The Lower Ordovician Tongzi Formation containing abundant shoal sediments is the most promising stratum for the petroleum exploration in the Sichuan Basin. However, the current studies mainly focus on the central part of the Basin, the systematic analysis of the southeastern part with well-developed shoal facies is lacking. This paper aims to clarify the characteristics and genesis of the Tongzi Formation reservoir in Southeastern Sichuan Basin, following an analysis of sedimentary facies within the sequence stratigraphic framework. The research shows that the main types of reservoir rocks are oolitic, intraclastic and bioclastic dolostones. And the reservoir spaces consist mainly of intergranular (dissolved) pores, intercrystalline (dissolved) pores, intragranular dissolved pores and fractures. Among them, intergranular pores account for the highest proportion, which is followed by intragranular dissolved pores. In addition, most throats are necking and flaky which mainly connect intergranular and intercrystalline pores, respectively. The diagenetic sequence shows that penecontemporaneous karst well improved the porosity of the reservoir in the early-stage although the cementation and compaction reduced parts of pores. The reservoir formation is associated with the tectonic-depositional settings, diageneses and terrigenous contamination. Paleohighlands and submerged uplifts, forming in the early Ordovician amalgamation between Yangtze and Cathaysia blocks, accumulated shoal sediments as the material basis for the reservoir formation. Penecontemporaneous karst forming intragranular dissolved pores and the dolomitization aiding grainstones to resist the pressure solution are the key to increasing porosity and preserving pores. The absence of terrigenous contamination prevented intergranular pores from being strongly cemented, which resulted in the reservoir difference between the central and southeastern Sichuan Basin. The study can be used as a reference for the further exploration of Ordovician petroleum in the Sichuan Basin and other regions owning similar geological settings.

Unnecessary exposure to ionizing radiation (IR) often causes acute and chronic oxidative damages to normal cells and organs, leading to serious physiological and even life-threatening consequences. Amifostine (AMF) is a validated radioprotectant extensively applied in radiation and chemotherapy medicine, but the short half-life limits its bioavailability and clinical applications, remaining as a great challenge to be addressed. DNA-assembled nanostructures especially the tetrahedral framework nucleic acids (tFNAs) are promising nanocarriers with preeminent biosafety, low biotoxicity, and high transport efficiency. The tFNAs also have a relative long-term maintenance for structural stability and excellent endocytosis capacity. We therefore synthesized a tFNA-based delivery system of AMF for multi-organ radioprotection (tFNAs@AMF, also termed nanosuit). By establishing the mice models of accidental total body irradiation (TBI) and radiotherapy model of Lewis lung cancer, we demonstrated that the nanosuit could shield normal cells from IR-induced DNA damage by regulating the molecular biomarkers of anti-apoptosis and anti-oxidative stress. In the accidental total body irradiation (TBI) mice model, the nanosuit pretreated mice exhibited satisfactory alteration of superoxide dismutase (SOD) activities and malondialdehyde (MDA) contents, and functional recovery of hematopoietic system, reducing IR-induced pathological damages of multi-organ and safeguarding mice from lethal radiation. More importantly, the nanosuit showed a selective radioprotection of the normal organs without interferences of tumor control in the radiotherapy model of Lewis lung cancer. Based on a conveniently available DNA tetrahedron-based nanocarrier, this work presents a high-efficiency delivery system of AMF with the prolonged half-life and enhanced radioprotection for multi-organs. Such nanosuit pioneers a promising strategy with great clinical translation potential for radioactivity protection.

Abstract Background Lung cancer, a leading cause of cancer mortality, poses significant treatment challenges. The use of immune checkpoint inhibitors (ICIs) has revolutionized therapy, but it is associated with immune-related pneumonitis (IRP). This study systematically reviews and analyzes the impact of Chronic Obstructive Pulmonary Disease (COPD) on the risk of IRP in lung cancer patients undergoing immunotherapy. Methods Adhering to PRISMA guidelines and using the PICO framework, a comprehensive search across PubMed, Embase, Web of Science, and the Cochrane Library was conducted. Inclusion criteria encompassed peer-reviewed studies involving lung cancer patients treated with ICIs, comparing those with and without COPD. The primary outcome was the incidence and risk of IRP. The Newcastle-Ottawa Scale evaluated study quality. The effect size was calculated using random or fixed-effects models based on the observed heterogeneity. We assessed the heterogeneity between studies and conducted a sensitivity analysis. Results The search identified 1026 articles, with six meeting the criteria for inclusion. The structure and function of microbial communities in aquaculture water bodies are important in the aquaculture ecosystem. To comprehensively and systematically evaluate the structure and functional composition of the microbial community in the water during Chinese mitten crab (Eriocheir sinensis) cultivation, the water quality indicators in the crab pond in Chongming District, Shanghai were monitored monthly from June to October 2022. The microbial species and functional structural characteristics in the water environment during the breeding period were analyzed based on metagenomics technology, and their relationships with environmental factors were discussed. The results showed that the major water quality indicators that exceeded the standard in the breeding pond were pH, permanganate index, total phosphorus, and total nitrogen. During the breeding period, the diversity of the microbial community in the water body from June to August and the richness of the microbial community from July to August were high, and the dominant microbial phyla in the water body were mainly Proteobacteria, Bacteroidetes, Actinobacteria, Verrucomicrobia, Cyanobacteria, and Uroviricota. At the genus level, the top 10 dominant genera regarding abundance were significantly different among multiple groups. For example, the abundances of Microcystis bacteria in July and unclassified_o_Caudovirales viruses in August–October were significantly higher than those in other months. The primary function of microorganisms is metabolism, including energy metabolism, global and overview maps, and amino acid metabolism. Notable differences are present in the functional composition of different months; particularly, the abundance of metabolic pathways in June–July was significantly higher than that in August–October, and the dominant bacteria (Proteobacteria, Actinobacteria, and Bacteroidetes) were major contributors to the aforementioned functions. The influence trends of environmental factors on the structure and functional composition of microbial communities were consistent. Chlorophyll a and pH had the most significant impact on the structure and functional composition of microbial communities, whereas dissolved oxygen and total phosphorus had a slightly weaker impact. The pathogenic bacteria with a relatively large abundance in aquaculture water bodies are Salmonella enterica, Edwardsiella ictaluri, and Staphylococcus aureus. The research results provide basic data for the study of microbial community structure and functional composition in water bodies of Chinese mitten crab breeding ponds and can provide a theoretical basis for aquaculture water quality control and ecosystem construction.

Alpine mountain ecosystems are highly sensitive to global warming and threatened by climate change. Elevation gradients are frequently used as space-for-time surrogates for climate change in mountain ecosystems due to their correspondence with climate variables over short geographic distances. However, the distribution patterns of fungal communities in perennial woody plants along elevations in this ecosystem and how elevation and host identity affect them remain unclear. In this work, we used Illumina MiSeq DNA sequencing technology to study the rhizosphere soil fungal community and root and leaf endophytic fungal community assembly of alpine Rhododendron communities across a 3,100- to 4,100-m elevation gradient on Jiaozi Mountain, Southwest China. The abundance and diversity of fungi in different compartments varied among elevation gradients and hosts. The fungal community assembly in each compartment was mainly influenced by stochastic processes. However, the contribution of homogeneous selection, dispersal limitation, and drift varied among host identities and elevations in different compartments. The diversity and composition of rhizosphere soil, root, and leaf guilds and the overall abundance of fungal functional groups tracked the Rhododendron host identity more closely than elevation did. Most indicator taxa of fungi were putative saprotrophs. These findings will improve our understanding of the maintenance of alpine Rhododendron communities along elevation gradients. Abstract Purpose This study aimed to identify and quantify the association and investigate whether serum vitamin B12 alone or vitamin B12 combined with folate and plasma total homocysteine (tHcy) levels could be used to predict the risk of acute ischemic stroke. Materials and methods This retrospective case-control study was conducted in the Department of Neurology, First Affiliated Hospital of Chongqing Medical University. It included 259 inpatients experiencing their first-ever acute ischemic stroke and 259 age-matched, sex-matched healthy controls. Patients were categorized into groups based on the etiology of their stroke: large-artery atherosclerosis (LAAS, n = 126), cardio embolism (CEI, n = 35), small vessel disease (SVD, n = 89), stroke of other determined etiology (ODE, n = 5), and stroke of undetermined etiology (UDE, n = 4). The associations of serum vitamin B12, folate, and plasma tHcy levels with the risk of ischemic stroke were evaluated using multivariable logistic regression analysis. Receiver operator characteristic (ROC) curves were used to assess the diagnostic power of vitamin B12, folate, and tHcy levels for ischemic stroke. Results Serum vitamin B12 and folate levels were significantly lower in ischemic stroke patients compared to controls, while plasma tHcy levels were significantly higher. The first quartile of serum vitamin B12 levels was significantly associated with an increased risk of LAAS (aOR = 2.289, 95% CI = 1.098–4.770), SVD (aOR = 4.471, 95% CI = 1.110–4.945) and overall ischemic stroke (aOR = 3.216, 95% CI = 1.733–5.966). Similarly, the first quartile of serum folate levels was associated with an increased risk of LAAS (aOR = 3.480, 95% CI = 1.954–6.449), CEI (aOR = 2.809, 95% CI = 1.073–4.991), SVD (aOR = 5.376, 95% CI = 1.708–6.924), and overall ischemic stroke (aOR = 3.381, 95% CI = 1.535–7.449). The fourth quartile of tHcy levels was also significantly associated with an increased risk of LAAS (aOR = 2.946, 95% CI = 1.008–5.148), CEI (aOR = 2.212, 95% CI = 1.247–5.946), SVD (aOR = 2.957, 95% CI = 1.324–6.054), and overall ischemic stroke (aOR = 2.233, 95% CI = 1.586–4.592). For predicting different types of ischemic stroke, vitamin B12 alone demonstrated the best diagnostic value for SVD, evidenced by a sensitivity of 71.0% and negative predictive value of 90.3%, along with the highest positive likelihood ratio (+ LR) for SVD. Vitamin B12 + tHcy + folate are valuable in predicting different types of ischemic stroke, with the most significant effect observed in SVD, followed by LAAS, and the weakest predictive effect in CEI. Additionally, vitamin B12 alone in combination with other indicators, such as folate alone, tHcy alone, and folate + tHcy could reduce negative likelihood ratio (-LR) and improve + LR. Conclusions Vitamin B12 was an independent risk factor for acute ischemic stroke. The risk calculation model constructed with vitamin B12 + tHcy + folate had the greatest diagnostic value for SVD.

Abstract Background Despite evidence showing a connection between inflammation and endometrial cancer (EC) risk, the surveys on genetic correlation and cohort studies investigating the impact on long-term outcomes have yet to be refined. We aimed to address the impact of inflammation factors on the pathogenesis, progression and consequences of EC. Methods For the genetic correlation analyses, a two-sample of Mendelian randomization (MR) study was applied to investigate inflammation-related single-nucleotide polymorphisms involved with endometrial cancer from GWAS databases. The observational retrospective study included consecutive patients diagnosed with EC (stage I to IV) with surgeries between January 2010 and October 2020 at the Cancer Hospital of Shantou University Medical College. Results The 2-sample MR surveys indicated no causal relationship between inflammatory cytokines and endometrial cancer. 780 cases (median age, 55.0 years ) diagnosed with EC were included in the cohort and followed up for an average of 6.8 years. Increased inflammatory parameters at baseline were associated with a higher FIGO stage and invasive EC risk (odds ratios [OR] 1.01 to 4.20). Abstract Background The risk assessment for survival in heart failure (HF) remains one of the key focuses of research. This study aims to develop a simple and feasible nomogram model for survival in HF based on the Heart Failure-A Controlled Trial Investigating Outcomes of Exercise TraiNing (HF-ACTION) to support clinical decision-making. Methods The HF patients were extracted from the HF-ACTION database and randomly divided into a training cohort and a validation cohort at a ratio of 7:3. Multivariate Cox regression was used to identify and integrate significant prognostic factors to form a nomogram, which was displayed in the form of a static nomogram. Bootstrap resampling (resampling = 1000) and cross-validation was used to internally validate the model. The prognostic performance of the model was measured by the concordance index (C-index), calibration curve, and the decision curve analysis. Results There were 1394 patients with HF in the overall analysis. Seven prognostic factors, which included age, body mass index (BMI), sex, diastolic blood pressure (DBP), exercise duration, peak exercise oxygen consumption (peak VO2), and loop diuretic, were identified and applied to the nomogram construction based on the training cohort. The C-index of this model in the training cohort was 0.715 (95% confidence interval (CI): 0.700, 0.766) and 0.662 (95% CI: 0.646, 0.752) in the validation cohort. The area under the ROC curve (AUC) value of 365- and 730-day survival is (0.731, 0.734) and (0.640, 0.693) respectively in the training cohort and validation cohort. The calibration curve showed good consistency between nomogram-predicted survival and actual observed survival. The decision curve analysis (DCA) revealed net benefit is higher than the reference line in a narrow range of cutoff probabilities and the result of cross-validation indicates that the model performance is relatively robust. Conclusions This study created a nomogram prognostic model for survival in HF based on a large American population, which can provide additional decision information for the risk prediction of HF.

Abstract Background Heart failure (HF) is a complex cardiovascular syndrome with high mortality. Santalum album L. (SAL) is a traditional Chinese medicine broadly applied for various diseases treatment including HF. However, the potential active compounds and molecular mechanisms of SAL in HF treatment are not well understood. Methods The active compounds and possible mechanisms of action of SAL were analyzed and validated by a systems pharmacology framework and an ISO-induced mouse HF model. Results We initially confirmed that SAL alleviates heart damage in ISO-induced HF model. A total of 17 potentially active components in SAL were identified, with Luteolin (Lut) and Syringaldehyde (SYD) in SAL been identified as the most effective combination through probabilistic ensemble aggregation (PEA) analysis. These compounds, individually and in their combination (COMB), showed significant therapeutic effects on HF by targeting multiple pathways involved in anti-oxidation, anti-inflammation, and anti-apoptosis. The active ingredients in SAL effectively suppressed inflammatory mediators and pro-apoptotic proteins while enhancing the expression of anti-apoptotic factors and antioxidant markers. Furthermore, the synergistic effects of SAL on YAP and PI3K-AKT signaling pathways were further elucidated. Conclusions Mechanistically, the anti-HF effect of SAL is responsible for the synergistic effect of anti-inflammation, antioxidation and anti-apoptosis, delineating a multi-targeted therapeutic strategy for HF. Abstract In this study, the effects of sea buckthorn oil (SBO), fish oil (FO) and an enzymatically synthesized structured lipid (SL) on serum, short-chain fatty acids (SCFAs) and intestinal microbiota in Sprague–Dawley (SD) rats were investigated. The results demonstrated that FO, SBO, and SL effectively reduced the levels of high-density lipoprotein cholesterol and low-density lipoprotein cholesterol in the serum of SD rats. SBO increased serum triglyceride levels, while FO elevated total cholesterol levels. Furthermore, all three dietary lipids decreased short-chain fatty acid production and enhanced intestinal microbiota diversity. FO increased the abundance of intestinal microbiota including Romboutsia, Lactobacillus, Escherichia-Shigella, and Lachnospiraceae_NK4A136_group. Conversely, all three dietary lipids reduced the abundance of Klebsiella and Blautia. These findings provide a foundation for understanding the functionality of SBO and FO as well as their potential application in synthesizing novel SLs to regulate intestinal microbiota. In the case of electrical contact, copper alloys are often unable to withstand arc ablation and premature failure. To solve this issue, Ni-8wt.%Al (Ni–8Al) coatings were successfully prepared on copper substrates utilizing the high-speed laser cladding. In addition, the microstructure evolution, microhardness, electric conductivity and arc ablation resistance of the prepared coatings were investigated in detail. As a result, the achieved Ni–8Al coatings were structurally dense with good metallurgical bonding to the substrate, of which the microstructure showed a bottom-up evolution of fine sub-grain structure. Notably, the microhardness of the coating was 191.7 HV0.2, representing an increase of 82.57% compared to the substrate, and the electrical conductivity was 18.03 %IACS, approaching that of pure nickel. Additionally, the increase in ablation currents led to changes in the ablation behaviors and failure mechanisms of both the substrate and coating. At high currents, a huge arc ablation crater appeared in the center of the substrate due to the reaction force of the anode flame and the difference in surface tension between the liquid metal at the center and the edge of the molten pool. Nevertheless, the coatings showed better arc ablation resistance due to the improvement of molten pool stability during arc ablation by the in situ formed Al2O3 layer on the coating surface.

In the context of sustainable urban development, effective garbage management plays a crucial role. However, traditional methods encounter limitations in terms of data quality and quantity. The research on automatic garbage image recognition and classification methods based on deep learning has been gaining attention. This study proposes an integrated garbage image recognition and classification method that combines ResNet-50, YOLOv5, and weakly supervised CNN algorithms. The aim is to enhance both the accuracy and efficiency of image recognition, optimize intelligent garbage management, and promote urban sustainable development planning. The ResNet-50 model is employed to extract meaningful features from images and train weakly supervised CNN models for subsequent training and prediction. This enables the analysis of urban environmental development trends and the formulation of planning measures. Through evaluation on four representative public datasets, the proposed method outperforms several traditional algorithms in terms of accuracy, efficiency, and stability in garbage image recognition systems. Notably, on the HGI-30 dataset, the algorithm achieves significant improvements by reducing inference time by over 48.6%, FLOPs by over 46.5%, and MAPE by over 41%. These enhancements greatly enhance the accuracy and robustness of garbage image classification, highlighting the substantial significance of this method in the realms of garbage management and environmental protection.

The harvest moisture content (MC) of maize (Zea mays L.) is an important factor affecting industrial processing and harvest quality. However, higher MC levels may escalate drying expenses during production. While augmented plant density has shown potential to enhance maize yield, limited information exists regarding its impact on plant density and MC interaction. This study amassed four year of field trial data from four primary maize ecological areas in China (Longitude: 89°E to 125°E, Latitude: 35°N to 44°N) to examine the relationship between plant density and MC of maize. Our analysis revealed that increasing plant density exerted an effect on the MC of maize. This effect stemmed from a combination of uncertain grain development and premature plant senescence due to overcrowding. We categorized the relationship between plant density and MC into six types: NC-Type (No-changing), I-Type (ever-increasing), L-Type (ever-lessening), V-Type (like the valley), P-Type (like the peak), and W-Type (like a wave). On average, a variation approximately 0.053 % in MC was observed when the plant population changed by 1000 plants/ha. Moreover, different maize varieties contributed to a more substantial variation in MC. Despite the negligible impact of MC variation caused by plant density on grain quality, subsequent harvest management, and industrial production of maize, there was a significant enhancement in maize yield with increased plant density. Thus, increasing plant density presents an opportunity to boost maize output without compromising quality or grain moisture levels.

A novel lightweight, radiation-shielding Mg-Ta-Al layered metal-matrix composite (LMC) was successful designed by doping the extremely refractory metal (Ta) into Mg sheets. These Mg-based LMCs sheets shows excellent radiation-dose shield effect, about 145 krad·a−1, which is about 17 times of traditional Mg alloy, while its surface density is only about 0.9 g·cm−2, reducing by 60% than that of pure Ta. The quantitate relationship between radiation-dose and the materials’ thickness was also confirmed to the logistic function when the surface density is in the range of 0.6–1.5 g·cm−2. Meantime, the rolling parameters, interface microstructure and mechanical properties in both as-rolled and annealing treated samples were evaluated. The sheets possess a special dissimilar atoms diffusion transitional zone containing an obvious inter-diffusion Mg-Al interface and the unique micro-corrugated Ta-Al interface, as well as a thin Al film with a thickness of about 10 µm. The special zone could reduce the stress concentration and enhance the strength of Mg-Ta-Al LMCs. The interface bonding strength reaches up to 54–76 MPa. The ultimate tensile strength (UTS) and yield strength (TYS) of the Mg-Ta-Al sheet were high to 413 MPa and 263 MPa, respectively, along with an elongation of 5.8%. The molecular dynamics (MD) analysis results show that the two interfaces exhibit different formation mechanism, the Mg-Al interface primarily depended on Mg/Al atoms diffusion basing point defects movement, while the Ta-Al interface with a micro-interlock pining shape formed by close-packed planes slipping during high temperature strain-induced deformation process.

Context information is significant for semantic extraction and recovery of messages in semantic communication. However, context information is not fully utilized in the existing semantic communication systems since relationships between sentences are often ignored. In this paper, we propose an Extended Context-based Semantic Communication (ECSC) system for text transmission, in which context information within and between sentences is explored for semantic representation and recovery. At the encoder, self-attention and segment-level relative attention are used to extract context information within and between sentences, respectively. In addition, a gate mechanism is adopted at the encoder to incorporate the context information from different ranges. At the decoder, Transformer-XL is introduced to obtain more semantic information from the historical communication processes for semantic recovery. Simulation results show the effectiveness of our proposed model in improving the semantic accuracy between transmitted and recovered messages under various channel conditions.

Increasing research has focused on semantic communication, the goal of which is to convey accurately the meaning instead of transmitting symbols from the sender to the receiver. In this paper, we design a novel encoding and decoding semantic communication framework, which adopts the semantic information and the contextual correlations between items to optimize the performance of a communication system over various channels. On the sender side, the average semantic loss caused by the wrong detection is defined, and a semantic source encoding strategy is developed to minimize the average semantic loss. To further improve communication reliability, a decoding strategy that utilizes the semantic and the context information to recover messages is proposed in the receiver. Extensive simulation results validate the superior performance of our strategies over state-of-the-art semantic coding and decoding policies on different communication channels.

Abstract Background The global aging population presents challenges that are particularly acute in China. Older Chinese adults’ attitudes towards death significantly impact their quality of life. Death education is crucial for promoting positive perspectives on life and death. Narrative education offers a promising approach to facilitating death education. Integrating the Knowledge-Attitude-Practice (KAP) model into death education will enhance the feasibility and acceptability of death education programs. Methods This mixed-methods feasibility study included a quasi-experimental trial and semi-structured interviews. Older adults in the intervention group (N = 27) received a 6-week KAP-based narrative life education program in addition to standard community health education; participants in the control group (N = 20) received only the normal community health education. In both groups, attitudes toward death and the meaning of life were assessed at baseline and immediately after the intervention. A post-intervention semi-structured interview and satisfaction survey were also conducted for the intervention group. Results Forty out of 47 older adults completed the program for an 85.1% retention rate. All of the older adults in the experiment were very satisfied and satisfied with the life education program, and no adverse events were reported. Compared to the control group, participants in the intervention group had a significant decrease in the fear of death (P = 0.028), and substantial improvement in their value of life (P = 0.031), goal of life (P = 0.035), freedom of life (P = 0.003), and the total score for purpose in life (P = 0.017). The qualitative results yielded four themes: profound recognition of life and death, contradiction between thoughts and action, conflict between one’s acceptance and others’ avoidance, and evaluation of the life education program. Conclusions The KAP-based narrative life education program is feasible and acceptable for older Chinese community-dwelling adults. It is also potentially effective in improving attitudes toward death attitudes and the meaning of life in this cohort. Abstract SARS-CoV-2 infection is initiated by Spike glycoprotein binding to the human angiotensin-converting enzyme 2 (ACE2) receptor via its receptor binding domain. Blocking this interaction has been proven to be an effective approach to inhibit virus infection. Here we report the discovery of a neutralizing nanobody named VHH60, which was directly produced from an engineering nanobody library based on a commercialized nanobody within a very short period. VHH60 competes with human ACE2 to bind the receptor binding domain of the Spike protein at S351, S470-471and S493-494 as determined by structural analysis, with an affinity of 2.56 nM. It inhibits infections of both ancestral SARS-CoV-2 strain and pseudotyped viruses harboring SARS-CoV-2 wildtype, key mutations or variants at the nanomolar level. Furthermore, VHH60 suppressed SARS-CoV-2 infection and propagation 50-fold better and protected mice from death for twice as long as the control group after SARS-CoV-2 nasal infections in vivo. Therefore, VHH60 is not only a powerful nanobody with a promising profile for disease control but also provides evidence for a highly effective and rapid approach to generating therapeutic nanobodies.

Abstract Background Increased intake of specific vitamins has been linked to a decreased prevalence of osteoporosis. However, the association between dietary folate intake and the risk of osteoporosis in the general population remains incompletely understood. Therefore, we aimed to determine the association between dietary folate intake and the risk of osteoporosis in the general population of the USA. Methods In this cross-sectional study, data from the National Health and Nutrition Examination Survey (2017–2020) were collected. Osteoporosis was considered to be indicated by a bone mineral density greater than 2.5 standard deviations below the mean of the young adult reference group. Dietary folate intake was measured by a 24-hour dietary recall. Multivariate logistic regression models and restricted cubic spline models were used. Results The study included 2297 participants (mean age: 63.69 ± 0.35 years), 49.92% of whom were female. In the general population, increased dietary folate intake was directly associated with a decreased risk of osteoporosis (P for trend = 0.005). Abstract Background Photodegradation of plant litter plays a pivotal role in the global carbon (C) cycle. In temperate forest ecosystems, the exposure of plant litter to solar radiation can be significantly altered by changes in autumn phenology and snow cover due to climatic change. How this will affect litter decomposition and nutrient dynamic interacting with forest canopy structure (understorey vs. gaps) is uncertain. In the present study, we conducted a field experiment using leaf litter of early-fall deciduous Betula platyphylla (Asian white birch) and late-fall deciduous Quercus mongolica (Mongolian oak) to explore the effect of change in autumn solar radiation on dynamics of litter decomposition in a gap and understorey of a temperate mixed forest. Results Exposure to the full-spectrum of not only significantly increased the loss of mass, C, and lignin, but also modified N loss through both immobilization and mineralization during the initial decomposition during autumn canopy opening, irrespective of canopy structure and litter species. These effects were mainly driven by the blue-green spectral region of sunlight. Short-term photodegradation by autumn solar radiation had a positive legacy effect on the later decomposition particularly in the forest gap, increasing mass loss by 16% and 19% for Asian white birch and Mongolia oak, respectively. Conclusions Our results suggest that earlier autumn leaf-fall phenology and/or later snow cover due to land-use or climate change would increase the exposure of plant organic matter to solar radiation, and accelerate ecosystem processes, C and nutrient cycling in temperate forest ecosystems. The study provides a reference for predictive research on carbon cycling under the background of global climate change.

Abstract Polyploid is considered an advantage that has evolved to be more environmentally adaptable than its diploid. To understand if doubled chromosome of diploid rice can improve drought tolerance, we evaluated the diploid (2X) and autotetraploid (4X) plants of three indica and three japonica varieties. Drought stress in the plastic bucket of four-leaf stage revealed that the drought tolerance of 4X plants was lower than that of its diploid donor plants. The assay of photosynthetic rate of all varieties showed that all 4X varieties had lower rates than their diploid donors. The capacity for reactive oxygen species production and scavenging varied among different 2X and 4X varieties. Further, transcriptomic analysis of 2X and 4X plants of four varieties under normal and drought condition showed that the wide variation of gene expression was caused by difference of varieties, not by chromosome ploidy. However, weighted gene co-expression network analysis (WGCNA) revealed that the severe interference of photosynthesis-related genes in tetraploid plants under drought stress is the primary reason for the decrease of drought tolerance in autotetraploid lines. Consistently, new transcripts analysis in autotetraploid revealed that the gene transcription related with mitochondrion and plastid of cell component was influenced most significantly. The results indicated that chromosome doubling of diploid rice weakened their drought tolerance, primarily due to disorder of photosynthesis-related genes in tetraploid plants under drought stress. Maintain tetraploid drought tolerance through chromosome doubling breeding in rice needs to start with the selection of parental varieties and more efforts.

Abstract Background YTHDC1, a key m(6)A nuclear reader, plays a crucial role in regulating mRNA splicing, export, and stability. However, the functional significance and regulatory mechanisms of YTHDC1 in inflammatory bowel disease (IBD) remain to be explored. Methods We established a dextran sulfate sodium (DSS)-induced murine colitis model in vivo and LPS/IFN-γ-stimulated macrophage inflammation in vitro. The expression of YTHDC1 was determined. Colocalization of YTHDC1 and macrophages was assayed by immunofluorescence staining. LV-YTHDC1 or shYTHDC1 lentiviruses were applied for YTHDC1 overexpression or inhibition. For NF-κB inhibition, JSH-23 was utilized. The interaction of YTHDC1 and Beclin1 mRNA was determined by RIP, and the m6A modification of Beclin1 was confirmed by MeRIP. Results In DSS-induced colitis and LPS/IFN-γ-treated RAW264.7 macrophages, we observed a significant downregulation of YTHDC1. Overexpression of YTHDC1 resulted in decreased levels of iNOS, CD86, and IL-6 mRNA, along with inhibited NF-κB activation in LPS/IFN-γ-treated RAW264.7 cells. Conversely, downregulation of YTHDC1 promoted iNOS expression and inhibited autophagy. Additionally, the effect of YTHDC1 knockdown on CD86 and IL-6 mRNA induced by LPS/IFN-γ was abolished by the NF-κB inhibitor JSH-23. Mechanistically, YTHDC1 interacted with Beclin1 mRNA, thereby stabilizing Beclin1 mRNA and enhancing Beclin1 expression and autophagy. These effects ultimately led to the inhibition of NF-κB signaling in LPS/IFN-γ-challenged macrophages. Conclusions YTHDC1 inhibited the macrophage-mediated inflammatory response by stabilizing Beclin1 mRNA, which may be a potential therapeutic target for the treatment of IBD.

Abstract Meat intake, particularly from oily fish, has been associated with various chronic diseases. However, its relationship with acne has always been controversial. Therefore, we have adopted Mendelian randomization (MR) analysis to investigate the causal relationship between different types of meat intake and acne. The exposure and outcome datasets for this study were obtained from the Integrative Epidemiology Unit (IEU) Open GWAS project. Seven datasets on meat intake were included, which consisted of non‐oily fish, oily fish, lamb/mutton, poultry, pork, beef, and processed meat. The main methods used for MR analysis were inverse variance weighted, weighted median, and MR‐egger. To ensure the accuracy of the results, heterogeneity, pleiotropy, and Mendelian randomization pleiotropy residual sum and outlier (MR‐PRESSO) analyses were conducted. Additionally, an analysis of four risk factors (fasting insulin, insulin resistance, total testosterone level, and estradiol level) was performed to investigate the underlying mechanisms linking statistically significant meat intake to acne. Abstract Background Limited evidence exists regarding the link between platelet count and 30-day in-hospital mortality in acute respiratory failure (ARF) patients. Thus, this study aims to investigate this association among ICU patients experiencing acute respiratory failure. Methods We conducted a retrospective cohort study across multiple centers, utilizing data from the US eICU-CRD v2.0 database covering 22,262 patients with ARF in the ICU from 2014 to 2015. Our aim was to investigate the correlation between platelet count and 30-day in-hospital mortality using binary logistic regression, subgroup analyses, and smooth curve fitting. Results The 30-day in-hospital mortality rate was 19.73% (4393 out of 22,262), with a median platelet count of 213 × 109/L. After adjusting for covariates, our analysis revealed an inverse association between platelet count and 30-day in-hospital mortality (OR = 0.99, 95% CI 0.99, 0.99). Subgroup analyses supported the robustness of these findings. Furthermore, a nonlinear relationship was identified between platelet count and 30-day in-hospital mortality, with the inflection point at 120 × 109/L. Below the inflection point, the effect size (OR) was 0.89 (0.87, 0.91), indicating a significant association. However, beyond this point, the relationship was not statistically significant. Conclusion This study establishes a clear negative association between platelet count and 30-day in-hospital mortality among ICU patients with ARF. Furthermore, we have identified a nonlinear relationship with saturation effects, indicating that among ICU patients with acute respiratory failure, the lowest 30-day in-hospital mortality rate occurs when the baseline platelet count is approximately 120 × 109/L.

Abstract Background Chronic kidney disease (CKD) carries a high public health burden yet little is known about the relationship between metalworking fluid (MWF) aerosols, occupational noise and CKD. We aimed to explore the relationship between occupational MWF aerosols, occupational noise and CKD. Methods A total of 2,738 machinists were sampled from three machining companies in Wuxi, China, in 2022. We used the National Institute for Occupational Safety and Health (NIOSH) method 5524 to collect individual samples for MWF aerosols exposure, and the Chinese national standard (GBZ/T 189.8–2007) method to test individual occupational noise exposure. Glucagon-like peptide-1 receptor agonists (GLP-1 RAs) protect against diabetic cardiovascular diseases and nephropathy. However, their activity in diabetic retinopathy (DR) remains unclear. Our retrospective cohort study involving 1626 T2DM patients revealed superior efficacy of GLP-1 RAs in controlling DR compared to other glucose-lowering medications, suggesting their advantage in DR treatment. By single-cell RNA-sequencing analysis and immunostaining, we observed a high expression of GLP-1R in retinal endothelial cells, which was down-regulated under diabetic conditions. Treatment of GLP-1 RAs significantly restored the receptor expression, resulting in an improvement in retinal degeneration, vascular tortuosity, avascular vessels, and vascular integrity in diabetic mice. GO and GSEA analyses further implicated enhanced mitochondrial gene translation and mitochondrial functions by GLP-1 RAs. Additionally, the treatment attenuated STING signaling activation in retinal endothelial cells, which is typically activated by leaked mitochondrial DNA. Expression of STING mRNA was positively correlated to the levels of angiogenic and inflammatory factors in the endothelial cells of human fibrovascular membranes. Further investigation revealed that the cAMP-responsive element binding protein played a role in the GLP-1R signaling pathway on suppression of STING signaling. This study demonstrates a novel role of GLP-1 RAs in the protection of diabetic retinal vasculature by inhibiting STING-elicited inflammatory signals.

In the present work, the type, morphology and size of secondary phase particles in a Mg–4Al–2Sn-0.5Ca (ATX420) alloy are tailored through sub-rapid solidification (SRS) and aging treatment before friction stir processing (FSP) to systematically investigate their impact on microstructures and mechanical properties of the FSP ATX420 Mg alloy. The smaller eutectic CaMgSn phase with micro-defects in the as-cast SRS alloy is more likely to be broken during FSP, compared to the as-cast conventional solidification (CS) alloy. Additionally, nano-sized CaMgSn and Mg17Al12 particles precipitate in as-aged SRS alloy due to a high supersaturation solubility of solutes in the as-cast SRS alloy. These densely-distributed nano-sized precipitates in the as-aged SRS alloy increase the processing temperature by 60 °C during FSP, which results in less nano-sized precipitates in the stirring zone and grain growth of the α-Mg matrix. This leads to a decrease in hardness in the FSP-aged-SRS alloy (∼10 HV). Besides, the grain structure evolutions in the stirring zone and transition zone are systematically investigated for understanding the microstructure evolution during FSP in Mg alloys with eutectic phase.

The influence of B and Sb on the microstructure and corrosion behavior of T6-treated Al–Si–Mg alloys has been specifically studied. It is found that the addition of 0.03 wt% B to Al–Si–Mg alloys results in the lowest corrosion rate (0.02451 mm/y) when immersed in 3.5 wt% NaCl solution for 3 days. This improvement can be attributed to the transformation of coarse dendritic α-Al grains into refined equiaxed grains, which is beneficial for the formation of uniform and compact passive film. In contrast, the Al–Si–Mg alloy with addition of 0.2 wt% Sb shows the highest corrosion rate (0.05795 mm/y), which can be attributed to two reasons. First, the addition of Sb contributes to the transformation of rod-like eutectic Si particles into a spherical morphology, increasing the number of micro-galvanic corrosion sites and thus accelerating the corrosion process. Second, the generated Mg3Sb2 phase, being more noble than the eutectic Si phase, strengthens the micro-galvanic corrosion.

Abstract Background Functional endoscopic sinus surgery is a principal option for treating chronic rhinosinusitis with nasal polyps (CRSwNP) after medication failures. Unfortunately, some patients still have unsatisfactory postoperative recovery. The type of inflammatory cell infiltration in nasal polyp tissue has been reported available for recurrence prediction. As it is invasive and time-consuming, this technique is hard to promote clinically under the existing technical conditions. And during the course of clinical treatment, we have noted that differences in the postoperative recurrence rate of patients present among different traditional Chinese medicine syndrome types. Methods and analysis This is a non-randomized, single-center, and prospective cohort study started in Chengdu Sichuan Province, People’s Republic of China, in January 2021. A total of 200 participants will be recruited from patients who are diagnosed with CRSwNP and prepared for functional endoscopic sinus surgery. We collect preoperative data which includes general information, medical history, TCM syndromes, visual analogue scale (VAS) of subjective symptoms, Lund-Kennedy endoscopic score, and Lund-Mackay score of computed tomography (CT) scanning of sinuses. We acquire the VAS score and Lund-Kennedy score of subjective symptoms through multiple planned follow-up after surgery. After 1 year of follow-up, the recurrence rate will be calculated, and the curative effect will be assessed. Meanwhile, the patients’ pathological sections will be sorted out, and inflammatory cell infiltration will be analyzed. Statistical analysis will be carried out to evaluate the correlation among CRSwNP recurrence and TCM syndrome types and tissue inflammatory cell infiltration types. Then we will establish a predictive model for CRSwNP recurrence. Analyses of survey data include descriptive and inferential statistical approaches. Discussion This is the first prospective cohort study on investigating the correlation of CRSwNP recurrence with TCM syndrome types and tissue inflammatory cell infiltration types. Through this study, we hope to discover a new and simple, effective, and noninvasive way to predict the recurrence rate rapidly after CRSwNP and provide reference for the intervention timing of traditional Chinese medicine application, thereby achieving customized diagnosis and treatment, minimizing risks of surgical events, and delaying postoperative recurrence of CRSwNP. Abstract Previous studies through targeted mutagenesis of K-D-K-E motif have demonstrated that 2′-O-MTase activity is essential for efficient viral replication and immune evasion. However, the K-D-K-E catalytic motif of 2′-O-MTase is highly conserved across numerous viruses, including flaviviruses, vaccinia viruses, coronaviruses, and extends even to mammals. Here, we observed a stronger 2′-O-MTase activity in SARS-CoV-2 compared to SARS-CoV, despite the presence of a consistently active catalytic center. We further identified critical residues (Leu-36, Asn-138 and Ile-153) which served as determinants of discrepancy in 2′-O-MTase activity between SARS-CoV-2 and SARS-CoV. These residues significantly enhanced the RNA binding affinity of 2′-O-MTase and boosted its versatility toward RNA substrates. Of interest, a triple substitution (Leu36 → Ile36, Asn138 → His138, Ile153 → Leu153, from SARS-CoV-2 to SARS-CoV) within nsp16 resulted in a proportional reduction in viral 2′-O-methylation and impaired viral replication. Furthermore, it led to a significant upregulation of type I interferon (IFN-I) and proinflammatory cytokines both in vitro and vivo, relying on the cooperative sensing of melanoma differentiation-associated protein 5 (MDA5) and laboratory of genetics and physiology 2 (LGP2). In conclusion, our findings demonstrated that alterations in residues other than K-D-K-E of 2′-O-MTase may affect viral replication and subsequently influence pathogenesis. Monitoring changes in nsp16 residues is crucial as it may aid in identifying and assessing future alteration in viral pathogenicity resulting from natural mutations occurring in nsp16.

Abstract Fibrosis is a reparative and progressive process characterized by abnormal extracellular matrix deposition, contributing to organ dysfunction in chronic diseases. The tumor suppressor p53 (p53), known for its regulatory roles in cell proliferation, apoptosis, aging, and metabolism across diverse tissues, appears to play a pivotal role in aggravating biological processes such as epithelial-mesenchymal transition (EMT), cell apoptosis, and cell senescence. These processes are closely intertwined with the pathogenesis of fibrotic disease. In this review, we briefly introduce the background and specific mechanism of p53, investigate the pathogenesis of fibrosis, and further discuss p53’s relationship and role in fibrosis affecting the kidney, liver, lung, and heart. In summary, targeting p53 represents a promising and innovative therapeutic approach for the prevention and treatment of organ fibrosis.

The sensory quality and metabolite profiles of the fresh and dried buds and leaves of Lycium from 26 cultivars (lines) were analyzed by sensory evaluation and metabolomics combined with multivariate statistical analysis. The results of sensory evaluation showed that the dried buds and leaves of Lycium had a mellow, fresh and sweet taste, and by liquid chromatography-mass spectrometry (LC-MS), 879 metabolites were identified in Lycium buds and leaves, including amino acids and their derivatives, flavonoids, phenolic acids, saccharides, nucleotides and their derivatives, lignans and coumarins, alkaloids, terpenoids, organic acids, and lipids. Using partial least square discriminant analysis (PLS-DA) model and one-way analysis of variance (ANOVA), 211 differential metabolites were selected, which were mainly enriched in the linoleic acid metabolism, flavonoid biosynthesis, flavonoids and flavanols biosynthesis, phenylpropanol biosynthesis, and galactose metabolism pathways. In addition, it was found that amino acids and their derivatives, flavonoids, sugars, and lipids were important components to discriminate the metabolites of different cultivars (lines). Partial least squares regression (PLSR) analysis showed that 26 metabolites including epicatechin, isoquercitrin, and rutin collectively affected the taste of Lycium tea such as mellow, thick, fresh, sweet and bitter. The results of sensory evaluation and PLSR analysis showed that seven cultivars (lines) of Lycium including ‘Ningqicai No. 1’, ‘Ningqi No. 9’, Z90, Z68, Q3, Q6-13, and Z48 had better sensory quality and rich flavor substances.
