Objective: To describe left recurrent laryngeal neurectomy (LRLn) performed under standing sedation and evaluate the effect of LRLn on upper respiratory tract function using a high-speed treadmill test (HST). We hypothesized that (1) unilateral LRLn could be performed in standing horses, resulting in ipsilateral arytenoid cartilage collapse (ACC); and (2) HST after LRLn would be associated with alterations in upper respiratory function consistent with dynamic ACC., Animals: 6 Thoroughbred horses., Methods: The horses were trained and underwent a baseline HST up to 14 m/s at 5% incline until fatigue. Evaluation included; airflow, pharyngeal and tracheal pressures, and dynamic upper respiratory tract endoscopy. Trans-laryngeal impedance (TLI) and left-to-right quotient angle ratio (LRQ) were calculated after testing. The following day, standing LRLn was performed in the mid-cervical region. A HST was repeated within 4 days after surgery., Results: Standing LRLn was performed without complication resulting in Havemayer grade 4 ACC at rest (complete paralysis) and Rakestraw grade C or D ACC (collapse up to or beyond rima glottis midline) during exercise. Increasing treadmill speed from 11 to 14 m/s increased TLI (P < .001) and reduced LRQ (P < .001). Neurectomy resulted in an increase in TLI (P = .021) and a reduction in LRQ (P < .001)., Clinical Relevance: Standing LRLn induces laryngeal hemiplegia that can be evaluated using a HST closely after neurectomy. Standing LRLn may be useful for future prospective evaluations of surgical interventions for laryngeal hemiplegia.

Background: Holter electrocardiographic monitoring is a cornerstone of diagnostic testing for arrhythmogenic cardiomyopathy (ACM) in Boxer dogs, but physical activity during monitoring is not controlled. In humans, exercise testing (ExT) can identify latent tachyarrhythmias associated with cardiomyopathy, and exercise increases serum cardiac troponin-I concentrations ([hs-cTnI]). These effects have not yet been investigated in Boxer dogs., Hypothesis/objectives: Subjecting Boxer dogs to brief, moderate-intensity ExT can identify changes in Holter recordings and [hs-cTnI] compared to baseline results., Animals: Thirty overtly healthy, client-owned Boxer dogs., Methods: Prospective interventional study. Dogs underwent baseline diagnostic testing including 24-hour Holter monitoring and [hs-cTnI], followed by brief ExT (accompanied, brisk stair-climbing and -descending for <5 minutes)., Results: Eleven dogs (37%) had >100 premature ventricular complexes (PVCs)/24 hours at baseline (3), ExT (3), or both (5). After ExT, these dogs had more PVCs/24 hours and greater increases in [hs-cTnI] compared to those with ≤100 PVCs/24 hours. Dogs with the striatin mutation had more PVCs/24 hours and a greater increase in [hs-cTnI] after ExT than did dogs without the striatin mutation., Conclusions and Clinical Importance: Exercise testing may improve the binary classification of Boxer dogs with or without ACM by increasing the number of PVCs and [hs-cTnI] in affected dogs to a greater degree than in unaffected dogs. This effect also is associated with presence or absence of the striatin mutation. Exercise should be a controlled variable when screening Boxer dogs for ACM., (© 2023 The Authors. Background: Incline treadmill and underwater treadmill (UWTM) exercises are common canine rehabilitation modalities , which are often used in isolation in dogs recovering from spinal surgery. Early use of an incline during UWTM exercise may have the potential to improve rehabilitation outcomes in dogs, but, it is hypothesised that dorsoventral movement of the spine may be excessive meaning it is unsuitable in some circumstances., Objectives: The purpose of this study was to identify changes in canine spinal kinematics in dogs when using a dry treadmill at different angles of incline compared to an underwater treadmill using the same inclines., Methods: Eight dogs were encouraged to walk on a dry, horizontal, underwater treadmill as well as under the same conditions with both a 10% and 20% incline. This was then repeated at a 10% and 20% incline with the addition of water to hock level. Data were collected using reflective anatomical markers placed at the occipital protuberance, T1, T13, L3, L7 and sacral apex, captured by a high-speed camera facing the lateral aspect of the treadmill. Dorsoventral motion of the spine as well as flexion, extension and range of motion (ROM) of T1, T13, L3 and L7 were recorded., Results: We found significant differences in dorsoventral spinal ROM at T1, L3 and L7, but no significant differences in T13 ROM. No significant differences were found in flexion and extension of any of the joints assessed when comparing dry conditions to the use of water (P>0.05)., Conclusions: The lack of significant differences in joint flexion and extension at T1, T13, L3 and L7 indicates the potential safe use of combining underwater treadmill and incline exercise in canine rehabilitation. However, a lack of uniformity in results makes distinguishing any patterns of significance difficult. Background: Despite its importance, there is no agreed definition of recovery of ambulation in dogs with spinal cord injury., Objectives: To validate a new walking test in dogs recovering from thoracolumbar spinal cord injury., Animals: Two hundred twenty-four dogs weighing <20 kg: 120 normally ambulatory dogs, plus 104 dogs undergoing decompressive surgery for acute thoracolumbar intervertebral disc herniation., Methods: Prospective cohort studies. The distance each freely-ambulatory dog walked during 50 step cycles was regressed on ulna length. For each postsurgical dog, we recorded when the calculated 50-step distance was completed without falling, or their inability to complete this distance by 4 months or more after surgery. Bayesian analysis compared outcomes for presurgical neurologic categories; association of recovery with several preoperative variables was explored using logistic and time-to-event regression., Results: For control dogs, 50-step distance (m) = 1.384 × ulnar length (cm) + 2.773. In postsurgical dogs, the 50-step test provided decisive evidence that deep pain-negative dogs were less likely to recover ambulation than dogs with intact pain perception (12/29 recovered vs 71/75; Bayes factor [BF] = 5.9 × 10 6 ) and, if they did recover, it took much longer (median 91 days vs median 14 days; BF = 1.5 × 10 3 ). Exploratory analysis suggested that presurgical neurologic status (subhazard ratio [SHR] = 0.022; P < .001) and duration of presurgical anesthesia (SHR = 0.740; P = .04) were associated with rapidity of recovery., Conclusions and Clinical Importance: This straightforward 50-step walking test provides robust data on ambulatory recovery well-suited to large scale pragmatic trials on treatment of thoracolumbar spinal cord injury in dogs., (© 2022 The Authors. Objective: To evaluate the association between the severity of dynamic upper airway obstruction (DUAO) and selected fitness parameters in Standardbred racehorses measured by treadmill test., Animals: 191 Standardbred racehorses that underwent a standardized diagnostic protocol for poor performance evaluation between 2002 and 2021 at the Veterinary Teaching Hospital, University of Milan., Procedures: Horses underwent a diagnostic protocol including clinical examination, lameness evaluation, laboratory analyses, ECG, resting upper airway endoscopy, metabolic incremental treadmill test, high-speed treadmill endoscopy, postexercise tracheobronchoscopy, and bronchoalveolar lavage fluid cytology. On the basis of findings of the high-speed treadmill endoscopy, horses were divided into the following groups: no DUAO, mild DUAO (medial deviation of the aryepiglottic folds or epiglottic entrapment), severe DUAO (dorsal displacement of the soft palate, nasopharyngeal collapse, dynamic laryngeal collapse, or epiglottic retroversion), and multiple DUAOs (different forms of DUAO occurring concomitantly). Fitness parameters, measured by incremental treadmill test, included speed at a heart rate of 200 bpm (V200), speed and heart rate at a lactate concentration of 4 mmol/L (VLa4, HRLa4), peak lactate, maximum speed, maximum hematocrit, and minimum pH. Data were compared between groups by use of descriptive and statistical analyses., Results: Horses affected by severe or multiple DUAOs showed lower V200 and VLa4, compared to the no-DUAO group; horses with multiple DUAOs also had higher peak lactate concentrations compared to horses without DUAOs., Clinical Relevance: While mild forms of DUAO do not affect athletic capacity, severe forms of DUAO and multiple DUAOs may lead to reduced fitness in Standardbred racehorses by impairing aerobic capacity.

Salivary biomarkers could be useful to evaluate stress, fitness level, and skeletal muscle damage associated to exercise in horses in an easy and non-painful way. Therefore, this study aims to evaluate if cortisol in saliva (sCor), salivary alpha-amylase (sAMY) and butyrylcholinesterase (sBChE) and lactate (sLA) and creatine kinase (sCK) in saliva of horses can show changes during a standardized exercise test, and if they are related to heart rate variability (HRV) parameters related to sympathetic and parasympathetic tone, fitness level or skeletal muscle damage. For this purpose, ten endurance horses were submitted to a standardized exercise test in field conditions. Saliva and blood were obtained at basal time (TB), after the seven bouts of velocity (T + 01 to T + 07), and 5, 15, 30, and 45 min later (T + 5, T + 15, T + 30, and T + 45). Five endurance horses in resting condition (control group) were also enrolled. HRV and fitness level parameters, and plasma CK as a marker of muscle damage were also evaluated. Salivaryalpha-amylase increased at T + 30 (P = 0.03), sBChE at T + 5 (P = 008), and sCK at T + 07 (P = 0.009) after the exercise test, with significant differences between the exercise and control groups' results. The sCor did not show significant changes during the exercise test in the exercise group but higher concentration compared to the control horses (P < 0.001) were observed. sCor, sAMY, sBChE, and sCK showed a positive correlation (r values between 0.47 and 0.64) with the sympathetic tone and a negative correlation (r values between -0.37 and -0.56) with the parasympathetic tone. In conclusion, sAMY, sBChE, and sCK showed significant increases in ten endurance horses after an increasing intensity velocity exercise. Values of sCor, sAMY, sBChE, and sCK were associated with HRV, which is used to evaluate stress, and therefore, they could be potentially used to assess the exercise-related stress after a physical effort., (Copyright © 2021 The Authors. Background: Glycogen in skeletal muscle is a major source of energy during exercise and an important determinant of endurance capacity, so that its measurement may provide a meaningful marker of athletes' preparation and a possible predictor of performance, both in humans and in equines. Gold standard of glycogen concentration measurement is the histochemical and biochemical analysis of biopsy-derived muscle tissue, an invasive and potentially injuring procedure. Recently, high-frequency ultrasound (US) technology is being exploited in human sports medicine to estimate muscle glycogen content. Therefore, aim of the present study is to evaluate the feasibility of US assessment of muscle glycogen in equines., Results: US images of gluteus medius (GL) and semitendinosus (ST) muscles were obtained on eight healthy horses (3-10 years) before and after a steady-state exercise on treadmill (velocity: 4.0-12.5 m/s; duration: 2-20 min; heart rate: 137-218 b/min). Average image greyscale intensity was significantly different between GL and ST, both before and after exercise (p < 0.001). Comparing baseline and post-exercise US images, significant increase in greyscale intensity has been observed in ST (p < 0.001), but not in GL (p = 0.129). The volume of the exercise was significantly correlated with exercise-dependent change in image intensity (R 2  = 0.891), consistent with a reduction of glycogen muscle stores resulting from aerobic activity., Conclusions: US technique evidences also in horses muscle changes possibly associated to glycogen utilisation during exercise. Present results on a small sample need to be further confirmed and provide preliminary data warranting future validation by direct glycogen measurement through biopsy technique.

We investigated whether horses trained in moderate and mild hypoxia demonstrate greater improvement in performance and aerobic capacity compared to horses trained in normoxia and whether the acquired training effects are maintained after 2 weeks of post-hypoxic training in normoxia. Seven untrained Thoroughbred horses completed 4 weeks (3 sessions/week) of three training protocols, consisting of 2-min cantering at 95% maximal oxygen consumption V ˙ O 2 max under two hypoxic conditions (H16, F I O 2  = 16%; H18, F I O 2  = 18%) and in normoxia (N21, F I O 2  = 21%), followed by 2 weeks of post-hypoxic training in normoxia, using a randomized crossover study design with a 3-month washout period. Incremental treadmill tests (IET) were conducted at week 0, 4, and 6. The effects of time and groups were analyzed using mixed models. Run time at IET increased in H16 and H18 compared to N21, while speed at V ˙ O 2 max was increased significantly only in H16. V ˙ O 2 max in all groups and cardiac output at exhaustion in H16 and H18 increased after 4 weeks of training, but were not significantly different between the three groups. In all groups, run time, V ˙ O 2 max , V V ˙ O 2 max , Q ˙ max , and lactate threshold did not decrease after 2 weeks of post-hypoxic training in normoxia. These results suggest that 4 weeks of training in moderate (H16), but not mild (H18) hypoxia elicits greater improvements in performance and running economy than normoxic training and that these effects are maintained for 2 weeks of post-hypoxic training in normoxia., (© 2021 The Authors. Objective: To investigate the effects of novel legwear designed to limit metacarpophalangeal joint (MCPJ) extension and redirect loading forces from the flexor apparatus through analyses of 2-D kinematic and kinetic data., Animals: 6 adult horses without musculoskeletal disease., Procedures: Horses were subjected to 4 treatments: control (no legwear), inactive legwear (unlimited legwear extension), and active legwear with mild (30°) and moderate (20°) legwear extension limitation. Two-dimensional kinematic data were collected for the right forelimb (FL) during walk and trot and from leading and trailing FLs during canter on a treadmill. Ground reaction force (GRF) data were collected from FLs during overground walk and trot. Peak MCPJ angle and angular velocity were calculated from kinematic data, and peak force and average loading rate were calculated from vertical GRF data during the stance phase of the gait. Interactions between gait and treatment were determined via ANOVA., Results: Interactions between gait and treatment for peak MCPJ angle were significant. Significant reductions in MCPJ angle were noted between the control treatment and legwear with moderate extension limitation for trot and canter (leading and trailing FL) and between inactive legwear and legwear with moderate extension limitation for trot and leading FL during canter. Interactions among peak MCPJ angular velocity, peak vertical GRF, and average loading rate of the vertical GRF showed nonsignificance., Conclusions and Clinical Relevance: Significant reductions in MCPJ extension without significant alterations to peak vertical GRF suggested the legwear's ability to redistribute internal forces. Findings suggested that the legwear may be beneficial for horses rehabilitating from flexor apparatus injuries.

Objective: To evaluate intrasession and intersession repeatability of measurements for temporospatial and kinetic variables obtained with a pressure-sensitive treadmill designed for gait analysis of dogs., Animals: 16 client-owned dogs., Procedures: The influence of treadmill speed on accuracy of ground reaction force (GRF) measurements was assessed by simulated gait analysis at 0 to 7.5 km/h with a custom test device. A similar test was performed with 1 client-owned dog ambulating on the treadmill at 5 speeds (3 to 7 km/h) for GRF calculations. Fifteen client-owned dogs were then walked on the treadmill at 3 km/h for collection of temporospatial and kinetic data. Intrasession repeatability was determined by comparing 2 sets of measurements obtained ≤ 2 hours apart. Intersession repeatability was determined by comparing the first set of these measurements with those for a second session ≥ 4 days later. Intraclass correlation coefficients (ICCs; consistency test) and difference ratios were calculated to assess repeatability., Results: Increases in treadmill speed yielded a mean 9.1% decrease in weight-normalized force data at belt speeds of up to 7.5 km/h for the test device, compared with the value when the treadmill belt was stationary. Results were similar for the dog at increasing treadmill speeds (mean decrease, 12.4%). For temporospatial data, intrasession ICCs were > 0.9 and intersession ICCs ranged from 0.75 to 0.9; for GRFs, intrasession and intersession ICCs ranged from 0.68 to 0.97 and from 0.35 to 0.78, respectively., Conclusions and Clinical Relevance: Repeatability of temporospatial data for healthy dogs was good to excellent; results for kinetic data varied. Further research is needed to investigate use of this system for gait analysis with larger samples of dogs and dogs with lameness.

Objective: To investigate whether an actual improvement in gait could be differentiated from physiologic differences or habituation effects during gait analysis of dogs., Animals: 11 healthy dogs., Procedures: On 4 examination days, kinetic parameters were measured while dogs were walking on a treadmill. Differences in mean parameter values and habituation effects (ie, effect sizes) were quantified and compared among examination days. Coefficients of variation for repeated measurements were calculated to determine measurement reproducibility, and minimum differences were calculated to distinguish between physiologic fluctuation and an actual change in gait pattern., Results: Among the 4 examination days, mean absolute differences in peak vertical force and vertical impulse (VI) varied from 1.5% to 5.3% of body weight (BW) and 0.9% to 1.8% of BW·s, respectively. Mean absolute differences in the percentage of stance-phase duration (%SPD) and relative stride length (RSL) varied from 0.9% to 3.2% and 1.7% to 3.0%, respectively. Reproducibility of parameter measurements was good. Values for %SPD had the lowest amount of dispersion and largest effect size, suggesting a habituation effect for this parameter. Calculated minimum differences among the days for peak vertical force, VI, %SPD, and RSL did not exceed 9.9% of BW, 3.3% of BW·s, 5.8 percentage points, and 5.2 percentage points, respectively., Conclusions and Clinical Relevance: The %SPD of healthy dogs walking on a treadmill was the most sensitive and diagnostically reliable of the measured kinetic parameters, in contrast to VI and RSL. Findings suggested that actual changes can be distinguished from random physiologic fluctuations during gait analysis of dogs.

Background: A greater understanding of exercise physiology and biochemistry is required for the sport horse disciplines, including show jumping. Conditioning of horses for show jumping is empirical because they are primarily trained on flat ground, however the equivalent workload between jumping and flat work is currently unknown. The objectives of the study were therefore to compare the physiological demands of Warmblood show jumpers over a standardized 1.10 m course vs a 600 m standardized incremental exercise test on flat ground, and to report reference field test values for competitive show jumping horses. In this prospective field study, 21 healthy, actively competing Warmblood show jumping horses were assessed to determine physiological variables after a standardized jumping course at 6.4 m/s (average speed) and track standardized incremental exercise test at 5 m/s, 8 m/s and 11 m/s. Heart rate, velocity, blood lactate, blood pH, pCO2, bicarbonate, PCV and TP concentrations were recorded. V200, V170 and VLa4 were calculated. Parametric statistics were performed on analysis of all 21 horses' variables., Results: Contrary to exercise at 5 m/s and 11 m/s, cantering at 8 m/s did not induce any significant difference in blood lactate, mean heart rate or mean venous blood pH compared to after completion of the jumping course., Conclusions: Jumping a 1.10 m course demands a statistically similar workload to cantering around a flat track at 8 m/s. This study will help to test fitness and design conditioning programs for Warmblood show jumping horses.

The objective of this study was to explore the mathematical relationships between independent variables (patient morphometrics and treadmill speed) and dependent variables (accelerometer or pedometer output) when evaluating data from accelerometers and pedometers in dogs. Twenty dogs took part in 3 randomized activities, consisting of exercise on a treadmill at 1.0, 1.5, and 2.0 m/s for a total distance of 1 km at each speed. Dogs simultaneously wore both an accelerometer and a pedometer. Statistical analysis used multiple regression models to discover the relationships between independent and dependent variables. A formula was developed to predict the distance traveled by a dog based on its morphometrics and activity monitor output. Shoulder height had stronger correlations to accelerometer and pedometer outputs than other morphometric variables. As shoulder height increased, all accelerometer and pedometer outputs decreased. As treadmill speed increased, both accelerometer and pedometer step counts decreased, while accelerometer activity counts increased. According to a formula derived to predict the total distance traveled using patient shoulder height and accelerometer or pedometer output, pedometer steps were the most accurate predictor of distance traveled. Accelerometer steps were less accurate when using the same model. Accelerometer activity counts did not reveal a meaningful predictive formula. The results of this study indicate that patient morphometrics and treadmill speed (as a measure of intensity) influenced accelerometer and pedometer data. The pedometer data more precisely and accurately estimated the distance traveled based on step counts and patient shoulder height. Brachycephalic syndrome (BS) is a pathophysiological disorder caused by excessive soft tissue within the upper airways of short-nosed dog breeds, causing obstruction of the nasal, pharyngeal and laryngeal lumen, resulting in severe respiratory distress. As the prevalence of BS appears to be high among some of the affected breeds, there is an urgent need for breeding efforts to improve the health status of those dogs. In the present study, we evaluated correlations between morphometric and other phenotypic characteristics and BS in a population of 69 French bulldogs from Denmark to identify parameters that could serve as a basis for breeding against BS. Furthermore, the genetic variation was monitored to determine whether it would be possible to breed based on these characteristics without simultaneously causing a critical reduction in genetic variation. Six phenotypic characteristics were correlated with the Brachycephalic Syndrome Functional (BSF) score. Among the morphometric risk factors, nostril stenosis (NS) and neck girth (NG) had the highest impact on the BSF score, accounting for 32% and 4% of the variation, respectively. The genetic variation in the population was comparable to other pure breeds, i.e. estimated and observed heterozygosity was 0.60 and the average inbreeding coefficient was 0.01. If only dogs with Grades 1 and 2 NS (no or only mild NS) were selected for breeding the mean BSF score would be reduced significantly. However, it would result in the exclusion of 81% of the population for breeding and this is not prudent. Excluding only dogs with severe stenosis (Grade 4) would exclude 50% of the population without any adverse impact on genetic variation within the population. Although exclusion of dogs with Grade 4 would result in an apparent reduction in the mean BSF score, this reduction is not significant. As NS accounts for 32% of the variation in BSF score, a possible long term strategy to reduce the prevalence of the BS in French bulldogs would seem to be a selection scheme that first excluded dogs with the most severe NS from breeding, gradually moving towards selecting dogs with lower NS grades. Background: Exercise testing in conjunction with measurement of cardiac biomarkers NT-proBNP and cTnI is a useful tool for monitoring the effect of treatment on cardiac patients. Administering Pimobendan in dogs with degenerative mitral valve disease (DMVD) and cardiomegaly results in delaying the onset of clinical symptoms and prolonging life. Its effect in dogs with DMVD without cardiomegaly has not been well examined. The aim of the current study was to investigate the effect of administering Pimobendan in dogs with DMVD without cardiomegaly using exercise testing in conjunction with measuring cardiac biomarkers in addition to echocardiography. Twenty-one dogs with asymptomatic DMVD without echocardiographic signs of cardiomegaly participated in a randomised, double-blinded trial. Dogs were divided into a Pimobendan-group (n = 11) and a placebo-group (n = 10) in a double-blinded study design and underwent a standardised submaximal exercise test (SSET). One dog in the Pimobendan-group was retrospectively removed from the study after being diagnosed with Leishmaniosis. Cardiac biomarkers NT-proBNP and cTnI were measured before and after exercise. Follow-up appointments were performed at days 90 and 180., Results: Dogs in the Pimobendan-group had significantly lower post-exercise NT-proBNP-levels after being administered Pimobendan than at the beginning of the study. They also had lower pre- and post-exercise-NT-proBNP-levels than those dogs in the placebo-group. There was neither a significant difference regarding the measured cTnI levels nor an increase in cTnI between the groups at any time., Conclusions: Pimobendan lowers NT-proBNP in dogs with presymptomatic mitral valve disease without cardiomegaly before and after submaximal exercise. This indicates a reduction in cardiac wall stress. If dogs with asymptomatic DMVD without cardiomegaly benefit from treatment with Pimobendan (for example, through a longer survival time) warrants further investigation.

Background: Little is known about normal heart rate variability (HRV) in horses during exercise. It can be difficult to separate premature beats from normal beat-to-beat variation at higher heart rates., Objectives: The aim was to quantify HRV in healthy horses during a high-speed treadmill-standardized exercise test (HSET) and to compare with the HRV in horses observed to have arrhythmias during exercise., Animals: Thirteen healthy horses (Group H), 30 horses with arrhythmias (Group A), and 11 horses with poor performance but no observed arrhythmias (Group O)., Methods: Prospective, observational study. All horses performed a HSET with simultaneous electrocardiograph (ECG) recorded. The ECGs were corrected for artifacts, and arrhythmias noted. Percent instantaneous beat-to-beat cycle length variation (% R-R variation) was calculated, and HRV analyses were performed on trot, canter, and recovery segments., Results: Group H showed between -4.4 and +3.8% R-R variation during trot and between -6.1 and +5.4% R-R variation during the canter phase of the HSET. Group A had significantly larger maximum and 1st percentile R-R shortening and lengthening compared with Group H and Group O during the recovery phase where most arrhythmias were observed. During recovery, a cutoff of 6% maximum % R-R shortening predicted the presence of arrhythmia with 88% sensitivity and 97% specificity and likelihood ratio of 26., Conclusions and Clinical Importance: Healthy horses have little instantaneous R-R variation during exercise. If a cardiac cycle shortens more than 6% from the previous cycle during the recovery phase, this R-R interval is likely to represent an arrhythmic event., (© 2018 The Authors. OBJECTIVE To quantify fatigue-induced electromyographic changes in hind limb muscles in horses. ANIMALS 8 Thoroughbreds. PROCEDURES The left and right hind limb longissimus dorsi, tensor fasciae latae, gluteus medius, and biceps femoris muscles were instrumented for surface electromyography. Hoof strain gauges were attached to confirm stride cycle. Each horse was galloped on a treadmill (grade, 3%) at a constant speed (12.6 to 14.7 m/s) to achieve fatigue after approximately 360 seconds. Before and after this exercise, the horses were trotted at 3.5 m/s. At 30-second intervals during galloping an integrated electromyography (iEMG) value for a stride and the median frequency of muscle discharge (MF) in each limb were measured. The mean of stride frequency (SF), iEMG value, and MF of 5 consecutive strides at the start and end of galloping for the lead and trailing limbs were compared. For trotting, these variables were compared at 60 seconds before and after galloping. RESULTS The mean ± SD value for SF decreased over time (2.14 ± 0.06 to 2.05 ± 0.07 stride/s). In both the lead and trailing limbs, fatigue decreased the iEMG values of the gluteus medius and biceps femoris muscles but not those of the longissimus dorsi and tensor fasciae latae muscles. The MF did not change for any muscle during galloping with fatigue. The SF, iEMG value, and MF did not change during trotting with fatigue. CONCLUSIONS AND CLINICAL RELEVANCE Fatigue induced by high-speed galloping decreased the gluteus medius and biceps femoris muscles' iEMG values in Thoroughbreds. Fatigue of these less fatigue-resistant hind limb muscles would affect a horse's speed.

Background: Atrial fibrillation (AF) is the most common arrhythmia affecting performance in horses. However, no previous studies have quantified the performance reduction in horses suffering from AF., Objectives: To quantify the effect of AF on maximum velocity (V max ), maximum heart rate (HR max ), heart rate recovery (T 100 ), hematologic parameters and development of abnormal QRS complexes., Animals: Nine Standardbred trotters., Methods: Two-arm controlled trial. Six horses had AF induced by means of a pacemaker and 3 served as sham-operated controls. All horses were subjected to an exercise test to fatigue before (SET1) and after (SET2) 2 months of AF or sham. The V max and HR max were assessed using a linear mixed normal model. Abnormal QRS complexes were counted manually on surface ECGs., Results: Atrial fibrillation resulted in a 1.56 m/sec decrease in V max (P < .0001). In the AF group, HR max  ± SD increased from 226 ± 11 bpm at SET1 to 311 ± 27 bpm at SET 2. The AF group had higher HR max at SET2 compared with controls (P < .0001), whereas no difference between the control and AF groups was observed at SET1 (P = .96). Several episodes of wide complex tachycardia were observed during exercise in 3 of the AF horses during SET2., Conclusions and Clinical Importance: Atrial fibrillation resulted in a significant reduction in performance, an increase in HR and development of abnormal QRS complexes during exercise, which may be a risk factor for collapse or sudden cardiac death., (Copyright © 2018 The Authors. Background: Lameness due to paw injuries is common in the clinical practice. Although many studies investigated gait adaptations to diseases or injuries, mainly of the hip and knee, our understanding of the biomechanical coping mechanisms that lame dogs utilize is limited. Therefore, this study evaluated the kinematic changes associated with an induced, load-bearing pelvic limb lameness in healthy dogs trotting on a treadmill. Kinematic analysis included spatio-temporal comparisons of limb, joint and segment angles of all limbs. Key parameters compared between sound and lame conditions were: angles at touch-down and lift-off, minimum and maximum joint angles and range of motion., Results: Significant differences were identified in each limb during both stance and swing phases. The most pronounced differences concerned the affected pelvic limb, followed by the contralateral pelvic limb, the contralateral thoracic limb and, to the least degree, the ipsilateral thoracic limb. The affected limb was retracted more, while the contralateral limb was protracted more, consistent with this limb bearing more body weight in lame dogs., Conclusions: Kinematic adaptations involved almost all segment and joint angles in the pelvic limbs, while they exclusively concerned distal parts of the thoracic limbs. Comparisons with tripedal locomotion reveal several striking similarities, implying that dogs use similar principles to cope with a partial or a total loss in limb function. Because kinematic alterations occurred in all limbs and not just the affected one, all limbs should be included in routine follow-ups and be part of the diagnostic and therapeutic care of canine patients.

Background: The equine m. gluteus medius (GM) is the largest muscle of the horse, its main movement function is the extension of the hip joint. The objective of the present study was to measure equine GM activity in three adjacent locations on GM during walk and trot on a treadmill, in order to document potential differences. Fourteen Haflinger mares were measured using surface electromyography and kinematic markers to identify the motion cycles on three occasions over 16 weeks. The electrodes were placed on left and right gluteus medius muscle over the middle of its widest part and 5 cm lateral and medial of it. For data processing, electrical activity was normalised to its maximum value and timing was normalised to the motion cycle. A Gaussian distribution approach was used to determine up to 10 modes of focussed activity, and results were analysed separately for stance and swing phase of the ipsilateral hindlimb., Results: Fair reliability was found for mean mode values (Cronbach's alpha = 0.66) and good reliability was found for mean mode locations (Cronbach's alpha = 0.71) over the three data collection days. The magnitude of muscle activity identified as mean mode value was much larger at trot than at walk, and mean mode value was significantly different between stance phases of walk and trot for all electrode positions (p < 0.01). The pattern of muscle activity identified as mean mode location was significantly different for walk and trot at all electrode positions, both during stance and swing phases (p < 0.001). This indicates the different timing pattern between the gaits. Results of the three electrode positions on the same muscle during each gait were not significantly different when comparing the same measurement., Conclusions: The middle of the equine GM does not show any indication of functional differentiation during walk and trot on a treadmill; this might be due to lack of segmentation as such, or due to lack of need for segmented use for these very basic main tasks of the muscle. The reliability of the sEMG measurements over several weeks was fair to good, an indication for the robustness of the methodology.

OBJECTIVE To assess effects of speed on kinematic variables measured by use of extremity-mounted inertial measurement units (IMUs) in nonlame horses performing controlled exercise on a treadmill. ANIMALS 10 nonlame horses. PROCEDURES 6 IMUs were attached at predetermined locations on 10 nonlame Franches Montagnes horses. Data were collected in triplicate during trotting at 3.33 and 3.88 m/s on a high-speed treadmill. Thirty-three selected kinematic variables were analyzed. Repeated-measures ANOVA was used to assess the effect of speed. RESULTS Significant differences between the 2 speeds were detected for most temporal (11/14) and spatial (12/19) variables. The observed spatial and temporal changes would translate into a gait for the higher speed characterized by increased stride length, protraction and retraction, flexion and extension, mediolateral movement of the tibia, and symmetry, but with similar temporal variables and a reduction in stride duration. However, even though the tibia coronal range of motion was significantly different between speeds, the high degree of variability raised concerns about whether these changes were clinically relevant. For some variables, the lower trotting speed apparently was associated with more variability than was the higher trotting speed. CONCLUSIONS AND CLINICAL RELEVANCE At a higher trotting speed, horses moved in the same manner (eg, the temporal events investigated occurred at the same relative time within the stride). However, from a spatial perspective, horses moved with greater action of the segments evaluated. The detected changes in kinematic variables indicated that trotting speed should be controlled or kept constant during gait evaluation.

Background: Despite the use of water treadmills (WT) in conditioning horses, the intensity of WT exercise has not been well documented. The workload on a WT is a function of water height and treadmill speed. Therefore, the purpose of this study was to determine the effects of these factors on workload during WT exercise. Fifteen client-owned Quarter Horses were used in a randomized, controlled study. Three belt speeds and three water heights (mid cannon, carpus and stifle), along with the control condition (dry treadmill, all three speeds), were tested. Measured outcomes were oxygen consumption (V̇O 2 ), ventilation (respiratory frequency, tidal volume (V T )), heart rate (HR), and blood lactate. An ergospirometry system was used to measure V̇O 2 and ventilation. Linear mixed effects models were used to examine the effects of presence or absence of water, water height and speed (as fixed effects) on measured outcomes., Results: Water height and its interaction with speed had a significant effect on V̇O 2 , V T and HR, all peaking at the highest water level and speed (stifle at 1.39 m/s, median V̇O 2  = 16.70 ml/(kg.min), V T  = 6 L, HR = 69 bpm). Respiratory frequency peaked with water at the carpus at 1.39 m/s (median 49 breaths/min). For a given water height, the small increments in speed did not affect the measured outcomes. Post-exercise blood lactate concentration did not change., Conclusions: Varying water height and speed affects the workload associated with WT exercise. The conditions utilized in this study were associated with low intensity exercise. Water height had a greater impact on exercise intensity than speed.

This study investigated the effects of transport and diet on metabolic response during a subsequent race-like test in Standardbred horses in training fed a forage-only diet and a 50:50 forage:oats diet. Six trained and raced Standardbred trotter mares were used. Two diets, 1 forage-only diet (FONLY) and 1 diet with 50% of DM intake from forage and 50% from oats (FOATS), were fed for two 29-d periods in a crossover design. At Day 21, the horses were subjected to transport for 100 km before and after they performed an exercise test (transport test [TT]). At Day 26, the horses performed a control test (CT), in which they were kept in their stall before and after the exercise test. Blood samples were collected throughout the study, and heart rate and water intake were recorded. Heart rate and plasma cortisol, glucose, and NEFA concentrations were greater for the TT than for the CT ( = 0.008, = 0.020, = 0.010, and = 0.0002, respectively) but were not affected by diet. Plasma acetate concentration was lower during the TT than during the CT ( = 0.034) and greater for the FONLY than for the FOATS ( = 0.003). There were no overall effects of the TT compared with the CT on total plasma protein concentration (TPP), but TPP was lower with the FONLY than with the FOATS ( = 0.016). There was no overall effect of the TT compared with the CT on water intake, but water intake was greater with the FONLY than the FOATS ( = 0.011). There were no overall effects of transport or diet on BW, plasma lactate, or plasma urea concentration. It was concluded that both transport and diet affect metabolic response during exercise in horses. Aerobic energy supply was most likely elevated by transportation and by the FONLY. The FONLY also decreased exercise-induced effects on extracellular fluid regulation. These results highlight the importance of experimental design in nutrition studies. If the aim is to examine how a diet affects exercise response in competition horses, transport should preferably be included in the experimental design, because horses are likely to be transported before a competition.

Background: Severity of pulmonary hypertension (PH) in dogs is related to clinical signs and prognosis., Hypothesis/objectives: We hypothesized that Doppler echocardiographic (DE) indices of pulmonary artery pressure (PAP) and pulmonary vascular resistance (PVR) are influenced by independent factors that create clinically important variability of DE-based estimates of PH in dogs., Animals: Thirty-eight client owned dogs with naturally acquired degenerative atrioventricular valve disease and tricuspid regurgitation (TR)., Methods: Dogs were prospectively enrolled, and target variables were acquired during 4 echocardiographic study periods (lateral recumbency, standing, lateral recumbency after a 6-minute walk test [6MWT], and lateral recumbency after sedation with butorphanol 0.25 mg/kg IM). Statistical methods included repeated measures ANOVA, mixed model analysis, and Chi-squared test of association., Results: There was a significant increase in peak TR flow velocity (TRFV; P < 0.01) after sedation in 78% of dogs, with TRFV increasing by >0.4 m/s in 42% of dogs, independent of stroke volume. A significant effect of study period on DE-estimated PVR was not found (P = 0.15). There were negligible effects of sonographer, body position, and 6MWT on echocardiographic variables of PH. Clinically relevant cyclic variation of TRFV was found. There was an association between estimation of right atrial pressure based on subjective assessment and estimation based on cranial vena cava collapsibility (P = 0.03)., Conclusions and Clinical Importance: The increase in TRFV observed with sedation could change assessment of PH severity and impact prognostication and interpretation of treatment response. This study examined the effect of increasing BW ratio (BWR) between rider and horse, in the BWR range common for Icelandic horses (20% to 35%), on heart rate (HR), plasma lactate concentration (Lac), BWR at Lac 4 mmol/l (W4), breathing frequency (BF), rectal temperature (RT) and hematocrit (Hct) in Icelandic horses. In total, eight experienced school-horses were used in an incremental exercise test performed outdoors on an oval riding track and one rider rode all horses. The exercise test consisted of five phases (each 642 m) in tölt, a four-beat symmetrical gait, at a speed of 5.4±0.1 m/s (mean±SD), where BWR between rider (including saddle) and horse started at 20% (BWR20), was increased to 25% (BWR25), 30% (BWR30), and 35% (BWR35) and finally decreased to 20% (BWR20b). Between phases, the horses were stopped (~5.5 min) to add lead weights to specially adjusted saddle bags and a vest on the rider. Heart rate was measured during warm-up, the exercise test and after 5, 15 and 30 min of recovery and blood samples were taken and BF recorded at rest, and at end of each of these aforementioned occasions. Rectal temperature was measured at rest, at end of the exercise test and after a 30-min recovery period. Body size and body condition score (BCS) were registered and a clinical examination performed on the day before the test and for 2 days after. Heart rate and BF increased linearly (P0.05), but negative correlations (P<0.05) existed between body size measurements and Hct. While HR, Hct and BF recovered to values at rest within 30 min, Lac and RT did not. All horses had no clinical remarks on palpation and at walk 1 and 2 days after the test. In conclusion, increasing BWR from 20% to 35% resulted in increased HR, Lac, RT and BF responses in the test group of experienced adult Icelandic riding horses. The horses mainly worked aerobically until BWR reached 22.7%, but considerable individual differences (17.0% to 27.5%) existed that were not linked to horse size, but to back BCS.

This study investigated the effects of rider weight in the BW ratio (BWR) range common for Icelandic horses (20% to 35%), on stride parameters in tölt in Icelandic horses. The kinematics of eight experienced Icelandic school horses were measured during an incremental exercise test using a high-speed camera (300 frames/s). Each horse performed five phases (642 m each) in tölt at a BWR between rider (including saddle) and horse starting at 20% (BWR20) and increasing to 25% (BWR25), 30% (BWR30), 35% (BWR35) and finally 20% (BWR20b) was repeated. One professional rider rode all horses and weight (lead) was added to saddle and rider as needed. For each phase, eight strides at speed of 5.5 m/s were analyzed for stride duration, stride frequency, stride length, duty factor (DF), lateral advanced placement, lateral advanced liftoff, unipedal support (UPS), bipedal support (BPS) and height of front leg action. Stride length became shorter (Y=2.73-0.004x; P0.05). In conclusion, increased BWR decreased stride length and increased DF proportionally to the same extent in all limbs, whereas BPS increased at the expense of decreased UPS. These changes can be expected to decrease tölt quality when subjectively evaluated according to the breeding goals for the Icelandic horse. However, beat, symmetry and height of front leg lifting were not affected by BWR.

OBJECTIVE To perform 3-D inverse dynamics analysis of the entire forelimb of healthy dogs during a walk and trot. ANIMALS 5 healthy adult Beagles. PROCEDURES The left forelimb of each dog was instrumented with 19 anatomic markers. X-ray fluoroscopy was used to optimize marker positions and perform scientific rotoscoping for 1 dog. Inverse dynamics were computed for each dog during a walk and trot on the basis of data obtained from an infrared motion-capture system and instrumented quad-band treadmill. Morphometric data were obtained from a virtual reconstruction of the left forelimb generated from a CT scan of the same dog that underwent scientific rotoscoping. RESULTS Segmental angles, torque, and power patterns were described for the scapula, humerus, ulna, and carpus segments in body frame. For the scapula and humerus, the kinematics and dynamics determined from fluoroscopy-based data varied substantially from those determined from the marker-based data. The dominant action of scapular rotation for forelimb kinematics was confirmed. Directional changes in the torque and power patterns for each segment were fairly consistent between the 2 gaits, but the amplitude of those changes was often greater at a trot than at a walk. CONCLUSIONS AND CLINICAL RELEVANCE Results indicated that control of the forelimb joints of dogs is similar for both a walk and trot. Rotation of the forelimb around its longitudinal axis and motion of the scapula should be reconsidered in the evaluation of musculoskeletal diseases, especially before and after treatment or rehabilitation.

OBJECTIVE To determine cardiorespiratory responses of Thoroughbreds to uphill and downhill locomotion on a treadmill at identical gradients. ANIMALS 5 highly trained Thoroughbred geldings. PROCEDURES Thoroughbreds were exercised for 2-minute intervals on a treadmill at 1.7, 3.5, 6.0, 8.0, and 10.0 m/s at a 4% incline, 0% incline (horizontal plane), and 4% decline in random order on different days. Stride frequency, stride length, and cardiopulmonary and O 2 -transport variables were measured and analyzed by means of repeated-measures ANOVA and Holm-Šidák pairwise comparisons. RESULTS Horses completed all treadmill exercises with identical stride frequency and stride length. At identical uphill speeds, they had higher (vs horizontal) mass-specific O 2 consumption (mean increase, 49%) and CO 2 production (mean increase, 47%), cardiac output (mean increase, 21%), heart rate (mean increase, 11%), and Paco 2 (mean increase, 1.7 mm Hg), and lower Pao 2 (mean decrease, 5.8 mm Hg) and arterial O 2 saturation (mean decrease, 1.0%); tidal volume was not higher. Downhill locomotion (vs horizontal) reduced mass-specific O 2 consumption (mean decrease, 24%), CO 2 production (mean decrease, 23%), and cardiac output (mean decrease, 9%). Absolute energy cost during uphill locomotion increased linearly with speed at approximately twice the rate at which it decreased during downhill locomotion. CONCLUSIONS AND CLINICAL RELEVANCE Findings suggested that for Thoroughbreds, downhill locomotion resulted in a lower energy cost than did horizontal or uphill locomotion and that this cost changed with speed. Whether eccentric training induces skeletal muscle changes in horses similar to those in humans remains to be determined.

Background: Most Friesian horses reach their anaerobic threshold during a standardized exercise test (SET) which requires lower intensity exercise than daily routine training., Aim: to study strengths and weaknesses of an alternative SET-protocol. Two different SETs (SETA and SETB) were applied during a 2 month training period of 9 young Friesian dressage horses. SETB alternated short episodes of canter with trot and walk, lacking long episodes of cantering, as applied in SETA. Following parameters were monitored: blood lactic acid (BLA) after cantering, average heart rate (HR) in trot and maximum HR in canter. HR and BLA of SETA and SETB were analyzed using a paired two-sided T-test and Spearman Correlation-coefficient (p* < 0.05)., Results: BLA after cantering was significantly higher in SETA compared to SETB and maximum HR in canter was significantly higher in SETA compared to SETB. The majority of horses showed a significant training response based upon longitudinal follow-up of BLA. Horses with the lowest fitness at start, displayed the largest training response. BLA was significantly lower in week 8 compared to week 0, in both SETA and SETB. A significantly decreased BLA level after cantering was noticeable in week 6 in SETA, whereas in SETB only as of week 8. In SETA a very strong correlation for BLA and average HR at trot was found throughout the entire training period, not for canter., Conclusions: Young Friesian horses do reach their anaerobic threshold during a SET which requires lower intensity than daily routine training. Therefore close monitoring throughout training is warranted. Longitudinal follow up of BLA and not of HR is suitable to assess training response. In the current study, horses that started with the lowest fitness level, showed the largest training response. During training monitoring HR in trot rather than in canter is advised. SETB is best suited as a template for daily training in the aerobic window.

OBJECTIVE To determine whether racehorses undergoing regular exercise at 2 intensities or stall rest during a period of reduced training (detraining) would differentially maintain their cardiopulmonary and oxygen-transport capacities. ANIMALS 27 Thoroughbreds. PROCEDURES Horses trained on a treadmill for 18 weeks underwent a period of detraining for 12 weeks according to 1 of 3 protocols: cantering at 70% of maximal rate of oxygen consumption ([Formula: see text]o 2max ) for 3 min/d for 5 d/wk (canter group); walking for 1 h/d for 5 d/wk (walk group); or stall rest (stall group). Standardized treadmill exercise protocols (during which cardiopulmonary and oxygen-transport variables were measured) were performed before and after detraining. RESULTS Mass-specific [Formula: see text]o 2max , maximal cardiac output, and maximal cardiac stroke volume of all groups decreased after 12 weeks of detraining with no differences among groups. After detraining, arterial-mixed-venous oxygen concentration difference did not decrease in any group, and maximal heart rate decreased in the walk and stall groups. Run time to exhaustion and speeds eliciting [Formula: see text]o 2max and maximal heart rate and at which plasma lactate concentration reached 4mM did not change in the canter group but decreased in the walk and stall groups. CONCLUSIONS AND CLINICAL RELEVANCE Horses following the cantering detraining protocol maintained higher values of several performance variables compared with horses following the walking or stall rest protocols. These results suggested that it may be possible to identify a minimal threshold exercise intensity or protocol during detraining that would promote maintenance of important performance-related variables and minimize reductions in oxygen-transport capacity in horses.

Gait scoring is the most popular method for assessing the walking ability of poultry species. Although inexpensive and easy to implement, gait scoring systems are often criticized for being subjective. Using a treadmill performance test we assessed whether observable differences in Pekin duck walking ability identified using a gait scoring system translated to differences in walking performance. One hundred and eighty ducks were selected using a three-category gait scoring system (GS0 = smooth gait, n = 55; GS0.5 = labored walk without easily identifiable impediment, n = 56; GS1 = obvious impediment, n = 59) and the amount of time each duck was able to sustain walking on a treadmill at a speed of 0.31 m/s was evaluated. The walking test ended when each duck met one of three elimination criteria: (1) The duck walked for a maximum time of ten minutes, (2) the duck required support from the observer's hand for more than three seconds in order to continue walking on the treadmill, or (3) the duck sat down on the treadmill and made no attempt to stand despite receiving assistance from the observer. Data were analyzed in SAS 9.4 using PROC GLM. Tukey's multiple comparison test was used to compare differences in time spent walking between gait scores. Significant differences were found between all gait scores (P < 0.05). Behavioral correlates of walking performance were investigated. Video recorded during the treadmill test was analyzed for counts of sitting, standing, and leaning behaviors. Data were analyzed in SAS 9.4 using a negative binomial model for count data. No differences were found between gait scores for counts of sitting, standing, and leaning behaviors (P > 0.05). In conclusion, the amount of time spent walking on the treadmill corresponded to gait score and was an effective measurement for quantifying Pekin duck walking ability. Background: Noninvasive methods of quantitating exercise tolerance in dogs with neuromuscular disease are needed both for clinical and research use. The 6-minute walk test (6MWT) has been validated as a reliable test of exercise tolerance in dogs with pulmonary and cardiac disease, but not in dogs with neuromuscular disease., Hypothesis/objectives: Distance walked and number of steps taken during 6MWT will differ between Labrador retriever dogs with centronuclear myopathy (CNM) and control (ie, healthy) littermates., Animals: Eight purebred Labrador retrievers were drawn from a purpose-bred research colony (status: 3 clear, 2 carrier, and 3 homozygous mutants for the protein tyrosine phosphatase-like A (PTPLA) gene mutation associated with CNM)., Methods: Pilot, prospective, Case-controlled study. Researchers were blinded to disease status. Each dog was leash-trained and acclimatized to the testing area (length, 12.8 m). At the start of testing, each animal was fitted with a pedometer, a timer was started, and dogs were allowed to walk at their own pace for 6 minutes. Distance walked and pedometer readings were recorded., Results: Degree of paresis varied among affected dogs, and was reflected by significant differences in distance walked between CNM-affected dogs and those with clear and carrier genotypes (P = .048). Pedometer readings did not vary according to genotype (P = .86)., Conclusions: The 6MWT appears to differentiate between the ambulatory capacity of normal and CNM-affected dogs. Additional studies are needed to confirm this relationship in a larger number of dogs, and to evaluate the ability of the 6MWT to differentiate between dogs with variable severity of neuromuscular disease-associated exercise intolerance., (Copyright © 2016 The Authors. Objective: To compare effects of training on conventional and underwater treadmills on fiber properties and metabolic responses of the superficial digital flexor (SDF) and gluteal muscles to high-speed exercise in horses., Sample: 6 unconditioned Quarter Horse-type horses., Procedures: 6 horses were walked on underwater and conventional treadmills for 5 d/wk (maximum, 40 min/d) for 8 weeks in a randomized crossover design (60-day detraining period). Horses underwent a standardized exercise test (SET) at high speed before and after training. Analyte concentrations and fiber characteristics were measured in muscle biopsy specimens obtained from horses before and after each SET., Results: Lactate concentration increased 2- to 3-fold in SDF and gluteal muscle after SETs. No training effect was identified on muscle fiber type composition, type II fiber diameter, muscle analyte concentrations, blood lactate concentration, or heart rate responses. Maximum diameters of type I fibers decreased significantly in gluteal muscle with conventional treadmill training and decreased in SDF muscle with both types of training, with maximum diameters greater for horses after underwater versus conventional treadmill training. No change was identified in minimum fiber diameters., Conclusions and Clinical Relevance: SETs involving near-maximal exertion resulted in an anaerobic response in SDF and gluteal muscles of horses. Eight weeks of conventional or underwater treadmill training resulted in minor changes in type I muscle fiber sizes, with no effect on muscle metabolic or heart rate responses to SETs. After rehabilitation involving underwater treadmills, training at progressing speeds is recommended for horses to develop the required fitness for speed work.

Background: Systolic dysfunction is associated with poor outcomes in dogs with myxomatous mitral valve disease. However, assessment of systolic variables by conventional echocardiographic methods is difficult in these dogs because of mitral regurgitation (MR)., Hypothesis: We hypothesized that assessment of systolic function by dobutamine stress may identify systolic dysfunction in dogs with MR, and that 2-dimensional speckle-tracking echocardiography (2D-STE) could quantitatively evaluate myocardial function., Animals: Anesthetized dogs with experimentally induced MR., Methods: Dogs were examined for systolic myocardial deformations using 2D-STE during dobutamine infusion before and 3 and 6 months after MR induction. We evaluated peak systolic rotation and rotation rate in each basal and apical view; peak systolic torsion and torsion rate were also calculated., Results: Invasive peak positive first derivatives of left ventricular pressure (dp/dt) were significantly decreased in dogs 6 months after induction of MR compared with pre-MR results. After 3 and 6 months of MR, dogs had diminished peak systolic torsion values and torsion rates in response to dobutamine infusion compared with pre-MR results (3 months, P < .001 and P = .006; 6 months, P = .003 and P = .021). These results were significantly correlated with overall invasive dp/dt (r = 0.644, P < .001; r = 0.696, P < .001)., Conclusions and Clinical Importance: Decreased torsion during dobutamine infusion in dogs with MR may reflect latent systolic dysfunction. Background: Idiopathic pulmonary fibrosis (IPF) is an incurable interstitial lung disease occurring mainly in West Highland White Terriers (WHWTs). The effects of IPF on survival and on exercise tolerance in WHWTs are unknown., Objectives: To evaluate survival, prognostic factors, and exercise tolerance in WHWTs with IPF., Animals: Privately owned WHWTs; 15 with IPF and 11 healthy controls., Methods: Prospective case-control study conducted in 2007-2012. For survival, descriptive statistics and Kaplan-Meier (KM) survival curves with Cox proportional hazard ratios were performed. For the prognostic factor study, KM curves, Cox regression analysis, and logistic regression models were used. The 6-minute walk test (6MWT) was used for measurement of exercise tolerance., Results: The median IPF-specific survival of deceased WHWTs (7/15) with IPF was 32 (range 2-51) months from onset of clinical signs. The risk of death from birth in WHWTs with IPF in age-adjusted Cox model was significantly higher (hazard ratio 4.6; 95% confidence interval 1.05-19.74, P = .04) than in control WHWTs. No significant prognostic factors were identified. In 6MWT, WHWTs with IPF walked a shorter distance, median 398 m (range 273-519 m), than healthy controls, median 492 m (420-568 m), P = .05, and the partial pressure of oxygen in arterial blood in diseased dogs had a moderate positive correlation with walking distance (Kendall's tau-b = 0.69, P = .06)., Conclusion and Clinical Importance: IPF had a negative impact on life expectancy, but individual survival varied considerably. 6MWT proved to be a well-tolerated, noninvasive test to evaluate exercise tolerance., (Copyright © 2014 by the American College of Veterinary Internal Medicine.)

Background: Few studies show the detrimental effect of canine obesity on cardiopulmonary function (CPF). The 6-Minute Walk Test (6MWT) is a noninvasive exercise test easy to perform in clinical settings., Objective: The aim of this study was to investigate the effect of obesity and body weight loss (BWL) on CPF assessed by the 6MWT and arterial blood gas analysis., Animals: Six experimental Beagles and 9 privately owned obese dogs were enrolled in a diet-induced BWL program., Methods: Arterial blood gas analysis and 6MWT were repeated in obese subjects (BCS 8-9/9), in the middle of BWL (overweight, BCS 6-7/9), and in lean dogs (BCS 5/9). Heart rate (HRp) and oxygen saturation (SpO2 ) were measured by pulse oximetry before the 6MWT, at midtest, and during a 5-minute recovery period., Results: Twelve dogs completed the BWL program (initial BW, 27.3 ± 2.9 kg; final BW, 20.85 ± 2.9, lsmeans ± SE, P ≤ .001). BWL caused a significant increase in 6MWT walked distance (WD; obese: 509 ± 35 m; overweight: 575 ± 36 m; lean: 589 ± 36 m; P ≤ .05). Resting arterial blood gas results were not influenced by BWL. Including all time points, obese dogs showed higher HRp and lower SpO2 compared to overweight and lean dogs. SpO2 at the end of the walk was significantly lower in obese dogs., Conclusion and Clinical Importance: Obesity negatively affects 6MWT performances in dogs. The 6MWT may be used to demonstrate the efficacy of BWL to improve CPF and quality of life in obese dogs. Although BWL induced significant improvement of cardiopulmonary parameters before ideal BW, WD improved until the end of the BWL program., (Copyright © 2013 by the American College of Veterinary Internal Medicine.)

The novel aim of this study was to describe the reference values of different haematological and biochemical parameters in the Spanish purebred horse (Andalusian, SPB) in each of the stages of a programmed exercise on a treadmill system, and to establish heritability and genetic correlations for these haematological and biochemical parameters. For this, 94 young SPB male horses (4.22 ± 2.27 years old) were used. An increasing intensity exercise test at 4, 5, 6 and 7 m/s was carried out on a treadmill (6% inclination). Total red blood cells, total white blood cells, neutrophils and lymphocytes counts; haematocrit, haemoglobin, lactate, uric acid, creatinine and total plasma proteins concentrations and aspartate transaminase, lactate dehydrogenase, creatine-quinase activities were determined. To conclude: (i) the reference values for each parameter were determined for each of the exercise test stages (ii) all the parameters analysed manifested a medium-high heritability and a high repeatability. These results will, in the near future, determine the measuring guidelines for improving the SPB horse's athletic ability on an objective treadmill system and for selecting these animals in response to those parameters.

Objective: To evaluate the effect of underwater treadmill exercise on static postural sway in horses with experimentally induced carpal joint osteoarthritis under various stance conditions., Animals: 16 horses., Procedures: On day 0, osteoarthritis was induced arthroscopically in 1 randomly selected middle carpal joint of each horse. Beginning on day 15, horses were assigned to either underwater or overground (without water) treadmill exercise at the same speed, frequency, and duration. Two serial force platforms were used to collect postural sway data from each horse on study days -7, 14, 42, and 70. Horses were made to stand stationary on the force platforms under 3 stance conditions: normal square stance, base-narrow placement of the thoracic limbs, and removal of visual cues (blindfolded) during a normal square stance. The mean of 3 consecutive, 10-second trials in each condition was calculated and used for analysis., Results: Displacement of the center of pressure differed significantly depending on the stance condition. Among horses exercised on the underwater treadmill, postural stability in both the base-narrow and blindfolded stance conditions improved, compared with findings for horses exercised on the overground treadmill. Horses exercised on the overground treadmill were only successful at maintaining a stable center of pressure during the normal square stance position., Conclusions and Clinical Relevance: Variations in stance position had profound effects on the mechanics of standing balance in horses with experimentally induced carpal joint osteoarthritis. Underwater treadmill exercise significantly improved the horses' postural stability, which is fundamental in providing evidence-based support for equine aquatic exercise.

Objective: To compare overground and treadmill-based gaits of dogs., Animals: 5 clinically normal adult mixed-breed dogs., Procedures: To obtain dynamic gait data, 30 retroreflective markers were affixed bilaterally to specific regions of the hind limbs and pelvis of each dog. For each dog, 3-D joint motion data (sagittal [flexion and extension], transverse [internal and external rotation], and frontal [abduction and adduction] planes of motion) for the hip, femorotibial, and tarsal joints were acquired during walking and trotting through a calibrated testing space overground or on a treadmill. Comparison of data was performed via generalized indicator function analysis and Fourier analysis., Results: Both overground and treadmill-based gaits produced similar waveforms in all planes of motion. Fourier analysis revealed no difference between overground and treadmill-based gaits in the sagittal plane of motion; however, small differences were detected between overground and treadmill-based gaits in the other 2 planes of motion. Additionally, femorotibial joint motion during walking did not differ among planes of motion. Generalized indicator function analysis was able to detect differences between overground and treadmill-based gait waveforms in all planes of motion for all joints during walking and trotting., Conclusions and Clinical Relevance: In dogs, overground and treadmill-based gaits produced similar waveform shapes. Of the 3 planes of motion evaluated, only sagittal plane kinematic gait data were unaffected by mode of ambulation as determined via Fourier analysis. Sagittal kinematic gait data collected from dogs during overground or treadmill-based ambulation were comparable. However, analysis methods may affect data comparisons.

Objective: To determine the maximum amount of flexion and extension of the carpal, tarsal, metacarpophalangeal, and metatarsophalangeal joints and the percentage duration of the stance and swing phases of the stride for horses walking on an underwater treadmill in various water depths., Animals: 9 healthy adult horses., Procedures: Zinc oxide markers were placed on the forelimbs and hind limbs of the horses. Video was recorded of horses walking (0.9 m/s) on an underwater treadmill during baseline conditions (< 1 cm of water) or in various amounts of water (level of the metatarsophalangeal, tarsal, and stifle joints). Maximum amount of joint flexion and extension, range of motion (ROM), and the percentage durations of the stance and swing phases of the stride were determined with 2-D motion analysis software., Results: The ROM was greater for all evaluated joints in any amount of water versus ROM for joints in baseline conditions (primarily because of increases in amount of joint flexion). The greatest ROM for carpal joints was detected in a tarsal joint water depth, for tarsal joints in a stifle joint water depth, and for metacarpophalangeal and metatarsophalangeal joints in metatarsophalangeal and tarsal joint water depths. As water depth increased, the percentage durations of the stance and swing phases of the stride significantly decreased and increased, respectively., Conclusions and Clinical Relevance: Results of this study suggested that exercise on an underwater treadmill is useful for increasing the ROM of various joints of horses during rehabilitation and that the depth of water affects the amount of flexion and extension of joints.

Horses lose considerably more electrolytes through sweating during prolonged exercise than can be readily replaced through feeds. The present study tested an oral electrolyte supplement (ES) designed to replace sweat electrolyte losses. We measured gastric emptying of 3 litres of ES (using gamma imaging of (99)Tc-sulfide colloid), the absorption of Na(+) and K(+) from the gastrointestinal tract using (24)Na(+) and (42)K(+), and the distribution of these ions in the body by measuring radioactivity within plasma and sweat during exercise. Three litres of ES emptied from the stomach as fast as water, with a half-time of 47 min, and appeared in plasma by 10 min after administration (n = 4 horses). Peak values of plasma (24)Na(+) and (42)K(+) radioactivity occurred at 20-40 min, and a more rapid disappearance of K(+) radioactivity from plasma was indicative of movement of K(+) into cells (n = 3 horses). In a randomized crossover experiment (n = 4 horses), 1 h after administration of placebo (water), 1 or 3 litres of ES containing (24)Na(+), horses exercised on a treadmill at 30% of peak oxygen uptake until voluntary fatigue. The (24)Na(+) appeared in sweat at 10 min of exercise, and when horses received 3 litres of ES the duration to voluntary fatigue was increased in all horses by 33 ± 10%. It is concluded that an oral ES designed to replace sweat ion losses was rapidly emptied from the gastrointestinal tract, rapidly absorbed in the upper intestinal tract and rapidly distributed within the body. The ES clearly served as a reservoir to replace sweat ion losses during exercise, and administration of ES prior to exercise resulted in increased duration of submaximal exercise.

Objective: To evaluate the load redistribution mechanisms in walking and trotting dogs with induced forelimb lameness., Animals: 7 healthy adult Beagles., Procedures: Dogs walked and trotted on an instrumented treadmill to determine control values for peak and mean vertical force as well as vertical impulse for all 4 limbs. A small sphere was attached to the ventral pad of the right forelimb paw to induce a reversible lameness, and recordings were repeated for both gaits. Additionally, footfall patterns were assessed to test for changes in temporal gait variables., Results: During walking and trotting, peak and mean vertical force as well as vertical impulse were decreased in the ipsilateral forelimb, increased in the contralateral hind limb, and remained unchanged in the ipsilateral hind limb after lameness was induced. All 3 variables were increased in the contralateral forelimb during trotting, whereas only mean vertical force and vertical impulse were increased during walking. Stance phase duration increased in the contralateral forelimb and hind limb during walking but not during trotting., Conclusions and Clinical Relevance: Analysis of the results suggested that compensatory load redistribution mechanisms in dogs depend on the gait. All 4 limbs should be evaluated in basic research and clinical studies to determine the effects of lameness on the entire body. Further studies are necessary to elucidate specific mechanisms for unloading of the affected limb and to determine the long-term effects of load changes in animals with chronic lameness.

Objective: To investigate forelimb hoof wall strains and shape changes in unshod horses undergoing regular moderate exercise on a treadmill at selected speeds and gaits., Animals: 6 horses of various body types., Procedures: Each horse was exercised on a treadmill (walking, trotting, and cantering, with or without galloping at 12.5 m/s) 3 times a week for 4 consecutive weeks; duration of each exercise session ranged from 10 to 14 minutes. During the 4-week period, the proximal hoof circumference (PHC) and toe angle (TA) of each forelimb hoof were measured weekly with a flexible measuring tape and a hoof gauge, respectively. Forelimb hoof wall strains were measured bilaterally at the toe and each quarter (3 strain gauges) immediately before the first and after the last exercise session., Results: Strain measurements revealed a consistent pattern of deformation of the hoof wall in both forelimbs at all gaits; strains increased during the stance phase of the stride. Strain values were dependent on site and gait. Compared with initial findings, mean TA increased significantly, whereas mean PHC did not, after the 4-week exercise period. A relationship between TA changes and hoof wall strains could not be established., Conclusions and Clinical Relevance: In unshod horses, forelimb hoof wall strains were affected by site and gait, but not by discrete changes in TA; PHC did not change in response to moderate regular exercise. The pattern of hoof loading was consistent despite significant changes in TA.

The welfare consequences of long-distance transportation of animals remain a controversial topic. Animals that stand for most of the long journey (especially if additional muscular activity is required to deal with postural instability) are at risk of developing fatigue. Previous observational studies of behaviour and physiology suggested either that sheep do not become markedly fatigued by long journeys or that previous methods did not adequately identify fatigue. A range of behavioural and physiological measures were made on eight pairs of sheep during and after treadmill exercise. Within each pair of sheep, a treatment sheep was walked on a treadmill at 0.5 m/s for up to 5 h or until the sheep voluntarily stopped exercising or showed other signs of reduced performance, and a control sheep was exercised for two 10-min periods on either side of the exercise period for the treatment sheep. With the exception of one sheep that only walked for 4.5 h, all treatment sheep walked for 5 h without apparent difficulty. After exercise, the plasma cortisol concentration of treatment sheep was significantly greater than that of control sheep. However, there were no significant treatment effects on plasma creatine kinase activity or blood lactate concentration. After 5 h of exercise, there was a proportionate decrease in the median frequency of the electromyogram recorded over the m. semitendinosus, and this was significantly different from control sheep. There was no evidence that treatment sheep lay down sooner or for longer after treadmill exercise than controls. In sheep tested in a maze to examine whether there was increased motivation to rest after exercise, there was no significant difference between the times taken by treatment and control sheep to obtain a food reward. Qualitative behavioural assessment of the sheep by a panel of observers identified two main dimensions of sheep demeanour, but among descriptors elicited from observers only one person used a term associated with fatigue. No significant difference was found between the scores of treatment and control sheep on these two demeanour dimensions. Thus, there was little evidence that prolonged gentle walking exercise fatigues sheep. Further development of methods to both repeatedly induce and to identify fatigue in sheep is required.

Objective: To identify gait characteristics during trotting on a treadmill in nonlame Labrador Retrievers presumed predisposed or not predisposed to cranial cruciate ligament disease (CCLD)., Animals: Clinically normal Labrador Retrievers presumed predisposed (n = 10) or not predisposed (7) to CCLD., Procedures: The right hind limb of each dog was classified by use of a predictive score equation that combined tibial plateau angle and femoral anteversion angle as presumed predisposed (high score [> -1.5]) or not predisposed (low score [≤ -1.5]) to CCLD. Tarsal joint, stifle joint, and hip joint kinematics, net moments, and powers were computed., Results: The stifle joint was held at a greater degree of flexion in limbs presumed predisposed to CCLD (130.9° vs 139.3°). More power was generated by muscles acting on the stifle joint in the early stance phase of limbs presumed to be predisposed to CCLD (2.93 vs 1.64 W/kg). The tarsal joint did not reach the same degree of extension in limbs presumed predisposed to CCLD, compared with that in limbs presumed not predisposed to CCLD (179.0° vs 161.0°). Velocity, stance time, vertical and craniocaudal forces, angular velocities, and net joint muscle moments did not differ between groups., Conclusions and Clinical Relevance: Gait mechanics of dogs with high (> -1.5) and low (≤ -1.5) tibial plateau angle and femoral anteversion angle scores were characterized on a treadmill, which may help in the identification of dogs predisposed to CCLD.

A field test and a standardized treadmill test were used to assess fitness in endurance horses. These tests discriminated horses of different race levels: horses participating in races of 120 km and more showed higher values of VLA4 (velocity at which blood lactate reached 4 mmol/L) and V200 (velocity at which heart rates reached 200 beats per min) than horses of lower race levels.

The aim of the present study was to evaluate the modifications of some physiological parameters during moderate treadmill exercise in seven healthy Beagle dogs. All animals were submitted to treadmill exercise consisting of walking (15 min), trotting (20 min) and walking (10 min). At every step, rectal temperature (RT) was measured, and the mean heart rate (HR) was assessed. Venous blood samples were collected immediately before starting the treadmill exercise session (at rest), after the end of walking (15 min), trotting (20 min) and walking (10 min), and after 30 min of passive recovery. For immediate assessment of lactate and glucose concentration, blood was analyzed with portable blood lactate and blood glucose analyzers, respectively. Blood was also transferred into sterile glass tubes containing K(3)-ethylenediaminetetraacetic acid (K(3)-EDTA) for evaluation of red blood cells (RBC), white blood cells (WBC), platelets (PLT), hemoglobin (Hb), hematocrit (Hct), mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC). One-way repeated measures analysis of variance (ANOVA) showed a significant effect of treadmill exercise (P<0.05) on RT, HR, lactate, glucose, RBC and Hct. Considering these significant variations, the knowledge of RT, HR, glucose and lactate concentrations, RBC, and Hct, the most suitable and sensitive indicators of response to treadmill exercise in untrained dogs, is essential in order to evidence the individual levels of exercise tolerance, to investigate exercise-related problems and to design specific and individual treadmill protocols.

Hampshire pigs carrying the PRKAG3 mutation in the AMP-activated protein kinase (AMPK) γ3 subunit exhibit excessive skeletal muscle glycogen storage and an altered glycogen synthesis signalling response following exercise. AMPK plays an important role as a regulator of carbohydrate and fat metabolism in mammalian cells. Exercise-trained muscles are repeatedly exposed to glycogen degradation and resynthesis, to which the signalling pathways adapt. The aim of this study was to examine the effect of acute exercise on glycogen synthesis signalling pathways, and the levels of insulin and other substrates in blood in exercise-trained pigs with and without the PRKAG3 mutation. After 5 weeks of training, pigs performed two standardized treadmill exercise tests, and skeletal muscle biopsies were obtained immediately after exercise and 3 h postexercise in the first test, and 6 h postexercise in the second test. The PRKAG3 mutation carriers had higher glycogen storage, and resynthesis of glycogen was faster after 3 h but not after 6 h of recovery. Alterations in the concentrations of insulin, glucose, lactate and free fatty acids after exercise did not differ between the genotypes. The carriers showed a lower expression of AMPK and increased phosphorylation of Akt Ser(473) after exercise, compared with non-carriers. Acute exercise stimulated the phosphorylation of AS160 in both genotypes, and the phosphorylation of GSK3α Ser(21) and ACC Ser(79) in the non-carriers. In conclusion, exercise-trained pigs carrying the PRKAG3 mutation show an altered Akt and AMPK signalling response to acute exercise, indicating that glucose metabolism is associated with faster resynthesis of muscle glycogen in this group.

The aim of this study was to investigate the effects of training and detraining on the monocarboxylate transporter (MCT) 1 and MCT4 levels in the gluteus medius muscle of Thoroughbred horses. Twelve Thoroughbred horses were used for the analysis. For 18 weeks, all the horses underwent high-intensity training (HIT), with running at 90-110% maximal oxygen consumption (VO2 max ) for 3 min, 5 days week(-1). Thereafter, the horses either underwent detraining for 6 weeks by either 3 min of moderate-intensity training (MIT) at 70% VO2 max, 5 days week(-1) (HIT-MIT group) or stall rest (HIT-SR group). The horses underwent an incremental exercise test, VO2 max was measured and resting muscle samples were obtained from the middle gluteus muscle at 0, 18 and 24 weeks. The content of MCT1 and MCT4 proteins increased after 18 weeks of HIT. At the end of this period, an increase was noted in the citrate synthase activity, while phosphofructokinase activity remained unchanged. After 6 weeks of detraining, all these indexes returned to the pretraining levels in the HIT-SR group. However, in the HIT-MIT group, the increase in the MCT1 protein content and citrate synthase activity was maintained after 6 weeks of MIT, while the MCT4 protein content decreased to the pretraining value. These results suggest that the content of MCT1 and MCT4 proteins increases after HIT in Thoroughbred horses. In addition, the increase in the MCT1 protein content and oxidative capacity induced by HIT can be maintained by MIT of 70% VO2 max, but the increase in the MCT4 protein content cannot be maintained by MIT.

Background: The 6-minute walk test (6MWT) is widely used in human medicine to objectively assess the degree of impairment, and to provide objective evidence of disease progression or response to therapy., Hypothesis/objectives: The 6MWT will be easy to perform and well-tolerated in pet dogs. Dogs with pulmonary disease will walk shorter distances than healthy dogs., Animals: Sixty-nine healthy dogs were recruited from the hospital community. Six dogs with mild to moderate pulmonary disease were recruited from animals presented for evaluation at the teaching hospital., Methods: Prospective study. Dogs walked for 6 minutes in a hallway and the distance covered was measured. Pulse oximetry and heart rate were recorded before and after walking. Physical characteristics of the dogs, including age, leg length, body condition score, and weight, were recorded. Healthy dogs were compared with affected dogs by a Student's t-test (P < .05). Correlations were calculated between the age, physical characteristics, and distances walked in the healthy dogs., Results: Healthy dogs walked 522.7 ± 52.4 m, while sick dogs (n = 6) walked 384.8 ± 41.0 m (P < .001). There was low (r = 0.13) to moderate (r= 0.27) correlation in the healthy dogs between physical characteristics and distances walked., Conclusions and Clinical Importance: The 6MWT was easy to perform and discriminated between healthy dogs and dogs with pulmonary disease., (Copyright © 2011 by the American College of Veterinary Internal Medicine.)
