1. CriteriaMapper: establishing the automatic identification of clinical trial cohorts from electronic health records by matching normalized eligibility criteria and patient clinical characteristics

Abstract

Abstract The use of electronic health records (EHRs) holds the potential to enhance clinical trial activities. However, the identification of eligible patients within EHRs presents considerable challenges. We aimed to develop a CriteriaMapper system for phenotyping eligibility criteria, enabling the identification of patients from EHRs with clinical characteristics that match those criteria. We utilized clinical trial eligibility criteria and patient EHRs from the Mount Sinai Database. The CriteriaMapper system was developed to normalize the criteria using national standard terminologies and in-house databases, facilitating computability and queryability to bridge clinical trial criteria and EHRs. The system employed rule-based pattern recognition and manual annotation. Our system normalized 367 out of 640 unique eligibility criteria attributes, covering various medical conditions including non-small cell lung cancer, small cell lung cancer, prostate cancer, breast cancer, multiple myeloma, ulcerative colitis, Crohn’s disease, non-alcoholic steatohepatitis, and sickle cell anemia. About 174 criteria were encoded with standard terminologies and 193 were normalized using the in-house reference tables. The agreement between automated and manual normalization was high (Cohen’s Kappa = 0.82), and patient matching demonstrated a 0.94 F1 score. Our system has proven effective on EHRs from multiple institutions, showing broad applicability and promising improved clinical trial processes, leading to better patient selection, and enhanced clinical research outcomes.

Abstract

The Weihe Basin, enclosed by the Chinese Loess Plateau to the north and the Qinling Mountains to the south, is an outstanding, world-class continental site for obtaining high-resolution multi-proxy records that reflect environmental changes spanning most of the Cenozoic. Previous geophysical and sedimentary studies indicate that the basin hosts 6000–8000 m thick fluvial–lacustrine sedimentary successions spanning the Eocene to Holocene. This sedimentary record provides an excellent and unique archive to decipher long-term tectonic–climate interactions related to the uplift of the Tibetan Plateau, the onset/evolution of the Asian monsoon, and the development of the biogeography of East Asia. Owing to its location at the interface of the opposing westerly and Asian monsoon circulation systems, the Weihe Basin also holds enormous promise for providing a record of changes in these circulation systems in response to very different boundary conditions since the Eocene. To develop an international scientific drilling programme in the Weihe Basin, the Institute of Earth Environment, Chinese Academy of Sciences, organized a dedicated workshop with 55 participants from eight countries. The workshop was held in Xi'an, China, from 15 to 18 October 2019. Workshop participants conceived the key scientific objectives of the envisaged Weihe Basin Drilling Project (WBDP) and discussed technical and logistical aspects as well as the scope of the scientific collaboration in preparation for a full drilling proposal for submission to the International Continental Scientific Drilling Program (ICDP). Workshop participants mutually agreed to design a two-phase scientific drilling programme that will in a first phase target the upper 3000 m and in a second phase the entire up to 7500 m thick sedimentary infill of the basin. For the purpose of the 7500 m deep borehole, the world's only drill rig for ultra-deep scientific drilling on land, Crust 1, which previously recovered the entire continental Cretaceous sediments in the Songliao Basin, will be deployed in the WBDP.

Abstract

Ion Cyclotron Range of Frequencies (ICRF) heating and Neutral Beam Injection (NBI) can have synergy due to the acceleration of NBI beam ions by ICRF wave fields at their harmonics. To understand the influence of ICRF-NBI synergy on fast ion distribution and plasma performance, dedicated experiments and TRANSP simulations have been carried out on EAST. The simulation results are consistent with the experimental results. They show that the ICRF-NBI synergy not only accelerates the NBI beam ions with energy lower than 80 keV to energy larger than 300 keV, but also generates fusion neutrons with energy larger than 3 MeV. Moreover, ICRF-NBI synergy improves the plasma performance by increasing the poloidal beta, plasma stored energy, core ion temperature, total neutron yield and kinetic pressure. In a typical H-mode plasma with 1.0 MW NBI and 1.5 MW ICRF power, it was observed that ICRF-NBI synergy increases the poloidal beta, plasma stored energy, core ion temperature and neutron yield by ∼35%, 33%, 22% and 80%, respectively. Various parameter scans show that the ICRF-NBI synergetic effects can be enhanced by decreasing the minority ion concentration or the distance between the harmonic resonance and magnetic axis, or by increasing the ICRF heating power or NBI beam energy. Consequently, this leads to a generation of fast ions with higher energy. For instance, the maximum energy of the fast ion tail increases from 300 to 600 keV as n(H) decreases from 5% to 0.1%.

Abstract

Dried pollen grains of three date palm (male) cultivars grown in Oman were stored either in a freezer (-18°C), a refrigerator (4 to 5°C), or at room temperature (23 to 25°C) for 6 or 12 months. The three cultivars include Khori, Medjahdil, and Bahlani. Germination percentage was determined after 6 hours incubation in aerated and non- aerated liquid media. The results showed that date palm pollen was better adapted to refrigerator storage than freezing for all three cultivars, while room temperature storage significantly reduced pollen germination. Aeration was found to enhance germination of refrigerator- and freezer-stored pollen, but decreased that of pollen stored at room temperature for 6 months. The results also indicated that refrigeration might have caused the inactivation of pollen germination. The latter could either be overcome by aeration or reversed if pollen is refrigerated for a longer period. Room temperature storage directly reduced viability and did not induce temporary inactivation of pollen.

Abstract

The larvae of Echinococcus (hydatidcyst) can parasitize humans and animals, causing a serious zoonotic disease-echinococcosis. The life history of Echinococcus is complicated, and as the disease progresses slowly after infection, early diagnosis is difficult to establish. Due to the limitations of imaging and immunological diagnosis in this respect, domestic and foreign scholars have established a variety of molecular detection techniques for the pathogen Echinococcus over recent years, mainly including nested polymerase chain reaction (PCR), multiplex PCR, real-time quantitative PCR, and nucleic acid isothermal amplification technology. In this article, the research progress of molecular detection technology for Echinococcus infection currently was reviewed and the significance of these methods in the detection and diagnosis of hydatid and hydatid diseases was also discussed.

9. Exploring the impact of media use on wellbeing following a natural disaster

Abstract

Media use can be beneficial in many ways, but little is known about how it might improve wellbeing outcomes following a traumatic natural disaster. Survivors (n = 491) of deadly Hurricane Michael, which struck the Florida (USA) coastline in 2018, completed an online survey, reporting indicators of post-traumatic growth (PTG) and stress (PTSS). A serial mediation model explored how hurricane-related stressors were related to both outcomes, as mediated by approach, avoidant, and support-seeking coping strategies and post-hurricane hedonic, eudaimonic, and self-transcendent media use as coping tools. Factors contributing to each type of post-hurricane media use were also explored. Results indicate that hurricane-related stressors were associated with PTG, serially mediated through approach coping strategies and self-transcendent media use, thus providing some of the first empirical evidence of the longer-term, beneficial wellbeing effects of media use on survivors of trauma. Additionally, hurricane-related stressors were associated with avoidant coping strategies, which were associated with increased eudaimonic media use. However, hedonic and eudaimonic media use were not associated with PTSS or PTG. Finally, factors known to be associated with media use were not predictive of post-hurricane media use, perhaps suggesting that media play a different role in survivors' lives in the months following a traumatic event.

Abstract

Effective marine management requires balancing conservation and sustainable use of resources. Food web models are useful for simulating direct and indirect effects of management scenarios on ecosystem functioning by using multiple indicators. However, a key challenge is consolidating these indicators into a single, comprehensive, measure, which is often required to guide management decisions, such as in Strategic Environmental Assessment. This study applies spatial multi-criteria analysis to food web model outputs to develop a single index for different marine management applications. We applied this framework to the case of the “Tegnùe di Chioggia”, a Special Area of Conservation (SAC; IT3250047) under the Natura 2000 European network, located in the northern Adriatic Sea (Italy). This area, characterised by the presence of biogenic rocky outcrops, currently lacks a formal management plan. Using the Ecospace module of the Ecopath with Ecosim software, we simulated three management scenarios: 1) SAC expansion; 2) winter artisanal fishing in the SAC; 3) a combination of both. In line with ecosystem-based management, we focused on economically important trophic groups in the region, such as the Mediterranean mussel, Mytilus galloprovincialis, and striped venus clam, Chamelea gallina, which are present near the SAC. We also considered the efforts of the local fishing fleets. Ecosystem structure and functioning indicators, generated by the spatial food-web model, were linked to three criteria aligned with the management priorities of the area: nature conservation, aquaculture productivity, and fishing productivity. These criteria were aggregated into a final score to compare the management scenarios. The results showed that none of the scenarios would significantly alter community composition or ecosystem functioning compared to the current situation. However, they did show contrasting responses in the food web model. The SAC expansion scenario notably increased total biomass and commercial fish biomass, especially pectinids and cephalopods. The fishing scenario had a minimal impact on trophic groups. Ecosystem resilience and structure indicators were less sensitive to management scenarios than biomass indicators. However, the multi-criteria analysis revealed that the fishing scenario limited the benefits of expanding the SAC, due to reduced catches. The final score effectively ranked proposed scenarios, highlighting key indicators that influenced these variations. The proposed approach shows potential for supporting participatory modelling and engaging stakeholders in developing management scenarios.

Abstract

Over the last 2 decades, there have been significant advances in the 3D modelling of geological structures via the incorporation of geological knowledge into the model algorithms. These methods take advantage of different structural data types and do not require manual processing, making them robust and objective. Igneous intrusions have received little attention in 3D modelling workflows, and there is no current method that ensures the reproduction of intrusion shapes comparable to those mapped in the field or in geophysical imagery. Intrusions are usually partly or totally covered, making the generation of realistic 3D models challenging without the modeller's intervention. In this contribution, we present a method to model igneous intrusions in 3D considering geometric constraints consistent with emplacement mechanisms. Contact data and inflation and propagation direction are used to constrain the geometry of the intrusion. Conceptual models of the intrusion contact are fitted to the data, providing a characterisation of the intrusion thickness and width. The method is tested using synthetic and real-world case studies, and the results indicate that the method can reproduce expected geometries without manual processing and with restricted datasets. A comparison with radial basis function (RBF) interpolation shows that our method can better reproduce complex geometries, such as saucer-shaped sill complexes.

Abstract

SDSS J134244.4+053056 is a tidal disruption event candidate with strong temporal coronal line emitters and a long fading, mid-infrared dust echo. We present detailed analyses of X-ray emission from a Swift/XRT observation in 2009 and the most recent XMM-Newton/pn observation in 2020. The two spectra can be modeled with hard and soft components. While no significant variability is detected in the hard component above 2 keV between these two observations, the soft X-ray emission in 0.3-2 keV varies by a factor of $\sim5$. The luminosity of this soft component fades from $\sim1.8\times10^{41}$ to $\sim3.7\times10^{40}$ erg s$^{-1}$ from the observation in Swift to that of XMM-Newton, which are 8 and 19 years after the outburst occurred, respectively. The evolution of luminosity matches with the $t^{-5/3}$ decline law well; there is a soft X-ray peak luminosity of 10$^{44}$ erg s$^{-1}$ at the time of the optical flare. Furthermore, the spectra of the soft component harden slightly in the decay phase, in which the photon index $\Gamma$ varies from $4.8^{+1.2}_{-0.9}$ to $3.7\pm0.5$, although they are consistent with each other if we consider the uncertainties. Additionally, by comparing the BH mass estimate between the $M-\sigma$ correlation, the broad H$\alpha$ emission, and the fundamental plane relation of BH accretion, we find that a value of $\sim10^{5}$Msun is favored. If so, taking its X-ray spectral variation, luminosity evolution, and further support from theory into account, we suggest that SDSS J134244.4+053056 is a long-lived tidal disruption event candidate lasting more than 18 years with an intermediate-mass black hole., Comment: 7 pages, 2 figures, accepted by Astronomy & Astrophysics

Abstract

Stress exposure during the sensitive period of early development has been shown to program the brain and increases the risk to develop cognitive deficits later in life. We have shown earlier that early-life stress (ES) leads to cognitive decline at an adult age, associated with changes in adult hippocampal neurogenesis and neuroinflammation. In particular, ES has been shown to affect neurogenesis rate and the survival of newborn cells later in life as well as microglia, modulating their response to immune or metabolic challenges later in life. Both of these processes possibly contribute to the ES-induced cognitive deficits. Emerging evidence by us and others indicates that early nutritional interventions can protect against these ES-induced effects through nutritional programming. Based on human metabolomics studies, we identified various coffee-related metabolites to be part of a protective molecular signature against cognitive decline in humans. Caffeic and chlorogenic acids are coffee-polyphenols and have been described to have potent anti-oxidant and anti-inflammatory actions. Therefore, we here aimed to test whether supplementing caffeic and chlorogenic acids to the early diet could also protect against ES-induced cognitive deficits. We induced ES via the limited nesting and bedding paradigm in mice from postnatal(P) day 2–9. On P2, mice received a diet to which 0.02% chlorogenic acid (5-O-caffeoylquinic acid) + 0.02% caffeic acid (3′,4′-dihydroxycinnamic acid) were added, or a control diet up until P42. At 4 months of age, all mice were subjected to a behavioral test battery and their brains were stained for markers for microglia and neurogenesis. We found that coffee polyphenols supplemented early in life protected against ES-induced cognitive deficits, potentially this is mediated by the survival of neurons or microglia, but possibly other mechanisms not studied here are mediating the effects. This study provides additional support for the potential of early nutritional interventions and highlights polyphenols as nutrients that can protect against cognitive decline, in particular for vulnerable populations exposed to ES.

17. Insights into the novel function of system Xc- in regulated cell death

Abstract

System Xc-, also named cystine/glutamate antiporter, is an important intracellular antioxidant element. It is composed of the light chain SLC7A11 (xCT) and the heavy chain SLC3A2 (4F2hc) and functions as raw materials for the synthesis of glutathione (GSH). Recent studies have demonstrated that system Xc- plays an important role in different types of regulated cell death, which is referred to cell death controlled by dedicated molecular machinery. It has been shown that system Xc- involves in ferroptosis, apoptosis, and autophagy-dependent cell death, contributing to different diseases and drug resistance, such as cancer, neurological disorders, and cisplatin resistance to cancers. To date, the intervention of system Xc- by its inhibitors or activators displays a beneficial effect on the treatment of certain diseases. In this review, we summarize recent findings on the role of system Xc- in regulated cell death, including molecular mechanisms and potential therapeutic applications.

Abstract

Generation of tolerogenic peripheral regulatory T (pTreg) cells is commonly thought to involve CD103+ gut dendritic cells (DCs), yet their role in commensal-reactive pTreg development is unclear. Using two Helicobacter-specific T cell receptor (TCR) transgenic mouse lines, we found that both CD103+ and CD103– migratory, but not resident, DCs from the colon-draining mesenteric lymph node presented Helicobacter antigens to T cells ex vivo. Loss of most CD103+ migratory DCs in vivo using murine genetic models did not affect the frequency of Helicobacter-specific pTreg cell generation or induce compensatory tolerogenic changes in the remaining CD103– DCs. By contrast, activation in a Th1-promoting niche in vivo blocked Helicobacter-specific pTreg generation. Thus, these data suggest a model where DC-mediated effector T cell differentiation is ‘dominant’, necessitating that all DC subsets presenting antigen are permissive for pTreg cell induction to maintain gut tolerance.

Abstract

X-ray emission from quasars has been detected up to redshift $z=7.5$, although only limited to a few objects at $z>6.5$. In this work, we present new Chandra observations of five $z>6.5$ quasars. By combining with archival Chandra observations of six additional $z>6.5$ quasars, we perform a systematic analysis on the X-ray properties of these earliest accreting supermassive black holes (SMBHs). We measure the black hole masses, bolometric luminosities ($L_{\rm bol}$), Eddington ratios ($\lambda_{\rm Edd}$), emission line properties, and infrared luminosities ($L_{\rm IR}$) of these quasars using infrared and sub-millimeter observations. Correlation analysis indicates that the X-ray bolometric correction (the factor that converts from X-ray luminosity to bolometric luminosity) decreases with increasing $L_{\rm bol}$, and that the UV/optical-to-X-ray ratio, $\alpha_{\rm ox}$, strongly correlates with $L_{\rm 2500}$, and moderately correlates with $\lambda_{\rm Edd}$ and blueshift of CIV emission lines. These correlations are consistent with those found in lower-$z$ quasars, indicating quasar accretion physics does not evolve with redshift. We also find that $L_{\rm IR}$ does not correlate with $L_{\rm 2-10 keV}$ in these luminous distant quasars, suggesting that the ratio of the SMBH growth rate and their host galaxy growth rate in these early luminous quasars are different from those of local galaxies. A joint spectral analysis of the X-ray detected $z>6.5$ quasars yields an average X-ray photon index of $ \Gamma=2.32^{+0.31}_{-0.30}$, steeper than that of low-$z$ quasars. By comparing it with the $\Gamma-\lambda_{\rm Edd}$ relation, we conclude that the steepening of $\Gamma$ for quasars at $z>6.5$ is mainly driven by their higher Eddington ratios., Comment: 16 pages, 10 figures, Accepted for publication in ApJ

Abstract

Tumor metastasis remains the main cause of breast cancer-related deaths, especially the later breast cancer distant metastasis. This study assessed CD44−/CD24−tumor cells in 576 tissue specimens for associations with clinicopathological features and metastasis and then investigated the underlying molecular events. The data showed that level of CD44−/CD24−cells was associated with later postoperative distant tumor metastasis. Furthermore, CD44−/CD24−triple negative cells could spontaneously convert into CD44+/CD24−cancer stem cells (CSCs) with properties similar to CD44+/CD24−CSCs from parental MDA-MB-231 cells in terms of gene expression, tumor cell xenograft formation, and lung metastasisin vitroandin vivo. Single-cell RNA sequencing identified RHBDL2 as a regulator that enhanced spontaneous CD44+/CD24−CSC conversion, whereas knockdown of RHBDL2 expression inhibited YAP/NF-κB signaling and blocked spontaneous CD44−/CD24−cell conversion to CSCs. These data suggested that the level of CD44−/CD24−tumor cells could predict breast cancer prognosis, metastasis, and response to adjuvant therapy.

Abstract

We report observations of a transient source \fermi\ from radio to \grs. \fermi\ was discovered by the {\it Fermi-LAT} in May 2017. Follow-up {\it Swift-XRT} observations revealed three flaring episodes through March 2018, and the peak X-ray flux is about $10^3$ higher than the {\it ROSAT all-sky survey (RASS)} flux upper limit. Optical spectral measurements taken by the {\it Magellan 6.5-m telescope} and the {\it Lick-Shane telescope} both show a largely featureless spectrum, strengthening the BL Lac interpretation first proposed by \citet{Bruni18}. The optical and mid-infrared (MIR) emission goes to a higher state in 2018, when the flux in high energies goes down to a lower level. Our {\it RATAN-600m} measurements at 4.8~GHz and 8.2~GHz do not indicate any significant radio flux variation over the monitoring seasons in 2017 and 2018, nor deviate from the archival {\it NVSS} flux level. During GeV flaring times, the spectrum is very hard ($\Gamma_\gamma\sim$1.7) in the GeV band and at times also very hard (($\Gamma_{\rm X}\lesssim2$) in the X-rays, similar to a high-synchrotron-peak (or even an extreme) BL Lac object, making \fermi\ a good target for ground-based {\it Cherenkov telescopes}., Comment: Submitted in Journal of High Energy Astrophysics

Abstract

Radio frequency (RF) power in the ion cyclotron range of frequencies (ICRF) is one of the primary auxiliary heating techniques for Experimental Advanced Superconducting Tokamak (EAST). In this paper, we will present an overview of the main experimental results from recent campaign on EAST. 1) A dedicated experiment for more than 10MW operations has been carried out on EAST. The major aspects linked to the use of the EAST auxiliary heating systems: ICRF,LHCD and NBI are presented with emphasis on ICRF and LH. The challenges for ICRF coupling on EAST will be described at the low densities typically used for long pulse operation with lithium wall conditioning. 2)This paper also summarizes effects of ICRF power on LH coupling and modification of heat loads on EAST. 3) Co-current rotation driven by ICRF are observed in both L-mode and H-mode discharges. Co-current rotation can be generated by ICRF heating in LHCD plasma. Different rotation behavior observed in LHCD discharges. ICRF can also induce counter-current rotation in LHCD L-mode discharges. 4) A high-frequency magnetic probes (HFBs) system has been developed and successfully applied to detect ion cyclotron emission (ICE) in D-NBI injection experiments in the 2018 experimental campaign of EAST. ICE driven by fast D-NBI ions and fusion born ions are observed on EAST.5) In Hydrogen minority in Deuterium plasma, the interaction of fast wave and fast D-NBI ions is detected on EAST. Acceleration of fast D ions is confirmed by increased neutron yield.

Abstract

Ion cyclotron emission (ICE) detection system has been successfully designed and used on the Experimental Advanced Superconducting Tokamak (EAST). Experiments were performed in deuterium target plasmas, with the neutral heating beam consisting of 100% deuterium atoms accelerated up to 55-70 keV. The spectrum analyzer was used to detect the spectrogram of ICE signals with the operation frequencies of 10 MHz and 100 MHz. The spectrogram of ICE signals contains very strong and narrow emission lines, which correspond to successive cyclotron harmonics of deuterons at the outer midplane (major radius R∼2.20 m). In addition, experimental observations of correlations with edge-localized modes also support that the excitation region of the ICE is located near the plasma edge.

Abstract

ICRF-enhanced plasma potentials are observed in the scrape off layer (SOL) that map to active antennas on EAST. In near field, the sheath potential is mainly induced by the slow wave. The coupling of the antenna to plasma is related to the fast wave component. From our experiment results, it is very clear that as the power of ICRF antenna increases, the intensity of slow wave increases sharply, but the intensity of fast wave does not increase obviously. Meanwhile, the fast wave signal and slow wave increase when core electron density increases. The magnetic probe is used to investigate ICRF wave in inner side of EAST.

Abstract

A new analytical model is developed for three types of 2-D periodic star-shaped re-entrant lattice structures that possess the orthotropic symmetry and exhibit negative Poisson's ratios. Contributions from both the re-entrant and connection struts are considered using an energy method based on Castigliano's second theorem. Each re-entrant strut is treated as a Timoshenko beam, and stretching, transverse shearing and bending deformations are all incorporated in the formulation. Unlike existing studies, the overlapping of struts at joints is included in determining the relative density, which is analytically expressed for each lattice type. Closed-form formulas are derived for the effective Young's moduli and Poisson's ratios of each type of lattice structure, which contain three non-dimensional length ratios, two re-entrant angles, one shear correction factor, and Young's modulus and Poisson's ratio of the strut material. The new analytical model is validated against finite element simulations conducted in the current study and two existing analytical models for simpler square lattice structures without re-entrant struts. To illustrate the newly developed analytical model, a parametric study is conducted to quantitatively show the effects of the five geometrical parameters on the effective properties of each type of lattice structure. It is found that for the effective Poisson's ratios the key controlling parameters are the two re-entrant angles, while for the effective Young's moduli all of the geometrical parameters can have significant effects except for the external connection length ratio. It is demonstrated that through a proper selection of the geometrical parameters, vertex connections and strut material, it is possible to tailor the effective Poisson's ratios and Young's moduli of each type of lattice structure over wide ranges to satisfy different needs in various applications.

Abstract

Four types of three-dimensional (3-D) metallic metamaterials with tailorable thermo-mechanical properties are designed. For the first three types, the structure-property relations are studied by adjusting design parameters including two length parameters, one angle parameter and two material combinations. For the fourth type, one additional angle parameter is involved. It is shown that each of the four types of metamaterials designed exhibits the cubic symmetry and thus needs three independent elastic constants to characterize its elastic behavior and one coefficient of thermal expansion to describe its isotropic thermal expansion. The effects of the design parameters on the effective Poisson's ratio (PR), coefficient of thermal expansion (CTE), Young's modulus, shear modulus and the relative density are systematically investigated for each of the four types of designed metamaterials by using unit cell-based finite element simulations that incorporate periodic boundary conditions. It is found that 3-D metallic metamaterials with positive, near-zero or negative PR and CTE can be obtained by tailoring the bi-material lattice structures and material combinations. Also, it is revealed that metamaterial # 1 can achieve both a negative PR and a non-positive CTE while maintaining a high stiffness and a low relative density (and thus a lightweight). The good tunability of thermo-mechanical properties of the four types of metamaterials provides an avenue of enabling the expansion of Ashby's material chart to produce more material options for engineering applications.

Abstract

Warner et al. show that knock-in mice expressing a human disease–associated STING mutation spontaneously develop inflammatory lung and skin disease, hypercytokinemia, and T cell cytopenia, which occurs independently of IRF3., Patients with stimulator of interferon genes (STING)–associated vasculopathy with onset in infancy (SAVI) develop systemic inflammation characterized by vasculopathy, interstitial lung disease, ulcerative skin lesions, and premature death. Autosomal dominant mutations in STING are thought to trigger activation of IRF3 and subsequent up-regulation of interferon (IFN)-stimulated genes (ISGs) in patients with SAVI. We generated heterozygous STING N153S knock-in mice as a model of SAVI. These mice spontaneously developed inflammation within the lung, hypercytokinemia, T cell cytopenia, skin ulcerations, and premature death. Cytometry by time-of-flight (CyTOF) analysis revealed that the STING N153S mutation caused myeloid cell expansion, T cell cytopenia, and dysregulation of immune cell signaling. Unexpectedly, we observed only mild up-regulation of ISGs in STING N153S fibroblasts and splenocytes and STING N154S SAVI patient fibroblasts. STING N153S mice lacking IRF3 also developed lung disease, myeloid cell expansion, and T cell cytopenia. Thus, the SAVI-associated STING N153S mutation triggers IRF3-independent immune cell dysregulation and lung disease in mice.

Abstract

A brief Chandra observation of the ultraluminous quasar, SDSS J010013.02+280225.8 at redshift 6.326, showed it to be a relatively bright, soft X-ray source with a count rate of about 1 ct/ks. In this paper we present results for the quasar from a 65ks XMM-Newton observation, which well constrains its spectral shape. The quasar is clearly detected with a total of $\sim$ 460 net counts in the 0.2-10 keV band. The spectrum is characterised by a simple power-law model with photon index of $\Gamma = 2.30^{+0.10}_{-0.10}$, and the intrinsic 2-10 keV luminosity is $3.14\times10^{45}$ erg $\text{s}^{-1}$. The 1 $\sigma$ upper limit to any intrinsic absorption column density is $N_{H} = 6.07\times 10^{22} {\text{cm}}^{-2}$. No significant iron emission lines were detected. We derive the X-ray-to-optical flux ratio $\alpha_{\text{ox}}$ of $-1.74\pm$0.01, consistent with the values found in other quasars of comparable ultraviolet luminosity. We did not detect significant flux variations either in the XMM-Newton exposure or between XMM-Newton and XMM-Newton observations, which are separated by $\sim$ 8 months. Four metallic metamaterials with tailorable mechanical properties are designed using bi-material star-shaped re-entrant planar lattice structures, which do not involve pins, adhesive, welding or pressure-fit joints and can be fabricated through laser-based additive manufacturing. Three length parameters, one angle parameter and three material combinations are used as adjustable design parameters to explore structure-property relations. For each of the four designed metamaterials, the effects of the design parameters on the Poisson’s ratio (PR), coefficient of thermal expansion (CTE), Young’s modulus and relative density are systematically investigated using unit cell-based finite element simulations that incorporate periodic boundary conditions. It is found that the bi-material lattice structures can be tailored to obtain 3-D printable metallic metamaterials with positive, near-zero or negative PR and CTE together with an uncompromised Young’s modulus. In particular, it is shown that metamaterial # 1 can exhibit both a negative PR and a non-positive CTE simultaneously. These metallic metamaterials can find applications in structures or devices such as antennas and precision instruments to reduce thermomechanical stresses and extend service lives.

Abstract

We previously generated STING N153S knock-in mice that have a human disease-associated gain-of-function mutation in STING. Patients with this mutation (STING N154S in humans) develop STING-associated vasculopathy with onset in infancy (SAVI), a severe pediatric autoinflammatory disease characterized by pulmonary fibrosis. Since this mutation promotes the upregulation of antiviral type I interferon-stimulated genes (ISGs), we hypothesized that STING N153S knock-in mice may develop more severe autoinflammatory disease in response to a virus challenge. To test this hypothesis, we infected heterozygous STING N153S mice with murine gammaherpesvirus 68 (γHV68). STING N153S mice were highly vulnerable to infection and developed pulmonary fibrosis after infection. In addition to impairing CD8+ T cell responses and humoral immunity, STING N153S also promoted the replication of γHV68 in cultured macrophages. In further support of a combined innate and adaptive immunodeficiency, γHV68 infection was more severe in Rag1–/– STING N153S mice than in Rag1–/– littermate mice, which completely lack adaptive immunity. Thus, a gain-of-function STING mutation creates a combined innate and adaptive immunodeficiency that leads to virus-induced pulmonary fibrosis. IMPORTANCE A variety of human rheumatologic disease-causing mutations have recently been identified. Some of these mutations are found in viral nucleic acid-sensing proteins, but whether viruses can influence the onset or progression of these human diseases is less well understood. One such autoinflammatory disease, called STING-associated vasculopathy with onset in infancy (SAVI), affects children and leads to severe lung disease. We generated mice with a SAVI-associated STING mutation and infected them with γHV68, a common DNA virus that is related to human Epstein-Barr virus. Mice with the human disease-causing STING mutation were more vulnerable to infection than wild-type littermate control animals. Furthermore, the STING mutant mice developed lung fibrosis similar to that of patients with SAVI. These findings reveal that a human STING mutation creates severe immunodeficiency, leading to virus-induced lung disease in mice.

Abstract

Early flowering plants (ephemers and ephemeroids) are an important component of the biosystem of steppes and deserts. These species form perennial pasture communities, for early grazing. The present work was conducted to evaluate the floristic checklist and ecological uniqueness of early flowering plants of Central Kazakhstan, in the forest-steppe regions of Em-Bulak region and Edige mountains, during 2021 and 2022. A total of 26 species of ephemers and ephemeroids were recorded, which belong to 16 families: Liliaceae – 3 species, Asteraceae – 3 species, Ranunculaceae – 3 species, Rosaceae – 3 species, Brassicaceae – 2 species, Boraginaceae – 2 species, the other families are represented by one species. According to ecomorphological characteristics: 8 species (31%) belong to the xerophyte group, 6 (23%) to the xeromesophyte group, 7 (27%) to the mesoxerophyte group and 5 (19%) to the mesophyte group. Also, as a result of our research we found two species of plants which are included in the Red Book of the Republic of Kazakhstan – Pulsatilla patens (L.) Mill. and Tulipa patens Agardh. ex Schult. f. The data obtained can be used for environmental monitoring and issues of rational nature management of wild plants of Central Kazakhstan.

Abstract

The study was conducted on four islands located west of Bejaia (north-eastern Algeria) to collect baseline information about diversity and composition of insects. Data were collected during summer and spring of 2015 using traps, sweep nets and the Japanese umbrella sampling methods. A total of 1691 individuals belonging to 112 species were collected. Totals of 72, 56, 41 and 34 species were recorded in Pisans Island, El Euch Island, Sahel Islet and Ail Islet, respectively. Of the 1,691 individuals recorded, 39.85 %, 30.51 %, 18.03 % and 11.59 % were recorded in Pisans Island, El Euch Island, Sahel Islet and Ail Islet, respectively. Shannon-Weaver index ranged from 2.96 to 3.34 and Equitability between 0.77 and 0.87. The composition of the insect community differed between sampled islands (NMDS, Stress =. 0.013). Eleven (11) insect species composed over 50 % of dissimilarity between islands. Two species (Cassida viridis (L.) and Pheidole pallidula (Nyl.) accounted for more than 5 % of the diversity on all islands. Sorensen's similarity coefficient showed that Pisans and El Euch islands have the highest similarity (45.37 %), while the lowest similarity was recorded between Pisans Island and Sahel Islet (15.53 %).

Abstract

Obesity is a public health epidemic, particularly among underrepresented populations. With a large proportion of immigrants, Latino Americans comprise the largest minority population in the United States. This study examined the association of acculturation factors with obesity among Latino American men ( n = 1,127) using the National Latino and Asian American Study. The result identified two acculturation-related factors (being U.S.-born and living in the United States for the longest period/5-10 years) as positive correlates. In contrast, a different study on obesity in Latino American women demonstrated discrimination, but not the above factors, as significant correlates. The men’s pattern suggests that the Hispanic/Latino paradox might have greater implications for men with respect to weight issues. Furthermore, Mexican American and Other Latino American men presented a greater likelihood of being obese than Cuban and Puerto Rican men. The findings, if replicated in prospective research, suggest the need for gender- and ethnic-specific intervention for obesity in Latino American men, particularly for the largest subgroup, Mexican Americans.

Abstract

We analyze the X-ray, optical, and mid-infrared data of a “changing-look” Seyfert galaxy SDSS J155258+273728 at z ≃ 0.086. Over a period of one decade (2009–2018), its broad Hα line intensity increased by a factor of ∼4. Meanwhile, the X-ray emission in 2014 as observed by Chandra was about five times brighter than that in 2010 by Suzaku, and the corresponding emissions in the V-band, mid-infrared W1 band brighten by ∼0.18, 0.32 mag, respectively. Moreover, the absorption in X-rays is moderate and stable, i.e., N H ∼ 10 21 cm − 2 , but the X-ray spectrum becomes harder in the 2014 Chandra bright state (i.e., photon index Γ = 1.52 − 0.06 + 0.06 ) than that of the 2010 Suzaku low state ( Γ = 2.03 − 0.21 + 0.22 ). With an Eddington ratio being lower than a few percent, the inner region of the accretion disk in SDSS J155258+273728 is likely a hot accretion flow. We then compile from literature the X-ray data of “changing-look” active galactic nuclei (AGNs), and find that they generally follow the well-established “V”-shaped correlation in AGNs, that is, above a critical turnover luminosity the X-ray spectra soften with the increasing luminosity, and below that luminosity the trend is reversed in the way of “harder when brighter.” This presents direct evidence that CL-AGNs have distinctive changes in not only the optical spectral type, but also the X-ray spectral shape. The similarity in the X-ray spectral evolution between CL-AGNs and black hole X-ray binaries indicates that the observed CL-AGNs phenomena may relate to the state transition in accretion physics.

Abstract

Two-dimensional (2-D) micro-architectured mechanical metamaterials are designed using a topology optimization approach that integrates a parametric level set method (PLSM) with a meshfree method based on compactly supported radial basis functions (CS-RBF). The PLSM is employed as the optimization algorithm to achieve desired microstructures with targeted material properties. The effective elastic properties, including the bulk modulus, shear modulus and Poisson’s ratio, are predicted using a strain energy-based homogenization method and the CS-RBF meshfree algorithm. Two sets of optimizations are implemented: one is for the case of a single solid material, and the other is for the case of two solid materials, each involving an additional void phase. Three numerical examples are provided for each case with the same optimization objectives: maximizing the effective bulk modulus, maximizing the effective shear modulus, and minimizing the effective Poisson’s ratio under given volume fraction constraints. The numerical results reveal that the newly proposed approach can generate smooth topological boundaries and optimal microstructures. In particular, the current method can topologically optimize auxetic metamaterials with a negative Poisson’s ratio. It can also be extended to design other periodic metamaterials, including those with a negative coefficient of thermal expansion or frequency bandgaps.

Abstract

This is the second installment for the Large Sky Area Multi-Object Fibre Spectroscopic Telescope (LAMOST) Quasar Survey, which includes quasars observed from September 2013 to June 2015. There are 9024 confirmed quasars in DR2 and 10911 in DR3. After cross-match with the SDSS quasar catalogs and NED, 12126 quasars are discovered independently. Among them 2225 quasars were released by SDSS DR12 QSO catalogue in 2014 after we finalised the survey candidates. 1801 sources were identified by SDSS DR14 as QSOs. The remaining 8100 quasars are considered as newly founded, and among them 6887 quasars can be given reliable emission line measurements and the estimated black hole masses. Quasars found in LAMOST are mostly located at low-to-moderate redshifts, with a mean value of 1.5. The highest redshift observed in DR2 and DR3 is 5. We applied emission line measurements to H$\alpha$, H$\beta$, Mg{\sc ii} and C{\sc iv}. We deduced the monochromatic continuum luminosities using photometry data, and estimated the virial black hole masses for the newly discovered quasars. After dry pressing molding, using solid phase sintering method sinter them at 1380?C,1410?C,1440?C,1410?C,1500?C,1530?C, respectively. A performance of the material is characterized by using SEM and XRD, such as microscopic morphology, phase composition and the compressive strength, etc. Combining results of experiments, explore the mechanism of sintering temperature on the properties of ceramic materials.

Abstract

Medication therapy management (MTM) is one form of a medication benefit program offered by public and private health providers and insurers. Although the term was first coined in 2003, MTM in its earlier forms has been used since the 1990s as a mechanism to improve health metrics for medically complex patients. Its role expanded with the passage of Medicare Part D, as a mandated component to help patients with multiple chronic conditions, high drug costs, and high utilization to improve the effectiveness and safety of their medication treatments. To review the evidence on MTM effectiveness in order to (a) provide information on its establishment and goals and (b) summarize research findings under 3 outcomes: economic, clinical, and humanistic. PubMed, a search engine service of the National Center for Biotechnology Information was utilized by trained research assistants to search for articles with the following key words: MTM, randomized controlled trials on MTM, evaluation of MTM, comprehensive medication review, medication action plan, special needs population, special needs plans, Medicaid, disease management, adherence, non-adherence, compliance, chronic conditions, disabling chronic conditions, and disability. Additional searches were conducted for key articles in references listed in the most recent review articles. The initial search identified nearly 300 articles.When evaluated, most studies found economic benefits, but the quality of research design and end point measures varied considerably across evaluations. Clinical outcomes encompassed a wide range of potential metrics, from service utilization, to individual patient and population outcomes, and quality of care. Quality measures such as provider-prescribing habits and medication adherence were frequently found to improve. As noted with the economic outcomes studies, overall rigor of study design was suboptimal, and often underpowered. Few studies have focused on humanistic outcomes such as improved patient quality of life. Evidence suggests that MTM services are a promising way to manage complex patients, but there are gaps in the literature largely because of the limited number of studies with strong designs. Stronger evaluation of MTM programs is warranted.

46. Quantitative analysis of factors controlling sediment yield in mountainous watersheds

Abstract

Sediment and turbidity are primary causes of impaired river ecosystems; remedial action for these impairments requires the identification of their sources and controlling factors. This paper examines the combined effects of watershed complexity in terms of land use and physiography on the specific sediment yield of the upper Du River watershed (8973 km2) in China. The land use composition, land use pattern, morphometric variables, and soil properties of the watershed were calculated at the subwatershed scale and considered to be potentially influential factors. Because these watershed characteristics are highly codependent, a partial least-squares regression (PLSR) was used to elucidate the linkages between the specific sediment yield and metrics composed of 19 selected watershed characteristics. The first-order factors were identified by calculating the variable importance for the projection (VIP). The results revealed that the land use composition and land use pattern exerted the largest effects on the specific sediment yield and explained 65.2% of the variation in the specific sediment yield. A set of physiographic indices was also found to have a large effect on the specific sediment yield and explained 17.7% of the observed variation in the specific sediment yield. The following are the dominant first-order factors of the specific sediment yield at the subwatershed scale: the areal percentages of agriculture and forest, patch density, value of the Shannon's diversity index, contagion, value of the hypsometric integral, and saturated soil hydraulic conductivity. The watershed size exerted a substantial effect on the sediment delivery ratio (SDR). The VIP values also suggested that the Shannon's diversity index, contagion, and hypsometric integral are important factors in the SDR. With a readily available digital spatial database and rapid developments in geographic information system (GIS) technology, this practical and simple PLSR approach could be applied to a variety of watersheds.

Abstract

On September 14, 2015 at 09:50:45 UTC the two detectors of the Laser Interferometer Gravitational-Wave Observatory simultaneously observed a transient gravitational-wave signal. The signal sweeps upwards in frequency from 35 to 250 Hz with a peak gravitational-wave strain of 1.0×10-21. It matches the waveform predicted by general relativity for the inspiral and merger of a pair of black holes and the ringdown of the resulting single black hole. The signal was observed with a matched-filter signal-to-noise ratio of 24 and a false alarm rate estimated to be less than 1 event per 203 000 years, equivalent to a significance greater than 5.1σ. The source lies at a luminosity distance of 410-180+160 Mpc corresponding to a redshift z=0.09-0.04+0.03. In the source frame, the initial black hole masses are 36-4+5M⊙ and 29-4+4M⊙, and the final black hole mass is 62-4+4M⊙, with 3.0-0.5+0.5M⊙c2 radiated in gravitational waves. All uncertainties define 90% credible intervals. These observations demonstrate the existence of binary stellar-mass black hole systems. Abstract

Specific gut commensal bacteria improve host health by eliciting mutualistic regulatory T (T[subscript reg]) cell responses. However, the bacteria that induce effector T (T[subscript eff]) cells during inflammation are unclear. We addressed this by analyzing bacterial-reactive T cell receptor (TCR) transgenic cells and TCR repertoires in a murine colitis model. Unexpectedly, we found that mucosal-associated Helicobacter species triggered both T[subscript reg] cell responses during homeostasis and T[subscript eff] cell responses during colitis, as suggested by an increased overlap between the T[subscript eff]/T[subscript reg] TCR repertoires with colitis. Four of six T[subscript reg] TCRs tested recognized mucosal-associated Helicobacter species in vitro and in vivo. By contrast, the marked expansion of luminal Bacteroides species seen during colitis did not trigger a commensurate T[subscript eff] cell response. Unlike other T[subscript reg] cell-inducing bacteria, Helicobacter species are known pathobionts and cause disease in immunodeficient mice. Thus, our study suggests a model in which mucosal bacteria elicit context-dependent T[subscript reg] or T[subscript eff] cell responses to facilitate intestinal tolerance or inflammation. ©2017, NIH (grant no. Abstract

The present study investigated psychosocial predictors for major depressive disorder (MDD), general anxiety disorder (GAD), and suicidal ideation (SI) of Latino American men identified in the first national mental health epidemiological survey of Latinos. Three separate sets of logistic regression analyses were performed for 1,127 Latinos, following preplanned two steps (Model 1—Known Demographic and Acculturation Predictors as controls, Model 2—Psychosocial Risk and Protective Factors). Results show that Negative Interactions with family members significantly predicted the likelihood of both MDD and SI, while SI was also associated with Discrimination. Acculturation Stress was associated with that of GAD (alongside more Income, Education of 12 years, and Years in the United States for less than 11 years). Other potential protective factors (social support, racial/ethnic identity, religious involvement) were not influential. The differential predictors for mental health issues among Latino men imply that assessment and intervention for them may need certain gender-specific foci in order to improve mental health disparities in this population.
