25 páginas, 6 figuras, 2 tablas, Characterization of the genetic landscape of Alzheimer's disease (AD) and related dementias (ADD) provides a unique opportunity for a better understanding of the associated pathophysiological processes. We performed a two-stage genome-wide association study totaling 111,326 clinically diagnosed/'proxy' AD cases and 677,663 controls. We found 75 risk loci, of which 42 were new at the time of analysis. Pathway enrichment analyses confirmed the involvement of amyloid/tau pathways and highlighted microglia implication. Gene prioritization in the new loci identified 31 genes that were suggestive of new genetically associated processes, including the tumor necrosis factor alpha pathway through the linear ubiquitin chain assembly complex. We also built a new genetic risk score associated with the risk of future AD/dementia or progression from mild cognitive impairment to AD/dementia. The improvement in prediction led to a 1.6- to 1.9-fold increase in AD risk from the lowest to the highest decile, in addition to effects of age and the APOE ε4 allele., This work was funded by a grant (EADB) from the EU Joint Programme – Neurodegenerative Disease Research. - Abstract
Genetic discoveries of Alzheimer’s disease are the drivers of our understanding, and together with polygenetic risk stratification can contribute towards planning of feasible and efficient preventive and curative clinical trials. We first perform a large genetic association study by merging all available case-control datasets and by-proxy study results (discovery n = 409,435 and validation size n = 58,190). Here, we add six variants associated with Alzheimer’s disease risk (near APP, CHRNE, PRKD3/NDUFAF7, PLCG2 and two exonic variants in the SHARPIN gene). Assessment of the polygenic risk score and stratifying by APOE reveal a 4 to 5.5 years difference in median age at onset of Alzheimer’s disease patients in APOE ɛ4 carriers. Because of this study, the underlying mechanisms of APP can be studied to refine the amyloid cascade and the polygenic risk score provides a tool to select individuals at high risk of Alzheimer’s disease. © 2021, The Author(s).

- Abstract
Genetic discoveries of Alzheimer’s disease are the drivers of our understanding, and together with polygenetic risk stratification can contribute towards planning of feasible and efficient preventive and curative clinical trials. We first perform a large genetic association study by merging all available case-control datasets and by-proxy study results (discovery n = 409,435 and validation size n = 58,190). Here, we add six variants associated with Alzheimer’s disease risk (near APP, CHRNE, PRKD3/NDUFAF7, PLCG2 and two exonic variants in the SHARPIN gene). Assessment of the polygenic risk score and stratifying by APOE reveal a 4 to 5.5 years difference in median age at onset of Alzheimer’s disease patients in APOE ɛ4 carriers. Because of this study, the underlying mechanisms of APP can be studied to refine the amyloid cascade and the polygenic risk score provides a tool to select individuals at high risk of Alzheimer’s disease.

Methods: We constructed a non-polar mutant of the sensor protein NtrY of the system, and then analysed the symbiosis of this mutant with soybean plants. Results: We found that the ntrY mutant was defective in plant dry weight, nitrogen content and nitrogenase activity. Haem c-staining of cbb3 oxidase components showed a clear reduction in the expression of this terminal oxidase in ntrY bacteroids with respect to wild type bacteroids. Such defect in cbb3 expression correlated with a decreased respiratory capacity of the bacteroids produced by the ntrY mutant. The objectives were to determine the effectiveness of these actions, identify variables associated with success, and propose a successful indirect free kick model. After registering 447 indirect free kicks during the 64 games of the World Cup in South Africa 2010, and performing statistical analysis, results indicate a low efficiency in the shot, shot between the three posts and goal. 64.3 % of goals that came from indirect free kick contributed to score points in the final result. The mode of sending, number of attackers involved and the offensive organization are variables that have been revealed as modulators of effectiveness. These statements could be a starting point to equip coaches with new tactical tools.

Las tablas tradicionales y rígidas en socorrismo acuático han sido rechazadas en muchos servicios de socorrismo por los problemas que generaban. La experiencia con tablas hinchables (AIRSUPRA) ha demostrado muchas ventajas y las objeciones sobre este material están desapareciendo y ya comienzan a utilizarse en servicios de socorrismo, que antes eran reticentes. El objetivo de este estudio fue analizar el efecto del uso de la tabla AIRSUPRA en el tiempo de aproximación en una distancia de 100 metros, con una muestra de 16 socorristas profesionales (13 hombres y 3 mujeres), certificados y actualizados en técnicas de rescate, pero sin experiencia con tablas AIRSUPRA. Los resultados demuestran que cuando se realiza la aproximación con la tabla AIRSUPRA (TR1) se obtiene un tiempo de 54.13 + 8.58, mientras que cuando se realiza la aproximación a nado el tiempo es 93.19 + 25.52, con una diferencia de 39.06 segundos, estadísticamente significativa (p, The use of traditional and rigid boards in aquatic lifesaving cause more problems than advantages, for this reason was frequently rejected in many lifesaving services. El ahogamiento es una de las principales causas de muerte en el mundo y en España. Los socorristas ejercen una labor de prevención y vigilancia pero su labor es estacional y temporal. En muchos lugares, la primera respuesta a la emergencia, cuando los socorristas no están de servicio, depende de la policía, a la que se le requiere habilidades de rescate y reanimación cardiopulmonar (RCP). El objetivo de este estudio es determinar la capacidad de rescate y efecto de la fatiga sobre la calidad de la RCP de un grupo de diez policías costeros cuya área de influencia es el litoral de Vigo. El diseño fue cuasi-experimental con dos factores (pretest basal/postest rescate). Los policías pudieron realizar el rescate acuático rápido y seguro 417 ± 54,5 seg, a nivel de lactacidema se registró 12,27 ± 2,36 mmol. La fatiga inducida por el rescate afectó negativamente a la calidad de la compresiones en la RCP (p = 0,002), Drowning is a leading cause of death worldwide and in Spain. Lifeguards exert vigilance and prevention efforts but their work is seasonal and temporary. In many places the first emergency response when lifeguards are not on duty, depends on the police, which are required rescue skills and cardiopulmonary resuscitation (CPR). The objective of this study is to determine the ability of lifesaving and effect of fatigue on the quality of CPR of a group of ten coastal police whose area of influence is the coast of Vigo. The design was quasiexperimental with two factors (basal pretest / posttest rescue). Cops with basic training could perform fast and safe water rescue 417 ± 54.5 seconds, lactate level was recorded 12.27 ± 2.36 mmol. Induced fatigue during resuce effort had a nevative effect on the quality of compressions in CPR (p = 0.002)

[Resumen]: Objetivo: Alcanzar un conocimiento más profundo (y con el soporte empírico adecuado) de los saques de esquina en el fútbol de máximo nivel. Se pretende conocer la incidencia real de este tipo de acciones durante los partidos, así como describir cuáles son las prácticas habituales. A continuación, se pretende identificar aquellas variables que puedan estar asociadas a la eficacia de estas acciones. Método: Se plantean dos tipos de aproximaciones complementarias en el análisis de los datos: una de carácter univariada, y otra bivariada. A nivel univariado se trata de describir cuáles son las características de la ejecución de estas acciones (número, forma de ejecución...). A nivel bivariado, mediante la reali-zación de tablas de contingencia (acompa ˜nadas del contraste Chi-cuadrado y medidas de asociación) se intentará identificar aquellas variables que puedan estar asociadas a la eficacia alcanzada. Resultados: Después del registro de 345 saques de esquina ejecutados durante la Eurocopa 2012, los resultados indican una baja eficacia en el remate de este tipo de acciones. En cambio, los goles que proceden de saque de esquina sí presentan una transcendencia importante en el resultado final. Para alcanzar el éxito en este tipo de acciones, deben intervenir entre tres y cuatro atacantes y producirse una organización ofensiva dinámica. Conclusiones: Los datos empíricos disponibles han puesto en valor la necesidad de un trabajo táctico más refinado de estas acciones, basado en asociaciones entre los jugadores y movimientos de engaño., [Abstract]: Objective: Reaching a deeper knowledge (and with proper empirical support) of the corners in the top-level football. It is intended to determine the real impact of this type of actions during matches and describe what the current practices are. Then, it is intended to identify those variables that may be associated with the effectiveness of these actions. Method: Two complementary approaches arise in the analysis of data: one univariate and other bivariate. At univariate level, it comes to describe what are the characteristics of the execution of these actions (number, form of execution...). At bivariate level, by making contingency tables (accompanied by Chi-square contrast and association measures) it will try to identify those variables that may be associated with the achieved efficiency. Results: After registration of 345 corners executed during Euro 2012, the results indicate a low efficiency in the auction of such actions. Instead, the goals coming from corner have an important significance in the final result. To succeed in this kind of actions should intervene between three and four attackers and a dynamic offensive organization should occur. A seguir, se pretende identificar as variáveis que podem estar associados com a eficácia destas ac¸ ões. Método: Surgem dois tipos de abordagens complementares na análise de dados: uma de caráter univa-riada e outra bivariada. A univariada trata-se de descrever quais são as características da execuc¸ ão destas ac¸ ões (número, forma de execuc¸ão ...). A bivariada, mediante a realizac¸ ão de tabelas de contingência (acompanhada do contraste qui-quadrado e medidas de associac¸ ão) tentará identificar as variáveis que podem estar associados com a eficiência alcanc¸ ada. Resultados: Após o registro de 345 cobranc¸ as de escanteios executadas durante a Eurocopa 2012, os resultados indicam uma baixa eficiência na finalizac¸ ão de tais ac¸ ões. Entretanto, os objetivos que prece-dem de cobranc¸ as de escanteios têm uma transcendência importante no resultado final. Este estudio nace con la necesidad de establecer criterios básicos y descripción de recomendaciones para mejorar el nivel de atención de los socorristas acuáticos profesionales en los espacios acuáticos naturales, concretamente en el trabajo realizado en las playas, con el objetivo de aumentar la seguridad de estas zonas de baño. El estudio se ha desarrollado en la Comunidad Autónoma de Cataluña, concretamente en tres playas de varios municipios de la provincia de Barcelona. Se puede afirmar que tras esta investigación la mayor parte de los socorristas acuáticos que han participado en el estudio valoraron positivamente las medidas y criterios propuestos, asumiendo que con los mismos se puede desempeñar mejor su trabajo, This study arises from the need to establish basic criteria and the description of recommendations to improve the level of responsiveness of lifeguards in natural aquatic areas, specifically in the work done on the beaches, in order to increase the safety of these bathing areas. The study was developed Cataluña, in three beaches of different municipalities in the province of Barcelona. We can aver after this research that most of the lifeguards who participated in the study value positively the proposed measures and criteria, through the assumption that these features help improve their work

Objetivo: El objetivo es analizar hasta qué edad podría influir una actitud radical en la esperanza de vida de los pacientes con cánceres de próstata clínicamente localizados Gleason 8-10. Material y métodos: Se hace un estudio retrospectivo de 80 pacientes con cáncer de próstata localizado T1-2N0-XM0 Gleason 8-10. Los pacientes se estratificaron en 2 grupos: grupo A 34 pacientes que por diversos motivos, edad superior a 75 años, riesgo quirúrgico, elección del paciente u otros motivos, habían sido tratados con bloqueo androgénico y grupo B, 46 pacientes sometidos a prostatectomía radical. Las variables analizadas son: mortalidad por cáncer y supervivencia cáncer específica. Resultados: Características de los pacientes: mediana de edad grupo A 75 años(66-84) y grupo B 64 años(56-75)(p=0,033); mediana de tiempo de seguimiento grupo A 54 meses(20-180) y grupo B 37 meses(12-140)(p=0,016); mediana de Gleason grupo A 8(8-10) y grupo B 8(8-10)(p=0,144); porcentaje de T1 grupo A 24% y grupo B 41%(p=0,096); mediana de PSA grupo A 10(4-91) y grupo B 12(4-71)(p=0,269). La mortalidad cáncer específica del grupo A es de 24(71%) y en el grupo B 3(7%) (p=0.000). A los 5 años han fallecido 20(59%) pacientes como consecuencia del cáncer en el grupo A y 1(2%) de los pacientes en el grupo B. Las curvas de Kaplan-Meier y test de Log-Rank muestran diferencias significativas en la supervivencia cáncer específicas entre los 2 grupos. La regresión de Cox muestra que las posibilidades de morir por cáncer de próstata son superiores en el grupo de pacientes que no recibieron tratamiento con intención curativo, el riesgo relativo (HR 95% IC) en el grupo A es de 6.826(2.032-22.931). Conclusiones: Algo más de la mitad de los pacientes con cáncer de próstata localizado Gleason 8-10 tratados de forma conservadora, mueren por cáncer dentro de los 5 años siguientes al diagnóstico. Los pacientes con una expectativa de vida superior a 5 años pueden beneficiarse de una actitud curativa. Objective: The aim is to analyse until which age a radical procedure could have an influence on life expectancy of patients with localized prostate cancer Gleason 8-10. El objetivo de este estudio consiste en recabar información sobre los conocimientos y actitudes de una población universitaria acerca de la investigación genética, sus opiniones acerca de cómo y para qué debería ser usada la información personal de este tipo y su disposición a someterse a pruebas genéticas predictivas. La metodología consistió en la administración colectiva, anónima y voluntaria de un cuestionario adaptado de otros similares a una muestra de 232 estudiantes de primeros cursos de Enfermería y Psicología de la Universidad de Murcia. Los resultados muestran un amplio apoyo a la investigación en genética, sobre todo en lo que respecta a las nuevas posibilidades para prevenir, diagnosticar y tratar enfermedades. No obstante hay ciertas áreas que son percibidas como límites éticos, por lo que se percibe necesario un control riguroso en el uso y el almacenaje de la información genética. ABSTRACT The objective of this study consists on the gathering of information about knowledge and attitudes regarding genetic research from a university student sample, its opinions about the use of personal information of this type and its disposition to undergo predictive genetic testing. Methodology consisted on a collective, anonymous and voluntary administration of an adapted questionnaire to a sample of 232 students from first courses of Infirmary and Psychology at Universidad de Murcia. Results show a wide support of genetic research, mainly regarding the new possibilities for preventing, diagnosing and treating disease.Nevertheless there are certain areas which are perceived as ethical limits. A rigorous control in the use and the storage of the genetic information is perceives as necessary.

Aquesta guia d’acompanyament s’integra en una de les línies estratègiques que defineix l’atenció sanitària accessible, resolutiva i integral per donar resposta a aquesta necessitat d’atenció per a les mares i pares que es troben en la situació de viure la mort d’un fill. La pérdida de un hijo durante la gestación, o poco tiempo después de nacer, no es un hecho habitual, pero sí es un hecho prevalente en nuestra sociedad. Tanto la intensidad como la complejidad de este hecho traumático hacen que el sistema sanitario considere esta situación de interés especial. Esta guía de acompañamiento se integra en una de las líneas estratégicas que define la atención sanitaria accesible, resolutiva e integral para dar respuesta a esta necesidad de atención para las madres y padres que se encuentran en la situación de vivir la muerte de un hijo. The loss of a child during pregnancy, or shortly after birth, is not a common occurrence, but it is a prevalent fact in our society. Both the intensity and the complexity of this traumatic event make the health system consider this situation of special interest. This accompanying guide is integrated into one of the strategic lines that defines affordable, comprehensive and comprehensive health care to respond to this need for care for mothers and parents who are in the situation of living the death of a son.

45. External Load Evaluation in Elite Futsal: Influence of Match Results and Game Location with IMU Technology.

- Abstract
The purpose of this study was to assess the external load demands in futsal, considering both home and away matches and their outcomes, in order to plan microcycles throughout the season based on the external load of each match. The external load of 10 players from a First Division team in the Spanish Futsal League was recorded throughout 15 official matches in the first half of the league championship. The players' external load was monitored using OLIVER devices. To analyse the influence of the match outcome and location on the external load, a univariate general linear model (GLM) analysis was conducted with Bonferroni post hoc. There are no differences between the variables neither comparing results nor location factors, except for accelerations of 2 to 3 m/s
2 (m) per minute and the number of accelerations of 2 to 3 m/s2 per minute, reporting higher value winnings at home than away ( p < 0.05). The location and results are not factors that influence on external load in futsal matches, except the number and distance performed in accelerations and distance covered at a low to medium speed. These findings are important for planning microcycles and providing the appropriate dosage to each player to achieve optimal performance in matches.- Published

46. Cornerball: a new alternative sport proposal for school physical education.

Physical Education has had to evolve and change throughout history to adapt to the demands of society. As a result, teachers have had to seek pedagogical alternatives to ensure that students are active, motivated, and engaged in the classroom. This approach allows for the development of motor, cognitive, and socio-emotional skills in students, ultimately contributing to the holistic development of the individual, which is the primary goal of education. Out of this intrinsic need for Physical Education, Alternative Sports have emerged, providing opportunities for different types of learning compared to more traditional sports. Cornerball, a hybrid between split-court and wall-based sports, played in a somewhat unique playing area-a 90° corner, is introduced with the aim of offering a new Alternative Sport. The objective of this descriptive study is to present a new pedagogical proposal designed for the educational context, highlighting its characteristics and fundamental aspects to consider, such as rules, the playing field, participants, and methodological strategies for its implementation within a Physical Education classroom. Three different concentrations of eucalyptol and menthol were prepared following ISO 10993-5 guidelines (1, 5, and 10 mM). GFs were isolated from extracted teeth from healthy donors. The following parameters were assessed: cell viability via MTT, Annexin-V-FITC and 7-AAD staining, and IC
50 assays; cell migration via horizontal scratch wound assay; and cell oxidative stress via reactive oxygen species assay. Data were analyzed using one-way ANOVA and Tukey's post hoc test. Statistical significance was established at p < 0.05. Eucalyptol and Menthol exhibited high cytotoxicity on gingival fibroblasts, as evidenced by cytotoxicity assays. Eucalyptol showed lower levels of cytotoxicity than menthol, compared to the control group. The cytotoxicity of the tested substances increased in a concentration-dependent manner. The same occurred in a time-dependent manner, although even 10 min of exposure to the tested substances showed a high cytotoxicity to the GFs. Commercially available products for oral application with these substances in their composition should be tested for cytotoxicity before their use.- Published

- Abstract
Plasma biomarkers for Alzheimer's disease (AD) are a promising tool that may help in early diagnosis. However, their levels may be influenced by physiological parameters and comorbidities that should be considered before they can be used at the population level. For this purpose, we assessed the influences of different comorbidities on AD plasma markers in 208 cognitively unimpaired subjects. We analyzed both plasma and cerebrospinal fluid levels of Aβ40, Aβ42, and p-tau181 using the fully automated Lumipulse platform. The relationships between the different plasma markers and physiological variables were studied using linear regression models. The mean differences in plasma markers according to comorbidity groups were also studied. The glomerular filtration rate showed an influence on plasma Aβ40 and Aβ42 levels but not on the Aβ42/Aβ40 ratio. The amyloid ratio was significantly lower in diabetic and hypertensive subjects, and the mean p-tau181 levels were higher in hypertensive subjects. The glomerular filtration rate may have an inverse relationship on plasma Aβ40 and Aβ42 levels but not on the amyloid ratio, suggesting that the latter is a more stable marker to use in the general population. Cardiovascular risk factors might have a long-term effect on the amyloid ratio and plasma levels of p-tau181.

- Abstract
Background: Video-oculography constitutes a highly-sensitive method of characterizing ocular movements, which could detect subtle premotor changes and contribute to the early diagnosis of Parkinson's disease (PD)., Objective: To investigate potential oculomotor differences between idiopathic PD (iPD) and PD associated with the G2019S variant of LRRK2 (L2PD), as well as to evaluate oculomotor function in asymptomatic carriers of the G2019S variant of LRRK2., Methods: The study enrolled 129 subjects: 30 PD (16 iPD, 14 L2PD), 23 asymptomatic carriers, 13 non-carrier relatives of L2PD patients, and 63 unrelated HCs. The video-oculographic evaluation included fixation, prosaccade, antisaccade, and memory saccade tests., Results: We did not find significant differences between iPD and L2PD. Compared to controls, PD patients displayed widespread oculomotor deficits including larger microsaccades, hypometric vertical prosaccades, increased latencies in all tests, and lower percentages of successful antisaccades and memory saccades. Non-carrier relatives showed oculomotor changes with parkinsonian features, such as fixation instability and hypometric vertical saccades. Asymptomatic carriers shared multiple similarities with PD, including signs of unstable fixation and hypometric vertical prosaccades; however, they were able to reach percentages of successful antisaccade and memory saccades similar to controls, although at the expense of longer latencies. Classification accuracy of significant oculomotor parameters to differentiate asymptomatic carriers from HCs ranged from 0.68 to 0.74, with BCEA, a marker of global fixation instability, being the parameter with the greatest classification accuracy., Conclusions: iPD and LRRK2-G2019S PD patients do not seem to display a differential oculomotor profile. Several oculomotor changes in asymptomatic carriers of LRRK2 mutations could be considered premotor biomarkers.
