4. Screening and Fermentation Performance of Highly Efficient Starter Cultures for Semi-Dry Douchi

Abstract

To select high-efficiency strains suitable for semi-dry black douchi fermentation, this study compared and analyzed the koji-making effects of six Aspergillus oryzae strains and one Lichtheimia corymbifera strain with large differences in enzyme-producing capacity. The results showed that A. oryzae HN3.042, DM1 and DM2 had a strong ability to metabolize amylase, and the fermented douchi had a lower L* value. A. oryzae 24M-1, 30M-1, and PNM003 had a strong ability to produce protease, aminopeptidase, and carboxypeptidase, resulting in greater utilization of protein substrates and higher contents of total acid and amino nitrogen in the fermented douchi. In terms of volatile flavor substances, the douchi fermented with stains HN3.042, DM1, and DM2 had a relatively higher content of 1-octen-3-ol, isovaleric acid, and 4-methylpentanoic acid. The douchi fermented with strains 24M-1, 30M-1, and PNM003 had a relatively higher content of trans-2-nonenal, 3-octanone, and isophorone, resulting in richer and better flavor quality. L. corymbifera QM3 had lower enzymatic activity when compared with A. oryzae, resulting in poorer physicochemical indicators, lower contents of free amino acids and volatile flavor substances and poorer quality in douchi. The study suggests that the quality of douchi is related to the enzyme-producing capacity of the koji mold. The predominance of amylase in the koji enzyme system is conducive to the formation of color and acidic flavor substances in douchi, while with the predominance of proteolytic enzymes is conducive to the formation of amino nitrogen and good flavor quality in douchi.

Abstract

Pentatrichomonas hominis, a flagellated parasitic protozoan, predominantly infects the mammalian digestive tract, often causing symptoms such as abdominal pain and diarrhea. However, studies investigating its pathogenicity are limited, and the mechanisms underlying P. hominis-induced diarrhea remain unclear. Establishing an in vitro cell model for P. hominis infection is imperative. This study investigated the interaction between P. hominis and IPEC-J2 cells and its impact on parasite growth, adhesion, morphology, and cell viability. Co-cultivation of P. hominis with IPEC-J2 cells resulted in exponential growth of the parasite, with peak densities reaching approximately 4.8 × 105 cells/mL and 1.2 × 106 cells/mL at 48 h for initial inoculation concentrations of 104 cells/mL and 105 cells/mL, respectively. The adhesion rate of P. hominis to IPEC-J2 cells reached a maximum of 93.82% and 86.57% at 24 h for initial inoculation concentrations of 104 cells/mL and 105 cells/mL, respectively. Morphological changes in IPEC-J2 cells co-cultivated with P. hominis were observed, manifesting as elongated and irregular shapes. The viability of IPEC-J2 cells exhibited a decreasing trend with increasing P. hominis concentration and co-cultivation time. Additionally, the mRNA expression levels of IL-6, IL-8, and TNF-α were upregulated, whereas those of CAT and CuZn-SOD were downregulated. These findings provide quantitative evidence that P. hominis can promote its growth by adhering to IPEC-J2 cells, inducing morphological changes, reducing cell viability, and triggering inflammatory responses. Further in vivo studies are warranted to confirm these results and enhance our understanding of P. hominis infection.

Abstract

In this thesis, heat powered ejector cooling systems was investigated in two ways: to store the cold energy with energy storage system and to utilize low grade energy to provide both electricity and cooling effect. A basic ejector prototype was constructed and tested in the laboratory. Water was selected as the working fluid due to its suitable physical properties, environmental friendly and economically available features. The computer simulations based on a 1-0 ejector model was carried out to investigate the effects of various working conditions on the ejector performance. The coefficients of performance from experimental results were above 0.25 for generator temperature of lI5°C-130 °C, showing good agreements with theoretical analysis. Experimental investigations on the operating characteristics of PCM cold storage system integrated with ejector cooling system were conducted. The experimental results demonstrated that the PCM cold storage combined with ejector cooling system was practically applicable. The effectiveness-NTU method was applied for characterizing the tube-in-container PCM storage system. The correlation of effectiveness as the function of mass flow rate was derived from experimental data, and was used as a design parameter for the PCM cold storage system. In order to explore the possibility of providing cooling effect and electricity simultaneously, various configurations of combined power and ejector cooling system were studied experimentally and theoretically. The thermal performance of the combined system in the range of 0.15-0.25 and the turbine output between 1200W -1400W were obtained under various heat source temperatures, turbine expansion ratios and condenser temperatures. Such combined system was further simulated with solar energy as driving force under Shanghai climates, achieving a predicted maximum thermal efficiency of 0.2. By using the methods of Life Saving Analysis, the optimized solar collector area was 30m2 and 90m2 respectively for the system without and with power generation. The environmental impacts and the carbon reductions of these two systems were discussed.

32. A novel Energy-Flow based Ensemble Calibration model for rapid and accurate energy-economic performance prediction of deep energy retrofit in single-family houses

Abstract

In Europe, 68% of residential building stocks are single-family houses, of which roughly 54% are constructed before the 1970 s with poor energy efficiency. However, few studies have addressed the issue of ‘Rapid and accurate energy-economic performance prediction of deep energy retrofit in single-family houses’ over the past decades. Recently, this issue has been discussed in limited research via Black- or White-box based building energy models to predict post-retrofit energy-economic performance. Applying the Black-box models, low accuracy occurs due to inadequate data, neglect of coupling effects in deep energy retrofit combinations and lack of complex heat transfer phenomena in single-family houses. Conversely, the White-box models with time-consuming and numerous dynamic simulations are very hard to be implemented at a building cluster or urban scale. This paper proposes a novel Energy-Flow based Ensemble Calibration model to tackle this issue, with a maximum discrepancy of 6% compared to other building performance simulation results and 0.72 s computing time on a single combination. This model consists of three layers 1) input neurons, 2) complex heat and electrical transfer models, and 3) output neurons. By replacing the layer of hidden neurons with complex heat and electrical transfer models in conventional Black-box models, the coupling effects in deep energy retrofit combinations and complex heat transfer phenomena are thoroughly considered in this research. Besides, the computational efforts are substantially reduced with only a few building performance simulations required for baseline building and identified individual retrofit measures. A semi-detached 1960 s Italian single-family house is selected as a case study to validate the calculation results from this model against real energy consumption of baseline and post-retrofit scenario, with a discrepancy of −0.66% and 1.03%, respectively.

Abstract

The global policy debate on just transitions is concerned with how to achieve a socially just and acceptable transition toward a climate-neutral and climate-resilient global economy. At the core of this debate is the assumption that efforts to combat environmental threats will not succeed unless combined with measures to reduce poverty and inequality. Our research explores the potential of carbon fiscal reforms, combining a carbon tax of levels deemed appropriate to achieve climate targets and the transfer of the revenues raised to vulnerable households. The current energy and cost-of-living crisis shows the importance of protecting the poorest and most vulnerable households from price increases. It also shows the difficulty of achieving short- and long-term policy priorities. Despite the current spikes in energy prices, carbon fiscal reforms can achieve both social and environmental goals through simultaneously decreasing emissions and reducing poverty and inequality. They should act as an effective enabler of just transitions. Carbon fiscal reform can avoid some environmental impacts by incentivising reductions in emissions. Carbon pricing has been increasingly advocated and is now at the centre of policy debates, including the UNFCCC Conference of the Parties (COP) and the recent German presidency of the world's leading industrial nations (G7). But carbon fiscal reforms can also be used to raise revenue from carbon pricing instruments to offset the negative effects of higher prices on poorer households as well as further reaching distributional targets and poverty alleviation. Climate targets are negotiated every year, including at COP, hence it is critical to re-evaluate and improve estimates of the distributional impacts of climate policies such as carbon pricing. Public acceptability of climate policies is key to their implementation, but it depends to a large extent on the perceived fairness of such policies. Recycling revenues from carbon taxes directly back to vulnerable households is likely to gain the approval of a large number of people, especially in low-income countries where the high proportion of the population involved in the informal economy means that lowering income tax does not benefit the poorest and most vulnerable sections of society. But the targeting of these direct transfers needs careful consideration. Here, we assess the impact on poverty and inequality of a global carbon tax and national redistribution of revenues to vulnerable households. We look at different options for such redistribution, including a lump sum payment, the use of current social assistance programmes, and an expansion of social assistance following COVID-19. We find that a carbon tax of US$50/tCO2 without revenue redistribution could increase global extreme poverty, but the redistribution of revenue from such a carbon tax could substantially reduce poverty by between 16% and 27% (110 to 190 million people), and reduce inequality (the average Gini coefficient would decline by between 4% and 8%), depending on the scenario. This shows that the way in which revenue from a carbon tax is redistributed greatly affects its impact, underlining the importance of policy design and targeting mechanisms. The recycling of revenues should also take into account the specific political economy of a country and consider international transfers. These findings provide policy makers with a strong basis for informing discussions, starting off with those at COP27, in which ambitious climate targets and just transition should both remain central goals in the context of the ongoing international energy crisis.

Abstract

Objective: This study was designed to analyze the clinical effectiveness of Deanxit (DEA) for treating chronic subjective dizziness (CSD). Methods: A total of 110 CSD patients (all from a community survey) admitted to our hospital from August 2018 to August 2020 were recruited as the study cohort. Among them, 60 who underwent DEA treatment were placed in the research group (RG) and 50 who underwent basic treatment were placed in the control group (CG). The two groups’ efficacy, their dizziness disability rating scale (DHI) scores, the improvement in their clinical symptoms (duration of dizziness, frequency of dizziness attacks), their anxiety and depression (i.e., their Hamilton Anxiety Scale (HAMA) scores) and their Hamilton Depression Scale (HAMD)) scores, their sleep quality and their quality of life (short form 36 (SF-36) health survey scores) were observed and compared. Results: Compared with the CG, the DHI scores, the dizziness durations, the number of dizziness attacks and the HAMA and HAMD scores in the RG were markedly lower than they were in the CG, and the sleep quality levels and SF-36 scores were higher than they were in the CG. Conclusion: DEA treatment helps to improve the dizziness, anxiety, and quality of life of CSD patients.

40. Analysing unbalanced ageing in EV battery Packs using the Low-Cost Lumped Single Particle Model (LSPM): the impact of temperature gradients among parallel-connected cells

Abstract

Lithium-ion battery cells are widely used in electric vehicles (EVs) due to their high energy density and long lifespan. However, these cells are sensitive to temperature and can age at different rates depending on the temperature they are subjected to. While EV battery packs are equipped with a battery thermal management system (BTMS) to regulate the temperature of the pack, it cannot completely eliminate temperature gradient within the pack due to the limited cooling capabilities. As a result, a maximum temperature gradient of 5°C is allowed in BTMS designs for EVs, which leads to uneven ageing speeds within the pack. Due to practical limitations of battery management system (BMS), it is difficult to directly measure the currents flowing through each parallel-connected cell, making it challenging to actively balance the cells. To investigate the pack-level current distribution and ageing trends caused by temperature gradient, this paper adopts the low computational cost Lumped Single Particle Model (LSPM) coupled with Arrhenius dependency to investigate the influence of temperature on current distribution as well as the resulting unbalanced ageing rates among parallel-connected cells in a 4s6p Panasonic NCR18650PF battery pack.
