Therapeutic drug monitoring (TDM) is increasingly relevant for an individualized antibiotic therapy and subsequently a necessary tool to reduce multidrug-resistant pathogens, especially in light of diminishing antimicrobial capabilities. Critical illness is associated with profound pharmacokinetic and pharmacodynamic alterations, which challenge dose finding and the application of particularly hydrophilic drugs such as β-lactam antibiotics. Methods: Implementation strategy, potential benefit, and practicability of the developed standard operating procedures were retrospectively analyzed from January to December 2020. Furthermore, the efficacy of the proposed dosing target of piperacillin in critically ill patients was evaluated. Results: In total, 160 patients received piperacillin/tazobactam therapy and were subsequently included in the study. Of them, 114 patients received piperacillin/tazobactam by continuous infusion and had at least one measurement of piperacillin serum level according to the standard operating procedure. In total, 271 measurements were performed with an average level of 79.0 ± 46.0 mg/L. Seventy-one piperacillin levels exceeded 100 mg/L and six levels were lower than 22.5 mg/L. The high-level and the low-level group differed significantly in infection laboratory parameters (CRP (mg/dL) 20.18 ± 11.71 vs. 5.75 ± 5.33) and renal function [glomerular filtration rate (mL/min/1.75 m2) 40.85 ± 26.74 vs. 120.50 ± 70.48]. Conclusions: Piperacillin levels are unpredictable in critically ill patients. TDM during piperacillin/tazobactam therapy is highly recommended for all patients. Although our implementation strategy was effective, further strategies implemented into the daily clinical workflow might support the health care staff and increase the clinicians’ alertness.

This research investigated the influence of the layered structure of paper on its selected strength properties. Two types of cellulose pulp were used: unbleached kraft pine pulp (UBSK) and unrefined bleached birch kraft pulp (BHK). Multi-ply papers were obtained from layers of appropriate grammage formed separately and bonded in a wet state. Paper layers were formed from primary pulps (UBSK and BHK) or their mixtures (three-ply UBSK-BHK paper and three-ply UBSK-BHK-UBSK paper), from which various variants of laboratory paper samples were made. This work focused on three-layer papers. The obtained laboratory sheets were analysed for changes in tear resistance, tensile strength, bursting strength, short span compression strength (SCT), and corrugating medium test (CMT). In most cases, three-layer structures were characterised by higher strength parameters than single-layer papers. The tear strength and SCT of papers consisting of three layers formed from both mixed pulps were slightly lower than those of solid papers. The results demonstrate the feasibility of producing three-ply paper with significantly reduced variability in parameters such as elongation and bursting strength, which are inherently subject to high variability.

The SARS-CoV-2 Omicron variant, with 15 mutations in Spike receptor-binding domain (Spike-RBD), renders virtually all clinical monoclonal antibodies against WT SARS-CoV-2 ineffective. We recently engineered the SARS-CoV-2 host entry receptor, ACE2, to tightly bind WT-RBD and prevent viral entry into host cells ("receptor traps"). Here we determine cryo-EM structures of our receptor traps in complex with stabilized Spike ectodomain. We develop a multi-model pipeline combining Rosetta protein modeling software and cryo-EM to allow interface energy calculations even at limited resolution and identify interface side chains that allow for high-affinity interactions between our ACE2 receptor traps and Spike-RBD. Our structural analysis provides a mechanistic rationale for the high-affinity (0.53-4.2 nM) binding of our ACE2 receptor traps to Omicron-RBD confirmed with biolayer interferometry measurements. Finally, we show that ACE2 receptor traps potently neutralize Omicron and Delta pseudotyped viruses, providing alternative therapeutic routes to combat this evolving virus.

The Strategic Research Agenda (SRA; https://www.ejp-eurad.eu/publications/eurad-sra) of the European Joint Programme on Radioactive Waste Management (EURAD; https://www.ejp-eurad.eu/) describes the scientific and technical domains and sub-domains and knowledge management needs of common interest between EURAD participant organizations. Theme number 7 is entitled “Performance assessment, safety case development and safety analyses.” A list of research and development priorities and activities of common interest to be addressed within EURAD for theme 7 have been established. Amongst others, the Understanding and modelling of multi-physical Thermo-Hydro-Mechanical-Chemical coupled processes (THMC) occurring in radioactive waste disposal is a major and permanent issue to support optimization of design and safety case abstraction. To tackle this challenge a research work package entitled “DONUT: Development and improvement of numerical methods and tools for modelling coupled processes” has been conducted within the EURAD join programming initiative. The purpose of this work package is to improve/develop methods or numerical tools in order to go a step further in development of (i) relevant, performant and cutting-edge numerical methods that can easily be implemented in existing or new tools, in order to carry out high-performance computing to facilitate the study of highly coupled processes in large systems, (ii) numerical scale transition schemes for coupled processes, (iii) innovative numerical methods to carry out uncertainty and sensitivity analyses. In this paper the work carried out within the DONUT work package is put in perspective regarding the existing concept and literature on the field. It does not pretend to be exhaustive but rather to put emphasis on particular issues tackled during the project.

Background: Acral actinic keratosis (AK) lesions are considered difficult to treat, and published data for photodynamic therapy (PDT) on these lesions is limited. Thus, we evaluated sustained efficacy, safety, and satisfaction after PDT for AK on the hands. Methods: We analysed subgroup data for treatment on the hands from a randomised, double-blind, intra-individual phase III study. All participants previously underwent up to two field-directed red light PDTs with 10 % 5-aminolevulinic acid nanoemulsion gel (BF-200 ALA). Assessments included pain during PDT, clearance and recurrence rates, and satisfaction. Results: 24 participants treated on the hands were included; 21 participants were analysed. Complete clearance rates with BF-200 ALA were 90.9 % (lesion-based) and 76.2 % (per participant's side), both markedly higher than with vehicle. The lesion recurrence rate with BF-200 ALA was 29.0 %. Adverse events reflected the mode of action. Mean pain intensities were 4.8 ± 3.8 (BF-200 ALA) and 0.8 ± 2.1 (vehicle) on an 11-point numeric rating scale. Most participants (81.0 %) rated their satisfaction with BF-200 ALA as very good or good. Conclusion: This subgroup analysis indicates that PDT with BF-200 ALA provides a suitable treatment for AK lesions on the hands.

Chemical reactivity of the superheavy elements nihonium (Nh, element 113) and moscovium (Mc, element 115) has been studied by the gas-solid chromatography method using a new combined chromatography and detection setup. The Mc isotope, 288Mc, was produced in the nuclear fusion reaction of 48Ca ions with 243Am targets at the GSI Helmholtzzentrum Darmstadt, Germany. After isolating 288Mc ions in the gas-filled separator TASCA, adsorption of 288Mc and its decay product 284Nh on silicon oxide and gold surfaces was investigated. As a result of this work, the values of the adsorption enthalpy of Nh and Mc on the silicon oxide surface were determined for the first time, −∆HadsSiO2Mc=54−5+11 kJ/mol and −∆HadsSiO2Nh=58−3+8 kJ/mol (68% c.i.). The obtained −ΔHads values are in good agreement with results of advanced relativistic calculations. Both elements, Nh and Mc, were shown to interact more weakly with the silicon oxide surface than their lighter homologues Tl and Bi, respectively. However, Nh and Mc turned out to be more reactive than the neighbouring closed-shell and quasi-closed-shell elements copernicium (Cn, element 112) and flerovium (Fl, element 114), respectively. The established trend is explained by the influence of strong relativistic effects on the valence atomic orbitals of these elements.

Edible insects have been officially classified as food in the European Union since 2015. Currently, four insect species are approved for food use. However, no official method for detecting insects in food has been recognized to date. To establish a simple analytical method for insect detection in food, we developed an indirect sandwich (enzyme-linked lectin sorbent assay) ELLA specific for N-acetylglucosamine in chitin and chitosan polymers. The validation of the method demonstrated that the ELLA developed in this study is reliable for insect detection. The limit of detection (LOD) and quantification (LOQ) were 0.006 and 0.028 mg/mL, respectively. Intra-day precision ranged from 2.45% to 30.29%, and inter-day precision from 0.36% to 12.87%. Significant differences in the total amount of chitin and chitosan were observed among the insect products, processed insect products, and samples without any insect addition (p < 0.05).

Impinging air jets can be used to dewater, heat, and dry the web of tissue paper. High velocities of the air jets degrade the paper, and appropriate adjustments to the jet velocity and the distance of the nozzle from the surface of the wet web are crucial to obtain the highest quality product. This work investigated the correlation between the velocity of the air jet and the strength of paper subjected to the impingement method. Papers with an initial moisture content of 20% and various pulp mixes were tested, and the physical properties of papers were explored. After impinging an air jet, different tensile strength limits were obtained in the machine and cross directions. The paper had lower apparent density and higher roughness compared to classical pressing. The dependence of tensile strength and roughness on the fibers composition also was determined. Increasing the amount of eucalyptus fibers in impingement dewatered paper resulted in a decrease in its tensile strength and roughness. The value of elongation before breaking was the highest for softwood papers after the impingement method. The maximum velocity of an air jet that can be used to dewater or dry paper without the risk of damage to the papers was determined.

Background & Aims: Hepatic fibrosis is characterized by enhanced deposition of extracellular matrix (ECM), which results from the wound healing response to chronic, repeated injury of any etiology. Upon injury, hepatic stellate cells (HSCs) activate and secrete ECM proteins, forming scar tissue, which leads to liver dysfunction. Monocyte-chemoattractant protein-induced protein 1 (MCPIP1) possesses anti-inflammatory activity, and its overexpression reduces liver injury in septic mice. In addition, mice with liver-specific deletion of Zc3h12a develop features of primary biliary cholangitis. In this study, we investigated the role of MCPIP1 in liver fibrosis and HSC activation. Methods: We analyzed MCPIP1 levels in patients’ fibrotic livers and hepatic cells isolated from fibrotic murine livers. In vitro experiments were conducted on primary HSCs, cholangiocytes, hepatocytes, and LX-2 cells with MCPIP1 overexpression or silencing. Results: MCPIP1 levels are induced in patients’ fibrotic livers compared with their nonfibrotic counterparts. Murine models of fibrosis revealed that its level is increased in HSCs and hepatocytes. Moreover, hepatocytes with Mcpip1 deletion trigger HSC activation via the release of connective tissue growth factor. Overexpression of MCPIP1 in LX-2 cells inhibits their activation through the regulation of TGFB1 expression, and this phenotype is reversed upon MCPIP1 silencing. Conclusions: We demonstrated that MCPIP1 is induced in human fibrotic livers and regulates the activation of HSCs in both autocrine and paracrine manners. Our results indicate that MCPIP1 could have a potential role in the development of liver fibrosis.

Polymeric single chloride-ion conductor networks based on acrylic imidazolium chloride ionic liquid monomers AACXImCYCl as reported previously are prepared. The chemical structure of the polymers is varied with respect to the acrylic substituents (alkyl spacer and alkyl substituent in the imidazolium ring). The networks are examined in detail with respect to the influence of the chemical structure on the resulting properties including thermal behavior, rheological behavior, swelling behavior, and ionic conductivity. The ionic conductivities increase (by two orders of magnitude from 10−6 to 10−4 S·cm−1 with increasing temperature), while the complex viscosities of the polymer networks decrease simultaneously. After swelling in water for 1 week the ionic conductivity reaches values of 10−2 S·cm−1. A clear influence of the spacer and the crosslinker content on the glass transition temperature was shown for the first time in these investigations. With increasing crosslinker content, the Tg values and the viscosities of the networks increase. With increasing spacer length, the Tg values decrease, but the viscosities increase with increasing temperature. The results reveal that the materials represent promising electrolytes for batteries, as proven by successful charging/discharging of a p(TEMPO-MA)/zinc battery over 350 cycles.

Laser resonance ionization spectroscopy was performed on the rare einsteinium isotopes Es253-255 at the RISIKO mass separator in Mainz. With low sample sizes ranging down to femtograms, the prominent 352 nm-ground-state transition was measured in all three einsteinium isotopes, and four additional ground-state transitions were measured in Es254. Hyperfine-structure analysis resulted in assigned spin values of I(Es254)=7 and I(Es255)=7/2. From the extracted coupling constants, nuclear magnetic dipole moments of μI(Es254)=3.42(7)μN and μI(Es255)=4.14(10)μN as well as spectroscopic electric quadrupole moments of Qs(Es254)=9.6(1.2)eb and Qs(Es255)=5.1(1.7)eb were derived. Our value for Es254 deviates from the value of |μI(Es254)|=4.35(41)μN extracted from the angular anisotropy of α-radiation emitted by Es254.

The teaching and learning of quantum physics has recently become a topic of increasing interest in physics education research. In particular, the study of two-state systems is gaining importance as a means of teaching quantum physics at various educational levels. Meanwhile, a number of approaches have been developed that are also suitable for high school students. It can be assumed that the different approaches have different degrees of effectiveness in teaching central quantum concepts. However, suitable evaluation instruments to test this are still lacking. Therefore, as a first step, a short questionnaire on quantum measurement, suitable for both research and classroom use, was developed in several steps. First, a questionnaire with open and closed items was created and piloted with a total of N=120 learners. The responses were evaluated qualitatively using a comprehensive coding manual, which provided insights into learners’ conceptions. These results led to the development of an eight-item questionnaire that could be adapted to different teaching approaches. This questionnaire was subjected to expert review and, finally, successfully tested for its psychometric properties with a sample of N=201 learners. Overall, our results provide initial empirical evidence that context (i.e., which two-state approach is used) does matter for student learning, but in general, two-state approaches appear to be particularly conducive to learning quantum concepts (specified in this article for quantum measurement) compared to traditional instruction.

The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) virus enters host cells via an interaction between its Spike protein and the host cell receptor angiotensin-converting enzyme 2 (ACE2). By screening a yeast surface-displayed library of synthetic nanobody sequences, we developed nanobodies that disrupt the interaction between Spike and ACE2. Cryo-electron microscopy (cryo-EM) revealed that one nanobody, Nb6, binds Spike in a fully inactive conformation with its receptor binding domains locked into their inaccessible down state, incapable of binding ACE2. Affinity maturation and structure-guided design of multivalency yielded a trivalent nanobody, mNb6-tri, with femtomolar affinity for Spike and picomolar neutralization of SARS-CoV-2 infection. mNb6-tri retains function after aerosolization, lyophilization, and heat treatment, which enables aerosol-mediated delivery of this potent neutralizer directly to the airway epithelia.

The SARS-CoV-2 protein Nsp2 has been implicated in a wide range of viral processes, but its exact functions, and the structural basis of those functions, remain unknown. Here, we report an atomic model for full-length Nsp2 obtained by combining cryo-electron microscopy with deep learning-based structure prediction from AlphaFold2. The resulting structure reveals a highly-conserved zinc ion-binding site, suggesting a role for Nsp2 in RNA binding. Mapping emerging mutations from variants of SARS-CoV-2 on the resulting structure shows potential host-Nsp2 interaction regions. Using structural analysis together with affinity tagged purification mass spectrometry experiments, we identify Nsp2 mutants that are unable to interact with the actin-nucleation-promoting WASH protein complex or with GIGYF2, an inhibitor of translation initiation and modulator of ribosome-associated quality control. Our work suggests a potential role of Nsp2 in linking viral transcription within the viral replication-transcription complexes (RTC) to the translation initiation of the viral message. Collectively, the structure reported here, combined with mutant interaction mapping, provides a foundation for functional studies of this evolutionary conserved coronavirus protein and may assist future drug design.

Abstract Background CD36 has been identified as a potential therapeutic target both in leukemic cells and in the tumor immune microenvironment. In acute myeloid leukemia (AML), we found that APOC2 acts with CD36 to promote leukemia growth by activating the LYN-ERK signaling. CD36 also plays a role in lipid metabolism of cancer associated T-cells leading to impaired cytotoxic CD8+ T-cell and enhanced Treg cell function. To establish CD36 as a viable therapeutic target in AML, we investigated whether targeting CD36 has any detrimental impact on normal hematopoietic cells. Methods Differential expression data of CD36 during human and mouse normal hematopoiesis were examined and compared. Cd36 knockout (Cd36-KO) mice were evaluated for blood analysis, hematopoietic stem cells and progenitors (HSPCs) function and phenotype analyses, and T cells in vitro expansion and phenotypes in comparison with wild type (WT) mice. In addition, MLL-PTD/FLT3-ITD leukemic cells were engrafted into Cd36-KO and WT mice, and leukemia burden was compared between groups. Results RNA-Seq data showed that Cd36 expression was low in HSPCs and increased as cells matured. Abstract It has been well-established that mutations in BRCA1 and BRCA2, compromising functions in DNA double-strand break repair (DSBR), confer hereditary breast and ovarian cancer risk. Importantly, mutations in these genes explain only a minor fraction of the hereditary risk and of the subset of DSBR deficient tumors. Our screening efforts identified two truncating germline mutations in the gene encoding the BRCA1 complex partner ABRAXAS1 in German early-onset breast cancer patients. To unravel the molecular mechanisms triggering carcinogenesis in these carriers of heterozygous mutations, we examined DSBR functions in patient-derived lymphoblastoid cells (LCLs) and in genetically manipulated mammary epithelial cells. By use of these strategies we were able to demonstrate that these truncating ABRAXAS1 mutations exerted dominant effects on BRCA1 functions. Interestingly, we did not observe haploinsufficiency regarding homologous recombination (HR) proficiency (reporter assay, RAD51-foci, PARP-inhibitor sensitivity) in mutation carriers. However, the balance was shifted to use of mutagenic DSBR-pathways. The dominant effect of truncated ABRAXAS1 devoid of the C-terminal BRCA1 binding site can be explained by retention of the N-terminal interaction sites for other BRCA1-A complex partners like RAP80. In this case BRCA1 was channeled from the BRCA1-A to the BRCA1-C complex, which induced single-strand annealing (SSA). Further truncation, additionally deleting the coiled-coil region of ABRAXAS1, unleashed excessive DNA damage responses (DDRs) de-repressing multiple DSBR-pathways including SSA and non-homologous end-joining (NHEJ). Our data reveal de-repression of low-fidelity repair activities as a common feature of cells from patients with heterozygous mutations in genes encoding BRCA1 and its complex partners.

Honey contains a wide range of inorganic substances. Their content can be influenced, i.e., by the type of soil on which the bee pasture is located. As part of this study, the mineral profile of 32 samples of honey from hobby beekeepers from the Czech Republic wasevaluated and then compared with soil types in the vicinity of the beehive location. Pearson’s correlation coefficient was used to express the relationship between mineral substances and soil type. There was a high correlation between antroposol and Zn (R = 0.98), Pb (R = 0.96), then between ranker and Mn (0.95), then regosol and Al (R = 0.97) (p < 0.05). A high negative correlation was found between regosol and Mg (R = −0.97), Cr (R = −0.98) and between redzinas and Al (R = −0.97) (p < 0.05). Both positive and negative high correlations were confirmed for phaeozem. The CART method subsequently proved that the characteristic elements for individual soil types are B, Ca, Mg, Ni, and Mn. The soil types of cambisol, fluvisol, gleysol, anthrosol, and kastanozem had the closest relationship with the elements mentioned, and it can therefore be assumed that their occurrence indicates the presence of these soil types within the range of beehive location.

This article reports on the extraction of palladium(II) from hydrochloric acid (HCl) solutions using polymer inclusion membranes (PIMs) containing tetrabutylammonium bromide (TBAB) as the ion carrier. The membranes were based on cellulose triacetate (CTA) as the polymer support. The main aim of this study is to determine the possibility of TBAB’s application as the effective ion carrier/extractant of Pd(II) from hydrochloric acid solutions. At first, the effect of the hydrochloric acid concentration in the aqueous phase on palladium(II) extraction was investigated. Next, cellulose triacetate membranes with TBAB as the carrier were prepared and applied for the recovery of Pd(II) from HCl solutions. As a result of the investigations, the optimal composition of the receiving phase was determined to be 0.5 M thiourea in 0.1 M hydrochloric acid. The effect of the acid concentration in the source phase was investigated. The results show a linear decrease in the permeability coefficient and initial flux of palladium(II) with an increase in the hydrochloric acid concentration in the source phase. The separation of Pd(II) from Pt(IV) from the hydrochloric acid solution was also studied. The transport rate of Pd(II) was higher than Pt(IV). The separation coefficient SPd/Pt was 1.3. The results show that transport through PIMs with TBAB can be used as an effective method to recover Pd(II) from hydrochloric acid, especially at a low concentration of this acid.

ABSTRACTThe aim of this study was evaluated the effect of selected bioactive substances and nanoparticles on the immunoreactivity of edible packages based on chitosan using the ELISA. The analysed protein was the tropomyosin. The results confirmed the presence of the tropomyosin (3.77 ± 0.79–5.75 ± 0.01 µg/g) in control samples. This study demonstrated that the bioactive substances in the form of grape (0.61 ± 0.34–0.43 ± 0.16 µg/g), blueberry (0.58 ± 0.32–0.39 ± 0.27 µg/g), and parsley extracts (2.09 ± 1.28–0.79 ± 0.40 µg/g) reduces immunoreactivity (p 0.05) on immunoreactivity. An upgraded TASISpec decay station was placed behind the gas-filled separator TASCA at the GSI Helmholtzzentrum für Schwerionenforschung in Darmstadt, Germany. The fusion-evaporation reactions ^{48}Ca+^{242}Pu and ^{48}Ca+^{244}Pu provided a total of 32 flerovium-candidate decay chains, of which two and eleven were firmly assigned to ^{286}Fl and ^{288}Fl, respectively. A prompt coincidence between a 9.60(1)-MeV α particle event and a 0.36(1)-MeV conversion electron marked the first observation of an excited state in an even-even isotope of the heaviest man-made elements, namely ^{282}Cn. Spectroscopy of ^{288}Fl decay chains fixed Q_{α}=10.06(1)  MeV. In one case, a Q_{α}=9.46(1)-MeV decay from ^{284}Cn into ^{280}Ds was observed, with ^{280}Ds fissioning after only 518  μs. The impact of these findings, aggregated with existing data on decay chains of ^{286,288}Fl, on the size of an anticipated shell gap at proton number Z=114 is discussed in light of predictions from two beyond-mean-field calculations, which take into account triaxial deformation.

Neutralizing agents against SARS-CoV-2 are urgently needed for the treatment and prophylaxis of COVID-19. Here, we present a strategy to rapidly identify and assemble synthetic human variable heavy (VH) domains toward neutralizing epitopes. We constructed a VH-phage library and targeted the angiotensin-converting enzyme 2 (ACE2) binding interface of the SARS-CoV-2 Spike receptor-binding domain (Spike-RBD). Using a masked selection approach, we identified VH binders to two non-overlapping epitopes and further assembled these into multivalent and bi-paratopic formats. These VH constructs showed increased affinity to Spike (up to 600-fold) and neutralization potency (up to 1,400-fold) on pseudotyped SARS-CoV-2 virus when compared to standalone VH domains. The most potent binder, a trivalent VH, neutralized authentic SARS-CoV-2 with a half-maximal inhibitory concentration (IC50) of 4.0 nM (180 ng ml-1). A cryo-EM structure of the trivalent VH bound to Spike shows each VH domain engaging an RBD at the ACE2 binding site, confirming our original design strategy.

The impact of these findings, aggregated with existing data on decay chains of ^{286,288}Fl, on the size of an anticipated shell gap at proton number Z=114 is discussed in light of predictions from two beyond-mean-field calculations, which take into account triaxial deformation.

An upgraded TASISpec setup, with the addition of a veto DSSD and the new Compex detector-germanium array, has been employed with the gas-filled recoil separator TASCA at the GSI Helmholtzzentrum für Schwerionenforschung Darmstadt, to study flerovium (element 114) decay chains. The detector upgrades along with development of new analytical techniques have improved the sensitivity of the TASISpec setup for measuring α-photon coincidences. These improvements have been assessed with test reactions. The reaction 48Ca+206,207Pb was used for verification of experimental parameters such as transmission to implantation DSSD and target-segment to α-decay correlations. The reaction 48Ca+ nat Hf was used to produce several short-lived nuclei with multiple-α decay chains to investigate pile-up event deconvolution.

We study the equation of state of symmetric nuclear matter at zero temperature over a wide range of densities using two complementary theoretical approaches. At low densities, up to twice nuclear saturation density, we compute the energy per particle based on modern nucleon-nucleon and three-nucleon interactions derived within chiral effective field theory. For higher densities, we derive for the first time constraints in a Fierz-complete setting directly based on quantum chromodynamics using functional renormalization group techniques. We find remarkable consistency of the results obtained from both approaches as they come together in density and the natural emergence of a maximum in the speed of sound c_{S} at supranuclear densities. The presence of this maximum appears tightly connected to the formation of a diquark gap. Notably, this maximum is observed to exceed the asymptotic value c_{S}^{2}=1/3 while its exact position in terms of the density cannot yet be determined conclusively.

BackgroundAllogeneic hematopoietic stem cell transplant remains the most effective strategy for patients with high-risk acute myeloid leukemia (AML). Leukemia-specific neoantigens presented by the major histocompatibility complexes (MHCs) are recognized by the T cell receptors (TCR) triggering the graft-versus-leukemia effect. A unique TCR signature is generated by a complex V(D)J rearrangement process to form TCR capable of binding to the peptide-MHC. The generated TCR repertoire undergoes dynamic changes with disease progression and treatment.MethodHere we applied two different computational tools (TRUST4 and MIXCR) to extract the TCR sequences from RNA-seq data from The Cancer Genome Atlas (TCGA) and examine the association between features of the TCR repertoire in adult patients with AML and their clinical and molecular characteristics.ResultsWe found that only ~30% of identified TCR CDR3s were shared by the two computational tools. Yet, patterns of TCR associations with patients’ clinical and molecular characteristics based on data obtained from either tool were similar. The numbers of unique TCR clones were highly correlated with patients’ white blood cell counts, bone marrow blast percentage, and peripheral blood blast percentage. Multivariable regressions of TCRA and TCRB median normalized number of unique clones with mutational status of AML patients using TRUST4 showed significant association of TCRA or TCRB with WT1 mutations, WBC count, %BM blast, and sex (adjusted in TCRB model). We observed a correlation between TCRA/B number of unique clones and the expression of T cells inhibitory signal genes (TIGIT, LAG3, CTLA-4) and foxp3, but not IL2RA, CD69 and TNFRSF9 suggestive of exhausted T cell phenotypes in AML.ConclusionBenchmarking of computational tools is needed to increase the accuracy of the identified clones. The utilization of RNA-seq data enables identification of highly abundant TCRs and correlating these clones with patients’ clinical and molecular characteristics. This study further supports the value of high-resolution TCR-Seq analyses to characterize the TCR repertoire in patients.

Abstract The interest in bio‐based alternatives to classical polyesters such as poly(ethylene terephthalate) (PET) and poly(butylene terephthalate) (PBT) is steadily growing to achieve a more sustainable approach to polymer materials. In this study, PBT/poly(butylene furanoate) (PBF) blends are prepared, characterized and extrusion foamed. PBF as a bio‐based polyester offers two advantages. The ecological footprint of the material is reduced, and additionally, it can be used in Diels‐Alder reactions at the blend surface to support fusion of the foamed beads. The blending behavior of the polyesters is investigated using samples prepared in a microcompounder, particularly focused on the miscibility of the blends and transesterification reactions. The blends are thermodynamically immiscible but show a certain degree of transesterification according to nuclear magnetic resonance (NMR) spectroscopy. The morphology of blend beads produced by an extrusion foaming process is analyzed regarding their cell density, cell size distribution, and open‐cell content. It is shown that PBF has a positive effect on the bead foam morphology. The use of a bifunctional linker designed for chemical fusion of the bead surfaces allows to obtaining of molded parts, in contrast to beads containing pure PBT.

The possible origin of the short-decay chains is discussed in comparison with the known experimental data. They are found to fit with the decay chain patterns attributed to Ts293.
