Organic electrochemical transistors (OECTs) are receiving significant attention due to their ability to efficiently transduce biological signals. A major limitation of this technology is that only p-type materials have been reported, which precludes the development of complementary circuits, and limits sensor technologies. Here, we report the first ever n-type OECT, with relatively balanced ambipolar charge transport characteristics based on a polymer that supports both hole and electron transport along its backbone when doped through an aqueous electrolyte and in the presence of oxygen. This new semiconducting polymer is designed specifically to facilitate ion transport and promote electrochemical doping. Stability measurements in water show no degradation when tested for 2 h under continuous cycling. This demonstration opens the possibility to develop complementary circuits based on OECTs and to improve the sophistication of bioelectronic devices.

LCST-type synthetic thermoresponsive polymers can reversibly respond to certain stimuli in aqueous media with a massive change of their physical state. When fluorophores, that are sensitive to such changes, are incorporated into the polymeric structure, the response can be translated into a fluorescence signal. Based on this idea, this thesis presents sensing schemes which transduce the stimuli-induced variations in the solubility of polymer chains with covalently-bound fluorophores into a well-detectable fluorescence output. Benefiting from the principles of different photophysical phenomena, i.e. of fluorescence resonance energy transfer and solvatochromism, such fluorescent copolymers enabled monitoring of stimuli such as the solution temperature and ionic strength, but also of association/disassociation mechanisms with other macromolecules or of biochemical binding events through remarkable changes in their fluorescence properties. For instance, an aqueous ratiometric dual sensor for temperature and salts was developed, relying on the delicate supramolecular assembly of a thermoresponsive copolymer with a thiophene-based conjugated polyelectrolyte. Alternatively, by taking advantage of the sensitivity of solvatochromic fluorophores, an increase in solution temperature or the presence of analytes was signaled as an enhancement of the fluorescence intensity. A simultaneous use of the sensitivity of chains towards the temperature and a specific antibody allowed monitoring of more complex phenomena such as competitive binding of analytes. The use of different thermoresponsive polymers, namely poly(N-isopropylacrylamide) and poly(meth)acrylates bearing oligo(ethylene glycol) side chains, revealed that the responsive polymers differed widely in their ability to perform a particular sensing function. In order to address questions regarding the impact of the chemical structure of the host polymer on the sensing performance, the macromolecular assembly behavior below and above the phase transition temperature was evaluated by a combination of fluorescence and light scattering methods. It was found that although the temperature-triggered changes in the macroscopic absorption characteristics were similar for these polymers, properties such as the degree of hydration or the extent of interchain aggregations differed substantially. Therefore, in addition to the demonstration of strategies for fluorescence-based sensing with thermoresponsive polymers, this work highlights the role of the chemical structure of the two popular thermoresponsive polymers on the fluorescence response. The results are fundamentally important for the rational choice of polymeric materials for a specific sensing strategy. Als Reaktion auf bestimmte äußere Stimuli ändern bestimmte wasserlösliche Polymere reversibel ihren physikalischen Zustand. Dieser Vorgang kann mithilfe von Fluorophoren, die in die Polymerstrukturen eingebaut werden und deren Fluoreszenzeigenschaften vom Polymer¬zustand abhängen, detektiert werden. Diese Idee ist der Ausgangspunkt der vorliegenden Arbeit, die sich damit beschäftigt, wie äußerlich induzierte Änderungen der Löslichkeit solcher Polymere mit kovalent gebundenen Fluorophoren in Wasser in ein deutlich messbares Fluoreszenzsignal übersetzt werden können. Dazu werden photophysikalische Phänomene wie Fluoreszenz-Resonanz¬energie¬transfer und Solvatochromie ausgenutzt. In Kombination mit einem responsiven Polymergerüst wird es möglich, verschiedene Stimuli wie Lösungs¬temperatur oder Ionenstärke, oder auch Assoziation-Dissoziation Reaktionen mit anderen Makromolekülen oder biochemische Bindungs¬reaktionen über die Änderung von Fluorezenz¬farbe bzw. –Intensität autonom mit bloßem Auge zu detektieren. Unter anderem wurde ein wässriger ratiometrischer Temperatur- und Salzsensor entwickelt, der auf der komplexen supramolekularen Struktur eines thermoresponsiven Copolymers und eines thiophenbasierten konjugierten Polyelektrolyts beruht. Die Anbindung solvato¬chromer Fluorophore erlaubte den empfindlichen Nachweis einer Temperatur¬änderung oder des Vorhandenseins von Analyten. Komplexere Phänomene wie das kompetitive Anbinden von Analyten ließen sich hochempfindlich steuern und auslesen, indem gleichzeitig die Sensitivität dieser Polymeren gegenüber der Temperatur und spezifischen Antikörpern ausgenutzt wurde. Überraschenderweise wiesen die hier untersuchten thermoresponsiven Polymere wie poly-N-isopropylacrylamid (pNIPAm) oder poly-Oligoethylenglykolmethacrylate (pOEGMA) große Unterschiede bzgl. ihrer responsiven optischen Eigenschaften auf. Dies erforderte eine ausführliche Charakterisierung des Fluoreszenz- und Aggregationsverhaltens, unter- und oberhalb des Phasenübergangs, im Bezug auf die chemische Struktur. Ein Ergebnis war, dass alle drei Polymertypen sehr ähnliche temperaturabhängige makroskopische Absorptionseigenschaften aufweisen, während sich die Eigenschaften auf molekularer Ebene, wie der Hydratisierungsgrad oder die intermolekulare Polymerkettenaggregation, bei diesen Polymeren sehr unterschiedlich. Diese Arbeit zeigt damit anhand zweier sehr etablierter thermoresponsiver Polymere, nämlich pNIPAm und pOEGMA, das die chemische Struktur entscheidend für den Einsatz dieser Polymere in fluoreszenzbasierten Sensoren ist. Diese Ergebnisse haben große Bedeutung für die gezielte Entwicklung von Polymermaterialien für fluoreszenzbasierte Assays.

From established to emergent technologies, doping plays a crucial role in all semiconducting devices. Doping could, theoretically, be an excellent technique for improving repressively low transconductances in n-type organic electrochemical transistors – critical for advancing logic circuits for bioelectronic and neuromorphic technologies. However, the technical challenge is extreme: n-doped polymers are unstable in electrochemical transistor operating environments, air and water (electrolyte). Here, the first demonstration of doping in electron transporting organic electrochemical transistors is reported. The ammonium salt tetra-n-butylammonium fluoride is simply admixed with the conjugated polymer poly(N,N’-bis(7-glycol)-naphthalene-1,4,5,8-bis(dicarboximide)-co-2,2’-bithiophene-co-N,N’-bis(2-octyldodecyl)-naphthalene-1,4,5,8-bis(dicarboximide), and found to act as a simultaneous molecular dopant and morphology-additive. The combined effects enhance the n-type transconductance with improved channel capacitance and mobility. Furthermore, operational and shelf-life stability measurements showcase the first example of water-stable n-doping in a polymer. Overall, the results set a precedent for doping/additives to impact organic electrochemical transistors as powerfully as they have in other semiconducting devices., Improving electron transport and stability of n-type organic electrochemical transistors (OECTs) is required to realize a commercially-viable technology for bioelectronics applications. Here, the authors report water-stable doped n-type OECTs with enhanced transconductance and record stability.

Organic mixed conductors have garnered significant attention in applications from bioelectronics to energy storage/generation. Their implementation in organic transistors has led to enhanced biosensing, neuromorphic function, and specialized circuits. While a narrow class of conducting polymers continues to excel in these new applications, materials design efforts have accelerated as researchers target new functionality, processability, and improved performance/stability. Materials for organic electrochemical transistors (OECTs) require both efficient electronic transport and facile ion injection in order to sustain high capacity. In this work, we show that the product of the electronic mobility and volumetric charge storage capacity (µC*) is the materials/system figure of merit; we use this framework to benchmark and compare the steady-state OECT performance of ten previously reported materials. This product can be independently verified and decoupled to guide materials design and processing. OECTs can therefore be used as a tool for understanding and designing new organic mixed conductors., Organic materials that support both electronic and ionic transport hold promise for applications in bioelectronics and energy storage. Here, Inal et al. use transistors to quantify the materials performance of organic mixed conductors in terms of the product of charge mobility and volumetric capacitance.

Stem cell osteogenic differentiation is a complex process, associated with a number of events such as the secretion of collagen type I, osteopontin, osteonectin, osteocalcin and Bone Morphogenic Protein 2 (BMP-2). These molecules can be used as markers to monitor stem cell fate while studying the effects of a specific osteogenic differentiation treatment (e.g. electrical stimulation). Currently available techniques, such as the evaluation of the expression levels of specific genes and end-point biochemical assays, do not allow real-time monitoring of cellular processes, therefore overlooking potentially interesting information. This study explores a promising functionalization strategy towards on-line electrical monitoring of stem cell osteogenic differentiation process, using an organic electrochemical transistor (OECT) to detect cytokines of interest, secreted by cells during the osteogenic differentiation process, such as BMP-2. In this work, antibodies against BMP-2 were anchored on the poly(3,4-ethylenedioxythiophene):polystyrene sulfonate (PEDOT:PSS) gate electrode of an OECT. The biofunctionalization process was evaluated using multiple techniques such as atomic force microscopy, electrochemical Raman spectroscopy, quartz crystal microbalance. Electrode properties were assessed by running chronoamperometric studies, as well as by characterizing the PEDOT:PSS thin film resistance to ion flow by electrochemical impedance spectroscopy and OECT performance using transient (AC) measurements. Finally, a proof-of-concept, biosensor measurement was performed to test our functionalization strategy for sensing, proving that the antibody-functionalized OECTs were able to detect recombinant BMP-2 at levels that are comparable to those used for in vitro stimulation of bone regeneration via soluble osteoinductive factors.

This is the first ever demonstration of an accumulation mode OECT for biosensing and n-type polymer in electrocatalysis., The inherent specificity and electrochemical reversibility of enzymes poise them as the biorecognition element of choice for a wide range of metabolites. To use enzymes efficiently in biosensors, the redox centers of the protein should have good electrical communication with the transducing electrode, which requires either the use of mediators or tedious biofunctionalization approaches. We report an all-polymer micrometer-scale transistor platform for the detection of lactate, a significant metabolite in cellular metabolic pathways associated with critical health care conditions. The device embodies a new concept in metabolite sensing where we take advantage of the ion-to-electron transducing qualities of an electron-transporting (n-type) organic semiconductor and the inherent amplification properties of an ion-to-electron converting device, the organic electrochemical transistor. The n-type polymer incorporates hydrophilic side chains to enhance ion transport/injection, as well as to facilitate enzyme conjugation. The material is capable of accepting electrons of the enzymatic reaction and acts as a series of redox centers capable of switching between the neutral and reduced state. The result is a fast, selective, and sensitive metabolite sensor. The advantage of this device compared to traditional amperometric sensors is the amplification of the input signal endowed by the electrochemical transistor circuit and the design simplicity obviating the need for a reference electrode. The combination of redox enzymes and electron-transporting polymers will open up an avenue not only for the field of biosensors but also for the development of enzyme-based electrocatalytic energy generation/storage devices.
