linkers filamin and α‐actinin and ii) an indirect cortical mechanosensitivity that triggers actin cortex reinforcement via enhanced polymerization of actin. Thereby a “molecular safety belt” mechanism that protects the cortex from injury upon mechanical challenges is disclosed.

Streamflow regime management is critical for preserving species in riverine and riparian habitats. Quantifying flow-ecology relationships can help to characterize the effect of flow regime change on species; however, dealing with this in ungauged catchments is challenging. By regionalizing simulation of streamflow to ungauged catchments, the Indicators of Hydrological Alterations (IHA) are calculated and used as a proxy for quantifying ecological impacts. High-resolution Regional Climate Models are used to derive the future streamflow in both gauged and ungauged catchments of the Ethiopian Rift Valley Lakes Basin (RVLB). Consequently, 32 IHAs representing the magnitude, duration, frequency, rate, and timing of streamflow events during their historical and future periods are calculated. The number of cases for future IHAs deviating from their historical range is calculated, which is called the non-attainment rate of future IHAs. A threshold value for the non-attainment rate is set to classify the impact as no-impact, low, medium, or high. The non-attainment rate for the future period indicates that, high ecological impact resulted in October, which is the local high flow month. Spatially, a shift towards a later date of the Julian date indicating the maximum flow resulted in a high ecological impact. This approach can be adapted to other regions with reliable regionalization results and suitable future climate models, which would be useful for the quantification and management of ecological impacts in ungauged regions.

Data scarcity in many areas around the world represents a major problem for hydrological model calibrations. Global parameter estimates and global forcing can provide possibilities to access hydrological responses in ungauged regions. In this study, we applied HBV global parameter estimates considering uncertainty in the Upper Neckar and Upper Danube catchments, Germany, to answer what are the influencing factors and how good are their local applications. We tested simulations with precipitation in spatial resolutions from 0.05 to 0.2° and with local/global sources. Results show that the general performance is acceptable to good (Kling-Gupta efficiency, KGE: 0.51–0.79) in both catchments using local or global precipitation. The influence of spatial resolutions is insignificant while using local precipitation slightly increases performance in both catchments. Catchment properties such as complex topography and special karst subsurface may lead to a deterioration of performance by 0.2 of median KGE in the Upper Danube compared with the Upper Neckar catchment. The median correlation coefficient, runoff ratio and relative error suggest that using global parameter estimates can reproduce seasonality and long-term water balance in our studied region. Our study highlights the potential of using global parameter estimates and global forcing in ungauged areas. HIGHLIGHTS Global parameter estimates with global forcing can produce acceptable local discharge simulations in our study area.; Catchment properties have more impacts on local applications than sources and spatial resolutions of forcing when applying global parameter estimates.; Our study highlights the potential of using global forcing to understand hydrological behavior and water balance at a monthly scale in ungauged areas.;

Abstract Tourette Syndrome (TS) is a complex neurodevelopmental disorder characterized by vocal and motor tics lasting more than a year. It is highly polygenic in nature with both rare and common previously associated variants. Epidemiological studies have shown TS to be correlated with other phenotypes, but large-scale phenome wide analyses in biobank level data have not been performed to date. In this study, we used the summary statistics from the latest meta-analysis of TS to calculate the polygenic risk score (PRS) of individuals in the UK Biobank data and applied a Phenome Wide Association Study (PheWAS) approach to determine the association of disease risk with a wide range of phenotypes. A total of 57 traits were found to be significantly associated with TS polygenic risk, including multiple psychosocial factors and mental health conditions such as anxiety disorder and depression. Additional associations were observed with complex non-psychiatric disorders such as Type 2 diabetes, heart palpitations, and respiratory conditions. Cross-disorder comparisons of phenotypic associations with genetic risk for other childhood-onset disorders (e.g.: attention deficit hyperactivity disorder [ADHD], autism spectrum disorder [ASD], and obsessive-compulsive disorder [OCD]) indicated an overlap in associations between TS and these disorders. ADHD and ASD had a similar direction of effect with TS while OCD had an opposite direction of effect for all traits except mental health factors. Sex-specific PheWAS analysis identified differences in the associations with TS genetic risk between males and females. Type 2 diabetes and heart palpitations were significantly associated with TS risk in males but not in females, whereas diseases of the respiratory system were associated with TS risk in females but not in males. This analysis provides further evidence of shared genetic and phenotypic architecture of different complex disorders.

Understanding rainwater dispersion in a spatiotemporal context is invaluable toward resourceful water management and a food-secure society. This study, therefore, assessed the variations in rainfall at a spatiotemporal scale in the Oti River Basin of West Africa for observed (1981–2010) and future periods (2021–2050) under the representative concentration pathways (RCPs) 4.5 and 8.5 emission scenarios. Rainfall data from meteorological stations and Climate Hazards Group Infrared Precipitation with Stations (CHIRPS) were used. The percentage changes in rainfall for the peak month as well as for rainy and dry seasons under the two climate scenarios were determined. The coefficient of variation (CV) and the standardized anomaly index (SAI) were used to assess annual variations in rainfall. In general, under both emission scenarios, rainfall is projected to decrease over the study area. However, the amount of rainfall during the peak month (August) for RCP4.5 and RCP8.5 could increase by 0.26 and 9.3%, respectively. The highest SAIs for the observed period were +1.58 (2009) and −2.29 (1983) with the latter showing a relationship with historic drought in the basin. The projected SAI under RCP4.5 and RCP8.5 indicated extremely wet (+2.12) and very wet (+1.91) periods for the years 2037 and 2028, respectively. The study provides relevant information and a chance to aid the design of innovative adaptation measures toward efficient water management and agricultural planning for the basin. Abstract Background For translational genomics, a roadmap is needed to know the molecular similarities or differences between species, such as model species and crop species. This knowledge is invaluable for the selection of target genes and pathways to alter downstream in response to the same stimuli. Here, the transcriptomic responses to six treatments including hormones (abscisic acid - ABA and salicylic acid - SA); treatments that cause oxidative stress (3-amino-1,2,4-triazole - 3AT, methyl viologen - MV); inhibit respiration (antimycin A - AA) or induce genetic damage (ultraviolet radiation -UV) were analysed and compared between Arabidopsis (Arabidopsis thaliana), barley (Hordeum vulgare) and rice (Oryza sativa). Results Common and opposite responses were identified between species, with the number of differentially expressed genes (DEGs) varying greatly between treatments and species. At least 70% of DEGs overlapped with at least one other treatment within a species, indicating overlapping response networks. Remarkably, 15 to 34% of orthologous DEGs showed opposite responses between species, indicating diversity in responses, despite orthology. Orthologous DEGs with common responses to multiple treatments across the three species were correlated with experimental data showing the functional importance of these genes in biotic/abiotic stress responses. The mitochondrial dysfunction response was revealed to be highly conserved in all three species in terms of responsive genes and regulation via the mitochondrial dysfunction element. Conclusions The orthologous DEGs that showed a common response between species indicate conserved transcriptomic responses of these pathways between species. However, many genes, including prominent salt-stress responsive genes, were oppositely responsive in multiple-stresses, highlighting fundamental differences in the responses and regulation of these genes between species. This work provides a resource for translation of knowledge or functions between species.

Quantifying the spatial variability of groundwater recharge is crucial for water resource management under changing land use conditions. Hydrological model performance is highly influenced particularly for peak discharge simulation for the wetland-dominated watersheds. This study aimed at improving the performance of a numerical model at a wetland-dominated watershed by explicitly accounting for wetland processes. The Soil and Water Assessment Tool (SWAT) was used to simulate the spatial variability of groundwater recharge for the two Land Use and Land Cover (LULC) maps classified in 1996 and 2017. An acceptable model performance during the calibration and validation periods is found by incorporating wetland parameters. The trend of LULC change showed a significant reduction for Wetland and Forest going along with an increase of agricultural land at the rate of 9.56 ​km2/yr. Accounting for the LULC changes in the model, the simulated median groundwater recharge to the aquifer has reduced from 506.25 ​mm to 483.34 ​mm per year. During these periods, the extent of Wetland extremely converted towards Grassland, resulting in a reduction of median groundwater recharge from 508.6 ​mm to 495.3 ​mm per year in wetland-dominated sub-watersheds. In addition, this study demonstrates spatial variability in groundwater recharge with differences in catchment properties such as elevation and slope. Therefore, this study provides spatially quantified values for groundwater recharge under the LULC conditions, which are useful in the management and development of groundwater recharge regions.

Abstract Reward sensitivity has been suggested as one of the central pathophysiological mechanisms in Tourette disorder. However, the subjective valuation of a reward by introduction of delay has received little attention in Tourette disorder, even though it has been suggested as a trans-diagnostic feature of numerous neuropsychiatric disorders. We aimed to assess delay discounting in Tourette disorder and to identify its brain functional correlates. We evaluated delayed discounting and its brain functional correlates in a large group of 54 Tourette disorder patients and 31 healthy controls using a data-driven approach. We identified a subgroup of 29 patients with steeper reward discounting, characterised by a higher burden of impulse-control disorders and a higher level of general impulsivity compared to patients with normal behavioural performance or to controls. Reward discounting was underpinned by resting-state activity of a network comprising the orbito-frontal, cingulate, pre-supplementary motor area, temporal and insular cortices, as well as ventral striatum and hippocampus. Within this network, (i) lower connectivity of pre-supplementary motor area with ventral striatum predicted a higher impulsivity and a steeper reward discounting and (ii) a greater connectivity of pre-supplementary motor area with anterior insular cortex predicted steeper reward discounting and more severe tics. Overall, our results highlight the heterogeneity of the delayed reward processing in Tourette disorder, with steeper reward discounting being a marker of burden in impulsivity and impulse control disorders, and the pre-supplementary motor area being a hub region for the delay discounting, impulsivity and tic severity.

Tourette syndrome (TS) is characterized by multiple motor and vocal tics, and high-comorbidity rates with other neuropsychiatric disorders. Obsessive compulsive disorder (OCD), attention deficit hyperactivity disorder (ADHD), autism spectrum disorders (ASDs), major depressive disorder (MDD), and anxiety disorders (AXDs) are among the most prevalent TS comorbidities. To date, studies on TS brain structure and function have been limited in size with efforts mostly fragmented. This leads to low-statistical power, discordant results due to differences in approaches, and hinders the ability to stratify patients according to clinical parameters and investigate comorbidity patterns. Here, we present the scientific premise, perspectives, and key goals that have motivated the establishment of the Enhancing Neuroimaging Genetics through Meta-Analysis for TS (ENIGMA-TS) working group. The ENIGMA-TS working group is an international collaborative effort bringing together a large network of investigators who aim to understand brain structure and function in TS and dissect the underlying neurobiology that leads to observed comorbidity patterns and clinical heterogeneity. Previously collected TS neuroimaging data will be analyzed jointly and integrated with TS genomic data, as well as equivalently large and already existing studies of highly comorbid OCD, ADHD, ASD, MDD, and AXD. Our work highlights the power of collaborative efforts and transdiagnostic approaches, and points to the existence of different TS subtypes. ENIGMA-TS will offer large-scale, high-powered studies that will lead to important insights toward understanding brain structure and function and genetic effects in TS and related disorders, and the identification of biomarkers that could help inform improved clinical practice.

Abstract Tourette syndrome (TS) is a neuropsychiatric disorder of complex genetic architecture involving multiple interacting genes. Here, we sought to elucidate the pathways that underlie the neurobiology of the disorder through genome-wide analysis. We analyzed genome-wide genotypic data of 3581 individuals with TS and 7682 ancestry-matched controls and investigated associations of TS with sets of genes that are expressed in particular cell types and operate in specific neuronal and glial functions. We employed a self-contained, set-based association method (SBA) as well as a competitive gene set method (MAGMA) using individual-level genotype data to perform a comprehensive investigation of the biological background of TS. Our SBA analysis identified three significant gene sets after Bonferroni correction, implicating ligand-gated ion channel signaling, lymphocytic, and cell adhesion and transsynaptic signaling processes. MAGMA analysis further supported the involvement of the cell adhesion and trans-synaptic signaling gene set. The lymphocytic gene set was driven by variants in FLT3, raising an intriguing hypothesis for the involvement of a neuroinflammatory element in TS pathogenesis. The indications of involvement of ligand-gated ion channel signaling reinforce the role of GABA in TS, while the association of cell adhesion and trans-synaptic signaling gene set provides additional support for the role of adhesion molecules in neuropsychiatric disorders. This study reinforces previous findings but also provides new insights into the neurobiology of TS.

IntroductionThe aim of this study was to create a new version of the French GTS-QOL adapted to adolescents with GTS aged 12-16 years (GTS-QOL-French-Ado) and to evaluate its psychometric properties.MethodsWe assessed the psychometric properties of the GTS-QOL-French-Ado in 84 adolescents (mean age 13.6 years, standard deviation 1.2) in terms of factor structure, internal consistency, reliability and convergent validity with the Child Depression Inventory (CDI), the Multidimensional Anxiety Scale for Children (MASC), the Motor tic, Obsessions and compulsions, Vocal tic Evaluation Survey (MOVES) and the French "Vécu et Santé Perçue de l'Adolescent" (VSP-A), a generic self-administered measure of health-related quality of life (HRQoL) in adolescents.ResultsExploratory factor analysis of the GTS-QOL-French-Ado resulted in a 5-factor solution. The GTS-QOL-French-Ado demonstrated good acceptability with missing values per subscale ranging from 0% to 1.2%, good internal consistency for four of the five subscales with Cronbach's alpha ranging from 0.56 to 0.87 and good test-retest reliability with intraclass correlation coefficients ranging from 0.74 (95% CI: 0.52-0.86) to 0.82 (95% CI: 0.66-0.91). Convergent validity was supported by correlations with CDI, MASC, MOVES, VSP-A and clinical variables.DiscussionThe GTS-QOL-French-Ado is the first disease-specific HRQoL tool for French-speaking adolescents with GTS aged 12-16 years, and shows good psychometric properties. Further psychometric testing on responsiveness to change would be of great interest.

Currently, no treatments available for Parkinson’s disease (PD) can slow PD progression. At the early stage of the disease, only a subset of individuals with PD progress quickly, while the majority have a slowly progressive form of the disease. In developing treatments that aim to slow PD progression, clinical trials aim to include individuals who are likely to progress faster, such that a treatment effect, if one exists, can be identified easier and earlier. The aim of the present study was to identify baseline predictors of clinical progression in early PD. We analyzed 12-month data acquired from the PASADENA trial Part 1 (NCT03100149, n = 76 participants who were allocated to the placebo arm and did not start symptomatic therapy) and the Parkinson’s Progression Markers Initiative (PPMI) study (n = 139 demographically and clinically matched participants). By using ridge regression models including clinical characteristics, imaging, and non-imaging biomarkers, we found that Hoehn and Yahr stage and dopamine transporter single-photon emission computed tomography specific binding ratios (Dat-SPECT SBR) in putamen ipsilateral to the side of motor symptom onset predicted PD progression at the early stage of the disease. Further studies are needed to confirm the validity of these predictors to identify with high accuracy individuals with early PD with a faster progression phenotype.

Based on its prevalence, Tourette syndrome cannot be considered a rare condition. However, in this opinion article, we make the claim that it should nonetheless be considered as an orphan or neglected disease.

Branaplam is a therapeutic agent currently in clinical development for the treatment of infants with type 1 spinal muscular atrophy (SMA). Since preclinical studies showed that branaplam had cell-cycle arrest effects, we sought to determine whether branaplam may affect postnatal cerebellar development and brain neurogenesis. Here, we describe a novel approach for developmental neurotoxicity testing (DNT) of a central nervous system (CNS) active drug. The effects of orally administered branaplam were evaluated in the SMA neonatal mouse model (SMNΔ7), and in juvenile Wistar Hannover rats and Beagle dogs. Histopathological examination and complementary immunohistochemical studies focused on areas of neurogenesis in the cerebellum (mice, rats, and dogs), and the subventricular zone of the striatum and dentate gyrus (rats and dogs) using antibodies directed against Ki67, phosphorylated histone H3, cleaved caspase-3, and glial fibrillary acidic protein. Additionally, image-analysis based quantification of calbindin-D28k and Ki67 was performed in rats and dogs. The patterns of cell proliferation and apoptosis, and neural migration and innervation in the cerebellum and other brain regions of active adult neurogenesis did not differ between branaplam- and control-treated animals. Quantitative image analysis did not reveal any changes in calbindin-D28k and Ki67 expression in rats and dogs. The data show that orally administered branaplam has no impact on neurogenesis in juvenile animals. Application of selected immunohistochemical stainings in combination with quantitative image analysis on a few critical areas of postnatal CNS development offer a reliable approach to assess DNT of CNS-active drug candidates in juvenile animal toxicity studies.

Background: Currently available treatments for Parkinson's disease (PD) do not slow clinical progression nor target alpha-synuclein, a key protein associated with the disease.Objective: The study objective was to evaluate the efficacy and safety of prasinezumab, a humanized monoclonal antibody that binds aggregated alpha-synuclein, in individuals with early PD.Methods: The PASADENA study is a multicenter, randomized, double-blind, placebo-controlled treatment study. Individuals with early PD, recruited across the US and Europe, received monthly intravenous doses of prasinezumab (1,500 or 4,500 mg) or placebo for a 52-week period (Part 1), followed by a 52-week extension (Part 2) in which all participants received active treatment. Key inclusion criteria were: aged 40–80 years; Hoehn & Yahr (H&Y) Stage I or II; time from diagnosis ≤2 years; having bradykinesia plus one other cardinal sign of PD (e.g., resting tremor, rigidity); DAT-SPECT imaging consistent with PD; and either treatment naïve or on a stable monoamine oxidase B (MAO-B) inhibitor dose. Study design assumptions for sample size and study duration were built using a patient cohort from the Parkinson's Progression Marker Initiative (PPMI). In this report, baseline characteristics are compared between the treatment-naïve and MAO-B inhibitor-treated PASADENA cohorts and between the PASADENA and PPMI populations.Results: Of the 443 patients screened, 316 were enrolled into the PASADENA study between June 2017 and November 2018, with an average age of 59.9 years and 67.4% being male. Mean time from diagnosis at baseline was 10.11 months, with 75.3% in H&Y Stage II. Baseline motor and non-motor symptoms (assessed using Movement Disorder Society—Unified Parkinson's Disease Rating Scale [MDS-UPDRS]) were similar in severity between the MAO-B inhibitor-treated and treatment-naïve PASADENA cohorts (MDS-UPDRS sum of Parts I + II + III [standard deviation (SD)]; 30.21 [11.96], 32.10 [13.20], respectively). The overall PASADENA population (63.6% treatment naïve and 36.4% on MAO-B inhibitor) showed a similar severity in MDS-UPDRS scores (e.g., MDS-UPDRS sum of Parts I + II + III [SD]; 31.41 [12.78], 32.63 [13.04], respectively) to the PPMI cohort (all treatment naïve).Conclusions: The PASADENA study population is suitable to investigate the potential of prasinezumab to slow disease progression in individuals with early PD.Trial Registration: NCT03100149.

Background: Genes involved in Tourette syndrome (TS) remain largely unknown. We aimed to identify genetic factors contributing to TS in a French cohort of 120 individuals using a combination of hypothesis-driven and exome-sequencing approaches. Methods: We first sequenced exons of SLITRK1-6 and HDC in the TS cohort and subsequently sequenced the exome of 12 individuals harboring rare variants in these genes to find additional rare variants contributing to the disorder under the hypothesis of oligogenic inheritance. We further screened three candidate genes (OPRK1, PCDH10, and NTSR2) preferentially expressed in the basal ganglia, and three additional genes involved in neurotensin and opioid signaling (OPRM1, NTS, and NTSR1), and compared variant frequencies in TS patients and 788 matched control individuals. We also investigated the impact of altering the expression of Oprk1 in zebrafish. Results: Thirteen ultrarare missense variants of SLITRK1-6 and HDC were identified in 12 patients. Exome sequencing in these patients revealed rare possibly deleterious variants in 3,041 genes, 54 of which were preferentially expressed in the basal ganglia. Comparison of variant frequencies altering selected candidate genes in TS and control individuals revealed an excess of potentially disrupting variants in OPRK1, encoding the opioid kappa receptor, in TS patients. Accordingly, we show that downregulation of the Oprk1 orthologue in zebrafish induces a hyperkinetic phenotype in early development. Discussion: These results support a heterogeneous and complex genetic etiology of TS, possibly involving rare variants altering the opioid pathway in some individuals, which could represent a novel therapeutic target in this disorder.

δ18O of speleothems are a widely used paleoclimate proxy. Here, the authors conduct a global analysis of cave drip water δ18O compositions and find that drip waters from warmer climates have a seasonal bias toward precipitation δ18O of recharge periods, unlike in cooler climates where drip waters match well with recharge-weighted δ18O.

Quantifying uncertainties in water resource prediction in data-scarce regions is essential for resource development. We use globally available datasets of precipitation and potential evapotranspiration for the regionalization of model parameters in the data-scarce regions of Ethiopia. A regional model was developed based on 14 gauged catchments. Three possible parameter sets were tested for regionalization: (1) the best calibration parameters, (2) the best validation parameter set derived from behavioral parameters during the validation period, and (3) the stable parameter sets. Weighted multiple linear regression was applied by assigning more weight to identifiable parameters, using a novel leave-one-out cross-validation technique for evaluation and uncertainty quantification. The regionalized parameter sets were applied to the remaining 35 ungauged catchments in the Ethiopian Rift Valley Lake Basin (RVLB) to provide regional water balance estimations. The monthly calibration of the gauged catchments resulted in Nash Sutcliffe Efficiencies (NSE) ranging from 0.53 to 0.86. The regionalization approach provides acceptable regional model performances with a median NSE of 0.63. The results showed that, other than the commonly used best-calibrated parameters, the stable parameter sets provide the most robust estimates of regionalized parameters. As this approach is model-independent and the input data used are available globally, it can be applied to any other data-scarce region.

The novel coronavirus disease (COVID-19) was identified as the cause of an outbreak of respiratory disease in China at the end of 2019. It then spread with enormous rapidity and by mid-March 2020 was declared a world pandemic. Gilles de la Tourette Syndrome (GTS) is a childhood-onset neurodevelopmental disorder with a worldwide prevalence of about 1% of the population. The clinical symptoms include multiple motor and one or more phonic (vocal) tics. Germane to this communication is that 85% of patients with GTS have associated psychiatric co-morbidities, many of which are being exacerbated in the current global health crisis. In addition, several symptoms of GTS may mimic COVID-19, such as a dry cough and sniffing (phonic tics), while other symptoms such as spitting, inappropriate touching of others and “non-obscene socially inappropriate symptoms” can potentially get patients with GTS into trouble with the law. We suggest that a clear explanation of the COVID-19 illness and GTS is important to enable colleagues of various specialities who tend to patients with GTS. It is important to acknowledge at the outset that the information available on the COVID-19 pandemic changes daily, including cases infected, deaths reported, and how various national health systems are planning and or coping or not. It is fair to say that having read the current medical and lay press we conclude that it is not easy to reassure our patients with absolute certainty. However, notwithstanding that, we hope our documentation is of some assistance.

BackgroundType 2 diabetes is a major risk factor for cardiovascular diseases, e.g. coronary artery disease (CAD). But it has also been shown that diabetes can cause heart failure independently of ischemic heart disease (IHD) by causing diabetic cardiomyopathy. In contrast to diabetes and IHD, limited data exist regarding patients with diabetes and dilated cardiomyopathy (DCM).MethodsEVIdence based TreAtment in Heart Failure (EVITA-HF) comprises web-based case report data on demography, diagnostic measures, adverse events and 1-year follow-up of patients hospitalized for chronic heart failure and an ejection fraction ≤40%. In the present study we focused on the results of patients with diabetes and heart failure.ResultsBetween February 2009 and November 2015, 4101 patients with chronic heart failure were included in 16 tertiary care centers in Germany. The mortality in patients with diabetes and DCM (n = 323) was more than double (15.2%) than that of DCM patients without diabetes (6.5%, pConclusionThe influence of diabetes on the mortality rate is only significant in patients with DCM not in patients with CAD. Therefore, the underlying mechanisms of this effect should be studied in greater detail to improve patient care and outcome.

IntroductionThe Gilles de la Tourette Syndrome-Quality of Life Scale (GTS-QOL) is a self-rated disease-specific questionnaire to assess health-related quality of life of subjects with GTS. Our aim was to perform the cross-cultural adaptation of the GTS-QOL into French and to assess its psychometric properties.MethodsThe GTS-QOL was cross-culturally adapted by conducting forward and backward translations, following international guidelines. The psychometric properties of the GTS-QOL-French were assessed in 109 participants aged 16 years and above with regard to factor structure, internal consistency, reliability and convergent validity with the MOVES (Motor tic, Obsessions and compulsions, Vocal tic Evaluation Survey) and the WHOQOL-BREF (World Health Organization Quality of Life Brief).ResultsExploratory factor analysis of the GTS-QOL-French resulted in a 6-factor solution and did not replicate the original structure in four subscales. The results showed good acceptability (missing values per subscale ranging from 0% to 0.9%), good internal consistency (Cronbach's alpha ranging from 0.68 to 0.94) and good test-retest reliability (intraclass correlation coefficients ranging from 0.70 to 0.81). Convergent validity with the MOVES and WHOQOL-BREF scales showed high correlations.DiscussionOur study provides evidence of the good psychometric properties of the GTS-QOL-French. The cross-cultural adaptation and validation of this specific instrument will make it possible to assess health-related quality of life in French-speaking subjects with GTS. The GTS-QOL-French could be recommended for use in future research.

Abstract The beta‐site amyloid precursor protein cleaving enzyme‐1 (BACE‐1) initiates the generation of amyloid‐β (Aβ), and the amyloid cascade leading to amyloid plaque deposition, neurodegeneration, and dementia in Alzheimer's disease (AD). Clinical failures of anti‐Aβ therapies in dementia stages suggest that treatment has to start in the early, asymptomatic disease states. The BACE‐1 inhibitor CNP520 has a selectivity, pharmacodynamics, and distribution profile suitable for AD prevention studies. CNP520 reduced brain and cerebrospinal fluid (CSF) Aβ in rats and dogs, and Aβ plaque deposition in APP‐transgenic mice. Animal toxicology studies of CNP520 demonstrated sufficient safety margins, with no signs of hair depigmentation, retina degeneration, liver toxicity, or cardiovascular effects. In healthy adults ≥ 60 years old, treatment with CNP520 was safe and well tolerated and resulted in robust and dose‐dependent Aβ reduction in the cerebrospinal fluid. Thus, long‐term, pivotal studies with CNP520 have been initiated in the Generation Program.

Abstract Gilles de la Tourette syndrome (TS) is a complex developmental neuropsychiatric condition in which motor manifestations are often accompanied by comorbid conditions that impact the patient’s quality of life. In the DSM-5, TS belongs to the “neurodevelopmental disorders” group, together with other neurodevelopmental conditions, frequently co-occurring. In this study, we searched the PubMed database using a combination of keywords associating TS and all neurodevelopmental diagnoses. From 1009 original reports, we identified 36 studies addressing TS and neurodevelopmental comorbidities. The available evidence suggests the following: (1) neurodevelopmental comorbidities in TS are the rule, rather than the exception; (2) attention deficit/hyperactivity disorder (ADHD) is the most frequent; (3) there is a continuum from a simple (TS + ADHD or/and learning disorder) to a more complex phenotype (TS + autism spectrum disorder). We conclude that a prompt diagnosis and a detailed description of TS comorbidities are necessary not only to understand the aetiological basis of neurodevelopmental disorders but also to address specific rehabilitative and therapeutic approaches.

Abstract The dopamine partial agonist aripiprazole is increasingly used to treat pathologies for which other antipsychotics are indicated because it displays fewer side effects, such as sedation and depression-like symptoms, than other dopamine receptor antagonists. Previously, we showed that aripiprazole may protect motivational function by preserving reinforcement-related signals used to sustain reward-maximization. However, the effect of aripiprazole on more cognitive facets of human reinforcement learning, such as learning from the forgone outcomes of alternative courses of action (i.e., counterfactual learning), is unknown. To test the influence of aripiprazole on counterfactual learning, we administered a reinforcement learning task that involves both direct learning from obtained outcomes and indirect learning from forgone outcomes to two groups of Gilles de la Tourette (GTS) patients, one consisting of patients who were completely unmedicated and the other consisting of patients who were receiving aripiprazole monotherapy, and to healthy subjects. We found that whereas learning performance improved in the presence of counterfactual feedback in both healthy controls and unmedicated GTS patients, this was not the case in aripiprazole-medicated GTS patients. Our results suggest that whereas aripiprazole preserves direct learning of action-outcome associations, it may impair more complex inferential processes, such as counterfactual learning from forgone outcomes, in GTS patients treated with this medication.

The study of motor timing informs on how temporal information integrates with motor acts. Cortico-basal ganglia and cortico-cerebellar circuits control this integration, whereas transcallosal interhemispheric connectivity modulates finely timed lateralized or bimanual actions. Motor timing abilities are under-explored in Tourette syndrome (TS). We adopted a synchronization-continuation task to investigate motor timing in sequential movements in TS patients. We studied 14 adult TS patients and 19 age-matched healthy volunteers. They were asked to tap in synchrony with a metronome cue (SYNC) and then, when the tone stopped, to keep tapping, maintaining the same rhythm (CONT). We tested both a sub-second and a supra-second inter-stimulus interval between the cues. Subjects randomly performed a single-hand task with the right hand and a bimanual task using both hands simultaneously wearing sensor-engineered gloves. We measured the temporal error and the interval reproduction accuracy index. We also performed MRI-based diffusion tensor imaging and probabilistic tractography of inter-hemispheric corpus callosum (CC) connections between supplementary motor areas (SMA) and the left SMA-putamen fiber tract. TS patients were less accurate than healthy individuals only on the single-hand version of the CONT task when asked to reproduce supra-second time interval. Supra-second time processing improved in TS patients in the bimanual task, with the performance of the right hand on the bimanual version of the CONT task being more accurate than that of the right hand on the single-hand version of the task. We detected a significantly higher fractional anisotropy (FA) in both SMA-SMA callosal and left-sided SMA-putamen fiber tracts in TS patients. In TS patients only, the structural organization of transcallosal connections between the SMAs and of the left SMA-putamen tract was higher when the motor timing accuracy of the right hand on the bimanual version of the task was lower. Abnormal timing performance for supra-second time processing is suggestive of a defective network inter-connecting the striatum, the dorsolateral prefrontal cortex and the SMA. An increase in accuracy on the bimanual version of the CONT task may be the result of compensatory processes linked to self-regulation of motor control, as witnessed by plastic rearrangement of inter-hemispheric and cortical-subcortical fiber tracts.

We introduce a two-parameter approximate counter-diabatic term into the Hamiltonian of the transverse-field Ising model for quantum annealing to accelerate convergence to the solution, generalizing an existing single-parameter approach. The protocol is equivalent to unconventional diabatic control of the longitudinal and transverse fields in the transverse-field Ising model and thus makes it more feasible for experimental realization than an introduction of new terms such as nonstoquastic catalysts toward the same goal of performance enhancement. We test the idea for the p-spin model with p=3, which has a first-order quantum phase transition, and show that our two-parameter approach leads to significantly larger ground-state fidelity and lower residual energy than those by traditional quantum annealing and by the single-parameter method. We also find a scaling advantage in terms of the time-to-solution as a function of the system size in a certain range of parameters as compared to the traditional methods in the sense that an exponential time complexity is reduced to another exponential complexity with a smaller coefficient. Although the present method may not always lead to a drastic exponential speedup in difficult optimization problems, it is useful because of its versatility and applicability for any problem after a simple algebraic manipulation, in contrast to some other powerful prescriptions for acceleration such as nonstoquastic catalysts in which one should carefully study in advance if it works in a given problem and should identify a proper way to meticulously control the system parameters to achieve the goal, which is generally highly nontrivial.

Quantum refrigerators pump heat from a cold to a hot reservoir. In the few-particle regime, counter-diabatic (CD) driving of, originally adiabatic, work-exchange strokes is a promising candidate to overcome the bottleneck of vanishing cooling power. Here, we present a finite-time many-body quantum refrigerator that yields finite cooling power at high coefficient of performance, that considerably outperforms its non-adiabatic counterpart. We employ multi-spin CD driving and numerically investigate the scaling behavior of the refrigeration performance with system size. We further prove that optimal refrigeration via the exact CD protocol is a catalytic process.

Seed germination is a critical process for completion of the plant life cycle and for global food production. Comparing the germination transcriptomes of barley (Hordeum vulgare) to Arabidopsis thaliana revealed the overall pattern was conserved in terms of functional gene ontology; however, many oppositely responsive orthologous genes were identified. Conserved processes included a set of approximately 6000 genes that peaked early in germination and were enriched in processes associated with RNA metabolism, e.g., pentatricopeptide repeat (PPR)-containing proteins. Comparison of orthologous genes revealed more than 3000 orthogroups containing almost 4000 genes that displayed similar expression patterns including functions associated with mitochondrial tricarboxylic acid (TCA) cycle, carbohydrate and RNA/DNA metabolism, autophagy, protein modifications, and organellar function. Biochemical and proteomic analyses indicated mitochondrial biogenesis occurred early in germination, but detailed analyses revealed the timing involved in mitochondrial biogenesis may vary between species. More than 1800 orthogroups representing 2000 genes displayed opposite patterns in transcript abundance, representing functions of energy (carbohydrate) metabolism, photosynthesis, protein synthesis and degradation, and gene regulation. Differences in expression of basic-leucine zippers (bZIPs) and Apetala 2 (AP2)/ethylene-responsive element binding proteins (EREBPs) point to differences in regulatory processes at a high level, which provide opportunities to modify processes in order to enhance grain quality, germination, and storage as needed for different uses.

Excess nitrogen (N) deposition and gaseous N emissions from industrial, domestic, and agricultural sources have led to increased nitrate leaching, the loss of biological diversity, and has affected carbon (C) sequestration in forest ecosystems. Nitrate leaching affects the purity of karst water resources, which contribute around 50% to Austria’s drinking water supply. Here we present an evaluation of the drivers of dissolved inorganic N (DIN) concentrations and fluxes from a karst catchment in the Austrian Alps (LTER Zöbelboden) from 27 years of records. In addition, a hydrological model was used together with climatic scenario data to predict expected future runoff dynamics. The study area was exposed to increasing N deposition during the 20th century (up to 30 to 35 kg N ha−1 y−1), which are still at levels of 25.5 ± 3.6 and 19.9 ± 4.2 kg N ha−1 y−1 in the spruce and the mixed deciduous forests, respectively. Albeit N deposition was close to or exceeded critical loads for several decades, 70–83% of the inorganic N retained in the catchment from 2000 to 2018, and NO3- concentrations in the runoff stayed −1 unless high-flow events occurred or forest stand-replacing disturbances. We identified tree growth as the main sink for inorganic N, which might together with lower runoff, increase retention of only weakly decreasing N deposition in the future. However, since recurring forest stand-replacement is predicted in the future as a result of a combination of climatically driven disturbance agents, pulses of elevated nitrate concentrations in the catchment runoff will likely add to groundwater pollution.

Quantum heat engines are modeled by thermodynamic cycles with quantum-mechanical working media. Since high engine efficiencies require adiabaticity, a major challenge is to yield a nonvanishing power output at finite cycle times. Shortcuts to adiabaticity using counter-diabatic (CD) driving may serve as a means to speed up such, otherwise infinitely long, cycles. We introduce local approximate CD protocols for many-body spin quantum heat engines and show that this method improves the efficiency and power for finite cycle times considerably. The protocol does not require a priori knowledge of the system eigenstates and is thus realistic in experiments.

Topographically delineated catchments are the common spatial unit to connect human activities and climate change with their consequences for water availability as a prerequisite for sustainable water management. However, inter-catchment groundwater flow and limited connectivity within the catchment results in effective catchment areas different from those suggested by surface topography. Here, we introduce the notion of effective catchment area quantified through an effective catchment index (ECI), derived from observed streamflow, precipitation and actual evapotranspiration estimates, to understand the prevalence and significance of substantial differences between topographic and effective catchment areas in a global dataset. We evaluate our ECI analysis by comparing it to hydraulic head simulations of a global groundwater flow model and to the Budyko framework. We find that one in three studied catchments exhibit an effective catchment area either larger than double or smaller than half of their topographic area. These catchments will likely be affected by management activities such as groundwater pumping or land use change outside their topographic boundaries. Or alternatively, they affect water resources beyond their topographic boundaries. We find that the magnitude of the observed differences is strongly linked to aridity, mean slope, distance to coast, and topographic area. Our study provides a first-order identification of catchments where additional in-depth analysis of subsurface connectivity is needed to support sustainable water management.

The paper explores the potential of data mining in bibliographic records. Quantitative analsysis of co-occurrences of keywords in bibliographic records makes it possible to create a semantic network. In contrast to the rigid hierarchy of traditional thesauri, this network maps polyvalent semantic relations between keywords across the distinct thematic branches of a thesaurus tree. Analysis of a comprehensive data pool made up from two major bibliographies (Gnomon Bibliographic Database and ZENON DAI) should allow for the semantic mapping of a core set of central topics (people, places, social structures, concepts) of classical scholarship.

BackgroundPneumonia is among the most common acute complications after stroke and is associated with poor long-term outcome. Biomarkers may help identifying stroke patients at high risk for developing stroke-associated pneumonia (SAP) and to guide early treatment.AimsThis trial investigated whether procalcitonin (PCT) ultrasensitive (PCTus)-guided antibiotic treatment of SAP can improve functional outcome after stroke.MethodsIn this international, multicenter, randomized, controlled clinical trial with blinded assessment of outcomes, patients with severe ischemic stroke in the middle cerebral artery territory were randomly assigned within 40 h after symptom onset to PCTus-based antibiotic therapy guidance in addition to stroke unit care or standard stroke unit care alone. The primary endpoint was functional outcome at 3 months, defined according to the modified Rankin Scale (mRS) and dichotomized as acceptable (≤4) or unacceptable (≥5). Secondary endpoints included usage of antibiotics, infection rates, days of fever, and mortality. The trial was registered with http://ClinicalTrials.gov (Identifier NCT01264549).ResultsIn the intention-to-treat-analysis based on 227 patients (112 in PCT and 115 in control group), 197 patients completed the 3-month follow-up. Adherence to PCT guidance was 65%. PCT-guided therapy did not improve functional outcome as measured by mRS (odds ratio 0.79; 95% confidence interval 0.45–1.35, p = 0.47). Pneumonia rate and mortality were similar in both groups. Days with fever tended to be lower (p = 0.055), whereas total number of days treated with antibiotics were higher (p = 0.004) in PCT compared to control group. A post hoc analysis including all PCT values in the intention-to-treat population demonstrated a significant increase on the first day of infection in patients with pneumonia and sepsis compared to patients with urinary tract infections or without infections (p

Changes in molecular conformations are one of the major driving forces of complex biological processes. Many studies based on single-molecule techniques have shed light on conformational dynamics and contributed to a better understanding of living matter. In particular, single-molecule FRET experiments have revealed unprecedented information at various time scales varying from milliseconds to seconds. The choice and the attachment of fluorophores is a pivotal requirement for single-molecule FRET experiments. One particularly well-studied millisecond conformational change is the opening and closing of DNA hairpin structures. In this study, we addressed the influence of base- and terminal-labeled fluorophores as well as the fluorophore DNA interactions on the extracted kinetic information of the DNA hairpin. Gibbs free energies varied from ∆G0 = −3.6 kJ/mol to ∆G0 = −0.2 kJ/mol for the identical DNA hairpin modifying only the labeling scheme and design of the DNA sample. In general, the base-labeled DNA hairpin is significantly destabilized compared to the terminal-labeled DNA hairpin and fluorophore DNA interactions additionally stabilize the closed state of the DNA hairpin. Careful controls and variations of fluorophore attachment chemistry are essential for a mostly undisturbed measurement of the underlying energy landscape of biomolecules.

We present a coherent counter-diabatic quantum protocol to prepare ground states in the lattice gauge mapping of all-to-all Ising models (LHZ) with considerably enhanced final ground state fidelity compared to a quantum annealing protocol. We make use of a variational method to find approximate counter-diabatic Hamiltonians that has recently been introduced by Sels and Polkovnikov (2017 Proc. Natl. Acad. Sci. 114 3909). The resulting additional terms in our protocol are time-dependent local on-site y-magnetic fields. These additional Hamiltonian terms do not increase the minimal energy gap, but instead compensate for the Berry curvature. A single free parameter is introduced which is optimized via classical updates. The protocol consists only of local and nearest-neighbor terms which makes it attractive for implementation in near term experiments.
