Abstract

The 2013 decree defining the missions of higher education institutions in the Wallonia-Brussels Federation of Belgium identified applied research as a full-fledged mission of the university college. However, the professional project of teachers who evolve in university college is initially guided by teaching. In order to align its teaching staff with this new mission, one university of applied sciences has set up a call for internal projects. The study presented in this article takes as its object the novel professional development of teachers involved in research projects. With a view to the continuous improvement of the call process, its comprehensive and exploratory aim is to document the process of identity construction in which these teacher-researchers are involved, to identify the learning achieved as well as the facilitators and obstacles perceived. Finally, 51 teachers were interviewed by means of an individual online questionnaire. The data were analysed with a qualitative method. The results indicate, among other things, that the desire to do research is linked to the desire to improve and inform one's teaching practices. In this respect, the Scholaship of Teaching and learning approach (Brew, 2010) could shed light on the specificity of the professional development process in HE. The main obstacle remains the time allocated to the research project, which is both too short and difficult to combine with the time allocated to teaching. Levers are identified in the collective dynamics within and between project teams as well as in the institutional support that is necessary.

Abstract

Monoclonal antibodies are playing an increasing role in both human and animal health. Different strategies of protein and chemical engineering, including humanization techniques of non-human antibodies were applied successfully to optimize clinical performances of antibodies. Despite the emergence of techniques allowing the development of fully human antibodies such as transgenic Xeno-mice, antibody humanization remains a standard procedure for therapeutic antibodies. An important prerequisite for antibody humanization requires standardized numbering methods to define precisely complementary determining regions (CDR), frameworks and residues from the light and heavy chains that affect the binding affinity and/or specificity of the antibody-antigen interaction. The recently generated deep-sequencing data and the increasing number of solved three-dimensional structures of antibodies from human and non-human origins have led to the emergence of numerous databases. However, these different databases use different numbering conventions and CDR definitions. In addition, the large fluctuation of the variable chain lengths, especially in CDR3 of heavy chains (CDRH3), hardly complicates the comparison and analysis of antibody sequences and the identification of the antigen binding residues. This review compares and discusses the different numbering schemes and “CDR” definition that were established up to date. Furthermore, it summarizes concepts and strategies used for numbering residues of antibodies and CDR residues identification. Finally, it discusses the importance of specific sets of residues in the binding affinity and/or specificity of immunoglobulins.

Abstract

Bovine respiratory syncytial virus (BRSV) is associated with severe respiratory disease in cattle. BRSV infection frequently leads to the death of young infected animals. The presence of BRSV in postmortem specimens is routinely detected using indirect immunofluorescence (IIF). However, this technique requires special equipment and considerable expertise. The present paper describes the development of a 1-step ELISA for rapid (1.5 hours) detection of BRSV antigen in organ homogenates. The performance of the new 1-step ELISA was evaluated using bovine postmortem specimens ( n = 108) in comparison with 3 other BRSV diagnostic techniques: indirect immunofluorescence, the Clearview respiratory syncytial virus (RSV) test, and real-time reverse transcriptase polymerase chain reaction (RT-PCR). The relative sensitivity, specificity, and the kappa coefficient of 1-step ELISA, the Clearview RSV electroimmunoassay (EIA), and IIF were calculated, using real-time RT-PCR as the reference test. The new 1-step ELISA was the most sensitive and specific of the 3 tests. Thus, the new 1-step ELISA is a reliable test for detecting BRSV antigen in organ homogenates.

Abstract

Mycobacteria are important causes of infectious diseases. Although Mycobacterium tuberculosis and Mycobacterium leprae, two slow-growing species, are responsible for the most serious diseases, some fast-growing species, such as Mycobacterium avium, Mycobacterium kansasii, Mycobacterium chelonae, and Mycobacterium fortuitum, may cause opportunistic infections among AIDS patients (24). In addition, M. fortuitum has been reported to be responsible for a wide spectrum of clinical diseases, such as skin or soft tissue infections following surgery, pulmonary infections, accidental penetrating trauma, and wounds that come in contact with soil or water contaminated with these mycobacteria, though nosocomial infections are by far the most common (41, 44). The cell wall of mycobacteria contains mycolic acid, arabinogalactan, and peptidoglycan, forming a covalent complex. The influx of small hydrophilic agents through the resulting low-permeability envelope is extremely slow and is thought to be one of the major factors involved in the resistance of mycobacteria to β-lactam antibiotics (21, 41). β-Lactamase production, catalyzing β-lactam antibiotic hydrolysis, appears to be the second mechanism by which mycobacteria express β-lactam resistance (9, 21, 35). Chromosomally encoded β-lactamases have been detected in most, but not all, mycobacterial species, including M. fortuitum and M. tuberculosis (43). The class A β-lactamase of M. fortuitum (MFO) is a chromosomally encoded broad-spectrum β-lactamase hydrolyzing both cephalosporins and penicillins. Its substrate profile includes ureidopenicillins (piperacillin, azlocillin, and mezlocillin), carbenicillin, cephalothin, cefotaxime, and cefuroxime, but not ceftazidime. Cefoxitin, dicloxacillin, imipenem, and aztreonam are poorly recognized by the enzyme, and the protein is inhibited by the penem inhibitor BRL42715 and, to a lesser extent, clavulanic acid. The specificity profile is similar overall to those of CTX-M-type enzymes; some TEM-derived extended-spectrum β-lactamases (ESBLs); the chromosomally encoded ESBLs from Citrobacter sedlakii and Yersinia enterocolitica; and β-lactamases identified in the genera Nocardia, Kluyvera, and Burkholderia (1, 12, 31, 33, 35, 37). At the amino-acid level, the mature protein shares 70% identity with the β-lactamase of Mycobacterium smegmatis and between 40 and 45% with the aforementioned proteins. MFO also shares 41% amino acid identity with the ESBL Toho-1. Many recent studies have been concerned with the structure solution of TEM, SHV, or CTX-M-type ESBLs, but few structural data are available on chromosomally encoded broad-spectrum class A β-lactamases. MFO is the first solved structure of a mycobacterial β-lactamase. Its structure-activity relationship was analyzed on the basis of a comparison between its structure and recently solved ESBL structures (5, 27, 29, 38).

Introduction Le froment est une source allergenique complexe contenant un grand nombre de proteines qui sont difficiles a isoler et a identifier. Le but de ce projet est de developper une methode de diagnostic capable de relier les allergenes specifiques a une clinique particuliere via western blot (WB) 2D. Ensuite, la technique de spectrometrie de masse (LC-MS/MS) a ete utilisee pour identifier ces allergenes. Methodes Une extraction proteique suivie d’une separation sur base du point isoelectrique et du poids moleculaire des proteines ont ete realisees. Vingt-cinq patients presentant des IgE specifiques (IgEs) pour le froment ont ete selectionnes et classes en 3 groupes : anaphylaxie alimentaire induite par l’effort (AAIE), dermatite atopique (DA) et rhinite pollinique (RP). Leur serum a ete analyse par WB 2D afin d’evaluer la reactivite de leurs IgEs. Les profils de sensibilisation ont ete compares et les spots proteiques d’interet identifies par LC-MS/MS. Resultats Des profils de sensibilisation specifiques ont ete identifies pour chaque groupe. Pour le groupe AAIE, les spots proteiques sont situes a 35 kDa, pH6-9 et a 37–50 kDa, pH5-6. Pour le groupe DA, le profil est localise autour de 50 kDa, pH9 ; 10 kDa, pH9 et a 20–75 kDa, pH3. Pour le groupe RP, le profil est situe vers 90 kDa, pH9. Les analyses en LC-MS/MS de ces spots proteiques ont mis en evidence differents allergenes potentiellement interessants tels que tri a 26, tri a bA, tri a 34 et tri a tritin. Discussion L’etude presentee repond a une demande clinique afin de prevoir les reactions croisees, d’adapter les regimes et de limiter le risque de choc anaphylactique. Neanmoins, les resultats restent a correler a l’anamnese. Les manifestations cliniques dependent des allergenes impliques et de la voie d’exposition. Ce projet vise a mettre en evidence de nouveaux allergenes qui pourraient etre associes a une symptomatologie clinique specifique. Conclusion Des profils de sensibilisation specifiques ont ete identifies pour les 3 groupes, mais l’identification de certains spots proteiques est encore en cours d’analyse. A terme, il sera possible d’etablir un lien entre la symptomatologie specifique et les allergenes responsables nouvellement identifies.

Abstract

Insertion of heterologous peptide sequences into a protein carrier may impose structural constraints that could help the peptide to adopt a proper fold. This concept could be the starting point for the development of a new generation of safe subunit vaccines based on the expression of poorly immunogenic epitopes. In the present study, we characterized the tolerance of the TEM-1 class A beta-lactamase to the insertion of two different peptides, the V3 loop of the gp120 protein of HIV, and the thermostable STa enterotoxin produced by enterotoxic Escherichia coli. Insertion of the V3 loop of the HIV gp120 protein into the TEM-1 scaffold yielded insoluble and poorly produced proteins. By contrast, four hybrid beta-lactamases carrying the STa peptide were efficiently produced and purified. Immunization of BALB/c mice with these hybrid proteins produced high levels of TEM-1-specific antibodies, together with significant levels of neutralizing antibodies against STa.

Abstract

Xyl1 from Streptomyces sp. S38 belongs to the low molecular mass family 11 of endo-beta-1,4-xylanases. Its three-dimensional structure has been solved at 2.0 A and its optimum temperature and pH for enzymatic activity are 60 degrees C and 6.0, respectively. Aspergillus kawachii xylanase XynC belongs to the same family but is an acidophilic enzyme with an optimum pH of 2.0. Structural comparison of Xyl1 and XynC showed differences in residues surrounding the two glutamic acid side chains involved in the catalysis that could be responsible for the acidophilic adaptation of XynC. Mutations W20Y, N48D, A134E, and Y193W were introduced by site-directed mutagenesis and combined in multiple mutants. Trp 20 and Tyr 193 are involved in substrate binding. The Y193W mutation inactivated Xyl1 whereas W20Y decreased the optimum pH of Xyl1 to 5.0 and slightly increased its specific activity. The N48D mutation also decreased the optimum pH of Xyl1 by one unit. The A134E substitution did not induce any change, but when combined with N48D, a synergistic effect was observed with a 1.4 unit decrease in the optimum pH. Modeling showed that the orientations of residue 193 and of the fully conserved Arg 131 are different in acidophilic and "alkaline" xylanases whereas the introduced Tyr 20 probably modifies the pKa of the acid-base catalyst via residue Asn 48. Docking of a substrate analog in the catalytic site highlighted striking differences between Xyl1 and XynC in substrate binding. Hydrophobicity calculations showed a correlation between acidophilic adaptation and a decreased hydrophobicity around the two glutamic acid side chains involved in catalysis.

Abstract

International audience; Mycobacterium fallax (M. fallax) is naturally sensitive to many beta-lactam antibiotics (MIC < 2 microg/ml) and devoid of beta-lactamase activity. In this paper, we show that the production of the beta-lactamase of Mycobacterium fortuitum by M. fallax significantly increased the MIC values for good substrates of the enzyme, whereas the potency of poor substrates or transient inactivators was not modified. The rates of diffusion of beta-lactams through the mycolic acid layer were low, but for all studied compounds the half-equilibration times were such that they would only marginally affect the MIC values in the absence of beta-lactamase production. These results emphasize the importance of enzymatic degradation as a major factor in the resistance of mycobacteria to penicillins.

The beta-lactamase of Mycobacterium smegmatis mc(2)155 has been purified to protein homogeneity. Its N-terminal sequence and catalytic properties are similar to those of the beta-lactamase produced by Mycobacterium fortuitum D316 and establish this new enzyme as a member of molecular class A.

Abstract

Sixteen different compounds usually considered beta-lactamase stable or representing potential beta-lactam inhibitors and inactivators were tested against the beta-lactamase produced by Mycobacterium fortuitum. The compounds exhibiting the most interesting properties were BRL42715, which was by far the best inactivator, and CGP31608 and ceftazidime, which were not recognized by the enzyme. These compounds thus exhibited adequate properties for fighting mycobacterial infections. Although cloxacillin, dicloxacillin, cefoxitin, and CP65207-2 exhibited poor inhibitory efficiency against the enzyme, they were also rather poor substrates and might be considered potential antimycobacterial agents. By contrast, CGP31523A and ceftamet were good substrates.
