Uważa go za książkę ratunkową: to jedna z niewielu lektur aktywujących uczniowską emergencję. Wskazuje także na tropy możliwe do podjęcia podczas zajęć języka polskiego w szkole ponadpodstawowej.

The adoption of Directive 2010/63/EU on the protection of animals used for scientific purposes has given a major push to the formation of Three Rs initiatives in the form of centres and platforms. These centres and platforms are dedicated to the so-called Three Rs, which are the Replacement, Reduction and Refinement of animal use in experiments. ATLA’s 50th Anniversary year has seen the publication of two articles on European Three Rs centres and platforms. The first of these was about the progressive rise in their numbers and about their founding history; this second part focuses on their current status and activities. This article takes a closer look at their financial and organisational structures, describes their Three Rs focus and core activities (dissemination, education, implementation, scientific quality/translatability, ethics), and presents their areas of responsibility and projects in detail. This overview of the work and diverse structures of the Three Rs centres and platforms is not only intended to bring them closer to the reader, but also to provide role models and show examples of how such Three Rs centres and platforms could be made sustainable. The Three Rs centres and platforms are very important focal points and play an immense role as facilitators of Directive 2010/63/EU ‘on the ground’ in their respective countries. They are also invaluable for the wide dissemination of information and for promoting the implementation of the Three Rs in general.

1-Methylnaphthalene (1-MN) is composed of 2 benzene rings and belongs to polycyclic aromatic hydrocarbons. The metabolism of 1-MN in laboratory animals and bacteria leads to the formation of 1-naphthoic acid (1-NA).In this study the distribution of 1-NA in lung, liver, spleen, kidney and urinary excretion of 1-NA in rats after single and repeated inhalation exposure to 1-MN vapors were investigated. The activity of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and cytochrome were measured of the rats. Genotoxic effects were evaluated with theThe concentrations of 1-NA in the tissues of rats after single and repeated exposure to 1-MN were dependent on the exposure dose. High levels of 1-NA were found in kidneys of animals after the single and repeated exposure to 1-MN. With an increase of 1-MN dose, an increase in the activity of cytochrome P450 (CYP1A1 and CYP1A2) was observed in the liver of rats. Compared to control animals, significantly higher ALT activity was noted in serum of rats exposed to 1-MN. The micronuclei frequency in V79 cells exposed to 1-MN (in the range of analyzable concentrations; i.e., 5-25 μg/ml) did not differ significantly from the vehicle control, whereas urine extracts from rats exposed to 1-MN induced a significant increase in the frequency of micronuclei compared to urine extracts from the group of control animals.Metabolism of 1-MN in rats after the inhalation exposure leading to 1-NA was mainly observed during the first day after the end of exposure. It is likely that 1-MN metabolites present in rat urine can induce the increased micronuclei frequency as was shown in V79 cells. Int J Occup Med Environ Health. 2022;35(6):731-46.

Public awareness and discussion about animal experiments and replacement methods has greatly increased in recent years. The term ‘the Three Rs’, which stands for the Replacement, Reduction and Refinement of animal experiments, is inseparably linked in this context. A common goal within the Three Rs scientific community is to develop predictive non-animal models and to better integrate all available data from in vitro, in silico and omics technologies into regulatory decision-making processes regarding, for example, the toxicity of chemicals, drugs or food ingredients. In addition, it is a general concern to implement (human) non-animal methods in basic research. Toward these efforts, there has been an ever-increasing number of Three Rs centres and platforms established over recent years — not only to develop novel methods, but also to disseminate knowledge and help to implement the Three Rs principles in policies and education. The adoption of Directive 2010/63/EU on the protection of animals used for scientific purposes gave a strong impetus to the creation of Three Rs initiatives, in the form of centres and platforms. As the first of a series of papers, this article gives an overview of the European Three Rs centres and platforms, and their historical development. The subsequent articles, to be published over the course of ATLA’s 50th Anniversary year, will summarise the current focus and tasks as well as the future and the plans of the Three Rs centres and platforms. The Three Rs centres and platforms are very important points of contact and play an immense role in their respective countries as ‘on the ground’ facilitators of Directive 2010/63/EU. They are also invaluable for the widespread dissemination of information and for promoting implementation of the Three Rs in general.

The article attempts to analyse Hannah Arendt’s short essay “We Refugees” published in 1943 in a Jewish magazine The Menorah Journal in the context of the current humanitarian crisis (a crisis of reception policies) and peace studies. It also reconstructs refugee themes in the life trajectory of the author of Eichmann in Jerusalem. The author is confident that the essay “We Refugees” from seventy-seven years ago allows one to speak up repeating the questions concerning one of the key problems of contemporary world – the condition of the outcasts.

Cadmium (Cd), a heavy metal with strong carcinogenic properties has been linked with breast cancer risk. Epidemiological data on the association between Cd exposure and breast cancer are not consistent and suggest that this relationship may be modulated by a number of different factors. The mechanisms of action underlying the molecular effects of Cd, especially in terms of its carcinogenicity, are generally not well understood. Specifically, in the mammary gland, the effects of Cd are considered to be related mainly to its oestrogenic potential, however, several other mechanisms have also been suggested, such as epigenetic alterations, inhibition of DNA repair pathways, induction of oxidative stress, interference with metallothioneins, cadherins and integrins, as well as interactions with xenobiotics. This review summarizes the current state of knowledge in this field, including potential mechanisms of action of Cd in breast cancer initiation and progression, as well as possible ways of protection against its toxicity. Importantly, there are many research gaps in this area since limited evidence is available from experimental studies. Important issues to be further investigated concern exact molecular mechanisms of Cd accumulation in the tissues and Cd-induced activation of eostrogen receptors. Impact on DNA damage and epigenome upon Cd exposure in breast cancer development remains still highly unexplored area and should gain more interest.

Do tetrady Betty Reardon, uznającej za fundamenty idealnej edukacji na rzecz pokoju to, co: personalne, polityczne, pedagogiczne i poetyckie, proponuje ona dołączyć jeszcze jedno P – performatywne. Przedmiotem interpretacji uczyniono dwa musicale: Imagine This oraz Korczak, ujawniające zresztą wieloaspektowe zbieżności, m.in. występuje w nich motyw teatru w teatrze, akcja obu rozgrywa się w warszawskim getcie w 1942 roku.

The expanding applications of nanotechnology seem to be a response to many technological, environmental, and medical challenges. The unique properties of nanoparticles allow for developing new technologies and therapies. Among many investigated compounds is manganese and its oxides, which in the form of nanoparticles, could be a promising alternative for gadolinium-based contrast agents used in diagnostic imaging. Manganese, which is essential for living organisms as an enzyme cofactor, under excessive exposure—for example, due to water contamination or as an occupational hazard for welders—can lead to neurological disorders, including manganism—a condition similar to Parkinson’s disease. This review attempts to summarise the available literature data on the potential applications of manganese and manganese oxide nanoparticles and their biological activity. Some of the published studies, both in vitro and in vivo, show negative effects of exposure to manganese, mainly on the nervous system, whereas other data suggest that it is possible to develop functionalised nanoparticles with negligible toxicity and novel promising properties.

Autorka artykułu akcentuje potrzebę konfrontowania młodzieży z tematem Innych (etnicznych, narodowościowych, religijnych), co wiąże się ze stosunkowo nową sytuacją społeczną i polityczną, z nasilającymi się ruchami migracyjnymi. Polska szkoła powinna zobaczyć dla siebie istotne zadanie w kontekście światowego nomadyzmu i eskalującego kryzysu humanitarnego. Podczas etapu zbierania materiałów przeprowadzono wywiady skategoryzowane z nauczycielami, bibliotekarzami i uczniami dziesięciu poznańskich szkół. W artykule dokonano analizy i interpretacji zawartości podręczników do nauczania języka polskiego oraz zaproponowano realizację tematów powiązanych z wątkiem uchodźczym. Płynie z niego wniosek, że najważniejszym zadaniem, stojącym przed polską edukacją na wszystkich szczeblach, wydaje się wprowadzenie, nieobecnej dotąd w programie, edukacji na rzecz pokoju.

In the article I have been studying what premises constitute in the work of this German-speaking Nobel Prize winner and how the author brings the wounded world to the poem. I am aiming at interpreting her works in the relation to the refugees theme. I read her neurotic poetry also in the context of the contemporary nomadological discourse and I perceive it as a signpost to the language of solidarity. In the works of the Nobel Prize winner from 1966 one can see a preview of the style of the poems written by contemporary refuges (Breyten Breytenbach or Ashur Etwebi), and the artistic community of people deprived of their homes. I close the article with the conclusion related to the use of poetry in education for peace.

Omówienie najsłynniejszej powieści antywojennej wszechczasów, Na Zachodzie bez zmian Ericha Marii Remarque’a, przedstawiciela lost generation, staje się pretekstem, by zestawić niemieckiego pisarza z urodzonym w Warszawie fizykiem, Józefem Rotblatem. Obaj znaleźli własną drogę, by walczyć z powikłaniami wojennymi. Pierwszego z nich, jako weterana wojny światowej z lat 1914-1918 i autora antywojennej powieści, nominowano w roku 1931 do Pokojowej Nagrody Nobla, drugi w 1995 został jej laureatem. O ile jednak Na Zachodzie bez zmian to powieść najsilniej powiązana asocjacyjnie z I wojną światową, o tyle Rotblat, kojarzony na zachodzie Europy z wieloletnimi działaniami nawołującymi do światowego pokoju i rozbrojenia nuklearnego, w Polsce jest niemal nieznany, a w szkolnych programach nauczania nie znaleziono dla niego miejsca. Artykuł poświęcony jest ich odrębnym drogom uprawiania „wojującego pacyfizmu”.

This study was undertaken to assess cytotoxic effects of selected aluminium compounds, parabens and phthalates in combination with silver nanoparticles (AgNP, 15 and 45 nm by STEM, Ag15 and Ag45, respectively) on cell lines of the human breast epithelium, normal (MCF-10A) and transformed (MDA-MB-231 and MCF-7). Combination indices were the most spectacular at effective concentrations (ED) inducing 25 % decrease in viability for the combinations of Ag15 with AlCl3 for MDA-MB-231 cells or aluminium zirconium tetrachlorohydrex Gly (AlZr) for MCF-10A and MCF-7 cells, where rather strong antagonism was revealed. As the ED values increased, those effects were enhanced (e.g. Ag15+AlCl3 for MDA-MB-231) or reversed into synergism (e.g. Ag15+AlZr for MCF-7). Another strong effect was observed for aluminium chloride hydroxide, which increasing ED, induced synergistic effect with both Ag15 and Ag45 on MCF-10A cells. Another interesting synergistic effect was observed for DBPh, but only in combination with Ag45 on MCF-10A and MCF-7. The results on cytotoxicity, cell cycle and oxidative stress induction indicate complex response of the cell lines to combined treatment with silver nanoparticles and the chemicals, which were influenced by diverse factors, such as physico-chemical characteristics of AgNP, method of their synthesis, concentrations used, and finally cell type.

In June 1942 Emanuel Ringelblum asked: “What kind of books do people read? This topic has always been interesting for each Jew and after the war it also became interesting to the world”. This topic hasn’t received a comprehensive study yet. The author of the paper The Remains of the Letters. Three Reading Paradigms in the Ghettos examined this aspect of history of the ghettos. The literature allows us to shed light on the topic of the life and deaths of their inhabitants. But, taking up this topic enables us also to activate new ways of interpreting the books. The volumes they spent time on in the ghetto allow to capture the way the residents of the ghettos built or broke the bond with the reality, how they taught their own children to read or found solace in someone else’s pain. The author considers, however, whether the studies of ordinary life in extraordinary times of the Holocaust can be considered micro-historical.

Arsenic trioxide (ATO) is a well-recognized antileukemic drug used for the treatment of newly diagnosed and relapsed acute promyelocytic leukemia (APL). A major drawback of therapy with ATO is the development of APL cell resistance, the mechanisms of which are still not clear. The aim of this study was to investigate the role of the PI3K/Akt signaling pathway in ATOtreated human acute myeloid leukemia (HL-60) cells and in ATO-resistant clones. The cytotoxicity of ATO was assessed using Trypan blue staining or a WST-1 reduction assay. The Akt phosphorylation level was measured by immunofluorescent staining and flow cytometry. Gene expression analysis was performed using real-time polymerase chain reaction (PCR). The clones derived by culturing for 8-12 weeks in the presence of 1.75, 2.5, and 5 μM ATO were characterized by high viability but a slower growth rate compared to the parental HL-60 cells. The flow cytometry analysis showed that in the parental cells the levels of p-Akt were undetectable or very low, and that ATO had no effect on the level of p-Akt in either the ATO-treated parental cells or the clones. The gene expression analysis revealed that some of the genes involved in the Akt pathway may play a key role in the induction of resistance to ATO, e.g., genes encoding cyclin D1 (CCND1), fork head box O1 (FOXO1), Jun oncogene (JUN), protein kinase C isoform B1 (PRKCB1), because their expression profiles were predominantly changed in the clones and/or the ATO-treated parental HL-60 cells. The overall results indicate that CCND1, FOXO1, and JUN may contribute to the induction of resistance to ATO, and that the C-Jun N-terminal kinase (JNK) signaling pathway may have greater significance than the phosphoinositide 3-kinase (PI3K)/Akt pathway in mediating the cytotoxic effects of ATO and the development of resistance to ATO in the HL-60 cell line. In this study the role of PI3K/Akt signaling pathway in arsenic trioxide (ATO)-treated parental Jurkat cells and also in derived ATO-resistant clones grown in the presence of given ATO concentration was investigated. ATO-resistant clones (cultured for 8-12weeks in the presence of 1, 2.5 and 5μM ATO) were characterized by high viability in the presence of ATO but slower growth rate compared to the parental cells. Morphological and functional characterization of derived ATO-resistant clones revealed that they did not differ fundamentally from parental Jurkat cells in terms of cell size, level of GSH, the lysosomal fluorescence or CD95/Fas surface antigen expression. However, a slight increase in the mitochondrial potential (JC-1 staining) was detected in the clones compared to parental Jurkat cells. Side population analysis (Vybrant DyeCycle Violet™ staining) in ATO resistant clones did not indicate any enrichment withcancer stem cells. Akt1/2, AktV or wortmannin inhibitors decreased viability of ATO-resistant clones grown in the presence of ATO, with no effect on ATO-treated parental cells. Flow cytometry analysis showed that ATO decreased the level of p-Akt in ATO-treated parental cells, while the resistant clones exhibited higher levels of p-Akt immunostaining than parental Jurkat cells. Expression analysis of 84 genes involved in the PI3K/Akt pathway revealed that this pathway was predominantly active in ATO-resistant clones. c-JUN seems to play a key role in the induction of cell death in ATO-treated parental Jurkat cells, as dose-dependent strong up-regulation of JUN was specific for the ATO-treated parental Jurkat cells. On the other hand, changes in expression of cyclin D1 (CCND1), insulin receptor substrate 1 (IRS1) and protein kinase C isoforms (PRKCZ,PRKCB and PRKCA) may be responsible for the induction of resistance to ATO. The changes in expression of growth factor receptor-bound protein 10 (GRB10) observed in ATO-resistant clones suggest a possibility of induction of different mechanisms in development of resistance to ATO depending on the drug concentration and thus involvement of different signaling mediators.

The effect of inhibitors of phosphatidylinositol-3-kinase-related kinases (PIKK): ataxia-telangiectasia mutated (ATM), ATM- and Rad3-related (ATR) and DNA-dependent protein kinase (DNA-PK) on response of HepG2 human liver cancer cells to benzo[a]pyrene (BaP) was investigated. PIKK inhibitors: KU55933 (5 μM), NU7026 (10 μM) or caffeine (1 and 2mM) when used as single agents or in combinations (KU55933/NU7026 and caffeine/NU7026) did not significantly influence the BaP (3 μM) cytotoxicity (MTT reduction test). BaP induced a weak proapoptotic effect which was moderately enhanced by both inhibitor combinations. HepG2 cells exposed to BaP showed a strong S-phase arrest which was considerably diminished by both inhibitor combinations. The DNA damage (comet assay) induced after continuous 24h exposure to BaP was significantly diminished by both inhibitor combinations. Weak induction of reactive oxygen species by BaP was observed, which was not modulated by the inhibitor combinations. Similarly, no modulation of the glutathione levels was observed.

The article is a reflection on the wall being built since 2002 between Israel and the Palestinian Authority. The paper presents this structure, its history and how it is reflected in poetry (Yehuda Amichai and Mahmud Darwish), reportage (Raja Shehadeh) and film. Artists’ initiatives once focussed on the Berlin Wall and more recently on the initiatives related to the Palestinian wall prompt the question how the media report on the present situation of the excluded. It is worth bearing in mind that memory or post-memory of our European wall and ghettos does not shape the discourse about the Middle-East. The preferred method of talking about walls has become geocritics (based more broadly on cultural and postcolonial studies; it is significant that the foundations of the theory of postcolonialism were put forward by a Palestinian – Edward Said).
