Abstract Background Non-invasive and high-throughput monitoring of drought in plants from its initiation to visible symptoms is essential to quest drought tolerant varieties. Among the existing methods, chlorophyll a fluorescence (ChlF) imaging has the potential to probe systematic changes in photosynthetic reactions; however, prerequisite of dark-adaptation limits its use for high-throughput screening. Results To improve the throughput monitoring of plants, we have exploited their light-adaptive strategy, and investigated possibilities of measuring ChlF transients under low ambient irradiance. We found that the ChlF transients and associated parameters of two contrasting Arabidopsis thaliana accessions, Rsch and Co, give almost similar information, when measured either after ~20 min dark-adaptation or in the presence of half of the adaptive growth-irradiance. The fluorescence parameters, effective quantum yield of PSII photochemistry (ΦPSII) and fluorescence decrease ratio (R FD) resulting from this approach enabled us to differentiate accessions that is often not possible by well-established dark-adapted fluorescence parameter maximum quantum efficiency of PSII photochemistry (F V/F M). Further, we screened ChlF transients in rosettes of well-watered and drought-stressed six A. thaliana accessions, under half of the adaptive growth-irradiance, without any prior dark-adaptation. Relative water content (RWC) in leaves was also assayed and compared to the ChlF parameters. As expected, the RWC was significantly different in drought-stressed from that in well-watered plants in all the six investigated accessions on day-10 of induced drought; the maximum reduction in the RWC was obtained for Rsch (16%), whereas the minimum reduction was for Co (~7%). Drought induced changes were reflected in several features of ChlF transients; combinatorial images obtained from pattern recognition algorithms, trained on pixels of image sequence, improved the contrast among drought-stressed accessions, and the derived images were well-correlated with their RWC. Conclusions We demonstrate here that ChlF transients and associated parameters measured even in the presence of low ambient irradiance preserved its features comparable to that of measured after dark-adaptation and discriminated the accessions having differential geographical origin; further, in combination with combinatorial image analysis tools, these data may be readily employed for early sensing and mapping effects of drought on plant’s physiology via easy and fully non-invasive means.

Discharge of livestock effluents and poor management of livestock manures are important reasons for the eutrophication of water bodies. Dissolved organic matter (DOM) originated from livestock manure is ubiquitous in water, but the effects of such DOM and the organic substances on the growth and cyanotoxins synthesis of bloom-forming cyanobacteria are still unknown. In order to elucidate these issues, the growth, microcystins synthesis, and cell viability of Microcystis aeruginosa exposed to cattle manure originated DOM (CM-DOM) were investigated in this study. The results indicated that the growth of M . aeruginosa FACHB905 was slightly improved by CM-DOM at a concentration of 90 mg l −1 (TOC). CM-DOM slightly decreased the electron transfer efficiency of photosystem II (PS II) of M . aeruginosa FACHB905. The synthesis and secretion of microcystins were promoted by CM-DOM. The cell viability (TTC test) increased in response to CM-DOM addition. Results presented in this study suggested that CM-DOM may increase the ecological and public health risks of M . aeruginosa when poor management of livestock manure is anticipated. Impaired carbon precursor supply through cpPDC is detrimental for TAG biosynthesis in Chlamydomonas reinhardtii under conditions of photoautotrophy and nitrogen starvation.The chloroplast pyruvate dehydrogenase complex (cpPDC) catalyses the oxidative decarboxylation of pyruvate forming acetyl-CoA, an immediate primer for the initial reactions of de novo fatty acid (FA) synthesis. Little is known about the source of acetyl-CoA in the chloroplasts of photosynthetic microalgae, which are capable of producing high amounts of the storage lipid triacylglycerol (TAG) under conditions of nutrient stresses. We generated Chlamydomonas reinhardtii CC-1618 mutants with decreased expression of the PDC2_E1α gene, encoding the putative chloroplast pyruvate dehydrogenase subunit E1α, using artificial microRNA. A comparative study on the effects of PDC2_E1α silencing on FAs and TAG production in C. reinhardtii, grown photoautotrophically and mixotrophically, with and without a nitrogen source in the nutrient medium, was carried out. Reduced expression of PDC2 _E1α led to a severely hampered photoautotrophic growth phenotype with drastic impairment in TAG accumulation under nitrogen deprivation. In the presence of acetate, downregulation of PDC2_E1α exerted little to no effect on TAG production and photosynthetic activity. In contrast, under photoautotrophic conditions, especially in the absence of a nitrogen source, a dramatic decline in photosynthetic oxygen evolution and photosystem II quantum yield against a background of the apparent over-reduction of the photosynthetic electron chain was recorded. Our results suggest an essential role of cpPDC in the supply of carbon precursors for de novo FA synthesis in microalgae under conditions of photoautotrophy. A shortage of this supply is detrimental to the nitrogen-starvation-induced synthesis of storage TAG, an important carbon and energy sink in stressed Chlamydomonas cells, thereby impairing the acclimation ability of the microalga. Abstract: The higher plant Wolffia arrhiza is regarded to be well suited concerning the provision of photosynthetic products in the cycle of matter of a Controlled Ecological Life Support System (CELSS) to be established in the context of extraterrestrial, human-based colonization and long-term space flight. Since UV radiation is one major extraterrestrial environmental stress for growth of any plant, effects of UV-B radiation on W. arrhiza were assessed in the present study. We found that UV-B radiation significantly inhibited photosynthetic CO2 assimilation activity, and the contents of chlorophyll a, chlorophyll b (Chl a, Chl b) and carotenoids considerably decreased when plants were exposed to UV-B radiation for 12h. High UV-B radiation also declined the quantum yield of primary photochemistry (φ po), the quantum yield for electron transport (φ Eo) and the efficiency per trapped excitation (Ψ o) in W. arrhiza simultaneously, while the amount of active PSII reaction centers per excited cross section (RC/CS) and the total number of active reaction centers per absorption (RC/ABS) had comparative changes. These results indicate that the effects of UV-B radiation on photosynthesis of W. arrhiza is due to an inhibition of the electron transport and via inactivation of reaction centers, but the inhibition may take place at more than one site in the photosynthetic apparatus. The phytogenic algicide sanguinarine shows strong inhibitory effects on some bloom-forming cyanobacteria and exhibits great potential in cyanobacterial bloom mitigation. To evaluate the possible ecological effects of sanguinarine on microalgae, the effects and possible mechanisms of sanguinarine on the competition between bloom-forming cyanobacterium Raphidiopsis raciborskii (formerly named Cylindrospermopsis raciborskii) and green alga Scenedesmus obliquus were investigated through co-culture competition test and comparative toxicological study including growth characteristics, chlorophyll fluorescence transients, activities of antioxidant enzymes, and lipid peroxidation. The results of Raphidiopsis-Scenedesmus co-culture competition test showed that sanguinarine decreased the competition ability of R. raciborskii , which benefitted S. obliquus in winning the competition. Toxicological studies have shown that sanguinarine exhibited high inhibitory effects on the growth and photosynthesis of R. raciborskii but no obvious toxicity on S. obliquus at concentrations of no more than 80 μg L−1. Oxidative damage partially contributed but was not the primary mechanism for the toxicity of sanguinarine on R. raciborskii. The results presented in this study indicate that sanguinarine may be a good algicidal candidate in mitigation of Raphidiopsis -based water bloom.
