Abstract

Objective This study aimed to systematically assess the effect of the metastasis associated lung adenocarcinoma transcript 1 ( MALAT1) long noncoding RNA rs619586 polymorphism on cancer risk. Methods We conducted a literature search of the PubMed, Embase, and China National Knowledge Internet databases to identify relevant studies, and calculated the pooled odds ratios (ORs) and 95% confidence intervals (CIs) for the retrieved studies using RevMan software. Results Nine eligible studies including 5968 cases and 7439 controls were included in the meta-analysis. The pooled results showed that MALAT1 rs619586 polymorphism was significantly associated with cancer risk [(AG + GG) vs. AA: OR = 0.88; GG vs. (AG + AA): OR = 0.64; GG vs. AA: OR = 0.63; AG vs. AA: OR = 0.91; G vs. A: OR = 0.87]. However, subgroup analyses based on ethnicity and cancer type showed significant associations between MALAT1 rs619586 polymorphism and cancer risk in Asians and for cancers other than hepatocellular carcinoma, but not for Caucasians and hepatocellular carcinoma. Conclusions MALAT1 rs619586 polymorphism may play a role in cancer risk. However, further studies are needed to confirm these findings.

Abstract

Objective: MicroRNAs-associated single nucleotide polymorphisms are thought to play an important role in cancer development. We investigated the association of miR-146a rs2910164 polymorphism with the risk of colorectal cancer (CRC) in Chinese population.Materials and Methods: In this hospital-based case-control study of 560 cases and 780 age- and gender-matched healthy controls, the miR-146a rs2910164 polymorphism was genotyped by polymerase chain reaction-restriction fragment length polymorphism.Results: We found that miR-146a rs2910164 polymorphism was not associated with CRC risk. However, the subgroup analysis by gender showed significant associations in male subjects (GG vs. CC: Odds ratios [OR] = 1.54, 95% confidence intervals [CI] = 1.04-2.29, P = 0.03; G vs. C: OR = 1.21, 95% CI = 1.01-1.45, P = 0.04).Conclusion: Our study provides evidence that miR-146a rs2910164 polymorphism may play an important role in susceptibility to CRC in Chinese male population. Abstract

Transcription factor 21 (TCF21) functions as a tumor suppressor and is inactivated in several types of cancer. The purpose of this study is to investigate whether TCF21 genetic polymorphisms(rs2327429 T>C, rs2327430 T>C, rs2327433 A>G, rs12190287 C>G, rs7766238 G>A, rs4896011 T>A) are associated with the risk of breast cancer in Chinese women. Logistic regression analyses showed that TCF21 rs12190287 polymorphism was significantly associated with the reduced risk of breast cancer. Stratified analyses based on pathological type indicated that TCF21 rs12190287 polymorphism was only associated with the reduced risk of infiltrative ductal carcinoma. Real-time quantitative PCR analyses revealed that compared with those carrying rs12190287 CC genotype, subjects with GG genotype had higher expression levels of TCF21 mRNA in normal breast tissues. Furthermore, luciferase activity assay showed that the rs12190287 G allele weakened the binding affinity of hsa-miR-224 to TCF21 3' UTR. These findings suggest that TCF21 rs12190287 polymorphism can regulate TCF21 expression and may serve as a potential marker for genetic susceptibility to breast cancer.
