[spa] ANTECENDENTES: La fibrilación auricular (FA) es la arritmia más común, afectando a un 1-2% de la población general. Aunque la FA se ha asociado con numerosas alteraciones electrofisiológicas, moleculares y estructurales, su tratamiento no es eficaz en todos los pacientes. Desde hace una década, diversos estudios de “genome wide association” han asociado varios single nucleotide polymorphisms (SNP)s en las regiones cromosómicas 4q25, 1q21 y 16q22, entre otras, a un mayor riesgo de presentar FA. No obstante, los mecanismos que están detrás de cada uno de ellos todavía son desconocidos. Por otro lado, varios estudios han demostrado que factores como tener enfermedad en la válvula mitral, la edad o la función del ventrículo izquierdo pueden afectar a la homeostasis intracelular del calcio y a la actividad eléctrica en miocitos auriculares humanos. Sin embargo, aunque hay estudios que demuestran que la epidemiología de la FA es diferente entre hombres y mujeres, sólo se ha analizado el efecto del sexo sobre la homeostasis intracelular del calcio en modelos animales. HIPÓTESIS: La asociación específica de los diferentes SNPs con mecanismos moleculares y consecuencias electrofisiológicas concretas podría ser utilizada como biomarcador para la prevención del desarrollo de FA, así como para el tratamiento personalizado de pacientes teniendo en cuenta el efecto del sexo. OBJETIVOS: • Profundizar en los mecanismos moleculares que alteran la homeostasis del calcio en los portadores de múltiples variantes de riesgo de la región 4q25. • Investigar el efecto que tienen las variantes de riesgo de la región 1q21 sobre la corriente SK3 y la actividad eléctrica, así como la influencia de la homeostasis del calcio sobre la corriente SK3 en miocitos auriculares humanos. • Analizar las posibles interacciones que se pueden dar entre las diferentes variantes de riesgo 4q25 y 1q21, y cómo afectarían estas a la función de los miocitos auriculares humanos. • Determinar la influencia del sexo en la homeostasis intracelular del calcio en pacientes con FA. MÉTODOS: Los experimentos se llevaron a cabo en miocitos auriculares humanos aislados. Se utilizó la técnica de patch-clamp (voltaje-clamp o current-clamp en función del protocolo) para medir la corriente de calcio (ICa) y sus características electrofisiológicas, así como las corrientes transitorias de entrada (ITI) o las despolarizaciones espontáneas de membrana. Para visualizar el calcio intracelular se utilizó microscopia confocal. La expresión de proteínas se determinó mediante la técnica de western blot y su distribución se analizó con marcaje inmunofluorescente RESULTADOS: El análisis del efecto de múltiples variantes de riesgo de 4q25 sobre la ICa y la actividad eléctrica en miocitos auriculares humanos, mostró que los pacientes portadores de la variante de riesgo en rs1448818 (la más próxima a PITX2) presentaban una reducción significativa de la ICa, mientras que los pacientes portadores de las variantes de riesgo en rs2200733, rs13143308 y rs10033464 (aproximadamente a 150 kb de PITX2) presentaban un incremento significativo en la frecuencia de ITI. Teniendo en cuenta estos hallazgos en conjunto, el efecto parece estar asociado a la localización en la que se encuentran las variantes de riesgo, sugiriéndose un efecto inhibidor en el extremo de 4q25 en la que se encuentra la variante de riesgo rs1448818, y un efecto estimulador en la región central donde se encuentran las variantes de riesgo rs2200733, rs13143308 y rs10033464. Por otro lado, el análisis del efecto de la alteración en la homeostasis del calcio y de las variantes de riesgo 1q21 sobre la corriente SK3 y la actividad eléctrica en miocitos auriculares humanos mostró que los pacientes con FA, los portadores de la variante de riesgo en rs1448818 de 4q25 y los portadores de la variante rs13376333 de 1q21 presentaban una reducción en la corriente SK sensible a apamina. Además, el bloqueo de los canales SK3 con apamina demostró que una disminución de la función de estos se traducía en un incremento de las despolarizaciones espontáneas de la membrana. Finalmente, aunque la FA se ha asociado con una disminución de la ICa y un aumento de la frecuencia de ITI, el análisis del efecto del sexo sobre la homeostasis intracelular del calcio en miocitos auriculares humanos demostró que la ICa sólo se vio reducida en hombres con FA; mientras que permitió observar una mayor incidencia de ITI y despolarizaciones espontáneas de la membrana únicamente en mujeres, exacerbándose en aquellas con FA. CONCLUSIONES: 1. Las variantes de riesgo de la región 4q25 asociadas a FA tienen un efecto diferencial sobre la regulación del calcio intracelular en función de su localización. • Los pacientes portadores de la variante de riesgo en rs1448818, que se encuentra en el extremo más próximo a PITX2, presentan una reducción de la ICa que favorecería un acortamiento del periodo refractario y la reentrada eléctrica. • Por otro lado, los pacientes portadores de las variantes de riesgo en rs2200733, rs13143308 o rs10033464, situadas a unas 150 kb de PITX2, presentan un incremento en la frecuencia de ITI que favorecería las despolarizaciones espontáneas de la membrana y actividad ectópica. Esto sugiere que la elección del tratamiento para prevenir o tratar la FA debería tener en cuenta las variantes de riesgo que presenta el paciente, ya que sería beneficioso restaurar la ICa en portadores de rs1448818 y reducir la liberación espontánea de calcio en portadores de rs13143308. 2. Las alteraciones en la homeostasis del calcio modifican la corriente SK sensible a apamina. • Así, la reducción de la ICa observada en pacientes con FA o en portadores de la variante de riesgo en rs1448818 reduce la corriente SK sensible a apamina. • Por otro lado, la variante de riesgo rs13376333 de la región1q21, ubicada en el intrón 1 del gen KCNN3, también reduce la corriente SK sensible a apamina. • Los efectos de las variantes de riesgo rs1448818 y rs13376333 no son complementarios. Estas reducciones de la corriente SK3 retrasarían la repolarización de las despolarizaciones espontáneas de membrana, facilitando la inducción de potenciales de acción espontáneos y arritmias inducidas por actividad ectópica. Por tanto, la restauración de la función de los canales SK3 podría ser una terapia farmacológica específica para la prevención o el tratamiento de los pacientes portadores de la variante rs13376333. 3. El sexo es un factor a tener en cuenta ya que tiene un efecto diferencial sobre la ICa y la frecuencia de ITI en miocitos auriculares de pacientes con FA. • La ICa sólo se ve reducida en hombres con FA. • La incidencia de ITI y despolarizaciones espontáneas de la membrana es mayor únicamente en mujeres. Estos resultados sugieren que las terapias dirigidas a reestablecer la ICa serían más efectivas en hombres con FA, mientras que las terapias dirigidas a reestablecer la actividad del RyR2 serían más efectivas en mujeres con FA., [eng] BACKGROUND: Atrial fibrillation (AF) is the most common arrhythmia, affecting 1-2% of the general population. Although AF has been associated with numerous electrophysiological, molecular and structural alterations, its treatment is not effective in all patients. For a decade, various genome wide association studies have associated several single nucleotide polymorphisms (SNP)s in the chromosomal regions 4q25, 1q21 and 16q22 with an increased risk of developing AF. Nevertheless, the mechanisms behind each of them are still unknown. On the other hand, several studies have shown that clinical factors such as mitral valve disease, age, or left ventricular function can affect intracellular calcium homeostasis and electrical activity in human atrial myocytes. However, although there are studies showing differences in the epidemiology of AF between men and women, the effect of sex on intracellular calcium homeostasis has only been analyzed in animal models. HYPOTHESIS: The specific association of the different SNPs with molecular mechanisms and specific electrophysiological responses could be used as biomarkers for the prevention of AF, as well as for personalized treatment of patients with AF, taking into account the influence of sex. AIMS OF THE THESIS: • Analyze the molecular mechanisms that alter calcium homeostasis in carriers of multiple risk variants of the 4q25 region. • Investigate the effects of 1q21 risk variants on SK3 current and electrical activity, as well as the influence of calcium homeostasis on SK3 current in human atrial myocytes. • Analyze the possible interactions that can occur between the different 4q25 and 1q21 risk variants, and their combined effect on electrophysiological function of human atrial myocytes. • Determine the influence of sex on intracellular calcium homeostasis in patients with AF. METHODS: Experiments were carried out on isolated human atrial myocytes. Patch-clamp technique (voltage-clamp or current-clamp depending on the protocol) was used to measure calcium current and its electrophysiological characteristics, as well as transient inward currents (ITI) or spontaneous membrane depolarizations. Confocal microscopy was used to visualize intracellular calcium. Protein expression was determined using the western blot technique and their distribution was analyzed with immunofluorescent marking. RESULTS: Analysis of the effect of all 4q25 risk variants on ICa and electrical activity in human atrial myocytes showed a significant reduction in ICa amplitude in patients carrying the risk variant in rs1448818 (the variant closest to PITX2), while patients carrying the risk variants in rs2200733, rs13143308 and rs10033464 (approximately located 150 kb upstream from PITX2) presented a significant increase in the ITI frequency. Taking these findings into account, the functional effect seems to be associated with the location of the risk variants, suggesting an inhibitory effect on ion channel activity in the region close to PITX2 where the risk variant rs1448818 is found, and a stimulatory effect in the area further away from PITX2 where the risk variants rs2200733, rs13143308 and rs10033464 are found. On the other hand, analysis of the effect of the alterations in calcium homeostasis and 1q21 risk variants on SK3 current and electrical activity in human atrial myocytes showed that patients with AF, carriers of the risk variant rs1448818 and carriers of the risk variant rs13376333 at 1q21 all exhibited a reduction in apamine-sensitive SK current. Furthermore, blocking the SK3 channels with apamin led to an increase in spontaneous membrane depolarizations. Finally, AF has generally been associated with a decrease in ICa and an increase in ITI frequency, but analysis of the effect of sex on intracellular calcium homeostasis in human atrial myocytes showed that ICa is only reduced in men with AF; while higher frequency of ITI and spontaneous membrane depolarizations is only observed in women and exacerbated in those with AF. CONCLUSIONS: 1. Risk variants in the 4q25 region associated with risk of AF have a differential effect on intracellular calcium regulation depending on their location. • Patients with risk variant at rs1448818, which is closest to PITX2, present a reduction in ICa that would favor a shortening of refractory period and electrical reentry. • On the other hand, patients with risk variants at rs2200733, rs13143308 or rs10033464, located approximately 150 kb upstream from PITX2, present an increase in the ITI frequency that favors spontaneous membrane depolarizations and ectopic activity. This demonstrates that the choice of treatment to prevent or treat AF should take into account the risk variants carried by the patient, suggesting that it would be beneficial to restore ICa in carriers of rs1448818 and to reduce spontaneous calcium release in carriers of rs13143308. 2. Altered calcium homeostasis and 1q21 variants modify the apamine-sensitive current. • The reduction in ICa observed in patients with AF or in carriers of the risk variant in rs1448818 reduces the apamine-sensitive SK current. • The risk variant rs13376333 in the 1q21 region, located in intron 1 of the KCNN3 gene, also reduce the apamine-sensitive current. • The effects of the risk variants rs1448818 and rs13376333 are not complementary. These reductions in SK3 current would delay the repolarization of spontaneous membrane depolarizations, facilitating the induction of spontaneous action potentials and arrhythmias induced by ectopic activity. Therefore, restoration of SK3 channel function might represent a specific therapy for the prevention or treatment of AF in carriers of the rs13376333 risk variant. 3. Sex is also a factor to take into account since it has a differential effect on ICa amplitude and ITI frequency in atrial myocytes of patients with AF. • ICa is only reduced in men with AF. • The incidence of ITI and spontaneous membrane depolarizations is higher in women only and exacerbated in those with AF. These results suggest that therapies aimed at reestablishing ICa would be more effective in men with AF, while therapies aimed at restoring RyR2 activity would be more effective in women with AF.

[cat] Les proteïnes de la família de les CaM quinases (CAMK en anglès), l’activitat de les quals és dependent del complex calci-calmodulina, estan implicada en funcions cel·lulars com la regulació de l’expressió gènica, la contracció, l’apoptosi i en la neurotransmissió. A S. pombe, s’ha descrit una proteïna homòloga a CaMKI de mamífers, Cmk1, que és fosforilada per una quinasa upstream, Ckk2, que és homòloga a la CaMKK de mamífers. Es va observar que l’expressió de Ckk2 en condicions de deprivació nutricional com absència de nitrogen o glucosa s’incrementava. El tractament amb drogues com latrunculina B, que causa alteracions dels microfilaments d’actina, compromet la viabilitat de la soca sense ckk2 (Δckk2). Es van identificar possibles substrats en un assaig de fosfo-proteòmica com Cgs1, subunitat reguladora del complex PKA a S. pombe; Int6, homòloga a la subunitat E del complex eIF3, implicat en la traducció; i Gad8, substrat del TORC2 (TOR complex 2). En aquest treball, s’ha identificat que la funció de Ckk2 és diferent segons el tipus d’estrès, en la deprivació de nitrogen no és important per mantenir la viabilitat però si participa en la re-entrada al cicle cel·lular des de la fase G0. En el cas de la deprivació nutricional de glucosa, la soca Δckk2 mostra una reducció de viabilitat en créixer en condicions de deprivació de glucosa, indicant que podria estar participant en la supervivència en aquestes condicions o que podria ser important per activar mecanismes de restricció calòrica. S’ha observat també que l’increment de l’expressió causat per la deprivació nutricional, retorna a nivells basals en reintroduir els nutrients en el medi, indicant que estava restringit a les condicions de deprivació. L’increment de l’expressió en condicions de deprivació es devia a que en aquestes condicions TORC1 es troba deprivat, i s’ha observat que la inhibició de TORC1 amb rapamicina causa un increment en els nivells de Ckk2. L’augment de l’expressió de Ckk2 en deprivació de nitrogen, es produeix gràcies a la acció de Gaf1, un factor de transcripció que es torna actiu en aquestes mateixes condicions, ja que en absència de Gaf1, l’expressió de Ckk2 no s’indueix amb la mateixa intensitat. No es va detectar la fosforilació dels substrats Cgs1, Int6 i Gad8, però si que es van identificar fosforilacions en proteïnes que co-precipiten amb elles. En analitzar les interaccions genètiques entre Ckk2 i aquests possibles substrats, s’ha observat que el doble mutant Δckk2 Δpka1 presenta un fenotip de letalitat sintètica en condicions de deprivació de glucosa, el que ens indica que la funció de les dos proteïnes és necessària per sobreviure en aquestes condicions i potser convergeixen en una tercera que encara desconeixem. També s’ha observat que l’activitat proteosòmica de Int6 podria estar controlant els nivells de Ckk2 en condicions normals. L’anàlisi de la viabilitat en resposta a latrunculina va demostrar que la funció de Ckk2 no està relacionada amb la de Cmk1, i que podria presentar altres substrats. La composició d’actina en cèl·lules Δckk2 es veu alterada en deprivació de nitrogen, formant nòduls corticals més petits i dispersos. La formació d’aquests nòduls es veu alterada en mutants endocítics i la latrunculina B és capaç d’inhibir l’endocitosi. L’addició de sorbitol, que facilita l’endocitosi, és capaç de revertir la sensibilitat de la soca Δckk2 a la latrunculina B, indicant que Ckk2 podria estar participant en l’endocitosi., [eng] TCaMK are proteins whose activity depends on the calcium-calmodulin complex, who are implicated in different cellular functions like contraction, apoptosis, neurotransmission and gene expression. In S. pombe, it has been described a homologue of the mammal CaMKI, Cmk1, which is phosphorylated by an upstream kinase Ckk2 (which is a homologue for CaMKK). It has been observed that under nutrient deprivation caused by depletion of nitrogen or glucose, Ckk2 expression is increased. Treatment with cytoskeleton altering drugs like latrunculin B (LatB) compromises viability of strains without Ckk2 (Δckk2). Possible substrates of Ckk2 have been identified in a phosphoproteomic assay, like Cgs1, Int6 and Gad8. In this thesis, it has been determined that Ckk2 function depends on the stress; for example, Ckk2 is not important to maintain viability in G0 phase, but it is important in the re-entry of cell cycle from G0. Ckk2 could be participating in the response to glucose starvation, as Δckk2 strains show sensitivity in low glucose concentration. Changes in the expression of Ckk2 are restricted to starvation conditions, as the protein level rapidly decreases when nutrients are re-introduced in the media. Ckk2 increase under nutrient starvation is caused by TORC1 inhibition, and is also observed when treating cells with rapamycin, a TORC1 inhibitor. It has been identified that Ckk2 expression is caused by Gaf1, a transcription factor that is active during the first hours of nitrogen deprivation. No phosphorylations were caused by Ckk2 in vitro in Cgs1, Int6 or Gad8, but some were observed in co-precipitated proteins. Ckk2 and Pka1 could be participating together in the response to glucose starvation, as the double deletion show a phenotype of synthetic lethality under these conditions. Steady state levels of Ckk2 might be controlled by proteosomic activity of Int6. Genetic interaction analysis of deletions of CAMK pathway in the response to LatB revealed that targets other than Cmk1 might be involved in the response. Lack of Ckk2 causes alterations in the formation of actin cortical patches under nitrogen starvation and sorbitol treatment is able to rescue Δckk2 mutants from sensitivity to LatB, indicating a possible role for Ckk2 in endocytosis.

A pesar de la importancia del Ca2+ como segundo mensajero intracelular, aun existen discrepancias en la literatura sobre los niveles de Ca2+ que se alcanzan en el interior de la mitocondria tras un estímulo fisiológico. En esta tesis se ha realizado una comparativa detallada de los diferentes métodos de medida del Ca2+mitocondrial y además se han desarrollado nuevos métodos de medida para dicho Ca2+. Gracias a estas nuevas técnicas hemos podido realizar un estudio exhaustivo de los flujos de entrada y salida del Ca2+, tanto en células HeLa intactas como permeabilizadas. Hemos esclarecido algunos puntos tales como la dependencia de Ca2+ de la apertura del poro de transición de permeabilidad, la relación intramitocondrial entre Ca2+-libre/Ca2+ ligado, el efecto del fosfato en los niveles de Ca2+ mitocondriales o la dependencia d la salida del Ca2+ mitocondrial con el Na+ citosólico., Departamento de Bioquímica y Biología Molecular y Fisiología, Instituto de Biología y Genética Molecular (IBGM)

La excesiva proliferación celular contribuye al desarrollo de enfermedades como la aterosclerosis y la restenosis. Los anti-inflamatorios no esteroideos (AINEs) inhiben esa excesiva proliferación independientemente de su actividad anti-inflamatoria. La entrada de Ca2+ operada por depósitos (SOCE) es importante en la proliferación de las células de músculo liso vascular (VSMCs), por lo que estudiamos los canales de Ca2+ de estas células y el efecto antiproliferativo de ciertos AINEs. Utilizando las técnicas de medida de Ca2+ citosólico y mitocondrial, registros electrofisiológicos, qRT-PCR y western-blotting realizamos nuestro estudio en células A10 y un cultivo primario de células de músculo liso de arteria coronaria humana. Los resultados obtenidos muestran que SOCE, y las mitocondrias, que modulan esta vía, son esenciales en la proliferación de las VSMCs. Además, los AINEs actúan directamente sobre las mitocondrias y consecuentemente sobre SOCE, explicando de esta forma su efecto antiproliferativo., Departamento de Bioquímica y Biología Molecular y Fisiología

Producción Científica, Intracellular Ca2+ plays a pivotal role in the control of a large series of cell functions in all types of cells, from neurotransmitter release and muscle contraction to gene expression, cell proliferation and cell death. Ca2+ is transported through specific channels and transporters in the plasma membrane and subcellular organelles such as the endoplasmic reticulum and mitochondria. Therefore, dysregulation of intracellular Ca2+ homeostasis may lead to cell dysfunction and disease. Accordingly, chemical compounds from natural origin and/or synthesis targeting directly or indirectly these channels and proteins may be of interest for the treatment of cell dysfunction and disease. In this review, we show an overview of a group of marine drugs that, from the structural point of view, contain one or various heterocyclic units in their core structure, and from the biological side, they have a direct influence on the transport of calcium in the cell. The marine compounds covered in this review are divided into three groups, which correspond with their direct biological activity, such as compounds with a direct influence in the calcium channel, compounds with a direct effect on the cytoskeleton and drugs with an effect on cancer cell proliferation. For each target, we describe its bioactive properties and synthetic approaches. Producción Científica, We have used an aequorin chimera targeted to the membrane of the secretory granules to monitor the free [Ca2+] inside them in neurosecretory PC12 cells. More than 95% of the probe was located in a compartment with an homogeneous [Ca2+] around 40 M. Cell stimulation with either ATP, caffeine or high-K+ depolarization increased cytosolic [Ca2+] and decreased secretory granule [Ca2+] ([Ca2+]SG). Inositol-(1,4,5)- trisphosphate, cyclic ADP ribose and nicotinic acid adenine dinucleotide phosphate were all ineffective to release Ca2+ from the granules. Changes in cytosolic [Na+] (0–140 mM) or [Ca2+] (0–10 M) did not modify either ([Ca2+]SG). Instead, [Ca2+]SG was highly sensitive to changes in the pH gradient between the cytosol and the granules. Both carbonyl cyanide 4-(trifluoromethoxy)phenylhydrazone (FCCP) and nigericin, as well as cytosolic acidification, reversibly decreased [Ca2+]SG, while cytosolic alcalinization reversibly increased [Ca2+]SG. These results are consistent with the operation of a H+/Ca2+ antiporter in the vesicular membrane. This antiporter could also mediate the effects of ATP, caffeine and high-K+ on [Ca2+]SG, because all of them induced a transient cytosolic acidification. The FCCP-induced decrease in [Ca2+]SG was reversible in 10–15 min even in the absence of cytosolic Ca2+ or ATP, suggesting that most of the calcium content of the vesicles is bound to a slowly exchanging Ca2+ buffer. This large store buffers [Ca2+]SG changes in the long-term but allows highly dynamic free [Ca2+]SG changes to occur in seconds or minutes.

We use here a new very low-Ca2+-affinity targeted aequorin to measure the [Ca2+] in the endoplasmic reticulum ([Ca2+]ER). The new aequorin chimera has the right Ca2+-affinity to make long-lasting measurements of [Ca2+]ER in the millimolar range. Moreover, previous Ca2+-depletion of the ER is no longer required. The steady-state [Ca2+]ER obtained is 1-2mM, higher than previously reported. In addition, we find evidence that there is significant heterogeneity in [Ca2+]ER among different regions of the ER. About half of the ER had a [Ca2+]ER of 1mM or below, and the rest had [Ca2+]ER values above 1mM and in some parts even above 2mM. About 5% of the ER was also found to have high [Ca2+]ER levels but to be thapsigargin-insensitive and inositol trisphosphate insensitive. The rate of refilling with Ca2+ of the ER was almost linearly dependent on the extracellular [Ca2+] between 0.1 and 3mM, and was only partially affected by mitochondrial membrane depolarization. Instead, it was significantly reduced by loading cells with chelators, and the fast chelator BAPTA was much more effective than the slow chelator EGTA. Cellular Ca 2+signaling results from a complex interplay among a variety of Ca 2+ fluxes going across the plasma membrane and across the membranes of several organelles, together with the buffering effect of large numbers of Ca 2+-binding sites distributed along the cell architecture. Endoplasmic and sarcoplasmic reticulum, mitochondria and even nucleus have all been involved in cellular Ca 2+ signaling, and the mechanisms for Ca 2+ uptake and release from these organelles are well known. In neuroendocrine cells, the secretory granules also constitute a very important Ca 2+-storing organelle, and the possible role of the stored Ca 2+ as a trigger for secretion has attracted considerable attention. However, this possibility is frequently overlooked, and the main reason for that is that there is still considerable uncertainty on the main questions related with granular Ca 2+ dynamics, e.g., the free granular [Ca 2+], the physical state of the stored Ca 2+ or the mechanisms for Ca 2+ accumulation and release from the granules. 9 páginas, 9 figuras.-- El pdf del artículo es la versión manuscrita de autor., Secretory vesicles have low pH and have been classically identified as those labelled by a series of acidic fluorescent dyes such as acridine orange or neutral red, which accumulate into the vesicles according to the pH gradient. More recently, several fusion proteins containing enhanced green fluorescent protein (EGFP) and targeted to the secretory vesicles have been engineered. Both targeted fluorescent proteins and acidic dyes have been used, separately or combined, to monitor the dynamics of secretory vesicle movements and their fusion with the plasma membrane. We have now investigated in detail the degree of colocalization of both types of probes using several fusion proteins targeted to the vesicles (synaptobrevin2-EGFP, Cromogranin A-EGFP and neuropeptide Y-EGFP) and several acidic dyes (acridine orange, neutral red and lysotracker red) in chromaffin cells, PC12 cells and GH3 cells. We find that all the acidic dyes labelled the same population of vesicles. However, that population was largely different from the one labelled by the targeted proteins, with very little colocalization among them, in all the cell types studied. Our data show that the vesicles containing the proteins more characteristic of the secretory vesicles are not labelled by the acidic dyes, and vice versa. Peptide glycyl-L-phenylalanine 2-naphthylamide (GPN) produced a rapid and selective disruption of the vesicles labelled by acidic dyes, suggesting that they could be mainly lysosomes. Therefore, these labelling techniques distinguish two clearly different sets of acidic vesicles in neuroendocrine cells. We show here that activation of mitochondrial Ca2+ uptake by 4,4′,4″-(4-propyl-[1H]-pyrazole-1,3,5-triyl)trisphenol (PPT) or kaempferol stimulates histamine-induced Ca2+ release from the endoplasmic reticulum (ER) and that this effect is enhanced if the mitochondrial Na+–Ca2+ exchanger is simultaneously inhibited with CGP37157. This suggests that both Ca2+ uptake and release from mitochondria control the ability of local Ca2+ microdomains to produce feedback inhibition of inositol 1,4,5-trisphosphate receptors (InsP3Rs). In addition, the ability of mitochondria to control Ca2+ release from the ER allows them to modulate cytosolic Ca2+ oscillations. In histamine stimulated HeLa cells and human fibroblasts, both PPT and kaempferol initially stimulated and later inhibited oscillations, although kaempferol usually induced a more prolonged period of stimulation. Both compounds were also able to induce the generation of Ca2+ oscillations in previously silent fibroblasts. [spa] FUNDAMENTO: Los movimientos del Ca2+ a través de la membrana celular están mediados en parte por la bomba de Ca2+ por lo que es posible que una alteración genética o adquirida en esta bomba puede explicar el aumento de la concentración intracelular de calcio presente en los pacientes afectos de HTA esencial. METODOS: Se ha estudiado la cinética de la bomba de CA2+ en los hematíes de 49 pacientes hipertensos esenciales y 27 sujetos control. Se ha utilizado el Sr2+ como análogo del Ca2+ por la dificultad de medir en condiciones estables el flujo de CA2+ por su baja concentración libre y su rapidez en el flujo de salida. RESULTADOS: El grupo de pacientes hipertensos mostró unos valores medios (DE) de la velocidad máxima de flujo de Sr2+ (Vmax) significativamente superiores a los observados en los individuos del grupo control, 6,6 (2,3) frente a 5,2(1,6)mmol/L.cels/h., p=0,0067. Los valores medios de la constante de disociación aparente para el Ca2+ intracelular total (Kca) fueron superiores en el grupo de hipertensos respecto al grupo control 80,36 (53,46) frente a 55,25 (15,13) umol/L.cels., p=0,066. La Vmax mostró una relación directa significativa con la edad (r=0,342, p=0,016) y con las cifras de creatinina sérica (r=0,446, p=0,001). La Kca únicamente mostró una relación directa signficativa con las cifras de creatinina sérica (r=0,402, p=0,004). Se pudieron diferenciar dos grupos de pacientes hipertensos según presentaran una Kca normal (33 pacientes, 67,3%) o alterada (16 pacientes, 32,6%). La edad de los pacientes, 50,8 (13,5) frente a 43 (10,2) años, p=0,028, la creatinina sérica, 1,13 (0,17) frente a 0,95 (0,17) mg/dl p=0,0019 y la uricemia, 7,27 (3,32) frente a 6,14 (1,49) mg/dl, p=0,0482, fueron superiores en los hipertensos con una Kca más elevada. Finalmente, los pacientes con Kca elevada presentaron una Vmax mayor que los pacientes con una Kca normal, 9,136 (2,02) frente a 5,38 (12,81) mmol/L.cels.
