Speaking English in a face-to-face classroom versus an online classroom may make EFL students feel uneasy in different ways, depending on a variety of factors. To help students reduce anxiety, teachers must understand the different levels of anxiety they experience in different situations. This study investigated the differences in English-speaking anxiety between face-to-face and an online classroom for Thai students, and used questionnaires and structured interviews to identify the factors that may contribute to the anxiety of different students. The questionnaire was completed by 26 first-year undergraduates from a Thai public university who had both face-to-face and online classroom experience in order to compare anxiety levels in the two learning environments. Seven students were then interviewed in order to determine the factors that may have contributed to these differences. The data from the questionnaire were presented as mean scores and standard deviation (SD) in order to compare the differences in students' anxiety between on-site and online classrooms, whereas the data from the interviews were used to explain the factors that could cause these differences. The results of the questionnaire revealed that, with the exception of confidence, students' speaking anxiety in face-to-face classrooms is greater in every respect than in online classrooms. In the meantime, the results of the interviews revealed that nervousness, a limited vocabulary, and an inability to remember vocabulary could be significant contributors to anxiety differences. The discussion explained how each aspect and factor makes speaking anxiety in on-site classrooms greater than in online classrooms, and suggested and encouraged teachers to reduce students' anxiety in both types of classrooms. Teachers can improve the language learning outcomes of their students by creating a less stressful environment in the English classroom and by incorporating activities that motivate or engage students to practice speaking.

Since late 2007, several outbreaks of porcine epidemic diarrhea virus (PEDV) infection have emerged in Thailand. Phylogenetic analysis places all Thai PEDV isolates during the outbreaks in the same clade as the Chinese strain JS-2004-2. Abstract Porcine circovirus type 2 (PCV2) is the major swine pathogen associated with Porcine circovirus associated disease (PCVAD) including post-weaning multisystemic wasting syndrome (PMWS). Currently, there are 4 subtypes of PCV2 (PCV2a, b, c and d) and some epidemiological evidences demonstrated that virulence of PCV2 may relate to its subtypes. Recently, PMWS was observed more frequently in swine farms in Thailand; however, the information regarding to PCV2 subtype involved was limited. Therefore, this study was aimed to determine the association between occurrence of PMWS and PCV2 subtypes as well as genetically characterize PCV2 in Thailand. PCV2 DNA was isolated from faecal swabs and whole blood of piglets from PMWS-affected and -negative farms. The full length ORF2 sequences were compared using multiple alignment. The results showed that PCV2 DNA was detected more frequently in PMWS-affected farms. The nucleotide identities of the ORF2 from 9 PCV2 isolates representing each PMWS-affected farm and one from the negative farm ranged from 92.4 to 99.5% suggesting that there is some genetic variation of PCV2 in Thai swine. The 10 PCV2 isolates were classified into 2 clusters, in which the 7 isolates from PMWS-positive farms were in PCV2b cluster 1 A/B. The remaining isolates were separated in the new subtype called PCV2e. The results suggest the presence of new PCV2 subtypes in addition to PCV2a and PCV2b in Asian swine population. However, correlation between subtypes and virulence of PCV2 infection is not conclusive due to limited number of the PCV2 sequences from PMWS negative farms.

In this paper, we propose hot-wire plasma welding, a combination of the plasma welding (PAW) process and the hot-wire process in the additive manufacturing (AM) process. Generally, in plasma welding for AM processes, the deposit grain size increases, and the hardness decreases as the wall height increases. The coarse microstructure, along with the large grain size, corresponds to an increase in deposit temperature, which leads to poorer mechanical properties. At the same time, the hot-wire laser process seems to contain an overly high interstitial amount of oxygen and nitrogen. With an increasing emphasis on sustainability, the hot-wire plasma welding process offers significant advantages: deeper and narrow penetration than the cold-wire plasma welding, improved design flexibility, large deposition rates, and low dilution percentages. Thus, the hot-wire plasma welding process was investigated in this work. The wire used in the welding process was a titanium American Welding Society (AMS) 4951F (Grade 2) welding wire (diameter 1.6 mm), in which the welding was recorded in real time with a charge-coupled device camera (CCD camera). We studied three parameters of the hot-wire plasma welding process: (1) the welding speed, (2) wire current, and (3) wire feeding speed. The mechanical and physical properties (porosity, Vickers hardness, microstructure, and tensile strength) were examined. It was found that the number of layers, the length and width of the molten pool, and the width of the deposited bead increased, while the height of the layer increased, and the hot-wire current played an important role in the deposition. In addition, these results were benchmarked against specimens created by a hot-wire plasma welding/wire-based additive manufacturing process with an intention to develop the hot-wire PAW process as a potential alternative in the additive manufacturing industry.

Background and Aim: The emerging of antimicrobial-resistant foodborne bacteria is a serious public health concern worldwide. This study was conducted to determine the association between farm management systems and antimicrobial resistance profiles of Escherichia coli isolated from conventional swine farms and natural farms. E. coli isolates were evaluated for the minimum inhibitory concentration (MIC) of 17 antimicrobials, extended-spectrum beta-lactamase (ESBL)-producing enzymes, and plasmid-mediated colistin-resistant genes., Materials and Methods: Fecal swabs were longitudinally collected from healthy pigs at three stages comprising nursery pigs, fattening pigs, and finishers, in addition to their environments. High-generation antimicrobials, including carbapenem, were selected for the MIC test. DNA samples of colistin-resistant isolates were amplified for mcr-1 and mcr-2 genes. Farm management and antimicrobial applications were evaluated using questionnaires., Results: The detection rate of ESBL-producing E. coli was 17%. The highest resistance rates were observed with trimethoprim/sulfamethoxazole (53.9%) and colistin (48.5%). All isolates were susceptible to carbapenem. Two large intensive farms that used colistin-supplemented feed showed the highest colistin resistance rates of 84.6% and 58.1%. Another intensive farm that did not use colistin showed a low colistin resistance rate of 14.3%. In contrast, a small natural farm that was free from antimicrobials showed a relatively high resistance rate of 41.8%. The majority of colistin-resistant isolates had MIC values of 8 mg/mL (49%) and ≥16 mg/mL (48%). The genes mcr-1 and mcr-2 were detected at rates of 64% and 38%, respectively, among the colistin-resistant E. coli ., Conclusion: Commensal E. coli were relatively sensitive to the antimicrobials used for treating critical human infections. Colistin use was the primary driver for the occurrence of colistin resistance in swine farms having similar conventional management systems. In the natural farm, cross-contamination could just occur through the environment if farm biosecurity is not set up carefully, thus indicating the significance of farm biosecurity risk even in an antimicrobial-free farm., (Copyright: © Ketkhao, et al.)

Long-term use of colistin for preventing Gram-negative bacterial infections in food animals was prohibited in Thailand in 2017, but it is permitted for short-term treatment. This study aimed to investigate association between the use of colistin for short-term treatment of infection and the emergence of colistin-resistant Enterobacteriaceae in swine. The current study was conducted at 2 selected swine farms in Thailand. Neither farm has used colistin to prevent infection for longer than 1 year. Rectal swabs were collected from the same 66 pigs at birth, and on days 7, 14, 21, 28, and 60. Colistin was used to treat sick pigs for up to 3 days. Additional rectal swabs were collected during colistin treatment. Rectal swabs were analyzed for colistin-resistant Enterobacteriaceae and the mcr-1 gene. Results revealed that colistin-resistant Enterobacteriaceae were absent at birth. Some pigs at both farms had diarrhea and received colistin treatment during days 2-27. Colistin-resistant Enterobacteriaceae were detected in 13.3-50.0% of sick and healthy pigs. No sick pigs were observed during days 28-60, and colistin was not used during that period. Colistin-resistant Enterobacteriaceae were detected in 2.8-10.0% of healthy pigs on day 28, and in 0-3.4% of healthy pigs on day 60. The mcr-1 gene was detected in 57.6% of colistin-resistant Enterobacteriaceae isolates. Short-term treatment with colistin was found to be associated with the emergence of colistin-resistant Enterobacteriaceae in swine. Colistin-resistant Enterobacteriaceae rapidly emerged after colistin use, and rapidly decreased or disappeared after its discontinuation., Competing Interests: The authors have declared that no competing interests exist.

A previous study demonstrated that a recombinant outer membrane protein H (rOmpH)-based intranasal fowl cholera vaccine elicited efficient homologous protection against the Pasteurella multocida strain X-73 (A:1) in chickens. The present study aimed to determine the cross-protectivity against heterologous P. multocida strains. The rOmpH was purified via electroelution and formulated with two kinds of adjuvants. The vaccine formulations in a total volume of 100 µl were 100 µg rOmpH with 3 µg of Escherichia coli enterotoxin B or 10 µg of CpG ODN2007. Chickens were assigned to three experimental groups depending on bacterial strain challenge exposure as well as three control groups. The chickens were immunized intranasally three times at three-week intervals. Challenge exposures were conducted by inoculation with homologous strain X-73 or heterologous strains P-1059 (A:3) or P-1662 (A:4) at four weeks after the final immunization. The specific antibody against rOmpH was produced in vaccinated birds. Sera IgY and secretory IgA antibody titres were significantly increased (P < 0.05) post-immunization. The stimulation index values of the vaccinated groups were significantly different from stimulation index values of the non-vaccinated groups (P < 0.05). Chicken survival rates after exposure to avian P. multocida strains ranged from 70% to 100%. There was no significant difference in protection between two kinds of adjuvants in vaccine formulations. Statistical analysis indicated no significant differences in protection among avian P. multocida strains challenge exposure. We conclude that an in-house rOmpH-based intranasal fowl cholera vaccine produced efficient cross-protectivity against heterologous strains of P. multocida.

Recombinant outer membrane protein H (rOmpH) of Pasteurella multocida strain X-73 can be purified using affinity chromatography but this adversely affects its immunogenicity. The current study presents the results from an intervention study comparing the immunogenicity of rOmpH purified using electroelution with rOmpH purified using affinity chromatography and native OmpH purified using electroelution and a nonimmunized control group. Chickens immunized with rOmpH purified using electroelution produced the highest ELISA antibody levels against P. multocida strains. Chickens in each of the 5 treatment groups were split into two subgroups for challenge with two different P. multocida strains. The average number of adhesions to CEF cells was statistically significantly lower in sera from chickens immunized with rOmpH or native OmpH purified using electroelution than in those of the three other treatment groups. The survival amongst chickens immunized with rOmpH or native OmpH purified using electroelution indicated high levels of protection. In contrast, survival probability was zero or low in the groups immunized with rOmpH purified using affinity chromatography and in the nonimmunized group. These findings show that the rOmpH purified using electroelution retains its immunogenicity and stimulates high levels of protection in chickens against P. multocida infection.

Recombinant outer membrane protein H (rOmpH) is a potential fowl cholera vaccine candidate. The present study was aimed at developing rOmpH formulations for intranasal administration. The rOmpH was purified and formulated with either Escherichia coli enterotoxin B (LTB) or CpG oligodeoxynucleotides (ODN) as an adjuvant. Antibody responses in chickens intranasally immunized with rOmpH in combination with 2 different adjuvants were significantly increased (P<0.05) post immunization. Chicken survival rates showed that rOmpH formulated with ODN and LTB elicited 90% and 70% protection, respectively. Our findings indicated that rOmpH formulated with ODN elicited protection better than that formulated with LTB. Therefore, the vaccines formulations in the present study can be considered new intranasal vaccine formulations for fowl cholera in chickens.
