2. Determination of proteins in different feeds using the in sacco method in comparison to the CNCPS method

Abstract

The aim of this study was to determine the correlations between the output parameters of the in sacco method and the CNCPS method (Cornell Net Carbohydrate and Protein System). Both methods were tested on 106 samples from the feeds classified into following categories: extracted meal and cakes, cereals, legumes, maize and alfalfa silage, other silages, DDGS (dried distiller’s grains with solubles), oilseeds and various hays. The effective degradability of crude protein (EDg CP) by the in sacco method varied depending on the type of feed from 44.97% (hays) to 82.59% (alfalfa silage). The lowest degradation parameter of rapidly degraded fraction (a=24.3%) and the highest potentially degraded fraction (b=70.52%) were found in oilseeds. Five nitrogen fractions (A, B1, B2, B3, C) were determined according to the CNCPS method. Fraction A (NPN – non-protein nitrogen) was different in examined feed with the highest in silages (44.95% of the total CP). Fraction B2 represented in tested feeds the highest part of the total CP, (except for silages, legumes, and oilseeds). Fraction B3 ranged from 1.96% (legumes) to 19.25% (hays) of the total CP. The correlation between EDg CP and soluble fractions was low (r=0.5464) in concentrate feeds, the correlation between EDg and soluble fractions was (r=0.6323) also low in forages., Cieľom tohto príspevku bolo stanoviť korelačné vzťahy medzi výstupnými parametrami metódy in sacco a metódy CNCPS (Cornell Net Carbohydrate and Protein System). Metódy in sacco a CNCPS boli testované na rôznych krmivách (n=106). Krmivá boli rozdelené do jednotlivých kategórií: extrahované šroty a výlisky, obilniny, strukoviny, kukuričná a lucernová siláž, ostatné siláže, DDGS (sušené liehovarnícke výpalky s rozpustnými zložkami), olejniny a rôzne seno. Efektívna degradovateľnosť dusíkatých látok (EDg N-látok) metódou in sacco sa pohybovala v závislosti od typu krmiva od 44,97 % (seno) do 82,59 % (lucernová siláž). Najnižší parameter rozpustnej a degradovateľnej frakcie (a) a najvyššia nerozpustná a degradovateľná frakcia (b) bola zistená v olejnatých semenách (70,52 %). Päť dusíkových frakcií (A, B1, B2, B3, C) bolo stanovených podľa metódy CNCPS. Frakcia A (NPN - nebielkovinový dusík) bola v skúmanom krmive odlišná, najvyššia v silážach (44,94 % z celkových dusíkatých látok). Frakcia B2 predstavovala v testovaných krmivách najvyšší podiel z celkových dusíkatých látok, okrem siláží. Frakcia B3 sa pohybovala od 1,96 % (strukoviny) do 19,25 % (seno) z celkových dusíkatých látok. Korelácia pre jadrové krmivá medzi EDg N-látok a rozpustnými frakciami bola slabá (r=0,5464). Podobne to bolo aj u objemových krmív, kde korelácia medzi EDg N-látok a rozpustnými frakciami bola r=0,6323.

6. Effect of Diet Supplementation with Enterococcus DuransED26E/7 and its Durancin ED26E/7 on Growth Performance, Caecal Enzymatic Activity, Jejunal Morphology and Meat Properties of Broiler Rabbits

Abstract

The present study investigates the effects of Enterococcus duransED26E/7 beneficial strain and its enterocin – durancin (Ent) ED26E/7 on selected parameters in rabbits: growth performance, caecal enzymatic activity, jejunal morphometry and meat physico-chemical characteristics. Seventytwo rabbits (aged five weeks, M91 meat line, both sexes) were divided into experimental groups E1 (E. duransED26E/7 strain; dose 500 μL/animal/day, concentration 109CFU/mL) and E2 (durancin EntED26E/7; dose 50 μL/animal/day, with activity 12 800 AU/mL) and control group (C). The additives were administered in drinking water for a period of 21 days. All animals remained in good health during the experiment. The highest body weight gain (increase by 1.5% compared to C) was noted in E1 group during ED26E/7 strain application (P<0.001). Both bioactive compounds positively influenced (reduced) the feed conversion ratio (P<0.001). The ED26E/7 strain and its EntED26E/7 application stimulated the activity of most enzymes tested in the caecum; only the amylolytic and inulolytic activity in E2 group decreased during durancin ED26E/7 addition. Both additives, but mainly the ED26E/7 strain, showed a tendency to improve the jejunal morhological parameters till the end of the experiment (day 42). The meat physico-chemical parameters were not negatively influenced by the application of E. duransED26E/7 strain and its durancin ED26E/7. The diet supplementation with bacteriocinogenic and probiotic E. duransED26E/7 strain and its EntED26E/7 may improve the growth performance, caecal enzymatic activity and jejunal morphometry of rabbits, without any negative effect on rabbit meat quality.

Abstract

Corn contains 72% of DM as starch. In ruminant nutrition corn grain is typically processed prior to feeding because a whole grain has an intact pericarp that prevents bacterial attachment and subsequent digestion of the seed constituents. Most common processing techniques use various combinations of heat, moisture, time, and mechanical action that may affect starch availability by altering starch properties. In the present study influence of pelleting, steam flaking, micronization and extrusion processing methods on water absorption index (WAI), water solubility index (WSI), degree of gelatinization and organic matter digestibility (OMD) of corn in ruminants was examined. All hydrothermal treatments used in this study caused gelatinization of normal semi-crystalline structure of starch granules but in various extents. Low extent of gelatinization was achieved using steam flaking (21,33%) and pelleting (25,47%). Higher extent of gelatinization was achieved using micronization (63.58%) and extrusion (100%). Statistically significant increase in OMD value compared to unprocessed corn was obtained only after extrusion processing indicating that that all nutrients became more available after extrusion.

11. Dairy-Derived and Bacteriocin-Producing Strain Lactiplantibacillus plantarum LP17L/1: An Assessment of Its Safety and Effect Using Broiler Rabbits.

Abstract

Background: Using bacterial (probiotic) strains can influence beneficial health statuses, e.g., through dairy products; however, they must be assessed as safe before consumption; broiler rabbits represent a suitable model for this purpose. This study evaluated the safety and effect (functionality) of the dairy-derived, plantaricin-producing strain Lactiplantibacillus plantarum LP17L/1 using broiler rabbits. The following parameters were assessed to evaluate safety and functionality: microbiota, phagocytic activity (non-specific immunity parameter), blood serum biochemistry, oxidative stress enzyme, growth performance, cecal hydrolytic activity, and jejunal morphometry., Methods: Previously validated methods were used for the analyses. The appropriate administrations approved the experiments., Results: Although only up to 1.0 colony forming unit (CFU)/g (log10) LP17L/1 reached the gastrointestinal tract of the rabbits, the total lactic acid bacteria and amylolytic streptococci were significantly increased ( p < 0.001). The other microbiota were not influenced, meaning there was no negative influence on phagocytic activity, growth parameters, and biochemistry in the analyzed blood serum. Conversely, lower GPx values were measured in the experimental group than in the control group, meaning LP17L/1 did not induce oxidative stress. LP17L/1 caused an increase in villi length to crypt depth ratio, while hydrolytic activity was also significantly increased ( p < 0.01, p < 0.001)., Conclusions: L. plantarum LP17L/1 was confirmed as safe. Regarding the functionality, no negative influence on the microbiota was noted, and the tested parameters were not negatively influenced. Demonstrating that the LP17L/1 strain is safe provides further chances for its industrial application.
