The major challenge involved is to preserve, during the immobilization process, both the biological activity and the structural integrity of the enzymes. Here we report, for the first time, a proof-of-concept study for embedding active carbonic anhydrase (CA) into polyacrylamide (PAAm) nanogels. By immobilizing CA in these nanogels, we hope to provide important advantages, such as matrix protection of the CA as well as its targeted delivery, and also for potentially using these nanogels as zinc nano-biosensors, both in-vitro and in-vivo.Methods and Results: Two methods are reported here for CA immobilization: encapsulation and surface conjugation. In the encapsulation method, the common process was improved, so as to best preserve the CA, by 1) using a novel biofriendly nonionic surfactant system (Span 80/Tween 80/Brij 30) and 2) using an Al2O3 adsorptive filtration purification procedure. In the surface conjugation method, blank PAAm nanogels were activated by N-hydroxysuccinimide and the CA was cross-linked to the nanogels. The amount of active CA immobilized in the nanoparticles was quantified for both methods. Per 1 g nanogels, the CA encapsulated nanogels contain 11.3 mg active CA, while the CA conjugated nanogels contain 22.5 mg active CA. Also, the CA conjugated nanoparticles successfully measured free Zn2+ levels in solution, with the Zn2+ dissociation constant determined to be 9 pM.Conclusion: This work demonstrates universal methods for immobilizing highly fragile bio-macromolecules inside nanoparticle carriers, while preserving their structural integrity and biological activity. 2. Spontaneous Charge aggregation and Polarization Model for Electric Discharges in Clouds

Abstract

In present models cloud based lightning forms as a consequence of the earth's gravitational and/or electromagnetic fields. Our simplified field-free model probes the random aggregation of a neutral ensemble consisting of a random distribution of equal numbers of equally charged positive and negative "frozen droplets", given a bivariate sticking probability. Over a wide range of parameters, one maximal cluster (called "cloud") is formed, with time-increasing mass, net charge and electric dipole moment, where all relations follow a simple argument, and the only parameter is the fractal dimension of the cloud: In this overall neutral system we find that the net charge of the cloud increases with the square root of the cloud's mass, while the electric dipole moment increases quasi-linearly and even superlinearly. Thus an internal electric field can be self-created by a random process, with a field-strength that increases with time and cloud size, using just a two-dimensional random walk. Potential implications of this toy model on "real life" clouds are discussed, i.e., the electric dipole moment based possibility of having intra-cloud lightning discharges, and the net electric charge based possibility of having extra-cloud lightning discharges, all in the absence of gravitational or other external fields or forces. Possible implications are discussed for both water and dust based clouds, atmospheric and extraterrestrial. 3. Magnetic particle biosensors

Abstract

With the recent technological advances in the synthesis and characterization of nano- and microparticles, there has been a growing interest in applying this technology to different areas, such as diagnosis of diseases and detection of pollutants. The use of magnetic particles as biosensors is favored due to the mobility of magnetic particles under magnetic fields and the low magnetic background observed in most biological samples. This chapter briefly discusses the basic ways to modify the surface of magnetic particles to enhance interaction with biological targets, and thoroughly reviews the different applications of magnetic particles as biosensors. The use of magnetic particles is divided into three categories by the functionality of the magnetic particles in these biosensors, namely magnetic particles as carriers for signal intensification, magnetic particles as labels to detect magnetic contents, and bio-sensing based on the dynamics of magnetic particles. The strategy and theory of different magnetic particle biosensors are elucidated in details and illustrated through examples.

Abstract

Disorders of mineral metabolism, including secondary hyperparathyroidism, are thought to contribute to extraskeletal (including vascular) calcification among patients with chronic kidney disease. It has been hypothesized that treatment with the calcimimetic agent cinacalcet might reduce the risk of death or nonfatal cardiovascular events in such patients.In this clinical trial, we randomly assigned 3883 patients with moderate-to-severe secondary hyperparathyroidism (median level of intact parathyroid hormone, 693 pg per milliliter [10th to 90th percentile, 363 to 1694]) who were undergoing hemodialysis to receive either cinacalcet or placebo. All patients were eligible to receive conventional therapy, including phosphate binders, vitamin D sterols, or both. The patients were followed for up to 64 months. The primary composite end point was the time until death, myocardial infarction, hospitalization for unstable angina, heart failure, or a peripheral vascular event. The primary analysis was performed on the basis of the intention-to-treat principle.The median duration of study-drug exposure was 21.2 months in the cinacalcet group, versus 17.5 months in the placebo group. The primary composite end point was reached in 938 of 1948 patients (48.2%) in the cinacalcet group and 952 of 1935 patients (49.2%) in the placebo group (relative hazard in the cinacalcet group vs. the placebo group, 0.93; 95% confidence interval, 0.85 to 1.02; P=0.11). Hypocalcemia and gastrointestinal adverse events were significantly more frequent in patients receiving cinacalcet.In an unadjusted intention-to-treat analysis, cinacalcet did not significantly reduce the risk of death or major cardiovascular events in patients with moderate-to-severe secondary hyperparathyroidism who were undergoing dialysis. Abstract

Background— Patients with kidney disease have disordered bone and mineral metabolism, including elevated serum concentrations of fibroblast growth factor-23 (FGF23). These elevated concentrations are associated with cardiovascular and all-cause mortality. The objective was to determine the effects of the calcimimetic cinacalcet (versus placebo) on reducing serum FGF23 and whether changes in FGF23 are associated with death and cardiovascular events. Methods and Results— This was a secondary analysis of a randomized clinical trial comparing cinacalcet to placebo in addition to conventional therapy (phosphate binders/vitamin D) in patients receiving hemodialysis with secondary hyperparathyroidism (intact parathyroid hormone ≥300 pg/mL). The primary study end point was time to death or a first nonfatal cardiovascular event (myocardial infarction, hospitalization for angina, heart failure, or a peripheral vascular event). This analysis included 2985 patients (77% of randomized) with serum samples at baseline and 2602 patients (67%) with samples at both baseline and week 20. The results demonstrated that a significantly larger proportion of patients randomized to cinacalcet had ≥30% (68% versus 28%) reductions in FGF23. Among patients randomized to cinacalcet, a ≥30% reduction in FGF23 between baseline and week 20 was associated with a nominally significant reduction in the primary composite end point (relative hazard, 0.82; 95% confidence interval, 0.69–0.98), cardiovascular mortality (relative hazard, 0.66; 95% confidence interval, 0.50–0.87), sudden cardiac death (relative hazard, 0.57; 95% confidence interval, 0.37–0.86), and heart failure (relative hazard, 0.69; 95% confidence interval, 0.48–0.99). Conclusions— Treatment with cinacalcet significantly lowers serum FGF23. Treatment-induced reductions in serum FGF23 are associated with lower rates of cardiovascular death and major cardiovascular events. Clinical Trial Registration— URL: http://www.clinicaltrials.gov . Abstract

A fluorescent chemical sensor platform, integrating an organic light-emitting device (OLED) light-source with a fluorescent probe, is demonstrated for a subsecond-fast oxygen sensor. The integration results in strong light coupling and negligible heating of the sensor film or analyte. The potential in vivo operation of compact, stand-alone, battery-powered, OLED-based miniaturized sensor arrays for chemical and biological applications is discussed. © 2002 American Institute of Physics.

The fourth symposium on 'Dynamics in Small Confining Systems,' held as part of the 1996 MRS Fall Meeting, covered a broad range of topics related to static and dynamic properties of confining systems: probing of confined systems, structure and dynamics of liquids at interfaces, nanorheology and tribology, adsorption, diffusion in pores, and reactions. Participants from various disciplines shared different points of view on the questions of how ultrasmall geometries can force a system to behave in ways significantly different from its behavior in the bulk, how this difference affects molecular properties, and how it is probed. There appears to be a continuing interest in the dynamics and thermodynamics of confined molecular systems. The symposium was an effective way to bring together different disciplines interested in. The contributions in the book reflect the broad range of topics discussed at the meeting, all related to dynamics in small confining systems. The papers appear in the book in the order of their presentation during the symposium.

18. Spatially and Temporally Resolved Studies of Convectionless Photobleaching Kinetics: Line Trap

Abstract

The kinetics of the growth of depletion zones around a static trap in an effective one-dimensional geometry were studied with a new fluorescence-based setup. The experiment consists of a photobleaching reaction of fluorescein dye by a strong laser beam, which served as a phototrap in the experiment, inside a 150-μm gap between two parallel microscope slides. The kinetics of the growth of the depletion zone were monitored by the previously defined θ distance, which can be directly measured experimentally. The effect of trap strengths on the kinetics was tested experimentally by changing the photobleaching laser power. The strong laser power acted as a perfect trap, which gives a t1/2 scaling behavior for the θ distance over most of the time range. However, the experiment with a weak laser power produced an anomalous early-time behavior for the θ distance, which is faster than the t1/2 time scaling. A crossover behavior was observed with an intermediate laser power. The experimental results are consistent with an exact one-dimensional analytical solution and were also reproduced in Monte Carlo simulations as well as by exact enumeration calculations. The latter two methods show that the asymptotic results for an exact one-dimensional lattice are still valid in a quasi-one-dimensional system, for both perfect and imperfect trapping reactions.

19. Correlated Excimer Formation and Molecular Rotational Dynamics in Phenylacetylene Dendrimers

Abstract

Steady-state and time-resolved fluorescence data are used to examine the photophysical characteristics of a series of phenylacetylene dendrimers. Correlated excimer formation is observed in some solvents which is found to depend strongly upon initial excited-state placement within the dendrimer framework. The results suggest two distinct solvated molecular environments which give rise to independent photophysical behaviors. Models of excimer formation dynamics and relaxation kinetics are described. Additionally, fluorescence anisotropy measurements are examined in the context of simultaneous probe wobbling-in-a-cone and macromolecular dendrimeric rotation as the mechanisms leading to fluorescence depolarization. Approximate solutions to the model are shown to provide good descriptions of the solvated molecular dynamics.

20. Monte Carlo Simulation of Surface Adsorption−Diffusion−Reaction Kinetics

Abstract

A surface adsorption−diffusion−reaction model is proposed and tested by Monte Carlo simulations. The solvent shell effect is considered as a geometric constraint and the Langmuir adsorption mechanism is also incorporated. An anomalous power law of t1/2 is obtained for the amount of material removed from the surface vs time. The result agrees well with etching experiments of SAM on a silica surface. We conclude that the unusual power law results from an interplay of vacancy growth and fusion at the surface.

21. Spatially Resolved Anomalous Kinetics of a Catalytic Reaction:  Enzymatic Glucose Oxidation in Capillary Spaces

Abstract

A reaction involving the catalytic oxidation of glucose by the enzyme glucose oxidase is employed as a first spatially resolved experimental realization of a nonclassical A + C → C + Products chemical reaction. This heterogeneous kinetics experiment models the operation of optical glucose sensors. One-dimensional nonclassical reaction kinetics, manifested in anomalous rate laws, are observed. The time evolution of the density gradient of O2 in the zone near the catalytic trap was monitored. The temporal dependence of the growth of this depletion zone M, predicted to scale as t1/2, was measured to be M ~ t0.50±0.05. This time dependence of the growth in M is directly proportional to the anomalous, time-dependent, reaction rate coefficient.

22. Directed Energy Transfer Funnels in Dendrimeric Antenna Supermolecules

Abstract

We have studied the dynamics of directed, multistep energy transport in a class of fractal-like dendrimeric molecules. For particular forms of these highly branched phenylacetylene dendrimers, both theory and experiment put the lowest excitation energy at the center (locus) of the supermolecule. This results in a structurally symmetric and ordered exciton funnel, with a well-directed energy gradient. We have designed and synthesized a derivative of these dendrimers with a perylene moiety at the locus, which acts as an energy trap for the directed exciton funnel. Spectroscopic evidence indicates transfer efficiency of 98% from the photoabsorbing dendrimer backbone to the perylenic trap.

23. Anomalous Etching Kinetics of Self-Assembled Monolayers on Silica−Water Interfaces:  Experiment and Modeling

Abstract

We have studied the etching of siloxane/silica self-assembled monolayers in basic alcohol solutions using second harmonic generation. The kinetics of the etching process takes an unusual algebraic power law time dependence, with an exponent of 0.5. On the basis of the chemical nature of our system, we propose a surface cleavage mechanism and a surface diffusion based reaction model. We also perform Monte Carlo simulations on the system. When the solvent shell effect is considered as a geometric constraint for surface adsorption, we reproduce the anomalous power law.

24. Spatiotemporal Patterns and Nonclassical Kinetics of Competing Elementary Reactions:  Chromium Complex Formation with Xylenol Orange in a Capillary

Abstract

An experimental investigation of chemical reaction fronts, created by an initial separation of reactants, is reported for a system of two competing reactions. Spatiotemporal patterns are observed experimentally for the competing reaction front and are accounted for quantitatively by a reaction-diffusion model. We use the reaction of xylenol orange with Cr3+ in aqueous solution. Different oligomers of Cr3+ provide the two kinetically different species that react competitively with xylenol orange. The parameters that determine whether pattern formation is observable at the front are the ratios of (1) the microscopic reaction constants of the competing reactions and (2) the concentrations of the competing species. Under the parameter values studied, which allowed clear spatiotemporal separation of the two competing reactions, we find that the behavior of the reaction front at early times follows a perturbation theory developed for a simple elementary A + B → C reaction with initially separated reactants. The global reaction rate, observed over the entire time scale of the experiments, is highly non-monotonic. Overall, with no free parameters, our theoretical model is quantitatively consistent with the experimental observations of the spatiotemporal patterns, the unusual scaling laws, and the crossover behaviors. The geometrical constraints and nonclassical behavior of the reaction rate allow a quantitative determination of the reaction probability of the chromium ion monomer relative to that of the higher order oligomers.

25. Diffusion-Controlled Elementary Reactions in Tubular Confinement:  Extreme Nonclassicality, Segregation, and Anomalous Scaling Laws for Dimensional Crossovers

Abstract

To understand reaction kinetics in capillaries, pores, and tubules, we performed Monte Carlo simulations of random walk based exploration volumes and bimolecular A + A and A + B reactions on baguette-like lattices. The emphasis is on the scaling of the dimensional crossover times with tube width. We find that the exponents range between 1 and 4, i.e., that the global information propagates either slower or faster than single-particle diffusion, depending on the reaction type (e.g. A + A or A + B ) and on the dimensionality (2 or 3). The time evolutions of the A + A reactions approximately mimic those of the average exploration volumes, within the simulation uncertainties. All asymptotic time behaviors exhibit truely one-dimensional character, i.e., extremely nonclassical kinetics. Rapid and complete A, B reactant segregation is illustrated. The nonuniversal scaling powers present a new theoretical and experimental challenge.

Abstract

Background andobjectivesThecalcimimeticcinacalcet reduced therisk of death or cardiovascular (CV) events in older, but not younger, patients with moderate to severe secondary hyperparathyroidism (HPT) who were receiving hemodialysis. To determine whether the lower risk in younger patients might be due to lower baseline CV risk and more frequent use of cointerventions that reduce parathyroid hormone (kidney transplantation, parathyroidectomy, and commercial cinacalcet use), this study examined the effects of cinacalcet in older ($65 years, n=1005) and younger (,65 years, n=2878) patients. Design, setting, participants, & measurements Evaluation of Cinacalcet HCl Therapy to Lower Cardiovascular Events (EVOLVE) was a global, multicenter, randomized placebo-controlled trial in 3883 prevalent patients on hemodialysis, whose outcomes included death, major CV events, and development of severe unremitting HPT. The age subgroup analysis was prespecified. ResultsOlderpatients hadhigher baselineprevalenceof diabetesmellitusandCV comorbidity. Annualizedrates of kidney transplantation and parathyroidectomy were .3-fold higher in younger relative to older patients and were more frequent in patients randomized to placebo. In older patients, the adjusted relative hazard (95% confidence interval) for the primary composite (CV) end point (cinacalcet versus placebo) was 0.70 (0.60 to 0.81); in younger patients, the relative hazard was 0.97 (0.86 to 1.09). Corresponding adjusted relative hazards for mortality were 0.68 (0.51 to 0.81) and 0.99 (0.86 to 1.13). Reduction in the risk of severe unremitting HPT was similar in both groups. Conclusions In the EVOLVE trial, cinacalcet decreased the risk of death and of major CV events in older, but not younger, patients with moderate to severe HPT who were receiving hemodialysis. Effect modification by age may be partly explained by differences in underlying CV risk and differential application of cointerventions that reduce parathyroid hormone. Abstract

Purpose: Early detection and diagnosis of cancer is critical for achieving positive therapeutic outcomes. Biomarkers that can provide clinicians with clues to the outcome of a given therapeutic course are highly desired. Oxygen is a small molecule that is nearly universally present in biological tissues and plays a critical role in the effectiveness of radiotherapies by reacting with DNA radicals and subsequently impairing cellular repair of double strand breaks.Techniques for measuring oxygen in biological tissues often use blood oxygen saturation to approximate the oxygen partial pressure in surrounding tissues despite the complex, nonlinear, and dynamic relationship between these two separate oxygen populations., Methods and Materials: We combined a directly oxygen-sensitive, tumor-targeted, chemical contrast nanoelement with the photoacoustic lifetime-based (PALT) oxygen imaging technique to obtain image maps of oxygen in breast cancer tumors in vivo. The oxygen levels of patient-derived xenografts in a mouse model were characterized before and after a course of radiotherapy., Results: We show that, independent of tumor size, radiotherapy induced an increase in the overall oxygenation levels of the tumor. Further, this increase in the oxygenation of the tumor significantly correlated with a positive response to radiotherapy, as demonstrated by a reduction in tumor volume over the twenty-day monitoring period following therapy and histological staining., Conclusion: Our PALT imaging presented here is simple, fast, and non-invasive. Facilized by the PALT approach, imaging of tumor reoxygenation may be utilized as a simple, early indicator for evaluating cancer response to radiotherapy. Further characterization of the reoxygenation degree, temporal onset, and possible theragnostic implications are warranted.

Abstract

Sodium has many vital and diverse roles in the human body, including maintaining the cellular pH, generating action potential, and regulating osmotic pressure. In cancer, sodium dysregulation has been correlated with tumor growth, metastasis, and immune cell inhibition. However, most in vivo sodium measurements are performed via Na 23 NMR, which is handicapped by slow acquisition times, a low spatial resolution (in mm), and low signal-to-noise ratios. We present here a plasticizer-free, ionophore-based sodium-sensing nanoparticle that utilizes a solvatochromic dye transducer to circumvent the pH cross-sensitivity of most previously reported sodium nano-sensors. We demonstrate that this nano-sensor is non-toxic, boasts a 200 μM detection limit, and is over 1000 times more selective for sodium than potassium. Further, the in vitro photoacoustic calibration curve presented demonstrates the potential of this nano-sensor for performing the in vivo chemical imaging of sodium over the entire physiologically relevant concentration range.

Abstract

Evaluating the aggressiveness of prostate cancer (PCa) is crucial for PCa diagnosis and prognosis. Previously, studies have shown that photoacoustic spectral analysis (PASA) can assess prostate tissue microarchitecture for evaluating the aggressiveness of PCa. In this study, in a transgenic mouse (TRAMP) model of PCa, we utilized methylene blue polyacrylamide nanoparticles (MB PAA NPs) to label the cancer cells in prostate in vivo. MB PAA NPs can specifically target proliferating cancer cells as a contrast agent, allowing photoacoustic (PA) imaging to better detect PCa tumors, and also assessing prostate glandular architecture. With the PA signals from the prostates measured simultaneously by a needle hydrophone and a PA and ultrasound (US) dual-imaging system, we conducted PASA and correlated the quantified spectral parameter slopes with the cancer grading from histopathology. The PASA results from 18 mice showed significant differences between normal and cancer, and also between low-score cancer and high-score cancer. This study in the clinically relevant TRAMP model of PCa demonstrated that PA imaging and PASA, powered by MB PAA NPs that can label the PCa microarchitectures in vivo after systemic administration, can detect PCa and, more importantly, evaluate cancer aggressiveness.

37. Tumor Biochemical Heterogeneity and Cancer Radiochemotherapy: Network Breakdown Zone-Model.

Abstract

Breakdowns of two-zone random networks of the Erdős-Rényi type are investigated. They are used as mathematical models for understanding the incompleteness of the tumor network breakdown under radiochemotherapy, an incompleteness that may result from a tumor's physical and/or chemical heterogeneity. Mathematically, having a reduced node removal probability in the network's inner zone hampers the network's breakdown. The latter is described quantitatively as a function of reduction in the inner zone's removal probability, where the network breakdown is described in terms of the largest remaining clusters and their size distributions. The effects on the efficacy of radiochemotherapy due to the tumor micro-environment (TME)'s chemical make-up, and its heterogeneity, are discussed, with the goal of using such TME chemical heterogeneity imaging to inform precision oncology.

38. Tumor Hypoxia Heterogeneity Affects Radiotherapy: Inverse-Percolation Shell-Model Monte Carlo Simulations.

Abstract

Tumor hypoxia was discovered a century ago, and the interference of hypoxia with all radiotherapies is well known. Here, we demonstrate the potentially extreme effects of hypoxia heterogeneity on radiotherapy and combination radiochemotherapy. We observe that there is a decrease in hypoxia from tumor periphery to tumor center, due to oxygen diffusion, resulting in a gradient of radiative cell-kill probability, mathematically expressed as a probability gradient of occupied space removal. The radiotherapy-induced break-up of the tumor/TME network is modeled by the physics model of inverse percolation in a shell-like medium, using Monte Carlo simulations. The different shells now have different probabilities of space removal, spanning from higher probability in the periphery to lower probability in the center of the tumor. Mathematical results regarding the variability of the critical percolation concentration show an increase in the critical threshold with the applied increase in the probability of space removal. Such an observation will have an important medical implication: a much larger than expected radiation dose is needed for a tumor breakup enabling successful follow-up chemotherapy. Information on the TME's hypoxia heterogeneity, as shown here with the numerical percolation model, may enable personalized precision radiation oncology therapy.

Abstract

We define cell morphodynamics as the cell's time dependent morphology. It could be called the cell's shape shifting ability. To measure it we use a biomarker free, dynamic histology method, which is based on multiplexed Cell Magneto-Rotation and Machine Learning. We note that standard studies looking at cells immobilized on microscope slides cannot reveal their shape shifting, no more than pinned butterfly collections can reveal their flight patterns. Using cell magnetorotation, with the aid of cell embedded magnetic nanoparticles, our method allows each cell to move freely in 3 dimensions, with a rapid following of cell deformations in all 3-dimensions, so as to identify and classify a cell by its dynamic morphology. Using object recognition and machine learning algorithms, we continuously measure the real-time shape dynamics of each cell, where from we successfully resolve the inherent broad heterogeneity of the morphological phenotypes found in a given cancer cell population. In three illustrative experiments we have achieved clustering, differentiation, and identification of cells from (A) two distinct cell lines, (B) cells having gone through the epithelial-to-mesenchymal transition, and (C) cells differing only by their motility. This microfluidic method may enable a fast screening and identification of invasive cells, e.g., metastatic cancer cells, even in the absence of biomarkers, thus providing a rapid diagnostics and assessment protocol for effective personalized cancer therapy., Competing Interests: The authors have declared that no competing interests exist.

40. In vivo photoacoustic potassium imaging of the tumor microenvironment.

Abstract

The accumulation of potassium (K + ) in the tumor microenvironment (TME) has been recently shown to inhibit immune cell efficacy, and thus immunotherapy. Despite the abundance of K + in the body, few ways exist to measure it in vivo . To address this technology gap, we combine an optical K + nanosensor with photoacoustic (PA) imaging. Using multi-wavelength deconvolution, we are able to quantitatively evaluate the TME K + concentration in vivo , and its distribution. Significantly elevated K + levels were found in the TME, with an average concentration of approximately 29 mM, compared to 19 mM found in muscle. Abstract

With the capability of presenting endogenous tissue contrast or exogenous contrast agents in deep biological samples at high spatial resolution, photoacoustic (PA) imaging has shown significant potential for many preclinical and clinical applications. However, due to strong background signals from various intrinsic chromophores in biological tissue, such as hemoglobin, achieving highly sensitive PA imaging of targeting probes labeled by contrast agents has remained a challenge. In this study, we introduce a novel technique called transient triplet differential (TTD) imaging which allows for substantial reduction of tissue background signals. TTD imaging detects directly the triplet state absorption, which is a special characteristic of phosphorescence capable dyes not normally present among intrinsic chromophores of biological tissue. Thus, these triplet state absorption PA images can facilitate "true" background free molecular imaging. We prepared a known phosphorescent dye probe, methylene blue conjugated polyacrylamide nanoparticles, with peak absorption at 660 nm and peak lowest triplet state absorption at 840 nm. We find, through studies on phantoms and on an in vivo tumor model, that TTD imaging can generate a superior contrast-to-noise ratio, compared to other image enhancement techniques, through the removal of noise generated by strongly absorbing intrinsic chromophores, regardless of their identity.

Abstract

Changes of physiological pH are correlated with several pathologies, therefore the development of more effective medical pH imaging methods is of paramount importance. Here, we report on an in vivo pH mapping nanotechnology. This subsurface chemical imaging is based on tumor-targeted, pH sensing nanoprobes and multi-wavelength photoacoustic imaging (PAI). The nanotechnology consists of an optical pH indicator, SNARF-5F, 5-(and-6)-Carboxylic Acid, encapsulated into polyacrylamide nanoparticles with surface modification for tumor targeting. Facilitated by multi-wavelength PAI plus a spectral unmixing technique, the accuracy of pH measurement inside the biological environment is not susceptible to the background optical absorption of biomolecules, i.e., hemoglobins. As a result, both the pH levels and the hemodynamic properties across the entire tumor can be quantitatively evaluated with high sensitivity and high spatial resolution in in vivo cancer models. The imaging technology reported here holds the potential for both research on and clinical management of a variety of cancers.Background optical absorption of several biomolecules impedes an effective in vivo pH imaging in tumors. Here, the authors developed a visible light-based in vivo pH mapping method by coupling photoacoustic imaging and pH-responsive modified nanoparticles that selectively target tumor cells.

44. Matrix Density Engineering of Hydrogel Nanoparticles with Simulation-Guided Synthesis for Tuning Drug Release and Cellular Uptake.

Abstract

The use of a nanoparticle (NP)-based antitumor drug carrier has been an emerging strategy for selectively delivering the drugs to the tumor area and, thus, reducing the side effects that are associated with a high systemic dose of antitumor drugs. Precise control of drug loading and release is critical so as to maximize the therapeutic index of the NPs. Here, we propose a simple method of synthesizing NPs with tunable drug release while maintaining their loading ability, by varying the polymer matrix density of amine- or carboxyl-functionalized hydrogel NPs. We find that the NPs with a loose matrix released more cisplatin, with up to a 33 times faster rate. Also, carboxyl-functionalized NPs loaded more cisplatin and released it at a faster rate than amine-functionalized NPs. We performed detailed Monte Carlo computer simulations that elucidate the relation between the matrix density and drug release kinetics. We found good agreement between the simulation model and the experimental results for drug release as a function of time. Also, we compared the cellular uptake between amine-functionalized NPs and carboxyl-functionalized NPs, as a higher cellular uptake of NPs leads to improved cisplatin delivery. The amine-functionalized NPs can deliver 3.5 times more cisplatin into cells than the carboxyl-functionalized NPs. The cytotoxic efficacy of both the amine-functionalized NPs and the carboxyl-functionalized NPs showed a strong correlation with the cisplatin release profile, and the latter showed a strong correlation with the NP matrix density.

Abstract

We utilized a novel ratiometric nanoquantum dot fluorescence resonance energy transfer (NQD-FRET) optical sensor to quantitatively measure oxygen dynamics from single cell microdomains during hypoxic episodes as well as during 4-aminopyridine (4-AP)-induced spontaneous seizure-like events in rat hippocampal slices. Coupling oxygen sensing with electrical recordings, we found the greatest reduction in the O2 concentration ([O2]) in the densely packed cell body stratum (st.) pyramidale layer of the CA1 and differential layer-specific O2 dynamics between the st. pyramidale and st. oriens layers. These hypoxic decrements occurred up to several seconds before seizure onset could be electrically measured extracellularly. Without 4-AP, we quantified a narrow range of [O2], similar to the endogenous hypoxia found before epileptiform activity, which permits a quiescent network to enter into a seizure-like state. We demonstrated layer-specific patterns of O2 utilization accompanying layer-specific neuronal interplay in seizure. None of the oxygen overshoot artifacts seen with polarographic measurement techniques were observed. We therefore conclude that endogenously generated hypoxia may be more than just a consequence of increased cellular excitability but an influential and critical factor for orchestrating network dynamics associated with epileptiform activity., (Copyright © 2014 the American Physiological Society.)

46. Asynchronous Magnetic Bead Rotation (AMBR) Microviscometer for Label-Free DNA Analysis.

Abstract

We have developed a label-free viscosity-based DNA detection system, using paramagnetic beads as an asynchronous magnetic bead rotation (AMBR) microviscometer. We have demonstrated experimentally that the bead rotation period is linearly proportional to the viscosity of a DNA solution surrounding the paramagnetic bead, as expected theoretically. Simple optical measurement of asynchronous microbead motion determines solution viscosity precisely in microscale volumes, thus allowing an estimate of DNA concentration or average fragment length. The response of the AMBR microviscometer yields reproducible measurement of DNA solutions, enzymatic digestion reactions, and PCR systems at template concentrations across a 5000-fold range. The results demonstrate the feasibility of viscosity-based DNA detection using AMBR in microscale aqueous volumes.

Abstract

In this report we demonstrate the outstanding advantages of multifunctional nanoplatforms for cancer-imaging and therapy. The non-toxic polyacrylamide (PAA) nanoparticles (size:18-25 nm) formulation drastically changed the pharmacokinetic profile of the ¹²⁴I- labeled chlorophyll-a derivative (formulated in 10% ethanol in PBS) with a remarkable enhancement in tumor uptake, and significantly reduced uptake in spleen and liver. Among the various nanoformulations investigated, the ¹²⁴I- labeled photosensitizer (dose: 0.6142 MBq), and the cyanine dye-nanoparticles (CD-NP) conjugate (dose 0.3 μmol/kg) in combination showed great potential for tumor imaging (PET/NIR fluorescence) in BALB/c mice bearing Colon26 tumors. Compared to free non-labeled photosensitizer, the corresponding PAA nanoformulation under similar treatment parameters showed a remarkable enhancement in long-term tumor cure by PDT (photodynamic therapy) and provides an opportunity to develop a single nanoplatform for tumor-imaging (PET/fluorescence) and phototherapy, a practical "See and Treat" approach.

Abstract

This paper demonstrates a proof-of-principle for a new signal transduction method for protein detection called Bead Assembly Magnetorotation (BAM). BAM is based on using the target protein to mediate the formation of aptamer-coated 1 μm magnetic beads into a bead assembly, formed at the bottom of a 1 μL hanging droplet. The size, shape and fractal dimension of this bead assembly all depend on the protein concentration. The protein concentration can be measured in two ways: by magnetorotation, in which the rotational period of the assembly correlates with the protein concentration, or by fractal analysis. Additionally, a microscope-free magnetorotation detection method is introduced, based on a simple laser apparatus built from standard laboratory components. The determination of oxygen levels in blood and other tissues in vivo is critical for ensuring proper body functioning, for monitoring the status of many diseases, such as cancer, and for predicting the efficacy of therapy. Here we demonstrate, for the first time, a lifetime-based photoacoustic technique for the measurement of oxygen in vivo, using an oxygen sensitive dye, enabling real time quantification of blood oxygenation. The results from the main artery in the rat tail indicated that the lifetime of the dye, quantified by the photoacoustic technique, showed a linear relationship with the blood oxygenation levels in the targeted artery.

50. Targeted Blue Nanoparticles as Photoacoustic Contrast Agent for Brain Tumor Delineation.

Abstract

Distinguishing a tumor from non-neoplastic tissue is a challenging task during cancer surgery. Several attempts have been made to use visible or fluorescent agents to aid in the visualization of a tumor during surgery. We describe a novel method to delineate brain tumors, using a highly sensitive photoacoustic imaging technique that is enhanced by tumor-targeting blue nanoparticles serving as a contrast agent. Experiments on phantoms and on rat brains, ex vivo , demonstrate the high sensitivity of photoacoustic imaging in delineating tumors containing contrast agent at a concentration much lower than needed for visualization by the naked eye. The limit of detection of the system for the nanoparticles is about 0.77 μg/mL in water (equivalent to 0.84 μmol/L Coomassie Blue dye). The present exploratory study suggests that photoacoustic imaging, when used with strongly optical absorbing contrast agents, could facilitate cancer surgery intraoperatively by revealing the distribution and extent of the tumor.
