Background: In this experimental study, we aimed to investigate the effects of hesperetin, a natural flavonoid, on a lipopolysaccharideinduced acute lung injury model in rats. Methods: Between March 2019 and May 2019, a total of 18 adult male Wistar albino rats, weighing approximately 250 to 300 g, were randomly divided into three groups as control, lipopolysaccharide, and lipopolysaccharide + hesperetin groups (n=6 in each group). The wet/dry weight ratio of lung tissue was determined. Histopathological changes were examined using light and scanning electron microscopy. Pulmonary nuclear factor-kappa beta, inducible nitric oxide synthase, and alpha-smooth muscle antigen activity were determined with indirect immunohistochemical methods. Pulmonary apoptosis was detected with the terminal deoxynucleotidyl transferase dUTP nick-end labeling method. Tumor necrosis factor-alpha, interleukin-1 beta, interleukin-6, and interleukin-10 concentrations were measured with enzyme-linked immunosorbent assay. Results: Treatment with hesperetin significantly improved the architecture of lung tissue and reduced the wet/dry weight ratio, nuclear factor-kappa beta, inducible nitric oxide synthase, and alphasmooth muscle antigen expression, pulmonary apoptosis, and levels of proinflammatory cytokines. Conclusion: Our study results suggest that hesperetin has a potent protective effect against lipopolysaccharide-induced acute lung injury in rats via suppression of the proinflammatory cytokine cascade, nuclear factor-kappa beta, signaling pathway activation, and apoptosis., Amaç: Bu deneysel çalışmada, doğal bir flavonoid olan hesperetinin sıçanlarda lipopolisakkarit uyarımlı akut akciğer hasar modeli üzerindeki etkileri incelendi. Ça¬lış¬ma pla¬nı: Mart 2019 - Mayıs 2019 tarihleri arasında ağırlıkları yaklaşık 250-300 g olan toplam 18 erişkin erkek Wistar albino sıçan, kontrol, lipopolisakkarit ve lipopolisakkarit + hesperetin grubu olmak üzere rastgele üç gruba ayrıldı (her grupta n=6). Akciğer dokusunda ıslak/kuru ağırlık oranı belirlendi. Histopatolojik değişiklikler ışık ve elektron mikroskobu ile incelendi. Pulmoner nükleer faktör-kappa beta, indüklenebilir nitrik oksit sentaz ve alfa-düz kas antijeni indirekt immünohistokimyasal yöntem ile belirlendi. Pulmoner apoptoz, terminal deoksinükleotidil transferaz dUTP çentik uç işaretleme yöntemi ile belirlendi. Tümör nekroz faktör-alfa, interlökin-1 beta, interlökin-6 ve interlökin-10 konsantrasyonları enzim bağlı immunosorbent testi ile belirlendi. Bulgular: Hesperetin tedavisi akciğer doku mimarisini anlamlı düzeyde iyileştirdi ve ıslak/kuru ağırlık oranı, nükleer faktör-kappa beta, indüklenebilir nitrik oksit sentaz ve alfa-düz kas antijeni ekspresyonu, pulmoner apoptoz ve proinflamatuvar sitokin düzeylerini azalttı. So¬nuç: Çalışma sonuçlarımız, hesperetinin sıçanlarda proinflamatuvar sitokin kaskadı, nükleer faktör-kappa beta sinyal yolağı aktivasyonu ve apoptozun baskılanması ile lipopolisakkarit uyarımlı akut akciğer hasarına karşı güçlü bir koruyucu etkiye sahip olduğunu göstermektedir.

Bu tez çalışmasında, sıçanlara LPS (Serotip E.coli: O111:B4; 250 µg/kg, ip) uygulanarak hipotermi oluşturduk. Oluşturulan hipoterminin girişinde, en derin noktasında ve çıkışta intrakardiyak delinme ile kan örnekleri alındı. Yine aynı çalışmada kontrol grubu sıçanlara tuzlu su (% 0.09'luk NaCl, 0.5 ml/kg ip) uygulanarak hipotermi giriş, en derin noktasına ve çıkışına dek gelen zaman dilimlerinde kan örnekleri alındı. Uygulamalar sonucunda hipoterminin en derin (ΔT: -1.48±0.09 ºC) noktasına ortalama 104.46±4.32 dakikada ulaşıldı. Alınan kan serumu örneklerinin analizleri sonucunda aldosteron, Na, K, Cl, Ca ve P düzeyleri kontrol grubuna göre LPS indüklü hipotermi grupları (giriş, dip ve çıkış) arasındaki farklar istatiksel olarak önemsiz olduğu belirlenmiştir. Elde etiğimiz sonuçlar, rodentlerde LPS ile termoregülatuvar anlamda ateşle birlikte hipotermininde gelişebileceği göstermiştir. Bu çalışmadaki LPS dozunun endotoksik etkileşim süresinin aldosteron seviyesini değiştirmediğini göstermiştir. Yine bu araştırma sonucunda elde edilen Na, K, Cl, Ca ve P düzeylerinin değişmemesinin LPS indüklü hipotermi süresince aldosteron hormon düzeylerinin değişmemesi sonucu olduğunu düşündürdü., In this thesis study, hypothermia was induced by administration LPS (Serotype E. coli: O111: B4; 250 μg / kg, ip) to rats. Blood samples were taken by intracardiac puncture at the initial phase, nadir and the end of the created hypothermia. In the same study, blood samples were taken at the time of hypothermia the initial phase, nadir and the end to the control group rats by saline (0.09% NaCl, 0.5 ml / kg ip). As a result of the applications, the nadir (ΔT: -1.48 ± 0.09 ºC) point of the hypothermia was reached at an average of 104.46 ± 4.32 minutes. The differences between the groups of LPS-induced hypothermia (initial phase, nadir and end) were statistically insignificant compared to the control group of aldosterone, Na, K, Cl, Ca and P levels as a result of analysis of the blood serum samples. Our results show that in rodents, thermoregulator with LPS may develop in hypothermia with fever. The LPS dose in this study did not change the level of aldosterone during the endotoxic interaction period. Again, the results of this study suggested that the changes of Na, K, Cl, Ca and P levels were the result of not changing the aldosterone hormone levels during LPS-induced hypothermia.

Amaç: Hücrelerin LPS (Lipopolisakkarit)’ye enflamatuvaryanıtları in-vitro deneylerde sıklıkla kullanılmaktadır. Buna rağmen hücre kültürü çalışmaları için LPS ’nin doğru dozunun belirlenmesinde zorluklarla karşılaşılmaktadır. Maksimum yanıt veren LPS konsantrasyonunun belirlenmesi in vitro enflamatuvar deneyler için kritik bir noktadır. Bu çalışmanın amacı LPS’nin 293T insan renal hücre dizisindeki konsantrasyon bağımlı etkisinin yeniden değerlendirilmesidir.Yöntem: Bu çalışmada LPS ile stimule edilmiş 293T hücre hattı Xcelligence Real-Time Cell Analyzer (RTCA) sisteminde incelendi. LPS ’nin artmış konsantrasyonlarının 293T hücre hattı üzerindeki etkisi RTCA cihazında hücre proliferasyonu ölçümleri yapılarak araştırıldı.Bulgular: Sonuçlarımıza göre 2, 4 ve 8 μg’lık LPS konsantrasyonlarını hücre bölünmesini inhibe ederek hücreleri kararlı duruma yönlendirdiğini gösterdi. 1 μg ’lık LPS konsantrasyonu hücreleri mitotik faza sürüklerken daha düşük LPS konsantrasyonlarının ise hücre proliferasyonu üzerinde bir etki göstermedi.Sonuç: Bu çalışmadaki sonuçlara göre LPS konsantrasyonlarının hücre proliferasyonu üzerinde farklı etkilerinin olduğu ve deneysel çalışmalardan önce LPS konsantrasyonlarının optimize edilmesi gerektiği gösterilmiştir., Objective: The inflammatory responsiveness of the cells to Lipopolysaccharides (LPS) is commonly used for in vitro experiments. However, the correct dose of the LPS for cell line experiments is elusive. The LPS concentration that gives the maximal response is a critical point of in vitro inflammatory experiments. The aim of this study was to reevaluate the concentration dependent effect of LPS on the 293T human renal cell line.Methods: We evaluated the cell-detachment assay of LPS-stimulated 293T cell line monitored by xCELLigence Real-Time Cell Analyzer (RTCA) system. We applied increasing concentration of LPS followed by Roche xCELLigence Instrument based on the Real-Time Cell Analysis System.Results:Our results demonstrated that the 2, 4 and 8μg of LPS inhibit cell division which diverts cells to a steady-state phase, 1 μg acts as mitogen. Lower concentrations are no effect on cells.Conclusion:These work showed that LPS concentrations had various effects on cell proliferation and need to be estimated for each experiment before carry out the experiments.

İnflamasyon, fiziksel ve enfeksiyöz etkilerin yanında vücutta otoimmün olarak meydana gelebilen sistemik ya da bölgesel yanıttır. İnflamasyon sırasında oluşan serbest radikaller ve lipid peroksidasyonu ürünleri, genlerde mutasyonlara ve kanser de dahil olmak üzere çoğu inflamatuar hastalıkla ilgili anahtar proteinlerin translasyon sonrası değişiklik geçirmesine neden olmaktadır. Lipopolisakkarit (LPS), Gram-negatif bakterilerin dış zarının bir parçası olup, inflamatuar sitokinlerin, serbest oksijen radikallerinin, nitrik oksit ve araşidonik asit metabolitlerinin aşırı üretimine yol açarak inflamasyon oluşumuna sebep olur. Yüksek flavanoid ve fenolik asit içeriği ile Salvia officinalis (Lamiaceae) serbest radikalleri ortadan kaldırma özelliği sayesinde kuvvetli bir antioksidan ve antiinflamatuardır. Bu çalışmada lipopolisakkaritle oluşturulan deneysel inflamasyon modelinde Salvia officinalis ekstresinin inflamasyon ve antioksidan sistem üzerine olan etkileri araştırılmıştır. Çalışmamızda 42 adet 4-5 aylık Wistar albino ırkı dişi sıçanlar ile 6 deney grubu oluşturulmuştur. Üç gruba intraperitoneal olarak 1 mg/kg LPS (E.coli, serotipe 055-B5) uygulandı. LPS enjeksiyonundan 24 saat sonra tedavi gruplarına 10 mg/kg ve 30 mg/kg Salvia officinalis ekstresi gavaj yoluyla verildi. FDG-PET taraması ile inflamasyon durumunu belirlemek için PET taramasından 1 saat önce ketamin anestezisi altında intrakardiyak olarak 18F-fluoro-deoksi-D-glukoz (FDG) uygulandı. Hayvanlardan kan ve doku örnekleri toplandı. Akciğer ve karaciğer FDG-PET tutulumu hesaplandı. Karaciğer, böbrek, akciğer dokuları ve eritrosit malondialdehit (MDA) seviyeleri, katalaz (KAT), süperoksit dismutaz (SOD) ve glutatyon peroksidaz (GPx) enzim aktiviteleri belirlendi. Çalışma sonucunda, eritrosit hemolizatında, karaciğer, böbrek ve akciğer dokularında MDA seviyeleri inflamasyon grubunda, tedavi gruplarına göre anlamlı derecede yüksek bulundu. İnflamasyon grubu SOD aktiviteleri karaciğer, akciğer dokuları ve hemolizatta Salvia officinalis ile tedavi edilen gruplardan anlamlı düzeyde düşük olarak tespit edildi. Aynı şekilde KAT ve GPx aktivitelerinin de inflamasyon grubunda tedavi grubu enzim aktivitelerine oranla istatistiksel açıdan anlamlı derecede düşük bulunduğu sonuçlar elde edilmiştir. Akciğer ve karaciğer 18F-FDG tutulumunun inflamasyon grubunda, kontrol grubuna göre yüksek olduğu, Salvia officinalis tedavi gruplarında ise normale döndüğü saptandı. Sonuç olarak Salvia officinalis’ in sıçanlarda LPS ile oluşturulan inflamasyonun ve oksidatif stresin etkilerini azalttığı belirlenmiştir. Inflammation is a systematic or a local response that may occur as autoimmune in organism besides physical and infectious effects. Free radicals and lipid peroxidation products produced during inflammation causes harmful mutations and post translational changes of key proteins which involves in many inflammatory disorders including cancer. Lipopolisaccharide (LPS) is a part of the external membrane of gram-negative bacteria that causes inflammation by causing overproduction of inflammatory cytokines, free oxygen radicals, nitric oxide and arachidonic acid metabolites. Salvia officinalis (Lamiaceae) is a powerful antioxidant and anti-inflammatory herb thanks to its feature of removing free radicals with its high flavonoid and phenolic acid content. The aim of this study is to investigate the effects of Salvia officinalis extract on inflammation and antioxidant system in an experimental inflammation model induced by LPS in rats. In our study, 4-5 months old 42 female Wistar albino rats were divided into 6 groups. In three groups, 1 mg/kg LPS (E.coli, serotipe 055-B5) was applied to rats intraperitoenally. After 24 hours from LPS injection 10 mg/kg and 30 mg/kg Salvia officinalis extract was orally administrated to treatment groups. 18F-fluoro- deoxy-D-glucose (18FDG) was applied intracardially under ketamine anesthasia 1 hour before PET scanning in order to determine status of inflammation with FDG-PET scanning. Rats were sacrificed and blood and tissue samples were collected. Lung and liver FDG-PET uptake was calculated. Malondialdehyde (MDA) levels, catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GPx) enzyme activities of liver, kidney, lung tissues and erythrocyte hemolysate were determined. As a result of the study, MDA levels in erythrocyte hemolysate, liver, lung and kidney tissues were found to be significantly higher in inflammation group compared to treatment groups, whereas SOD activities of liver and lung tissues and hemolysate were determined significantly lower in inflammation group compared to group treated with Salvia officinalis. Likewise CAT and GPx activities in inflammation control group were found to be significantly lower compared to treatment groups. Lung and liver 18F-FDG uptake were found to be higher in inflammation group than control group. In treatment groups 18F-FDG uptake was at normal level. In conclusion, Salvia officinalis was found to be efficient in reducing free oxygen radical production and ameliorating inflammation induced by LPS in rats.

Amaç: Lipopolisakkarit ile deneysel akciğer enfeksiyon modeli oluşturulmuş ratların serum/plazmalarında yangı mediatörleri ile organ hasar belirteçlerine veteriner hekimlik alanında kullanılan makrolid antibiyotiklerin etkisini belirlemektir. Gereç ve Yöntem: Toplam 198 adet rat kullanıldı. 6 adet rat 0. saat örnekleme zamanı için ayrıldı. 192 adet SpragueDawley ırkı rat; lipopolisakkarit (Escherichia coli 0111:B4, 0.5 mg/200 μl, intratraheal), lipopolisakkarit + tilozin (10 mg/kg, derialtı), lipopolisakkarit + tilmikosin (20 mg/kg, derialtı) ve lipopolisakkarit + tulatromisin (2.5 mg/kg, derialtı) uygulama grubu olmak üzere 4 eşit gruba ayrıldı. Uygulamalardan sonraki 0.5, 1, 1.5, 2, 4, 6, 12 ve 24. saatlerde genel anestezi altında kalpten kan örnekleri alındı. Serum tümör nekrozis faktör-α, interlökin-1β, interlökin-6 ve interlökin-10 düzeyleri ve plazma 13, 14-dihydro-15- keto-prostaglandin F2α düzeyleri ELISA ile belirlendi. Serum C-reaktif protein, kreatin kinaz-MB, alkalen fosfataz, aspartat aminotransferaz, alanin aminotransferaz, üre, kreatinin, kolesterol ve trigliserit düzeyleri ise otoanalizörde belirlendi. Eğri altında kalan alan (EAA0-24), maksimum konsantrasyon (Cmax) ve maksimum konsantrasyona ulaşma zamanı (tmax) farmakokinetik programla belirlendi. Bulgular: Makrolid antibiyotiklerin serum tümör nekrozis faktör-α ve 13, 14-dihydro-15-keto-prostaglandin F2α düzeylerini artırdığı belirlendi. Öneri: Veteriner sahada kullanılan makrolid antibiyotiklerin savunma sistemi üzerinde baskılayıcı etkileri olmayabileceği belirlendi. Ancak makrolid antibiyotiklerin immun sistem üzerinde etkilerinin net olarak anlaşılabilmesi için özellikle doza bağlı etkilerinin araştırılması gerektiği kanaatine varıldı., Aim: The aim of this study was to determine the effect of macrolide antibiotics using in veterinary area on serum/ plasma inflammatory mediators and organ damage markers in lipopolysaccharide-induced pulmonary damage rats. Materials and Methods: Total 198 Sprague-Dawley rats were used. Six rats were used for a 0th sampling point for all groups. 192 rats were divided into four equal groups as lipopolysaccharide (Escherichia coli 0111:B4, 0.5 mg/200 μl, intratracheal), lipopolysaccharide + tylosin (10 mg/kg, subcutaneously), lipopolysaccharide + tilmicosin (20 mg/ kg, subcutaneously) and lipopolysaccharide + tulathromycin (2.5 mg/kg, subcutaneously). After the treatments, blood samples were collected by cardiac puncture under anesthesia at 0.5, 1, 1.5, 2, 4, 6, 12 and 24 hours. Serum tumor necrosis factor α, interleukin-1β, interleukin-6, interleukin-10 levels, and plasma 13, 14-dihydro-15-ketoprostaglandin F2α levels were determined by ELISA, while serum C-reactive protein, creatine kinase-MB, aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, cholesterol, triglyceride, blood urea nitrogen and creatinine levels were determined by otoanalyzer. Area under the concentration time curve (AUC0-24), maximal concentration in plasma or serum (Cmax) and time to reach Cmax (tmax) values were determined by pharmacokinetic computer program. Results: Macrolide antibiotics increased serum tumor necrosis factor α and plasma 13, 14-dihydro-15-keto-prostaglandin F2α levels. Conclusion: Macrolide antibiotics used in veterinary area may no shown depression on immune system. Hovewer, investigation is need that especially dose-depended manner effects of macrolide antibiotics on immunosystem.
