Abstract

As a green, safe and eco-friendly solvent, ionic liquids (ILs) have been widely used in organic syntheses. In this paper, polyamide-imides (PAIs) based on different chain length of amino acids were synthesized by two methods, i.e. in the presence of tetrabutyl ammonium bromide (TBAB) as IL and via conventional polycondensation. The obtained results have been compared. It was confirmed that by the polymerization method in the IL, the PAIs can get higher molecular weight and viscosity and better heat resistance and crystallinity after annealing treatment. All polymers showed higher tensile strength (122–134 MPa) and tensile modulus (2.4–3.0 GPa). Among them, the molecular weight of PAIs based on glycine was characterized by gel permeation chromatography.

Abstract

The ionic-complementary self-assembling peptides discovered by Zhang Shuguang have solution-to-gel (sol-gel) transition capacity and one such peptide RADA16 has been commercialized into hemostatic agents. However, their sol-gel transition ability was not obvious because the peptide aqueous solution with a concentration greater than 1% w/v appeared to be thick and viscous. The current report describes PP-type self-assembling peptides. In addition to the ionic-complementary sequence, they have prolines at both ends of the sequence. This feature has led to better solubility, lower viscosity of the peptide solution, and simplified synthesis and purification processes while maintaining the great gelling performance of the ionic-complementary peptides. The PP-type peptides self-assembled into a well-organized nanofiber scaffold as shown by TEM. Among the PP-type peptides, the PRVDP9 sequence peptide was tested as a hemostatic agent and a mucosal elevating agent. The results were comparable to the classic RADA16. The PP-type self-assembling peptides have superior sol-gel transition ability. Therefore, it is predicted that they will be more suitable to be transported through catheters or endoscopes and have higher commercialization potential as compared with the classic self-assembling peptide sequences.

42. Fusing Syntax and Semantics-Based Graph Convolutional Network for Aspect-Based Sentiment Analysis

Abstract

Aspect-based sentiment analysis (ABSA) aims to classify the sentiment polarity of a given aspect in a sentence or document, which is a fine-grained task of natural language processing. Recent ABSA methods mainly focus on exploiting the syntactic information, the semantic information and both. Research on cognition theory reveals that the syntax an*/874d the semantics have effects on each other. In this work, a graph convolutional network-based model that fuses the syntactic information and semantic information in line with the cognitive practice is proposed. To start with, the GCN is taken to extract syntactic information on the syntax dependency tree. Then, the semantic graph is constructed via a multi-head self-attention mechanism and encoded by GCN. Furthermore, a parameter-sharing GCN is developed to capture the common information between the semantics and the syntax. Experiments conducted on three benchmark datasets (Laptop14, Restaurant14 and Twitter) validate that the proposed model achieves compelling performance comparing with the state-of-the-art models.

Abstract

An enzymatic intramolecular asymmetric reductive amination has been developed for the synthesis of chiral 1,4-diazepanes. Several enantiocomplementary IREDs were identified for the synthesis of (R)- and (S)-5-chloro-2-(5-methyl-1,4-diazepan-1-yl)benzo[d]oxazole with high enan- tioselectivity. The catalytic efficiency of (R)-selective IRED from Leishmania major (IR1) and (S)-selective IRED from Micromonospora echinaurantiaca IR25) was 0.027 and 0.962 s(-1) mM(-1), respectively. To further improve the catalytic efficiency of IR1, its double mutant Y194F/D232H was identified by saturation mutagenesis and iterative combinatorial mutagenesis, which exhibited 61-fold in the catalytic efficiency relative to that of wild-type enzyme. The density functional calculations and molecular dynamics simulations provided some insights into the molecular basis for the improved activity of mutant Y194F/D232H. Furthermore, Y194F/D232H and IR25 were applied to access a range of different substituted 1,4-diazepanes with high enantiomeric excess (from 93 to >99%). This study offers an effective method for construction of chiral 1,4-diazepanes of pharmaceutical importance via imine reductase-catalyzed intramolecular reductive amination of the corresponding aminoketones.

Abstract

3-Ketosteroid-Δ1-dehydrogenases (KstDs [EC 1.3.99.4]) catalyze the Δ1-dehydrogenation of steroids and are a class of important enzymes for steroid biotransformations. In this study, nine putative kstD genes from different origins were selected and overexpressed in Escherichia coli BL21(DE3). These recombinant enzymes catalyzed the Δ1-desaturation of a variety of steroidal compounds. Among them, the KstD from Propionibacterium sp. (PrKstD) displayed the highest specific activity and broad substrate spectrum. The detailed catalytic characterization of PrKstD showed that it can convert a wide range of 3-ketosteroid compounds with diverse substituents, ranging from substituents at the C9, C10, C11 and C17 position through substrates without C4-C5 double bond, to previously inactive C6-substituted ones such as 11β,17-dihydroxy-6α-methyl-pregn-4-ene-3,20-dione. Reaction conditions were optimized for the biotransformation of hydrocortisone in terms of pH, temperature, co-solvent and electron acceptor. By using 50 g/L wet resting E. coli cells harboring PrKstD enzyme, the conversion of hydrocortisone was about 92.5% within 6 h at the substrate concentration of 80 g/L, much higher than the previously reported results, demonstrating the application potential of this new KstD.
