Abstract Guizhou Province is one of the most important karst regions of southwest China, with 22 Primulina species. These species are highly habitat-specialized and dependent on the soils of the karst region, and many inter-species classifications are unclear. Therefore, studying the chloroplast genomes and estimating the divergence times of there species can not only provide a better understanding of interspecific relationships but also help to know the species speciation and divergence in the karst environment. So, we sequenced and assembled the chloroplast genomes of nine Primulina species (including six endemic species of Guizhou) and conducted chloroplast genome comparison analysis and phylogenetic study. The chloroplast genome structures of the nine Primulina species were quadripartite with total lengths of 152,869–153,364 base pairs (bp) and GC content of 37.55–37.64%. There are 132 functional genes annotated, respectively. A total of 375 simple sequence repeats and 375 interspersed nuclear elements were identified. The 30 highly preferred codons identified were used at similar frequencies in different species, respectively. A phylogenetic tree constructed on the basis of the 38 chloroplast genomes showed that Primulina species form a monophyletic group. Eleven mutational hotspot regions that could serve as potential molecular markers were identified, of which two regions near the 3′ and 5′ ends of the ycf1 gene were of appropriate size and can serve as molecular markers for phylogenetic studies of Primulina. The results of molecular clock analyses indicate that the three major branches of Primulina begin to diverge in the Miocene, and the number of species proliferated in the Pliocene and Pleistocene. Most of the species of Primulina in Guizhou Province were formed in the Pleistocene and rapidly diverged within a short period of time. This research study enriches the genetic resource information of Primulina and deepens the understanding of the phylogenetic relationships of the genus.

Abstract Due to the multiple influences of unique physicochemical properties of helium, petrographic characteristics and temperature and pressure conditions, little is known about the helium adsorption behaviors in minerals and rocks at geological conditions. Based on the grand canonical Monte Carlo simulations, this study revealed the adsorption characteristics of pure helium and the competitive adsorption of binary mixtures with different proportions of methane and helium under geological temperature and pressure conditions in quartz slit model. Molecular simulation of pure helium shows that physical adsorption of helium exists in mineral surfaces, which indicates a preservation mechanism of helium in helium source rocks. Binary mixtures simulations indicate that the adsorption capacity of methane in quartz is stronger than that of helium, and the competitive adsorption of methane increases with decreasing burial depth. This means that during the upwards migration processes of natural gas, the adsorbed helium that distributed in the migration pathway will be gradually displaced by methane, then concentrate in the hydrocarbon gases and subsequently accumulate together in favorable traps to form helium-rich natural gas reservoirs. Our results provide a molecular-scale insight into the preservation and accumulation of helium in helium source rocks and are significant for assessing the helium resource potential.

Abstract Background Hepatitis B virus (HBV) infection is a major concern regarding blood safety in countries with a high HBV prevalence, such as China. We aimed to understand the prevalence of HBV infection among blood donors in Chongqing and provide an important basis for developing appropriate blood screening strategies. Methods Dual enzyme-linked immunosorbent assays (ELISAs) for hepatitis B surface antigen (HBsAg) were conducted in parallel with nucleic acid testing (NAT) of donors. All HBsAg-reactive and/or HBV DNA-positive blood samples were tested for HBsAg and hepatitis B DNA levels. Results A total of 117,927 blood donor samples were collected from the Chongqing Blood Center between April 2020 and November 2020. In total, 473 HBV-ineligible samples were retained for HBsAg and DNA confirmation. A total of 272 samples were confirmed to be HBsAg+, including 2 HBV DNA − and 270 HBV DNA + samples. Objective To conduct Rh blood group serological testing and third-generation sequencing(TGS) on 22 RhD variant voluntary blood donors in Chongqing and explore the phenotypic distribution and genotyping of RhD variants in Chongqing. Methods From January to August 2023, individuals who participated in blood donation in our blood center were selected as the study objects. RhD variant phenotype identification was performed using routine serological methods. Once the RhD variants were identified, tests on different antigenic epitopes of RhD were conducted using a D-screen assay kit. Furthermore, after the genomic DNA from 22 RhD variant blood samples was extracted, imbraided primers design and multi-segment amplification and splicing were used to sequence the full-length RHD gene for TGS. The RHD gene sequence was analyzed using SnapGene software. Results Among the 22 RhD variants, 8 were DVI type 3 (36.36%), with the main mutation of RHD-CE (3-6)-D hybrid allele. Six cases (27.27%) showed partial weak D15 type, with the main mutation of c.845G>A. There were 6 cases of Asia type Del (27.27%), with the main mutation of c.1227G>A. One case was weak D17 type with a mutation of c.340C>T and 1 case speculated to be partial D (c.491A>T, p. Asp164Val, missense mutation). Conclusion The most common RhD variant phenotype among blood donors in Chongqing is DVI type 3, and the full-length haplotype sequence of RHD variant alleles can be obtained by Pacific Bioscience single-molecule real-time sequencing(SMRT).

The Fushan Depression is a hydrocarbon-rich depression in the Beibuwan Basin, South China Sea. In this study, 14 source rocks and 19 crude oils from the Chaoyang Step-Fault Zone and Southern Slope Zone were geochemically analyzed to determine their origins. The hydrocarbon generation, migration, and accumulation processes were also determined using two-dimensional basin modeling. Crude oils from the low-step area show a close relationship with the source rocks of the first and second members of the Eocene Liushagang Formation (Els1 and Els2). The oils from the middle-step area and the Southern Slope Zone are derived from the local source rocks in those areas, in the third member of the Eocene Liushagang Formation (Els3). Hydrocarbons generated from the Els3 source rocks of the Southern Slope Zone migrated along sand bodies to the Els3 reservoir. The fault system of the Chaoyang Step-Fault Zone controls hydrocarbon migration and accumulation in the low-step and middle-step areas. The resource potential of the middle-step area is limited by its shallow burial depth. The low-step area is a more favorable exploration area due to its proximity to the source kitchen.

Abstract Background Lung cancer is one of the most common malignant cancers worldwide. Previous studies have shown that Afzelin, a flavonoid, possesses anticancer activity. The aim of this study was to explore Afzelin’s effect on lung cancer cells and delineate potential anti-cancer mechanism. Methods The effect of Afzelin on cell viability, proliferation, and apoptosis of lung cancer cells i.e., A549 and H1299 cells, was studied. The targets for Afzelin in lung cancer were predicted using SwissTargetPrediction, Next, the GO analysis and pathway enrichment were analyzed using String. For in vitro studies, the overexpression plasmid of NQO2, the identified target of Afzelin, was transfected into Afzelin-treated cells to verify the regulatory role of Afzelin on its target and signaling pathway. Results In in vitro studies, Afzelin markedly inhibited cell viability, proliferation, and raised apoptotic rate of A549 and H1299 cells. In addition, Afzelin activated endoplasmic reticulum (ER) stress and increased ATP, HMGB1, and CRT levels in lung cancer cells, indicating that Afzelin induced immunogenic cell death (ICD). SwissTargetPrediction identified NQO2 as a target of Afzelin. Further, Afzelin markedly inhibited NQO2 protein expression and in turn, overexpression of NQO2 attenuated the effect of Afzelin on A549 and H1299 cells. Conclusion Afzelin inhibits lung cancer progression by targeting NQO2, in turn, activating ER stress and inducing ICD.

Soil hydraulic parameters are influenced by various inherent soil properties, such as pore structure and organic matter content, which can vary with changes in the ecosystem. However, identifying the temporal variations of soil hydraulic parameters in a co-evolving soil-vegetation system remains a challenge. This study focused on a tropical forest with significant seasonal variations in vegetation attributes, evaporation, and carbon fluxes over a five-year monitoring period. The particle batch smoother algorithm was integrated with an unsaturated flow model to identify the seasonally varied soil hydraulic parameters through assimilation of in-situ measured soil moisture. As a benchmark, the Generalized Likelihood Uncertainty Estimation method was applied to optimize soil hydraulic parameters without considering temporal variation. The results indicated that the temporally varying soil hydraulic parameters exhibited regular seasonal patterns and outperformed the unvaried soil hydraulic parameters in terms of reducing the errors in modeling of soil moisture and evaporation. Moreover, the seasonal variations in soil hydraulic parameters were closely linked to changes in the litterfall and terrestrial carbon fluxes over time. Specifically, due to the hysteresis of the transformation from litterfall to soil organic matter, the accumulated litterfall in Hot-dry season can replenish the soil organic matter, resulting in an increase in field capacity and saturated hydraulic conductivity in the Hot-rainy season. However, the intense decomposition of soil organic matter under high temperature in Hot-dry season led to a decrease in field capacity and saturated hydraulic conductivity. This study emphasizes the value of the particle batch smoother algorithm in detecting temporal variations in soil hydraulic parameters within a coevolving soil-vegetation system, thereby contributing to a more comprehensive understanding of the intricate dynamics within the ecohydrological system under a changing environment.

【Objective】In order to establish the reasonable planting density under the double-row planting model of large ridges and build a high-yielding peanut group model, the impact of the double-row planting density of large ridges (90 cm) on the characteristics, group structure changes and yield of peanut plants was studied.【Method】The semi-trailing type peanut variety 'Silihong' and the erect type 'Jihua26'were used as experimental materials. Set 6 planting densities: M1 (31.71×104 plants/hm2), M2 (27.75×104 plants/hm2), M3 (24.67×104 plants/hm2), M4 (22.20×104 plants/hm2), M5 (18.50×104 plants/hm2), M6 (15.85×104 plants/hm2), based on single ridge (60 cm) double-seed sowing (11.93×104 holes/hm2) as a comparison (CK). Determine the seedling rate, plant growth, SPAD value, dry matter accumulation and yield of each peanut variety under different density, and establish a reasonable group structure of peanuts and the optimal planting density of different varieties.【Result】The emergence time of different varieties was different under different planting densities, the emergence time of 'Silihong' was earlier than 'Jihua 26'. 25 days after sowing, the seedling rate of 'Silihong' was up to 100% at M3 density, 30 days after sowing, the seedling rate of 'Jihua26' was up to 100% at M3 density, which were significantly higher than other densities. With the decrease of planting density, the height of the main stem and the length of the side branch gradually increase with the decrease of the number of plants per unit area; the SPAD value tends to increase first and then decrease with the decrease of planting density; 'Jihua 26' with the decrease of density, the quality of dry matter showed a tendency to rise first and then decrease, and 'Silihong' the accumulated amount showed a trend of gradually increasing. With the reduction of planting density, the number of pods per plant, pod yield per plant and weight of 100-pod increased first and then decreased of the two experimental varieties, the highest value was under M4 density of 'Jihua26' and M5 density of 'Silihong', which were significantly higher than other densities. The yield of the two experimental varieties continues to increase, Jihua 26 at M4 density had highest yield was 7 956.67 kg/hm2 and 'Silihong' at M5 density had highest yield was 4 790.73 kg/hm2, compare to control increase 96.14% and 65.42% respectively.【Conclusion】Compared with the single-row double-seed planting mode, the large ridge double-row + single-seed planting mode can increase the peanut height, SPAD and dry matter accumulation. At the same time, the yield is improved. Under the condition that the density remains unchanged, peanuts can be changed from double-seed to single-seed precision sowing, which increases the yield. When planting in two rows of large ridges, the appropriate sowing density should be selected according to the type of peanut varieties. The M5 density of the semi-trailing type peanut varieties is the best, and the M4 density of the erect type peanut is the best.

To study the efficiency of flue gas sequestration with enhanced shale gas recovery in quartz slit, Grand Canonical Monte Carlo (GCMC) and Molecular Dynamics (MD) simulations were adopted to investigate the influence of burial depth, formation water content and the injection ratio of flue gas on the recovery efficiency of shale gas (CH4) in quartz slit by flue gas (CO2/N2). The density distribution, loading, adsorption heat and interaction energy of each component were systematically analyzed to reveal their adsorption mechanisms and the recovery efficiency of CH4. It indicates that the loadings of mixed CH4 and N2 (ΓCH4 and ΓN2) exhibit negative correlation with formation water content. With the increasing burial depth, both ΓCH4 and ΓN2 increase at first and then tend to be constant when the burial depth is over 2 400 m. The maximum loading of CO2 (ΓCO2) occurs at the burial depth of 2 400 m. There is a positive correlation of ΓCO2 with formation water content when the burial depth is below 2 400 m, and a negative correlation when the burial depth is over 2 400 m. The recovery efficiency of CH4 (η) reaches the maximum point at the burial depth of 400-600 m, which increases with the increasing mole fraction of CO2 in flue gas, showing that CO2 in flue gas can promote the displacement of CH4 significantly.

Petrocodon Hance is one of the famous ornamental flowers, but most of them are assessed as extremely dangerous (CR), or at least above vulnerable (VU) due to climate turbulence and strong interference of human activities. To reconstruct the temporal and spatial changes of the potential suitable areas of Petrocodon since the last interglacial period, and explore the response of the suitable areas to environmental changes, which provided theoretical guidance for the origin of Petrocodon, the study of geographical differentiation, the conservation of unique germplasm resources in China, and the development and utilization of gardens. In this paper, combined with 120 distribution records and 17 environmental variables, the optimal MaxEnt model and geographic information technology (ArcGIS) were used to simulate the suitable areas and distribution pattern of Petrocodon in China and Indo-China Peninsula. Based on stepwise multiple linear regression analysis, redundancy analysis and Monte-Carlo test, the dominant variables affecting the current geographical distribution of Petrocodon were evaluated. The results were as follows: (1) The prediction accuracy of the optimized MaxEnt model was high, and the AUC value was greater than 0.96. The potential suitable areas of Petrocodon for the present distribution are continuous from Southwest China to North Vietnam, scattered in central and southern China and blocky in North Myanmar, of which the South Yunnan-Guizhou Plateau in China was the most suitable. (2) The environmental variables which restrict the geographical distribution of Petrocodon included precipitation of the driest month (bio14), precipitation of the warmest quarter (bio18), precipitation of the wettest quarter (bio16), SD of the temperature seasonality (bio4), min temperature of the coldest month (bio6) and altitude (alt). (3) Under the climate change scenario, the expansion and contraction areas of the suitable habitat of Petrocodon were located in the north and northeast of the current potential distribution area, which were sensitive areas susceptible to climate change. During the last interglacial period, the suitable area of Petrocodon expanded on a large scale, but there was almost no suitable distribution area of Petrocodon in the dry and cold environment during the last glacial maximum. After that, with the aggravation of climate warming, the suitable habitats of Petrocodon increased rapidly to high latitudes, while the suitable habitat in low latitudes decreased. (4) The centroid position of the suitable area for Petrocodon migrated northwards from Yongfu County, Guangxi (110.10° E, 24.69° N) to Chengbu County, Hunan (110.29° E, 26.05° N). To sum up, global warming has a positive impact on the potential distribution area of Petrocodon, but extreme warming will cause the suitable habitat of Petrocodon to shrink, and the niche of Petrocodon will narrow. Southwest China to North Vietnam, which has the advantage of mature karst landform, may be its main refuge.

Light oil, a high-quality energy resource abundant in deep basins, is prevalent in the northern zone of the Dongying Depression. To elucidate the formation mechanism of light oil reservoirs, this study investigates the molecular and stable isotope composition, biomarkers, light hydrocarbons, and diamondoid compositions of petroleum. The results reveal that the gas primarily consists of oil-cracking gas from a late filling event, mixed with oil-associated gas generated during the source rock’s “oil window” maturity phase. Methane exhibits enriched light carbon isotopes, indicating noticeable migration fractionation effects. The crude oil in the same deep strata exhibits high maturity, originating from both terrestrial and aquatic organic matter in the source materials. Molar proportions of n-alkanes and light hydrocarbon indices (Tol/nC7, nC7/MCH) indicate significant evaporative fractionation in the petroleum reservoirs. This fractionation process modified early-formed oil reservoirs due to the late filling of highly mature gas reservoirs. The evaporative fractionation at different stages has varying effects on the diamondoid ratio (1- + 2-MA)/(3- + 4-MD). It is considered a pivotal mechanism in the formation of deep condensate reservoirs and volatile oil reservoirs.

Objective: We examined the relationships between infants’ growth trajectories and prenatal exposure to air pollution, which is still under-investigated. Methods: A birth cohort study was constructed using medical records of pregnant women and infants born between 2015 and 2019 in Foshan, China. Using satellite-based spatial-temporal models, prenatal exposure to air pollutants including particulate matter with an aerodynamic dimension of < 2.5 µm (PM2.5), sulfur dioxide (SO2), nitrogen dioxide (NO2), and ozone (O3) was assessed at each woman's residence. Latent class growth modeling was used to identify trajectories of physical (body length and weight) growth and neurodevelopment, which were repeatedly measured within 1 year after birth. Logistic regression models were used to investigate the associations between prenatal exposure to air pollution and the risks of growth disorders, adjusting for an array of potential confounders. Results: We identified two growth trajectories for body length [normal: 3829 (93%); retardation: 288 (7%)], three for weight [normal: 2475 (59.6%); retardation: 390 (9.4%); overgrowth: 1287 (31%)], and two for neurodevelopment [normal: 956 (66.1%); retardation: 491 (33.9%)]. For exposure over whole pregnancy, SO2 was associated with an increased risk of body length retardation (OR for per 1 µg/m3 increment: 1.09, 95%CI: 1.01–1.17); PM2.5 (OR: 1.05, 95%CI: 1.03–1.07), SO2 (OR: 1.15, 95%CI: 1.08–1.22), and NO2 (OR: 1.05, 95%CI: 1.03–1.07) were positively associated with neurodevelopmental retardation. Such associations appeared stronger for exposures over the first and second trimesters. No significant associations were detected for weight growth. Conclusions: Maternal exposure to air pollution during pregnancy was associated with higher risks of impairments in both physical growth, particularly body length, and neurodevelopment.

The hydrocarbon phase state of deep to ultra-deep reservoirs in the Tarim and Sichuan basins has been of great interest in oil and gas exploration. Based on a combination of molecular dynamics simulation, gold-tube pyrolysis experiments, and geological-geochemical theory, this study discusses the mechanisms governing the stability of oils in deep reservoirs from the perspectives of their reservoir accumulation histories and chemical reactions. Generally, the reason for the existence of liquid oil in the Tarim Basin is widely considered to be only controlled by external geological conditions, mainly including low geothermal gradient, absence of thermal events, low maximum reservoir temperatures, and late hydrocarbon generation process. However, this study firstly proposed that the chemical composition of oil is an internal factor for its thermal stability. The simulation results reveal that the polycondensation reactions of asphaltene will release hydrogen atoms, which can provide a necessary hydrogen source for cracking of liquid chain hydrocarbons. It means that the presence of asphaltene components can promote the cracking of chain hydrocarbons and generate methane. The normal mature oil in the Sichuan Basin generally has higher contents of asphaltenes than that of the high-mature light oil of the Tarim Basin, so more hydrogen has historically been available for the cracking of oil to gas. By looking at the accumulation histories and chemical compositions of the crude oils, this study first explains the stable long-term storage of liquid hydrocarbons in the Tarim Basin, providing important guidance for future deep to ultra-deep oil and gas exploration.

Crude oils from the Baiyun Sag of Pearl River Mouth Basin have high abundance of diagnostic biomarkers including novel C15 sesquiterpene, special configurated tricyclic sesquiterpenes and tetracyclic sesquiterpenes (X, Y, Z and X1, Y1, Z1), and bicadinanes, which indicate the contributions from terrestrial higher plants to the organic matters. These oils are also rich in oleanane, which is a diagnostic biomarker for angiosperm organic matter. Based on the analysis of 13 crude oils from the Baiyun Sag, the compositions and geochemical significance of typical biomarkers were revealed. Results indicate that C24-des-oleanane (Y1) and C27 tetracyclic (Z1) may share a similar biological origin. Novel C15 sesquiterpene, C24-des-oleanane and C27 tetracyclic, and bicadinanes may be derived from different higher plants according to the molecular structure and abundance correlation analysis of the related compounds. In addition, the biomarker compositions of crude oils from different regions are significantly varied, which will benefit the oil group classification in the Baiyun Sag. The distribution of novel C15 sesquiterpene and bicadinanes was influenced by sedimentary environment, and may be more enriched in oxidized environment. The distribution patterns of C24-des-oleanane, C27 tetracyclic and oleanane are principally affected by biogenic input, but are less influenced by sedimentary environment fluctuation. The relatively high abundance of C24-des-oleanane, C27 tetracyclic and oleanane in the crude oil in the northeastern and eastern Baiyun area reflects that the Dongsha Uplift and the Yunli Low Uplift may be an important source of terrigenous organic matter in this area.

Western Hunan province and its surrounding areas are significant targets for shale gas exploration and development in southern China, where the black shale of the lower Cambrian Niutitang Formation and Wunitang Formation is extensively distributed. Geochemical analysis was conducted on the lower Cambrian black shale from a new exploration well of XAD1 located at the southeast margin of the Yangtze paraplatform, followed by a discussion on gas-bearing properties using molecular dynamics simulation. The geochemical characteristics indicate that the black shale in well XAD1 was primarily deposited in a strongly reducing marine environment, with organic matter predominantly composed of type I kerogen derived from algae. Currently, it has reached a stage of high to over maturity with limited potential for liquid hydrocarbon generation. The recovery of the original hydrocarbon generation potential shows that they are excellent source rocks and have completed the main hydrocarbon generation evolution. Despite the favorable conditions for shale gas formation observed in well XAD1, the low measured gas content within the Niutitang Formation suggests that other geological factors may have contributed to a substantial loss of shale gas. Gas adsorption simulation reveals that the maximum methane adsorption capacity (15.77 m3/t) was achieved by Niutitang shale during the late Silurian period when there was an abundant source of natural gas without any influence from CO2, H2O or other molecules. However, due to a lack of natural gas replenishment and subsequent tectonic uplift and subsidence causing variations in temperature and pressure, the methane adsorption capacity gradually decreased (to 6.56 m3/t). Furthermore, water occurrence within the shale reservoir further reduced the methane adsorption capacity (below 2 m3/t), while tectonic activities exacerbated the loss of shale gas potential within this study area. The findings indicate that the dynamic alteration of gas-bearing properties in shale reservoirs due to tectonic movements is a crucial factor influencing the success rate of shale gas exploration in the study area, provided that there are sufficient gas resources and superior reservoir conditions.

Abstract Background The safety assessment of ulinastatin can guide clinical practice. The present study aimed to investigate the real-world safety of ulinastatin in China. Methods This multicenter study retrospectively analyzed the post-marketing surveillance data of consecutive patients treated with ulinastatin between August 2014 and June 2017 in the general wards and the intensive care units (ICU) of nine hospitals in China. Adverse drug reactions/adverse drug events (ADRs/ADEs) were collected and evaluated in a post-marketing database. Results A total of 11,252 consecutive patients were included in the study: 7009 ICU patients and 4243 general ward patients. Eleven patients with ADRs/ADEs were observed, including nine ICU patients and two general ward patients. The clinical manifestations were liver dysfunction (n = 5 ICU cases, n = 1 general case), thrombocytopenia (n = 2 ICU cases, n = 1 general case), leukopenia (n = 1 ICU case), and rash (n = 1 ICU case). During the study period, the drug ADR/ADE rate of ulinastatin injection was 0.98‰ (11/11,252 × 1000‰). Among the 11,252 valid patients, only 327 received ulinastatin in accordance with the drug specifications. After excluding unreasonable drug use, the calculated ADR rate was 3.06‰ (1/327 × 1000‰) (95% confidence interval: 0.0‰-17.1‰). Laser Raman spectroscopy has shown a good application prospect for maceral analysis. Raman spectroscopic analysis of different macerals (e.g., vitrinite, semifusinite and macrinite) in coal samples with different maturity (%Ro=0.49%-1.88%) was carried out in this study, and results show that three macerals have significantly different Raman spectrum parameters, which have the following implications for the macerals analysis of coal: (1) Macerals in coal samples can be distinguished by the combination of Raman spectrum parameters. There are 21 kinds of parameter combinations discussed in this study, which can be used as reference standard for the classification of these organic macerals; (2) Peak displacement (WD1) is the most critical parameter to distinguish the macerals of coal samples. The influence of thermal evolution should be considered, which may assistant for the study of maceral differences in the Lower Paleozoic which are in the high to over mature stage with optical properties gradually converging. Therefore, Raman spectrum parameters can be used as an effective method for maceral analysis.

Large amounts of natural gas hydrates have been discovered in the Qiongdongnan Basin (QDNB), South China Sea. The chemical and stable carbon isotopic composition shows that the hydrate-bound gas was a mixture of thermogenic and microbial gases. It is estimated that microbial gas accounts for 40.96% to 60.58%, showing a trend of decrease with the increase in burial depth. A significant amount of gas hydrates is thought to be stored in the mass transport deposits (MTDs), exhibiting vertical superposition characteristics. The stable carbon isotopic values of methane (δ13C1) in the MTD1, located near the seabed, are less than −55‰, while those of the methane below the bottom boundary of MTD3 are all higher than −55‰. The pure structure I (sI) and structure II (sII) gas hydrates were discovered at the depths of 8 mbsf and 145.65 mbsf, respectively, with mixed sI and sII gas hydrates occurring in the depth range 58–144 mbsf. In addition, a series of indigenous organic matters and allochthonous hydrocarbons were extracted from the hydrate-bearing sediments, which were characterized by the origin of immature terrigenous organic matter and low-moderate mature marine algal/bacterial materials, respectively. More allochthonous (migrated) hydrocarbons were also discovered in the sediments below the bottom boundary of MTD3. The gas hydrated is “wet gas” characterized by a low C1/(C2 + C3) ratio, from 2.55 to 43.33, which was mainly derived from a deeply buried source kitchen at a mature stage. There is change in the heterogeneity between the compositions of gas and biomarkers at the site GMGS5-W08 along the depth and there is generally a higher proportion of thermogenic hydrocarbons at the bottom boundary of each MTDs, which indicates a varying contribution of deeply buried thermogenic hydrocarbons. Our results indicate that the MTDs played a blocking role in regulating the vertical transportation of hydrate-related gases and affect the distribution of gas hydrate accumulation in the QDNB.

Background: The relationship between air pollution and stroke has been extensively studied, however, the evidence regarding the association between air pollution and hospitalization due to stroke and its subtypes in coastal areas of China is limited. Objective: To estimate the associations between air pollution and hospitalizations of stroke and its subtypes in the Beibu Gulf Region of China. Methods: We conducted a time-stratified case-crossover study in 15 cities in Beibu Gulf Region in China from 2013 to 2016. Exposures to PM1, PM2.5, PM10, SO2, NO2, O3, and CO on the case and control days were assessed at residential addresses using bilinear interpolation. Conditional logistic regressions were constructed to estimate city-specific associations adjusting for meteorological factors and public holidays. Meta-analysis was further conducted to pool all city-level estimates. Results: There were 271,394 case days and 922,305 control days. The odds ratios (ORs) for stroke hospitalizations associated with each interquartile range (IQR) increase in 2-day averages of SO2 (IQR: 10.8 µg/m3), NO2 (IQR: 11.2 µg/m3), and PM10 (IQR: 37 µg/m3) were 1.047 (95 % CI [confidence interval]: 1.015–1.080), 1.040 (95 % CI: 1.027–1.053), and 1.018 (95 % CI: 1.004–1.033), respectively. The associations with hospitalizations of ischemic stroke were significant for all seven pollutants, while the association with hemorrhagic stroke was significant only for CO. The associations of SO2, NO2, and O3 with stroke hospitalization were significantly stronger in the cool season. Conclusions: Short-term increase in SO2, NO2, and PM10 might be important triggers of stroke hospitalization. All seven air pollutants were associated with ischemic stroke hospitalization, while only CO was associated with hemorrhagic stroke hospitalization. These results should be considered in public health policy.

Abstract Dysregulation of microRNAs (miRNAs) exerts key roles in the development of pancreatic cancer (PCa). miR-26a is reportedly a tumor suppressor in cancers. However, whether miR-26a modulates PCa progression is poorly understood. Here, we found that miR-26a was down-regulated in PCa. Overexpressed miR-26a suppressed PCa cell proliferation, colony formation, and tumor stem cell properties. Mechanically, the transcription factor E2F7 is a downstream target of miR-26a. miR-26a decreased E2F7 expression through binding to the 3’-untranslated region (UTR) of E2F7. Decreased miR-26a in PCa tissues was inversely correlated with E2F7. The inhibitory effects of miR-26a in PCa were reversed by E2F7 overexpression. Consistently, the knockout of E2F7 further significantly inhibited the growth of PCa cells combined with miR-26a overexpression. Further study revealed that E2F7 bound the promoter of vascular endothelial growth factor A (VEGFA), a key factor in angiogenesis, and transcriptionally activated the expression of VEGFA. miR-26a overexpression attenuated the effects of E2F7 on VEGFA promotion. Our results uncovered the novel function of miR-26a/E2F7/VEGFA in PCa, making miR-26a a possible target for PCa treatment.

The fragmentation of normal alkanes during the cracking of oil under geochemical conditions with high thermal stress is important for the preservation of crude oils in reservoirs and the formation of gaseous hydrocarbons. In this study, the kinetics behavior, cracking mechanisms, and products of n -tetradecane pyrolysis were investigated using reactive molecular dynamics (ReaxFF MD) simulations at high temperatures (2000K∼3000 K). The pyrolysis process itself, its main products and intermediates, and kinetic behavior were analyzed at an atomic/molecular level. Low molecular weight (C 1 –C 5 ) alkanes and olefins, together with H 2 , were the predominant intermediates and products of the simulations. Three distinct stages—a stable stage, initial decomposition, and secondary pyrolysis—can be distinguished during the decomposition process. The reactant n -tetradecane was exhausted when the simulation temperature reaches ∼2250 K, accompanied by a rapid increase in hydrocarbons with molecular numbers C 1 –C 5 . The yield of C 2 –C 5 peaked at ∼2500 K and then decreased. The number of CH 4 molecules increased continuously throughout the entire simulation process because of the contribution of secondary pyrolysis. The Arrhenius parameters obtained from ReaxFF MD simulations, on the basis of first-order kinetic analysis of n -tetradecane, were reasonably consistent with experimental data and generally in agreement with results from pyrolysis experiments on crude oils in the laboratory. The pyrolysis process and reaction mechanism of n -tetradecane were also reasonably consistent with laboratory pyrolysis experiments on whole oils and their individual components. ReaxFF molecular dynamics simulation is therefore considered to be a valid approach for the study of thermal cracking of subsurface crude oils.

Background: Atrial fibrillation (AF) has affected millions of adults in the world. It is important to monitor and manage blood pressure (BP) in AF patients. The accuracy of BP monitoring in AF patients with noninvasive methods remains questionable, however. Objectives: To compare the accuracy of different noninvasive BP devices (oscillographic sphygmomanometer and pulse wave device) for BP measurement in elderly patients with AF, with a mercury sphygmomanometer as a reference. Design: This study was an observational study. Methods: Patients with AF from the inpatient department of cardiology were included from 1 January to 31 December 2020. BP measurements were performed by two trained nurses using a tee junction connection on the cuff to connect three sphygmomanometers. The Bland–Altman plot analysis was conducted to compare the agreement of BP measurements. We also compared the agreement of BP measurements through metrics such as accuracy, bias, and precision. Results: A total of 202 patients (54.5% female) were included. The Bland–Altman plot analysis showed that the lower and upper limits of agreement (LoAs) of pulse wave/reference were similar to the predefined acceptable clinical limits (10/5 mmHg). The bias and precision in both systolic and diastolic BP were significantly less in pulse wave/reference (a bias of 1.8 and 0.77 mmHg and a precision of 5.20 and 4.66 mmHg, respectively), with corresponding higher accuracy readings (98.51% for P10 in systolic BP and 85.64% for P5 in diastolic BP). Conclusion: A novel noninvasive sphygmomanometer – pulse wave device has a good concordance with a mercury sphygmomanometer in BP monitoring, and may be applicable to perform BP measurements in the elderly with AF.

Weaning is considered to be one of the most critical periods in pig production, which is related to the economic benefits of pig farms. However, in actual production, many piglets are often subjected to weaning stress due to the sudden separation from the sow, the changes in diet and living environment, and other social challenges. Weaning stress often causes changes in the morphology and function of the small intestine of piglets, disrupts digestion and absorption capacity, destroys intestinal barrier function, and ultimately leads to reduced feed intake, increased diarrhea rate, and growth retardation. Therefore, correctly understanding the effects of weaning stress on intestinal health have important guiding significance for nutritional regulation of intestinal injury caused by weaning stress. In this review, we mainly reviewed the effects of weaning stress on the intestinal health of piglets, from the aspects of intestinal development, and intestinal barrier function, thereby providing a theoretical basis for nutritional strategies to alleviate weaning stress in mammals in future studies.

Buried hill zones in the rift basins have a significant impact on the enrichment of natural gas resources, and this is of great significance for exploration and development. This study aims to unravel the origins, migration, and dynamic accumulation process of natural gas in the Xinglongtai structural belt, Liaohe Subbasin. A comprehensive geological and geochemical analysis was performed on source rocks and natural gas samples from various geological structures within the Xinglongtai structural belt. Moreover, basin modeling techniques were employed to trace the genesis and migration of natural gas, offering an in-depth understanding of the dynamic process of accumulation. We identified the Fourth Mbr (Es4) and Third Mbr (Es3) of the Shahejie Fm as the main source rocks in the Qingshui and Chenjia Sags. The Es4, primarily Shallow Lacustrine Mudstones, contributed mainly type II organic matter, while the Es3, consisting of Nearshore Subaqueous Fan and Deep Lacustrine Mudstones, contributed mainly type III and type II organic matter, respectively. Two distinct hydrocarbon accumulation systems were observed, one inside and one outside the buried hills. The system outside the buried hill is governed by a complex fault system within the lacustrine basin, resulting in dual-source directions, dual-source rock types, two migration phases, and late-stage accumulation. In contrast, the system within the buried hill primarily involves reservoirs nested in the basement, exhibiting dual-source directions, dual-source rock types, a single migration phase, and early-stage charging. The understanding of this interplay, alongside dynamic simulation of generation, migration, and accumulation, can provide valuable insights for predicting natural gas distribution and accumulation patterns in terrestrial faulted lacustrine basins. This knowledge can guide more effective exploration and development strategies for natural gas.

In clinical practice, ovarian pregnancy is extremely rare and is always found to be ruptured. A definitive diagnosis for ruptured ovarian pregnancy is difficult to obtain. We present two cases of unruptured ovarian pregnancies detected during laparoscopy and review existing literature to better understand the clinical characteristics of ectopic pregnancies in this rare site. Intrauterine devices, assisted reproductive technology, and intrauterine operations are all high-risk factors in ovarian pregnancy. Moreover, menopause, abdominal pain, and vaginal bleeding are clinical manifestations. Ovarian pregnancy can be diagnosed using serum hCG, transvaginal ultrasound, and magnetic resonance imaging. Laparoscopy is the treatment of choice for ovarian pregnancy. It is recommended that the intact gestational sac be excised and the ovarian function be protected to the greatest extent possible during the operation. More definitive diagnosis of ovarian pregnancy must be reported in order to gain a better understanding of ovarian pregnancy.

The separation and preparation process of petroleum group components is the basis for the subsequent molecular composition and isotopic analysis. Due to the complex composition of isomers, the components of aromatic compounds prepared by traditional techniques still cannot meet the isotopic analysis requirements of specific compounds. Two different separating methods were used to finely separate and prepare polycyclic aromatic hydrocarbons in crude oils. The "one-step method" was to directly pass the de-asphalted crude oil samples through an alumina/silica gel chromatography column, and elute them with different ratios of petroleum ether/dichloromethane mixed solvent. The "two-step method" firstly used conventional separation methods to obtain aromatic fraction from crude oils, and then applied the aromatic fraction to a column filled with alumina and sub-fractions with different ring numbers then eluted with mixed solvent with different ratios of petroleum ether/dichloromethane. Gas chromatography-mass spectrometry (GC-MS) analysis for the sub-fractions were carried out for the comparison between the two separation methods. Results show that the "two-step method" can better realize the separation and preparation of monocyclic, bicyclic and tricyclic aromatic compounds. The mixed solvents of petroleum ether/dichloromethane of V/V: 99/1, 9/1 and 8/2 can obtain monocyclic, bicyclic and tricyclic aromatic compounds respectively.

Opisthorchiosis is a parasitic liver disease found in mammals that is widespread throughout the world and causes systemic inflammation. Praziquantel remains the drug of choice for the treatment of opisthorchiosis, despite its many adverse effects. An anthelmintic effect is attributed to the main curcuminoid of Curcuma longa L. roots—curcumin (Cur)—along with many other therapeutic properties. To overcome the poor solubility of curcumin in water, a micellar complex of curcumin with the disodium salt of glycyrrhizic acid (Cur:Na2GA, molar ratio 1:1) was prepared via solid-phase mechanical processing. In vitro experiments revealed a noticeable immobilizing effect of curcumin and of Cur:Na2GA on mature and juvenile Opisthorchis felineus individuals. In vivo experiments showed that curcumin (50 mg/kg) had an anthelmintic effect after 30 days of administration to O. felineus-infected hamsters, but the effect was weaker than that of a single administration of praziquantel (400 mg/kg). Cur:Na2GA (50 mg/kg, 30 days), which contains less free curcumin, did not exert this action. The complex, just as free curcumin or better, activated the expression of bile acid synthesis genes (Cyp7A1, Fxr, and Rxra), which was suppressed by O. felineus infection and by praziquantel. Curcumin reduced the rate of inflammatory infiltration, whereas Cur:Na2GA reduced periductal fibrosis. Immunohistochemically, a decrease in liver inflammation markers was found, which is determined by calculating the numbers of tumor-necrosis-factor-positive cells during the curcumin treatment and of kynurenine-3-monooxygenase-positive cells during the Cur:Na2GA treatment. A biochemical blood test revealed a normalizing effect of Cur:Na2GA (comparable to that of curcumin) on lipid metabolism. We believe that the further development and investigation of therapeutics based on curcuminoids in relation Opisthorchis felineus and other trematode infections will be useful for clinical practice and veterinary medicine.

Methylated 2-methyl-2-(4,8,12-trimethyltridecyl)chromans are salinity-sensitive biomarkers that have been detected in immature – early mature petroleum and sediments. In this study, the occurrence and distribution patterns of 2-methyl-2-(4,8,12-trimethyltridecyl)chromans were investigated in a set of lacustrine sediments from Nördlinger Ries of southern Germany and marine sediments from the South China Sea. Among all of the 2-methyl-2-(4,8,12-trimethyltridecyl)chroman isomers detected, 8-Me-2-methyl-2-(4,8,12-trimethyltridecyl)chroman presented with high abundance in sediments deposited in hypersaline environments, while absent in samples from normal marine environments. In contrast, 5,7,8-triMe-2-methyl-2-(4,8,12-trimethyltridecyl)chroman was more enriched in sediments from marine environments. This study also showed that the ratio of 5,7,8-triMe-/5,8-diMe-2-methyl-2-(4,8,12-trimethyltridecyl)chroman can be applied as a potential salinity indicator on account of a positive correlation with other 2-methyl-2-(4,8,12-trimethyltridecyl)chroman salinity indicators. This ratio can be an alternative indicator of paleosalinity when 8-Me-2-methyl-2-(4,8,12-trimethyltridecyl)chroman is absent or present in quite low abundance. The content of 2-methyl-2-(4,8,12-trimethyltridecyl)chroman isomers may be affected by freshwater supply and lithology. Molecular simulations showed that 5,8-diMe-2-methyl-2-(4,8,12-trimethyltridecyl)chroman has a higher thermal dynamic stability than 7,8-diMe-2-methyl-2-(4,8,12-trimethyltridecyl)chroman. Thus, the ratio of 5,8-diMe-2-methyl-2-(4,8,12-trimethyltridecyl)chroman/7,8-diMe-2-methyl-2-(4,8,12-trimethyltridecyl)chroman may be a potential maturity parameter for sediments at a low thermal mature stage.

This study was conducted to evaluate the effects of dietary supplementation of different recommended levels of Cu and Zn on antioxidant capacity, tissue mineral status, minerals excretion, meat quality, digestive enzyme activity, and metal transporters in finishing pigs. A total of 120 pigs (with an average initial body weight (BW) of 70.0 ± 2.1 kg) were randomly divided into four treatments: (1) basal diet without added Cu or Zn (control), (2) basal diet+35 mg cupreous N-carbamylglutamate chelate (NCG-Cu) +150 mg zinc-methionine chelate (Zn-Met) (AC), (3) basal diet + 3.0 mg of NCG-Cu + 43 mg Zn-Met (CN), and (4) basal diet + 3.5 mg NCG-Cu + 50 mg Zn-Met (NRC100). Pig growth performance was not affected by the level of Cu or Zn. Among the four treatments, the AC treatment had the highest concentration (P < 0.05) of glutathione peroxidase (GSH-Px). Pigs fed the AC diet had the highest (P < 0.05) liver Zn, fecal Cu, and fecal Zn among the four treatments. The protein levels of trypsin and aminopeptidase N (APN) in the intestinal mucosa showed their highest levels (P < 0.05) in the NRC100 and AC treatments. The mRNA levels of trypsinogen and APN were significantly up-regulated (P < 0.05) in the AC, CN, and NRC100 treatments compared with the control. The mRNA levels for the Zn transporter genes SLC30A1 (ZnT1) and SLC30A2 (ZnT2) were significantly up-regulated (P < 0.05) in the AC treatment, and the mRNA levels for SLC39A4 (ZIP4) and metallothionein 1 (MT) in the AC, CN, and NRC100 treatments were significantly up-regulated (P < 0.05) compared with the control. Meat quality were not affected (P > 0.05) by the different recommended levels of Cu and Zn. These results indicated that the supplemental Cu and Zn levels routinely used in AC diets in Chinese commercial feed enterprises should be reduced.

Hepatocellular carcinoma (HCC) is one of the most frequent malignancies and the third leading cause of cancer-related deaths worldwide. Besides, it has been revealed that long non-coding RNA (LncRNA) cancer susceptibility candidate 11 (CASC11) is involved in cancer progression. However, the functional role and underlying mechanism of CASC11 in HCC remains largely unknown. In this context, here, it was found that CASC11 was upregulated in HCC tissues and associated with tumor grades, metastasis, and prognosis of HCC patients. Functionally, CASC11 facilitated HCC cell proliferation, migration, and invasion in vitro, and enhanced tumor growth and metastasis in vivo. Mechanistically, CASC11 associated with and stabilized Ubiquitin-conjugating enzyme E2T (UBE2T) mRNA. To be specific, it decreased UBE2T N6-methyladenosine (m6A) level via recruiting ALKBH5. Moreover, CASC11 inhibited the association between UBE2T mRNA and m6A reader protein YTHDF2. Taken together, our findings demonstrate the epigenetic mechanism of CASC11 in the regulation of UBE2T expression and possibly provide a novel therapeutic target for HCC treatment.

Background: A recent study disclosed that ferroptosis was an important myocyte death style in myocardial infarction (MI). However, the diagnostic value of ferroptosis regulators and correlated underlying mechanisms in acute myocardial infarction (AMI) remain unknown.Methods: Bioinformatical analyses were conducted to identify the candidate biomarkers for AMI, and the collected local samples were used to validate the findings via real-time quantitative PCR. Bioinformatical analysis and luciferase reporter assay were implemented to identify the transcriptional factor. Transient transfection and ferroptosis characteristic measurement, including glutathione peroxidase 4, malondialdehyde, iron, and glutathione, was performed to verify the ability of the candidate gene to regulate the ferroptosis of cardiomyocytes. A meta-analysis was conducted in multiple independent cohorts to clarify the diagnostic value.Results: A total of 121 ferroptosis regulators were extracted from previous studies, and aldo-keto reductase family 1 member C3 (AKR1C3) was significantly downregulated in the peripheral blood samples of AMI cases from the analysis of GSE48060 and GSE97320. HOXB4 served as a transcriptional activator for AKR1C3 and could suppress the ferroptosis of the H9C2 cells treated with erastin. Besides this, peripheral blood samples from 16 AMI patients and 16 patients without coronary atherosclerotic disease were collected, where AKR1C3 and HOXB4 both showed a high diagnostic ability. Furthermore, a nomogram including HOXB4 and AKR1C3 was established and successfully validated in six independent datasets. A clinical correlation analysis displayed that AKR1C3 and HOXB4 were correlated with smoking, CK, CK-MB, and N-terminal-pro-B-type natriuretic peptide.Conclusion: Taken together, this study demonstrates that AKR1C3 and HOXB4 are promising diagnostic biomarkers, providing novel insights into the ferroptosis mechanisms of AMI.

The present study aimed firstly to use a set of crude oil samples and a dataset to provide new evidence for source input contribution in selected aromatic isomers for discrimination of oils from three oilfields from Tarim Basin and identify the key factor (s) controlling the isotope composition. Thus, the present research showed that the δ13C values of alkylnaphthalenes and alkylphenanthrenes plotted against P/DBT and Ga/C30H ratios is a reliable and convenient tool for discrimination of organic matter variations in different oilfields. More importantly, molecular ratios and different diagram plots revealed that the selected oil samples would be derived from a mixing of indigenous organic matter from the terrestrial (in Kuqa area) and marine (in the cratonic area) depositional environments prior the apparition of the Yakela Faulted Uplift. Thus, Daolaoba, Yakela, and Tahe oils were made up of organic materials from both marine and terrestrial sources. Furthermore, marine organic matter input dominates oils from the Tahe and Yakela, with a minor input from terrestrial sources. The oils from Daolaoba were assigned to be from a mixing of marine and terrestrial material inputs. The controlling factors assessment revealed that biodegradation has an insignificant effect on the set of oils; however, the source input and the thermal maturity together control the isotopic compositions of individual aromatic isomers from these three oilfields.

Background. At present, COVID-19 is sweeping the world, and all countries are actively responding. During the COVID-19 epidemic, the treatment of patients with acute myocardial infarction (AMI) may be affected. Methods. We reviewed data of patients with AMI from January 23 to April 23, 2020 (2020), and January 23 to April 23, 2019 (2019), who were admitted to two hospitals from Southern China. We collected clinical characteristics, comorbidities, treatment, prognosis, and key time segments to analyze. Results. The total number of patients that had been diagnosed with AMI in the two hospitals was 218 in 2020 and 260 in 2019. The number of AMI patients that were admitted to hospitals per day decreased in 2020. The percentage of patients with AMI who refused hospitalization in 2020 was significantly higher than that in 2019 (5.0% vs 1.5%, p=0.028). There is no statistical difference in symptoms of the first medical contact (S2FMC) time between 2020 and 2019 (p=0.552). Door-to-balloon (D2B) time of ST-elevation myocardial infarction (STEMI) patients who were treated with a primary percutaneous coronary intervention (pPCI) in 2020 was 79 (63.75–105.25) mins, while D2B time in 2019 was 57.5 (41.5–76.5) mins, which was statistically different from the two groups. Conclusions. COVID-19 had an impact on the number of AMI patients who were admitted to hospitals and the time of treatment. During the COVID-19 epidemic, the number of AMI patients that were admitted to hospitals per day was decreased, while the percentage of AMI patients that refused therapy in these two hospitals increased, and the D2B time of STEMI patients was also delayed.

The Paleogene lacustrine mudstone is one of the most important sets of source rocks in Termit Basin, Niger. However, studies on the Paleogene source kitchen are scarce. In this study, the source rock types, spatial distribution and their hydrocarbon generative potential within the Paleogene Sokor-1 and LV formations are systematically evaluated. A total of two third-order sequences (SSQ1 and SSQ2, from the bottom to top) and six systems tracts were identified in the Sokor-1 Formation, while the LV Formation mainly comprises pure shale and is regarded as a compositive stratigraphic sequence with no need of subdivision. Six types of source rocks could be distinguished within the three depositional environments of the sequence stratigraphic framework: (1) deep lake mudstones and shale deposited in the lacustrine deep-water facies, (2) shallow lake mudstones and carbonaceous mudstones occurring in the shallow lake environment, and (3) deltaic-front mudstones and prodeltaic mudstones developed in the deltaic facies. Deep lake mudstones/shale, which mainly occurred in the SSQ1-TST (transgressive systems tract), SSQ2-TST and LV Formation, are considered organic-rich source rocks with oil-prone generative potential. The shallow lake mudstones and deltaic mudstones were predominantly distributed in the tectonic slope and marginal areas of the Agadem Block within the lowstand systems tract and high stand systems tract of the SSQ1 and SSQ2. Nevertheless, geochemical results indicated that the deltaic mudstones are good gas-prone source rocks but the shallow lake mudstones were classified as poor potential source rocks. The variance in organic matter accumulation and preservation conditions within different depositional facies and the fluctuation of relative lake level are the controlling factors for the occurrence and distribution of high-quality source rocks. The effective Paleogene source kitchen is limitedly distributed around the depocenter of the Dinga Depression within a small scale, and therefore exploration for oil and gas resources from the Paleogene source kitchen should focus on the eastern Dinga fault-step zone adjacent to the depocenter of the Dinga Depression.

IntroductionKetamine, which is widely used in anesthesia, can induce cortical neurotoxicity in patients. This study aims to investigate the effects of long non-coding RNA LINC00641 on the ketamine-induced neural injury.Materials and MethodsIn this study, rat pheochromocytoma cells (PC12 cells) were used as a cell model and Sprague–Dawley postnatal day 7 rats were used for experiments in vivo. Ketamine-induced aberrant expression levels of LINC00641, miR-497-5p and brain-derived neurotrophic factor (BDNF) were examined by qRT-PCR. The effects of LINC00641 and miR-497-5p on ketamine-induced neural injury were then examined by MTT assays and TUNEL analysis. In addition, the activity of ROS and caspase-3 was measured. The regulatory relationships between LINC00641 and miR-497-5p, miR-497-5p and BDNF were detected by dual-luciferase reporter assay, respectively.ResultsKetamine induced the apoptosis of PC12 cells, accompanied by down-regulation of LINC00641 and BDNF, and up-regulation of miR-497-5p. LINC00641 overexpression enhanced the resistance to the apoptosis of PC12 cells, while transfection of miR-497-5p had opposite effects. Furthermore, LINC00641 could bind to miR-497-5p and reduce its expression, but indirectly increase the BDNF expression, which was considered as a protective factor in neural injury and activated TrkB/PI3K/Akt pathway.ConclusionCollectively, LINC00641/miR-497-5p/BDNF axis was validated to be an important signaling pathway in modulating ketamine-induced neural injury.

Oil reservoirs have been discovered in the Mesoproterozoic strata in the Liaoxi Depression, NE China. In order to determine the source of oil shows of the Mesoproterozoic Gaoyuzhuang Formation and their organic geochemical characteristics, eight source rocks and reservoir cores from the Mesoproterozoic Gaoyuzhuang Formation and four source rocks from the overlying Middle Jurassic Haifanggou Formation were geochemically analysed. The distribution patterns of normal alkanes, acyclic isoprenoids, hopanes, steranes and triaromatic steroids of the Mesoproterozoic hydrocarbons from Well N-1 are consistent with those of source rock extracts from the Mesoproterozoic Gaoyuzhuang Formation in the Well L-1. The molecular marker compositions of source rock extracts from the overlying Middle Jurassic Haifanggou Formation are distinctively different from those of the Mesoproterozoic hydrocarbons. The results suggest that the Mesoproterozoic source rocks have significant petroleum generation potential. The Mesoproterozoic paleo-reservoir may be prospecting exploration targets in the Liaoxi Depression, NE China.

The Paleogene sandstone reservoir in the Huangtong Sag of the Fushan Depression is one of the most commercially prolific oil accumulations in the Beibuwan Basin, South China Sea. In this study, twenty-seven crude oil samples from four oilfields in the sag were geochemically analyzed and classified into three oil groups. Oils in the Meitai and Hongguang oilfields show similar geochemical characteristics, with relatively abundant C30 4α-methyl-24-ethylcholestanes and low contents of oleanane and C19+20 tricyclic terpanes (TT), and therefore may be derived from the same source kitchen. Oils from the Yong’an oilfield have a greater proportion of oleanane and C19+20 TT compounds. Oils from the Chaoyang oilfield have intermediate contents of these biomarkers and proportionate values of other related parameters. The unimodal distribution pattern of homogenization temperatures of fluid inclusions found in all the Paleogene reservoirs indicates a single episode of charging. Combining this understanding with the reconstruction of stratigraphy-burial and geothermal histories by 1-D basin modeling, the petroleum charging time was determined to be between 8–2 Ma (the end of the middle Miocene to the early Pliocene). Hydrocarbon migration orientation and charging pathways were traced using molecular indicators (4-/1-methyldibenzothiophene and 1-/4-methodibenzofuran). In the Chaoyang oilfield, the values of these geochemical parameters decreased with decreasing burial depth, indicating vertical oil migration along faults, which are plentiful in the sag. Sandstone bodies with deeper burial depths may therefore be the most promising exploration targets in the Chaoyang area. The Yong’an oils generally migrate from the northwest toward the southeast, suggesting that the source kitchen for the Yong’an oilfield is in the northwest. By similar inference, the hydrocarbon source kitchen for the Hongguang-Meitai area is likely to be on the north side of the Hongguang-Meitai area. By tracing these hydrocarbon charging pathways, the northwest of the Yong’an area and the northern part of the Hongguang-Meitai area are identified as the most promising areas for exploration.

In-depth understanding and analysis of turbulent convection heat transfer of supercritical water under semicircular heating conditions play a major role in system design and security. The inaccurate numerical results on simulating the buoyancy effect under deterioration heat transfer cases are partly attributed to the invalidity of the turbulent model. An improved turbulence model, which is validated suitable to three-dimensional model, is adopted in the present paper to numerical simulated flow and heat transfer in a vertical tube under semicircular heating condition. Heat transfer deterioration phenomenon occurs under semicircular heating condition, while the degree of deterioration is weakened due to the influence of variable physical properties and buoyancy effect. The velocity profile is distorted into “M-shape” in the heating side and present parabolic distribution in the adiabatic side, leading to different deterioration mechanisms under semicircular heating condition compared with uniform heating. The larger density difference between the heating side and the adiabatic side increases the shear stress production of turbulent kinetic energy; turbulent development is much faster recovery than the phenomenon in uniform heating condition. The results show that the semicircular heating condition can effectively alleviate the degree of heat transfer deterioration in a vertical tube.
