1. Point-of-care lung ultrasound in the diagnosis of childhood pneumonia

Abstract: Introduction Lung ultrasound is emerging as a rapid, simple and safe alternative for diagnosing pneumonia since it has a higher sensitivity than X-rays and lower radiation exposure than computerized tomography. This is a prospective observational study done at a tertiary care centre in Chennai to study the diagnostic utility of lung ultrasound in pneumonia. Methods Children aged 1 month to 12 years who were admitted to the hospital with complaints of cough, fever and/or breathing difficulty and on examination had tachypnea and/or chest indrawing were included in the study. All children underwent chest X-rays which was a standard hospital protocol. At admission, an independent investigator who was blinded to the clinical and radiological features of the child performed lung ultrasound. In all children, the final diagnosis of pneumonia was made by another independent expert paediatrician on the basis of the clinical features and chest X-ray. The test characteristics of ultrasound and chest X-ray were compared against this gold standard of physician-diagnosed pneumonia. Results Out of the 252 children studied, 225 (89.3%) had pneumonia while the rest 27 (10.7%) had other diagnoses. Among the 225 children with pneumonia, 223 (99.1%) were detected by ultrasound while 157 (69.8%) were detected by chest X-ray. All the test characteristics such as sensitivity, specificity, positive and negative predictive values of ultrasound were higher than those of chest X-ray. The sensitivity and specificity of ultrasound to diagnose pneumonia were 99.11% and 81.48%, respectively, while the sensitivity and specificity of X-ray for the same were 69.77% and 74.07%, respectively. Overall diagnostic accuracy for chest ultrasonography was 97.22% (94.36% to 98.88%), whereas for chest radiography, it was found to be 70.24% (64.18% to 75.81%).While both modalities were able to diagnose pneumonia significantly, ultrasound had better strength of association (Cramer’s V value = 0.849) than X-ray to the final diagnosis. Conclusion Lung ultrasound can be employed as a point-of-care investigation to diagnose pneumonia in suspected cases and can even replace chest X-ray in such circumstances.

2. Evaluation of molecular effects associated with apoptosis, tumour progression, angiogenesis and metastasis by a novel combination of drugs with ormeloxifene in triple negative breast cancer cells

Abstract: Aim: To investigate the molecular effects of a novel combination [sertraline and plumbagin (comb) with ormeloxifene (Orm)] for anticancer activity in triple negative breast cancer cell line “MDA-MB-231”. Methods: The cytotoxic effect of the drugs was analyzed by the MTT assay and nuclear morphological changes by acridine orange/ethidium bromide (AO/EB) staining. Induction of apoptosis by annexin V-FITC staining, active caspase-3 detection and cell cycle analysis were studied in vitro on “MDA-MB-231” cells. The qRT-PCR was done to explore the upregulation and down regulation of targeted genes for angiogenesis, metastasis, tumor suppression and protein folding on the triple negative breast cancer cells. The preliminary anti-angiogenic effect of the drugs was assessed by chorioallantoic membrane (CAM) assay. Results: Orm showed inhibitory effects in “MDA-MB-231” cells in a dose and time dependent manner whereas; the drugs in combination gave better cytotoxic effects in the screening MTT assay. Orm + comb was more effective than Orm alone in eliciting apoptosis as well as inhibited the single cell to grow into a colony. CAM assay using Orm and Orm + comb suggested the anti-angiogenic potential which was further confirmed by the downregulation of VEGF in “MDA-MB-231” cells by qRT-PCR studies. The combination was found to effectively upregulate the expression of P53 and P21 and downregulate the gene expression of zinc finger E-box binding homeobox 1 (ZEB1) and heat shock protein 70 (HSP70) in “MDA-MB-231” cancer cells. Conclusions: Collectively this study reveals the efficacy of Orm + comb as more significant than the clinically used tamoxifen (Tam). The study elucidates the promising novelty of the combination as a potential chemotherapeutic intervention for mitigating the aggressiveness of triple negative breast cancer and it addresses the intrinsic resistance caused by single

3. An Asymptomatic Mandibular Radiolucency: First Sign of Follicular Thyroid Carcinoma

Abstract: Background Metastatic carcinomas to the jaws are a rare occurrence. Most notable primary sources within the literature are breast, lung and kidney, followed by GI, prostate and thyroid. In almost all instances, there is a primary site already identified, but very occasionally the metastases are the first sign of a problem. Notable other, less obvious, clinical expressions of malignancy are weight loss, loss of appetite, fever and chills. Again, these can present with no obvious site of a primary cancer. A unique feature of our case is the lack of primary symptoms.Case Description The patient initially presented for teeth extraction a year previously, followed by a slow but eventually large expansion of the left mandibular ramus. The underlying radiolucency gave no apparent signs or symptoms suggestive of malignancy. Incisional biopsy showed a glandular neoplastic proliferation with apparent thyroid follicles noted at the margins. Special imaging and immunostains confirmed a tissue origin in the thyroid gland. The patient was referred to Otolaryngology for definitive treatment. The jaw lesion was treated by radical surgical excision and the neck site was treated, after microscopic confirmation of a follicular thyroid carcinoma, via total thyroidectomy.Practical Implications This case illustrates the importance of interdisciplinary treatment and how it can lead to an optimal outcome for the patient's prognosis.

4. Simulation analysis with rock muons from atmospheric neutrino interactions in the ICAL detector at the India-based Neutrino Observatory

Abstract: Abstract The proposed magnetized Iron CALorimeter detector (ICAL) to be built in the India-based Neutrino Observatory (INO) laboratory aims to study atmospheric neutrinos and its properties such as precision measurements of oscillation parameters and the neutrino mass hierarchy. High energy charged current (CC) interactions of atmospheric neutrinos with the rock surrounding the detector produce so-called “rock muons” along with hadrons. While the hadron component of these events are absorbed in the rock itself, the rock muons traverse the rock and are detected in the detector. These rock muon events can be distinguished from cosmic muons only in the upward direction and can provide an independent measurement of the oscillation parameters. A simulation study of these events at the ICAL detector shows that, although reduced in significance compared to muons produced in direct CC neutrino interactions with the detector, these events are indeed sensitive to the oscillation parameters, achieving a possible $$1\sigma $$ 1 σ precision of 10% and 27% in determining $$\varDelta m_{32}^2$$ Δ m 32 2 and $$\sin ^2\theta _{23}$$ sin 2 θ 23 , respectively. Hence a combination of the standard atmospheric neutrino analysis which is the main goal of ICAL, with these rock muon events, will improve the precision reach of ICAL for these parameters.

5. Evaluation of Pain-Associated Behavioral Changes in Monoiodoacetate-Induced Osteoarthritic Rats Using Dynamic Weight Bearing Analysis

Abstract: Pain is the primary clinical indication of osteoarthritis (OA), and behavioral assessments in rodent pain models are widely used to understand pain patterns. These preclinical pain assessments can also help us to understand the effectiveness of emerging therapeutics for prolonged OA pain management. Along with evoked methods like mechanical allodynia and thermal hyperalgesia, non-evoked methods such as dynamic weight bearing (DWB) analysis are valuable tools for behavioral assessments of pain. Both these methods were utilized to study pain-induced behavioral changes in a monoiodoacetate (MIA)-induced osteoarthritic pain model, which is a well-established preclinical OA pain model. However, the utility of DWB analysis as an indicator of long-term pain sensitivity (more than 4 weeks) remains largely unexplored. Understanding the long-term sensitivity of DWB is valuable to study the effectiveness of novel prolonged pain-relieving therapeutics. Here, we studied the dynamic behavioral changes in MIA-induced OA rats over a period of 16 weeks using DWB measurements. Female Sprague Dawley rats were injected in the right knee joint with MIA (3 mg) using X-ray guidance. Multiple dynamic postural evaluations such as ipsilateral weight percentage, paw area, contralateral/ipsilateral weight ratio and area ratio were assessed to understand the behavioral changes. The data showed that the ipsilateral weight bearing percentage alone is not sufficient to assess pain-related behavior beyond 6 weeks. This study shows the advantages and limitations of dynamic weight bearing as an assessment tool for the long-term progression of pain behavior in MIA-induced OA rats.

6. Comparison of Time Taken for Intubation (TTI) in Conventional Laryngoscope with Video Laryngoscope for Endotracheal Intubation In Laproscopic Surgeries

Abstract: Background: Direct laryngoscopy necessitates the alignment of the oropharyngeal-laryngeal axis whereas video laryngoscope is an optical vision which doesn’t require alignment. This study aimed to compare direct laryngoscopy with a Macintosh blade to King-Vision Video laryngoscope for endotracheal intubation in patients who were scheduled for elective laproscopic surgeries under general anaesthesia. Methods: In this prospective randomised clinical study,118 adults with ASA I and II requiring endotracheal intubation for laproscopic surgeries under general anaesthesia were enrolled and randomised into either of the two groups by envelope method, Group DL-direct laryngoscope and Group VL-video laryngoscope where they were intubated using direct laryngoscope with Macintosh blade or King Vision videolaryngoscope. The Primary objective was to compare Time to intubate(TTI), Visualization of the laryngeal view by Cormack-Lehane grade and Successful first attempt. Secondary objective was to record the Number of intubation failure, Number of attempts, Change of anaesthesiologist and use of adjunct equipment and the complications such as oropharyngeal trauma, neck pain, dysphagia and hoarseness. Results: In comparison to group DL (21.67±4.318s), group VL took longer time to intubate (26.21± 4.150s) but had superior glottic vision than DL group(p=0.0177). Compared to DL group (72.4%), the VL (84.5%) patients had their first successful attempt, inspite of 2 failures. Complications such as pharyngeal pain (8.6%vs29.3%), hoarseness (5.2%vs29.3%), Use of adjunct equipment like bougie (19%vs 3.4%) were significantly higher in DL compared to VL group, while oropharyngeal injury, dysphagia, number of attempts and change of anaesthetists were similar in both groups. Conclusion: In comparison to the Macintosh laryngoscope, the King-vision VideoLaryngoscope took longer to intubate but had clearer glottis visualisation and a higher first-time success rate and can be used as a good teaching tool. In King-vision video laryngoscope, there was less use of auxiliary equipment and fewer complications.

7. Regulation of SPDEF expression by DNA methylation in advanced prostate cancer

Abstract: IntroductionProstate cancer (PCa) presents a significant health challenge in men, with a substantial number of deaths attributed to metastatic castration resistant PCa (mCRPC). Moreover, African American men experience disproportionately high mortality rates due to PCa. This study delves into the pivotal role of SPDEF, a prostate specific Ets transcription factor, and its regulation by DNA methylation in the context of PCa progression.MethodsWe performed Epigenetic reprogramming using daily treatment with non-toxic dose of 5Aza-2-deoxycytidine (5Aza-dC) for two weeks to assess its impact on PDEF expression in prostate cancer cells. Next, we conducted functional studies on reprogrammed cells, including cell migration (wound-healing assay), invasion (Boyden-Chamber test), and proliferation (MTT assay) to comprehensively evaluate the consequences of altered PDEF expression. We used bisulfite sequencing (BSP) to examine DNA methylation at SPDEF promoter. Simultaneously, we utilized siRNA-mediated targeting of key DNMTs (DNMT1, DNMT3A, and DNMT3B) to elucidate their specific role in regulating PDEF. We measured mRNA and protein expressions using qRT-PCR and immune-blotting techniques, respectively.ResultsIn this report, we observed that: a) there is a gradual decrease in SPDEF expression with a concomitant increase in methylated CpG sites within the SPDEF gene during prostate cancer progression from lower to higher Gleason grade; b) Expression of DNMT’s (DNMT1, 3a and 3b) is increased during prostate cancer progression, and there is an inverse correlation between SPDEF and DNMT expression; c) SPDEF levels are decreased in RC77/T, a line of PCa cells from African American origin similar to PC3 and DU145 cells (CRPC cells), as compared to LNCaP cells , a line of androgen dependent cells,; d) the 5′ CpG island of SPDEF gene are hypermethylated in SPDEF-negative CRPC ( PC3, DU145 and RC77/T) cell lines but the same regions are hypomethylated in SPDEF-positive castrate sensitive (LNCaP) cell line ; (e) expression of SPDEF in PCa cells lacking SPDEF decreases cell migration and invasion, but has no significant effect on cell proliferation, and; (f) treatment with the demethylating agent, 5-aza-2′-deoxycytidine, or silencing of the DNMT’s by siRNA, partially restores SPDEF expression in SPDEF-negative PCa cell lines, and decreases cell migration and invasion.DiscussionThese results indicate hypermethylation is a prevalent mechanism for decreasing SPDEF expression during prostate cancer progression. The data demonstrate that loss of SPDEF expression in prostate cancer cells, a critical step in cellular plasticity, results from a potentially reversible process of aberrant DNA methylation. These studies suggest DMNT activity as a potential therapeutic vulnerability that can be exploited for limiting cellular plasticity, tumor progression, and therapy resistance in prostate cancer.

8. A Case Report on Care-Seeking Type Illness Anxiety Disorder after COVID-19 Infection

Abstract: This case report highlights the diagnostic challenges presented by the overlapping symptoms of illness anxiety disorder (IAD) and long COVID-19 (LC-19). This case report focuses on a 58-year-old woman with care-seeking type IAD in the context of LC-19-associated symptoms. The patient experienced mild COVID-19 in August 2021. Since then, she has reported an increase in LC-19-associated symptoms, including cognitive deficits, breathlessness, fatigue, and anosmia. Despite largely normal laboratory results, imaging, and physical examinations, the patient’s distress and care-seeking behaviors persisted, resulting in the diagnosis of IAD. Accurately differentiating between LC-19 and IAD is crucial for appropriate patient care. We discuss the importance of recognizing and treating IAD in patients with LC-19-associated symptoms and the need for further research on the correlation between IAD and both COVID-19 and LC-19.

9. Characterizing inhibitors of human AP endonuclease 1

Abstract: AP endonuclease 1 (APE1) processes DNA lesions including apurinic/apyrimidinic sites and 3´-blocking groups, mediating base excision repair and single strand break repair. Much effort has focused on developing specific inhibitors of APE1, which could have important applications in basic research and potentially lead to clinical anticancer agents. We used structural, biophysical, and biochemical methods to characterize several reported inhibitors, including 7-nitroindole-2-carboxylic acid (CRT0044876), given its small size, reported potency, and widespread use for studying APE1. Intriguingly, NMR chemical shift perturbation (CSP) experiments show that CRT0044876 and three similar indole-2-carboxylic acids bind a pocket distal from the APE1 active site. A crystal structure confirms these findings and defines the pose for 5-nitroindole-2-carboxylic acid. However, dynamic light scattering experiments show the indole compounds form colloidal aggregates that could bind (sequester) APE1, causing nonspecific inhibition. Endonuclease assays show the compounds lack significant APE1 inhibition under conditions (detergent) that disrupt aggregation. Thus, binding of the indole-2-carboxylic acids at the remote pocket does not inhibit APE1 repair activity. Myricetin also forms aggregates and lacks APE1 inhibition under aggregate-disrupting conditions. Two other reported compounds (MLS000552981, MLS000419194) inhibit APE1 in vitro with low micromolar IC50 and do not appear to aggregate in this concentration range. However, NMR CSP experiments indicate the compounds do not bind specifically to apo- or Mg2+-bound APE1, pointing to a non-specific mode of inhibition, possibly DNA binding. Our results highlight methods for rigorous interrogation of putative APE1 inhibitors and should facilitate future efforts to discover compounds that specifically inhibit this important repair enzyme.

Abstract: A search for proton decay into $e^+/\mu^+$ and a $\eta$ meson has been performed using data from a 0.373 Mton$\cdot$year exposure (6050.3 live days) of Super-Kamiokande. Compared to previous searches this work introduces an improved model of the intranuclear $\eta$ interaction cross section, resulting in a factor of two reduction in uncertainties from this source and $\sim$10\% increase in signal efficiency. No significant data excess was found above the expected number of atmospheric neutrino background events resulting in no indication of proton decay into either mode. Lower limits on the proton partial lifetime of $1.4\times\mathrm{10^{34}~years}$ for $p\rightarrow e^+\eta$ and $7.3\times\mathrm{10^{33}~years}$ for $p\rightarrow \mu^+\eta$ at the 90$\%$ C.L. were set. These limits are around 1.5 times longer than our previous study and are the most stringent to date.

11. Effect of Fines on Strength of Granular Soil Reinforced with Geosynthetics

Abstract: Geosynthetics (GSN) are polymeric products used in Geotechnical Engineering for the purpose of improving the properties of the insitu soil. In this project, experimental work was carried out to determine the influence of presence of fines within granular soil on the CBR strength of soil reinforced with different GSN (Woven geotextile, Non-woven geotextile, Geogrid and Geonet). Two types of granular soil in disturbed state containing varying percentage of fines were selected from in and around the Chennai region. Soil type 1 contains high fines of about 40% (SHF) whereas soil type 2 contains less fines of about 7% (SLF). Unsoaked CBR (U-CBR) test was carried on unreinforced soil and on soil reinforced with different GSN placed at varying depths of D/5, 2D/5, 3D/5 and 4D/5 from top of soil (D is the total depth of soil), to determine the ideal depth of placement of GSN within SHF and SLF. Soaked CBR (S-CBR) test was carried on unreinforced soil and on soil reinforced with different GSN placed at an ideal depth determined from U-CBR test. Based on the experimental results, ideal depth of placement of GSN for SHF and SLF was found to be D/5 and 3D/5 respectively from top of the soil and Geogrid was found to be the best geosynthetic for reinforcing the granular soil containing high fines and less fines.

Cushing’s syndrome is a rare complication of Medullary thyroid carcinoma [MTC] and is due to ectopic Adrenocorticotropic [ACTH] secretion by tumor cells. Cushing’s syndrome presents a challenging diagnostic and management issue in patients with MTC. Entire clinical history, laboratory investigations, microscopic pictures are discussed in detail.

Abstract: SERMS like Tamoxifene, 5-hydroxy tamoxifene, raloxifene and endoxifene has been used for the treatment of hormonal imbalances and dependent cancers owing to their action via Estrogen receptors as in the treatment of estrogen sensitive breast cancers. Due to the adverse side effects, modifications and development of the existing or newer SERMS has always been of immense interest. Ormeloxifene, a SERM molecule manufactured by HLL Lifecare Ltd, India as birth control under the trade names Saheli, Novex, and Novex-DS which is also investigated against mastalgia, fibro-adenoma and abnormal uterine bleeding. Anti-cancer effects have been reported in estrogen dependent and independent cancers which shows its wide scope to be implemented in cancer therapy. Current investigation is a comprehensive effort to find the cytotoxic potential of Ormeloxifene in comparison with clinically used four SERMS in twenty six cancer cell lines of different origin using Adriamycin as positive control. Also the computational studies pertaining to selected target/ligand with respect to tumor progression, development, treatment responses and apoptosis. The studies proved effective cytotoxicity of Ormeloxifene on cancer cell lines with lower TGI, GI50 and LC50 values which are significantly comparable. Also the in silico studies proved that the docking score of the compound suggests the interaction of the compound which could tightly regulate key target genes controlling cancer like ER, EGFR kinase, EGFR-cSRC, HDAC-2, PARP-1 and BRAF. This study brings out the superior efficacy of Ormeloxifene compared to other SERMS with proven safety profile to be repositioned as an anti-cancer drug to treat diverse cancer types.

Abstract: Abstract We present the reach of the proposed INO-ICAL in measuring the atmospheric-neutrino-oscillation parameters $$\theta _{23}$$ θ23 and $$\varDelta m^2_{32}$$ Δm322 using full event-by-event reconstruction for the first time. We also study the fluctuations arising from the low event statistics and their effect on the precision measurements and mass-hierarchy analysis for a 5-year exposure of the 50 kton ICAL detector. We find a mean resolution of $$\varDelta \chi ^2 \approx 2.9$$ Δχ2≈2.9 , which rules out the wrong mass hierarchy of the neutrinos with a significance of approximately $$1.7\sigma $$ 1.7σ . These results on mass-hierarchy are similar to those presented in earlier studies that approximated the performance of the ICAL detector.

15. Serum and gingival crevicular fluid macrophage inflammatory protein-1β (MIP-1β): Their relationship in periodontal health, disease, and after treatment with nonsurgical therapy

Abstract: Objectives: Earlier studies have revealed high macrophage inflammatory protein-1β (MIP-1β) levels in the gingival crevicular fluid (GCF) and the serum of patients with chronic periodontitis. However, reports correlating effect of non surgical theraphy on levels of GCF and serum MIP-1βlevels are inadequate. Therefore, the present study estimates the GCF and serum MIP-1β levels in periodontal health, disease and after periodontal treatment with non surgical theraphy. Materials and Methods: Periodontal examination and collection of GCF and serum was performed for 60 subjects categorized into four groups with 20 subjects in each group: Group I (healthy); group II (gingivitis), group III (chronic periodontitis). Twelve weeks after scaling and root planing, the GCF and serum were collected from 20 patients of group III, who were considered as group IV. MIP-1β levels were estimated using the enzyme-linked immunosorbent assay. Results: MIP-1β was detected in all samples. However, the levels reduced significantly in group IV (P < 0.05). Mean MIP-1β levels in GCF and serum were the highest in group III (1.404 ng/μL,1.712 ng/μl) and the lowest in group I (0.342 ng/μl,0.465 ng/μl), and those in groups II and IV appeared between those of groups I and III. Conclusions: The GCF and serum MIP-1β levelsincreased proportionally with the progression of periodontal disease (PD) and decreased after treatment. Because MIP-1β levels in the GCF and serum correlated positively with clinical parameters, MIP-1βmay be considered a “novel biomarker” in PD progression. However, controlled longitudinal studies are required to confirm this possibility.

Abstract: The Super-Kamiokande and T2K collaborations present a joint measurement of neutrino oscillation parameters from their atmospheric and beam neutrino data. It uses a common interaction model for events overlapping in neutrino energy and correlated detector systematic uncertainties between the two datasets, which are found to be compatible. Using 3244.4 days of atmospheric data and a beam exposure of $19.7(16.3) \times 10^{20}$ protons on target in (anti)neutrino mode, the analysis finds a 1.9$\sigma$ exclusion of CP-conservation (defined as $J_{CP}=0$) and a preference for the normal mass ordering., Comment: 12 pages, 4 figures

Abstract: Preceding a core-collapse supernova, various processes produce an increasing amount of neutrinos of all flavors characterized by mounting energies from the interior of massive stars. Among them, the electron antineutrinos are potentially detectable by terrestrial neutrino experiments such as KamLAND and Super-Kamiokande via inverse beta decay interactions. Once these pre-supernova neutrinos are observed, an early warning of the upcoming core-collapse supernova can be provided. In light of this, KamLAND and Super-Kamiokande, both located in the Kamioka mine in Japan, have been monitoring pre-supernova neutrinos since 2015 and 2021, respectively. Recently, we performed a joint study between KamLAND and Super-Kamiokande on pre-supernova neutrino detection. A pre-supernova alert system combining the KamLAND detector and the Super-Kamiokande detector was developed and put into operation, which can provide a supernova alert to the astrophysics community. Fully leveraging the complementary properties of these two detectors, the combined alert is expected to resolve a pre-supernova neutrino signal from a 15 M$_{\odot}$ star within 510 pc of the Earth, at a significance level corresponding to a false alarm rate of no more than 1 per century. Abstract: Neutrinos from very nearby supernovae, such as Betelgeuse, are expected to generate more than ten million events over 10\,s in Super-Kamokande (SK). At such large event rates, the buffers of the SK analog-to-digital conversion board (QBEE) will overflow, causing random loss of data that is critical for understanding the dynamics of the supernova explosion mechanism. In order to solve this problem, two new DAQ modules were developed to aid in the observation of very nearby supernovae. The first of these, the SN module, is designed to save only the number of hit PMTs during a supernova burst and the second, the Veto module, prescales the high rate neutrino events to prevent the QBEE from overflowing based on information from the SN module. In the event of a very nearby supernova, these modules allow SK to reconstruct the time evolution of the neutrino event rate from beginning to end using both QBEE and SN module data. This paper presents the development and testing of these modules together with an analysis of supernova-like data generated with a flashing laser diode. We demonstrate that the Veto module successfully prevents DAQ overflows for Betelgeuse-like supernovae as well as the long-term stability of the new modules. During normal running the Veto module is found to issue DAQ vetos a few times per month resulting in a total dead time less than 1\,ms, and does not influence ordinary operations. Additionally, using simulation data we find that supernovae closer than 800~pc will trigger Veto module resulting in a prescaling of the observed neutrino data., Comment: 28 pages, 18 figures. Abstract: We present the results of the charge ratio ($R$) and polarization ($P^{\mu}_{0}$) measurements using the decay electron events collected from 2008 September to 2022 June by the Super-Kamiokande detector. Because of its underground location and long operation, we performed high precision measurements by accumulating cosmic-ray muons. We measured the muon charge ratio to be $R=1.32 \pm 0.02$ $(\mathrm{stat.}{+}\mathrm{syst.})$ at $E_{\mu}\cos \theta_{\mathrm{Zenith}}=0.7^{+0.3}_{-0.2}$ $\mathrm{TeV}$, where $E_{\mu}$ is the muon energy and $\theta_{\mathrm{Zenith}}$ is the zenith angle of incoming cosmic-ray muons. This result is consistent with the Honda flux model while this suggests a tension with the $\pi K$ model of $1.9\sigma$. We also measured the muon polarization at the production location to be $P^{\mu}_{0}=0.52 \pm 0.02$ $(\mathrm{stat.}{+}\mathrm{syst.})$ at the muon momentum of $0.9^{+0.6}_{-0.1}$ $\mathrm{TeV}/c$ at the surface of the mountain; this also suggests a tension with the Honda flux model of $1.5\sigma$. This is the most precise measurement ever to experimentally determine the cosmic-ray muon polarization near $1~\mathrm{TeV}/c$. Abstract: The first loading of gadolinium (Gd) into Super-Kamiokande in 2020 was successful, and the neutron capture efficiency on Gd reached 50\%. To further increase the Gd neutron capture efficiency to 75\%, 26.1 tons of $\rm Gd_2(\rm SO_4)_3\cdot \rm 8H_2O$ was additionally loaded into Super-Kamiokande (SK) from May 31 to July 4, 2022. As the amount of loaded $\rm Gd_2(\rm SO_4)_3\cdot \rm 8H_2O$ was doubled compared to the first loading, the capacity of the powder dissolving system was doubled. We also developed new batches of gadolinium sulfate with even further reduced radioactive impurities. In addition, a more efficient screening method was devised and implemented to evaluate these new batches of $\rm Gd_2(\rm SO_4)_3\cdot \rm 8H_2O$. Following the second loading, the Gd concentration in SK was measured to be $333.5\pm2.5$ ppm via an Atomic Absorption Spectrometer (AAS). From the mean neutron capture time constant of neutrons from an Am/Be calibration source, the Gd concentration was independently measured to be 332.7 $\pm$ 6.8(sys.) $\pm$ 1.1(stat.) ppm, consistent with the AAS result. Abstract: Among multi-messenger observations of the next galactic core-collapse supernova, Super-Kamiokande (SK) plays a critical role in detecting the emitted supernova neutrinos, determining the direction to the supernova (SN), and notifying the astronomical community of these observations in advance of the optical signal. On 2022, SK has increased the gadolinium dissolved in its water target (SK-Gd) and has achieved a Gd concentration of 0.033%, resulting in enhanced neutron detection capability, which in turn enables more accurate determination of the supernova direction. Accordingly, SK-Gd's real-time supernova monitoring system (Abe te al. 2016b) has been upgraded. SK_SN Notice, a warning system that works together with this monitoring system, was released on December 13, 2021, and is available through GCN Notices (Barthelmy et al. 2000). When the monitoring system detects an SN-like burst of events, SK_SN Notice will automatically distribute an alarm with the reconstructed direction to the supernova candidate within a few minutes. In this paper, we present a systematic study of SK-Gd's response to a simulated galactic SN. Assuming a supernova situated at 10 kpc, neutrino fluxes from six supernova models are used to characterize SK-Gd's pointing accuracy using the same tools as the online monitoring system. The pointing accuracy is found to vary from 3-7$^\circ$ depending on the models. However, if the supernova is closer than 10 kpc, SK_SN Notice can issue an alarm with three-degree accuracy, which will benefit follow-up observations by optical telescopes with large fields of view., Comment: 38 pages, 29 figures, 6 tables

Abstract: Oxidative stress (OS) in non-alcoholic fatty liver disease (NAFLD) promotes liver injury and inflammation. Treatment with vitamin E (α-tocopherol, αT), a lipid-soluble antioxidant, improves liver injury but also decreases steatosis, thought to be upstream of OS, through an unknown mechanism. To elucidate the mechanism, we combined a mechanistic human trial interrogating pathways of intrahepatic triglyceride (IHTG) accumulation and in vitro experiments. 50% of NAFLD patients (n = 20) treated with αT (200–800 IU/d) for 24 weeks had a ≥ 25% relative decrease in IHTG by magnetic resonance spectroscopy. Paired liver biopsies at baseline and week 4 of treatment revealed a decrease in markers of hepatic de novo lipogenesis (DNL) that strongly predicted week 24 response. In vitro, using HepG2 cells and primary human hepatocytes, αT inhibited glucose-induced DNL by decreasing SREBP-1 processing and lipogenic gene expression. This mechanism is dependent on the antioxidant capacity of αT, as redox-silenced methoxy-αT is unable to inhibit DNL in vitro. OS by itself was sufficient to increase S2P expression in vitro, and S2P is upregulated in NAFLD livers. In summary, we utilized αT to demonstrate a vicious cycle in which NAFLD generates OS, which feeds back to augment DNL and increases steatosis.Clinicaltrials.gov: NCT01792115.

Abstract: Background: Congenital diaphragmatic hernia (CDH) is a complex malformation with a challenging perioperative care. The outcome is still not very impressive in developing countries despite its remarkable improvement in developed countries. Methods: We analyzed outcomes, and factors associated with morbidity and mortality of 120 newborns with CDH, managed at our institution. Results: The overall survival was 73.4%. Chromosomal aberrations, pneumothorax within 24 hours, left ventricular hypoplasia, biventricular dysfunction, fetal lung to head ratio (LHR) Conclusion: This is the largest series of a retrospective cohort study of CDH reported in India. We have briefly reviewed the topic and the management guidelines along with prognostic markers derived from this study.

24. Evaluation of a bioengineered ACL matrix's osteointegration with BMP-2 supplementation.

Abstract: A poly (l-lactic) acid bioengineered anterior cruciate ligament (ACL) matrix has previously demonstrated the ability to support tissue regeneration in a rabbit ACL reconstruction model. The matrix was designed for optimal bone and ligament regeneration by developing a matrix with differential pore sizes in its bone and ligament compartments. Building upon past success, we designed a new bioengineered ACL matrix that is easier to install and can be used with endobutton fixation during ACL reconstruction. To achieve this, a new braiding procedure was developed to allow the matrix to be folded in half, making two-limbs, while maintaining its bone and ligament compartments. The osteointegration of the matrix with and without bone morphogenetic protein 2 (BMP-2) supplementation was evaluated in a rabbit ACL reconstruction model. Two doses of BMP-2 were evaluated, 1 and 10 μg, and delivered by saline injection into the bone tunnel at the end of surgery. A fibrous matrix-to-bone interface with occasional Sharpey's fibers was the primary mode of osteointegration observed. The matrix was also found to support a fibrocartilage matrix-to-bone interface. In some cases, the presence of chondrocyte-like cells was observed at the aperture of the bone tunnel and the center of the matrix within the bone tunnel. Treatment with BMP-2 was associated with a trend towards smaller bone tunnel cross-sectional areas, and 1 μg of BMP-2 was found to significantly enhance osteoid seam width in comparison with no BMP-2 or 10 μg of BMP-2 treatment. Regenerated tissue was well organized within the bioengineered ACL matrix and aligned with the poly (l-lactic) acid fibers. Disorganized tissue was found between the two-limbs of the bioengineered ACL matrix and hypothesized to be due to a lack of structural scaffolding. This study suggests that the bioengineered ACL matrix can undergo similar modes of osteointegration as current autografts and allografts, and that BMP-2 treatment may enhance osteoblastic activity within the bone tunnels.

Abstract: Background/Aims: Human enterocytic differentiation is altered during development, fasting, adaptation, and bariatric surgery, but its intracellular control remains unclear. We hypothesized that Schlafen 12 (SLFN12) regulates enterocyte differentiation. Methods: We used laser capture dissection of epithelium, qRT-PCR, and immunohistochemistry to evaluate SLFN12 expression in biopsies of control and fasting human duodenal mucosa, and viral overexpression and siRNA to trace the SLFN12 pathway in human Caco-2 and HIEC6 intestinal epithelial cells. Results: Fasting human duodenal mucosa expressed less SLFN12 mRNA and protein, accompanied by decreases in enterocytic markers like sucrase-isomaltase. SLFN12 overexpression increased Caco-2 sucrase-isomaltase promoter activity, mRNA, and protein independently of proliferation, and activated the SLFN12 putative promoter. SLFN12 coprecipitated Serpin B12 (SERPB12). An inactivating SLFN12 point mutation prevented both SERPB12 binding and sucrase-isomaltase induction. SERPB12 overexpression also induced sucrase-isomaltase, while reducing SERPB12 prevented the SLFN12 effect on sucrase-isomaltase. Sucrase-isomaltase induction by both SLFN12 and SERPB12 was attenuated by reducing UCHL5 or USP14, and blocked by reducing both. SERPB12 stimulated USP14 but not UCHL5 activity. SERPB12 coprecipitated USP14 but not UCHL5. Moreover, SLFN12 increased protein levels of the sucrase-isomaltase-promoter-binding transcription factor cdx2 without altering Cdx2 mRNA. This was prevented by reducing UCHL5 and USP14. We further validated this pathway in vitro and in vivo. SLFN12 or SERPB12 overexpression induced sucrase-isomaltase in human non-malignant HIEC-6 enterocytes. Conclusions: SLFN12 regulates human enterocytic differentiation by a pathway involving SERPB12, the deubiquitylases, and Cdx2. This pathway may be targeted to manipulate human enterocytic differentiation in mucosal atrophy, short gut or obesity.

Here, we report a case of TSC in a male patient who presented with oral and cutaneous manifestations without mental retardation or history of convulsive seizures, which is very rarely reported to the best of authors' knowledge.

Abstract: An analysis of solar neutrino data from the fourth phase of Super-Kamiokande~(SK-IV) from October 2008 to May 2018 is performed and the results are presented. The observation time of the data set of SK-IV corresponds to $2970$~days and the total live time for all four phases is $5805$~days. For more precise solar neutrino measurements, several improvements are applied in this analysis: lowering the data acquisition threshold in May 2015, further reduction of the spallation background using neutron clustering events, precise energy reconstruction considering the time variation of the PMT gain. The observed number of solar neutrino events in $3.49$--$19.49$ MeV electron kinetic energy region during SK-IV is $65,443^{+390}_{-388}\,(\mathrm{stat.})\pm 925\,(\mathrm{syst.})$ events. Corresponding $\mathrm{^{8}B}$ solar neutrino flux is $(2.314 \pm 0.014\, \rm{(stat.)} \pm 0.040 \, \rm{(syst.)}) \times 10^{6}~\mathrm{cm^{-2}\,s^{-1}}$, assuming a pure electron-neutrino flavor component without neutrino oscillations. The flux combined with all SK phases up to SK-IV is $(2.336 \pm 0.011\, \rm{(stat.)} \pm 0.043 \, \rm{(syst.)}) \times 10^{6}~\mathrm{cm^{-2}\,s^{-1}}$. Based on the neutrino oscillation analysis from all solar experiments, including the SK $5805$~days data set, the best-fit neutrino oscillation parameters are $\rm{sin^{2} \theta_{12,\,solar}} = 0.306 \pm 0.013 $ and $\Delta m^{2}_{21,\,\mathrm{solar}} = (6.10^{+ 0.95}_{-0.81}) \times 10^{-5}~\rm{eV}^{2}$, with a deviation of about 1.5$\sigma$ from the $\Delta m^{2}_{21}$ parameter obtained by KamLAND. Abstract: Cerebrospinal fluid (CSF) leak occurs due to a defect in the dura and skull base. Trauma remains the most common cause of CSF leak; however, a significant number of cases are iatrogenic, and result from a complication of functional endoscopic sinus surgery (FESS). Early diagnosis of CSF leak is of paramount importance to prevent life-threatening complications such as brain abscess and meningitis. Imaging plays a crucial role in the detection and characterization of CSF leaks. Three-dimensional, isotropic, high resolution computed tomography (HRCT) accurately detects the site and size of the bony defect. CT cisternography, though invasive, helps accurately identify the site of CSF leak, especially in the presence of multiple bony defects. Magnetic resonance imaging (MRI) accurately detects CSF leaks and associated complications such as the encephaloceles and meningoceles. In this review, we emphasize the importance and usefulness of 3D T2 DRIVE MR cisternography in localizing CSF leaks. This sequence has the advantages of effective bone and fat suppression, decreased artefacts, faster acquisition times, three-dimensional capability, y and high spatial resolution in addition to providing very bright signal from the CSF.

30. Drosophila Neuropeptide F Signaling Independently Regulates Feeding and Sleep-Wake Behavior

Abstract: Summary: Proper regulation of sleep-wake behavior and feeding is essential for organismal health and survival. While previous studies have isolated discrete neural loci and substrates important for either sleep or feeding, how the brain is organized to coordinate both processes with respect to one another remains poorly understood. Here, we provide evidence that the Drosophila Neuropeptide F (NPF) network forms a critical component of both adult sleep and feeding regulation. Activation of NPF signaling in the brain promotes wakefulness and adult feeding, likely through its cognate receptor NPFR. Flies carrying a loss-of-function NPF allele do not suppress sleep following prolonged starvation conditions, suggesting that NPF acts as a hunger signal to keep the animal awake. NPF-expressing cells, specifically those expressing the circadian photoreceptor cryptochrome, are largely responsible for changes to sleep behavior caused by NPF neuron activation, but not feeding, demonstrating that different NPF neurons separately drive wakefulness and hunger. : For optimal health, sleep and feeding behavior must be coordinated to not conflict with one another. However, it is unclear how the brain accomplishes this. Chung et al. demonstrate that enhanced Neuropeptide F (NPF) signaling promotes wakefulness and feeding behavior, presumably through different subsets of NPF-expressing neurons. Abstract: Knee osteoarthritis (OA) is the most common form of arthritis worldwide. The incidence of this disease is rising and its treatment poses an economic burden. Two early targets of knee OA treatment include the predominant symptom of pain, and cartilage damage in the knee joint. Current treatments have been beneficial in treating the disease but none is as effective as total knee arthroplasty (TKA). However, while TKA is an end-stage solution of the disease, it is an invasive and expensive procedure. Therefore, innovative regenerative engineering strategies should be established as these could defer or annul the need for a TKA. Several biomaterial and cell-based therapies are currently in development and have shown early promise in both preclinical and clinical studies. The use of advanced biomaterials and stem cells independently or in conjunction to treat knee OA could potentially reduce pain and regenerate focal articular cartilage damage. In this review, we discuss the pathogenesis of pain and cartilage damage in knee OA and explore novel treatment options currently being studied, along with some of their limitations.

Abstract: We present a measurement of neutrino oscillation parameters with the Super-Kamiokande detector using atmospheric neutrinos from the complete pure-water SK I-V (April 1996-July 2020) data set, including events from an expanded fiducial volume. The data set corresponds to 6511.3 live days and an exposure of 484.2 kiloton-years. Measurements of the neutrino oscillation parameters $\Delta m^2_{32}$, $\sin^2\theta_{23}$, $\sin^2 \theta_{13}$, $\delta_{CP}$, and the preference for the neutrino mass ordering are presented with atmospheric neutrino data alone, and with constraints on $\sin^2 \theta_{13}$ from reactor neutrino experiments. Abstract: We report the first measurement of the atmospheric neutrino-oxygen neutral-current quasielastic (NCQE) cross section in the gadolinium-loaded Super-Kamiokande (SK) water Cherenkov detector. In June 2020, SK began a new experimental phase, named SK-Gd, by loading 0.011% by mass of gadolinium into the ultrapure water of the SK detector. The introduction of gadolinium to ultrapure water has the effect of improving the neutron-tagging efficiency. Using a 552.2 day data set from August 2020 to June 2022, we measure the NCQE cross section to be 0.74 $\pm$ 0.22(stat.) $^{+0.85}_{-0.15}$ (syst.) $\times$ 10$^{-38}$ cm$^{2}$/oxygen in the energy range from 160 MeV to 10 GeV, which is consistent with the atmospheric neutrino-flux-averaged theoretical NCQE cross section and the measurement in the SK pure-water phase within the uncertainties. Furthermore, we compare the models of the nucleon-nucleus interactions in water and find that the Binary Cascade model and the Liege Intranuclear Cascade model provide a somewhat better fit to the observed data than the Bertini Cascade model. Since the atmospheric neutrino-oxygen NCQE reactions are one of the main backgrounds in the search for diffuse supernova neutrino background (DSNB), these new results will contribute to future studies - and the potential discovery - of the DSNB in SK., Comment: 8 pages, 3 figures

Abstract: We report a search for time variations of the solar $^8$B neutrino flux using 5804 live days of Super-Kamiokande data collected between May 31, 1996, and May 30, 2018. Super-Kamiokande measured the precise time of each solar neutrino interaction over 22 calendar years to search for solar neutrino flux modulations with unprecedented precision. Periodic modulations are searched for in a dataset comprising five-day interval solar neutrino flux measurements with a maximum likelihood method. We also applied the Lomb-Scargle method to this dataset to compare it with previous reports. The only significant modulation found is due to the elliptic orbit of the Earth around the Sun. The compressive strength of Reactive powder concrete (RPC) produced in this work was 88 MPa and the flexural strength was 20 MPa. Also the influence of different curing methods on the properties of RPC has been investigated. Durability studies also show that the material satisfies the durable parameters to be used as a construction material.

Abstract: Globulomaxillary cyst is a rare, spurious cyst. It was considered to be an inclusion or developmental cyst that arises from entrapped nonodontogenic epithelium in the globulomaxillary suture. Later, it was included as a fissural cyst secondary to proliferation of entrapped epithelium between the globular portion of the medial nasal and maxillary processes. Because of lack of evidence to support the theory of embryogenic epithelial entrapment in the site, most authors dispute the presence of the lesion. Herein, we are presenting a case of globulomaxillary cyst in a 20 year old female patient with an unusual radiographic appearance as a full moon.

Abstract: We report an updated measurement of the $\nu_{\mu}$-induced, and the first measurement of the $\bar{\nu}_{\mu}$-induced coherent charged pion production cross section on $^{12}C$ nuclei in the T2K experiment. This is measured in a restricted region of the final-state phase space for which $p_{\mu,\pi} > 0.2$ GeV, $\cos(\theta_{\mu}) > 0.8$ and $\cos(\theta_{\pi}) > 0.6$, and at a mean (anti)neutrino energy of 0.85 GeV using the T2K near detector. The measured $\nu_{\mu}$ CC coherent pion production flux-averaged cross section on $^{12}C$ is $(2.98 \pm 0.37 (stat.) \pm 0.31 (syst.) \substack{ +0.49 \\ -0.00 } \mathrm{ (Q^2\,model)}) \times 10^{-40}~\mathrm{cm}^{2}$. The new measurement of the $\bar{\nu}_{\mu}$-induced cross section on $^{12}{C}$ is $(3.05 \pm 0.71 (stat.) \pm 0.39 (syst.) \substack{ +0.74 \\ -0.00 } \mathrm{(Q^2\,model)}) \times 10^{-40}~\mathrm{cm}^{2}$. The results are compatible with both the NEUT 5.4.0 Berger-Sehgal (2009) and GENIE 2.8.0 Rein-Sehgal (2007) model predictions.

Abstract: Muon neutrino and antineutrino disappearance probabilities are identical in the standard three-flavor neutrino oscillation framework, but CPT violation and non-standard interactions can violate this symmetry. In this work we report the measurements of $\sin^{2} \theta_{23}$ and $\Delta m_{32}^2$ independently for neutrinos and antineutrinos. The aforementioned symmetry violation would manifest as an inconsistency in the neutrino and antineutrino oscillation parameters. The analysis discussed here uses a total of 1.97$\times$10$^{21}$ and 1.63$\times$10$^{21}$ protons on target taken with a neutrino and antineutrino beam respectively, and benefits from improved flux and cross-section models, new near detector samples and more than double the data reducing the overall uncertainty of the result. No significant deviation is observed, consistent with the standard neutrino oscillation picture.

41. Impact of errors in the magnetic field measurement on the precision determination of neutrino oscillation parameters at the proposed ICAL detector at INO

Abstract: The magnetised iron calorimeter (ICAL) detector proposed at the India-based Neutrino Observatory will be a 51 kton detector made up of 151 layers of 56 mm thick soft iron with 40 mm air gap in between where the RPCs, the active detectors, will be placed. The main goal of ICAL is to make precision measurements of the neutrino oscillation parameters using the atmospheric neutrinos as source. The charged current interactions of the atmospheric muon neutrinos and anti-neutrinos in the detector produce charged muons. The magnetic field, with a maximum value of $\sim$ 1.5 T in the central region of ICAL, is a critical component since it will be used to distinguish the charges and determine the momentum and direction of these muons. It is difficult to measure the magnetic field inside the iron. The existing methods can only estimate the internal field and hence will be prone to error. This paper presents the first simulations study of the effect of errors in the measurement of the magnetic field in ICAL on its physics potential, especially the neutrino mass ordering and precision measurement of oscillation parameters in the 2--3 sector. The study is a GEANT4-based analysis, using measurements of the magnetic field at the prototype ICAL detector. We find that there is only a small effect on the determination of the mass ordering. While local fluctuations in the magnetic field measurement are well-tolerated, calibration errors must remain well within 5\% to retain good precision determination of the parameters $\sin^2\theta_{23}$ and $\Delta m^2_{32}$., Comment: 25 pages, 10 figures latex; had uploaded Fig6(R) and Fig 7 (R) wrongly; now replaced in version2

Abstract: We report the first search result for the flux of astrophysical electron antineutrinos for energies O(10) MeV in the gadolinium-loaded Super-Kamiokande (SK) detector. In June 2020, gadolinium was introduced to the ultra-pure water of the SK detector in order to detect neutrons more efficiently. In this new experimental phase, SK-Gd, we can search for electron antineutrinos via inverse beta decay with efficient background rejection and higher signal efficiency thanks to the high efficiency of the neutron tagging technique. In this paper, we report the result for the initial stage of SK-Gd with a $22.5\times552$ $\rm kton\cdot day$ exposure at 0.01% Gd mass concentration. No significant excess over the expected background in the observed events is found for the neutrino energies below 31.3 MeV. Thus, the flux upper limits are placed at the 90% confidence level. The limits and sensitivities are already comparable with the previous SK result with pure-water ($22.5 \times 2970 \rm kton\cdot day$) owing to the enhanced neutron tagging.

Abstract: This paper reports the first measurement of muon neutrino charged-current interactions without pions in the final state using multiple detectors with correlated energy spectra at T2K. The data was collected on hydrocarbon targets using the off-axis T2K near detector (ND280) and the on-axis T2K near detector (INGRID) with neutrino energy spectra peaked at 0.6 GeV and 1.1 GeV respectively. The correlated neutrino flux presents an opportunity to reduce the impact of the flux uncertainty and to study the energy dependence of neutrino interactions. The extracted double-differential cross sections are compared to several Monte Carlo neutrino-nucleus interaction event generators showing the agreement between both detectors individually and with the correlated result., Comment: Updated discussion in Sec. Abstract: The T2K experiment presents new measurements of neutrino oscillation parameters using $19.7(16.3)\times10^{20}$ protons on target (POT) in (anti-)neutrino mode at the far detector (FD). Compared to the previous analysis, an additional $4.7\times10^{20}$ POT neutrino data was collected at the FD. Significant improvements were made to the analysis methodology, with the near-detector analysis introducing new selections and using more than double the data. Additionally, this is the first T2K oscillation analysis to use NA61/SHINE data on a replica of the T2K target to tune the neutrino flux model, and the neutrino interaction model was improved to include new nuclear effects and calculations. Frequentist and Bayesian analyses are presented, including results on $\sin^2\theta_{13}$ and the impact of priors on the $\delta_\mathrm{CP}$ measurement. Both analyses prefer the normal mass ordering and upper octant of $\sin^2\theta_{23}$ with a nearly maximally CP-violating phase. Assuming the normal ordering and using the constraint on $\sin^2\theta_{13}$ from reactors, $\sin^2\theta_{23}=0.561^{+0.021}_{-0.032}$ using Feldman--Cousins corrected intervals, and $\Delta{}m^2_{32}=2.494_{-0.058}^{+0.041}\times10^{-3}~\mathrm{eV^2}$ using constant $\Delta\chi^{2}$ intervals. The CP-violating phase is constrained to $\delta_\mathrm{CP}=-1.97_{-0.70}^{+0.97}$ using Feldman--Cousins corrected intervals, and $\delta_\mathrm{CP}=0,\pi$ is excluded at more than 90% confidence level. A Jarlskog invariant of zero is excluded at more than $2\sigma$ credible level using a flat prior in $\delta_\mathrm{CP}$, and just below $2\sigma$ using a flat prior in $\sin\delta_\mathrm{CP}$. When the external constraint on $\sin^2\theta_{13}$ is removed, $\sin^2\theta_{13}=28.0^{+2.8}_{-6.5}\times10^{-3}$, in agreement with measurements from reactor experiments. These results are consistent with previous T2K analyses.

Abstract: Cosmic-ray muons that enter the Super-Kamiokande detector cause hadronic showers due to spallation in water, producing neutrons and radioactive isotopes. Those are a major background source for studies of MeV-scale neutrinos and searches for rare events. Since 2020, gadolinium was introduced in the ultra-pure water in the Super-Kamiokande detector to improve the detection efficiency of neutrons. In this study, the cosmogenic neutron yield was measured using data acquired during the period after the gadolinium loading. The yield was found to be $(2.76 \pm 0.02\,\mathrm{(stat.) \pm 0.19\,\mathrm{(syst.)}}) \times 10^{-4}\,\mu^{-1} \mathrm{g^{-1} cm^{2}}$ at 259 GeV of average muon energy at the Super-Kamiokande detector., Comment: 10 pages, 10 figures, 3 tables

46. Study of nasal carriage of MRSA among the clinical staff and health care workers of a teaching hospital of Karnataka, India

Abstract: Objective: The present study was conducted to evaluate the rate of nasal carriage of Methicillin Resistant Staphylococcus aureus among the clinical staff and health careworkers working at our hospital with an aim to prevent the hospital acquired infections. Background: Methicillin resistant Staphylococcus aureus colonisation precedes infection, anterior nares being the ecological niches of Staphylcoccus aureus. Carriage of Staphylococcus aureus in the nose appears to play a key role in the epidemiology and pathogenesis of infection. Methicillin resistant Staphylococcus aureus is usually introduced into an institution by a colonised or infected patient or a healthcare worker.When nose is treated topically to eliminate nasal carriage, in most cases the organism also disappears from other areas of the body like groin, axilla, umbilicus, and hands. Methods: A total of 200 nasal swabs were collected, out of which 140 were from the nursing staff and 60 were from clinical staff. Sterile cotton swabs moistened with sterile saline were used for sample collection. Swabs were cultured on to blood agar, and mannitol salt agar, incubated at 37 oC for 24 hrs. Staphylococcus aureus was identified by standard methods according to CLSI guidelines. Methicillin resistance was detected by using cefoxitin disc 30µgm on Mueller Hinton agar with 4% NaCl. Results: Of the 200 samples screened 45 (43.6%) strains of Staphylococcus aureus were isolated, out of which 24 (12%) were. Methicillin resistant Staphylococcus aureus (MRSA) and 21 (10%) were methicillin sensitive Staphylococcus aureus (MSSA). The overall carriage rate of methicillin resistant Staphylococcus aureus in our study was 12% with the highest rate being seen among the nursing staff (12.2%) and clinical staff carriage rate was slightly less (11.7%) as compared to the nursing staff. Conclusion: Our study revealed that nursing staff were the potential colonisers of methicillin resistant Staphylococcus aureus when compared to clinical staff. These carriers may serve as reservoir and disseminator of MRSA, and should be treated with mupirocin 3 times daily for 5 days. So regular screening of carriers is required for the prevention of nosocomial infection.

47. ORIGINAL ARTICLE: Detection of in-vitro Activity of Linezolid in Methicillin Resistant Staphylococcus aureus Infections by E-test

Abstract: Aim: To determine the in-vitro activity of linezolid in methicillin resistant Staphylococcus aureus. Material and Methods: Samples such as pus, sputum, ear swab, pleural fluid that were received in microbiology laboratory were included in the study. Gram staining was performed on all the samples. Smears showing gram positive cocci in clusters were considered. 100 strains of Staphylococci were subsequently processed and identified using standard microbiological methods. Staphylococcus aureus so identified were subjected to screening for methicillin resistance using oxacillin disc (1µgm) and also in-vitro activity of linezolid by Kirby-Bauer disc diffusion method. Subsequently minimum inhibitory concentration for linezolid was determined by E-test using Hi-Comb strips obtained from Hi-Media. Results: Out of 100 strains of Staphylococcus aureus isolated, 42 (42%) were methicillin resistant and 58 (58%) were methicillin sensitive. Majority of methicillin resistant Staphylococcus aureus isolated were from pus [35] followed by sputum [7]. Among the 42 strains of methicillin resistant Staphylococcus aureus isolated, sensitivity to linezolid (30mcg) was 87.71% and minimum inhibitory concentration was in the range of 0.1 - 1µgm/ml. Conclusion: Linezolid is a novel alternative drug to vancomycin in the treatment of severe methicillin resistant Staphylococcus aureus infections.

Abstract: Neutrinos associated with solar flares (solar-flare neutrinos) provide information on particle acceleration mechanisms during the impulsive phase of solar flares. We searched using the Super-Kamiokande detector for neutrinos from solar flares that occurred during solar cycles $23$ and $24$, including the largest solar flare (X28.0) on November 4th, 2003. In order to minimize the background rate we searched for neutrino interactions within narrow time windows coincident with $\gamma$-rays and soft X-rays recorded by satellites. In addition, we performed the first attempt to search for solar-flare neutrinos from solar flares on the invisible side of the Sun by using the emission time of coronal mass ejections (CMEs). By selecting twenty powerful solar flares above X5.0 on the visible side and eight CMEs whose emission speed exceeds $2000$ $\mathrm{km \, s^{-1}}$ on the invisible side from 1996 to 2018, we found two (six) neutrino events coincident with solar flares occurring on the visible (invisible) side of the Sun, with a typical background rate of $0.10$ ($0.62$) events per flare in the MeV-GeV energy range. No significant solar-flare neutrino signal above the estimated background rate was observed. As a result we set the following upper limit on neutrino fluence at the Earth $\mathit{\Phi}<1.1\times10^{6}$ $\mathrm{cm^{-2}}$ at the $90\%$ confidence level for the largest solar flare. Abstract: We report a search for cosmic-ray boosted dark matter with protons using the 0.37 megaton$\times$years data collected at Super-Kamiokande experiment during the 1996-2018 period (SKI-IV phase). We searched for an excess of proton recoils above the atmospheric neutrino background from the vicinity of the Galactic Center. No such excess is observed, and limits are calculated for two reference models of dark matter with either a constant interaction cross-section or through a scalar mediator. This is the first experimental search for boosted dark matter with hadrons using directional information. The results present the most stringent limits on cosmic-ray boosted dark matter and exclude the dark matter-nucleon elastic scattering cross-section between $10^{-33}\text{ cm}^{2}$ and $10^{-27}\text{ cm}^{2}$ for dark matter mass from 10 MeV/$c^2$ to 1 GeV/$c^2$., Comment: With 1-page appendix.
