The PREDICTS project-Projecting Responses of Ecological Diversity In Changing Terrestrial Systems (www.predicts.org.uk)-has collated from published studies a large, reasonably representative database of comparable samples of biodiversity from multiple sites that differ in the nature or intensity of human impacts relating to land use. We have used this evidence base to develop global and regional statistical models of how local biodiversity responds to these measures. We describe and make freely available this 2016 release of the database, containing more than 3.2 million records sampled at over 26,000 locations and representing over 47,000 species. We outline how the database can help in answering a range of questions in ecology and conservation biology. To our knowledge, this is the largest and most geographically and taxonomically representative database of spatial comparisons of biodiversity that has been collated to date; it will be useful to researchers and international efforts wishing to model and understand the global status of biodiversity.

Land-use change and intensification threaten bee populations worldwide, imperilling pollination services. Global models are needed to better characterise, project, and mitigate bees' responses to these human impacts. The available data are, however, geographically and taxonomically unrepresentative; most data are from North America and Western Europe, overrepresenting bumblebees and raising concerns that model results may not be generalizable to other regions and taxa. To assess whether the geographic and taxonomic biases of data could undermine effectiveness of models for conservation policy, we have collated from the published literature a global dataset of bee diversity at sites facing land-use change and intensification, and assess whether bee responses to these pressures vary across 11 regions (Western, Northern, Eastern and Southern Europe; North, Central and South America; Australia and New Zealand; South East Asia; Middle and Southern Africa) and between bumblebees and other bees. Our analyses highlight strong regionally-based responses of total abundance, species richness and Simpson's diversity to land use, caused by variation in the sensitivity of species and potentially in the nature of threats. These results suggest that global extrapolation of models based on geographically and taxonomically restricted data may underestimate the true uncertainty, increasing the risk of ecological surprises.

Here we show that the micromolar threshold can easily be reached by employing laser-polarized xenon and cage molecules reversibly hosting it. Indirect detection of caged xenon is made via chemical exchange, using ultra-fast Z spectroscopy based on spatio-temporal encoding. On this non-dedicated low-field spectrometer, several ideas are proposed to improve the signal.

International audience; The nuclear polarization of noble gases, such as xenon, can be enhanced by several orders of magnitude through optical pumping, giving rise to very sensitive NMR probes. Moreover, due to its large and deformable electron cloud, xenon exhibits wide chemical shift range (more than 320 ppm for the monoatomic species) and is soluble in most biological fluids. It can be reversibly encapsulated in molecular host systems such as cage-molecules, decorated with ligands in order to target given receptors, and gives rise to sensitive detection of biological events. This two-step procedure, where the xenon host is first introduced and hyperpolarized xenon then delivered, benefits from the difference in resonance frequency between bound xenon and free xenon (in the gas phase or in the dissolved phase) [1]. This work has recently been published in Magnetic Resonance [2].

International audience; Investigation of systems in evolution such as chemical reactions via NMR can be rendered com-fortable if the intrinsic lack of sensitivity of this technique is circumvented and provided that quantitative data are obtained. Here we present an integrated device based on a 3D-printed mini bubble-pump associated with fluidics and micro-detection that overcomes the sensitivity problems inherent to slow return of magnetization to equilibrium in liquid-state NMR. The use of a closed-loop circuit of the solution near the NMR magnetic center presents two main advantages: pre-polarization is achieved for the whole solution volume, this volume can be reduced to tens of microliters. This device is installable into every commercial liquid probehead without modification; it is easily inserted from the top of the NMR magnet. A gas flow driven by a programmable syringe pump actuates a mini bubble-pump which leads to circulation of the liquid sample. A part of the solution circuit crosses the NMR detection region consisting of a micro-solenoid inductively coupled to the coil of the commercial probehead. In order to optimize this coupling a rod fixed on the upper part of the insert and ended by a Vernier placed on top of the magnet enables angular positioning of the micro-coil. The two resonance frequencies created by the coupling allow one to observe nuclei inaccessible with the host probe alone, or to study two different nuclei with the optimized detection allowed by the micro coil. To further increase the signal-to-noise ratio, this system can also be used to effi-ciently dispense gaseous species such as hyperpolarized xenon and parahydrogen to the solution. The performances of this device, in particular with cryoprobes, will be presented.

International audience; Xenon has several interesting properties for the NMR study of biological cells: 1) its nuclear polarization can beboosted via optical pumping, which increases the detection sensitivity by several orders of magnitude. 2) It is nontoxicand soluble in biologic medium and crosses the plasma membrane while keeping its polarization. 3) It has a wide rangeof chemical shift which makes its NMR signature sensitive to fine cell changes. In particular with cell suspensions twodistinct signals are observed on the $^{129}$Xe NMR spectrum, corresponding to xenon in the bulk and xenon inside thecells. This has led to promising applications, such as the discrimination of cells sensitive and resistant to chemotherapy.In this work, we studied Saccharomyces Cerevisiae cells at different times of a culture via hyperpolarized $^{129}$Xe NMR(xenon was hyperpolarized using our home made optical pumping setup) and controlled the number of cells by themeasure of the optical density at 600 nm. For each time stage of the yeast culture, a sample was withdrawn and afterhyperpolarized xenon addition, the corresponding $^{129}$Xe spectra were analyzed in a 11.7 Tesla spectrometer: thechemical shift splitting, the in-out exchange rate and the proportion of xenon inside the cells were extracted andcompared with classical analyses using optical density. The evolution of the spectral signature of $^{129}$Xe in S. Cerevisae isdependent of the culture time as presented in the figure. Hypotheses of explanations will be presented.In parallel, new methodologies, compatible with narrow-bore NMR spectrometers, were developed to allow the in situ follow-up of cell cultures by NMR, using microfluidic system and micro-NMR detection. - Abstract
We report the synthesis of C3-symmetric cryptophanes decorated with three aromatic amine groups on the same CTB cap and their interaction with xenon. The relative stereochemistry of these two stereoisomers synand antiwas assessed thanks to the determination of the X-ray structure of an intermediate compound. As previously observed with the tris-aza-cryptophanes analogs anti-1and syn-2(J. Org. Chem. 2021, 86, 11, 7648–7658), both compounds anti-5and syn-6show a slow in–out exchange dynamics of xenon at 11.7 T. Our work supports the idea that the presence of nitrogen atoms grafted directly onto the cryptophane backbone has a strong impact on the in–out exchange dynamics of xenon whatever their stereochemistry. This result contrasts with the case of other cryptophanes decorated solely with methoxy substituents. Finally, we demonstrate that these new derivatives can be used to design new anti/syncryptophanes bearing suitable ligands in order to constitute potent 129Xe NMR-based sensors. An example is reported here with the synthesis of the tris-iodo derivatives anti-13and syn-14from compounds anti-5and syn-6.

The availability of a benchtop nuclear magnetic resonance (NMR) spectrometer, of low cost and easily transportable, can allow detection of low quantities of biosensors, provided that hyperpolarized species are used. Here we show that the micromolar threshold can easily be reached by employing laser-polarized xenon and cage molecules reversibly hosting it. Indirect detection of caged xenon is made via chemical exchange, using ultra-fast Z spectroscopy based on spatio-temporal encoding. On this non-dedicated low-field spectrometer, several ideas are proposed to improve the signal. [ABSTRACT FROM AUTHOR]

The PREDICTS project—Projecting Responses of Ecological Diversity In Changing Terrestrial Systems (www.predicts.org.uk)—has collated from published studies a large, reasonably representative database of comparable samples of biodiversity from multiple sites that differ in the nature or intensity of human impacts relating to land use. We have used this evidence base to develop global and regional statistical models of how local biodiversity responds to these measures. We describe and make freely available this 2016 release of the database, containing more than 3.2 million records sampled at over 26,000 locations and representing over 47,000 species. We outline how the database can help in answering a range of questions in ecology and conservation biology. To our knowledge, this is the largest and most geographically and taxonomically representative database of spatial comparisons of biodiversity that has been collated to date; it will be useful to researchers and international efforts wishing to model and understand the global status of biodiversity.

Biodiversity continues to decline in the face of increasing anthropogenic pressures such as habitat destruction, exploitation, pollution and introduction of alien species. Existing global databases of species' threat status or population time series are dominated by charismatic species. The collation of datasets with broad taxonomic and biogeographic extents, and that support computation of a range of biodiversity indicators, is necessary to enable better understanding of historical declines and to project - and avert - future declines. We describe and assess a new database of more than 1.6 million samples from 78 countries representing over 28,000 species, collated from existing spatial comparisons of local-scale biodiversity exposed to different intensities and types of anthropogenic pressures, from terrestrial sites around the world. The database contains measurements taken in 208 (of 814) ecoregions, 13 (of 14) biomes, 25 (of 35) biodiversity hotspots and 16 (of 17) megadiverse countries. The database contains more than 1% of the total number of all species described, and more than 1% of the described species within many taxonomic groups - including flowering plants, gymnosperms, birds, mammals, reptiles, amphibians, beetles, lepidopterans and hymenopterans. The dataset, which is still being added to, is therefore already considerably larger and more representative than those used by previous quantitative models of biodiversity trends and responses. The database is being assembled as part of the PREDICTS project (Projecting Responses of Ecological Diversity In Changing Terrestrial Systems - www.predicts.org.uk). We make site-level summary data available alongside this article. The full database will be publicly available in 2015. Biodiversity continues to decline in the face of increasing anthropogenic pressures such as habitat destruction, exploitation, pollution and introduction of alien species. Existing global databases of species' threat status or population time series are dominated by charismatic species. The collation of datasets with broad taxonomic and biogeographic extents, and that support computation of a range of biodiversity indicators, is necessary to enable better understanding of historical declines and to project - and avert - future declines. We describe and assess a new database of more than 1.6 million samples from 78 countries representing over 28,000 species, collated from existing spatial comparisons of local-scale biodiversity exposed to different intensities and types of anthropogenic pressures, from terrestrial sites around the world. The database contains measurements taken in 208 (of 814) ecoregions, 13 (of 14) biomes, 25 (of 35) biodiversity hotspots and 16 (of 17) megadiverse countries. The database contains more than 1% of the total number of all species described, and more than 1% of the described species within many taxonomic groups - including flowering plants, gymnosperms, birds, mammals, reptiles, amphibians, beetles, lepidopterans and hymenopterans. The dataset, which is still being added to, is therefore already considerably larger and more representative than those used by previous quantitative models of biodiversity trends and responses. The database is being assembled as part of the PREDICTS project (Projecting Responses of Ecological Diversity In Changing Terrestrial Systems - ). We make site-level summary data available alongside this article. The full database will be publicly available in 2015.

International audience; The present study work was undertaken by SolData Acoustic and the Health regional Observatory (Southeastern France) within a research project aiming at the study of the links between road noise exposure and psychotropic drug consumption at a small-area level in the city of Marseilles. First, we built an average indicator of potential road noise exposure (PNEI) for the population living in each census block (the smallest sub municipal division with 1500 inhabitants on average). We calculated noise exposure using CadnaA software according to END method for noise mapping, and applied some improvements: we used observed mean speeds (instead of speed limitations), calculated mean noise level for each building (instead of maximum levels), and took floor levels into account. Second, we performed a sensitivity analysis to evaluate the impact of the variations of several parameters on model outputs. Sensitivity analysis shows that the most influent input parameters, at a global scale, are speed, then traffic flow. Propagation of uncertainties was performed on those parameters. Finally, uncertainties calculations on the PNEI will be taken into account in the analysis of the links between road noise exposure and psychotropic drug consumption.

- Abstract
Declining plant diversity and abundance have been widely reported in agroecosystems of North America and Europe. Intensive use of herbicides within cropfields and the associated drift in adjacent habitats are partly responsible for this change. The objectives of this work were to quantify the phenological stages of non-target plants in in-situ field situations during herbicide spray and to compare plant susceptibility at different phenological stages. Results demonstrated that a large number of non-target plants had reached reproductive stages during herbicide spray events in woodlots and hedgerows, both in Canada and Denmark where vegetation varies considerably. In addition, delays in flowering and reduced seed production occurred widely on plants sprayed at the seedling stage or at later reproductive periods, with plants sprayed at reproductive stages often exhibiting more sensitivity than those sprayed as seedlings. Ecological risk assessments need to include reproductive endpoints.

- Abstract
Fluorescence correlation spectroscopy (FCS) has been used to analyze the plasma membrane fluidity and heterogeneity of multidrug resistant cells. At the single cell level, the effects of different membrane agents present in the extra-cellular medium have been explored. Firstly, we reveal a modification of plasma membrane heterogeneities according to the addition of a fluidity modulator, benzyl alcohol. On the other hand, revertants such as verapamil and cyclosporin-A appear to act more specifically on the slow diffusion sites, such as lipids microdomains.

The PREDICTS project—Projecting Responses of Ecological Diversity In Changing Terrestrial Systems (www.predicts.org.uk)—has collated from published studies a large, reasonably representative database of comparable samples of biodiversity from multiple sites that differ in the nature or intensity of human impacts relating to land use. We have used this evidence base to develop global and regional statistical models of how local biodiversity responds to these measures. We describe and make freely available this 2016 release of the database, containing more than 3.2 million records sampled at over 26,000 locations and representing over 47,000 species. We outline how the database can help in answering a range of questions in ecology and conservation biology. To our knowledge, this is the largest and most geographically and taxonomically representative database of spatial comparisons of biodiversity that has been collated to date; it will be useful to researchers and international efforts wishing to model and understand the global status of biodiversity.

- Abstract
Biodiversity continues to decline in the face of increasing anthropogenic pressures such as habitat destruction, exploitation, pollution and introduction of alien species. Existing global databases of species' threat status or population time series are dominated by charismatic species. The collation of datasets with broad taxonomic and biogeographic extents, and that support computation of a range of biodiversity indicators, is necessary to enable better understanding of historical declines and to project - and avert - future declines. We describe and assess a new database of more than 1.6 million samples from 78 countries representing over 28,000 species, collated from existing spatial comparisons of local-scale biodiversity exposed to different intensities and types of anthropogenic pressures, from terrestrial sites around the world. The database contains measurements taken in 208 (of 814) ecoregions, 13 (of 14) biomes, 25 (of 35) biodiversity hotspots and 16 (of 17) megadiverse countries. The database contains more than 1% of the total number of all species described, and more than 1% of the described species within many taxonomic groups - including flowering plants, gymnosperms, birds, mammals, reptiles, amphibians, beetles, lepidopterans and hymenopterans. The dataset, which is still being added to, is therefore already considerably larger and more representative than those used by previous quantitative models of biodiversity trends and responses. The database is being assembled as part of the PREDICTS project (Projecting Responses of Ecological Diversity In Changing Terrestrial Systems - http://www.predicts.org.uk). We make site-level summary data available alongside this article. The full database will be publicly available in 2015.
