- Abstract
Marine biofouling is often associated with biofilm formation on submerged surfaces, which causes economic and environmental problems. Although several biocides and cleaners have been used to control biofouling, new eco‐friendly antifouling approaches are required. Chitosan (CS) is a polysaccharide widely used due to its outstanding properties, namely its antimicrobial activity. This work aimed to produce and characterise poly(lactic acid) (PLA)‐CS surfaces with CS of different molecular weight (Mw) at different concentrations for application in marine paints. The antimicrobial performance of these surfaces was assessed against Cobetia marina for 7 weeks under controlled hydrodynamic conditions. All CS surfaces exerted a significant antimicrobial action by reducing the number of culturable cells up to 69% compared to control, being this activity dependent on CS Mw. Optical Coherence Tomography analysis corroborated these results, showing reductions in biofilm thickness of up to 36%. Overall, CS coatings represent a promising approach to reducing biofouling in marine environments

- Abstract

The present work of professional experience took place in an industrial environment, describing step by step the quality control procedures of BOPP (Biaxially Oriented PolyPropylene) film in the company Poligal, supplier of BOPP, CPP (cast polypropylene) and metallized films for different application for the entire globe. The BOPP film production process consists of a biaxial plane stretching, that is, the film is oriented/stretched in the longitudinal and transverse directions. This is a sequential process that starts with the raw material, granulated polypropylene, which undergoes multiple stretching, heating and cooling throughout the sequence, ending with a polypropylene film wound with all the characteristics that BOPP film must present. This process is divided into different sections: silos, extrusion, chill roll, MDO (Machine Direction Orientation), TDO (Transverse Direction Orientation), pull roll and winder, in which the film undergoes changes in each segment, approaching the final product. This dissertation has as main object the characterization of a commercial BOPP film according to Poligal's quality control procedures. After characterizing the commercial film with different tests, the results obtained were compiled in order to identify the type of BOPP film, according to the films that the company has, and finally the specifications of the technical data sheet were compared (TDS, technical data sheet) for the selected film, with the results obtained from the commercial film. Finally, it was possible to conclude that the commercial BOPP film has the characteristics of CATE 30 SOL flexible packaging films, in which for procedures: thickness, treatment level, mechanical properties, shrinkage, resistance to thermal sealing, temperature of beginning of sealing and friction coefficient, the results are consistent with those specified in the TDS, except for the opacity/haze and gloss tests. For these properties, the TDS specification is for virgin/non-handled plastic, and the commercial film, in addition to being handled, was already found as an internal packaging involving “Maria cookie” from the Continente brand.

- Abstract
Marine biofouling is a natural process often associated with biofilm formation on submerged surfaces, creating a massive economic and ecological burden. Although several antifouling paints have been used to prevent biofouling, growing ecological concerns emphasize the need to develop new and environmentally friendly antifouling approaches such as bio-based coatings. Chitosan (CS) is a natural polymer that has been widely used due to its outstanding biological properties, including non-toxicity and antimicrobial activity. This work aims to produce and characterize poly (lactic acid) (PLA)-CS surfaces with CS of different molecular weight (Mw) at different concentrations for application in marine paints. Loligo opalescens pens, a waste from the fishery industry, were used as a CS source. The antimicrobial activity of the CS and CS-functionalized surfaces was assessed against Cobetia marina, a model proteobacterium for marine biofouling. Results demonstrate that CS targets the bacterial cell membrane, and PLA-CS surfaces were able to reduce the number of culturable cells up to 68% compared to control, with this activity dependent on CS Mw. The antifouling performance was corroborated by Optical Coherence Tomography since PLA-CS surfaces reduced the biofilm thickness by up to 36%, as well as the percentage and size of biofilm empty spaces. Overall, CS coatings showed to be a promising approach to reducing biofouling in marine environments mimicked in this work, contributing to the valorization of fishing waste and encouraging further research on this topic.

- Abstract
Leishmaniases have a broad spectrum of clinical manifestations, ranging from a cutaneous to a progressive and fatal visceral disease. Chemotherapy is nowadays the almost exclusive way to fight the disease but limited by its scarce therapeutic arsenal, on its own compromised by adverse side effects and clinical resistance. Cyclobenzaprine (CBP), an FDA-approved oral muscle relaxant drug has previously demonstrated in vitro and in vivo activity against Leishmania sp., but its targets were not fully unveiled. This study aimed to define the role of energy metabolism as a target for the leishmanicidal mechanisms of CBP. Methodology to assess CBP leishmanicidal mechanism variation of intracellular ATP levels using living Leishmania transfected with a cytoplasmic luciferase. Induction of plasma membrane permeability by assessing depolarization with DiSBAC(2)3 and entrance of the vital dye SYTOX® Green. Mitochondrial depolarization by rhodamine 123 accumulation. Mapping target site within the respiratory chain by oxygen consumption rate. Reactive oxygen species (ROS) production using MitoSOX. Morphological changes by transmission electron microscopy. CBP caused on L. infantum promastigotes a decrease of intracellular ATP levels, with irreversible depolarization of plasma membrane, the collapse of the mitochondrial electrochemical potential, mild uncoupling of the respiratory chain, and ROS production, with ensuing intracellular Ca2+ imbalance and DNA fragmentation. Electron microscopy supported autophagic features but not a massive plasma membrane disruption. The severe and irreversible mitochondrial damage induced by CBP endorsed the bioenergetics metabolism as a relevant target within the lethal programme induced by CBP in Leishmania. This, together with the mild-side effects of this oral drug, endorses CBP as an appealing novel candidate as a leishmanicidal drug under a drug repurposing strategy.

- Abstract
The growth location and plant variety may influence the active components and biological activities of plants used in phytomedicine. In this study, nine sets of different Epimedii Folium, from different representative cultivation locations and Epimedium species, were collected for comparison, using HPLC-DAD combined with multivariate analysis. The objective was to investigate the influence of geographical origin and Epimedium species on the quality of Epimedii Folium, and provide applicable guidance for cultivation and quality control of Epimedii Folium. Several Epimedium spp. sets were used to establish the HPLC-DAD fingerprints and 91 peaks (compounds) were selected for the multivariate analysis. Major compounds were analyzed by HPLC-DAD combined with principal component analysis (PCA). HPLC quantitative analysis of known bioactive compounds was performed. Application of PCA to HPLC data showed that Epimedium samples sharing the same geographical origin or species clustered together, indicating that both species and geographical origin have impacts on the quality of Epimedii Folium. The major bioactive flavonoid compounds, epimedin C, icariin and baohuoside I, were identified and quantified. The concentration of bioactive compounds was significantly influenced both by species and geographical origin. E. sagittatum from Sichuan showed the highest content of bioactive compounds. The results showed that both Epimedium species and geographical origin have strong impact into quality of Epimedii Folium. HPLC data combined with multivariate analysis is a suitable approach to inform the selection of cultivation areas and choose Epimedium spp. most suitable for different geographical areas, resulting in improved quality of Epimedii Folium.

- Abstract
Implantable medical devices (IMDs) are susceptible to microbial adhesion and biofilm formation, which lead to several clinical complications, including the occurrence of implant-associated infections. Polylactic acid (PLA) and its composites are currently used for the construction of IMDs. In addition, chitosan (CS) is a natural polymer that has been widely used in the medical field due to its antimicrobial and antibiofilm properties, which can be dependent on molecular weight (Mw). The present study aims to evaluate the performance of CS-based surfaces of different Mw to inhibit bacterial biofilm formation. For this purpose, CS-based surfaces were produced by dip-coating and the presence of CS and its derivatives onto PLA films, as well surface homogeneity were confirmed by contact angle measurements, Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM). The antimicrobial activity of the functionalized surfaces was evaluated against single- and dual-species biofilms of Staphylococcus aureus and Pseudomonas aeruginosa. Chitosan-based surfaces were able to inhibit the development of single- and dual-species biofilms by reducing the number of total, viable, culturable, and viable but nonculturable cells up to 79%, 90%, 81%, and 96%, respectively, being their activity dependent on chitosan Mw. The effect of CS-based surfaces on the inhibition of biofilm formation was corroborated by biofilm structure analysis using confocal laser scanning microscopy (CLSM), which revealed a decrease in the biovolume and thickness of the biofilm formed on CS-based surfaces compared to PLA. Overall, these results support the potential of low Mw CS for coating polymeric devices such as IMDs where the two bacteria tested are common colonizers and reduce their biofilm formation.

- Abstract
Drug repurposing affords the implementation of new treatments at a moderate cost and under a faster time-scale. Most of the clinical drugs against Leishmania share this origin. The antidepressant sertraline has been successfully assayed in a murine model of visceral leishmaniasis. Nevertheless, sertraline targets in Leishmania were poorly defined. In order to get a detailed insight into the leishmanicidal mechanism of sertraline on Leishmania infantum, unbiased multiplatform metabolomics and transmission electron microscopy were combined with a focused insight into the sertraline effects on bioenergetics metabolism of the parasite. Sertraline induced respiration uncoupling, a significant decrease of intracellular ATP level, and oxidative stress in L. infantum promastigotes. Metabolomics evidenced an extended metabolic disarray caused by sertraline. This encompasses a remarkable variation of the levels of thiol-redox and polyamine biosynthetic intermediates, as well as shortage of intracellular amino acids used as metabolic fuel by Leishmania Sertraline killed Leishmania through a multitarget mechanism of action, tackling essential metabolic pathways of the parasite. As such, sertraline is a valuable candidate for visceral leishmaniasis treatment under a drug repurposing strategy.

Fundação de Apoio a Pesquisa e à Inovação Tecnológica do Estado de Sergipe - FAPITEC/SE The purpose of this study is to analyze the presence of The Council of the Gods from Os Lusíadas in three adapted versions of Camões’ epic in prose and destined to children and juvenile public: one was written by Rubem Braga and Edson Rocha Braga; one by Luiz Maria Veiga; and one by Ricardo Vale, with the main objective of discussing the permanence or not of epic peculiarities from Os Lusíadas in the book adaptations in question through the recognition of the forms of retextualization used by the authors. It is also intended to measure the influence of adaptations of the epic genre on literary reading practices in childhood and adolescence. Firstly, the reflection will address the issue of literary reading in school, seeking to understand the "to be or not to be" of adapted literature. To do so, we will analyze how the book adaptations approached the selected episode of Os Lusíadas, taking into account both the aesthetic aspects and the resizing of content, with special emphasis on the presence of Roman gods in Camões’ epic. The analysis will be based on the detailed observation of the original work, in order to verify, in the adaptations, the processes of reduction of the original plot and the possible creative interferences, relating them to different forms of retextualization, based on the theoretical and critical contributions of Marilise Rezende Bertin (2008), Maria Alice Gonçalves Antunes (2007) and Dell'Isola (2007). Considerations about the epic genre, in turn, will be supported by Anazildo Vasconcelos da Silva's Teoria Épica do Discurso, in Christina Ramalho's Poemas Épicos: estratégias de leitura (2013), and also Leo Pollmann (1973) and his formulations about the epic and the Camonian epic. The following contributions are also noteworthy: Maria do Rosário Mortatti Magnani (2001), who offers a reflection on the formation of literary taste; Silvério Benedito (1997), who elaborates critical considerations about Camões epic; and José Cardoso Bernardes and Rui Afonso Mateus (2013), in their work Literatura e ensino do português. With this approach, we intend to contribute to the formation of a critical corpus on the phenomenon of literary adaptation and to measure it as one of the main segments of the mythological plan of Os Lusíadas makes sense in prose works of children's and juvenile’s literature. OBJECTIVES: Currently, the intake of fruits and vegetables is lower than the recommended amounts, despite the known association between consumption of plant foods and prevention of several chronic diseases. Grapes are a widely accepted and consumed fruit, and one of the richest in phytochemicals that are beneficial to human health. Previously, we reported that growing cold hardy table grapes on two different grapevine training systems, Modified Munson (MM) and Vertical Shoot Positioning (VSP), modulates their amount of total phenolics and antioxidant potential at harvest. However, consumers access to fruit is usually not immediate upon harvest, but could take weeks/months before reaching stores. The objective of this study was to evaluate whether phenolics and antioxidant potential of table grapes grown on different training systems could be maintained post-harvest in cold storage. METHODS: Cold-hardy table grapes variety Mars were grown on two grapevine training systems, MM and VSP. At harvest, randomly picked grape clusters from each training system were placed in clamshells (2 clusters per clamshell) and placed in cold storage (0 ± 1°C). After 1, 2 and 3 weeks post-harvest, grapes (triplicate clamshells) were homogenized and Brix, titratable acidity, antioxidant potential and total phenolics were analyzed. Statistical analysis was performed using 2-way ANOVA. RESULTS: Total phenolics were greater for MM compared with VSP (MM 2.56 vs. VSP 1.84 mM Catechin equivalents; P = 0.0001), but were reduced in cold storage irrespective of training system (P = 0.0002). The MM training system had greater Brix (MM 17.9 vs. VSP 15.8°Bx; P

This report is the result of a professional traineeship in the scope of the Master in Educational – Mediation and Supervision in Training, University of Minho. Outlined by Social and Community Mediation, the project is developed in the Social and Cultural Centre of S. Pedro de Bairro, with the target audience being the users of the social response of SAAS (Service of Social Attendance and Monitoring), aiming to improve the quality of life and the interpersonal relationships of the users of SAAS, by developing an intervention directed towards autonomy and civic participation in a perspective of active citizenship. This is a study whose aim goes beyond the improvement of interpersonal relationships, from a creative and transformative prevention action, capable of working out the communicative processes, developing interpersonal relationship competences and promoting positive conflict management. It is our belief that mediation comprises an alternative process that far from exhausts conflict resolution, but rather highlights its educational and transformative potential. From a perspective of preventive and transformative mediation, it is possible to create the conditions for an inclusive environment in interpersonal relationships through empowerment and awareness. The importance of mediation in this perspective should be emphasized, when we consider the need for a culture of pacification and engagement as active citizens, constituting itself as a process of enhancement and social cohesion. The study presented here is based on a qualitative approach, focusing on research-intervention as a methodology for an understanding of the complexity implicit in contexts of social vulnerability and acting with a transformation purpose. It is characterized by a close connection between theory and practice, applying processes of observation and reflection recording, enhancing professional development, provided by the spiral model of research-intervention. The intervention plan invoked two strategic vectors of action, pointing to the consolidation of a differentiated professional practice in the interpersonal and social-community nuclei, based on a communicational device. The results of this study reveal a significant contribution to a differentiating and transformative intervention through mediation, which responds to the objectives of the Local Social Intervention Network. To mediate action in these and these social responses to the community, with the mission of exploring and promoting active participation, democratic citizenship and social cohesion, assuming mediation as praxis, structuring practice and model of social regulation.
