Research on older people’s artistic participation has mainly focused on its benefits. Fewer studies have addressed the antecedents of older people’s artistic participation, especially the barriers to artistic practices, and particularly those related with contextual factors. In this study, we examined from a socioecological perspective which contextual barriers older artists perceive when they are carrying out their artistic practices. We conducted semi-structured individual interviews with 30 older visual artists and craftspeople. We found six themes relating to contextual barriers to the artistic practice: changes in cultural context; value of arts and crafts; dissemination of artistic work; financial difficulties; discrimination against women; and the covid-19 pandemic. Our study expands on previous research on antecedents of artistic participation among older people, and specifically on barriers. Finally, our study suggests the need to decrease these barriers by implementing programmes aimed at older artists maintaining their artistic practices for as long as possible.

Abstract Background FGFR1 regulates cell–cell adhesion and extracellular matrix architecture and acts as oncogene in several cancers. Potential cancer driver mutations of FGFR1 occur in neuroblastoma (NB), a neural crest-derived pediatric tumor arising in sympathetic nervous system, but so far they have not been studied experimentally. We investigated the driver-oncogene role of FGFR1 and the implication of N546K mutation in therapy-resistance in NB cells. Methods Public datasets were used to predict the correlation of FGFR1 expression with NB clinical outcomes. Whole genome sequencing data of 19 paired diagnostic and relapse NB samples were used to find somatic mutations. In NB cell lines, silencing by short hairpin RNA and transient overexpression of FGFR1 were performed to evaluate the effect of the identified mutation by cell growth, invasion and cologenicity assays. HEK293, SHSY5Y and SKNBE2 were selected to investigate subcellular wild-type and mutated protein localization. FGFR1 inhibitor (AZD4547), alone or in combination with PI3K inhibitor (GDC0941), was used to rescue malignant phenotypes induced by overexpression of FGFR1 wild-type and mutated protein. Results High FGFR1 expression correlated with low relapse-free survival in two independent NB gene expression datasets. In addition, we found the somatic mutation N546K, the most recurrent point mutation of FGFR1 in all cancers and already reported in NB, in one out of 19 matched primary and recurrent tumors. Loss of FGFR1 function attenuated invasion and cologenicity in NB cells, whereas FGFR1 overexpression enhanced oncogenicity. The overexpression of FGFR1N546K protein showed a higher nuclear localization compared to wild-type protein and increased cellular invasion and cologenicity. Moreover, N546K mutation caused the failure in response to treatment with FGFR1 inhibitor by activation of ERK, STAT3 and AKT pathways. The combination of FGFR1 and PI3K pathway inhibitors was effective in reducing the invasive and colonigenic ability of cells overexpressing FGFR1 mutated protein. Conclusions FGFR1 is an actionable driver oncogene in NB and a promising therapy may consist in targeting FGFR1 mutations in patients with therapy-resistant NB.

Articles Being socially connected is a universal human need, but a substantial number of older men and women are or become excluded from these connections in later life. Exclusion from social relations (ESR) is unwanted as it undermines people's ability to lead a healthy, active, and independent life. Policies to reduce this form of exclusion have been limited in effectiveness, due in part to a broader lack of knowledge about the dynamics of social exclusion in older ages and the intersection of social exclusion with gender constructions. To advance our understanding of ESR in later life, we develop a heuristic model based on theories and previous empirical studies. Considering the gendered constructing forces of ESR in older age that can potentially lead to loneliness and reduced health and wellbeing, the model identifies individual drivers, such as biopsychosocial conditions, personal standards and life- -course transitions, and macro-level drivers, such as norms and welfare state provisions. This model can serve as a conceptual platform for further theoretical development and empirical study on the gendered construction of ESR in later life. While our focus is on drivers of ESR and its outcomes, potential reversed effects are also discussed.

Abstract Background HIF1A (Hypoxia-Inducible-Factor 1A) expression in solid tumors is relevant to establish resistance to therapeutic approaches. The use of compounds direct against hypoxia signaling and HIF1A does not show clinical efficiency because of changeable oxygen concentrations in solid tumor areas. The identification of HIF1A targets expressed in both normoxia and hypoxia and of HIF1A/hypoxia signatures might meliorate the prognostic stratification and therapeutic successes in patients with high-risk solid tumors. Methods In this study, we conducted a combined analysis of RNA expression and DNA methylation of neuroblastoma cells silenced or unsilenced for HIF1A expression, grown in normoxia and hypoxia conditions. Results The analysis of pathways highlights HIF-1 (heterodimeric transcription factor 1) activity in normoxia in metabolic process and HIF-1 activity in hypoxia in neuronal differentiation process. HIF1A driven transcriptional response in hypoxia depends on epigenetic control at DNA methylation status of gene regulatory regions. Furthermore, low oxygen levels generate HIF1A-dependent or HIF1A-independent signatures, able to stratify patients according to risk categories. Conclusions These findings may help to understand the molecular mechanisms by which low oxygen levels reshape gene signatures and provide new direction for hypoxia targeting in solid tumor.

O direito à liberdade de expressão da identidade sexual e orientação de pessoas idosas que vivem em contextos institucionais é submetido a uma série de desafios específicos, que colocá-o em risco. O objetivo deste artigo é identificar e refletir sobre esses desafios e, refletir sobre as medidas para diminui-los ou eliminá-los. Ele descreve o contexto da violência sexual, o estigma associado com o grupo LGTB, e como este pode ser exacerbado entre os idosos. E, em seguida, discute os medos e expectativas das comunidades LGBT no que diz respeito a cuidados institucionais, bem como atitudes para com os membros deste grupo por parte de profissionais e residentes heterossexuais. Por último, várias propostas para garantir os direitos sexuais das pessoas LGBT que vivem em instituições.

Embryonic stem cells (ESCs) are derived from inner cell mass (ICM) of the blastocyst. In serum/LIF culture condition, they show variable expression of pluripotency genes that mark cell fluctuation between pluripotency and differentiation metastate. The ESCs subpopulation marked by zygotic genome activation gene (ZGA) signature, including Zscan4, retains a wider differentiation potency than epiblast-derived ESCs. We have recently shown that retinoic acid (RA) significantly enhances Zscan4 cell population. However, it remains unexplored how RA initiates the ESCs to 2-cell like reprogramming. Here we found that RA is decisive for ESCs to 2C-like cell transition, and reconstructed the gene network surrounding Zscan4. We revealed that RA regulates 2C-like population co-activating Dux and Duxbl1. We provided novel evidence that RA dependent ESCs to 2C-like cell transition is regulated by Dux, and antagonized by Duxbl1. Our suggested mechanism could shed light on the role of RA on ESC reprogramming.

En el presente artículo analizamos los significados existentes en torno a la participación en prácticas de apoyo colectivo en un grupo de hombres mayores integrantes de la comunidad de Axotlán, Estado de México. La muestra estuvo comprendida por ocho participantes, todos ellos seleccionados de manera intencional, buscando que fueran habitantes de la comunidad, que hubieran nacido antes de 1957 y que contaran con una trayectoria de participación en prácticas de apoyo colectivo dentro de su comunidad. Se realizó una entrevista semiestructurada a cada participante, la cual fue planeada desde la perspectiva de género y del curso de vida, adicionalmente se realizó un periodo de once meses de observación participante dentro de la comunidad. Los datos fueron trabajados a partir del análisis de contenido cualitativo, resaltando la influencia contextual, los procesos de socialización, las prácticas de apoyo colectivo y el compromiso de los entrevistados con su comunidad.

Addressing older people’s social exclusion is a major challenge for contemporary societies. However, policies designed to address it have tended to focus on poverty and unemployment. This paper explores the relationship between social exclusion and political participation from the perspective of those already holding responsible roles within seniors’ organisations. We aim to highlight the impact of later-life social exclusion in relation to politically active older individuals from two diverse socio-political contexts, Australia and Spain. Participants perceived a range of potential barriers for the inclusion of new members and their own continued involvement. These related to practical and resource ssues, beliefs and attitudes towards participation, and organisational and contextual issues. Members’ views of retention of existing members as well as the recruitment of new members highlight the complexity associated with building the diversity and representativeness that organisations need if they are to represent seniors’ views in the policy process.

El presente artículo tiene como objetivo presentar una panorámica actual de la aplicación de un enfoque narrativo al estudio del envejecimiento. En primer lugar se delinea cuál es el objeto de conocimiento de la gerontología narrativa, que se extiende en un doble nivel interrelacionado: la identidad narrativa en la vejez por una parte, y los relatos culturales sobre el envejecimiento por otra. En segundo lugar, se describen las principales metodologías utilizadas en la gerontología narrativa, diferenciando entre aquellas interesadas en el contenido de los relatos (qué se narra) de las interesadas en el proceso de elaboración dialogada de los relatos (cómo se narra). El artículo finaliza con algunas reflexiones sobre el futuro y desafíos a los que se enfrenta la gerontología narrativa.

Objetivo. Este estudio pretende explorar los objetivos vitales en diferentes momentos de la vida y determinar la influencia de la edad y el estado de salud en ellos. Método. En un estudio transversal se compararon tres muestras de voluntarios: una de jóvenes (78 participantes), una de mayores activos (67 participantes) y una de mayores con enfermedades crónicas (65 personas). Los objetivos vitales se obtuvieron mediante una pregunta abierta, cuyas respuestas fueron sometidas a un análisis de contenido. Resultados. Los objetivos vitales fueron similares para mayores sanos y con enfermedades crónicas, con un predominio de objetivos centrados en la salud y una orientación hacia el mantenimiento. En cambio, en los jóvenes la mayoría de objetivos tuvo que ver con el desarrollo y la obtención de nuevas ganancias. Entre los mayores, ni la edad, ni el estado de salud predijeron de manera significativa la probabilidad de mencionar objetivos centrados en la salud. Conclusión. Los resultados sugieren que las expectativas de edad tienen una influencia mayor que las circunstancias de salud concretas en la determinación de los objetivos vitales.

Traditionally, there has been no place for sexuality in older age. However, research has shown that sexuality plays an important role in older people’s life, even in situations such as dementia. The goal of the article is to review the scientific literature regarding the inappropriate sexual behavior that these kind of patients might present. In order to do so, we will firstly address the definition of inappropriate sexual behavior or, more precisely, its multiple definitions. After that, we will deal with other issues such as its prevalence, factors that can cause its appearance, its consequences and some of the available therapeutic options. Finally, in the last section some recommendations for future research will be provided, such as the need to clarify the concept of inappropriate sexual behavior, to find more efficient ways to address this problem, and the desirability of considering sexuality as a human dimension with a high adaptive potential in old age.

In this paper, we argue that the capacity for narrative agency is significantly compromised in individuals with dementia due to at least three factors: (a) Dementia itself, which causes increasing difficulties in constructing and articulating coherent and meaningful stories, and sharing them with others; (b) cultural narratives about dementia, which promote an extremely negative and pessimistic view of those with the disease; and (c) the convergence of these two last factors, which can lead to caregiving interactions that do not support storytelling and can even stop people with dementia from telling stories. We highlight the importance of narrative care, which involves interventions that focus on the person and their unique life narrative. In narrative care, people with dementia are treated not as impaired patients defined by the disease, but as human beings. In doing so, people with dementia can have their own voices back, which is silenced and discredited so many times.

The mutation I.3.2 was previously identified in a FLP/FRT screen of chromosome 2R for conditional growth regulators. Here we report the phenotypic characterization and genetic mapping of I.3.2 by undergraduate students participating in Fly-CURE, a pedagogical program that teaches the science of genetics through a classroom research experience. We find that creation of I.3.2 cell clones in the developing eye-antennal imaginal disc causes a headless adult phenotype, suggestive of both autonomous and non-autonomous effects on cell growth or viability. We also identify the I.3.2 mutation as a loss-of-function allele of the gene centromere identifier ( cid ), which encodes centromere-specific histone H3 variant critical for chromosomal segregation., (Copyright: © 2022 by the authors.)
