Few studies have been conducted on spatial and temporal trends in polychlorinated naphthalenes (PCNs) in coastal environments. Here, we describe 18 PCN congeners found in surface and dated sediment samples collected from highly industrialized bays of Korea. Measurable levels of PCN congeners were detected in all sediment samples, suggesting concurrent and historical contamination. The highest PCN concentrations were observed in sediment from rivers, streams, and the inner portions of the bays, which are surrounded by industrial complexes and commercial harbours. CNs 73, 66/67, and 52 were dominant in surface and dated sediment samples. Congener patterns and diagnostic ratios revealed that PCN contamination is originated from combustion processes and the use of polychlorinated biphenyl (PCB) technical mixtures. PCN concentrations in dated sediment increased from the 1980s to the mid 2000s and then decreased to 2015. Although the toxic equivalent (TEQ) levels of PCNs in our study did not exceed sediment quality guidelines proposed by international authorities, the cumulative risks from the TEQ concentrations of polychlorinated dibenzo-p-dioxins, furans, PCBs, and PCNs can be expected for benthic organisms.

Novel aryl hydrocarbon receptor (AhR) agonists were identified in coastal sediments in the Yellow and Bohai Seas by use of a combination of effect-directed analysis (EDA) and in silico prediction. A total of 125 sediments were screened for AhR-mediated potencies using H4IIE-luc bioassay. Great potencies were observed in organic extracts, mid-polar fraction (F2), and subfractions of F2 (F2.6-F2.9) of sediments collected from Nantong, Qinhuangdao, and Yancheng. Less than 15% AhR potencies could be explained by detected dioxin-like PAHs. Full-scan screening analysis was conducted for the more potent fractions using GC-QTOFMS to investigate the presence of unmonitored AhR agonists. A five-step prioritization strategy was applied; 92 candidate compounds satisfied all criteria. Among these chemicals, thirteen were evaluated for AhR efficacy. Six compounds; benz[b]anthracene, 6-methylchrysene, 2-methylbenz[a]anthracene, 1-methylbenz[a]anthracene, 1,12-dimethylbenzo[c]phenanthrene, and indeno[1,2,3-cd]fluoranthene, exhibited significant AhR-mediated efficacies. 1,12-dimethylbenzo[c]phenanthrene and indeno[1,2,3-cd]fluoranthene were identified as novel AhR agonists. Potency balance analysis showed that the six newly identified AhR agonists explained 0.4-100% of the total AhR-mediated potencies determined. Overall, combining EDA and in silico prediction applied in this study demonstrated the benefits of assessing the potential toxic effects of previously unidentified AhR agonists in sediments from the coasts of China and Korea.

Many studies have reported that neonates and infants are exposed to several per- and polyfluoroalkyl substances (PFASs) via breastfeeding; however, these studies have had small sample sizes. This study aimed to determine the concentrations and time-course trend of PFASs in breast milk and identify influencing factors governing PFAS concentrations. Between July and September (2018), 207 low-risk primiparous women were recruited from a lactation counseling clinic in Korea and their breast milk samples were tested for 14 PFASs, including four perfluoroalkyl sulfonic acids. A questionnaire survey, comprising 84 questions covering the women's demographic, obstetrical, dietary, lifestyle, behavioral, and neonatal information, was conducted to investigate associations. Twelve of the 14 PFASs were detectable in breast milk samples. Perfluorooctanesulfonic acid (PFOS), perfluorooctanoic acid (PFOA), and perfluorodecanoic acid were detected in 100% of the samples, followed by perfluorohexanesulfonic acid (detection rate: 87%), perfluorononanoic acid (87%), and perfluorohexanoic acid (73%); the median concentrations were 0.05, 0.10, 0.031, 0.007, and 0.033 ng/mL, respectively. The PFAS concentrations in breast milk measured in our study were higher than those reported in other studies or countries. In 12 years, from 2007 to 18, the mean concentration of PFOA in breast milk increased by approximately three times (278%). The major factors associated with PFAS concentrations in the bivariate association analysis were body mass index; living area (non-metropolitan); neonatal age; and frequency of fish, ice cream, and canned food consumption. In the multiple regression model, fish consumption significantly influenced the PFOS concentrations in breast milk (β = 0.88, p = 0.033). Frequently, fish consumption has been analyzed as the main dietary factor related to PFOS concentration. Our findings suggest the need for a comprehensive cohort study on PFAS exposure and its association with infant health.

Several studies have been conducted among South Korean to investigate effects of high blood mercury (Hg) levels. In this study fetal body burden of Hg in 344 pregnant women were estimated using a physiologically-based pharmacokinetic model (PBPK) and its associative effects on the growth of infants were analyzed. The association between cord blood Hg and growth variables at birth was analyzed based on parameters such as the sex of the newborn, living area, maternal age group, gestation day, maternal body mass index . We investigated the effects of Hg on infant growth through follow-ups, using a non-linear mixed model. The mean Hg levels in maternal and cord blood were 4.47 μg/L and 7.35 μg/L, respectively. Among the subjects, the corresponding fetal body burden for Methylmercury ranged between 26.3-86.9 mg. Cord blood Hg levels positively correlated with length at birth. Furthermore, the high cord blood group showed greater growth rates compared to the low cord blood group. Therefore, we suggest that pregnant women should make efforts to mitigate exposures to Hg, specifically from diet. Further research is suggested to investigate the relationship between the follow-up growth of the infants and Hg levels, considering fish consumption, diet information, and other environmental pollutants.

Despite the extensive usage of synthetic musk compounds (SMCs) and siloxanes in various personal care products (PCPs), trophic magnification of such chemicals in aquatic environments remains unexplored. In June and September 2020, eleven SMCs and nineteen siloxanes were measured in water, sediments, and biota. Samples were collected from two sites where levels were expected to be influenced by the distance from the wastewater treatment plant (WWTP) in the Geum River, Republic of Korea, were expected. High concentrations of SMCs and siloxanes entered through WWTP were measured in water, sediment, and biota at the both sites and both seasons. Synthetic antimicrobials known as parabens, triclosan (TCS), and triclocarban (TCC) are emerging environmental contaminants. Limited studies on these contaminants have been conducted in coastal environments. In our study, parabens, their metabolites, TCS, and TCC were measured in sediment collected along the Korean coast, to investigate contamination status, spatial distribution, and potential health risks to coastal environments. Methyl paraben and 4-hydroxybenzoic acid were detected in all sediment samples, suggesting widespread contamination. Total concentrations of parent parabens, their metabolites, TCS, and TCC ranged from 0.19 to 11.2 (mean: 2.40) ng/g dry weight, 9.65 to 480 (mean: 120) ng/g dry weight, and < limit of quantification (LOQ)-6.10 (mean: 0.41) ng/g dry weight, and from < LOQ-41.0 (mean: 2.78) ng/g dry weight, respectively. The overall contamination of parabens and antimicrobials in sediment was different from that reported for persistent organic pollutants due to different contamination sources among chemical groups. Significant correlation was found among target contaminants in sediment, suggesting the existence of a common source. Total organic carbon (TOC) was significantly correlated with the concentrations of target contaminants, implying a major factor for coastal distribution of parabens and antimicrobials. The concentrations of parabens and TCS measured in sediment did not exceed a hazard quotient (HQ), implying low potential health risks associated with exposure to these contaminants. This is the first study to report the nationwide distribution of parabens, their metabolites, and antimicrobials in the coastal environments of Korea.

Background: Chronic exposure to low-dose persistent organic pollutants (POPs) can induce mitochondrial dysfunction. This study evaluated the association between serum POP concentrations and oxygen consumption rate (OCR) as a marker of mitochondrial function in humans and in vitro cells.Methods: Serum concentrations of organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs) were measured in 323 adults. The OCRs of platelets and peripheral blood mononuclear cells (PBMCs) were assessed in 20 mL of fresh blood using a Seahorse XF analyzer. Additionally, the in vitro effects of Arochlor-1254, β-hexachlorocyclohexane, and p,p´-dichlorodiphenyltrichloroethane at concentrations of 0.1 pM to 100 nM were evaluated in human platelets, human PBMCs, and Jurkat T-cells.Results: The association between serum POP concentrations and OCR differed depending on the cell type. As serum OCP concentrations increased, basal platelet OCR levels decreased significantly; according to the OCP quintiles of summary measure, they were 8.6, 9.6, 8.2, 8.0, and 7.1 pmol/min/μg (P trend=0.005). Notably, the basal PBMC OCR levels decreased remarkably as the serum PCB concentration increased. Urine was used as a part of a human biomonitoring study based on the excretion kinetics of less-persistent contaminants, such as phthalates and bisphenol A (BPA). Despite the advantages of being non-invasive and easy to collect, urine can show a large variability of concentrations of phthalate metabolites and BPA within a person depending on sampling time. Therefore, it is essential to assess the variability of urinary concentrations for comprehensive sampling design in the context of exposure and risk assessments. In this study, 18 phthalate metabolites and eight BPs were measured in all spot urine (n = 401) collected from 12 participants for seven consecutive days to evaluate within- and between-person variabilities. The intraclass correlation coefficients (ICCs) for all spot urines were poor for monomethyl phthalate (ICC: 0.002) and BPA (0.121) but were moderate for monoethyl phthalate (0.514) and monobenzyl phthalate (0.462). Based on the results of di (2-ethylhexyl) phthalate (DEHP) metabolites, the half-life and differences in metabolic capability seem to affect the ICCs. Urinary mono (2-ethylhexyl) phthalate (MEHP), a primary metabolite of DEHP, was suggested as a short-term exposure marker of DEHP in our study. Creatinine- and specific gravity-adjusted concentrations of phthalate metabolites and BPs resulted in increased ICCs, implying requirements for randomly collected spot urine. Most analytes in the first morning voids (FMVs) were correlated significantly with those in the daily composites, suggesting the feasibility of FMVs to estimate the daily exposure dose. This study facilitates a more comprehensive sampling design and data interpretation strategy for human biomonitoring studies.

Novel halogenated flame retardants (HFRs) were introduced to industrial markets as alternatives to legacy brominated FRs (BFRs), such as polybrominated diphenyl ethers (PBDEs). In the present study, PBDEs and their brominated and chlorinated alternatives, novel BFRs (NBFRs) and dechlorane plus (DP), were measured in multiple environmental matrices in a highly industrialized lake in Korea. Legacy and novel HFRs were detected in multiple samples, indicating ubiquitous contamination. Concentrations of HFRs in water and sediment observed in creeks running through machine, textiles, and automobile industrial complexes were significantly higher than those observed in inside and outside of the lake. Higher bioaccumulation levels of HFRs were observed in inshore compared with offshore waters. Results suggest that multi-matrix distribution of legacy and novel HFRs was dependent on the geographical proximity to industrial sources. Compared with previous studies, the highest levels of PBDEs and NBFRs were recorded in water samples on a global scale, implying on-going emissions from industrial activities. Decabromodiphenyl ethane (DBDPE) was a dominant compound in water samples, whereas the concentrations of PBDEs, NBFRs, and DP in sediment were similar to each other. This suggests a shift in consumption from legacy to novel HFRs, preferentially in water environments. A significant declining trend in PBDEs was observed in water and sediment collected between 2008 and 2015, indicating the effectiveness of regulatory actions. Based on their environmental occurrence and bioaccumulation potential, pentabromoethylbenzene and bis(2-ethylhexyl) tetrabromophthalate may pose emerging concerns regarding contamination of aquatic environments.

Due to dilution status of the urine, chemical concentrations measured in spot urine are frequently adjusted using correction factors, such as creatinine, specific gravity (SG), or osmolarity of the urine. Urinary correction factors, however, can be influenced dramatically by physiological changes such as pregnancy. Details about the variation of urine dilution over the course of pregnancy are not well characterized. In the present study, we investigated the variation of urine correction factors over time among the pregnant women of Korea (n = 69) and Thailand (n = 102). Creatinine, SG, and osmolality were determined in the urine samples obtained in each trimester of the participating women, and were compared by sampling time and by nationality. Implication of the variation in these correction factors was studied using phthalate metabolites measured in the urine samples as model chemicals. Urinary correction factors significantly varied across the trimesters especially in Korean pregnant women: urinary creatinine and osmolality were significantly lower in the third trimester (T3) urine than the first trimester (T1) urine. Urinary creatinine and SG of the T3 urine of Korean pregnant women were also significantly lower than those reported from the non-pregnant women who participated in Korean National Environmental Health Survey (KoNEHS) 2015–2017. Among Thai women, however, these correction factors were rather stable across the pregnancy. Differences in ethnicity, or in behavior such as water consumption amount may partly explain the differences. Temporal changes in these urine correction factors influenced the urinary phthalate metabolite concentrations adjusted for dilution, in both Korean and Thai pregnant women. The present observations show that the variations of urinary correction factors should be considered in exposure assessment of urinary chemicals for pregnant women, in order to circumvent potential bias due to physiological changes occurring during pregnancy, and to reduce errors in exposure classification and association.

Cresyl diphenyl phosphate (CDP), as a kind of aryl substituted organophosphate esters (OPEs), is commonly used as emerging flame retardants and plasticizers detected in environmental media. Due to the accumulation of CDP in organisms, it is very important to discover the toxicological mechanism and metabolic process of CDP. Hence, liver microsomes of crucian carps (Carassius carassius) were prepared for in vitro metabolism kinetics assay to estimate metabolism rates of CDP. After 140 min incubation, the depletion of CDP accounted for 58.1%–77.1% (expect 0.5 and 2 μM) of the administrated concentrations. The depletion rates were best fitted to the Michaelis-Menten model (R2 = 0.995), where maximum velocity (Vmax) and Michaelis-Menten constant (Km) were 12,700 ± 2120 pmol min−1·mg−1 protein and 1030 ± 212 μM, respectively. Moreover, the in vitro hepatic clearance (CLint) of CDP was 12.3 μL min−1·mg−1 protein. Log Kow and bioconcentration factor (BCF) of aryl-OPEs were both higher than those of alkyl- and chlorinated-OPEs, indicating that CDP may easily accumulate in aquatic organisms. The results made clear that the metabolism rate of CDP was greater than those of other OPEs detected in liver microsomes in previous research. This paper was first of its kind to comprehensively investigate the in vitro metabolic kinetics of CDP in fish liver microsomes. The present study might provide useful information to understand the environmental fate and metabolic processes of these kinds of substances, and also provide a theoretical basis for the ecological risk assessment of emerging contaminants.

Phthalates and bisphenol A (BPA), widely applied in industrial and consumer products, can affect hormones associated with the human reproductive system. Because the incidence of reproductive diseases is increasing, a comprehensive exposure assessment of phthalates and bisphenol analogues (BPs) is required for childbearing-aged women. Phthalate metabolites and BPs were measured in urine samples collected from 509 childbearing-aged women (20–48 years) in Korea to investigate their current exposure status, profiles, influencing factors, and potential health risks. To date limited information's are available concerning unintentional productions, screening, profiling, and health risks of polychlorinated naphthalenes (PCNs) in ambient environment and occupational environment. Literature reveals that dust is a neglected environmental matrix never measured for PCNs. To our knowledge, this is the first study to investigate the concentrations and health risks of PCNs in indoor dust, air, and blood of major e-waste recycling hubs in Pakistan. Indoor air (n = 125), dust (n = 250), and serum (n = 250) samples were collected from five major e-waste hubs and their vicinity to measure 39 PCN congeners using GC-ECNI-MS. ∑ 39PCN concentrations in indoor air, dust, and serum (worker > resident > children) samples ranged from 7.0 to 9583 pg/m 3, from 0.25 to 697 ng/g, and from 0.15 to 401 pg/g lipid weight, respectively. Predominant PCN congeners in indoor air and dust were tri- and tetra-CNs, while tetra- and penta-CNs were dominant in human serum samples. The higher PCNs contribution was recorded at the recycling units, while the lower was observed at the shops of the major e-waste hubs. Higher contribution of combustion origin CNs in air, dust and human samples showed combustion sources at the major e-waste hubs, while Halowax and Aroclor based technical mixture showed minor contribution in these samples. Mean toxic equivalent (TEQ) concentrations of PCNs were 2.79E +00 pg-TEQ/m 3, 1.60E −02 ng-TEQ/g, 8.11E −01 pg-TEQ/g, 7.14E −01 pg-TEQ/g, and 6.37E −01 pg-TEQ/g for indoor air, dust, and serum samples from workers, residents, and children, respectively. In our study, CNs- 66/67 and −73 in indoor air, dust, and human serum were the great contributors to total TEQ concentrations of PCNs. This first base line data directs government and agencies to implement rules, regulation to avoid negative health outcomes and suggests further awareness in regard of provision of proper knowledge to the target population. Due to lack of data on production, screening, and profiling of PCNs, this is the first study to investigate concentrations of PCNs in indoor dust, air, and blood serum at the major e-waste recycling hubs in Pakistan where crude recycling methods were common and the potential health risks to human via its non-dietary pathways exposure would experience. This study results identified that Tri-to penta-CNs were dominant in the environmental samples and showed that higher PCNs contribution in the environmental samples was recorded by Combustion based activities as compared to technical mixture based CNs.

Background: T-cell immunosenescence, a hallmark of an aging immune system, is potentially linked to the risk of developing cancer and other aging-related diseases. Chronic infection by cytomegalovirus (CMV) has been widely studied as a risk factor for T-cell immunosenescence, but the role of persistent chemicals has never been examined. As a typical example of persistent chemicals, we evaluated whether organochlorine pesticides (OCPs) are related to T-cell immunosenescence in the general population. Methods: Serum concentrations of β-hexachlorocyclohexane, p,p′-DDT, p,p′-DDE, and trans-nonachlor were measured in 95 Korean adults ages 30 to 64 years. T-cell immunosenescence was assessed by the frequencies of CD8+CD57+, CD8+CD28−, CD4+CD57+, and CD4+CD28− T lymphocytes in 20 mL of fresh peripheral blood. Results: The senescence of CD8+ T lymphocytes was the most consistently associated with OCPs. For quartiles of measurements of OCPs, adjusted mean percentages of CD8+CD57+ and CD8+CD28− T lymphocytes in the CD8+ T lymphocyte population were 23.9, 27.6, 31.0, and 38.7 (Ptrend < 0.01) and 25.6, 27.3, 28.0, and 35.5 (Ptrend = 0.02), respectively. When we compared the strength of the associations among OCPs, CMV IgG titer, and age, OCPs showed the strongest association with markers of immunosenescence. Importantly, the association between OCPs and immunosenescence markers was more prominent among participants without known risk factors, such as a young age or low CMV immunoglobulin G titer. Conclusions: Chronic exposure to low-dose OCPs may be a new risk factor for T-cell immunosenescence. Bisphenol A (BPA) is one of the most widely used chemicals in various consumer products. In thermal papers such as receipts and tickets, BPA is used as a heat-activated developer. Cashiers are therefore suspected to be a vulnerable group of exposure to BPA, but neither contribution of receipt handling to the total body burden of BPA among cashiers, nor related health effects are well characterized. Female cashiers (n = 54) were recruited from seven retail shops of a major supermarket chain in Korea, and urinary levels of BPA and metabolic syndrome (MetS) related biomarkers were measured. In order to estimate the contribution of receipt handling to the body burden of BPA, an intervention was designed on the use of gloves: the subjects were asked not to wear gloves during the work for one week, and in the following week, to wear gloves. Urine samples were collected at pre-shift and post-shift for the first two consecutive days in each week, and urinary BPA concentrations were measured. In cashiers without gloves, about a two-fold increase in urinary BPA concentrations was observed after work-shift. When the cashiers wore gloves, however, urinary BPA levels showed no changes. Higher urinary BPA concentrations were associated with greater levels of fasting insulin and insulin resistance. Our observation shows that receipt handling among the cashiers could double the BPA exposure levels at post-shift compared to those at pre-shift, and use of simple protective equipment such as gloves could effectively reduce the BPA exposure levels. Low-dose persistent organic pollutants (POPs), especially organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs), have emerged as a new risk factor for type 2 diabetes. We evaluated whether chronic exposure to low-dose POPs affects insulin secretory function of β-cells in humans and in vitro cells. Serum concentrations of OCPs and PCBs were measured in 200 adults without diabetes. Mathematical model–based insulin secretion indices were estimated by using a 2-h seven-sample oral glucose tolerance test. Insulin secretion by INS-1E β-cells was measured after 48 h of treatment with three OCPs or one PCB mixture. Static second-phase insulin secretion significantly decreased with increasing serum concentrations of OCPs. Adjusted means were 63.2, 39.3, 44.1, 39.3, 39.7, and 22.3 across six categories of a summary measure of OCPs (Ptrend = 0.02). Dynamic first-phase insulin secretion remarkably decreased with increasing concentrations of OCPs among only insulin-sensitive individuals (Ptrend = 0.02); the insulin levels among individuals with high OCPs were ∼30% of those with low OCPs. Compared with OCPs, PCBs showed weaker associations. The decreased insulin secretion by INS-1E β-cells was observed for even 1 pmol/L OCP. The data from human and in vitro cell experiments suggest that chronic exposure to low-dose POPs, especially OCPs, can induce pancreatic β-cell dysfunction.

Although several studies have conducted maternal transfer of individual PBDE congener in experimental animals, there is a paucity of research on differences in maternal transfer of PBDE congeners. The purpose of the study was to investigate and compare placental and lactational transfer of BDE 47, -209 and its metabolites in rat dam-offspring pairs following repeated administration of BDE 47 and -209. 13C-BDE 47, BDE 209 and its debrominated congeners were detected both in dam serum and offspring body, which indicates that PBDEs can be maternally transferred. In addition, BDE 196 and -197 appeared in offspring body earlier than in maternal serum, which suggests that debromination can be occur in offspring body. BDE 209 increased in both dam and offspring while levels of 13C-BDE 47 was not increased in dam serum. 13C-BDE 47 seems to be stored in breast milk rather than in maternal serum, which can be assumed through the drastic increase of the congener in suckling pups. The magnitude of lactational transfer of the administered congeners was greater than that of placental transfer. And 13C-BDE 47 was relatively more transferred to suckling pups than BDE 209 through breastfeeding.

Exposure to endocrine disrupting chemicals is suggested to be responsible for the development or progression of uterine fibroids. However, little is known about risks related to emerging chemicals, such as organophosphate esters (OPEs) and alternative plasticizers (APs). A case-control study was conducted to investigate whether exposures to OPEs, APs, and phthalates, were associated with uterine fibroids in women of reproductive age. For this purpose, the cases (n = 32) and the matching controls (n = 79) were chosen based on the results of gynecologic ultrasonography among premenopausal adult women in Korea and measured for metabolites of several OPEs, APs, and major phthalates. Logistic regression models were employed to assess the associations between chemical exposure and disease status. Factor analysis was conducted for multiple chemical exposure assessments as a secondary analysis. Among OPE metabolites, diphenyl phosphate (DPHP), 2-ethylhexyl phenyl phosphate (EHPHP), and 1-hydroxy-2-propyl bis(1-chloro-2-propyl) phosphate (BCIPHIPP) were detected in >80% of the subjects. Among APs, metabolites of di-isononyl phthalate (DINP) and di(2-propylheptyl) phthalate (DPrHpP) were detected in >75% of the urine samples. The odds ratios (ORs) of uterine fibroids were significantly higher among the women with higher exposures to tris(1,3-dichlom-2-propyl) phosphate (TDCIPP) and tris(2-butoxyethyl) phosphate (TBOEP), di(2-ethylhexyl) terephthalate (DEHTP), DPrHpP, and di-(iso-nonyl)-cyclohexane-1,2-dicarboxylate (DINCH). In addition, urinary concentrations of mono(2-ethyl-5-oxohexyl) phthalate (MEOHP), a sum of five di(2-ethylhexyl) phthalate metabolites (Sigma 5DEHP), and mono(4-methyl-7-hydroxyoctyl) phthalate (OH-MINP) were significantly higher in the cases. In factor analysis, a factor heavily loaded with DPrHpP and DEHP was significantly associated with uterine fibroids, supporting the observation from the single chemical regression model. We found for the first time that several metabolites of OPEs and APs are associated with increased risks of uterine fibroids among pre-menopausal women. Further epidemiological and mechanistic studies are warranted to validate the associations observed in the present study.

This study utilized effect-directed analysis (EDA) combined with full-scan screening analysis (FSA) to identify aryl hydrocarbon receptor (AhR)-active compounds in sediments of inland creeks flowing into Lake Sihwa, South Korea. The specific objectives were to (i) investigate the major AhR-active fractions of organic extracts of sediments by using H4IIE-luc in vitro bioassay (4 h and 72 h exposures), (ii) quantify known AhR agonists, such as polycyclic aromatic hydrocarbons (PAHs) and styrene oligomers (SOs), (iii) identify unknown AhR agonists by use of gas chromatography-quadrupole time-of-flight mass spectrometry (GC-QTOFMS), and (iv) determine contributions of AhR agonists to total potencies measured by use of the bioassay. FSA was conducted on fractions F2.6 and F2.7 (aromatics with log Kow 5–7) in extracts of sediment from Siheung Creek (industrial area). Those fractions exhibited significant AhR-mediated potency as well as relatively great concentrations of PAHs and SOs. FSA detected 461 and 449 compounds in F2.6 and F2.7, respectively. Of these, five tentative candidates of AhR agonist were selected based on NIST library matching, aromatic structures and numbers of rings, and available standards. Benz[b]anthracene, 11H-benzo[a]fluorene, and 4,5-methanochrysene exhibited significant AhR-mediated potency in the H4IIE-luc bioassay, and relative potencies of these compounds were determined. Potency balance analysis demonstrated that these three newly identified AhR agonists explained 1.1% to 67% of total induced AhR-mediated potencies of samples, which were particularly great for industrial sediments. Follow-up studies on sources and ecotoxicological effects of these compounds in coastal environments would be required. A total of 20 organophosphate triesters (OPEs), including seven alkyl-OPEs, three chlorinated (Cl)-OPEs, seven aryl-OPEs, and three oligomeric-OPEs were measured in 341 house dust samples collected from 12 countries during the period 2010–2014. OPEs were ubiquitous in indoor dust, and the total concentrations of OPEs (∑OPEs; sum of 20 OPEs) ranged from 49.4 to 249,000 ng/g dry weight (dw). Generally, Cl-OPEs were the predominant compounds (51% of total) in indoor dust samples, with a median concentration of 800 ng/g, followed by alkyl-OPEs (31%), aryl-OPEs (17%), and oligomeric-OPEs (1%), with median concentrations of 480, 270, and 21.9 ng/g, respectively. ∑OPE concentrations in indoor dust from more industrialized countries (South Korea: median, 31,300; Japan: 29,800; and the United States: 26,500 ng/g dw) were one or two orders of magnitude higher than those from less industrialized countries (Greece: 7140, Saudi Arabia: 5310, Kuwait: 4420, Romania: 4110, Vietnam: 1190, China: 1120, Colombia: 374, India: 276, and Pakistan: 138 ng/g dw). Numerous chemicals have been manufactured through industrial activities and used as consumer products since the late 18th century. Non-target analysis is a new analytical tool to detect many chemicals in environmental samples and to prioritize emerging contaminants. In this study, suspect and non-target analytical methods were optimized using gas chromatography coupled with time-of-flight (GC/TOF) to propose contaminants of emerging concern for the Arctic environment. A suspect analytical method was developed with qualification and qualifier ions, isotopic ratios, and retention times of 215 contaminants including persistent organic pollutants (POPs) to establish an in-house library. Non-target analytical method was also optimized with a deconvoluted ion chromatogram, which is a form that can possibly match the mass spectrum of the NIST library. Multiple environmental samples, such as seawater, air, soil, sediment, sludge, and iceberg, collected from the Arctic region were analyzed with suspect and non-target analysis of GC/TOF after the clean-up procedure with a solid phase extraction (SPE) cartridge. The commonly detected contaminants in the Arctic environmental samples were siloxanes, organophosphate flame retardants, phthalates, synthetic musk compounds, polychlorinated biphenyls, and polycyclic aromatic hydrocarbons. Among them, siloxanes and organophosphate flame retardants were proposed to be contaminants of emerging concerns for the Arctic environment. This is the first report to prioritize emerging contaminants in the Arctic environment with suspect and non-target analysis of GC/TOF.

Few studies have been conducted on the alternatives to legacy flame retardants in avian species worldwide. In this study, polybrominated diphenyl ethers (PBDEs) and alternative flame retardants such as novel brominated flame retardants (NBFRs) and dechlorane plus (DP) were determined in livers of 10 species of birds from Korea to elucidate species-specific accumulation, biological factors that affect accumulation, and bioaccumulation potentials of these contaminants. Among the emerging alternative flame retardants, the highest occurrence was found for bis(2-ethylhexyl)-3,4,5,6-tetrabromo-phthalate (BEHTBP), syn-DP, anti-DP, and decabromodiphenyl ethane (DBDPE). PBDE concentrations (median: 17.1 ng/g lipid wt) measured in our study were within the ranges reported in previous studies, while the concentrations of BEHTBP, BTBPE and DP were greater than those reported earlier. Residential predatory birds showed significantly greater concentrations of PBDEs and NBFRs than migratory predators and passerine birds. The concentrations of PBDEs, BEHTBP, and DP in residential predatory birds were significantly correlated with increasing stable nitrogen isotope ratio (delta N-15), which indicated biomagnification potentials of these contaminants. Our results suggest that the concentrations and accumulation patterns of PBDEs, NBFRs, and DP depend on the feeding habits and migration patterns of avian species. Contamination by per- and polyfluoroalkyl substances (PFASs) is of great concern in global environments. Due to strong regulation of legacy PFASs, emerging PFASs including alternatives and precursors have been introduced to the industrial market. In this study, legacy and emerging PFASs were measured in seawater, sediment, and bivalves collected along the Korean coast to investigate the occurrence, distribution, contamination sources, and bioaccumulation potential of PFASs. Wide concentration ranges of legacy PFASs were detected in multiple environmental samples, indicating widespread contamination. C8-based PFASs (e.g., PFOA and PFOS) were still major contaminants in all of the environmental samples. Some precursors, such as 8:2 fluorotelomer sulfonate (8:2 FTS) and N-ethyl-perfluorooctane sulfonamidoacetic acid (N-EtFOSAA), and perfluoro-2-propoxypropanoic potassium 9-chlorohexadecafluoro-3-oxanonane-1-sulfonate (F-53B), an alternative to PFOS, were detected in sediment or bivalve samples, implying a shift in consumption patterns from legacy to emerging PFASs. The highest concentrations of PFASs in environmental samples were found at the locations near industrial complexes, such as those for the semi-conductor, paper mill, automobile, and metal-plating industry. This result indicates that PFAS contamination is associated with intensive industrial activities in the coastal environment. Matrix-dependent contamination and profiles of PFASs were observed. Seawater was dominated by short-chained PFASs as a prompt reflection of regulation, while the sediment and bivalves were dominated by long-chained PFASs. Carbon-chain length was a major factor governing environmental behavior and bioaccumulation of PFASs. This was the first nation-wide survey on legacy and emerging PFASs in the coastal environment of Korea.

Exposure to persistent organic pollutants (POPs) during pregnancy is associated with a disruption in thyroid hormone balance. The placenta serves as an important environment for fetal development and also regulates thyroid hormone supply to the fetus. However, epigenetic changes of thyroid regulating genes in placenta have rarely been studied. This study was conducted to evaluate the association between several POP concentrations in maternal serum and DNA methylation of thyroid hormone-related genes in the placenta. The placenta samples were collected from 106 Korean mother at delivery, and the promoter methylation of the placental genes was measured by a bisulfite pyrosequencing. The deiodinase type 3 (DIO3), monocarboxylate transporter 8 (MCT8), and transthyretin (TTR) genes were selected as the target genes as they play an important role in the regulation of fetal thyroid balance. Because people are exposed to multiple chemicals at the same time, a multiple-POP model using principal component analysis (PCA) was applied to evaluate the association between the multiple POPs exposure and the epigenetic change in placenta. In addition, a single-POP model which includes one chemical each in the statistical model for association was conducted.Based on the single-POP models, serum concentrations of p,p′-dichlorodiphenyldichloroethylene (p,p′-DDE) and brominated diphenyl ether-47 (BDE-47) were significantly associated with an increase in placental DIO3 methylation, but only among female infants. Among male infants, a positive association between serum p,p′-DDT and MCT8 methylation level was found. According to the multiple-POP models, serum DDTs were positively associated with DIO3 methylation in the placenta of female infants, while a positive association with MCT8 methylation was observed in those of the male infants. Our observation showed that in utero exposure to DDTs may influence the DNA methylation of DIO3 and MCT8 genes in the placenta, in a sexually dimorphic manner. These alterations in placental epigenetic regulation may in part explain the thyroid hormone disruption observed among the newborns or infants followed by in utero exposure to POPs. To estimate realistic exposure to a chemical, the aggregate exposure from multiple consumer products should be considered. A receptor-based aggregate exposure assessment was conducted according to individuals' exposure factors and simultaneous use patterns including co-use and non-use. A product-based aggregate exposure assessment was conducted by product usage rates of population and users’ exposure factors. Two aggregate exposure assessments were compared. Exposure factors for 31 cosmetic products were collected by face-to-face interviews with 1001 members of the Korean population through national representative sampling. The concentrations of phthalates in 214 cosmetic products were analyzed by GC-MS-MS. The average aggregate exposure dose (AED) determined by the receptor-based method for di(2-ethylhexyl)phthalate (DEHP), di-n-butyl phthalate (DnBP), and diethyl phthalate (DEP) were 0.68 ± 0.87, 1.08 ± 5.71, and 2.47 ± 9.05 μg/kg/day, respectively. The cosmetics that contributed most to the receptor-based AED were skin care and body care products for DEHP, nail care products for DnBP, and fragrance and hair care products for DEP. The young female group showed the highest exposure. The product-based aggregate exposure assessment method underestimated high exposure but overestimated average exposure for DnBP and DEP. The receptor-based aggregate exposure assessment method would be used to determine high exposure groups.

Ny-Alesund, one of four permanent settlements on Spitsbergen in Svalbard, is a research town that includes scientific institutes from many countries. Because of daily-used chemicals (e.g., pharmaceutical and personal care products (PPCPs)) used by residents in the area, generated sewage is considered as a point source in the Kongsfjorden. The aim of the present study was to identify and quantify organic pollutants in the effluent and along the shoreline and offshore via target, suspect, and non-target screening using liquid chromatography–high-resolution mass spectrometry. We tentatively identified 30 compounds using the suspect and non-target screening methods in effluent samples from our first visit to the settlement in 2016. Among these, 3 were false positive, 24 were confirmed, and the 3 remaining compounds were not confirmed because of a lack of reference standards. Of the confirmed, 21 were quantifiable and considered target compounds for the 2nd year study. The quantified compounds in the effluent samples in 2017 totaled 17, including PPCPs, pesticides, perfluorinated compounds, and their metabolites. Some of the compounds, such as caffeine, paraxanthine/theophylline, acetaminophen, cetirizine, diethyl toluamide (DEET), and icaridin, were also detected in the receiving seawater. The concentration range was from 4 to 280,000 ng/L in the effluent and 2–98 ng/L in the seawater. Other 24 compounds were tentatively identified in the second-year effluent samples. Five were further confirmed using reference standards. Prioritization was performed on the 47 substances screened in Ny-Alesund using the exposure and toxicity index. As the result, the top seven substances of concern present were perfluorooctanesulfonic acid (PFOS), triphenyl phosphate (TPHP), irbesartan, DEET, acetaminophen, caffeine, and paraxanthine/theophylline. As the effluent was identified as a source of the concerned organic pollutants, an emission reduction strategy should take place for protection of Arctic Fjorden environment.

Early life exposures to lead (Pb) and mercury (Hg) were reported to be associated with various adverse health outcomes. However, limited data was available for urinary Pb and Hg levels in young children and the proportion of children at risk by age, as well as inter- and intra-subject variations of urinary Pb and Hg levels. Therefore, we collected total 491 urine samples from 241 children by urine collection at birth and at intervals of 3 months until 27 months of age for each child (at 10 monitoring time points), measured urinary Pb and Hg levels, and then evaluated the proportion of children at risk by age and the intra-class correlation (ICC) of the urinary Pb and Hg levels. Both the urinary Pb and Hg levels were significantly different according to the monitoring time points (p 0.0001 for both Pb and Hg). The number of children with Hg level over the Human BioMonitoring (HBM) I (7 μg/L) and II (25 μg/L) in the first urine at birth were 3 (2.2%) and 1 (0.7%), respectively, while the urinary samples at the other time points did not show Hg level over HBM I or HBM II. However, the exceedance rate for urinary Pb based on HBM values was not calculated due to unavailable HBM values. Background Several studies have suggested potential links of phthalates to obesity in children and adults. Limited evidence, however, has been available for the relations between diethylhexyl phthalate (DEHP) and obesity-related markers or body mass change in early life. Methods 128 healthy pregnant women were recruited and, after delivery, their newborns’ first urine and umbilical cord blood samples were collected. We measured urinary levels of two DEHP metabolites, mono-(2-ethyl-5-hydroxyhexyl) phthalate (MEHHP) and mono-(2-ethyl-5-oxohexyl) phthalate (MEOHP). We also measured the levels of leptin, total cholesterol and triglyceride (TG) in cord serum, and used them along with weight, length, head circumference and ponderal index (PI, 100 g/cm 3 ) at birth, as obesity-related markers, and estimated the relations between DEHP metabolites and obesity-related markers using generalised linear models. For the evaluation of body mass increase by early life DEHP exposure, body mass index (BMI) z-score change during 3 months after birth by DEHP metabolites in the first urine samples of the newborns were evaluated using logistic regression. Results DEHP exposure was associated with decrease of PI and increase of TG (PI, β=−0.11, p=0.070 and TG, β=0.14, p=0.027), especially for boys (PI, β=−0.13, p=0.021; and TG, β=0.19, p=0.025). Moreover, DEHP exposure was positively associated with body mass increase during 3 months after birth (change of BMI z-scores, OR=4.35, p=0.025). Conclusions Our findings suggest that DEHP exposure may affect body mass change in early life through changes of obesity-related markers.

Concentrations of eight bisphenol analogues (Bps) including BPA, BPS, and BPF were determined in surface waters collected from select rivers in Japan, Korea, China, and India. BPA was found at a concentration in the range of several tens to several hundreds of nanograms per liter in most of the rivers surveyed and some of the highest concentrations (54-1950 ng/L) were found in rivers in Chennai, India. Concentrations of BPF were one to two orders of magnitude higher than those of BPA in river and sea waters collected from Japan, Korea and China, which suggested that BPF is a major contaminant in surface waters in several Southeast Asian countries. BPF concentrations as high as 2850 ng/L were found in the Tamagawa River in Japan. The flux of BPs through riverine discharges into Tokyo Bay was calculated to be approximately 5.5 t per year. Based on the flux estimates and the mass of BPF found in water column and sediment in Tokyo Bay, it was found that BPF degrades faster than BPA in the environment.
