Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) continues to evolve and recently emerging variants with substitutions in the Spike protein have led to growing concerns over increased transmissibility and decreased vaccine coverage due to immune evasion. Here, sera from recipients of a single dose of our Ad26.COV2.S COVID-19 vaccine were tested for neutralizing activity against several SARS-CoV-2 variants of concern. All tested variants demonstrated susceptibility to Ad26.COV2.S-induced serum neutralization albeit mainly reduced as compared to the B.1 strain. Most pronounced reduction was observed for the B.1.351 (Beta; 3.6-fold) and P.1 (Gamma; 3.4-fold) variants that contain similar mutations in the receptor-binding domain (RBD) while only a 1.6-fold reduction was observed for the widely spreading B.1.617.2 (Delta) variant.

While single-dose COVID-19 vaccines are preferred, two-dose regimens may improve efficacy. Immunogenicity of Ad26.COV2.S is improved in NHPs after a second dose, whereas both regimens protect animals against SARS-CoV-2 disease and elicit neutralizing antibodies against SARS-CoV-2 variants of concern to varying degrees independent of regimen., Safe and effective coronavirus disease–19 (COVID-19) vaccines are urgently needed to control the ongoing pandemic. While single-dose vaccine regimens would provide multiple advantages, two doses may improve the magnitude and durability of immunity and protective efficacy. We assessed one- and two-dose regimens of the Ad26.COV2.S vaccine candidate in adult and aged nonhuman primates (NHPs). A two-dose Ad26.COV2.S regimen induced higher peak binding and neutralizing antibody responses compared with a single dose. In one-dose regimens, neutralizing antibody responses were stable for at least 14 wk, providing an early indication of durability. Ad26.COV2.S induced humoral immunity and T helper cell (Th cell) 1–skewed cellular responses in aged NHPs that were comparable to those in adult animals. Aged Ad26.COV2.S-vaccinated animals challenged 3 mo after dose 1 with a SARS-CoV-2 spike G614 variant showed near complete lower and substantial upper respiratory tract protection for both regimens. Neutralization of variants of concern by NHP sera was reduced for B.1.351 lineages while maintained for the B.1.1.7 lineage independent of Ad26.COV2.S vaccine regimen.

Background In a Phase 3 trial, the Janssen COVID-19 vaccine, Ad26.COV2.S, showed robust efficacy against severe–critical COVID-19 in countries where different SARS-CoV-2 variants were circulating. We evaluated Ad26.COV2.S-elicited antibody neutralizing activity against variants of concern (VOC) B.1.1.7 (Alpha), B.1.351 (Beta), and B.1.617.2 (Delta) in sera from participants in clinical trials following a single dose of Ad26.COV2.S. Methods Neutralizing activities of Ad26.COV2.S (given at a dose level of 5 x 1010 viral particles [vp]) against VOC were assessed by wild-type virus neutralizing (wtVNA) and pseudovirion neutralization (psVNA) assays in sera from participants in Phase 1/2a and Phase 3 clinical trials, respectively. Geometric mean titers (GMTs) were determined at Days 29 and 71 after vaccination. Results In serum samples from Phase 1/2a participants (n = 6), at Day 29 after 1 dose of Ad26.COV2.S, wtVNA titers against VOC were lower than for the original strain (GMT = 573), with GMT = 65, 14, and 15 for Alpha, Beta, and Delta, respectively, representing 8.8-, 40.9-, and 37.7-fold decreases. By Day 71 after vaccination (n = 14), fold differences between the original strain (GMT = 375) and VOC (GMT = 113, 27, and 28) were smaller (3.3-, 13.9-, and 13.4-fold) than at Day 29, suggestive of B-cell maturation (Figure 1). Day 71 titers against the Delta variant were maintained for at least 8 months following a single dose of Ad26.COV2.S (5 x 1010 vp). In serum samples from Phase 3 participants (n = 8), psVNA titers against VOC were lower than the original strain at Day 71 after vaccination, with the lowest titers observed for the Beta variant (3.6-fold decrease vs original strain). Conclusion Ad26.COV2.S-elicited serum neutralizing activity against VOC showed an overall decrease in titers relative to the original strain that was largest for the Beta variant, even though vaccine efficacy against severe–critical COVID-19 was maintained in countries where these variants were circulating versus in countries where they were not circulating. Over time, titers against variants increased, suggesting B-cell affinity maturation leading to increasing coverage of VOC. Broadly reactive drugs for flu Drugs for influenza are limited. For those available, viral resistance is rife. Part of the problem is that the virus is constantly mutating. Kadam et al. tested the cell entry stage of the virus life cycle as a drug target (see the Perspective by Whitehead). Cell entry is mediated by the major surface glycoprotein hemagglutinin (HA). This stage can be blocked by broadly neutralizing antibodies binding to HA. The authors generated small cyclic peptides that bind to the same sites on HA as the antibodies and mimic their activity. The peptides are cheap and easy to synthesize, are nontoxic to mice, and prevented infection of cells by many types of influenza virus. Science , this issue p. 496 ; see also p. 450

A small molecule that targets influenza Many of us rely on seasonal vaccines for protection against influenza and are only too aware of their limited breadth. Broadly neutralizing antibodies (bnAbs) that target the conserved hemagglutinin (HA) stem of the influenza virus provide hope for the development of universal vaccines and are being evaluated in clinical trials. Van Dongen et al. selected and optimized a small-molecule lead compound that recapitulates key interactions of the bnAb with HA. Like the bnAb, the compound inhibited viral fusion in the endosomes of target cells. The compound protected mice from influenza after oral administration and neutralized virus infection in a 3D cell culture of human bronchial epithelial cells. Science , this issue p. eaar6221

Seasonal vaccines are currently the most effective countermeasure against influenza. However, seasonal vaccines are only effective against strains closely related to the influenza strains contained in the vaccine. Recently a new hemagglutinin (HA) stem-based antigen, the so-called “mini-HA”, has been shown to induce a cross-protective immune response in influenza-naive mice and non-human primates (NHP). However, prior exposure to influenza can have a profound effect on the immune response to subsequent influenza infection and the protective efficacy of vaccination. Here we show that mini-HA, compared to a trivalent influenza vaccine (TIV), elicits a broadened influenza-specific humoral immune response in NHP previously exposed to influenza. Serum transfer experiments showed that antibodies induced by both mini-HA and seasonal vaccine protected mice against lethal challenge with a H1N1 influenza strain heterologous to the H1 HA included in the TIV. However, antibodies elicited by mini-HA showed an additional benefit of protecting mice against lethal heterosubtypic H5N1 influenza challenge, associated with H5 HA-specific functional antibodies., Influenza: a novel vaccine offers broad protection A vaccine candidate developed from a novel flu-based protein induces antibodies able to protect against multiple influenza strains. Alongside a team of Dutch researchers, Janssen’s Joan van der Lubbe created a vaccine, dubbed “mini-HA”, based on a recently discovered influenza protein that induced neutralizing antibodies to many related influenza virus strains in non-human primates. Mini-HA vaccination targeted a surface protein region that is highly conserved between influenza strains and offered protection regardless of previous exposure to the disease, which is known to greatly affect vaccine efficacy. The immune response elicited by the novel vaccine surpassed that of a standard seasonal influenza vaccine. Future research is now warranted to verify whether mini-HA can act as a universal seasonal flu vaccine, potentially even effective against emergent virus strains.

Durable influenza protection Vaccines are indispensable for the control and prevention of influenza, but there are several challenges to efficacy. Some individuals respond poorly to vaccination, and virus variation makes targeting optimal antigens difficult. Broadly neutralizing antibodies are one solution, but they have their own pitfalls, including limited cross-reactivity to both influenza A and B strains and the need for repeated injections. Now, Laursen et al. have developed multidomain antibodies with breadth and potency. Administered intranasally to mice with an adeno-associated virus vector, the antibodies provided durable and continuous protection from a panoply of influenza strains. Science , this issue p. 598

Interactions with receptors for the Fc region of IgG (Fc gamma Rs) have been shown to contribute to the in vivo protection against influenza A viruses provided by broadly neutralizing antibodies (bnAbs) that bind to the viral hemagglutinin (HA) stem. In particular, Fc-mediated antibody-dependent cellular cytotoxicity (ADCC) has been shown to contribute to protection by stem-binding bnAbs. Fc-mediated effector functions appear not to contribute to protection provided by strain-specific HA head-binding antibodies. We used a panel of anti-stem and anti-head influenza A and B monoclonal antibodies with identical human IgG1 Fc domains and investigated their ability to mediate ADCC-associated Fc gamma RIIIa activation. Antibodies which do not interfere with sialic acid binding of HA can mediate Fc gamma RIIIa activation. However, the Fc gamma RIIIa activation was inhibited when a mutant HA, unable to bind sialic acids, was used. Antibodies which block sialic acid receptor interactions of HA interfered with Fc gamma RIIIa activation. The inhibition of Fc gamma RIIIa activation by HA head-binding and sialic acid receptor-blocking antibodies was confirmed in plasma samples of H5N1 vaccinated human subjects. Together, these results suggest that in addition to Fc-Fc gamma R binding, interactions between HA and sialic acids on immune cells are required for optimal Fc-mediated effector functions by anti-HA antibodies

The ability to directly visualize nanoscopic cellular structures and their spatial relationship in all three dimensions will greatly enhance our understanding of molecular processes in cells. Here, we demonstrated multicolor three-dimensional (3D) stochastic optical reconstruction microscopy (STORM) as a tool to quantitatively probe cellular structures and their interactions. To facilitate STORM imaging, we generated photoswitchable probes in several distinct colors by covalently linking a photoswitchable cyanine reporter and an activator molecule to assist bioconjugation. 3D localization was performed in conjunction with focal plane scanning and correction for refractive index mismatch to obtain whole-cell images with a spatial resolution of 20–30 nm and 60–70 nm in the lateral and axial dimensions, respectively. Using this approach, we imaged the entire mitochondrial network in fixed monkey kidney BS-C-1 cells, and studied the spatial relationship between mitochondria and microtubules. The 3D STORM images revealed mitochondrial morphologies as well as mitochondria-microtubule contacts that were obscured in conventional fluorescence images.

Protein transduction domains (PTDs) have been used to deliver a variety of biologically active cargo across cellular membranes. However the potential of PTDs to mediate transport of nanoparticular structures into the cytoplasm bypassing the endosomal compartment remains unclear. Cell-permeable virus-like particles (VLPs) harboring a marker gene based on hepatitis B virus nucleocaspids were established. Cell permeability was achieved by fusion with translocation motif (TLM)-PTD. Electron and confocal microscopy revealed that these VLPs translocate as complete particles across the plasma membrane and transverse the cytoplasm toward the nucleus. Inhibition of endocytosis did not affect translocation of these VLPs into the cytoplasm. Based on these particles, a gene transfer system was developed. To this end the particles were loaded with DNA-encoding small hepatitis B virus surface antigen (SHBs) or green fluorescence protein (eGFP) that served as marker genes. Although the DNA-packaging efficiency was very low, applying the appropriate number of VLPs to primary human hepatocytes a gene transfer efficiency of approximately 95% was observed. In conclusion, the TLM-PTD has the potential to mediate efficient transfer of assembled particles and its cargo, nucleic acids, into primary human hepatocytes. Die nachfolgend beschriebenen Forschungsprojekte der Nachwuchsgruppe I am Robert Koch-Institut (RKI) befassen sich am Beispiel des Hepatitis-B-Virus (HBV) mit der Charakterisierung intrazellularer Signalkaskaden und der Analyse ihrer Bedeutung fur den viralen Lebenszyklus (Replikaktionsprozess) sowie fur die Entstehung von HBV assoziierten Tumoren. Daruber hinaus werden auf der Basis eines neuen, aus dem HBV-Oberflachenprotein isolierten zellpermeablen Peptides neue Strategien zum Protein- und Gentransfer entwickelt.

The ability of antibodies binding the influenza hemagglutinin (HA) protein to neutralize viral infectivity is of key importance in the design of next-generation vaccines and for prophylactic and therapeutic use. The two antibodies CR6261 and CR8020 have recently been shown to efficiently neutralize influenza A infection by binding to and inhibiting the influenza A HA protein that is responsible for membrane fusion in the early steps of viral infection. Here, we use single-particle fluorescence microscopy to correlate the number of antibodies or antibody fragments (Fab) bound to an individual virion with the capacity of the same virus particle to undergo membrane fusion. To this end, individual, infectious virus particles bound by fluorescently labeled antibodies/Fab are visualized as they fuse to a planar, supported lipid bilayer. The fluorescence intensity arising from the virus-bound antibodies/Fab is used to determine the number of molecules attached to viral HA while a fluorescent marker in the viral membrane is used to simultaneously obtain kinetic information on the fusion process. We experimentally determine that the stoichiometry required for fusion inhibition by both antibody and Fab leaves large numbers of unbound HA epitopes on the viral surface. Kinetic measurements of the fusion process reveal that those few particles capable of fusion at high antibody/Fab coverage display significantly slower hemi-fusion kinetics. Overall, our results support a membrane fusion mechanism requiring the stochastic, coordinated action of multiple HA trimers and a model of fusion inhibition by stem-binding antibodies through disruption of this coordinated action

Human monoclonal antibodies have been identified which neutralize broad spectra of influenza A or B viruses. Here, we dissect the mechanisms by which such antibodies interfere with infectivity. We distinguish four mechanisms that link the conserved hemagglutinin (HA) epitopes of broadly neutralizing antibodies to critical processes in the viral life cycle. HA-stem binding antibodies can act intracellularly by blocking fusion between the viral and endosomal membranes and extracellularly by preventing the proteolytic activation of HA. HA-head binding antibodies prevent viral attachment and release. These insights into newly identified ways by which the human immune system can interfere with influenza virus infection may aid the development of novel universal vaccines and antivirals.

Current flu vaccines provide only limited coverage against seasonal strains of influenza viruses. The identification of VH1-69 antibodies that broadly neutralize almost all influenza A group 1 viruses constituted a breakthrough in the influenza field. Here, we report the isolation and characterization of a human monoclonal antibody CR8020 with broad neutralizing activity against most group 2 viruses, including H3N2 and H7N7, which cause severe human infection. The crystal structure of Fab CR8020 with the 1968 pandemic H3 hemagglutinin (HA) reveals a highly conserved epitope in the HA stalk distinct from the epitope recognized by the VH1-69 group 1 antibodies. During the course of an infection, viruses take advantage of a variety of mechanisms to travel in cells, ranging from diffusion within the cytosol to active transport along cytoskeletal filaments. To study viral motility within the intrinsically heterogeneous environment of the cell, we have developed a motility assay that allows for the global and unbiased analysis of tens of thousands of virus trajectories in live cells. Using this assay, we discovered that poliovirus exhibits anomalously rapid intracellular movement that was independent of microtubules, a common track for fast and directed cargo transport. Such rapid motion, with speeds of up to 5 μm/s, allows the virus particles to quickly explore all regions of the cell with the exception of the nucleus. The rapid, microtubule-independent movement of poliovirus was observed in multiple human-derived cell lines, but appeared to be cargo-specific. Other cargo, including a closely related picornavirus, did not exhibit similar motility. Furthermore, the motility is energy-dependent and requires an intact actin cytoskeleton, suggesting an active transport mechanism. The speed of this microtubule-independent but actin-dependent movement is nearly an order of magnitude faster than the fastest speeds reported for actin-dependent transport in animal cells, either by actin polymerization or by myosin motor proteins.

What could be a better way to study virus trafficking than ‘miniaturizing oneself’ and ‘taking a ride with the virus particle’ on its journey into the cell? Single-virus tracking in living cells potentially provides us with the means to visualize the virus journey. This approach allows us to follow the fate of individual virus particles and monitor dynamic interactions between viruses and cellular structures, revealing previously unobservable infection steps. The entry, trafficking and egress mechanisms of various animal viruses have been elucidated using this method. The combination of single-virus trafficking with systems approaches and state-of-the-art imaging technologies should prove exciting in the future.

Infectious entry of hepatitis B viruses (HBV) has nonconventional facets. Here we analyzed whether a cell-permeable peptide [translocation motif (TLM)] identified within the surface protein of human HBV is a general feature of all hepadnaviruses and plays a role in the viral life cycle. Surface proteins of all hepadnaviruses contain conserved functional TLMs. Genetic inactivation of the duck HBV TLMs does not interfere with viral morphogenesis; however, these mutants are noninfectious. TLM mutant viruses bind to cells and are taken up into the endosomal compartment, but they cannot escape from endosomes. Processing of surface protein by endosomal proteases induces their exposure on the virus surface. This unmasking of TLMs mediates translocation of viral particles across the endosomal membrane into the cytosol, a prerequisite for productive infection. The ability of unmasked TLMs to translocate processed HBV particles across cellular membranes was shown by confocal immunofluorescence microscopy and by infection of nonpermissive cell lines with HBV processed in vitro with endosomal lysate. Based on these data, we propose an infectious entry mechanism unique for hepadnaviruses that involves virus internalization by receptor-mediated endocytosis followed by processing of surface protein in endosomes. This processing activates the function of TLMs that are essential for viral particle translocation through the endosomal membrane into the cytosol and productive infection.

One of the first steps of influenza infection is fusion of the viral phospholipid envelope to a cellular membrane, opening a pore for the viral genome to enter a cell. The viral surface protein hemagglutinin (HA) mediates the fusion between these two lipid membranes and is a principal antigenic target for our immune system. Recently, antibodies capable of neutralizing both group 1 and group 2 influenza A viruses have been described. Here, we have visualized the interplay between a virus and antibodies during neutralization using our recently developed single-particle fusion assay. To this end, antibodies (mAb) or antibody fragments (Fab) and infectious viral particles were fluorescently labeled and incubated together. Viruses were then surface-immobilized to sialic acid decorated proteins incorporated into a planar, supported phospholipid bilayer and fusion induced by addition of an acidic buffer. Individual hemifusion events were observed as a dequenching spike, resulting from the rapid release of lipophilic dye from the viral membrane into the target bilayer. using this novel setup, we directly link the action of individual HA-binding mAb or Fab to viral fusion events and confirm that both act directly to inhibit HA-mediated membrane fusion in a dose-dependent fashion. Furthermore, we use the fluorescence intensity per virion to calculate the number of virus-bound mAb or Fab - information obfuscated in traditional cell-cell and bulk fusion assays. Our data indicate that total hemifusion inhibition occurs in a mAb/Fab coverage regime where hundreds of HA trimers remain fusion competent. Physical modeling further indicates that within this binding regime, these unbound HA can occur in spatial geometries allowing for the formation of fusogenic clusters. Overall, our results have explicit implications for the model of bilayer fusion arising from the coordinated efforts of multiple hemagglutinin molecules.

Influenza viruses have hundreds of hemagglutinin (HA) protein trimers embedded in their phospholipid envelope. The HA protein is the lynchpin in the first stage of influenza infection, first by facilitating binding and internalization into target host cells and then by mediating fusion with the membrane of late endosomes. Recently, antibodies capable of neutralizing both group 1 and group 2 influenza A viruses have been described [Science,v324,p246,2009; Science,v333,p843,2011]. These broadly neutralizing HA stem-binding antibodies appear to act by blocking the HA conformational change necessary for fusion. Here, we have visualized the interplay between a virus and antibodies during neutralization using our recently developed single-particle fusion assay [PNAS,v105(40),p15382,2008]. To this end, antibodies and infectious viral particles were fluorescently labeled and incubated together. Viruses were then bound to sialic acid decorated proteins incorporated into a planar, supported phospholipid bilayer and fusion induced by addition of an acidic buffer. Using TIRF microscopy, individual hemifusion events were observed as the rapid release of lipophilic dye from the viral membrane into the target bilayer. This novel approach allows us to directly link the action of individual HA-binding antibodies to viral fusion events, providing new insight into the mechanism and molecular stoichiometry involved in fusion and neutralization - information obfuscated in traditional cell-cell and bulk fusion assays. We see a concentration dependent effect of inhibiting antibodies on the time it takes for viruses to undergo hemifusion as well as on the total number of hemifusion events. Furthermore, we can correlate the fluorescence intensity per virion to the number of bound antibodies and thereby directly measure the number of HA molecules involved in fusion. More to the point, our results have explicit implications for the model of bilayer fusion arising from the coordinated efforts of multiple hemagglutinins.

A super-resolution image was then constructed by plotting the measured probe positions accumulated over time. With this we have generated 3D whole cell images, several micrometers thick, with a spatial resolution of 20 - 30 nm and 60 - 70 nm in the lateral and axial dimensions, respectively. Using this approach, we imaged the entire mitochondrial network in mammalian cells and studied the spatial relationship between mitochondria and the microtubule cytoskeleton. The 3D STORM images clearly resolved the hollow mitochondria outer membrane structures obscured in conventional fluorescence images. Distinct mitochondrial morphologies were observed, ranging from thin elongated tubes to globular compartments. Interestingly, while globular mitochondria are relatively dispersed in size, from 200 nm to 1500 nm, the tubular structures are more uniform in diameter, taking a narrow distribution around 200 nm. The images also displayed several distinct interaction modes between mitochondria and microtubules. Notably, elongated mitochondria were observed to “inchworm” along microtubules with discrete attachment sites, while such an interaction mode was completely unresolvable with conventional fluorescence. Super-resolution optical microscopy techniques such as STORM promise to significantly expand the understanding of biological structures and their interactions on a molecular level.
