This thesis was performed at the Institut de Chimie Moléculaire de d’Université de Bourgogne (France) and in the Département de chimie de l’Université de Sherbrooke (Québec). The aim of this work was to design new BODIPY derivatives in order to obtain optimal properties for in vivo bimodal optical/PET or SPECT imaging. The modifications concern the hydrosolubilisation of these organic dyes, as well as the shifting of the absorption and emission bands toward the therapeutic window, where the light is less attenuated by biological tissues. Solutions allowing the bioconjugation of BODIPY on a bombesin derivative, a peptide which targets receptors surexpressed in tumor cells, were also investigated. A strategy for BODIPY radiofluorination, implying the introduction of a leaving group on the boron atom, is also described. Several BODIPY based multichromophoric species have also been prepared. The photophysical studies of these derivatives, reported in the second part of the thesis, have evidenced a photoinduced energy transfer between the chromophores. In these systems, BODIPY-pophyrin and BODIPY-corolle, the BODIPY moiety is alternatively donor or acceptor., Ce travail de thèse en cotutelle a été réalisé à l’Institut de Chimie Moléculaire de l’Université de Bourgogne (France) et au département de chimie de l’Université de Sherbrooke (Québec). Ces travaux sont consacrés à la conception de nouveaux dérivés du BODIPY en vue d'une application in vivo pour l'imagerie bimodale optique/TEP ou TEMP. Ces modifications concernent l'hydrosolubilisation des fluorophores, mais aussi le décalage des domaines d’absorption et d’émission vers la fenêtre thérapeutique, zone dans laquelle la lumière est moins atténuée par les tissus biologiques. Des solutions permettant la bioconjugaison des fluorophores sur un dérivé de la bombésine, un peptide permettant de cibler des récepteurs membranaires surexprimés dans certains types de cellules cancéreuses, sont également présentées. Une stratégie de radiofluoration du, BODIPY, impliquant l’introduction sur l’atome de bore d’un groupe partant, est décrite.. Différents dérivés de BODIPY ont également été utilisés pour la construction d’édifices multichromophoriques à transfert d’énergie. Ces travaux sont décrits dans la seconde partie de cette thèse. Ils comprennent notamment des études photophysiques permettant de mettre en évidence le transfert d’énergie photo-induit donneur-accepteur entre différents chromophores. Dans les différents systèmes élaborés, de type BODIPY-pophyrine et BODIPY-corolle, le BODIPY est alternativement donneur, ou accepteur d’énergie.
