Abstract

Genetic variation is known to influence the amount of mRNA produced by a gene. Because molecular machines control mRNA levels of multiple genes, we expect genetic variation in components of these machines would influence multiple genes in a similar fashion. We show that this assumption is correct by using correlation of mRNA levels measured from multiple tissues in mouse strain panels to detect shared genetic influences. These correlating groups of genes (CGGs) have collective properties that on average account for 52–79% of the variability of their constituent genes and can contain genes that encode functionally related proteins. We show that the genetic influences are essentially tissue-specific and, consequently, the same genetic variations in one animal may upregulate a CGG in one tissue but downregulate the CGG in a second tissue. We further show similarly paradoxical behaviour of CGGs within the same tissues of different individuals. Thus, this class of genetic variation can result in complex inter- and intraindividual differences. This will create substantial challenges in humans, where multiple tissues are not readily available.

38. The design of unsurfaced roads using geosynthetics

Abstract

Current available methods for the design of unpaved roads, with and without geosynthetics, were used for sixteen full-scale test sections which were constructed at the Bothkennar Soft Clay Site, Airth, Scotland. The full-scale trials consisted of twelve pavements including geosynthetics and four control pavements incorporating two types of aggregate and two design life expectancies. The test pavements were instrumented to monitor the transient stress and strain distribution, permanent strain distribution, geosynthetic temperature and ground water level during the trafficking operation. Traffic loading was provided in two stages by a standard road-going vehicle. The vehicle used for Phase One applied an 80kN axle load and in Phase Two a 126kN axle load. Failure of the pavements was defined as a rut depth of 150mm. The passage of 2115 axles resulted in failure of three sections and significant deformations in many others. Back-calculation to compare predicted and measured performance was performed and hence the existing design methods were critically assessed. Where possible the measurements obtained from field trials were used to examine the assumptions made within the design methods. The existing design methods were found to be essentially static in approach and did not model transient stresses and strains or permanent strain development adequately. Pointers towards a new approach ensuring strain compatibility between the elements of the system are suggested. This should enhance the ability of the engineer to assess the value of differing products used in this application.

46. Structural Ambiguity and Reading Comprehension.

Abstract

This study questions the developmental nature of the ability to understand syntactic structures. An exploration is made of the possibility of learning more about reading comprehension and readability by examining responses made to sentences described by transformational grammarians as structurally ambiguous. A group of fifth grade students were asked to identify paraphrases of three kinds of sentences, ones with ambiguous surface structure, ones with ambiguous underlying structure, and ones considered to be unambiguous. (An example of a sentence with ambiguous surface structure is, "Small boys and girls are easily frightened." An example of a sentence with ambiguous underlying structure is, "Flying planes may be dangerous.") Although the fifth graders did not do well on the test, the order of difficulty of items was found to be similar to that found by other researchers. The order of difficulty ranged from unambiguous sentences as easiest, to surface structure ambiguities as next in difficulty, and underlying structural ambiguities as most difficult. Oral interviews with eight of the students after they took the test resulted in improved scores. A comparable study by Montague is reviewed and similarities in research findings are noted. (MKM)

50. Sentence Interpretation Test (SIT).

Abstract

Designed to measure the ability of children to identify the meanings of structurally ambiguous or unambiguous sentences of English, the test consists of forty lead sentences: ten with surface structure ambiguity, ten with underlying structure ambiguity, and twenty which are unambiguous. For each of the lead sentences three interpretative sentences were constructed, one, two, or all three of which gave a meaning of the lead sentence.
