The development of dual GIPR–GLP1R coagonists that bind to either one or the other receptor (sequence-mixed dual agonists) has emerged as an innovative therapeutic strategy for obesity and type 2 diabetes. Combined activation of both receptors may act synergistically providing additive effects on glucose and body weight in comparison of GLP1 analogues alone. Preclinical studies have confirmed that GIPR–GLP1R coagonists improve several hallmarks of metabolic syndrome, such as obesity, hyperglycemia, and dyslipidemia. These metabolic benefits have been translated from mice to nonhuman primates and humans. Recent clinical trials have shown that coagonists induce significant benefits on body weight, fasting, and postprandial glucose levels, insulin sensitivity, and total cholesterol. Abstract

Higher polygenic risk score for schizophrenia (szPGRS) has been associated with lower cognitive function and might be a predictor of decline in brain structure in apparently healthy populations. Age-related declines in structural brain connectivity—measured using white matter diffusion MRI —are evident from cross-sectional data. Yet, it remains unclear how graph theoretical metrics of the structural connectome change over time, and whether szPGRS is associated with differences in ageing-related changes in human brain connectivity. Here, we studied a large, relatively healthy, same-year-of-birth, older age cohort over a period of 3 years (age ∼ 73 years, N = 731; age ∼76 years, N = 488). From their brain scans we derived tract-averaged fractional anisotropy (FA) and mean diffusivity (MD), and network topology properties. We investigated the cross-sectional and longitudinal associations between these structural brain variables and szPGRS. Higher szPGRS showed significant associations with longitudinal increases in MD in the splenium (β = 0.132, pFDR = 0.040), arcuate (β = 0.291, pFDR = 0.040), anterior thalamic radiations (β = 0.215, pFDR = 0.040) and cingulum (β = 0.165, pFDR = 0.040). Significant declines over time were observed in graph theory metrics for FA-weighted networks, such as mean edge weight (β = −0.039, pFDR = 0.048) and strength (β = −0.027, pFDR = 0.048). No significant associations were found between szPGRS and graph theory metrics. These results are consistent with the hypothesis that szPGRS confers risk for ageing-related degradation of some aspects of structural connectivity., Highlights • Large longitudinal MRI study of brain structure in narrow age range older age cohort. • Longitudinal changes occur in white matter tracts from age 73 to 76. • Graph theory metrics also showed longitudinal reductions over that period. • Microstructural changes in white matter correlate with changes in brain’s network. • Higher genetic risk for schizophrenia is associated with changes in brain structure.

Abstract

We have carried out meta-analyses of genome-wide association studies (GWAS) (n = 23 784) of the first two principal components (PCs) that group together cortical regions with shared variance in their surface area. PC1 (global) captured variations of most regions, whereas PC2 (visual) was specific to the primary and secondary visual cortices. We identified a total of 18 (PC1) and 17 (PC2) independent loci, which were replicated in another 25 746 individuals. The loci of the global PC1 included those associated previously with intracranial volume and/or general cognitive function, such as MAPT and IGF2BP1. The loci of the visual PC2 included DAAM1, a key player in the planar-cell-polarity pathway. We then tested associations with occupational aptitudes and, as predicted, found that the global PC1 was associated with General Learning Ability, and the visual PC2 was associated with the Form Perception aptitude. These results suggest that interindividual variations in global and regional development of the human cerebral cortex (and its molecular architecture) cascade—albeit in a very limited manner—to behaviors as complex as the choice of one’s occupation.

Abstract

Subcortical brain structures are integral to motion, consciousness, emotions and learning. We identified common genetic variation related to the volumes of the nucleus accumbens, amygdala, brainstem, caudate nucleus, globus pallidus, putamen and thalamus, using genome-wide association analyses in almost 40,000 individuals from CHARGE, ENIGMA and UK Biobank. We show that variability in subcortical volumes is heritable, and identify 48 significantly associated loci (40 novel at the time of analysis). Annotation of these loci by utilizing gene expression, methylation and neuropathological data identified 199 genes putatively implicated in neurodevelopment, synaptic signaling, axonal transport, apoptosis, inflammation/infection and susceptibility to neurological disorders. This set of genes is significantly enriched for Drosophila orthologs associated with neurodevelopmental phenotypes, suggesting evolutionarily conserved mechanisms. Our findings uncover novel biology and potential drug targets underlying brain development and disease.

Abstract

OBJECTIVE: To explore genetic and lifestyle risk factors of MRI-defined brain infarcts (BI) in large population-based cohorts. METHODS: We performed meta-analyses of genome-wide association studies (GWAS) and examined associations of vascular risk factors and their genetic risk scores (GRS) with MRI-defined BI and a subset of BI, namely, small subcortical BI (SSBI), in 18 population-based cohorts (n = 20,949) from 5 ethnicities (3,726 with BI, 2,021 with SSBI). Top loci were followed up in 7 population-based cohorts (n = 6,862; 1,483 with BI, 630 with SBBI), and we tested associations with related phenotypes including ischemic stroke and pathologically defined BI. RESULTS: The mean prevalence was 17.7% for BI and 10.5% for SSBI, steeply rising after age 65. Two loci showed genome-wide significant association with BI: FBN2, p = 1.77 x 10(-8); and LINC00539/ZDHHC20, p = 5.82 x 10(-9). Both have been associated with blood pressure (BP)-related phenotypes, but did not replicate in the smaller follow-up sample or show associations with related phenotypes. Age- and sex-adjusted associations with BI and SSBI were observed for BP traits (p value for BI, p [BI] = 9.38 x 10(-25); p [SSBI] = 5.23 x 10(-14) for hypertension), smoking (p [BI] = 4.4 x 10(-10); p [SSBI] = 1.2 x 10(-4)), diabetes (p [BI] = 1.7 x 10(-8); p [SSBI] = 2.8 x 10(-3)), previous cardiovascular disease (p [BI] = 1.0 x 10(-18); p [SSBI] = 2.3 x 10(-7)), stroke (p [BI] = 3.9 x 10(-69); p [SSBI] = 3.2 x 10(-24)), and MRI-defined white matter hyperintensity burden (p [BI] = 1.43 x 10(-157); p [SSBI] = 3.16 x 10(-106)), but not with body mass index or cholesterol. Abstract

Depressive symptoms dramatically increase in the transition from childhood to adolescence (Hankin et al., 1998). Although friendships provide an important source of social support in this period and can protect youth against emotional problems (Bukowski et al., 1996), some characteristics of friendships may actually increase levels of depression rather than protect against such symptoms. Co-rumination, or excessively discussing personal problems within a dyadic relationship (Rose, 2002), has been found disadvantageous for emotional wellbeing, while also promoting friendship quality and closeness among best friends. The current dissertation aims to extend past research on co-rumination in several ways. First, although co-rumination is understood as the interpersonal form of rumination, no study hitherto has explored whether the intrapersonal rumination components 'brooding' and 'reflection' (Treynor et al., 2003), also apply to an interpersonal context. In the first two papers with 2- and 3-month intervals, we distinguished co-brooding from co-reflection and studied relations with outcomes. Interpersonal brooding was found to be maladaptive, predicting higher concurrent and prospective depressive symptoms in girls. Interpersonal reflection predicted less depressive symptoms in girls and higher concurrent friendship quality, and was thereby understood as adaptive. Second, although the relation between co-rumination and depressive symptoms has frequently been established, knowledge on its underlying mechanisms is scarce. The third paper investigated intrapersonal rumination as a mediator in this relation. An indirect effect was established, with co-rumination predicting relative increases in levels of depressive symptoms after two years, through rumination. No evidence was obtained for relations with co-rumination in opposite directions. Third, inconsistent findings on relations between co-rumination and depressive symptoms raise the question of possible interaction effects. The fourth paper investigated co-rumination from a vulnerability-stress perspective and studied whether it acts as a potentiating factor strengthening the associations between stress and depression. Girls high on co-rumination but boys low on co-rumination reported higher levels of depressive symptoms in the context of interpersonal stress across a one-year period. Fourth, although co-rumination has been shown to be maladaptive for emotional well-being, research on the antecedents of co-rumination is lacking. The fifth paper aimed to gain insight into who may be (more) prone to co-ruminate by studying temperament and attachment as two predictors of co-rumination trajectories. Higher positive affectivity, higher trust in the availability of a mother, and higher attachment anxiety were found to predict higher co-rumination in boys and girls. Higher effortful control and lower attachment avoidance related to higher co-rumination among boys. Higher positive affectivity in boys and girls and higher trust in boys predicted decreases in co-rumination across a 3-year interval. Fifth, although depression involves both increased negative affect and decreased positive affect, researchers studying best friend dyads so far solely associated depression with communication about problems. The sixth paper focused on different ways in which best friends talk about happy events and associated positive feelings. Concurrently, co-dampening was found to be maladaptive by predicting higher depressive symptoms and lower friendship quality. Co-enhancing appeared adaptive, showing concurrent associations with lower depressive symptoms and higher friendship quality. Over a 1-year interval, depressive symptoms predicted relative increases in co-dampening. In the final chapter, avenues for future research are discussed, together with clinical implications. In sum, the findings of the current dissertation further clarified how, when, and for whom co-rumination is related to depressive symptoms. In addition, preliminary evidence was obtained for the relevance of studying communication about uplifting events. Insights provided by the present dissertation may lead to more efficient use of prevention and intervention efforts. status: published

Abstract

Humans and animals use adaptive representations of movement dynamics, called internal models, which allow them to adapt motor commands to novel disturbances in the environment. To date this process has largely been described through learning curves characterizing changes in performance across movement repeats, yet recent evidence suggests that adaptive changes in reach representation may occur during a single movement. Here we sought to determine the latency of these rapid changes in movement representation based on electromyogram recordings. Healthy adults (n=18) performed standard reaching movement trials with a robotic handle randomly interleaved with force field trials (lateral force proportional to hand forward velocity) in unpredictable direction. First, we observed that the maximum hand displacement towards the direction of perturbation in a force field trial displayed very small improvement across trials, while the target overshoot was associated with a slower exponential decay. This result supports the presence of adaptive feedback control within each trial. Next, we identified in the main muscles involved in the motor corrections (shoulder flexors and extensors) a clear modulation explaining the reduction in target overshoot. Based on a sliding comparison of the difference between the first and last trials, we measured that the onset of the response tuned to the force field occurred on average at 237±15ms following reach onset (mean±SD). To further assess whether these corrections were tuned to the force field, we regressed the commanded force with the measured force and found highly significant improvements across trials. Moreover, the correlations became better than when measured force of a single trial was compared with commanded force of random trials. Abstract

Background: Acute lacunar ischaemic stroke, white matter hyperintensities, and lacunes are all features of cerebral small vessel disease. It is unclear why some small vessel disease lesions present with acute stroke symptoms, whereas others typically do not.\ud \ud Aim: To test if lesion location could be one reason why some small vessel disease lesions present with acute stroke, whereas others accumulate covertly.\ud \ud Methods: We identified prospectively patients who presented with acute lacunar stroke symptoms with a recent small subcortical infarct confirmed on magnetic resonance diffusion imaging. We compared the distribution of the acute infarcts with that of white matter hyperintensity and lacunes using computational image mapping methods.\ud \ud Results: In 188 patients, mean age 67 ± standard deviation 12 years, the lesions that presented with acute lacunar ischaemic stroke were located in or near the main motor and sensory tracts in (descending order): posterior limb of the internal capsule (probability density 0·2/mm3), centrum semiovale (probability density = 0·15/mm3), medial lentiform nucleus/lateral thalamus (probability density = 0·09/mm3), and pons (probability density = 0·02/mm3). Most lacunes were in the lentiform nucleus (probability density = 0·01–0·04/mm3) or external capsule (probability density = 0·05/mm3). The current study examined whether clusters of loneliness and depressive symptoms could be identified through latent profile analysis in two samples of 417 and 1140 adolescents (48.40 and 48.68 % male, respectively), on average 12.47 and 12.81 years old, respectively. Four clusters were identified, (1) low on loneliness and depressive symptoms, (2) low on loneliness and high on depressive symptoms, (3) high on loneliness and low on depressive symptoms, and (4) high on loneliness and depressive symptoms. We found that these four clusters were differentially related to friendship quantity and quality as well as to happiness and self-esteem. The current study stresses the importance of assessing both loneliness and depressive symptoms, as their mutual relation within individuals is differentially related to various aspects of adjustment.

Abstract

The Enhancing NeuroImaging Genetics through Meta-Analysis (ENIGMA) Consortium is a collaborative network of researchers working together on a range of large-scale studies that integrate data from 70 institutions worldwide. Organized into Working Groups that tackle questions in neuroscience, genetics, and medicine, ENIGMA studies have analyzed neuroimaging data from over 12,826 subjects. In addition, data from 12,171 individuals were provided by the CHARGE consortium for replication of findings, in a total of 24,997 subjects. By meta-analyzing results from many sites, ENIGMA has detected factors that affect the brain that no individual site could detect on its own, and that require larger numbers of subjects than any individual neuroimaging study has currently collected. ENIGMA's first project was a genome-wide association study identifying common variants in the genome associated with hippocampal volume or intracranial volume. Continuing work is exploring genetic associations with subcortical volumes (ENIGMA2) and white matter microstructure (ENIGMA-DTI). Working groups also focus on understanding how schizophrenia, bipolar illness, major depression and attention deficit/hyperactivity disorder (ADHD) affect the brain. We review the current progress of the ENIGMA Consortium, along with challenges and unexpected discoveries made on the way. © 2014 The Author(s).

Abstract

Age-associated disease and disability are placing a growing burden on society. However, ageing does not affect people uniformly. Hence, markers of the underlying biological ageing process are needed to help identify people at increased risk of age-associated physical and cognitive impairments and ultimately, death. Here, we present such a biomarker, ‘brain-predicted age’, derived using structural neuroimaging. Brain-predicted age was calculated using machine-learning analysis, trained on neuroimaging data from a large healthy reference sample (N=2001), then tested in the Lothian Birth Cohort 1936 (N=669), to determine relationships with age-associated functional measures and mortality. Having a brain-predicted age indicative of an older-appearing brain was associated with: weaker grip strength, poorer lung function, slower walking speed, lower fluid intelligence, higher allostatic load and increased mortality risk. Furthermore, while combining brain-predicted age with grey matter and cerebrospinal fluid volumes (themselves strong predictors) not did improve mortality risk prediction, the combination of brain-predicted age and DNA-methylation-predicted age did. This indicates that neuroimaging and epigenetics measures of ageing can provide complementary data regarding health outcomes. Our study introduces a clinically-relevant neuroimaging ageing biomarker and demonstrates that combining distinct measurements of biological ageing further helps to determine risk of age-related deterioration and death.

The volume of the hippocampus is a biomarker of incipient Alzheimer's disease and is reduced in schizophrenia, major depression and mesial temporal lobe epilepsy. Whereas many brain imaging phenotypes are highly heritable, identifying and replicating genetic influences has been difficult, as small effects and the high costs of magnetic resonance imaging (MRI) have led to underpowered studies. Here we report genome-wide association meta-analyses and replication for mean bilateral hippocampal, total brain and intracranial volumes from a large multinational consortium. The intergenic variant rs7294919 was associated with hippocampal volume (12q24.22; N = 21,151; P = 6.70 × 10 -16) and the expression levels of the positional candidate gene TESC in brain tissue. Additionally, rs10784502, located within HMGA2, was associated with intracranial volume (12q14.3; N = 15,782; P = 1.12 × 10 -12). We also identified a suggestive association with total brain volume at rs10494373 within DDR2 (1q23.3; N = 6,500; P = 5.81 × 10 -7). © 2012 Nature America, Inc. All rights reserved.
