2. Characterization of novel nitazene recreational drugs: Insights into their risk potential from in vitro µ-opioid receptor assays and in vivo behavioral studies in mice

2-Benzylbenzimidazole derivatives or ‘nitazenes’ are increasingly present on the recreational drug market. Here, we report the synthesis and pharmacological characterization of 15 structurally diverse nitazenes that might be predicted to emerge or grow in popularity. This work expands the existing knowledge about 2-benzylbenzimidazole structure-activity relationships (SARs), while also helping stakeholders (e.g., forensic toxicologists, clinicians, policymakers) in their risk assessment and preparedness for the potential next generation of nitazenes. In vitro µ-opioid receptor (MOR) affinity was determined via competition radioligand (3[H]DAMGO) binding assays in rat brain tissue. MOR activation (potency and efficacy) was studied by means of a cell-based β-arrestin 2 recruitment assay. For seven nitazenes, including etonitazene, opioid-like pharmacodynamic effects (antinociception, locomotor activity, body temperature changes) were evaluated after subcutaneous administration in male C57BL/6 J mice. The results showed that all nitazenes bound to MOR with nanomolar affinities, and the functional potency of several of them was comparable to or exceeded that of fentanyl. In vivo, dose-dependent effects were observed for antinociception, locomotor activity, and body temperature changes in mice. SAR insights included the high opioid-like activity of methionitazene, iso-butonitazene, sec-butonitazene, and the etonitazene analogues 1-ethyl-pyrrolidinylmethyl N-desalkyl etonitazene and ethylene etonitazene. The most potent analogue of the panel across all functional assays was α’-methyl etonitazene. Taken together, through critical pharmacological evaluation, this work provides a framework for strengthened preparedness and risk assessments of current and future nitazenes that have the potential to cause harm to users.

Introduction: 2-Benzylbenzimidazole opioids (‘nitazenes’) have become increasingly prevalent on the recreational drug market. We performed pharmacological characterization of various 'prophetic' nitazenes, allowing risk prioritization based on structure-activity relationships. Ethyleneoxynitazene, which we predicted to emerge and which was first found in January 2023, is presented as a case example. Methods: In vitro pharmacological characterization encompassed radioligand binding assays in rat brain tissue and a cell-based µ-opioid receptor activation (MOR-β-arrestin2) assay. Antinociception (hot plate assay), locomotor activity, and thermic effects were evaluated after subcutaneous administration in C57BL/6J mice. Results: Binding assays revealed a Ki of 57.9 nM; only slightly higher than that of etonitazene (38.4 nM). However, ethyleneoxynitazene had a >100-fold lower potency in the MOR-β-arrestin2 assay (EC50, ethyleneoxynitazene=70.0 nM; EC50, etonitazene=0.588 nM). Its efficacy (relative to the reference hydromorphone) was also lower (Emax,ethyleneoxynitazene=187% vs. Emax,etonitazene=254%). The strongly reduced activity was reflected in vivo, with an ED50, antinociception of 11.1 mg/kg and 0.0223 mg/kg for ethyleneoxynitazene and etonitazene, respectively. Hypothermia and locomotor assays revealed the same pattern. Conclusions: The a priori availability of pharmacological data upon the first emergence of ethyleneoxynitazene allowed to rapidly communicate that (compared to other nitazenes) this is likely not the opioid of highest concern. Similarly, pharmacological data for other anticipated nitazenes are readily available.

Summary: Transgender and gender-diverse individuals experience substantial health disparities across the cancer care continuum. Despite well recognized unique healthcare needs, there are barriers in accessing cancer prevention and treatment services, influenced by disadvantages in key social-economic determinants of health which result in worse clinical outcomes, as compared to the general population. The Italian Association of Medical Oncology (AIOM) acknowledges the critical relevance of this issue. The “Assisi Recommendations” here summarize the outcomes of the “AIOM Oncology Ethics Day” dedicated to gender differences in oncology and cancer care of transgender and gender-diverse people. The recommendations generated during a 2-day multidisciplinary discussion address the various aspects of cancer care experience of transgender and gender-diverse people. The promotion of research in this field, through the generation of new evidence and the collection of prospective data, has been identified as a priority action to mitigate these disparities. By acknowledging the challenges of cancer care in transgender and gender-diverse people and recognizing the need for dedicated policy and clinical recommendations, AIOM demonstrates its commitment to improving the health and well-being of all patients with cancer, regardless of their gender identity or any other personal or social circumstances, as part of health-for-all societal vision.

Abstract Background Escalation and de-escalation are a routine part of high-quality care that should be matched with clinical needs. The aim of this study was to describe escalation of care in relation to the occurrence and timing of Pediatric Intensive Care Unit (PICU) admission in a cohort of pediatric inpatients with acute worsening of their clinical condition. Methods A monocentric, observational cohort study was performed from January to December 2018. Eligible patients were children: 1) admitted to one of the inpatient wards other than ICU; 2) under the age of 18 years at the time of admission; 3) with two or more Bedside-Paediatric-Early-Warning-System (BedsidePEWS) scores ≥ 7 recorded at a distance of at least one hour and for a period of 4 h during admission. The main outcome -the 24-h disposition – was defined as admission to PICU within 24-h of enrolment or staying in the inpatient ward. Escalation of care was measured using an eight-point scale—the Escalation Index (EI), developed by the authors. The EI was calculated every 6 h, starting from the moment the patient was considered eligible. Analyses used multivariate quantile and logistic regression models. Results The 228 episodes included 574 EI calculated scores. The 24-h disposition was the ward in 129 (57%) and the PICU in 99 (43%) episodes. Antibiotic resistance has become more and more widespread over the recent decades, becoming a major global health problem and causing colistin to be increasingly used as an antibiotic of last resort. Acinetobacter baumannii, an opportunistic pathogen that has rapidly evolved into a superbug exhibiting multidrug-resistant phenotypes, is responsible for a large number of hospital infection outbreaks. With the intensive use of colistin, A. baumannii resistance to colistin has been found to increase significantly. In previous work, we identified a deflazacort derivative, PYED-1 (pregnadiene-11-hydroxy-16,17-epoxy-3,20-dione-1), which exhibits either direct-acting or synergistic activity against Gram-positive and Gram-negative species and Candida spp., including A. baumannii. The aim of this study was to evaluate the antibacterial activity of PYED-1 in combination with colistin against both A. baumannii planktonic and sessile cells. Furthermore, the cytotoxicity of PYED-1 with and without colistin was assessed. Our results show that PYED-1 and colistin can act synergistically to produce a strong antimicrobial effect against multidrug-resistant populations of A. baumannii. Interestingly, our data reveal that PYED-1 is able to restore the efficacy of colistin against all colistin-resistant A. baumannii isolates. This drug combination could achieve a much stronger antimicrobial effect than colistin while using a much smaller dosage of the drugs, additionally eliminating the toxicity and resistance issues associated with the use of colistin.

In recent years, multimodal biometric systems are increasingly employed in many application field due to several advantages in terms of universality, recognition rate. and security. Among various acquisition technologies, Ultrasound shows important merits, because it allows obtaining volumetric images of the human body and hence a more accurate description of characteristics and to verify liveness. In this work, a multimodal ultrasound recognition system based on the fusion between 3D hand geometry and 3D palmprint features is proposed and experimentally evaluated. The system acquires a volumetric image of the whole hand and for both characteristics, several 2D images are extracted at different depth levels. From each image, 2D features are extracted and then properly combined to achieve a 3D template. Recognition performances are evaluated through verification and identification experiments by employing a homemade database. Experiments are carried out first for the two unimodal biometrics and successively, by fusing the two modalities at score level. Results have shown that fusion is able to dramatically improve the recognition performances of the single biometrics, achieving an Equal Error Rate of 0.08% and an identification rate of 100%.

Belantamab-mafodotin is an innovative and selective treatment for multi-refractory/relapsed multiple myeloma (MM) patients; however, available real-life experiences on efficacy and safety are limited. In this real-world multicentric retrospective study, we enrolled 28 MM patients treated in four Hematology units of Campania region, Italy, who received a median of six treatment lines prior to belantamab-mafodotin. The overall response rate (ORR) was 40% (complete remission, CR, 11%; very good partial remission, VGPR, 11%; and partial remission, PR, 18%), with a median progression-free survival (PFS) and overall survival (OS) of 3 and 8 months, respectively. One of the most frequent drug-related adverse events was keratopathy observed in nine (32%) patients, leading to therapy discontinuation in only three (11%) of them. Moreover, 22 out of 28 total patients who were treated with at least two administrations achieved an ORR of 50% (CR, 14%; VGPR, 14%; and PR, 22%) with a median PFS and OS of 5 and 11 months, respectively. In conclusion, our multicentric study confirmed efficacy and safety of belantamab-mafodotin in triple-refractory MM patients even in the real-life setting.

18. A Feasible 3D Ultrasound Palmprint Recognition System for Secure Access Control Applications

Biometric recognition systems based on 3D palmprint captured with optical technology have been widely investigated in the last decade; however, they can provide information about the external skin surface only. This limit can be overcome by Ultrasound, which allows gaining information on the depth of palm lines and can verify the liveness of the sample, making the recognition systems very hard to fake. In this work, a feasible palmprint recognition system based on 3D ultrasound images is proposed. Unlike previous wet setups, the coupling between probe and human is realized through a gel pad, which permits a comfortable and precise positioning of the hand by the user. Collected 3D images are processed to generate 2D palmprint images at various under-skin depths. 2D features are then extracted from these images, experimenting with different procedures, and are merged to define a 3D template that contains lines’ depth information. Recognition performances were evaluated by performing verification and identification experiments on a home-made database composed of 423 samples from 55 volunteers. An EER rate of 0.36% and an identification accuracy of 100% are obtained. The suitability of the proposed system in secure access control applications is finally discussed.

The second half of the nineteenth century saw a proliferation of translations from romance languages into Romanian as a consequence of the economic and cultural development of the Romanian society. In this context, 1893 saw the publication of the first Romanian translation of De Amicis’s Cuore (Heart): An Italian Schoolboy’s Journal, by Clelia Bruzzesi (1836-1903). The twentieth century brought five other versions, two of them signed by women translators: Sofia Nădejde (1856-1946) in 1916 and Mia Frollo (1885-1962) in 1936. Until recently, Cuore was part of the primary school curriculum in Romania, so the text left a mark on the cultural history of the country. This paper aims to raise awareness of the often neglected translation activity of women and to reveal Bruzzesi, Nădejde and Frollo’s place in society as well their motivations and the public reception of their translations.

20. Recognition Performance Analysis of a Multimodal Biometric System Based on the Fusion of 3D Ultrasound Hand-Geometry and Palmprint

Multimodal biometric systems are often used in a wide variety of applications where high security is required. Such systems show several merits in terms of universality and recognition rate compared to unimodal systems. Among several acquisition technologies, ultrasound bears great potential in high secure access applications because it allows the acquisition of 3D information about the human body and is able to verify liveness of the sample. In this work, recognition performances of a multimodal system obtained by fusing palmprint and hand-geometry 3D features, which are extracted from the same collected volumetric image, are extensively evaluated. Several fusion techniques based on the weighted score sum rule and on a wide variety of possible combinations of palmprint and hand geometry scores are experimented with. Recognition performances of the various methods are evaluated and compared through verification and identification experiments carried out on a homemade database employed in previous works. Verification results demonstrated that the fusion, in most cases, produces a noticeable improvement compared to unimodal systems: an EER value of 0.06% is achieved in at least five cases against values of 1.18% and 0.63% obtained in the best case for unimodal palmprint and hand geometry, respectively. The analysis also revealed that the best fusion results do not include any combination between the best scores of unimodal characteristics. Identification experiments, carried out for the methods that provided the best verification results, consistently demonstrated an identification rate of 100%, against 98% and 91% obtained in the best case for unimodal palmprint and hand geometry, respectively.

This paper comprises results concluding the research activity done so far regarding enhanced web services and system integration. The objective of the paper is to define the software architecture for a coherent framework and methodology for enhancing existing web services into an integrated system. This document presents the research work that has been done so far in this aspect by applying the proposed architecture for system integration in the academic field. The basics concepts used are Enterprise Application Integration (EAI) and Service Oriented Architecture (SOA), as the most commonly used approaches to information systems integration lately. SOA is an Information Technology (IT) architectural style that supports the transformation of businesses into a set of linked services, or repeatable business tasks that can be accessed when needed over a network. This may be a local network, it may be the Internet, or it may be geographically and technologically diverse, combining services in New York, Paris, and Beijing as though they were all installed on a local desktop. These services can coalesce to accomplish a specific business task, enabling businesses to adapt quickly to changing conditions and requirements. (Contains 6 figures.)

Recognition systems based on palm veins are gaining increasing attention as they are highly distinctive and very hard to counterfeit. Most popular systems are based on infrared radiation; they have the merit to be contactless but can provide only 2D patterns. Conversely, 3D patterns can be achieved with Doppler or photoacoustic methods, but these approaches require too long of an acquisition time. In this work, a method for extracting 3D vascular patterns from conventional grayscale volumetric images of the human hand, which can be collected in a short time, is proposed for the first time. It is based on the detection of low-brightness areas in B-mode images. Centroids of these areas in successive B-mode images are then linked through a minimum distance criterion. Preliminary verification and identification results, carried out on a database previously established for extracting 3D palmprint features, demonstrated good recognition performances: EER = 2%, ROC AUC = 99.92%, and an identification rate of 100%. As further merit, 3D vein pattern features can be fused to 3D palmprint features to implement a costless multimodal recognition system.

Background and aims. Scedosporium species and Lomentospora prolificans are the filamentous fungi isolated more frequently from the cystic fibrosis (CF) lower airways, after Aspergillus fumigatus. Previous studies showed that, in CF patients, Scedosporium species and Lomentospora prolificans are responsible for airways colonization/ infection, enhancing pulmonary deterioration, and for severe invasive infections in CF patients undergoing lung transplantation. Aims of our project were to evaluate the prevalence of Scedosporium and Lomentospora species in Italian patients with CF, and to evaluate the interest of the Scedosporium-selective colture medium SceSel compared to routinely used agar-based culture media, in order to improve laboratory diagnostic performances. Materials and methods. A total of 1977 sputum samples from 1154 CF patients were collected and processed according to CF Italian recommendations in the year 2017 (January 1st to December 31st). The SceSel agar was used in addition to the routine culture procedures to burst growth and isolation of Scedosporium/ Lomentospora species. The fungal isolates were identified by classical mycological methods and polymerase chain reaction-based DNA sequencing of ITS1 and ITS4 regions. Susceptibility of the isolates to antifungal drugs was investigated by E-test method. Results. Among the 1154 enrolled patients, 62 (5.3%) were positive for Scedosporium/Lomentospora species (median age: 27y; range 8-64y; 33 male). Out of 1977 samples, 93 were positive for Scedosporium/Lomentospora species, and 13 (13.9%) were recovered only on SceSel agar. According to molecular analyses, isolation rates of each species were: S. apiospermum 53.1%, S. boydii 37.5%, L. prolificans 6.3% and S. aurantiacum 3.1%. Amphotericin B minimum inhibitory concentration (MIC) values were above or equal to 2 mg/L in all strains. Voriconazole MIC values were below or equal to 0.38 mg/L in almost all strains, proving this drug to be the most effective antimycotic for members of Scedosporium species. Otherwise, Lomentospora prolificans showed to be resistant to all considered antifungal drugs, only Voriconazole seems to be active on some Lomentospora prolificans isolates. Conclusions. Our results suggest that SceSel agar should be used in combination with routine media as standard microbiological protocol and procedures are not always adequate to isolate Scedosporium and Lomentospora species in CF respiratory samples. Molecular identification and susceptibility tests are needed, especially for strains isolated from critical patients and those considered for transplantation, as they might develop invasive scedosporiosis.

Pediatric acute myeloid leukemia is a clonal disorder characterized by malignant transformation of the hematopoietic stem cell. The incidence and the outcome remain inferior when compared to pediatric ALL, although prognosis has improved in the last decades, with 80% overall survival rate reported in some studies. The standard therapeutic approach is a combined cytarabine and anthracycline-based regimen followed by consolidation with allogeneic stem cell transplantation (allo-SCT) for high-risk AML and allo-SCT for non-high-risk patients only in second complete remission after relapse. In the last decade, several drugs have been used in clinical trials to improve outcomes in pediatric AML treatment.

The elicitation of plant secondary metabolism may offer interesting opportunities in the framework of sustainable approaches in plant science and in terms of their ability to prime resistance to biotic and abiotic stressors. The broad metabolic reprogramming triggered by different molecular elicitors, namely salicylate (SA), polyamines (PAs), and chitosan, was comprehensively investigated using a metabolomics approach and the tomato (Solanum lycopersicum L.) as the model crop. Six different treatments were compared: a negative control (no treatments), a second negative control treated with 1 M acetic acid (the reference for chitosan, since chitosan was solubilized in acetic acid), and four molecular elicitors, 1 mM 2,1,3-benzothiadiazole (BTH, a positive control), 10 mg/mL chitosan, 0.01 mM SA, and a 0.1 mM PA (putrescine, spermidine, and spermine). All treatments determined a slight increase in biomass, in particular following PA treatment. A broad reprogramming of secondary metabolism could be observed, including membrane lipid remodeling, phenylpropanoid antioxidants, and phytohormone crosstalk. Overall, our results suggest that PAs, SA, and BTH shared a systemic acquired resistance (SAR)-related response, whereas chitosan induced a more distinct induced systemic resistance (ISR)-like jasmonate-related response. These results pave the way towards the possible use of elicitors as a sustainable tool in plant science and agriculture by increasing crop resilience to biotic and abiotic stressors without detrimental effects on plant biomass.

Invasive Candida infections have become a global public health problem due to the increase of Candida species resistant against antifungal therapeutics. The glucocorticoid PYED-1 (pregnadiene-11-hydroxy-16α,17α-epoxy-3,20-dione-1) has antimicrobial activity against various bacterial taxa. Consequently, it might be considered for the treatment of Candida infections. The antifungal activity of PYED-1 was evaluated against several fungal strains that were representative of the five species that causes the majority of Candida infections—namely, Candida albicans, Candida glabrata, Candida tropicalis, Candida parapsilosis and Candida krusei. PYED-1 exhibited a weak antifungal activity and a fungistatic effect on all five Candida species. On the other hand, PYED-1 exhibited a good anti-biofilm activity, and was able to eradicate the preformed biofilms of all Candida species analyzed. Moreover, PYED-1 inhibited germ tube and hyphae formation of C. albicans and reduced adhesion of C. albicans to abiotic surfaces by up to 30%.

Arbuscular mycorrhiza and Trichoderma are well-known beneficial fungi whose plant growth promotion and defense elicitation effects are known. However, the molecular and biochemical processes underlying the beneficial effects of these priming microorganisms have not been fully elucidated yet. On this basis, the present work aimed to use metabolomics to dissect comprehensively the modulation of secondary metabolism induced by mycorrhiza and Trichoderma, using tomato as a model plant. To this aim, either mycorrhiza or Trichoderma were applied to tomato roots at transplanting using a commercial formulation and then harvested once the mutualistic relationship was well established. Shoots were analyzed using an MS-based untargeted metabolomics approach, and differential metabolites identified by multivariate statistics were subjected to pathway analysis. Together with promoting plant growth, the treatments induced a broad molecular reprogramming with the phenylpropanoid biosynthetic pathway (including defense phenolics like coumarins and glycosylated anthocyanins) being strongly elicited. An accumulation of auxins, cytokinins, and jasmonate (especially after treatment with Trichoderma) could be observed concerning phytohormone profiles. Overall, the broad and distinctive effects triggered by mycorrhiza and Trichoderma in tomato secondary metabolism supported both plant growth promotion and immunity.

Staphylococcus aureus is one of the major causes of hospital- and community-associated bacterial infections throughout the world, which are difficult to treat due to the rising number of drug-resistant strains. New molecules displaying potent activity against this bacterium are urgently needed. In this study, d- and l-deoxynojirimycin (DNJ) and a small library of their N-alkyl derivatives were screened against S. aureus ATCC 29213, with the aim to identify novel candidates with inhibitory potential. Among them, N-nonyloxypentyl-l-DNJ (l-NPDNJ) proved to be the most active compound against S. aureus ATCC 29213 and its clinical isolates, with the minimum inhibitory concentration (MIC) value of 128 μg/mL. l-NPDNJ also displayed an additive effect with gentamicin and oxacillin against the gentamicin- and methicillin-resistant S. aureus isolate 00717. Sub-MIC values of l-NPDNJ affected S. aureus biofilm development in a dose-dependent manner, inducing a strong reduction in biofilm biomass. Moreover, real-time reverse transcriptase PCR analysis revealed that l-NPDNJ effectively inhibited at sub-MIC values the transcription of the spa, hla, hlb and sea virulence genes, as well as the agrA and saeR response regulator genes.

In this work, the antibacterial activity of deflazacort and several of its synthetic precursors was tested against a panel of bacterial pathogens responsible for most drug-resistant infections including Staphylococcus aureus, Enterococcus spp., Acinetobacter baumannii, Pseudomonas aeruginosa, Klebsiella pneumoniae, Escherichia coli, and Enterobacter spp. The derivative of deflazacort, PYED-1 (pregnadiene-11-hydroxy-16α,17α-epoxy-3,20-dione-1) showed the best antibacterial activity in a dose-dependent way. We focused on the action of PYED-1 against S. aureus cells. PYED-1 exhibited an additive antimicrobial effect with gentamicin and oxacillin against the methicillin-resistant S. aureus isolate 00717. In addition to its antimicrobial effect, PYED-1 was found to repress the expression of several virulence factors of S. aureus, including toxins encoded by the hla (alpha-haemolysin), hlb (beta-haemolysin), lukE-D (leucotoxins E-D), and sea (staphylococcal enterotoxin A) genes, and cell surface factors (fnbB (fibronectin-binding protein B) and capC (capsule biosynthesis protein C)). The expression levels of autolysin isaA (immunodominant staphylococcal antigen) were also increased.

Background . Insulin resistance is associated with higher breast cancer (BC) penetrance in BRCA mutation carriers. Metabolic syndrome (MetS), an insulin resistance syndrome, can be reversed by adhering to the Mediterranean diet (MedDiet). In a dietary intervention trial on BRCA mutation carriers, we evaluated adherence to the MedDiet, and the association with the MetS, by analyzing data from the Mediterranean Diet Adherence Screener (MEDAS). Methods. BRCA mutation carriers, with or without BC, aged 18 to 70 years, were eligible for the trial. After the baseline examinations, women were randomized to a dietary intervention or to a control group. Both groups completed the MEDAS at baseline and at the end of the dietary intervention. Results . A total of 163 women completed the 6 months of dietary intervention. Compared with controls, the women in the intervention group significantly reduced their consumption of red meat ( P < .01) and commercial sweets ( P < .01) and their MEDAS score rose significantly (+1.3 vs +0.55, P = .02). The number of MetS parameters decreased with increasing points of adherence to the MEDAS score ( P = .01). In the intervention group, there was a significant association with the greater reduction of MetS. Conclusion. BRCA mutation carriers in the intervention group experienced greater improvement in their MedDiet and MetS parameters.

37. Autophagy Mediates Interleukin-1β Secretion in Human Neutrophils

Interleukin-1β (IL-1β), a major pro-inflammatory cytokine, is a leaderless cytosolic protein whose secretion does not follow the classical endoplasmic reticulum-to-Golgi pathway, and for which a canonical mechanism of secretion remains to be established. Neutrophils are essential players against bacterial and fungi infections. These cells are rapidly and massively recruited from the circulation into infected tissues and, beyond of displaying an impressive arsenal of toxic weapons effective to kill pathogens, are also an important source of IL-1β in infectious conditions. Here, we analyzed if an unconventional secretory autophagy mechanism is involved in the exportation of IL-1β by these cells. Our findings indicated that inhibition of autophagy with 3-methyladenine and Wortmannin markedly reduced IL-1β secretion induced by LPS + ATP, as did the disruption of the autophagic flux with Bafilomycin A1 and E64d. These compounds did not noticeable affect neutrophil viability ruling out that the effects on IL-1β secretion were due to cell death. Furthermore, VPS34IN-1, a specific autophagy inhibitor, was still able to reduce IL-1β secretion when added after it was synthesized. Moreover, siRNA-mediated knockdown of ATG5 markedly reduced IL-1β secretion in neutrophil-differentiated PLB985 cells. Upon LPS + ATP stimulation, IL-1β was incorporated to an autophagic compartment, as was revealed by its colocalization with LC3B by confocal microscopy. Overlapping of IL-1β-LC3B in a vesicular compartment peaked before IL-1β increased in culture supernatants. On the other hand, stimulation of autophagy by cell starvation augmented the colocalization of IL-1β and LC3B and then promoted neutrophil IL-1β secretion. In addition, specific ELISAs indicated that although both IL-1β and pro-IL-1β are released to culture supernatants upon neutrophil stimulation, autophagy only promotes IL-1β secretion. Furthermore, the serine proteases inhibitor AEBSF reduced IL-1β secretion. Moreover, IL-1β could be also found colocalizing with elastase, suggesting both some vesicles containing IL-1β intersect azurophil granules content and that serine proteases also regulate IL-1β secretion. Altogether, our findings indicate that an unconventional autophagy-mediated secretory pathway mediates IL-1β secretion in human neutrophils.

Biometric recognition systems are finding applications in more and more civilian fields because they proved to be reliable and accurate. Among the other technologies, ultrasound has the main merit of acquiring 3D images, which allows it to provide more distinctive features and gives it a high resistance to spoof attacks. This work reviews main research activities devoted to the study and development of ultrasound sensors and systems for biometric recognition purposes. Several transducer technologies and different ultrasound techniques have been experimented on for imaging biometric characteristics like fingerprints, hand vein pattern, palmprint, and hand geometry. In the paper, basic concepts on ultrasound imaging techniques and technologies are briefly recalled and, subsequently, research studies are classified according to the kind of technique used for collecting the ultrasound image. Overall, the overview demonstrates that ultrasound may compete with other technologies in the expanding market of biometrics, as the different commercial fingerprint sensors integrated in portable electronic devices like smartphones or tablets demonstrate.

- Abstract
Arbuscular mycorrhiza and Trichoderma are well-known beneficial fungi whose plant growth promotion and defense elicitation effects are known. However, the molecular and biochemical processes underlying the beneficial effects of these priming microorganisms have not been fully elucidated yet. On this basis, the present work aimed to use metabolomics to dissect comprehensively the modulation of secondary metabolism induced by mycorrhiza and Trichoderma, using tomato as a model plant. To this aim, either mycorrhiza or Trichoderma were applied to tomato roots at transplanting using a commercial formulation and then harvested once the mutualistic relationship was well established. Shoots were analyzed using an MS-based untargeted metabolomics approach, and differential metabolites identified by multivariate statistics were subjected to pathway analysis. Together with promoting plant growth, the treatments induced a broad molecular reprogramming with the phenylpropanoid biosynthetic pathway (including defense phenolics like coumarins and glycosylated anthocyanins) being strongly elicited. An accumulation of auxins, cytokinins, and jasmonate (especially after treatment with Trichoderma) could be observed concerning phytohormone profiles. Overall, the broad and distinctive effects triggered by mycorrhiza and Trichoderma in tomato secondary metabolism supported both plant growth promotion and immunity.

Several 2-benzylbenzimidazole opioids (also referred to as "nitazenes") recently emerged on the illicit market. The most frequently encountered member, isotonitazene, has been identified in multiple fatalities since its appearance in 2019. Although recent scheduling efforts targeted isotonitazene, many other analogues remain unregulated. Being structurally unrelated to fentanyl, little is known about the harm potential of these compounds. In this study, ten nitazenes and four metabolites were synthesized, analytically characterized via four different techniques, and pharmacologically evaluated using two cell-based beta-arrestin2/mini-Gi recruitment assays monitoring mu-opioid receptor (MOR) activation. On the basis of absorption spectra and retention times, high-performance liquid chromatography coupled to diode-array detection (HPLC-DAD) allowed differentiation between most analogues. Time-of-flight mass spectrometry (LC-QTOF-MS) identified a fragment with m/z 100.11 for 12/14 compounds, which could serve as a basis for MS-based nitazene screening. MOR activity determination confirmed that nitazenes are generally highly active, with potencies and efficacies of several analogues exceeding that of fentanyl. Particularly relevant is the unexpected very high potency of the N-desethylisotonitazene metabolite, rivaling the potency of etonitazene and exceeding that of isotonitazene itself. Supported by its identification in fatalities, this likely has in vivo consequences. These results improve our understanding of this emerging group of opioids by laying out an analytical framework for their detection, as well as providing important new insights into their MOR activation potential.

Background Cardiovascular diseases (CDs) were the leading cause of death worldwide in 2020. The risk of increasing CD is related to low Health Literacy (HL). HL can be defined as the individual’s ability to understand and process health information. Low HL has a risk ratio of 1.90 on mortality and 1.35 on readmission. A low level of HL was related to poorer use of health care services. Several inconsistencies were found about the relationship between HL and Length of stay (LOS). Although CDs are increasing because of the aging population, HL is often underestimated in the hospital setting. For this reason, HL assessment is important to identify patients at risk of poor HL. Therefore, this study aimed to describe the HL level in inpatients with CDs and to analyze the relationship between HL and LOS Method A retrospective design was used to randomly select 134 patients with CDs in eight cardiovascular and internal medicine units of an Italian hospital from 10 January to 10 May 2021. Clinical data were collected using a nursing information system (NIS) and the hospital discharge register. Single Item Literacy Screener (SILS) was used to measure HL and was systematically reported on NIS. Results The mean age of the sample was 71.28 (SD 13.19); 59% were male. HL‘s mean score was 2.43 (SD 1.2). A statistically significant linear association (Spearman’s correlation coefficient) was observed between the HL score and LOS. Patients with inadequate HL were 50.7% and had a longer LOS than patients with adequate HL (mean 9.1 ± 7.1 Vs 7.2 ± 5.9 days respectively, p = 0.05). Conclusion HL is poor and has been shown to be associated with LOS. These results should be confirmed after considering other variables through multivariate analyses. These findings suggest that the adverse effects of low HL may extend into the inpatient cardiovascular setting

Background HL plays a fundamental role in patients’ healthcare. According to the American Medical Association, HL can be viewed as a stronger predictor of a person’s health status than age, education level, and race. HL concept has many implications for nursing; they are responsible for identifying the patient’s barriers to their health information’s comprehension and providing specific nursing interventions in minimizing barriers. Moreover, HL assessment is necessary to plan tailored interventions, improve their efficacy through patient–centered communication. As a part of the nursing process, the nursing diagnosis (ND) classifies patients according to their level of nursing dependency and can be predictive for the nursing workload. No studies have been conducted on the relationship between NDs and HL. We aimed to describe the prevalence and distribution of NDs on admission in patients with CDs and to analyze the relationship between HL and NDs. Method A retrospective design was used to randomly select 134 patients with CDs in eight cardiovascular and internal medicine units of an Italian hospital from 10 January to 10 May 2021. NDs were collected using a nursing information system (NIS). Single Item Literacy Screener (SILS) was used to measure HL and was systematically reported on NIS. Results A mean of 3.5 ± 2.3 nursing diagnosis per patient was identified. A statistically significant linear association (Spearman’s correlation coefficient) was observed between HL score and the number of NDs. Conclusion HL was associated with the number of NDs.
